Automatic shape optimisation of pharmaceutical tablets using Partial Differential Equations

No / Pharmaceutical tablets have been the most dominant form for drug delivery and most of them are used in the oral administration of drugs. These tablets need to be strong enough so that they can tolerate external stresses. Hence, during the design process, it is important to produce tablets with maximum mechanical strength while conserving the properties of powder form components. The mechanical properties of these tablets are assessed by measuring the tensile strength, which is commonly measured using diametrical or axial compression tests. This work describes the parametric design and optimisation of solid pharmaceutical tablets in cylindrical and spherical shapes, which are obtained using a formulation based on the use of Partial Differential Equations (PDEs) for shape design. The PDE-based formulation is capable of parameterised complex shapes using the information from some boundary curves that describe the shape. It is shown that the optimal design of pharmaceutical tablets with a particular volume and maximum strength can be obtained using an automatic design optimisation which is performed by combining the PDE method and a standard method for numerical optimisation.

This study was developed with the students of elementary school. We also treat topics which are not normally worked in basic Education as Bézout's theorem and Euler function. We notice the use of these apps in the classroom brought more enthusiasm for students.

The thesis evaluates three types of anhydrous β-lactose combined with lubri- cants. The evaluation is made from the standpoint of compressibilty, tensile strength and ejection force, depending on the compression force used. Studied materials are Duralac H, SuperTab 21AN and Lactopress Anhydrous 250. Lubricants used in the concentration of 1% are magnesium stearate, glycerol-dibehenate and poloxamer 188. Compressibility is evaluated by the energy profile of compression. Total energy of compression increases with compression force used and de- pends on the type of anhydrous β-lactose and type of lubricant. Higher plasticity is shown by Duralac H and SuperTab 21AN. Ejection force is lowest in the case of Duralac H for all lubricants. Magnesium stearate appears to be the best lubricant for all types of anhydrous β-lactose. The effect of lubricants on the tensile strength of tablets depends on the compression force used. Highest values of disintegration time provides SuperTab 21AN, Duralac H with glycerol-dibehenate provides lowest values. 1

The thesis deals with the comparison of compressibility and properties of tablets containing silicified microcrystalline cellulose and physical mixtures of microcrystalline cellulose with different types of colloidal silicon dioxide. The used excipients were silicified microcrystalline cellulose Prosolv® SMCC 90, microcrystalline cellulose Avicel® PH-102 and colloidal silicon dioxide Aerosil® 200 and 255. Tablets were compressed at three compression forces 2.5, 3 and 3.5 kN. The energy profile of compression, the tensile strength of tablets and the disintegration time of tablets were evaluated. The effect of 1 % magnesium stearate on these parameters under various mixing conditions and its homogenity in the tablet were evaluated. Total energy of compression increased with the growing compression force, it was the highest at Prosolv® SMCC 90. Plasticity decreased with compression force, mixtures of Avicel® PH-102 with both types of Aerosil showed its the highest values. These mixtures provided tablets with lower strength than Avicel® PH-102 and Prosolv® SMCC 90. Disintegration time of tablets increased with the growing compression force and it was longer in the case of tablets from Prosolv® SMCC 90 and Avicel® PH-102. Desenvolvimento de comprimidos com propriedades mucoadesivas contendo anestésicos para aplicação bucal / Development of tablets with mucoadhesive properties containing anesthetics for buccal application

Os anestésicos locais têm sido utilizados na clínica médica e odontológica com o objetivo de atenuar a dor nos procedimentos cirúrgicos. Tais fármacos são administrados usualmente por meio do uso de agulhas, o que pode diminuir o número de pacientes nos consultórios dentários, afugentados pelo medo desses dispositivos invasivos. Comprimidos mucoadesivos de dissolução rápida podem ser promissores em incorporar esses anestésicos e assim liberá-los topicamente com a finalidade de promover a anestesia de forma não invasiva. Neste intuito, comprimidos de dissolução rápida contendo os anestésicos locais cloridratos de prilocaína (PCL) e lidocaína (LDC) foram desenvolvidos por três métodos: i) compressão direta; ii) compressão após pré-processamento dos adjuvantes em secagem por atomização e iii) liofilização. Estes foram comparados em relação ao tempo de hidratação e desintegração para a escolha de uma plataforma para estudos de liberação, permeação e mucoadesão in vitro. Filme polimérico oclusivo com Eudragit S100 foi moldável e flexível para a finalidade de revestimento parcial. A massa total de desintegrantes nos comprimidos modularam sua hidratação e desintegração. Os comprimidos obtidos por liofilização apresentaram menores valores de tempo de hidratação e desintegração. Os efeitos de diluente (manitol), desintegrante (glicolato sódico de amido), promotores químicos de permeação (ácido oleico, Tween® 80 e propilenoglicol) e polímeros mucoadesivos (HPMC e pullulan) sobre liberação em éster de celulose e permeação em esôfago suíno foram avaliados. Os comprimidos apresentaram rápida liberação em uma hora. A liberação obedeceu a cinética de primeira ordem e o mecanismo de liberação foi governado pelo transporte não fickiano. A proporção de manitol e tipo de polímero mucoadesivo não teve influência significativa nos estudos de liberação. A quantidade de fármacos liberada diminuiu em comprimidos com desintegrantes e promotores químicos de permeação. Um significante efeito sinérgico entre polímero mucoadesivo e promotores químicos no coeficiente de permeabilidade, fluxo e retenção de fármacos na mucosa foi observado. Quando comparado com comprimidos de HPMC com promotores químicos, o pullulam melhorou a permeação de fármacos através da mucosa. Mucoadesão dos comprimidos com diferentes polímeros foi avaliado. HPMC e pullulan melhoraram as propriedades mucoadesivas. Todos os comprimidos se mantiveram aderidos na região do epitélio durante uma hora. A nova plataforma de liberação de fármacos obtida pela combinação de tecnologias farmacêuticas de comprimidos por liofilização, com adição de promotores de permeação e pullulan como polímero mucoadesivo mostrou uma estratégia efetiva para o desenvolvimento de um sistema transbucal para LDC e PRC que pode ser usada para melhorar anestésica sem uso de agulhas / Local anesthetics have been used in medical and dental practice aiming at to become less painful some surgical procedures. Such drugs are typically administered through needles along surgical procedures, which decrease the number of patients in the dental offices, once many people are afraid of these invasive devices. Fast dissolving and mucoadhesive tablets could be promising for incorporating these anesthetics and thus releasing them topically in order to improve noninvasive anesthesia. Herein, it were developed fast dissolving tablets containing the local anesthetic drugs prilocaine (PRC) and lidocaine (LDC) hydrochloride. Three methods were used: a) direct compression of the mixture of components; b) pre-processing by spray drying adjuvants and c) freeze drying and these tablets. The tablets were compared in terms of wetting and disintegration time for the choice of a platform for further in vitro release, permeation and mucoadhesion studies. An occlusive polymeric film of Eudragit S100 showed suitable flexibility and plasticity according to purpose of coating the tablets. The total mass of disintegrants in formulations affected the wetting and disintegration of the tablets. Freeze dryed tablets had lower wetting and disintegration time values as compared to the others. The effects of diluent (mannitol), disintegrant (sodium starch glycolate), chemical enhancers (oleic acid, Tween® 80 and propylene glycol) and mucoadhesive polymers (hydroxypropyl methylcellulose and pullulan) on the drug release from cellulose ester membrane and drug permeation through porcine esophageal mucosa were evaluated. Dissolution test showed fast release on one hour. The drug release data fit well to the First order expression and the release mechanism was non-Fickian transport. No significant influence of proportion of mannitol and type of mucoadhesive polymer on release studies was observed. Release of drugs decreased in tablets with disintegrant and chemical enhancers. A significant synergic effect between the mucoadhesive polymer and chemical enhancers on the permeability coefficient, flux and retention of drugs on mucosa was observed. As compared to HPMC for tablets containing chemical enhancers, pullulan improved the drug permeation through the mucosa. Mucoadhesion to tablets with different polymers was evaluated. HPMC and pullulan improve mucoadhesive properties. All tablets maintained in the attachment site of the epithelium for at least one hour. It conclusion, the novel drug delivery platform achieved by combining the pharmaceutical technologies of freeze-dryed tablets comprising chemical enhancers and pullulan as mucoadhesive polymer displayed an effective strategy for the development of a transbuccal system for LDC and PRC that can be used to improve needle-free buccal anesthesia.

This plan is a public educational policy of social and digital inclusion implemented by the Uruguayan government in 2007, which aims to universalize access to digital technology (computer and Internet), improve the quality of education and bridge the digital divide. Currently, all students and teachers of public education have a portable computer (proprietary or library mode) with free Internet access in various public spaces in the country, such as educational centers, public squares, clubs, libraries, training centers and priority attention districts. The objective of the research is to analyze what kind of images and linkages CEIBAL plan is fomenting through its educational activities, since the electrified and instantaneous communication tends to privilege the hearing and the vision, to the detriment of the touch, smell and taste. This privilege ends up stimulating more distance linkages over those of proximity and intimacy. As we capture the world through our senses and link ourselves through images, the excess of mediations in the communicative process can modify our perception of the present, of the time, of the body and also of our way of socialization. We use as theoretical framework Vilém Flusser’s assertion of Abstraction Theory, Harry Pross’ notion of Theory Media, Boris Cyrulnik’s sense of belonging and the importance of a linkage tutor, Antonio Damásio’s emotion and feeling assertion, Dietmar Kamper’s theory of the loss of the present and the loss of the body, and Norval Baitello Jr.’s notion of linkage, capillaries and communicational environments. From a dialogical perspective to observe, describe and understand the object of research, this thesis presents evidences that even in environments saturated with digital technologies, flat images and abstractions, it is possible to promote communicational environments replete with images and proximity linkages. Therefore, distance images can be turned into proximity linkage factors / Esta tese tem como objeto de pesquisa o programa educativo CEIBAL en Inglés e o projeto Olimpíada de Robótica, Programación y Videojuegos desenvolvidos pelo Plano CEIBAL (Conectividad Educativa de Informática Básica para el Aprendizaje en Línea). Este plano é uma política educativa pública de inclusão social e digital decretada pelo governo uruguaio em 2007, que tem o objetivo de universalizar o acesso à tecnologia digital (computador e Internet), melhorar a qualidade da educação e diminuir a brecha digital. Atualmente, todos os alunos e professores do ensino público têm um computador portátil (modalidade proprietário ou biblioteca) com acesso gratuito à Internet em vários espaços públicos do país, como centros educativos, praças, clubes, bibliotecas, centros de formação e bairros de atenção prioritária. Como capturamos o mundo através de nossos sentidos e nos vinculamos através de imagens, o excesso de mediações no processo comunicativo pode modificar nossa percepção do presente, do tempo, do corpo e também a nossa forma de sociabilização. Utilizamos como marco teórico a noção de Escalada da Abstração, proposta por Vilém Flusser; a Teoria das Mediações, de Harry Pross; o sentimento de pertencimento e a importância do tutor de vinculação, de Boris Cyrulnik; a emoção e sentimento, de Antônio Damásio; a perda do presente e a perda do corpo, de Dietmar Kamper; e vinculação, capilaridades e ambientes comunicacionais, de Norval Baitello Jr. A partir de uma postura dialógica para se observar, descrever e compreender o objeto de pesquisa, esta tese registra indícios de que mesmo em ambientes saturados de tecnologias digitais, de imagens planas e abstrações, é possível promover ambientes de comunicação repletos de imagens e vínculos de proximidades.
