Viabilidade da utilização da terapia fotodinâmica no tratamento da estomatite protética. Estudos in vitro

Na primeira investigação, camundongos foram imunossuprimidos com injeções subcutâneas de prednisolona. Tetraciclina foi fornecida na água de beber para promover alteração da microbiota bucal dos camundongos. Os animais foram sedados com injeção de cloridrato de clorpromazina e um swab oral embebido em uma suspensão de C. albicans (107 UFC/mL) foi esfregado na cavidade bucal dos animais. Quatro dias após a inoculação, a PDT foi realizada no dorso lingual utilizando administração tópica de Photogem® a 400, 500 ou 1000 mg/L e, após 30 minutos, iluminação com 305 J/cm² de luz de LED a 455 ou 630 nm. Posteriormente, a quantidade de fungos viáveis foi determinada (UFC/mL) e analisada pelos testes de ANOVA e Holm- Sidak (P < 0,05). Os camundongos foram sacrificados, e as línguas foram removidas e processadas para avaliação histológica de presença de fungos e reação inflamatória. No estudo clínico, pacientes (n = 40) foram aleatoriamente atribuídos a um dos seguintes grupos de 20 indivíduos cada; grupo NYS: pacientes receberam tratamento tópico com nistatina (100.000 UI) quatro vezes ao dia por 15 dias e; grupo PDT: prótese total superior e o palato dos pacientes foram borrifados pelo Photogem® a 500 mg/L e, após 30 minutos de incubação, iluminado por luz de LED a 455 nm (37,5 e 122 J/cm2, respectivamente) três vezes por semana durante 15 dias. Investigação do efeito fotodinâmico da curcumina sobre espécies de Candida: estudos in vitro e in vivo

No primeiro estudo, suspensões planctônicas de C. albicans (ATCC 90028) foram expostas a nove concentrações de curcumina (0,005; 0,01; 0,05; 0,1; 0,5; 1, 5, 10 e 20μM) e oito doses de luz (1,32; 2,64; 3,96; 5,28; 6,60; 13,20 e 26,4J/cm2). O efeito de diferentes tempos de pré-irradiação (PIT), a possível captação de curcumina pelas células fúngicas e participação do oxigênio singlete na fotoinativação também foram avaliados. Adicionalmente, a PDT mediada pela curcumina foi avaliada em biofilmes formados pela cepa de referência de C. albicans e sobre culturas celulares de macrófagos. Além disso, as características ópticas da solução de curcumina foram investigadas em função da dose de luz utilizada. Os resultados foram submetidos à análise descritiva, Análise de Variância (ANOVA) e Kruskal-Wallis (α= 5%). As suspensões planctônicas de C. albicans foram completamente inativadas com a utilização de 20μM de curcumina e 5,28J/cm2. O efeito fotodinâmico foi acentuado pela presença da curcumina durante a iluminação... / The aim of this investigation was to evaluate: 1. the Photodynamic Therapy (PDT) mediated by curcumin for in vitro inactivation of a reference strain of Candida albicans and its citotoxic effects against a macrofage cell line; 2. the effectiveness of PDT mediated by curcumin to inactivate clinical isolates of C. albicans, Candida tropicalis and Candida glabrata, both in planktonic and biofilm phases; 3. the photoinactivation of C. albicans in a murine model of oral candidiasis using different curcumin concentrations. In the first study, suspensions of C. albicans were treated with nine curcumin concentrations (0.005; 0.01; 0.05; 0.1; 0.5; 1, 5, 10 and 20μM) and exposed to LED light at different fluences (1.32; 2.64; 3.96; 5.28; 6.60; 13.20 and 26.4J/cm2). The effect of the pre-irradiation time (PIT) on PDT effectiveness, the uptake of curcumin by C. albicans cells and the possible involvement of singlet oxygen in the photodynamic action was also evaluated. In addition, curcumin-mediated PDT was assessed against biofilms. Similar protocols were tested on a macrophage cell line and the effect was evaluated by MTT and SEM analysis. The optical properties of curcumin were investigated as a function of illumination fluence. Data were analyzed with Analysis of Variance (ANOVA), and Kruskal-Wallis test (α= 5%). The results showed a statistically significant reduction in C. albicans viability after PDT (p<0.05), for both planktonic and biofilm cultures. Photodynamic effect was greatly increased with the presence of curcumin in the surrounding media and the PIT of 20 minutes improved PDT effectiveness. Although PDT was phototoxic to macrophages, the therapy was more effective in inactivating the yeast cell than the defense cell. The spectral changes showed a high photobleaching rate of curcumin. Efetividade da irradiação por micro-ondas e da terapia fotodinâmica na inativação de micro-organismos presentes em próteses totais: estudo clínico

Esses micro-organismos podem ser transmitidos aos técnicos de laboratório por contato direto ou pelos aerossóis produzidos durante o desgaste ou o polimento de próteses contaminadas. Neste caso, os micro-organismos podem ser disseminados pelo ambiente, desencadeando infecção cruzada. Assim, para prevenir a contaminação cruzada, todas as próteses devem ser desinfetadas ao serem enviadas ao laboratório e ao retornarem ao consultório. Com base nessas considerações, o presente estudo avaliou a efetividade clínica da irradiação por micro-ondas e da terapia fotodinâmica (PDT) para desinfecção de próteses totais superiores. Para os procedimentos de irradiação por micro-ondas, amostras de biofilme das próteses foram coletadas de 30 pacientes, os quais foram divididos aleatoriamente em dois grupos experimentais com 15 indivíduos cada: Grupo 1 - as próteses foram expostas às micro-ondas durante 3 minutos (650 W), Grupo 2 - as próteses foram expostas às micro-ondas durante 2 minutos (650 W). Para os tratamentos com PDT, amostras de biofilme das próteses foram coletadas de 60 pacientes, os quais foram divididos aleatoriamente em quatro grupos experimentais (n = 15 cada): Grupos I e II - as próteses foram borrifadas com 50 mg/L e 100 mg/L do Photogem® em suspensão, respectivamente; Grupos III e IV - as próteses foram tratadas com 50 mg/L e 100 mg/L de Photogem® em gel, respectivamente. Após 30 minutos de pré-irradiação, todas as 60 próteses foram expostas à luz de LED azul com potência de 37,5 J/cm2 (26 minutos). Amostras das próteses foram obtidas utilizando-se swab estéril antes (nas superfícies do lado esquerdo) e após (nas superfícies do lado direito)... / Prosthetic appliances that have been in contact with oral tissues, saliva and blood can be contaminated with pathogenic microorganisms when they are removed from the patient’s mouth. These microorganisms can be transmitted to laboratory staff through direct contact with the infected denture or from aerosols that will be formed when the surfaces are ground or polished. In this case, microorganisms with varying degrees of virulence can be spread and disseminated into the air, leading to cross-infection. In order to prevent microbial cross-contamination among denture patients, all dental prostheses must be disinfected on entering and again on leaving the laboratory. Therefore, this investigation evaluated the clinical effectiveness of microwave irradiation and photodynamic therapy (PDT) on the inactivation of microorganisms present on complete dentures. For microwave irradiation procedures, biofilm samples were collected from dentures of 30 patients, who were randomly divided into two experimental groups of 15 subjects each: Group 1 - patients had their maxillary denture microwaved for 3 minutes (650 W); Group 2 - patients had their maxillary denture microwaved for 2 minutes (650 W). For PDT treatments, biofilm samples were collected from dentures of 60 patients, who were randomly divided into four experimental groups (n = 15 each): Groups I and II - patients had their maxillary denture sprayed with 50 mg/L and 100 mg/L of Photogem® suspension, respectively; Groups III and IV - patients had their maxillary denture treated with 50 mg/L and 100 mg/L of Photogem® gel, respectively. This work reports on the successful synthesis, characterisation and photophysical properties of new asymmetric metal free, magnesium and zinc phthalocyanines. The synthesis of symmetrical phthalocyanines is also reported. A selection of phthalocyanines have been conjugated, covalently linked, encapsulated or mixed with selected of drug delivery agents which include gold or silver nanoparticles, poly-L-lysine, liposomes and folic acid. The influence of delivery agent on the photophysical and photochemical properties of conjugated phthalocyanine is investigated. The studies showed that the Au nanoparticle significantly lowered the fluorescence quantum yield values of the phthalocyanines. The photodynamic activity of Zn phthalocyanine-ε-polylysine conjugates in the presence of nanoparticles towards the inactivation of Staphylococcus aureus showed high photoinactivation in the presence of silver nanoparticles. The presence of silver nanoparticels from the minimal inhibition concentration (MIC₅₀) studies showed remarkable growth inhibition for the tested conjugates even at low concentrations. The conjugate also showed no dark toxicity when evaluated using the chick choriallantoic membrane (CAM) assay. The efficiency of selected zinc phthalocyanine as photodynamic therapy (PDT) agents was investigated. The production of reactive oxygen species (ROS) and phototoxicity of the photosensitizers were assessed. Healthy fibroblast cells and breast cancer (MCF-7) cells were treated with either free phthalocyanine or phthalocyanine bound to either gold nanoparticles or encapsulated in liposomes. Cell viability studies showed the optimum phototoxic effect on non-malignant cells to be 4.5 J.cm⁻². The PDT effect of the liposome bound phthalocyanine showed extensive damage of the breast cancer cells. Gold nanoparticles only showed a modest improvement in PDT activity.

Low symmetry metallophthalocyanines and their nanoparticle conjugates for photodynamic antimicrobial chemotherapy

This thesis reports on the syntheses of novel low symmetrically substituted Zn, Sn, Ge and Ti MPc complexes containing a single carboxylic or cysteinyl group available for attachments to MNPs. The complexes were extensively characterized by various techniques to ensure their purity. Various metallic nanoparticles consisting of silver (AgNPs), gold (AuNPs) as well as quantum dots (QDs) were successfully prepared and conjugated to the low symmetry phthalocyanine complexes. The conjugates were successfully characterized using many techniques. The Q-band maxima of the MPcs were observed at completely different wavelength regions depending on the nature of the substituents and the central metal used. Blue shifting of the Q band in the absorption spectra was observed for the complexes in the presence of AuNPs, while aggregation was observed in the presence of quantum dots. The complexes were successfully electrospun into polymer fibers for the antimicrobial inhibition of bacteria. The photophysical and photochemical properties of these complexes were extensively investigated. Higher triplet and singlet oxygen quantum yields were achieved for the Ge Pc complexes, with all the complexes giving reasonable singlet oxygen quantum yields. An enhancement in triplet and singlet oxygen quantum yields was observed for all the complexes in the presence of metal nanoparticles. However, the singlet oxygen quantum yields decreased for all the complexes when incorporated into electrospun fibers. The antimicrobial behaviour of the complexes was investigated against Bacillus Subtilis and Staphylococcus Aureus in solution and in the fiber matrix. High antimicrobial inhibitions were observed for the Ge complexes followed by the ZnPc derivatives. All the low symmetry ZnPc derivatives were conjugated to AgNPs and their antimicrobial behaviour was compared to their symmetrical counterparts. The best antimicrobial inhibition behaviour was observed for the low symmetry Pcs when compared to their symmetrical counterparts. In the absence and in the presence of AgNPs, axially ligated SiPc also showed better antimicrobial activity when it was compared to the unsubstituted ZnPc complex.

Efeitos dos tratamentos mecânico, químico e fotodinâmico na proliferação de células da granulação óssea humana sobre raízes dentárias / The effects of mechanical, chemical and photodynamic treatment on the proliferation of osseous granulation cells on dental roots

A técnica do enxerto de granulação óssea tem demonstrado bons resultados na recuperação do periodonto e na melhora dos parâmetros clínicos dos dentes com comprometimento periodontal. Pouco se sabe porém, a respeito de qual tipo de tratamento de superfície radicular se faz mais condizente com o emprego dessa técnica. O objetivo dessa pesquisa foi avaliar a proliferação de células de granulação óssea sobre fragmentos radiculares com os seguintes tratamentos de superfície: Controle - somente raspagem, EDTA, terapia fotodinâmica (PDT- laser InGaAIP - 30mW, 30s, 45J/cm², 660nm + azul de toluidina), e ácido cítrico com tetraciclina. Todos os grupos teste receberam previamente tratamento com raspagem e alisamento com 20 golpes de cureta. Células de granulação óssea foram cultivadas em quadruplicata sobre os fragmentos por um período de 24, 48 e 72 horas. Após esse período de cultivo os fragmentos foram fixados para análise em microscópio eletrônico de varredura (MEV). Cinco campos por fragmento foram usados para a visualização e contagem de células aderidas a superfície radicular (centro, campo superior direito e esquerdo e campo inferior direito e esquerdo). A análise da calibração do examinador foi feita através de uma combinação de testes estatísticos como erro casual de Dahlberg, erro sistemático e correlação de Pearson (p<0,05). A análise da amostra foi realizada através do ANOVA de medidas repetidas complementado por Tukey, com nível de significância de 5% (p<0,05). Os resultados demostraram diferenças estatisticamente significantes, quanto ao numero de células, para as superfícies tratadas com terapia fotodinâmica no período de 72 h (p<0,05). Através de nossos resultados concluímos que o tratamento radicular com terapia fotodinâmica favorece a proliferação de células de granulação óssea humanas in vitro. / The osseous granulation graft has been demonstrating good results on the periodontal healing, resulting the improvement of clinical periodontal parameters. There are very few knowledge about what kind of dental surface would be more proper for the application of this technique. The aim of this study was to evaluate the proliferation of osseous granulation cells on human root fragments treated by different techniques as scaling and root planning (control), citric acid plus tetracycline, EDTA and photodynamic therapy (PDT InGaAIP, 45J/cm², 30mW, 30s, 660nm, toluidine blue O). All test groups were previously treated which 20 curette strikes. Osseous granulation cells was culture in quadruplicate on these fragments for 24h, 48h and 72h. After that, all fragments were fixed and prepared for analysis in Scanning Electron Microscopy (SEM). Aiming to counting the cells adhered on the roots, we obtained electron micrographs of 5 areas (center, upper right and left field, lower right and left field). The examiner calibration was analyzed by Dahlberg Casual Error measurement, systematic error test and Pearson correlation test (p<0.05). Statistical analysis was performed by ANOVA, followed by Tukey test, with a 5% level of significance (p<0.05). There were significant differences in cell number after 72h culture in favor of PDT group (p<0,05). We can conclude that the surface treatment of roots which PDT favor the proliferation of osseous granulation cells in vitro.

Efeito citotóxico da Terapia Fotodinâmica associando Photogem e LED azul e vermelho em cultura de células normais /

Resumo: Para considerar a Terapia Fotodinâmica (PDT) como tratamento clínico da estomatite protética, é necessário conhecer tanto o potencial antifúngico, como efeito citotóxico desta terapia sobre células normais do indivíduo. Assim, o objetivo desse estudo in vitro foi avaliar a citotoxicidade da PDT antifúngica com o fotossensibilizador Photogem® associado ao LED azul e ao LED vermelho em cultura de fibroblastos L929 e células odontoblastóides MDPC-23. As células foram cultivadas (30.000 células/cm2) em placas de 24 compartimentos por 48 horas e ambos os tipos celulares foram incubados com Photogem® (0, 10, 25, 50, 100 ou 150 mg/L) e irradiados ou não pelo LED azul (460 ± 3 nm; 25,5 ou 37,5 J/cm2; 22 mW/cm2) ou LED vermelho (630 ± 3 nm; 70 ou 100 J/cm2; 25 mW/cm2) . O metabolismo celular foi determinado 0, 12 e 24 horas após a PDT utilizando o teste do metiltetrazolium (MTT), e a morfologia celular avaliada pela microscopia eletrônica de varredura (MEV). A técnica de citometria de fluxo foi utilizada para avaliar o tipo de morte celular (necrose ou apoptose) assim como estimar os níveis intracelulares das espécies reativas de oxigênio (EROs). Observou-se redução do metabolismo celular estatisticamente significante para todas as concentrações do Photogem® quando irradiadas em qualquer dose de luz, sendo essa redução de 90 a 97% tanto para as células L929 quanto MDPC-23 (ANOVA and Dunnet's post hoc tests; p<0.05). Essa redução da atividade mitocondrial não foi dependente da concentração do fotossensibilizador e nem da dose de luz empregada. Também, foi demonstrado que a atividade mitocondrial das células submetidas a PDT não foi recuperada após 12 ou 24 horas, caracterizando um dano irreversível. A presença do Photogem® e da luz isoladamente não alterou estatisticamente a atividade mitocondrial de ambas as linhagens. As células submetidas... Therefore, the purpose of this in vitro study was to evaluate the cytotoxicity of PDT with Photogem® associated to blue and red LED on L929 and MDPC-23 cell cultures. The cells (30.000 cells/cm2) were seeded in 24-well plates for 48 hours, incubated with Photogem® (10, 25, 50, 100 or 150 mg/L) and were irradiated or not with a blue LED source (460 ± 3 nm; 25.5 or 37.5 J/cm2; 22 mW/cm2) or with a red LED source (630 ± 3 nm; 75 or 100 J/cm2; 22 mW/cm2). Cell metabolism was evaluated by the MTT assay (ANOVA and Dunnet's post hoc tests; p<0.05) and cell morphology was examined by scanning electron microscopy. Flow cytometry was employed to analyze the type of PDT-induced cell death (necrosis or apoptosis) as well as to estimate intracellular production of reactive oxygen species (ROS). There was a statistically significant decrease of mitochondrial activity for all Photogem® concentrations associated to blue or red LED regardless irradiation time; this reduction ranged from 90 to 97% for both cell lines. This reduction, however, was not dependent on the photosensitizer concentration. It was also demonstrated that the mitochondrial activity of the cells submitted to PDT was not recovered after 12 or 24 hours, characterizing irreversible cell damage. PDT-treated cells presented an altered morphology with ill-defined limits. In both cell lines, there was a predominance of necrotic cell death when in contact with the photosensitizer. The presence of Photogem® and exposure to blue and red LED increased the intracellular levels of ROS in both L929 and MDPC- 23 cells in a dose-dependent manner. Orientador: Ana Cláudia Pavarina / Abstract: Este estudo avaliou longitudinalmente a expressão de genes de virulência de isolados clínicos de Candida albicans de pacientes com candidose oral tratados com Terapia Fotodinâmica Antimicrobiana (aPDT) ou Nistatina (NIS). Inicialmente, a especificidade de primers da literatura e novos primers desenhados para os genes de virulência de C. albicans ALS1, CAP1, CAT1, EFG1, HWP1, LIP3, PLB1, SAP1, SAP4, SOD1, SOD5 e ACT1 (gene de controle) foi avaliada através de análises in silico e in vitro. Para a análise in silico, foi realizada uma busca no Pubmed por artigos com sequências de primers que avaliaram a expressão gênica de C. albicans. Em seguida, a homologia dos primers foi analisada (BLAST e ClustalW2) assim como a presença de estruturas secundárias (Mfold). Novos primers foram desenhados (Beacon Designer™) a partir de sequências obtidas do "Candida Genome Database". Os primers foram sintetizados e testados in vitro pela técnica de PCR utilizando um painel de DNA genômico de diferentes espécies de Candida, com seus produtos visualizados em gel de agarose. Reações de qPCR foram realizadas para determinar a concentração ótima e a eficiência dos primers. First, specificity of primers from the literature and newly designed primers for C. albicans virulence genes ALS1, CAP1, CAT1, EFG1, HWP1, LIP3, PLB1, SAP1, SAP4, SOD1, SOD5 and ACT1 (control gene) was evaluated through in silico and in vitro analyzes. Duzentas e setenta ratas foram divididas em 3 grupos de 90 animais: (A) SHAM; (B) Ovariectomizadas; (C) Ovariectomizadas tratadas com reposição hormonal. Nos primeiros molares inferiores esquerdos, de todos os animais, a doença periodontal foi induzida por ligadura. Após 7 dias, esta foi removida e nestes dentes procedeu-se a raspagem e alisamento corono-radicular (RAR). A seguir foram dividos em subgrupos de acordo com os tratamentos locais: I (n = 90) - Irrigação com 1 ml de soro fisiológico (RAR); II (n = 90) - Irrigação com 1 ml de soro fisiológico associado a aplicação de laser de baixa intensidade (LLLT) e III (n = 90) - Irrigação com 1 ml de azul de toluidina-O e, após 1 minuto, aplicação de laser em baixa intensidade (PDT). Dez animais de cada subgrupo foram sacrificados aos 7, 15 e 30 dias. Os animais do grupo A apresentaram perda óssea (PO) significativamente maior (p<0,01) no tratamento com RAR (1.11±0.26; 0.84±0.47) comparado à PDT (0.70±0.30; 0.42±0.20) nos períodos de 7 e 15 dias respectivamente. Os espécimes tratados com LLLT aos 30 dias demonstraram PO significativamente menor (p<0,01) no grupo A (0.35±0.18) comparado aos grupos B (0.82±0.21) e C (0.83±0.19). No tratamento com PDT, observou-se PO significativamente menor (p<0,01) no grupo A (0.42±0.20) comparado aos grupos B (0.95±0.20) e C (0.81±0.32) aos 15 dias. No entanto, no grupo C aos 30 dias a PDT demonstrou PO (0.52±0.23) em nível próximo ao observado nos espécimes do grupo A, no mesmo período (0.50±0.26). Na análise entre períodos, no mesmo grupo e tratamento, observou-se no grupo A, que o tratamento I apresentou PO significativamente maior... / The aim of this study was to evaluate histological, histometrically and immunohistochemistry the influence of laser and photodynamic therapy (PDT) as an adjuvant treatment on the experimentally induced periodontitis in ovariectomy rats with or without replacement hormone. Two hundred and seventy females rats were divided into 3 groups of 90 animals. (A) Normal female rats; (B) Control - Ovarectomy; (C) Ovarectomy treated with replacement hormone. The periodontal disease was induced by ligation and after 7 days was removed and the animals divided into subgroups who received the treatments: I - scaling and root planing (SRP) and irrigation with saline; II – SRP and irrigation with saline solution and LLLT (Laser -AsGaAl - 685 nm); III - SRP, irrigation with 1ml of Toluidine Blue (TBO) and, after 1 minute, application of laser (685 nm), performing photodynamic therapy (PDT). Ten animals of each subgroup were sacrificed at 7, 15 and 30 days. The animals of the group A presented bony loss (BL) significantly larger (p <0,01) in the treatment with RAR (1.11±0.26; 0.84±0.47) compared to PDT (0.70±0.30; 0.42±0.20) at 7 and 15 days respectively. In the specimens treated with LLLT at 30 days showed BL significantly less (p <0,01) in the group A (0.35±0.18) compared at groups B (0.82±0.21) and C (0.83±0.19). In the treatment with PDT, it was observed BL significantly less (p <0,01) in the group A (0.42±0.20) than groups B (0.95±0.20) and C (0.81±0.32) at 15 days. However, in the group C at 30 days PDT showed BL (0.52±0.23) in close level that observed in the specimens of the group A, in the same period (0.50±0.26). In the analysis among periods, in the same group and treatment, it was observed in the group A, that the treatment I presented BL significantly larger (p <0,01) at 7 days (1.11±0.26); that the treatment II showed larger BL (p <0,01) in the period of 7 days (0.90±0.29)... Ação da terapia fotodinâmica no tratamento da doença periodontal induzida: estudo histológico em ratos

Realizou-se a adaptação de uma ligadura de algodão na margem gengival do primeiro molar inferior esquerdo de cento e vinte ratos. Os animais foram divididos aleatoriamente em quatro grupos experimentais(n=30): Grupo I: nenhum tipo de tratamento realizado, somente a presença da ligadura; Grupo II (MB): tratamento com aplicação tópica de azul de metileno (100 æg/mL) dois dias após adaptação da ligadura; Grupo III (LLT): tratamento com Laser em baixa intensidade dois dias após adaptação da ligadura; e Grupo IV (PDT): tratamento com aplicação tópica de azul de metileno (100 æg/mL) seguido da aplicação do Laser em baixa intensidade dois dias após adaptação da ligadura. A superfície mesial dos dentes no Grupo III e IV foram irradiados com o Laser de diodo de GaAlAs, com 685 nm (50 mW, 120 s, 4.5 J/cm2). Todos os grupos foram subdivididos em subgrupos (n=10) e sacrificados nos períodos de 5, 15 e 30 dias pós-operatório. As peças foram processadas para análise histológica e coradas pela técnica de H&E. Através de um sistema de escores, vários parâmetros foram utilizados para avaliar o tecido conjuntivo, ligamento periodontal, superfície radicular e tecido ósseo. A análise estatística foi realizada pelo teste de Kruskal-Wallis, (p<0.05). Os resultados indicaram diferença no nível da inserção do ligamento periodontal entre o Grupo I (C) e Grupo III (LLT) (p=0,0327), Grupo II (MB) e Grupo III (LLT) (p=0,0118) e entre o Grupo III (LLT) e Grupo IV (PDT) (p=0,0076) no período de 15 dias de pó-operatório. Não ocorreu diferença estatisticamente significante... / The purpose of this study was to histologically evaluate the effect of photodynamic therapy (PDT) on the progression of experimentally induced periodontal disease in rats. Cotton ligatures were placed at the gingival margin level of the lower left first molar in one hundred and twenty rats. The animals were then randomly divided evenly into 4 groups (n=30): Group I (C): no treatment; Group II (MB): treated with topical application of methylene blue (100æg/ml) 2 days after ligature placement; Group III (LLT): treated by low level laser therapy 2 days after ligature placement; and Group IV (PDT): treated with topical application of methylene blue (100æg/ml) followed by low level laser therapy 2 days after ligature placement. The mesial aspect of the teeth in Groups III and IV were irradiated using a 685nm GaAlAs laser (50mW, 120s, 4.5J/cm2). All groups were subdivided into subgroups (n=10) and euthanized at either 5, 15 or 30 days post-operative. Block sections were routinely processed for histological analysis using H&E stain. Using a scoring system, several biological parameters were used to evaluate the connective tissue, periodontal ligament, root surface and bone. The data were statistically analyzed (Kruskal-Wallis, p<0.05).
