Influência de diferentes condições da ionosfera no posicionamento por ponto com GPS : avaliação na região brasileira /

Aquino / Banca: Inez Staciarini Batista / Banca: Claudio Antonio Brunini / Após a desativação da técnica SA, a ionosfera tornou-se a principal fonte de erro no posicionamento com GPS. O erro associado à ionosfera é diretamente proporcional ao conteúdo total de elétrons (TEC - Total Electron Content) presente ao longo do caminho da trajetória percorrida pelo sinal na ionosfera e inversamente proporcional ao quadrado da freqüência do sinal. O TEC, e conseqüentemente o erro devido à ionosfera, variam no tempo e no espaço e é influenciado por diversas variáveis, tais como: ciclo solar, época do ano, hora do dia, localização geográfica, atividade geomagnética, entre outros. A região brasileira é um dos locais que apresenta os maiores valores e variações espaciais do TEC e onde estão presentes diversas particularidades da ionosfera, tais como, a anomalia equatorial e o efeito da cintilação ionosférica. Desta forma, é importante a realização de pesquisas que visam estudar o comportamento do TEC, e conseqüentemente do erro devido à ionosfera no Brasil, que é um trabalho complexo devido aos diversos fatores que influenciam a variação do TEC, além das particularidades presentes na região brasileira. Estudos desta natureza podem auxiliar a comunidade geodésica brasileira, e demais usuários do GPS, no entendimento das limitações impostas pela ionosfera nas regiões de interesse. Devido à natureza dispersiva da ionosfera, o estudo do comportamento do TEC no Brasil pode ser realizado utilizando os dados GPS de receptores de dupla freqüência pertencentes à RBMC (Rede Brasileira de Monitoramento Contínuo). Adicionalmente, para uma melhor análise, pode-se também utilizar dados das estações da rede IGS (International GNSS Service) da América do Sul. / In the SA absence, the ionosphere is the largest error source in GPS positioning. The error due to the ionosphere in the GPS observables depends on the signal frequency and Total Electron Content (TEC) in the ionospheric layer. The TEC varies regularly in time and space in relation to the sunspot number, the season, the local time, the geographic position, and others. The Brazilian region is one of the regions of the Earth that presents largest values and space variations of the TEC, being influenced by the equatorial anomaly of ionization and ionospheric scintillation. Therefore, it is important to study the TEC behavior in the Brazilian region. Due to the ionosphere dispersive nature, the TEC behavior in Brazil can be studied using GPS data from RBMC (Rede Brasileira de Monitoramento Contínuo - Brazilian Network for Continuous Monitoring of GPS). Additionally, GPS data from IGS (International GNSS Service) network of the South America can also be used in the experiments. / Doutor

Utilising probabilistic techniques in the assessment of extreme coastal flooding frequency-magnitude relationships using a case study from south-west England

Recent events such as the New Orleans floods and the Japanese tsunami of 2011 have highlighted the uncertainty in the quantification of the magnitude of natural hazards. The research undertaken here has focussed on the uncertainty in evaluating storm surge magnitudes based on a range of statistical techniques including the Generalised Extreme Value distribution, Joint Probability and Monte Carlo simulations. To support the evaluation of storm surge frequency magnitude relationships a unique hard copy observed sea level data set, recording hourly observations, was acquired and digitised for Devonport, Plymouth, creating a 40 year data set. In conjunction with Devonport data, Newlyn (1915-2012) tide gauge records were analysed, creating a data set of 2 million data points. The different statistical techniques analysed led to an uncertainty range of 0.4 m for a 1 in 250 year storm surge event, and 0.7 m for a 1 in 1000 storm surge event. This compares to a 0.5 m uncertainty range between the low and high prediction for sea level rise by 2100. The Geographical Information system modelling of the uncertainty indicated that for a 1 in 1000 year event the level uncertainty (0.7 m) led to an increase of 100% of buildings and 50% of total land affect. Within the study area of south-west England there are several critical structures including a nuclear licensed site. Incorporating the uncertainty in storm surge and wave height predictions indicated that the site would be potentially affected today with the combination of a 1 in 1000 year storm surge event coincident with a 1 in 1000 wave. In addition to the evaluation of frequency magnitude relations this study has identified several trends in the data set. Over the data period sea level rise is modelled as an exponential growth (0.0001mm/yr2), indicating the modelled sea level rise of 1.9 mm/yr and 2.2 mm/yr for Newlyn and Devonport, will potentially increase over the next century by a minimum of 0.2 m by 2100.The increase in storm frequency identified as part of this analysis has been equated to the rise in sea level, rather than an increase in the severity of storms, with decadal variations in the observed frequency, potentially linked to the North Atlantic Oscillation. The identification as part of this study of a significant uncertainty in the evaluation of storm surge frequency magnitude relationships has global significance in the evaluation of natural hazards. Guidance on the evaluation of external hazards currently does not adequately consider the effect of uncertainty; an uncertainty of 0.7 m identified within this study could potentially affect in the region of 500 million people worldwide living close to the coast.

Pollution transport in the Boden garrison storm water

In 2012, the Swedish fortification agency started an environmental program for their real estate in northern Sweden. For the Boden garrison this meant an evaluation was needed for their storm water handling. The evaluation fell upon Grontmij AB in Boden. This thesis concerns the pollution from the storm water pipes into its recipient Lule River. Its aim was also to evaluate the areas for the event of a bigger oil spill, since there are many mechanical garages within the garrison. The pollution concerning oil was evaluated by calculating the flow in each outlet into the river, with different rain intensities. For other pollutants, the volumes of storm water each year, for every outlet were calculated. With concern for the snow melting process, it was also calculated for which period of the year would be interesting to keep an extra watch for pollutant concentrations and maximum flows. Results showed that for a 50-year period, none of the years would have had their peak flow because of the snow melt. The results for evaluation of an oil spill showed that none of the outlets were equipped for an accident of such sort, should it coincide with a rainfall of a high intensity. The results also showed that one of the outlets had an oil trap only dimensioned for rainfall of the lowest intensity. The calculating of pollutant volumes raises the concern for reliable data, but measures have already been taken to meet this concern.

In the present work, we dedicate to the study of the attenuation of these scintillations that is, an improvement in the signal, during the main magnetic storm phase during the period of October 2006 to February 2007. Using amplitude of scintillation 1.5GHz (L1) data of the net of satellites GPS, in the ionospheric station of Natal (5.84o S, 35.20o O, -20o dip) and geomagnetic indices, during the minimum solar cycle (referred to as cycle 23), demonstrating its anti-correlation between magnetic activity (Kp) and index of scintillation (<S4>) (Bonelli2005 method, that works for solar maximum, Bonelli(2005)). These results show that these storms correspond to category I of Aarons (1991). The magnetic storms can generate irregularities when the electric feld of penetration eastward on the pre-reversal hour intensificating that and can too generate irregularities on midnight and sunlight period. The limitation of the method applied here is that it is not considering some storms that had also occurred during equinox and summer in the Brazilian region. For this reason, we will use additional data of the stations of S?o Jo?o de Cariri (Imager and Photometer) and of the station at Fortaleza (Digissonde data), as aid to analyze these storms. The storms that had been left out by the applied method, fit in the other two categories of Aarons, with one only exception (storm of January, 02). We show that in the day of the main phase of the magnetic storm, and with presence of bubbles (according to data of the Imager and Photometer), the speed of vertical drift ~E ~B (hF=t) is below 20m/s, that is the threshold found for Anderson al., (2004) (>20m/s as condition favorable to the formation of irregularities and increase in the scintillation index). Uppföljning av reningsfunktionen i Steningedalens våtmark / Follow-Up of the Water Purification Function at Wetland of Steningedalen

Dagvattnet från Märsta är belastat med bland annat tungmetaller och näringsämnen. Vattnet hamnar i Märstaån och transporteras vidare ut till Mälaren. Steningedalens våtmark är ett dammsystem som renar och fördröjer en del av Märstaåns flöde före transport ut mot Mälaren.   Syftet med detta självständiga arbete på kandidatnivå har varit att undersöka anläggningens reningsfunktion med hjälp av sedimentprovtagning. Sedimentprover har tagits vid in- och utloppet och en kemisk analys av dessa har utförts i ett ackrediterat laboratorium. Resultaten från in- och utloppet har jämförts för att se om det sker en minskning av föroreningar genom systemet. Fem sedimentfällor har placerats ut i delar av dammsystemet för att undersöka sedimentfördelningen och muddringsbehovet i anläggningen. Resultatet av studien visade att dammsystemet har en god vattenreningsfunktion med avseende på tungmetallerna bly, nickel, kadmium, krom, koppar och zink samt näringsämnet fosfor. Anläggningen har bedömts att vara i behov av muddring. / In Märsta, the storm water is contaminated with heavy metals and nutrients (nitrogen and phosphorus) among other pollutants. The water emits to the river Märstaån and further to the lake Mälaren. The wetland of Steningedalen is a pond system that delays part of the stream in Märstaån and purifies the water on its way through the system. This independent project is based on investigation of the purification functioning at Steningedalen wetland. The investigation has been done by sediment sampling and analysis. The samples were taken from the inlet and the outlet of the pond system. Chemical analysis has been made on these samples in an accredited laboratory. The results were compared in order to see if there is a trend through the system. Five sediment traps were placed in parts of the system to study the distribution of the sediment and the need of dredging. The results of the study have concluded that the stormwater pond system has a good water purification function with regard to heavy metals lead, nickel, cadmium, chromium, copper and and zinc aswell as the nutrient phosphorous. The study has also shown that the system is in need of dredging.

The objective of this research is to evaluate infiltration structures for the control of excess drainage at the origin of runoff. These structures are constructed with the purpose of minimizing the impact of impervious surfaces to the urban environment. An experimental field was established consisting of several infiltration structures, such as small-diameter pilot wells and real scale wells. The real scale wells used perforated concrete pipe casing and brick lining. Falling-head infiltration tests were carried out during 15 months, encompassing the dry and rainy seasons. The infiltration tests were undertaken with initial conditions that correspond to the natural water content at the time of testing. Additional tests were undertaken with previously flooded conditions, reproducing several consecutive rainy days. The monitoring of pilot wells along with real scale wells allowed the evaluation of project parameters obtained from pilot wells. The values of average infiltration rate of different infiltration wells were compared and the performance of the several structures was compared. Laboratory test were carried out for the determination of the hydraulic parameters of the ground profile, including the saturated hydraulic conductivity the soil-water characteristic curve. A numerical model for the simulation of infiltration wells was implemented using the software FlexPDE. Special boundary conditions were developed that take into account the transient relationship between the varying hydraulic head at the well walls and the total volume infiltrated during a test. The numerical simulations contributed for a better understanding of the infiltration mechanism, the influence of the hydraulic conductivity to the infiltration rate, and the determination of the wetting front and pore-water pressures during infiltration. A design criterion for infiltration wells was proposed using daily compensation, considering the daily water balance during 25 years, and the precipitation historical series of the site. The proposed design criterion makes possible the selection of the group of infiltration structures that meet the project goals. Doctor of Philosophy / Department of History / Donald J. Mrozek / During the Cold War, the United States’ foreign policy relied heavily on its ability to project military power. More often than not, the central component of force projection rested on the United States military’s effectiveness in employing air power both by establishing air superiority and through accurate delivery of ordnance. As the primary service tasked with conducting aerial warfare, the United States Air Force (USAF) was expected to maintain this capability either to achieve deterrence or, when necessary, to military action. In January 1973, the USAF seemed incapable of performing the latter task due to the North Vietnamese Integrated Air Defense System’s (NV-IAD’s) effectiveness in Operation Rolling Thunder and its successor, Operation Linebacker. Eighteen years later, Air Force aircraft spearheaded the Coalition’s air attack on the Iraqi Integrated Air Defense System (I-IADS) in January 1991. Considered by many to be the most effective air defense system outside the Soviet Union’s, the I-IADS was expected to exact heavy casualties from the allied forces. Instead, in less than twenty days, the USAF’s dominance was so complete that politicians, analysts and military historians quickly proclaimed a “Revolution in Military Affairs” (RMA). The majority of the current historiography credits advances in precision-guided munitions (PGMs), airframes, and computer technology as the impetus for the RMA. Others have claimed that the USAF’s training methodology and construction of advanced training sites such as the Red Flag complex at Nellis Air Force Base were the primary drivers for the Air Force’s success. While acknowledging the role all of these factors played, this dissertation also demonstrates the key role played by the development of Suppression of Enemy Air Defense (SEAD) doctrine from January 1973 through August 1991. In the aftermath of the American war in Vietnam, the Air Force considered defense suppression a tactical task that was secondary to the primary mission of putting ordnance on target. At the end of Desert Storm, proponents of the Air Force’s SEAD doctrine had convincing evidence that an enemy IADS was not just an ancillary weapons array, but functioned a critical national system just like manufacturing, government, or the people’s will. The process by which this viewpoint changed had effects on the development of the United States Air Force’s Cold War conventional capability in general, and the development of training methods, electronic warfare platforms, and modern airframes specifically.

The role of synaptopodin for the diffusion of membrane protein in the dendritic spine neck / Le rôle de synaptopodine dans la diffusion des protéines membranaires dans la tige des épines dendritiques

Au sein des synapses comme dans les régions extra synaptiques, la diffusion latérale joue un rôle critique dans la densité membranaire des récepteurs. En face des zones actives, l’accumulation de récepteurs détermine en particulier l’efficacité de la transmission synaptique. Il est important de comprendre les paramètres cellulaires qui jouent sur l’accès au compartiment synaptique, qu’ils soient d’origine moléculaires ou morphologiques. Dans les synapses excitatrices, la tige de l’épine dendritique se comporte comme une barrière à la diffusion. Cette barrière pourrait être fonction de la longueur et du diamètre de la tige (paramètre géométrique), ou résider dans la présence d’éléments spécifiques constituant des obstacles à la diffusion. Une sous-population d’épines contient dans sa tige une forme spécialisée de réticulum endoplasmique, appelé appareil épineux et constituée d’un empilement des accules de réticulum. Une protéine liant l’actine, nommée synaptopodine, est associée de façon étroite à l’appareil épineux et participe aux mécanismes de plasticité synaptique. La question centrale de ce travail de thèse était de définir si la présence de synaptopodine influait sur les caractéristiques de la diffusion dans la tige de l’épine, et d’identifier les mécanismes sous-jacents. Afin d’étudier la diffusion membranaire, j’ai utilisé trois protéines recombinantes différentes: une protéine associée au feuillet extérieur de la membrane plasmique (GFP-GPI), une protéine avec un domaine transmembranaire et une courte séquence intracellulaire (TMD-pHluorin), et la sous-unitéGluR5 du récepteur métabotropique (mGluR5) contenant 7 domaines transmembranaires et une séquence intracellulaire volumineuse. Les trois constructions portent une étiquette (GFP ou pHluorin) du côté extracellulaire. Les propriétés diffusives de ces molécules ont été mesurées par un suivi de particules uniques, à base de quantum dots. Ces expériences ont révélé que la diffusion des protéines membranaires est fonction du diamètre de la structure cylindrique considérée, et par conséquent moins rapide dans la tige de l’épine que dans le tronc du dendrite. Mais les propritétés diffusives dépendent aussi de la taille et delà complexité des molécules membranaires considérées. En effet, la diffusion de molécules comportant des domaines transmembranaires est particulièrement faible dans les tiges contenant de la synaptopodine. Cet aspect a été approfondi par l’utilisation de traitements pharmacologiques, qui ont permis de modifier la structure interne de la tige dendritique. Les variations des tailles des domainesoccupés par l’actine-F, et par lesaggrégats de synaptopodine, ont été observées à l’échelle nanoscopique en utilisant l’imagerie PALM/STORM. En conditions contrôle, la synaptopodine occupe la partie centrale de la tige. La dépolymérisation indirecte de l’actine-F par le 4-Aminopyridineentraîne une diminution des zones occupées par ces deux composants, corrélée à une augmentation de la vitesse de diffusion de mGluR5. En revanche, la dépolymérisation par la latrunculin-A (effet direct sur l’actine) induit une augmentation de la taille des clusters de synaptopodine et donc de la surface occupée par ceux-ci dans la tige. Les mesures de la diffusion de la sous-unité mGluR5 réalisées dans ces conditions montrent une accélération de la vitesse de diffusion, indiquant que la mobilité de mGluR5 n’est pas régulée par une interaction directe avec la synaptopodine. En conclusion, je propose un rôle de stabilisation mutuel pourl’actine-F et la synaptopodinedans la tige des épines dendritiques de neurones d’hippocampe en culture. Les épines contenant de la synaptopodine dans leur tige auraient une organisation unique du cytosquelette qui agirait comme une barrière additionnelle pour la diffusion de récepteurs aux neurotransmetteurs. / Lateral diffusion in and outside synapses plays a key role in the accumulation of receptors at synapses, which critically determines the efficacy of synaptic neurotransmission. Therefore, to better understand the trapping of neurotransmitter receptors in synapses, it is important to investigate the mechanisms that may affect receptors diffusion and their capacity to reach synapses. The neck of dendritic spine imposes a diffusional barrier that is considered to depend on the length and diameter of the spine neck. The origin of this barrier could be purely geometrical or could be induced by the presence of specific barriers/obstacles for diffusion. A subpopulation of spines contains a specialized form of endoplasmic reticulum in the spine neck called spine apparatus. The actin-binding protein synaptopodin (SP) is tightly associated with the spine apparatus and participates in synaptic plasticity mechanisms. The central question of my research was to assess whether the presence of the SP affects the diffusion of receptors in the spine neck and to characterize the underlying molecular mechanisms. To study membrane diffusion, I have developed three different probes: a construct associated with the outer leaflet of the plasma membrane (GFP-GPI), a construct with one transmembrane domain and a short intracellular sequence (TMD-pHluorin), and a recombinant metabotropic mGluR5 receptor construct containing an extracellular domain tagged with pHluorin, seven transmembrane domains, as well as a large intracellular region. The diffusion properties of these molecules were measured by single particle tracking using quantum dots. My experiments revealed that the diffusion of membrane proteins was slower in the spine neck than in the dendrite as a result of the different diameter of the two compartments. Furthermore, the diffusion properties depended on the molecular size and complexity of the membrane proteins. Interestingly, the diffusion of membrane proteins with transmembrane domains was particular slow in spine necks containing SP. This could be the result of direct molecular interactions between the membrane proteins and SP or due to spatial constraints that are related to the structural organization of spine necks expressing SP. To address these questions further I used pharmacological treatments to change the internal organization of the spine neck, and measured their effect on the diffusion properties of mGluR5. The distribution of SP and F-actin in the spine neck was determined on the nanoscopic scale using PALM/STORM imaging. This showed that under control condition SP occupies only the central region of the spine neck. Activity-dependent depolymerization of F-actin by 4-Aminopyridine led to a simultaneous decrease of the amount of F-actin and SP and enhanced the diffusion of mGluR5 in all analyzed neck regions. Disruption of F-actin by latrunculin A induced the re-distribution of SP and the formation of larger SP clusters, occupying an increased region within the spine neck. The recruitment of SP was accompanied by an acceleration of mGluR5 diffusion in SP-positive spines, demonstrating that the mobility of mGluR5 is not controlled by direct interactions with SP. Instead, the diffusion of mGluR5 is dependent on the organization of the spine cytoskeleton. In conclusion, I propose that SP and the polymerization of actin filaments have a reciprocal effect on the stability of each other in the spine neck of cultured hippocampal neurons. Spine necks bearing SP have a unique F-actin cytoskeletal organization that acts as an additional diffusion barrier for neurotransmitter receptors such as mGluR5.

Restoration of the naturalness of boreal forests

Abstract Restoration is considered to be a crucial action in order to maintain and enhance ecosystem functioning and halt the decline of biodiversity. Within forested ecosystems, heavily exploited forests have lost a great part of their biodiversity values, and even nature conservation is insufficient to prevent the loss of habitats and the endangerment of species. The aim of this thesis was to provide new information on the impacts of forest restoration methods on boreal forest naturalness, including forest structure, forest-dwelling species and ecosystem processes and functions. This information is needed to develop the restoration methods and their cost-efficiency and to support decision-making concerning restoration and nature conservation. The restoration methods studied were felling with a chainsaw either without or with subsequent burning, and storm treatment, in which the trees were uprooted with an excavator. The variables measured were vegetation assemblages, deadwood dynamics, and deadwood-dependent insect assemblages. In addition to field measurements, a simulation approach was used so as to predict deadwood continuity after different restoration methods. The main finding of this thesis is that the aspects of naturalness studied were most enhanced by burning, whereas felling had the least effect. Restoration burning increased deadwood volume and diversity, enabled the establishment of pioneer plants, increased the relative cover of the forest keystone species Vaccinium myrtillus, enhanced the regeneration of the keystone tree Populus tremula, and provided habitat for red-listed, especially pyrophilous beetles (Coleoptera) and flat bugs (Heteroptera: Aradidae). Felling only increased the volume of deadwood. Storm treatment with tree uprooting was a more effective method than felling with a chainsaw, due to the additional disturbance it caused to ground, enhancing the regeneration of e.g. Pinus sylvestris. According to simulation models, compared to controls restored stands are predicted to have greater deadwood volumes at least for 40 years. The study shows that restoration can be used to accelerate the development of degraded forests towards a higher level of naturalness. The results can be used to choose appropriate restoration methods for forests, based on their initial stage and the goal set for the level of their naturalness. / Tiivistelmä Ennallistamista pidetään tärkeänä keinona palauttaa ja ylläpitää luonnontilaisten ekosysteemien toimintaa. Metsien ennallistamista tarvitaan, koska tehokkaasti hoidettujen metsien monimuotoisuus on köyhtynyt niin pitkälle, ettei pelkkä suojelu enää riitä pysäyttämään elinympäristöjen häviämistä ja lajien uhanalaistumista. Väitöskirjan tavoitteena oli tuottaa uutta tietoa siitä, miten erilaiset metsien ennallistamiskeinot vaikuttavat metsän luonnontilaisuuden elementteihin, kuten rakennepiirteisiin, lajistoon ja toiminnallisuuteen. Tietoa tarvitaan ennallistamismenetelmien ja niiden kustannustehokkuuden parantamiseksi sekä tukemaan ennallistamista ja luonnonsuojelua koskevaa päätöksentekoa. Tutkitut ennallistamismenetelmät olivat: lahopuun lisäys sahaamalla joko ilman polttoa tai polton kanssa ja myrskytuhokäsittely, jossa puut kaadettiin juurineen. Metsän aluskasvillisuus, lahopuusto ja lahopuusta riippuvaisten hyönteisten lajiyhteisöt olivat mitattuja muuttujia. Maastoinventointien ohella tehtiin simulaatiomallinnuksia, joilla ennustettiin ennallistamisen jälkeistä lahopuujatkumoa. Väitöskirjatyön keskeisin havainto on, että ennallistamispoltot paransivat tutkittuja luonnontilaisuuden elementtejä eniten, kun taas lahopuun lisäyksellä oli vähäisimmät vaikutukset. Ennallistamispoltto lisäsi lahopuun tilavuutta ja monimuotoisuutta, mahdollisti sukkession varhaisvaiheen kasvillisuuden kehittymisen, lisäsi mustikan (Vaccinium myrtillus) suhteellista peittävyyttä, paransi monimuotoisuudelle tärkeän puulajin, haavan (Populus tremula), uudistumista sekä lisäsi lahopuuriippuvaisten, erityisesti palanutta puuta suosivien kovakuoriaisten (Coleoptera) ja latikoiden (Heteroptera:Aradidae), laji- ja yksilömääriä. Myrskytuhon jäljittely oli tehokkaampi ennallistamiskeino kuin lahopuun lisäys sahaamalla, sillä juurilaikku lisäsi maanpinnan häiriötä parantaen samalla mm. männyn (Pinus sylvestris) taimettumista. Simulaatiomallinnus osoitti, että verrattuna kontrollialueisiin ennallistetuilla alueilla lahopuuta on runsaammin vähintään 40 vuoden ajan. Väitöskirja osoittaa, että kaikkia työssä tutkittuja ennallistamismenetelmiä voidaan käyttää nopeuttamaan metsän kehittymistä kohti luonnontilaa, mutta menetelmät eroavat toisistaan vaikutuksiltaan. Saatuja tuloksia voidaan käyttää, kun valitaan metsiin niihin parhaiten soveltuvaa ennallistamismenetelmää.
