Quantification of Two Isomeric Flavones in Rat Colon Tissue Using Reverse Phase High Performance Liquid Chromatography

Background: Antineoplastic activity has been previously shown for two isomeric flavones, 5,7-dihydroxy-3,6,8-trimethoxy flavone (flavone A) and 3,5-dihydroxy-6,7,8-trimethoxy flavone (flavone B), against colon cancer cell lines (Thomas et al. in PLoS ONE 7:e39806, 5). Here, we present modified methods for the extraction and quantification of flavones A and B in rat colon tissue after intravenous dosing via high performance liquid chromatography, from the originally described procedure for extraction and quantification in rat plasma (Whitted et al. in J Chromatogr B Analyt Technol Biomed Life Sci 1001:150-155, 7). Results: Modifications included tissue homogenization (1 g tissue: 2 mL water), filtration of the supernatant with a PVDF membrane, and the use of only one calibration curve to determine the concentration of each flavone in colon tissue. Good separation was achieved and representative equations were linear with r 2 ≥ 0.99 for both flavones. Precision and accuracy for flavone A ranged from 0.88-24.03 and 109-116%. Precision and accuracy for flavone B ranged from 1.62-33.56 and 98-113%. Concentrations of 1639 ± 601 ng/g flavone A and 5975 ± 2480 ng/g of flavone B were detected in rat colon tissue 6 h post dosing. Conclusions: Modifications to the extraction methods for flavone A and flavone B from rat colon tissue had good separation, precision, and accuracy.

Development of Reversed-Phase High Performance Liquid Chromatography Methods for Quantification of Two Isomeric Flavones and the Application of the Methods to Pharmacokinetic Studies in Rats

Isomers 5,7-dihydroxy-3,6,8-trimethoxy-2-phenyl-4H-chromen-4-one (5,7-dihydroxy-3,6,8 trimethoxy flavone) (flavone A) and 3,5-dihydroxy-6,7,8-trimethoxy-2-phenyl-4H-chromen-4-one (3,5-dihydroxy-6,7,8-trimethoxy flavone) (flavone B) have recently demonstrated differential antineoplastic activities against pancreatic cancer in vitro. These studies also indicated that these compounds target highly tumorigenic cells while sparing normal cells. The in vivo antitumor activities of these flavones have not been determined, and detection protocols for these compounds are needed to conduct pre-clinical assays following intravenous dosing. Here, we report methods developed using acetonitrile to extract two flavone isomers and corresponding internal standards, celecoxib and diclofenac, from rat plasma. Separation was achieved using a Shimadzu liquid chromatography system with a C18 column and mobile phase acetonitrile/water (60:40 and 70:30 for flavones A and B, respectively) containing 0.2% acetic acid and 0.05% triethylamine at a flow rate of 0.4mL/min and detection at 245nm. Calibration curves ranging from 250 to 2500ng/mL and 2500 to 100,000ng/mL for both flavones were linear (r2≥0.99) with the lower limits of quantification being 250ng/mL. Recovery of concentrations 250, 1000, 2500, 5000, and 100,000ng/mL ranged from 87 to 116% and 84 to 103% (n=3) for flavone A and B, respectively. Stability of both flavones after a freezing/thawing cycle yielded a mean peak ratio ≥0.92 when compared to freshly extracted samples. Intravenous administration of a 20mg/kg dose in rats yielded half-lives of 83.68±56.61 and 107.45±53.31min with clearance values of 12.99±13.78 and 80.79±35.06mL/min/kg for flavones A and B, respectively.

Tillsammans räddar vi liv? : En studie om HLR i skolan årskurs 7-9 / Together we save lives? : A study about CPR in school grades 7-9

Syfte och frågeställningar: Syftet med studien är att undersöka hur undervisning i hjärt-lungräddning kommer till uttryck i ämnet idrott och hälsa. För att uppnå studiens syfte kommer följande frågeställningar att besvaras: Hur tolkar lärare i ämnet idrott och hälsa styrdokumenten vad gäller HLR i årskurs 7-9? Hur undervisar lärare i HLR i årskurs 7-9? Vilka förutsättningar har lärare i idrott och hälsa att bedriva undervisning i HLR i årskurs 7-9?  Metod: Studiens metod är en kvalitativ ansats, varpå datainsamling har genomförts med semistrukturerade intervjuer. De sju lärare som deltar i studien har valts ut genom två olika urval, målstyrt urval och bekvämlighetsurval. Intervjuerna genomfördes digitalt via Zoom, Meet och Teams. Empirin transkriberades digitalt samt genom manuell hantering, och analyserades med hjälp av en tematisk analys. Studiens teoretiska ramverk är läroplansteori och ramfaktorteori. Resultat: Lärarna anser att läroplanen genom centralt innehåll är undervisningens kodex. HLR är ett explicit begrepp i centralt innehåll men saknar definition vilket lämnar utrymme för lärarna att göra sina tolkningar av begreppet. HLR sätts i relation till simning och anses vara ett viktigt moment i undervisningen. Undervisningen i HLR kommer till uttryck i varierande omfattning. Momentet undervisas såväl teoretiskt som praktiskt, där video är en återkommande metod i undervisningen. Eleverna ges möjlighet att öva praktiskt med hjälp av övningsdockor. Praktisk träning av bröstkompressioner ingår i samtliga lärares undervisning av HLR, medan inblåsningar är ett teoretiskt moment. Lärarnas förutsättningar ser olika ut där ramfaktorer som exempelvis kunskap, utrustning och tid påverkar undervisningen i HLR.  Slutsats: Lärarna tolkar HLR i det centrala innehållet sett från eget perspektiv och förmåga. Det är lärarnas kunskaper i och om HLR samt deras förutsättningar som är avgörande för hur HLR kommer till uttryck i undervisningen. Lärarna tolkar och navigerar styrdokumenten med hjälp av erfarenhet och kunskap, vilket innebär att undervisningen i HLR kommer till uttryck på olika sätt. Lärarna utmanas av en mängd olika ramfaktorer vilka har mer eller mindre inverkan på undervisningen. / Aim and research questions: The aim of the study is to investigate how teaching in cardiopulmonary resuscitation is expressed when teaching physical education and health. In order to achieve the aim of the study, the following questions will be answered: How do teachers in physical education and health interpret the curriculum regarding CPR in grades 7-9? How do teachers teach CPR in grades 7-9? What prerequisites do physical education and health teachers have to teach CPR in grades 7-9?  Method: The study's method is a qualitative approach, after which data has been collected with semi-structured interviews. The seven teachers participating in the study have been selected through two different samplings, goal-directed sampling and convenience sampling. The interviews were conducted digitally via Zoom, Meet and Teams. Data was transcribed digitally as well as through manual handling, and analyzed using a thematic analysis. The study's theoretical framework is curriculum theory and frame factor theory. Results: The teachers believe that the curriculum, through central content, is the teaching code. CPR is an explicit expression of the concept in central content but lacks a definition, which leaves room for the teachers to make their own interpretations of the concept. CPR is put in relation to swimming and is considered an important part of teaching. Teaching in CPR is expressed to varying extents. CPR is taught both theoretically and practically, where video is a recurring method in the teaching. The students are given the opportunity to practice practically with the help of a practice dummy. Practical training of chest compressions is included in all teachers' teaching of CPR, while inhalations are a theoretical part. The teachers' conditions look different where frame factors such as knowledge, equipment and time affect the teaching of CPR. Conclusions: The teachers interpret CPR in the central content from their own perspective and ability. It is the teachers' knowledge of and about CPR as well as their prerequisites that are decisive for how CPR is expressed in teaching. The teacher’s interpret and navigate the curriculum with help of experience and knowledge, which means that the teaching of CPR is expressed in different ways. Simulating the FTICR-MS Signal of a Decaying Beryllium-7 Ion Plasma in a 2D Electrostatic PIC Code

Beryllium-7 (Be-7) only decays by electron capture into lithium-7 (Li-7) with a half life of 53 days. We study the effect of ionization on this decay rate. We do so by trapping a Be-7 ion plasma in a cylindrical Malmberg-Penning trap and measuring Be-7 and Li-7 concentrations as functions of time by using Fourier transform ion cyclotron resonance mass spectrometry (FTICR-MS). We have simulated these signals in a 2-dimensional electrostatic particle-in-cell (PIC) code. The two spectrum peaks merge at high ion densities whereas at low ion densities they can be resolved. The merged peak shifts linearly according to the relative abundances of these species. We have also simulated singly-ionized beryllium-7 hydride (BeH+) and Li-7 ion plasmas at high densities. These two separate peaks shift according to their relative abundances. We describe an analytical model that explains how these peaks shift.

On reliability-based design of rock tunnel support

Tunneling involves large uncertainties. Since 2009, design of rock tunnels in European countries should be performed in accordance with the Eurocodes. The main principle in the Eurocodes is that it must be shown in all design situations that no relevant limit state is exceeded. This can be achieved with a number of different methods, where the most common one is design by calculation. To account for uncertainties in design, the Eurocode states that design by calculation should primarily be performed using limit state design methods, i.e. the partial factor method or reliability-based methods. The basic principle of the former is that it shall be assured that a structure’s resisting capacity is larger than the load acting on the structure, with high enough probability. Even if this might seem straightforward, the practical application of limit state design to rock tunnel support has only been studied to a limited extent. The aim of this licentiate thesis is to provide a review of the practical applicability of using reliability-based methods and the partial factor method in design of rock tunnel support. The review and the following discussion are based on findings from the cases studied in the appended papers. The discussion focuses on the challenges of applying fixed partial factors, as suggested by Eurocode, in design of rock tunnel support and some of the practical difficulties the engineer is faced with when applying reliability-based methods to design rock tunnel support. The main conclusions are that the partial factor method (as defined in Eurocode) is not suitable to use in design of rock tunnel support, but that reliability-based methods have the potential to account for uncertainties present in design, especially when used within the framework of the observational method. However, gathering of data for statistical quantification of input variables along with clarification of the necessary reliability levels and definition of “failure” are needed. / <p>QC 20170407</p>

Modelling the national electricity system of Rwanda : Contribution to the achievement of Sustainable Development Goal 7

För att ett samhälle ska utvecklas på ett modernt och framgångsrikt sätt är elektricitet en grundläggande faktor. Tillgång till elektricitet förhöjer levnadsstandarden för befolkningen genom att generera positiva synergieffekter såsom att fler utbildar sig, ekonomisk tillväxt och stabilitet, förbättrad välfärd och internationella samarbeten. När den globala populationstillväxten och användningen av elektricitet ökar blir det av ännu större betydelse att andelen förnyelsebara bränslen i energimixen blir större för att inte påskynda den negativa klimatförändringen. Rwanda är ett litet, kustlöst land i Östafrika med ambitiösa planer om att förse sin befolkning, om 11.9 miljoner, med elektricitet år 2024 och att bli en ekonomi med låga koldioxidutsläpp. Idag har endast 41% av befolkningen tillgång till elektricitet, där 30% är uppkopplade till det nationella elnätet och 11% förses med elektricitet från fristående ellösningar eller mikronät. Rwanda arbetar med nationella policys såsom Vision 2020, Energy Sector Strategic Plan and National Energy policy för att uppnå sina nationella mål. Dessa strategier inkluderar också Förenta Nationernas globala mål och andra internationella åtaganden. Rwanda är också medlem i det gränsöverskridande samarbetet Eastern African Power Pool, vilket kommer förbättra deras framtida möjligheter för handel med elektricitet med sina grannländer. Denna studie syftar till att analysera den nationella elsektorn i Rwanda, både det nuvarande läget och framtida prognoser, för att bidra till landets arbete mot det Globala Målet 7 - Hållbar energi för alla. Scenarierna speglar Rwandas arbete med nationella policys, potentiell export samt hur energimixen påverkas när priset på fotovoltaik reduceras. Resultatet påvisar vattenkraftens betydande roll i Rwandas elsystem och hur den i majoriteten av scenarierna utgör den största andelen av energimixen. De exportscenarios som är kombinerade med nationella policys genererar en högre total systemkostnad medan de med enbart export antingen genererar motsvarande utfall som i grundscenariot eller aningen lägre total systemkostnad. I fyra av sju scenarier får befolkningen tillgång till elektricitet år 2024 och mål 7 uppnås. Baserat på denna studie behöver Rwanda begränsa sina koldioxidutsläpp, investera mer i förnyelsebara energikällor och expandera sitt nationella elnät samt förse befolkningen på landsbygden med fristående ellösningar eller mikronät för att nå mål 7. / For a society to function in a modern and prosperous way, electricity is a fundamental necessity. Access to electricity enhances the living standard of the population by generating positive synergies such as more people proceeding in education, economic growth and stability, improved welfare and collaborations with other nations. As the global population grows and the use of electricity increases, it is of great importance to increase the share of renewable energy resources in the energy mix to decelerate the climate change altered by greenhouse gas emissions. Rwanda is a small, landlocked country in East Africa with ambitious plans to electrify its full population of about 11.9 million by 2024 and to become a low-carbon economy. Today merely 41% of the population has access to basic electricity, where 30% are connected to the national grid and 11% are electrified by off-grid solutions. Rwanda is working with national strategies such as Vision 2020, Energy Sector Strategic Plan and National Energy policy to achieve their goals. These policies comprise, among others, United Nations’ Sustainable Development Goals and their Intended Nationally Determined Contributions. Rwanda is also a member of the Eastern African Power Pool, which enhances their future possibilities of expanded collaboration with neighbouring countries in terms of electricity trading. This study aims to analyse the national electricity sector of Rwanda, both the current state and future outlooks, to contribute to their achievement of the Sustainable Development Goal 7 - Affordable and Clean Energy for All (SDG 7). The overall aim of the study is to find an energy mix which benefit Rwanda’s society, environment and economy. The objective of the thesis is to collect data about Rwanda’s electricity system and national policies through a field study in Kigali. The collected data is then compiled, and seven scenarios are modelled by using the tool OSeMOSYS through MoManI. The scenarios portray Rwanda’s work with national policies, potential exports and how the energy mix is affected when the price for photovoltaic decreases. The results show the importance of hydropower in Rwanda’s electrification system and how it in the majority of the scenarios constitutes for the largest share in the energy mix. The exports scenarios combined with national policies have a higher total system cost while scenarios with merely exports have either the same total system cost as the business as usual scenario or a bit lower. In four out of seven scenarios electricity access for all inhabitants is ensured by 2024 and SDG 7 is achieved. Based on this study, Rwanda needs to limit their national emissions, invest in more renewables and expand the national grid as well as providing rural areas with off-grid solutions to achieve SDG 7.

Processing activity of the miRNA maturation endonucleases Drosha and Dicer toward let-7 substrates

La famille des microARN (miARN) let-7 comprend treize membres qui jouent des rôles critiques dans de nombreux processus biologiques, notamment la différenciation et le développement cellulaires. Plus spécifiquement, ils fonctionnent comme des suppresseurs de tumeurs en ciblant plusieurs oncogènes. La dérégulation des niveaux de miARN let-7 a été associée à diverses maladies humaines, y compris des cancers et des troubles neurodégénératifs. Il est bien établi que Drosha et Dicer, appartenant à la famille des RNases III, sont deux enzymes clés de la voie de maturation des miARN, et qu'un traitement défectueux par ces endoribonucléases pourrait affecter l'expression des gènes. Au cours des dix dernières années, plusieurs recherches ont permis d'identifier les caractéristiques structurales de l'ARN et les protéines qui régulent la voie de maturation des miARN. Cependant, les détails moléculaires menant à la régulation des niveaux d’expression des miARN nécessitent des investigations supplémentaires. L'objectif principal de cette thèse est d'étudier l'activité de clivage in vitro des endoribonucléases Drosha et Dicer envers leurs substrats let-7, en se concentrant sur la façon dont diverses caractéristiques de séquence et de structure affectent leurs activités. Tout d'abord, un criblage structural de type SHAPE suivi d'investigations thermodynamiques et cinétiques détaillées pour les treize pré-miARN de la famille let-7 ont été réalisés avec une enzyme Dicer purifiée in vitro. Cette étude a révélé que malgré les différences structurales des membres de la famille let-7, Dicer ne discrimine pas entre ses substrats, y compris les pré-miARN avec une extension de 1-nt et 2-nt à leur extrémité 3'. L'ensemble de ces travaux met en évidence la remarquable promiscuité de Dicer vis-à-vis divers pré-miARN de la famille let-7. Deuxièmement, le mécanisme enzymatique du clivage du pré-let-7a-1 a été examiné. Les résultats de la cinétique de l'état stable, de l'état pré-stable et de l'impulsion-chase sont conformes à l'opinion dominante, soutenue par de récentes structures de cryo-EM, selon laquelle le ou les changements de conformation d'un complexe enzyme-substrat dans une conformation catalytiquement productive sont importants pour l'activité de clivage. Troisièmement, nous avons étudié la séquence et les déterminants structuraux du clivage du pri-let-7 par le complexe microprocesseur (MP) composé de Drosha et de son partenaire obligatoire DGCR8. Sur la base d'études de clivage de plusieurs substrats pri-let-7 avec un complexe MP reconstitué in vitro, il a été constaté que le clivage du pri-let-7g donne des produits multiples. En utilisant des variantes de pri-let-7g, il a été révélé qu'un élément structural conservé de pri-let-7g favorise un clivage improductif, peut-être en raison du clivage de son substrat par la MP dans l'orientation inverse. Cette étude fournit un cadre pour des investigations futures dans l'étude du clivage de pri-let-7g par Drosha et éventuellement l'identification de nouveaux mécanismes de régulation. Dans l'ensemble, nos résultats donnent un aperçu de la façon dont les caractéristiques structurales des pri-miARN et des pré-miARN de la famille let-7 modulent le traitement par Drosha et Dicer et ouvrent la voie à de futures études visant à examiner le rôle des facteurs protéiques dans la régulation de la maturation des miARN let-7. / The let-7 family of microRNAs (miRNAs) comprises of thirteen members that play critical roles in many biological processes, including cell differentiation and development. More specifically, they function as tumor suppressors by targeting several oncogenes. Deregulation in let-7 miRNA levels has been associated with various human diseases, including cancers and neurodegenerative disorders. It is well established that Drosha and Dicer are the two key enzymes of the miRNA maturation pathway, and that faulty processing by these endoribonucleases could affect gene silencing. Thus, it is crucial to better understand how Drosha and Dicer respectively process the primary miRNAs (pri-miRNAs) and precursor miRNAs (pre-miRNAs) to yield mature miRNAs, and how these enzymes are regulated. In the last decade of miRNA research, several investigations have identified RNA structural features and RNA-binding proteins that regulate the miRNA maturation pathway, adding another layer of regulation in this pathway. However, the molecular detail of this regulation requires further investigations. The main goal of this thesis is to investigate the in vitro processing activity of Drosha and Dicer toward their let-7 substrates, focusing on how diverse sequence and structural features affect their activities. First, SHAPE structural probing followed by detailed thermodynamic and kinetic investigations for all thirteen pre-miRNAs of the let-7 family were performed with in vitro purified Dicer. Surprisingly, this study revealed that despite structural differences in the pre-let-7 members, Dicer does not discriminate between these substrates, including pre-miRNAs with a 1 nt and a 2-nt overhang at their 3'-end. Additional binding and cleavage investigations of pre let-7 substrates carrying 3'-end modifications (mono- and oligo-uridylation, mono- and oligo-adenylation) were performed to clarify how these modifications affect Dicer binding and cleavage activities. Together, this work highlights the remarkable substrate promiscuity of Dicer toward diverse pre-miRNAs of the let-7 family. Second, the enzymatic mechanism of pre-let-7 cleavage by Dicer was examined using pre-let-7a-1 as a model substrate. The results from the steady-state, pre-steady state and pulse-chase kinetics are consistent with the prevailing view, supported by recent cryo-EM structures, that the conformational change(s) of an enzyme-substrate complex into a catalytically productive conformation are important for cleavage activity. Third, the sequence and structural determinants of pri-let-7 processing by the Microprocessor (MP) complex composed of Drosha and its obligatory partner DGCR8 were investigated. Based on cleavage studies of several pri-let-7 substrates with an in vitro reconstituted MP complex, it was found that cleavage of pri-let-7g yields multiple products. Using pri-let-7g variants, it was revealed that a conserved structural element of pri-let-7g promotes unproductive cleavage, possibly as a result of the MP cleaving its substrate in the reverse orientation. This study provides the framework for future investigations in studying pri-let-7g processing by Drosha and possibly identifying novel mechanisms of regulation. Overall, our findings provide insights on how the structural features of pri-miRNAs and pre-miRNAs of the let-7 family modulate processing by Drosha and Dicer and pave the way for future studies aimed at examining the role of protein factors in regulating the maturation of let-7 miRNAs.

Developing densified products to reduce transportation costs and improve the quality of rice straw feedstocks for cattle feeding

Densification of rice straw such as compacting and pelletizing is an important process to increase the density of rice straw resulting in a reduction of transportation cost. Within this research, we conducted a techno-economic investigation of rice straw densification to produce compressed bales and pellets, which are later used for animal feed. In line with the main deliverable of the performance of rice straw compacting and pelletizing processes, we also looked into the quality of the product in terms of uptake and digestibility of the cattle feed which could be improved by adding amendments such as urea during the compacting process. The compacting technology resulted in a 400% increase of bale density (from 94 to 390 kg.m-3). This could reduce transportation costs by about 60% for a 60 km driving distance using trucks. The net profit that resulted from compacted bales was USD 0.0062 kg-1. Although the pelletizing technology increased the cost of the densified product by 40–50%, its density increases by 700%, from 94 to 666 kg.m-3. The enriched-rice straw pellets contained 12.1% protein, 2.8% lipid, 32.7% raw fibre, and 11.2% ash. In addition, a test of this product for cattle feeding illustrated an increase in its eating desirability for cows. Findings from this study contribute to reducing feedstock cost and developing densified rice straw products. These, therefore, provide more alternative options to increase the benefits from rice production and thus, reduce the unsustainable burning of rice straw in the field. / Nén ép rơm là quá trình quan trọng và cần thiết để tăng khối lượng thể tích với mục đích giảm chi phí vận chuyển. Kết quả nén ép cuộn rơm đã làm tăng dung trọng của kiện rơm đến 400% (từ 94 đến 398.7 kg.m-3). Qua đó, đã giảm được chi phí vận chuyển 60% được ước tính cho 60 km khoảng cách vận chuyển. Đối với ép viên rơm, dung trọng tăng đến 700% (từ 94 đến 666 kg.m-3). Hàm lượng dinh dưỡng của viên nén hỗn hợp rơm gồm protein (12,1%), chất béo (2,8%), chất xơ (32,7%), và lượng tro tổng (11,2%). Ngoài ra, kết quả thử nghiệm cho bò ăn sản phẩm viên nén này cho thấy bò có cảm giác thích và ăn hết toàn bộ viên nén. Kết quả của nghiên cứu đã góp phần giảm chi phí khi sản xuất thức ăn cho bò, tăng thu nhập cho người nông dân và từ đó giảm tác động môi trường do việc đốt rơm trên đồng.

Effect of phytase on the water quality of Vietnamese pangasius ponds

Striped catfish (Pangasianodon hypophthalmus) is freshwater fish that is raising mainly in the Mekong Delta. The research was implemented at the Model Farm (College of Aquaculture and Fisheries), Can Tho University during 6 months from May to November of 2016 and aimed to evaluate the water quality between two treatments of feed in the pangasius production for proposing a better environmental management method. The treatment 1 (control treatment, CT) has two ponds used feed without adding phytase and treatment 2 (experiment treatment, ET) includes two ponds used feed with adding phytase. The results showed that the temperature, pH, DO, TSS, COD, BOD, N-NO2-, N-NO3-, TKN, TAN were not significantly different (p>0.05) between the CT and ET ponds. At the experiment pond (EP), the factors (TP: 2.28 mg/L; P-PO4 3-: 1.24 mg/L) which has P release to the environment are less than control pond (CP) (TP: 2.62 mg/L; P-PO4 3-: 2.13 mg/L). These results suggested that the addition of phytase in feed could contribute to reducing the nutrient to the water body of pangasius pond. / Cá tra (Pangasianodon hypophthalmus) là loài thủy sản nước ngọt được nuôi chủ yếu ở các tỉnh Đồng bằng sông Cửu Long. Nghiên cứu được thực hiện trong 6 tháng từ tháng 05 – 11/2016 tại Trang trại mẫu đặt tại Khoa Thủy sản, Trường Đại học Cần Thơ nhằm đánh giá chất lượng nước giữa 2 nghiệm thức thức ăn trong giai đoạn nuôi cá thịt để đề xuất biện pháp quản lý chất lượng nước ao nuôi tốt hơn. Nghiệm thức 1 (nghiệm thức đối chứng, CT) gồm 2 ao nuôi sử dụng thức ăn không bổ sung enzyme phytase, nghiệm thức 2 (nghiệm thức thí nghiệm, ET) gồm 2 ao nuôi sử dụng thức ăn có bổ sung enzyme phytase. Kết quả thí nghiệm cho thấy giá trị nhiệt độ, pH, DO, TSS, COD, BOD, H2S, N-NO2-, N-NO3-, TKN, TAN ở mức phù hợp cho ao đối chứng (CT). Tại ao thí nghiệm (ET) (TP:2,28 mg/L; P-PO4 3-: 1,24 mg/L) lượng P thải ra ít hơn so với ao CT (TP: 2,62 mg/L; P-PO4 3-: 2,13 mg/L) chứng tỏ việc bổ sung enzyme phytase trong thức ăn đã góp phần làm giảm lượng lân thải ra môi trường nước trong nuôi cá tra.

Anaerobic co-digestion cow dung and corn stalk - effect of corn stalk pre-treated timing

The study was aimed to investigate the effect of corn stalk pre-treatment duration on biogas production when cow dung and corn stalk was co-digested in an anaerobic digestion. Corn stalks were pre-treated in different durations: 2-days, 5-days, and 8-days before being added to cow dung into anaerobic co-digesters. The experiments were set up randomly by using triplicate batch anaerobic apparatus in 21 L containers that run in 60-days. The mixing ratio between a corn stalk and cow dung was 50%: 50% (based on the volatile solid value of each material), but corn stalk was cut into small pieces with around 10 cm length, while the cow dung was air dried. The results of the study indicated that all operation parameters such as temperature, pH, and alkalinity in the anaerobic batch were suitable for biogas production. The results showed that there was a significant improvement in total gas produced in the pre-treated 5-days treatment (206.4±8.4 L) compared to 2-days (153.4±9.6 L), and 8-days ones (174±11.1 L). The biogas yield of the pretreated 2-days, 5-days, and 8-days treatments were 392.7±9.8 L/kg VSfermented, 469.8±10.1 L/kg VSfermented and 497.1±13.3 L/kg VSfermented, respectively, that was not significantly different (5%). In all treatments, low concentration of methane in the beginning phase had been observed but increased and reached the optimum value for energy use after 10 days. The result of the study showed that it is preferable to have 5-days pre-treatment of corn stalk before the corn stalk is loaded to an anaerobic digester in combination with cow dung. / Nghiên cứu này nhằm đánh giá ảnh hưởng của thời gian xử lý thân cây bắp lên năng suất sinh khí khi ủ phối trộn phân bò và thân cây bắp trong điều kiện yếm khí. Ba mức thời gian xử lý thân cây bắp được chọn là 2 ngày, 5 ngày, và 8 ngày. Các thí nghiệm được bố trí ngẫu nhiên trong các bình ủ yếm khí theo mẻ 21 L, vận hành trong 60 ngày liên tiếp và có 3 lần lặp lại. Nguyên liệu ủ được phối trộn theo tỷ lệ 50% phân bò và 50% thân bắp, trong đó thân bắp được cắt nhỏ cỡ 10 cm. Kết quả thí nghiệm cho thấy tất cả các thông số pH, nhiệt độ, độ kiềm của mẻ ủ đều phù hợp để vận hành. Lượng khí sinh ra của các nghiệm thức xử lý ở 2 ngày, 5 ngày, 8 ngày được ghi nhận là 153,4±9,6 L, 206,4±8,4 L và 174±11,1 L; năng suất sinh khí của các nghiệm thức không khác biệt và đạt giá trị 392,7±9,8 L/kg VSphânhủy, 469,8±10,1 L/kg VSphânhủy và 497,1±13,3 L/kg VSphânhủy. Tất cả các nghiệm thức đều sản sinh lượng CH4 thấp ở giai đoạn đầu nhưng tăng dần theo thời gian ủ và đạt hiệu quả sử dụng sau 10 ngày ủ. Kết quả cho thấy có thể chọn mốc thời gian 5 ngày để xử lý thân cây bắp trước khi đưa vào hầm ủ biogas.
