Short Pulses in Engineered Nonlinear Media

Short optical pulses and engineered nonlinear media is a powerful combination. Mode locked pulses exhibit high peak powers and short pulse duration and the engineered ferro-electric KTiOPO4 facilitates several different nonlinear processes. In this work we investigate the use of structured, second-order materials for generation, characterization and frequency conversion of short optical pulses. By cascading second harmonic generation and difference frequency generation the optical Kerr effect was emulated and two different Nd-based laser cavities were mode locked by the cascaded Kerr lensing effect. In one of the cavities 2.8 ps short pulses were generated and a strong pulse shortening took place through the interplay of the cavity design and the group velocity mismatch in the nonlinear crystal. The other laser had a hybrid mode locking scheme with active electro-optic modulation and passive cascaded Kerr lensing incorporated in a single partially poled KTP crystal. The long pulses from the active modulation were shortened when the passive mode locking started and 6.9 ps short pulses were generated. High-efficiency frequency conversion is not a trivial task in periodically poled materials for short pulses due to the large group velocity mismatch. Optimization of parameters such as the focussing condition and the crystal temperature allowed us to demonstrate 64% conversion efficiency by frequency doubling the fs pulses from a Yb:KYW laser in a single pass configuration. Quasi phase matching also offers new possibilities for nonlinear interactions. We demonstrated that it is possible to simultaneously utilize several phase matched second harmonic interactions, resulting in a dual-polarization second harmonic beam. Short pulse duration of the fundamental wave is a key parameter in the novel method that we demonstrated for characterization of the nonlinearity of periodically poled crystals. The method utilizes the group velocity mismatch between the two polarizations in a type II second harmonic generation configuration. The domain walls of PPKTP exhibit second order nonlinearities that are forbidden in the bulk material. This we used in a single shot frequency resolved optical gating arrangement. The spectral resolution came from Čerenkov phase matching, a non-collinear phase matching scheme that exhibits a substantial angular dispersion. The second harmonic light was imaged upon a CCD camera and with the spectral distribution on one axis and the temporal autocorrelation on the other. From this image we retrieved the full temporal profile of the fundamental pulse, as well as the phase. The spectral dispersion provided by the Čerenkov phase matching was large enough to characterize optical pulses as long as ~200 fs in a compact setup. The Čerenkov frequency resolved optical gating method samples a thin stripe of the beam, i.e. the area close to the domain wall. This provides the means for high spatial resolution measurements of the spectral-temporal characteristics of ultrafast optical fields. / QC 20100831

" Locked Nucleic Acid " nanovectorisés pour la répression de l'activité de microARN impliqués dans la radiorésistance des cellules de glioblastome

Le glioblastome est la tumeur maligne primaire du cerveau la plus courante et la plus agressive chez l'homme. Son traitement conventionnel est palliatif et l'apparition de récidives est systématique. Dans le but de développer des thérapies innovantes basées sur le ciblage de nouvelles entités tumorales à l'aide de nanovecteur de médicaments, deux cibles ont été investiguées : le marqueur de radiorésistance AC133/1 et les onco-microARN. En utilisant des cellules de glioblastomes issues de patients, nous avons démontré que leur expansion in vitro à une pO2 non-physiologique (21%) altère leur agressivité tumorale in vivo et l'expression originelle d'AC133/1, au contraire d'une pO2 physiologique (3%) soulignant qu'AC133/1 est un marqueur précoce de non exposition à des pO2 élevées. Nous identifions par ailleurs un rôle pour AC133/1 dans l'endocytose du récepteur de la transferrine et son partenariat avec le métabolisme du fer. Enfin nous avons développé et caractérisé des immunonanoparticules capables de véhiculer des chimiothérapies ou des radiopharmaceutiques vers cet épitope fonctionnel. Dans un second axe de recherche, en parallèle d'établir le miRnome humain en réponse à l'action d'une radiothérapie, des nanocapsules lipidiques biomimétiques présentant à leur surface des peptides de papillomavirus et capables de se complexer avec des acides nucléiques antagonistes de microARN ont été développées et évaluées, démontrant leur intérêt en synergie d'une radiothérapie. Collectivement et en amont d'expérimentations in vivo en cours, ces résultats soulignent la pertinence d'appliquer de nouvelles nanomédecines ciblées pour le contournement de la radiorésistance dans le glioblastome.

Ce travail de thèse porte sur les systèmes de communication radio-sur-fibre (RoF) aux fréquences millimétriques dans la gamme de fréquences 57-66 GHz et leur génération par voie optique. La technique utilisée repose sur l’emploi de diodes laser à verrouillage de modes à boites quantiques. Au cours de cette thèse, plusieurs études ont été effectuées : la première porte sur la capacité d’intégrer ces types des lasers dans des systèmes de communication RoF avec leurs performances sous modulation directe ou externe. La deuxième étude a été consacrée aux effets de la propagation de ces signaux dans des systèmes basés sur les lasers à blocage de modes. Une technique originale a été mise en place afin de réduire la sensibilité à la dispersion chromatique dans la fibre optique. Une étude de la réduction du bruit de phase des lasers à verrouillage de modes basée sur l’observation de l’effet de la contre réaction et l’injection optique externe a été également présentée. / This thesis focuses on the radio-over-fiber (RoF) communication systems at millimeter frequencies in the frequency range 57-66 GHz and optical generation of a signal at millimeter-wave frequency band. The technique used is based on mode-locked laser diodes. The diodes employed in this work are in quantum dots (or quantum dashes) technology. In this thesis, several studies were conducted: the first relates the ability of integration these types of lasers in the RoF communication systems under direct or external modulation. The second study was devoted to propagation effects. An original technique was implemented to reduce the sensitivity to chromatic dispersion in an optical fiber. A study of the reduction of phase noise mode-locked lasers based on the observation of the reaction effect and against the external optical injection was presented.

Etude et réalisation de sources laser impulsionnelles en optique intégrée sur verre : application à la génération de supercontinuum / Study and realization of integrated optics pulsed lasers for supercontinuum generation

Le développement des sources supercontinuum a engendré des avancées majeures dans de nombreux domaines tels que la spectroscopie, la métrologie ou encore la biologie avec la tomographie optique cohérente. Récemment, l'arrivée des fibres à cristaux photoniques (FCPs) a permis la fabrication de telles sources avec des lasers impulsionnels de pompe moins puissants. Ainsi, de par leur compatibilité avec les fibres optiques, les lasers impulsionnels réalisés en optique intégrée sur verre semblent une alternative intéressante pour la réalisation d'une source supercontinuum intégrée. Notre travail a donc eu pour objectif l'étude et la réalisation d'un laser impulsionnel déclenché par modulation passive des pertes assez puissant et compatible avec les fibres à cristaux photoniques dans le but de générer un supercontinuum. Nous avons alors conçu et fabriqué ce laser constitué d'un guide d'onde amplificateur réalisé par échange d'ions dans un verre dopé au néodyme et d'une cavité Fabry-Perot fermée par un miroir diélectrique en entrée et sans miroir de sortie ce qui permet un couplage direct avec une fibre optique. L'hybridation de l'absorbant saturable sous la forme d'une couche mince d'acétate de cellulose dopée au BDN sur le guide d'onde a permis la génération d'impulsions ayant une puissance crête de (2,8 ± 0,6) kW. La source supercontinuum réalisé avec une FCP pompée par le laser préalablement étudié présente un spectre allant de 440 nm à 1600 nm. Les perspectives de cette étude portent sur le changement de matériau absorbant saturable pour permettre l'émission d'impulsions plus puissantes, ainsi que sur l'intégration monolithique de la FCP pour réaliser un dispositif encore plus compact. Elles portent aussi sur la réalisation de lasers intégrés à modes bloqués utilisant cette technologie. Nous avons démontré sa faisabilité ainsi que son régime de fonctionnement en soliton dissipatif. Des caractérisations préliminaires ont également été effectuées. / Supercontinuum sources made possible major break through in spectroscopy metrology and biology like the development of optical coherent tomography. Photonics crystal fiber have been useful for supercontinuum generation allowing the use of relatively low peak power pulsed laser sources. Regarding this kind of devices, pulsed lasers made by integrated optics on glass seemed to be a good candidate. The aims of this work were hence to develop and realize Q-switched lasers for photonic crystal fiber pumping. We demonstrated the possibility to realize a supercontinuum source pumped by an integrated Q-switched laser made by ion exchange on Neodymium doped glass. The Fabry-Perot cavity used is closed by a dielectric mirror on one hand and by the waveguide facet on the other side allowing direct coupling with optical fiber. The saturable absorber has been hybridized on the amplifying waveguide to interact with the evanescent wave. (2.8 ± 0.6) kW peak-power pulses have been generated, which coupled in a photonic crystal fiber, generated a 440 nm to 1600 nm large supercontinuum. As a perspective, a mode-locked laser realized with the same technology has been demonstrated. We show that this laser operates in dissipative soliton regime. This laser is then preliminary characterized.

Contribution à l'amélioration de la quantification des acides nucléiques par qPCR et RT-qPCR / Contribution to Improving of the Quantification of Nucleic Acids using qPCR and RT-qPCR

La qPCR est actuellement la technique de référence en matière de quantification d'ADN. Elle peut être définie comme une amplification exponentielle, cyclique et ciblée de la séquence d'ADN cible. Le caractère exponentiel de la qPCR est à la fois à l'origine de la sensibilité de la méthode mais aussi d'une potentielle variabilité inter-échantillons. Cette variabilité est compensée par le caractère cyclique de la méthode qui entraine une synchronisation de la réaction pour tous les échantillons à chaque cycle. L'amplification ciblée de la séquence choisie traduit quand à elle la spécificité de la méthode. Néanmoins, cette dernière propriété de la PCR reste la moins vérifiée. La spécificité de la qPCR est indiscutable lorsque la cible à détecter se trouve en quantité suffisante (>100 copies environ). En revanche, pour des quantités plus faibles ou en présence d'inhibiteurs, la spécificité diminue ou disparaît (limites de détection et de quantification). Cette perte de spécificité retentit de façon significative sur la précision et la reproductibilité du dosage à réaliser. Cependant, si l'hybridation non spécifique est inéluctable dans ces conditions, l'amplification non spécifique consécutive peut être limitée, voire supprimée au moyen d'amorces de PCR soit plus spécifiques, soit moins susceptibles de générer des différences inter-échantillons. La RT-qPCR, grâce à une étape initiale de transcription inverse (conversion d'un ARN en un ADN complémentaire) permet la quantification des ARN. Cependant, la transcription inverse reste moins reproductible que la PCR, générant des différences inter-échantillons délétères en diagnostic comme en recherche. Au cours de ces travaux, je propose des méthodes originales améliorant de façon significative différentes étapes de ces techniques. Premièrement, je propose une amélioration de la standardisation de l'étape de transcription inverse capable de diminuer de façon significative la variabilité inter-échantillons ; l'utilisation d'un volume constant d'extrait d'ARN pour chaque échantillon améliore considérablement la précision de la quantification d'ARN messagers. Deuxièmement, je propose une modification des amorces par incorporation de résidus d'acides nucléiques bloqués (LNA) ; cette modification permet d'augmenter la spécificité des amorces aux limites de la détection. Enfin, je propose une méthode simple et économique permettant la mesure directe de la température de fusion (Tm) dans les conditions réelles de la PCR. Ce paramètre, considéré comme capital pour la réalisation des dosages par qPCR, est généralement obtenu par des méthodes prédictives peu précises. De plus, cette méthode doit permettre de déterminer précisément les paramètres thermodynamiques des amorces (∆G, ∆H et ∆S) et ainsi avoir accès d'une part au pourcentage réel d'amorces hybridées en fonction de la température et d'autre part d'identifier la susceptibilité des amorces aux inhibiteurs. / QPCR is currently the gold standard for DNA quantification of DNA. It can be defined as an exponential, cyclic and targeted amplification of a known DNA sequence. The exponential character of qPCR is both the cause of the sensitivity of the method but also a potential variability between samples. This variability is offset by the cyclical nature of the method that causes a synchronization of the reaction for all samples in each cycle. The targeted amplification of the selected sequence translates the specificity of the method. Nevertheless, this last property of PCR remains the least tested. The specificity of qPCR is unquestionable when the target to detect is sufficient (> 100 copies). However, for smaller quantities or in the presence of inhibitors, the specificity decreases or disappears (limits of detection and quantification). This loss of specificity will have a significant effect upon the accuracy and reproducibility of the assay to be performed. However, if non-specific hybridizations are unavoidable in these conditions, consecutive nonspecific amplification may be limited or eliminated using more specific PCR primers or less likely to produce inter-sample differences. RT-qPCR allows RNA quantification because of an initial step of reverse transcription (conversion of an RNA into a complementary DNA). However, reverse transcription is less reproducible than PCR, generating harmful inter-sample differences both in diagnosis and in the research field. In this work, I propose innovative methods improving significantly different steps of these techniques. First, I propose an improved standardization of the reverse transcription step that can significantly reduce the variability between samples; using a constant volume of RNA extract for each sample substantially improves mRNA quantification accuracy. Second, I propose a primers modification by incorporating Locked Nucleic Acid residues (LNA); this modification increases the specificity of the primers to the limits of detection. Finally, I propose a simple and economical method for direct measurement of the melting temperature (Tm) in real PCR conditions. This essential parameter in the achievement of qPCR assays is generally obtained by predictive methods with low accuracy. Furthermore, this method should determine the thermodynamic parameters of the oligonucleotide sequence (∆G, ∆H and ∆S), on the one hand allowing access to the actual percentage of annealed primers according to the temperature and on the other hand, to identify the primers susceptibility in the presence of inhibitors.

So, an onboard GPS receiver could calculate navigation solutions if the Doppler effect is taken into consideration during the satellite signals acquisition and tracking. Thus, for the receiver subject to such dynamic cope these shifts in the frequency signal, resulting from this effect, it is imperative to adjust its acquisition bandwidth and increase its tracking loop to a higher order. This paper presents the changes in the GPS Orion s software, an open architecture receiver produced by GEC Plessey Semiconductors, nowadays Zarlink, in order to make it able to generate navigation fix for vehicle under high dynamics, especially Low Earth Orbit satellites. GPS Architect development system, sold by the same company, supported the modifications. Furthermore, it presents GPS Monitor Aerospace s characteristics, a computational tool developed for monitoring navigation fix calculated by the GPS receiver, through graphics. Although it was not possible to simulate the software modifications implemented in the receiver in high dynamics, it was observed that the receiver worked in stationary tests, verified also in the new interface. This work also presents the results of GPS Receiver for Aerospace Applications experiment, achieved with the receiver s participation in a suborbital mission, Operation Maracati 2, in December 2010, using a digital second order carrier tracking loop. Despite an incident moments before the launch have hindered the effective navigation of the receiver, it was observed that the experiment worked properly, acquiring new satellites and tracking them during the VSB-30 rocket flight. / Ve?culos espaciais se movem com velocidades elevadas e sofrem mudan?as bruscas de acelera??o. Controle de caos em PLL de terceira ordem. / Control of chaos in third-order PLL.

Inicialmente, apresentam-se características de dispositivos eletrônicos conhecidos como PLLs (phase-locked loops). PLLs são amplamente empregados para se extrair sinais de tempo em canais de comunicação e em aplicações nas quais se deseja controle automático de freqüência. O objeto principal é estudar PLLs analógicos descritos por uma equação diferencial ordinária de terceira ordem. Assim, deduzem-se condições de estabilidade assintótica e identifica-se um regime de caos conservativo, que ocorre sob certas combinações de valores de parâmetros. Três métodos de controle não-linear/caótico são então apresentados e aplicados. Os métodos são os seguintes: o Método de Pyragas via realimentação de variável de estado; o Método de Pyragas com atraso temporal na realimentação; e o Método de Sinha, o qual efetua o controle perturbando um parâmetro do sistema. Simulações numéricas são levadas a cabo a fim de ilustrar o comportamento dinâmico do sistema quando sujeito à ação desses métodos. Este trabalho termina com um estudo de uma rede formada por uma cadeia de PLLs. Condições para soluções síncronas, periódicas e caóticas (dissipativas e conservativas) são deduzidas para tal rede. / Firstly, features of electronic devices known as PLLs (Phase-Locked Loops) are presented. PLLs are widely employed to extract time signals in communication channels and in applications where automatic control of frequency is desired. The main goal is to study analog PLLs described by a third-order nonlinear ordinary differential equation. Thus, conditions for asymptotic stability are derived and a regime of conservative chaos occurring under certain combinations of parameter values is identified. Then, three methods of control of nonlinear/ chaotic dynamics are presented and applied. The methods are the following: the Pyragas method via feedback of state variable; the Pyragas method with time delay in the feedback; and the Sinhas method, which performs the control by disturbing a parameter of the system. Numerical simulations are accomplished in order to illustrate the dynamical behavior of the system when subjected to the action of these methods. This work ends with a study of a single-chain PLL network. Conditions for synchronous, periodic and chaotic (dissipative and conservative) solutions are derived for such a network.

Neste trabalho, são estudados os problemas de sincronismo de fase nas redes mestre-escravo de via única (OWMS), nas topologias Estrela Simples, Cadeia Simples e mista, através da Teoria Qualitativa de Equações Diferenciais, com ênfase no Teorema da Variedade Central. Através da Teoria das Bifurcações, analisa-se o comportamento dinâmico das malhas de sincronismo de fase (PLL) de segunda ordem que compõem cada rede, frente às variações nos seus parâmetros constitutivos. São utilizadas duas funções de excitação muito comuns na prática: o degrau e a rampa de fase, aplicadas pelo nó mestre. Em cada caso, discute-se a existência e a estabilidade do estado síncrono. A existência de pontos de equilíbrio não-hiperbólicos, não permite uma aproximação linear, e nesses casos é aplicado o Teorema da Variedade Central. Através dessa rigorosa técnica de simplificação de sistemas dinâmicos é possível fazer uma aproximação homeomórfica em torno desses pontos, preservando a orientação no espaço de fases. Desse modo, é possível determinar, localmente, suas estabilidades. / This work presents stability analysis of the syncronous state for three types of one-way master-slave time distribution network topologies: single star, single chain and both of them, mixed. Using bifurcation theory, the dynamical behavior of second-order phase-locked loops employed to extract the syncronous state in each node is analyzed in function of the constitutive parameters. Two usual inputs, the step and the ramp phase pertubations, are supposed to appear in the master node and, in each case, the existence and stability of the syncronous state are studied. For parameter combinations resulting in non hyperbolic synchronous states, the linear approximation does not provide any information, even about the local behaviour of the system. In this case, the center manifold theorem permits the construction of an equivalent vector field representing the asymptotic behaviour of the original system in the neighborhood of these points. Thus, the local stability can be determined.

CoordenaÃÃo de AperfeÃoamento de Pessoal de NÃvel Superior / In applications related to power converters, such as inverters, rectifiers and the use of active filters, the synchronization method represent a very important element in the performance of the control strategy of this equipment. The estimated values of the synchronism angle, frequency and amplitude determined by the synchronization algorithms present, facing strongly distorted signals with the presence of sub and interharmonics, high errors. This study presents two algorithms: one applied on single-phase electrical systems and one applied on three-phase electrical systems, with high immunity to interharmonics and subharmonics. First are presented the main synchronization systems that are used in the electrical power systems. In addition, will be presented the main causes and consequences of the presence of subharmonics and interharmnics in the system, as well as the mathematical modeling of the two algorithms with high rejection to these disturbances. Will be presented the simulation and the experimental results of the proposed algorithms and the comparison between these synchronization methods with particular methods present in the literature. As a result of the study, it can be seen that the proposed structures present a higher response time, but the error of the estimated signal with respect the fundamental component of the input signal is lower when compared to structures such as EPLL and structures based on SOGI. It was observed that the proposed synchronization methods are enabled to estimate the synchronism angle, the frequency and the fundamental component of the input signal adequately and can be used in control strategies of power converters. / Em aplicaÃÃes relacionadas à EletrÃnica de PotÃncia, como inversores, retificadores e a utilizaÃÃo de filtros ativos, o mÃtodo de sincronizaÃÃo representa um elemento chave no desempenho da estratÃgia de controle destes equipamentos. Os valores do Ãngulo de sincronismo, frequÃncia e amplitude estimados com determinados algoritmos de sincronizaÃÃo apresentam, diante de sinais fortemente distorcidos com a presenÃa de sub e inter-harmÃnicos, erros elevados. Neste trabalho sÃo apresentados dois algoritmos: um aplicado a sistemas elÃtricos monofÃsicos e outro aplicado a sistemas elÃtricos trifÃsicos, com elevada imunidade a inter-harmÃnicos e sub-harmÃnicos. Primeiramente sÃo apresentados os principais sistemas de sincronizaÃÃo utilizados em sistemas elÃtricos de potÃncia. AlÃm disso, sÃo apresentadas as principais causas e consequÃncias da presenÃa de sub-harmÃnicos e inter-harmÃnicos no sistema, bem como a modelagem matemÃtica dos dois algoritmos com elevada rejeiÃÃo a estes distÃrbios. SÃo apresentados os resultados de simulaÃÃo e experimentais dos algoritmos propostos e a comparaÃÃo entre estes mÃtodos de sincronizaÃÃo com determinados mÃtodos presentes na literatura. Como resultado do estudo, pode-se observar que as estruturas de sincronizaÃÃo propostas apresentam um tempo de resposta mais elevado, porÃm o erro do sinal estimado em relaÃÃo a componente fundamental do sinal de entrada à inferior quando comparado a estruturas como o EPLL e estruturas baseadas no SOGI. Com isso, tem-se que as mesmas estÃo habilitadas para estimar o Ãngulo de sincronismo, a frequÃncia e a componente fundamental do sinal de entrada adequadamente e podem serem utilizadas eficientemente em estratÃgias de controle de conversores de potÃncia.

Método para determinação dos pesos sinápticos em uma rede de PLLs reconhecedora de imagens

These networks are modeled with differential equation systems and can be studied by using Dynamical System Theory, which is used in this work in order to investigate the dynamical behavior related to a synaptic configuration of a neural network. As a result of such an investigation, two methods (Brute Force and Algebric) that help to build neural networks formed by PLLs are presented. These methods aim to relate the synaptic configuration of the network to the corresponding basin of attraction of fixed points, which represent the stored patterns on the network. Also general properties of synaptic configuration are presented in order to generate other useful configurations. Then a model of an image recognition machine able to store in its memory a monochromatic image and able to determine if other image is similar to the memorized one is proposed. / Reconhecimento de padrões pode ser feito usando redes neurais construídas com osciladores, como malhas de sincronismo de fase (PLLs). Essas redes são modeladas por sistemas de equações diferenciais e podem ser estudas pela Teoria de Sistemas Dinâmicos, que é usada neste trabalho para investigar o comportamento dinâmico associado a uma configuração sináptica de uma rede neural. Como resultado dessa investigação, são apresentados dois métodos (Força Bruta e Algébrico) que auxiliam na construção de redes neurais formadas por PLLs. Esses métodos têm como objetivo relacionar a configuração sináptica da rede às respectivas bacias de atração de pontos atratores, os quais representam os padrões memorizados na rede. Também são apresentadas propriedades gerais da configuração sináptica que podem ser usadas para compor outras configurações de interesse. Por fim, é proposto um modelo de máquina reconhecedora de imagem capaz de armazenar em sua memória uma figura monocromática e determinar se uma imagem qualquer apresentada a ela é semelhante à memorizada.
