Spatially guided angiogenesis by laser-bioprinting

L'ingénierie tissulaire est reconnue comme une méthode potentielle pour réparer ou régénérer les tissus endommagés. Malgré de grandes avancées dans l'ingénierie tissulaire, la réussite de la construction de tissus complexes avec des réseaux vascularisés reste un défi. Dans les modèles d'angiogenèse actuels, les cellules endothéliales sont ensemencées au hasard, n'offrant pas de structure organisée. La technologie de bioimpression par laser offre une résolution d'impression précise. Par cette technique, les structures microvasculaires peuvent être construites pour la fabrication d'organes complexes, ou pour modéliser la progression de la maladie ou les modèles de réponse aux médicaments. Dans cette étude, des techniques de bio-impression au laser ont été utilisées pour étudier le guidage de l'angiogenèse in vitro. Deux techniques basées sur le laser, le transfert direct induit par laser (LIFT) et le transfert latéral induit par laser (LIST) sont utilisées. Comparée à LIFT, la technologie LIST offrait des conditions idéales pour l'impression cellulaire telles que la concentration cellulaire requise pour la formation du tubes endothéliaux et l'uniformité du motif désiré. Nous avons réalisé le modelage de la formation de structures de type capillaire dans des motifs organisés via l'impression LIST. Les constructions de type capillaire formées présentent des motifs uniformes. Les structures formées ont été analysées par microscopie confocale et reconstruction d'images 3D. Bien que le développement de la lumière endothéliale soit incomplet, la technique développée possède le potentiel d'atteindre une stabilisation et un développement de la lumière si l'on recrute un deuxième type de cellule tel que les fibroblastes ou les péricytes. / Tissue engineering has been well acknowledged as a potential method to repair or regenerate damaged tissues in the human body, fulfilling the limitations and shortage in autologous and organ transplantations. Despite great advances in engineering tissues with simple geometry and low requirement for oxygen and blood supply such as cartilage, skin and cornea, success in constructing 3D complex tissues with vascularized networks remains a major challenge. Angiogenesis plays an important role in vascular development in vivo. In current angiogenesis models, endothelial cells are seeded randomly not offering precise and desired patterning. Laser-based bioprinting technology offers precise and high cell printing resolution. By using laser-based bioprinting technology, microvascular structures can be constructed as a platform for complex organ fabrication, disease progression and drug response models. In this study, laser-based bioprinting techniques are employed to study angiogenesis guidance in vitro by patterning endothelial cells. Two laser-based techniques, Laser-Induced Forward Transfer (LIFT) and Laser-Induced Side Transfer (LIST) are used as patterning tools. Compared to LIFT, LIST technology provided ideal conditions for cell printing such as required cell concentration for endothelial tube formation and pattern uniformity. In this study, we achieved the guidance of capillary-like structure formation in desired patterns via LIST printing. The formed capillary-like constructs featured precise patterns and uniformity. The structures were analyzed by confocal microscopy, 3D image reconstruction and frozen section procedure. Though lumen development was incomplete, it possesses the potential to attain further stabilization and lumen development if recruiting a second cell type such as fibroblast or pericyte.

Semantic Federation of Musical and Music-Related Information for Establishing a Personal Music Knowledge Base

Music is perceived and described very subjectively by every individual. Nowadays, people often get lost in their steadily growing, multi-placed, digital music collection. Existing music player and management applications get in trouble when dealing with poor metadata that is predominant in personal music collections. There are several music information services available that assist users by providing tools for precisely organising their music collection, or for presenting them new insights into their own music library and listening habits. However, it is still not the case that music consumers can seamlessly interact with all these auxiliary services directly from the place where they access their music individually. To profit from the manifold music and music-related knowledge that is or can be available via various information services, this information has to be gathered up, semantically federated, and integrated into a uniform knowledge base that can personalised represent this data in an appropriate visualisation to the users. This personalised semantic aggregation of music metadata from several sources is the gist of this thesis. The outlined solution particularly concentrates on users’ needs regarding music collection management which can strongly alternate between single human beings. The author’s proposal, the personal music knowledge base (PMKB), consists of a client-server architecture with uniform communication endpoints and an ontological knowledge representation model format that is able to represent the versatile information of its use cases. The PMKB concept is appropriate to cover the complete information flow life cycle, including the processes of user account initialisation, information service choice, individual information extraction, and proactive update notification. The PMKB implementation makes use of SemanticWeb technologies. Particularly the knowledge representation part of the PMKB vision is explained in this work. Several new Semantic Web ontologies are defined or existing ones are massively modified to meet the requirements of a personalised semantic federation of music and music-related data for managing personal music collections. The outcome is, amongst others, • a new vocabulary for describing the play back domain, • another one for representing information service categorisations and quality ratings, and • one that unites the beneficial parts of the existing advanced user modelling ontologies. The introduced vocabularies can be perfectly utilised in conjunction with the existing Music Ontology framework. Some RDFizers that also make use of the outlined ontologies in their mapping definitions, illustrate the fitness in practise of these specifications. A social evaluation method is applied to carry out an examination dealing with the reutilisation, application and feedback of the vocabularies that are explained in this work. This analysis shows that it is a good practise to properly publish Semantic Web ontologies with the help of some Linked Data principles and further basic SEO techniques to easily reach the searching audience, to avoid duplicates of such KR specifications, and, last but not least, to directly establish a \"shared understanding\". Due to their project-independence, the proposed vocabularies can be deployed in every knowledge representation model that needs their knowledge representation capacities. This thesis added its value to make the vision of a personal music knowledge base come true.

An assessment of student's English vocabulary levels and an exploration of the vocabulary profile of teacher's spoken discourse in an international high school

In many international schools where English is the language of learning and teaching there are large percentages of students whose first language is not English. Many of these students may have low vocabulary levels which inhibits their chances of taking full advantage of their education. Low vocabulary levels can be a particular problem for students in mainstream classes where fluent English speaking teachers are using English to teach content areas of Mathematics, Science and History. Not only do students have to comprehend the low-frequency, academic and technical vocabulary pertaining to the subject, but they also need to know the higher frequency vocabulary that makes up general English usage. If students’ vocabulary levels fall too far below the vocabulary levels with which their teachers are speaking, then their chance of comprehending the topic is small, as is their chance of succeeding in their subjects. This study has two broad aims. Firstly, I have set out to assess the English vocabulary levels of students at an international school where English is the language of learning and teaching. The majority of students at this school do not have English as their first language. The second aim of this study is to explore the vocabulary profile of the teachers’ spoken discourse at the research school. By gaining a better understanding of the nature of teacher discourse – specifically the percentage of high, mid and low-frequency vocabulary, as well as academic vocabulary that they use – English as a Second Language (ESL) teachers will be in a stronger position to identify what the vocabulary learning task is and be able to assist students in reaching the vocabulary levels necessary to make sense of their lessons. This study revealed a large gap between the generally low vocabulary levels of ESL students and the vocabulary levels spoken by their teachers. As a result the need for explicit vocabulary instruction and learning is shown to be very important in English medium (international) schools, where there are large numbers of students whose first language is not English. / Linguistics and Modern Languages / M.A. (Applied Linguistics)

Summaries in Afrikaans and English / Hierdie studie spreek die emosionele skoolgereedheid van voorskoolse kinders aan. Die voorskoolse kind met sy basiese emosies vanuit sielkundig opvoedkundige perspektief word in die studie van naderby beskou. Tydens die studie is die basiese emosies volgens Plutchik (1980) wat bepalend vir die emosionele skoolgereedheid van die skooltoetreder is, geidentifiseer. Na aanleiding van nuwe insigte waartoe gekom is, het die navorser 'n eie emosionele skoolgereedheidsevalueringslys volgens Plutchik (1980) se basiese emosies antwerp, wat as 'n maatstaf vir die evaluering van emosionele skoolgereedheid kan dien. 'n Empiriese ondersoek is onderneem om te bepaal of 'n aantal proefpersone volgens n emosionele skoolgereedheidsevalueringslys emosioneel skoolgereed is al dan nie. Emosionele tekorte is vanuit die emosionele skoolgereedheidsevalueringslys geidentifiseer, waarna diagnostiese en terapeutiese tegnieke vir emosionele skoolgereedmaking kortliks bespreek is. Opvoedkundige sielkundiges kan die diagnostiese en terapeutiese tegnieke as hulpmiddel gebruik om emosionele tekorte, wat moontlik 'n blokkasie vir emosionele skoolgereedheid inhou, aan te spreek. / This stidu addresses the emotional school preparedness (readiness) of pre-primary children. The pre-primary child with his basic emotions from a psychological educational perspective is examined more closely in this study. During this study the basic emotions according to Plutchik {1980) that determine the emotional school readiness of the school entrant, were identified. On account of new insights gained, the researcher designed a school readiness evaluation list of her own according to Plutchik's (1980) basic emotions. This list can serve as a guideline for the evaluation of emotional school readiness. An empirical study was undertaken to determine whether a number of experimental subjects {children) were ready for school or not. Emotional deficiencies were identified by means of the emotional school readiness evaluation list, whereafter diagnostic and therapeutic techniques for the emotional preparation of children were briefly discussed. Educational psychologists can use these diagnostic and therapeutic techniques as an aid to address emotional deficiencies, which may impede the emotional school readiness of children. / Educational Studies / M. 從台美兩地北京奧運新聞報導看中國國家形象 / China's image in the coverage of 2008 Beijing-Olympic: A comparison of major newspapers in the US and in Taiwan

本研究透過分析比較台灣與美國重要報紙媒體上的北京奧運相關報導,檢視中國主辦2008年北京奧運會,所投射出來的媒體國家形象(projected national image),進而探討舉辦全球性媒體活動(global media events)做為國家形象管理策略一環的應用性。本研究挑選台灣的《聯合報》、《自由時報》,與美國的《紐約時報》、《華盛頓郵報》為兩地重要媒體的代表,並經過搜尋篩選程序之後,共得到375則新聞報導。再以內容分析法與框架清單(the list of the frame)分析法,對構成媒體國家形象的各個變項進行相關統計分析。 本研究主要得到三點發現。首先,就台灣與美國報紙整體而言,中國國家形象偏向負面,且報導方向與新聞框架、報導型式之間皆存有顯著關係,而該負面形象的來源,未必是直接與奧運會相關的新聞主題。其次,若將美國報紙與台灣報紙進行比較,則發現兩地的中國國家形象均呈現負面,而台灣報紙的整體報導量以及負面新聞主題較多。另外,本研究發現,同一個國家內的不同媒體之間,也有顯著的差異,《華盛頓郵報》與《自由時報》的負面報導多過中立報導,而《紐約時報》及《聯合報》則仍然以中立報導最多,且《自由時報》的新聞框架運用偏重在政治的角度,而《聯合報》則較多元。 最後,本研究認為北京奧運成功帶來了許多相關的正面主題與正面框架報導,與奧運活動本身較不相關的負面主題,則顯示良好的國家形象需要長期多方面的努力建構。 / To host global media events is one of the important kinds of strategies for the national image management. This study focuses on news coverage of 2008 Beijing-Olympic from the New York Times, Washington Post in the US, and the United Daily, Liberty Times in Taiwan to examine China's projected national image. The content analysis, and the list of the frame analysis were carried out to analyze several variables forming the projected national image in the 375 news items. Through statistic analysis, this study found three main points. First, for four newspapers, China's projected national image was negative. The valence was related to news frames and news style evidently, and also a few indirect news topics were the source of the negative image as well. Secondly, in comparison of newspapers in the US and in Taiwan, both images of china were negative. The total number of stories and negative news topics in Taiwan's newspapers were more than in the US counterpart. Thirdly, in terms of valence, Washington Post and Liberty Times had more negative news than neutral ones, but the neutral news were still the most in the New York Times and United Daily. La Cathédrale de León a-t-elle des chances de remplir les conditions d’inscription à la Liste du patrimoine mondial de l’UNESCO?

La Liste du patrimoine mondial de l’UNESCO exerce une forte attraction à l’échelle internationale. Cette recherche aborde des démarches accomplies par l’État du Nicaragua en vue de l’inscription de la Cathédrale de León, et tente de déterminer le potentiel de réussite de cette candidature. En premier lieu, nous faisons une réflexion sur la Liste du patrimoine mondial, en regard de la Convention du patrimoine mondial et des États parties. En deuxième lieu, à partir de la décision du Comité concernant le projet du Nicaragua, nous mettons en lumière le manque de coordination entre les parties prenantes, soit entre les instances nationales, locales et diocésaines. Les données récoltées à León nous permettent d’observer que la coordination entre les parties prenantes est essentielle à la réalisation du projet d’inscription, tant pour la consolidation du dossier que pour la protection du bien. Nous soulevons ensuite les enjeux et les solutions envisageables. Afin de favoriser la participation de toutes les parties prenantes au projet, nous appliquons l’approche du « développement de consensus ». Les résultats de l’analyse révèlent néanmoins que la présence de conflits de valeurs empêche la création d’accords. Conséquemment, nous considérons que l’inscription de la Cathédrale de León est peu probable. Le processus d’inscription est une tâche ambitieuse, d’autant plus qu’il fait appel à la contribution d’une propriété privée sous l'égide d’une institution: l’Église Catholique. L’aboutissement du projet est confronté inévitablement à des enjeux de pouvoir, présents en toile de fond. / UNESCO’s World Heritage List is widely recognized and of great importance internationally. This research outlines the steps undertaken by the State of Nicaragua to register the Cathedral of León on the List and attempts to determine the chances of success of its nomination. Firstly, we study the World Heritage List, in regards to the World Heritage Convention and the States Parties. Second, looking at the Committee’s decision concerning the Nicaragua project, we highlight the lack of coordination between the principal stakeholders, namely the national, local and diocesan levels. Data collected in León allow us to observe that coordination between the stakeholders is essential for the registration project, affecting the consolidation of the dossier as well as the protection of property. We then analyze the issues and the possible solutions. To promote the participation of all stakeholders, we apply the approach of "consensus building". However, the results of the analysis show that ”conflicts in values” prevent the creation of agreements. We therefore conclude that the Cathedral of León has a low chance of being registered on the World Heritage List. The registration process is an ambitious task, all the more as the project requires the implication of a private property under the aegis of an institution: the Catholic Church. Its fulfillment is inevitably impacted by issues of power underlying the nomination project.

Les algorithmes de haute résolution en tomographie d'émission par positrons : développement et accélération sur les cartes graphiques

La tomographie d’émission par positrons (TEP) est une modalité d’imagerie moléculaire utilisant des radiotraceurs marqués par des isotopes émetteurs de positrons permettant de quantifier et de sonder des processus biologiques et physiologiques. Cette modalité est surtout utilisée actuellement en oncologie, mais elle est aussi utilisée de plus en plus en cardiologie, en neurologie et en pharmacologie. En fait, c’est une modalité qui est intrinsèquement capable d’offrir avec une meilleure sensibilité des informations fonctionnelles sur le métabolisme cellulaire. Les limites de cette modalité sont surtout la faible résolution spatiale et le manque d’exactitude de la quantification. Par ailleurs, afin de dépasser ces limites qui constituent un obstacle pour élargir le champ des applications cliniques de la TEP, les nouveaux systèmes d’acquisition sont équipés d’un grand nombre de petits détecteurs ayant des meilleures performances de détection. La reconstruction de l’image se fait en utilisant les algorithmes stochastiques itératifs mieux adaptés aux acquisitions à faibles statistiques. De ce fait, le temps de reconstruction est devenu trop long pour une utilisation en milieu clinique. Ainsi, pour réduire ce temps, on les données d’acquisition sont compressées et des versions accélérées d’algorithmes stochastiques itératifs qui sont généralement moins exactes sont utilisées. Les performances améliorées par l’augmentation de nombre des détecteurs sont donc limitées par les contraintes de temps de calcul. Afin de sortir de cette boucle et permettre l’utilisation des algorithmes de reconstruction robustes, de nombreux travaux ont été effectués pour accélérer ces algorithmes sur les dispositifs GPU (Graphics Processing Units) de calcul haute performance. Dans ce travail, nous avons rejoint cet effort de la communauté scientifique pour développer et introduire en clinique l’utilisation des algorithmes de reconstruction puissants qui améliorent la résolution spatiale et l’exactitude de la quantification en TEP. Nous avons d’abord travaillé sur le développement des stratégies pour accélérer sur les dispositifs GPU la reconstruction des images TEP à partir des données d’acquisition en mode liste. En fait, le mode liste offre de nombreux avantages par rapport à la reconstruction à partir des sinogrammes, entre autres : il permet d’implanter facilement et avec précision la correction du mouvement et le temps de vol (TOF : Time-Of Flight) pour améliorer l’exactitude de la quantification. Il permet aussi d’utiliser les fonctions de bases spatio-temporelles pour effectuer la reconstruction 4D afin d’estimer les paramètres cinétiques des métabolismes avec exactitude. Cependant, d’une part, l’utilisation de ce mode est très limitée en clinique, et d’autre part, il est surtout utilisé pour estimer la valeur normalisée de captation SUV qui est une grandeur semi-quantitative limitant le caractère fonctionnel de la TEP. Nos contributions sont les suivantes : - Le développement d’une nouvelle stratégie visant à accélérer sur les dispositifs GPU l’algorithme 3D LM-OSEM (List Mode Ordered-Subset Expectation-Maximization), y compris le calcul de la matrice de sensibilité intégrant les facteurs d’atténuation du patient et les coefficients de normalisation des détecteurs. Le temps de calcul obtenu est non seulement compatible avec une utilisation clinique des algorithmes 3D LM-OSEM, mais il permet également d’envisager des reconstructions rapides pour les applications TEP avancées telles que les études dynamiques en temps réel et des reconstructions d’images paramétriques à partir des données d’acquisitions directement. - Le développement et l’implantation sur GPU de l’approche Multigrilles/Multitrames pour accélérer l’algorithme LMEM (List-Mode Expectation-Maximization). L’objectif est de développer une nouvelle stratégie pour accélérer l’algorithme de référence LMEM qui est un algorithme convergent et puissant, mais qui a l’inconvénient de converger très lentement. Les résultats obtenus permettent d’entrevoir des reconstructions en temps quasi-réel que ce soit pour les examens utilisant un grand nombre de données d’acquisition aussi bien que pour les acquisitions dynamiques synchronisées. Par ailleurs, en clinique, la quantification est souvent faite à partir de données d’acquisition en sinogrammes généralement compressés. Mais des travaux antérieurs ont montré que cette approche pour accélérer la reconstruction diminue l’exactitude de la quantification et dégrade la résolution spatiale. Pour cette raison, nous avons parallélisé et implémenté sur GPU l’algorithme AW-LOR-OSEM (Attenuation-Weighted Line-of-Response-OSEM) ; une version de l’algorithme 3D OSEM qui effectue la reconstruction à partir de sinogrammes sans compression de données en intégrant les corrections de l’atténuation et de la normalisation dans les matrices de sensibilité. Nous avons comparé deux approches d’implantation : dans la première, la matrice système (MS) est calculée en temps réel au cours de la reconstruction, tandis que la seconde implantation utilise une MS pré- calculée avec une meilleure exactitude. Les résultats montrent que la première implantation offre une efficacité de calcul environ deux fois meilleure que celle obtenue dans la deuxième implantation. Les temps de reconstruction rapportés sont compatibles avec une utilisation clinique de ces deux stratégies. / Positron emission tomography (PET) is a molecular imaging modality that uses radiotracers labeled with positron emitting isotopes in order to quantify many biological processes. The clinical applications of this modality are largely in oncology, but it has a potential to be a reference exam for many diseases in cardiology, neurology and pharmacology. In fact, it is intrinsically able to offer the functional information of cellular metabolism with a good sensitivity. The principal limitations of this modality are the limited spatial resolution and the limited accuracy of the quantification. To overcome these limits, the recent PET systems use a huge number of small detectors with better performances. The image reconstruction is also done using accurate algorithms such as the iterative stochastic algorithms. But as a consequence, the time of reconstruction becomes too long for a clinical use. So the acquired data are compressed and the accelerated versions of iterative stochastic algorithms which generally are non convergent are used to perform the reconstruction. Consequently, the obtained performance is compromised. In order to be able to use the complex reconstruction algorithms in clinical applications for the new PET systems, many previous studies were aiming to accelerate these algorithms on GPU devices. Therefore, in this thesis, we joined the effort of researchers for developing and introducing for routine clinical use the accurate reconstruction algorithms that improve the spatial resolution and the accuracy of quantification for PET. Therefore, we first worked to develop the new strategies for accelerating on GPU devices the reconstruction from list mode acquisition. In fact, this mode offers many advantages over the histogram-mode, such as motion correction, the possibility of using time-of-flight (TOF) information to improve the quantification accuracy, the possibility of using temporal basis functions to perform 4D reconstruction and extract kinetic parameters with better accuracy directly from the acquired data. But, one of the main obstacles that limits the use of list-mode reconstruction approach for routine clinical use is the relatively long reconstruction time. To overcome this obstacle we : developed a new strategy to accelerate on GPU devices fully 3D list mode ordered-subset expectation-maximization (LM-OSEM) algorithm, including the calculation of the sensitivity matrix that accounts for the patient-specific attenuation and normalisation corrections. The reported reconstruction are not only compatible with a clinical use of 3D LM-OSEM algorithms, but also lets us envision fast reconstructions for advanced PET applications such as real time dynamic studies and parametric image reconstructions. developed and implemented on GPU a multigrid/multiframe approach of an expectation-maximization algorithm for list-mode acquisitions (MGMF-LMEM). The objective is to develop new strategies to accelerate the reconstruction of gold standard LMEM (list-mode expectation-maximization) algorithm which converges slowly. The GPU-based MGMF-LMEM algorithm processed data at a rate close to one million of events per second per iteration, and permits to perform near real-time reconstructions for large acquisitions or low-count acquisitions such as gated studies. Moreover, for clinical use, the quantification is often done from acquired data organized in sinograms. This data is generally compressed in order to accelerate reconstruction. But previous works have shown that this approach to accelerate the reconstruction decreases the accuracy of quantification and the spatial resolution. The ordered-subset expectation-maximization (OSEM) is the most used reconstruction algorithm from sinograms in clinic. Thus, we parallelized and implemented the attenuation-weighted line-of-response OSEM (AW-LOR-OSEM) algorithm which allows a PET image reconstruction from sinograms without any data compression and incorporates the attenuation and normalization corrections in the sensitivity matrices as weight factors. We compared two strategies of implementation: in the first, the system matrix (SM) is calculated on the fly during the reconstruction, while the second implementation uses a precalculated SM more accurately. The results show that the computational efficiency is about twice better for the implementation using calculated SM on-the-fly than the implementation using pre-calculated SM, but the reported reconstruction times are compatible with a clinical use for both strategies.

This thesis deals with the integration of the Vietnamese community in the Czech Republic which has its specifics and significantly differs from the integration of other foreigners living in the Czech Republic. The characteristics of the Vietnamese community are for example the high percentage of those who live here on the permanent residence permit, then very low unemployment and at last the majority of the community is doing their own business on the trade licence. The other important part is the issue of the acquisition of knowledge of the Czech language. The theoretical part of the thesis firstly defines the number of Vietnamese living in the Czech Republic, historical development of this community and their characteristics. Then the thesis deals with the term of integration; this term is properly defined and the particular dimensions of integration are described as well.
