INTOXICAÇÃO EM BOVINOS POR Senna occidentalis / POISONING IN CATTLE BY Senna occidentalis

Sixteen outbreaks of Senna occidentalis (coffee senna) occurring in cattle from the state of Rio Grande do Sul, Brazil, were reviewed. The great majority (75%) of the outbreaks occurred in adult cattle at pasture during the autumn and winter months with 50% occurring in May, evidencing a striking seasonality. Mortality rates varied from 4.2% to 55.2% and cattle died 2 days up to 2 weeks after showing clinical signs that included dry feces (occasionally diarrhea), muscle weakness, reluctance to move, tachypnea, instability of the hind limbs with dragging of the toes, tremors in muscles of the thighs, neck, and head, ear dropping, sternal recumbency, lateral recumbency and death. Myoglobinuria characterized by a dark red or black discolored urine was a consistent finding in cattle affected at pasture but not in those poisoned by ration contaminated with coffee senna beans. Creatine phosphokinase serum activity was markedly elevated. Main gross changes observed in 23 necropsies involved skeletal muscles of the hind limbs. These changes consisted of varying degrees of paleness of muscle groups. Subepicardial and subendocardial hemorrhages were present in the hearts of all affected cattle. Histologically a segmental degenerative myopathy of striated muscles was present in every case and had a multifocal polyphasic or monophasic character. Myocardial (3/23), hepatic (3/13), renal (3/10), and splenic (1/6) microscopic lesions were observed occasionally. Myocardial lesions were mild and consisted of vacuolation of cardiomyocytes or focal fibrosis. Hepatic changes consisted of diffuse hepatocellular vacuolation, cytosegrosomes within hepatocytes, and individual hepatocellular necrosis. Kidneys have vacuolar degeneration of tubular epithelium associated with acidophilic casts (proteinosis) within tubular lumina. In the spleen there was marked necrosis of lymphocytes of the white pulp. No histological changes were found in the brains of 13 affected cattle. The data of this study suggest that coffee senna poisoning is an important cause of death in cattle in southern Brazil. The epidemiological, clinical and pathological features described in the study shoud be useful for diagnosing the condition in the field. / Dezesseis surtos de intoxicação por Senna occidentalis (fedegoso) em bovinos do Rio Grande do sul foram revisados. A grande maioria dos surtos (75%) ocorreu em bovinos adultos em pastoreio durante o outono e inverno com 50% dos surtos ocorrendo em maio, evidenciando uma notável sazonalidade. Os coeficientes de mortalidade variaram de 4,2% a 55,2% e os bovinos morriam dois dias a duas semanas após mostrarem sinais clínicos que incluíam fezes ressecadas (ocasionalmente diarreia), fraqueza muscular, relutância em mover-se, taquipneia, instabilidade dos membros pélvicos com arrastamento das pinças, tremores nos músculos das coxas, pescoço e cabeça; orelhas caídas, decúbito esternal, decúbito lateral e morte. A atividade sérica da creatina fosfocinase estava acentuadamente elevada. As principais alterações macroscópicas observadas em 23 necropsias envolviam os músculos esqueléticos dos membros pélvicos. Essas alterações consistiam de graus variáveis de palidez em grupos musculares. Hemorragias subepicárdicas e subendocárdicas ocorreram nos corações de todos os bovinos afetados. Histologicamente, miopatia degenerativa dos músculos estriados esteve presente em todos os casos e tinha um caráter multifocal monofásico ou polifásico. Lesões microscópicas no miocárdio (3/23), fígado (3/13), rim (3/10) e baço (1/6) foram ocasionalmente observadas. As lesões miocárdicas eram discretas e consistiam de vacuolização dos cardiomiócitos ou fibrose focal. As lesões hepatocelulares consistiam de vacuolização difusa, formação de citossegrossomos e necrose individual. Nos rins havia degeneração vacuolar do epitélio tubular associada a cilindros eosinofílicos (proteinose) na luz tubular. No baço havia marcada necrose de linfócitos da polpa branca. Nenhuma alteração foi encontrada ao exame histológico do encéfalo de 13 bovinos afetados. Os dados deste estudo sugerem que a intoxicação por fedegoso é uma importante causa de morte em bovinos do sul do Brasil. Os aspectos epidemiológicos, clínicos e patológicos descritos neste estudo deverão ser úteis no diagnóstico a campo desta intoxicação.

In the first theoretical part, principle of electron microscopy and chloroplast morphology is described. In experimental part, electron microscopy images of chloroplast under different conditions of sample preparation are shown and discussed.

Petroleum ether, dichloromethane, and 80% ethanol extracts of 15 plant species collected in Nigeria were screened for in vitro antibacterial, anti-inflammatory and antimalarial activities. Antibacterial activity was tested using the agar diffusion method, while the minimum inhibitory concentrations (MIC) of the active extracts were determined using the microtitre serial dilution method. Most antibacterial activity detected was against Gram-positive bacteria with Staphylococcus aureus being the most susceptible. The highest activity was found in petroleum ether and dichloromethane leaf extracts of Mallotus oppositifolius; petroleum ether, dichloromethane and ethanolic root extracts of Newbouldia laevis; and ethanolic root extracts of Morinda lucida and Canthium subcordatum. Against the Gram-negative bacterium Escherichia coli, the highest activity was found in dichloromethane leaf extracts of Newbouldia laevis, ethanolic root extracts of Phyllanthus amarus, Mallotus oppositifolius, and Canthium subcordatum. A total of 60 plant extracts were screened for antiplasmodial activity. A chloroquine sensitive strain of Plasmodium falciparum (D10) was used. In the assay, the parasite lactate dehydrogenase (pLDH) activity was used to measure parasite viability. About 11 extracts showed promising activity with an IC₅₀ ranging from 2.5 to 13.4 µg/ml. The petroleum ether leaf extract of Hyptis suaveolens had the highest activity (IC₅₀ = 2.5 µg/ml). The cyclooxygenase (COX-1 and COX-2) assays were used to test for anti-inflammatory activity. All the plant species, with the exception of Hedranthera barteri and Picralima nitida showed anti-inflammatory activity. Apart for a few ethanolic extracts, all the activities were recorded with petroleum ether and dichloromethane extracts. Employing bioassay-guided activity fractionation, an antibacterial anthraquinone identified as emodin was isolated from ethanolic root extract of Senna occidentalis. Although this compound had been isolated from other sources, this was the first report of isolation from Senna occidentalis. Using a similar approach a novel antimalarial diterpenoid was isolated from the petroleum ether leaves extract of Hyptis suaveolens. It had IC₅₀ of 0.1 µg/ml. [EN] Tomato spotted wilt virus is one of the most widespread and economically important viruses worldwide. It infects a large number of plant species, being the tomato and pepper the most affected. TSWV is transmitted from one plant to another by various species of thrips in a circulative and propagative manner, being Flankliniella occidentalis the main vector. The best strategy for disease control in tomato and pepper has been breeding resistant cultivars, but only tomato with the gene Sw-5 and pepper with gene Tsw have been effective against a wide spectrum of TSWV isolates, but resistance-breaking isolates often arise. To obtain a more effective and durable control is necessary: A) the genetic and biological characterization of conventional and resistance-breaking isolates of TSWV; B) the understanding the evolutionary and epidemiological factors involved in the emergence and dispersion of resistance-breaking isolates; C) the development of tools for viral detection and quantification; and D) evaluation of new sources of resistance (total or partial, estimated as the difficulty to viral infection or/and accumulation) or tolerance (total or partial, estimated as the difficulty to symptoms development and damage without affecting viral infection). In this work, TSWV isolates from Spanish tomato and pepper crops have been biologically and molecularly characterized. The complete genome of three TSWV isolates with different biotypes were sequenced: N (unable to infect Sw-5 resistant tomato and Tsw resistant pepper), T (tomato Sw-5 resistance-breaking), and P (pepper Tsw resistance-breaking). No correlation between genetic variation and the ability of overcoming resistance was found. These nucleotide sequences and others retrieved from the GenBank database were used to develop RT-qPCR, with high sensitivity and dynamic range. Primers and one TaqMan MGB were designed from conserved sequence stretches with the aim of detecting and quantifying any TSWV isolate. It was not possible to develop a molecular method to differentiate between conventional and resistance-breaking isolates since there is no correlation between genotype and biotype. Instead, two TaqMan MGB probes were designed to quantify the two main genotypes (according to the M segment) in mixed infections. RT-qPCR was used to evaluate TSWV accumulation in non-resistant tomato, non-resistant pepper and Datura stramonium (an important reservoir), as well as in the main vector F. occidentalis, which was considered to evaluate transmission efficiency. The results showed that resistance breakdown was not associated to a fitness cost (tradeoff) in the infectivity of susceptible hosts or transmissibility by thrips and that the resistance-breaking isolates have the same potential for dispersion in field as the conventional isolates. Finally, the information obtained from the previous studies and the RT-qPCR technique were used to evaluate the resistance and tolerance to TSWV of a new accession of Capsicum baccatum. In addition to considering the genetic and biological variation of TSWV, a new approach was used based on analysis of longitudinal data (those measured in the same individuals over time). The resistance level was estimated with two variables: A) absolute fitness, calculated from the variation in time of the viral titer, quantified by RT-qPCR; and B) infectition survival, the median time in which the virus was detected in a plant by ELISA. The tolerance level was estimated as symptom survival, the median time in which a plant developed severe symptoms. This analysis showed that this new accession is partially resistant and totally tolerant to TSWV conventional and resistance-breaking isolates and therefore is a good candidate for a pepper breeding program / [ES] Tomato spotted wilt virus (TSWV) es uno de los virus más extendidos y de mayor importancia económica del mundo. Sin embargo, con frecuencia aparecen aislados del virus capaces de superar estas resistencias. Para obtener un control de la enfermedad más eficaz y duradero es necesario: A) la caracterización genética y biológica de los aislados del TSWV que superan y no superan las resistencias; B) el estudio de los factores evolutivos y epidemiológicos implicados en la aparición y establecimiento de los aislados que superan las resistencias; C) el desarrollo de herramientas que permitan la detección y cuantificación de estos aislados virales; y D) la evaluación de nuevas fuentes de resistencia o tolerancia. En este trabajo se han caracterizado biológicamente y molecularmente diferentes aislados del TSWV procedentes de cultivos de tomate y pimiento de España. Se determinó la secuencia nucleotídica del genoma completo de tres aislados españoles correspondientes a tres biotipos: N (incapaz de infectar variedades resistentes de tomate o pimiento); T (supera la resistencia Sw-5 de tomate); y P (supera la resistencia Tsw de pimiento). No se encontró correlación entre la variación genética y la capacidad de superar la resistencia. Estas secuencias nucleotídicas y otras obtenidas de la base de datos Genbank se utilizaron para desarrollar una técnica basada en la RT-PCR cuantitativa (RT-qPCR) que permite detectar el virus con un alto grado de sensibilidad y cuantificar en un amplio rango dinámico. Se diseñaron los iniciadores y una sonda TaqMan MGB a partir de segmentos de secuencia conservados para que fueran válidos para todos los aislados del TSWV. También se desarrollaron dos sondas Taqman que permitían cuantificar diferencialmente variantes genéticas del virus en infecciones mixtas. La RT-qPCR se utilizó para evaluar la acumulación de varios aislados del TSWV en tomate sin Sw-5, pimiento sin Tsw y Datura stramonium, así como, en su principal vector F. occidentalis que se usó para evaluar la eficiencia de la transmisión. Se observó que la superación de la resistencia no suponía un coste en la eficacia biológica (fitness) tanto en la multiplicación del virus en estas plantas como en la transmisión por trips. Por tanto, los aislados que superan las resistencias tienen la misma capacidad de dispersión en campo que los aislados convencionales. Por último, se utilizó la RT-qPCR y la información obtenida para evaluar la capacidad de resistencia y tolerancia al TSWV de una accesión de Capsicum baccatum. El nivel de resistencia se estimó a partir de dos variables: A) la eficacia absoluta, calculada a partir de la variación en el tiempo del título viral, cuantificado por RT-qPCR; y B) la infectividad, como la mediana del tiempo que tarda el virus en ser detectado en una planta por ELISA. El nivel de tolerancia se estimó como la mediana del tiempo que tarda una planta en mostrar síntomas graves. Esta nueva accesión es parcialmente resistente y totalmente tolerante tanto para aislados del TSWV convencionales como para aquellos capaces de infectar e inducir síntomas severos en variedades de pimiento con el gen Tsw. Es va determinar la seqüència nucleotídica del genoma complet de tres aïllats espanyols corresponents a tres biotips: N (incapaç d'infectar varietats resistents de tomaca o pebrot); T (supera la resistència Sw-5 de tomaca); i P (supera la resistència Tsw de pebrot). Es va observar que la superació de la resistència no suposava un cost en l'eficàcia biològica (fitness) tant en la multiplicació del virus en aquestes plantes com en la transmissió per trips. Per tant, els aïllats que superen les resistències tenen la mateixa capacitat de dispersió en camp que els aïllats convencionals. Finalment, es va utilitzar la RT-qPCR i la informació obtinguda per a avaluar la capacitat de resistència i tolerància d'una accessió de Capsicum baccatum al TSWV. El nivell de resistència es va estimar a partir de dues variables: A) l'eficàcia absoluta, calculada a partir de la variació en el temps del títol viral, quantificat per RT-qPCR; i B) la infectivitat, com la mitjana del temps que es tarda a detectar el virus en una planta per mitjèr d'ELISA. The Effects of Mechanical Site Preparation Treatment and Species Selection on Survival and Carbon Pools in 12-Year-Old American Sycamore (<i>Platanus occidentalis</i>) and Willow Oak (<i>Quercus phellos</i>) Riparian Plantations in the Southeastern U.S. Piedmont

Riparian wetlands may provide numerous ecosystem services, including water quality protection, food and fiber supply, wildlife habitat, and carbon sequestration. In recent years, riparian forests have received increased attention and funding for riparian forest restoration projects. Unfortunately, failures of riparian restoration efforts are likely due to mortality of planted trees. Tree mortality is commonly attributable to several factors, including selection of species that are not well suited to the wetland sites, inadequate planting densities, soil compaction associated with former agricultural activities, lack of microtopographic relief that allow small seedling to survive on wet sites, competition by herbaceous plants, and browse. Selection of well-suited species, dense planting and use of mechanical soil site preparations are all potential remedies to partially address success of wetland restoration plantings. Riparian restoration projects have historically been undertaken with goals of improving water quality and/or wildlife habitat, but in recent years there has been increased valuation of carbon sequestration. Carbon valuation appears to be increasing, but more research is needed to determine rates and pools of carbon accumulation in riparian areas. Our research quantifies forest establishment effects on multiple carbon pools in a densely planted, 12-year-old old-field riparian restoration. Our research evaluated the effects of four soil mechanical site preparations (bed, disk, pit, and mound and rip) and species selection (American sycamore (Platanus occidentalis) and willow oak, (Quercus phellos) on forest establishment and carbon storage across multiple pools, namely in planted trees, herbaceous vegetation, fine roots, organic soil horizons, and the mineral soil. At 12 years, we found that species selection was more important to carbon storage than site preparation. American sycamore was well suited to the site and had better survival than willow oak (64% vs 42% survival). American sycamore also stored more carbon across all site preparations than willow oak. Measured carbon storage averaged 74.8 Mg ha-1 for American sycamore treatments and 63.1 Mg ha-1 for willow oak treatments. The plots were densely planted (1.2 m (4ft) by 1.8 m (6ft) spacing), and forests were established even in higher mortality willow oak plots. These results indicate that high planting density is potentially a viable practice for establishing riparian forest cover, especially if desired species are marginally site suited or other survival inhibiting factors exist. / Master of Science / Riparian forests located within the floodplain of streams are known to protect stream water quality, provide wildlife habitat, and store carbon. Due to these benefits, trees may be replanted on riparian areas that were formerly used for agriculture. Unfortunately, trees planted on these restoration sites have often died. The tree mortality may be due to planting incorrect tree species that may struggle on wet floodplains, soil compaction from agricultural practices, not enough trees being planted, and restored floodplains lacking small, elevated areas common across natural wetlands known to favor tree rooting and survival. Reasonably, selecting species that are well suited to a specific site, planting trees more densely, and using plowing methods designed to break up compaction and create some relief in elevation would amend these issues. Traditionally, floodplain restorations have focused on improving water quality and providing habitat for wildlife, but the benefits of storing carbon have become increasingly valuable in recent years. While we know that wetlands store carbon, research is needed to examine how quickly and where carbon is stored. Our research quantifies forest establishment and carbon storage of very densely planted, 12-year-old American sycamore (Platanus occidentalis) and willow oak (Quercus phellos) plantations. This research examines the effects four soil tillage methods and tree species selection had on tree survival and carbon storage. Carbon storage was measured in vegetation, soil, and small roots. Twelve years after planting, species selection had a greater impact on carbon storage than soil tillage methods. No tillage method altered survival or total carbon after 12 years, but American sycamore was found to have better survival and more carbon than willow oak in all soil tillage treatments. Both species were successful in establishing planted forests on the floodplain, but the success was aided by the very high initial numbers of planted trees that ensured enough trees would survive on the site even after trees were lost to mortality. Creation of willow oak forests benefitted more from dense planting, as it was less suited to the site and more planted willow oaks died. We recommend tillage for compacted soil, selecting the most suitable tree species, and planting adequate numbers of trees.

Impatiens Necrotic Spot Virus Resistance in Transgenic Impatiens walleriana and Lycopersicon esculentum

This project evaluated I. walleriana and Micro-Tom transformed with Agrobacterium. The construct contained GFP (green fluorescent protein) and hygromycin antibiotic-resistant selectable markers, and the antisense sequence of open reading frame of INSV nucleocapsid protein (N). The N gene is expected to confer INSV resistance by RNA interference or gene silencing. The presence of transgenes was confirmed by PCR. Transgenic Impatiens was selfed for two generations. Transgenic Micro-Tom was selfed for 4 generations. Spinach was used as an INSV reservoir. Impatiens, spinach and Micro-Tom were mechanically inoculated with INSV and evaluated visually, with assay tests, ELISA testing, and PCR. It has a wide variety of flower colors and grows well in shade. Impatiens necrotic spot virus (INSV) is an incurable virus that causes disfiguring dead spots on plants. Micro-Tom is miniature tomato used for research due to its small size and short time to fruiting. This project tested impatiens and Micro-Tom transgenic plants that had been genetically modified using bacteria. The bacteria had been modified to contain ‘markers’ which allow researchers to confirm the modifications were successful. It also contained a small piece of genetic material from the virus, which was expected to make the plants resistant to the virus by interfering with virus movement and reproduction. These transgenic plants were self-pollinated for multiple generations and tested to confirm the transgene was present. “Wild-type” (not genetically modified) spinach was infected with the virus by hand and infected spinach leaves were used to try to infect impatiens and Micro-Tom. The plants were inspected visually and leaves were tested for presence of the virus. Spinach was successfully infected with INSV six times of seven attempts.
