Garantia de qualidade dos provedores de serviços de aplicativos (ASP), empregando os acordos dos níveis de serviços (SLA): uma pesquisa exploratória / Quality guarantee of Application Service Providers (ASP), employing Service Level Agreements (SLA): an exploratory research

A economia globalizada e os avanços tecnológicos propiciaram um novo modelo de Outsourcing de Tecnologia de Informação (TI): os provedores de serviços de aplicativos (ASP - Application Service Providers). As discussões a respeito de ASP intensificaram-se nos três últimos anos, com a proposta de comercialização do software como um serviço. Quanto maior a interdependência da área de TI com as metas de negócios das organizações, maior a ênfase dada à satisfação das expectativas do cliente quanto aos serviços prestados pelos provedores. No mercado, têm-se adotado os acordos dos níveis de serviços (SLA - Service Level Agreements) como mecanismo de controle das atividades de outsourcing. Porém, essa adoção é nova e imatura, e a literatura é escassa. Tanto os provedores quanto os cliente sentem a necessidade por estruturas de implantação semelhantes às de SLA. Essa carência é ressaltada no caso dos modelos ASP. Propondo-se a contribuir com a compressão do modelo ASP e de SLA para ASP, apresenta-se uma pesquisa exploratória das características desse modelo com SLA, de modelos de qualidade de serviços de outsourcing e finalmente de considerações que devem ser feitas para garantir a qualidade dos serviços ASP, empregando-se os SLA. / The global economy and technological advances propitiated a new model of Information Technology (IT) Outsourcing: the Application Service Providers - ASP. Discussions regarding ASP became more frequent in the last three years because of the new approach of selling software as a service. As the interdependency between the IT area and the business goals of organizations becomes greater, the emphasis on fulfilling the client\'s expectations regarding service rendered by providers becomes stronger. In the market, Service Level Agreements - SLA have been adopted as control mechanisms of outsourcing activities. However, this adoption is new and immature, and the literature on the matter is scarce. Providers and clients feel the need for implementation structures similar to SLA\'s. In lhe case of ASP models, this necessity is emphasized mainly due to the lack of conceptual information on the ASP model. In order to better comprehend lhe ASP and the SLA models for ASP, I am presenting an exploratory research about the characteristics of the new approach with SLA models, outsourcing models of service quality and considerations, which must be carried out to ensure quality of ASP services employing SLA.

Os venenos do gênero Bothrops induzem efeitos locais caracterizados por hemorragia, necrose, edema e dor intensa. Apesar da importância clínica do fenômeno de dor, os estudos sobre os mecanismos envolvidos na gênese deste fenômeno são ainda escassos. Além disso, não existem dados sobre a capacidade do antiveneno em neutralizar este fenômeno. Neste trabalho foi investigada, a capacidade dos venenos de Bothrops jararaca, Bothrops asper e da miotoxina III (Fosfolipase A2, variante Asp 49), uma toxina isolada do veneno de Bothrops asper, em induzir hiperalgesia em ratos, a mediação química deste fenômeno e a capacidade dos antivenenos em neutralizar esta ação dos venenos. A possível correlação entre a hiperalgesia e a resposta edematogênica causada pelos venenos ou miotoxina foi também avaliada. O limiar de dor foi determinado antes e em diferentes tempos após a administração dos venenos ou toxina, empregando o teste de pressão de pata de rato. Para o estudo da resposta edematogênica, o aumento do volume das patas posteriores foi determinado por pletismografia. Os venenos e a toxina, administrados por via intraplantar, nas doses de 5µg (VBj), 15µg (VBa) ou 10µg (MIII), induziram hiperalgesia e edema, com respostas máxima na 1a (VBj, MIII) ou 2a (VBa) hora, não sendo mais detectadas na 24a hora. Para o estudo da neutralização, foram utilizados o antiveneno botrópico produzido no Instituto Butantan e o antiveneno polivalente produzido no Instituto Clodomiro Picado da Costa Rica, administrados por via endovenosa, 15 min. ou imediatamente antes ou 15 min. após a injeção dos venenos. O AVIB, quando injetado 15 min. antes do VBj, foi capaz de reverter a hiperalgesia induzida pelo veneno. Em relação ao edema, esta inibição foi observada quando o antiveneno foi administrado 15 min. ou imediatamente antes do VBj. Por outro lado, o AVCP não interferiu com a dor e o edema acarretados pelo VBa. Quando o VBj e o VBa foram incubados, in vitro, por 30 min., a 370C com os AV correspondentes, a hiperalgesia e o edema foram abolidos. Estes resultados indicam que a incapacidade do AVCP, quando administrado in vivo, de bloquear a hiperalgesia e o edema induzidos pelo VBa, não é consequência da ausência de anticorpos específicos no antiveneno, uma vez que estes efeitos foram inibidos quando o veneno foi pré-incubado com o antiveneno. Para avaliação da mediação química da hiperalgesia e do edema, os animais foram submetidos a tratamentos com inibidores de síntese, antagonistas de receptores, anticorpos ou drogas depletoras destes mediadores. Os resultados mostraram que o Hoe-140, dexametasona e NDGA inibem a hiperalgesia induzida pelo VBa, enquanto que apenas a prometazina interferiu com o edema causado pelo veneno. A hiperalgesia induzida pela MIII foi revertida pelo tratamento com prometazina, metisergida, Hoe-140, dexametasona e por NDGA, enquanto que o edema foi inibido apenas por prometazina e dexametasona. Estes dados sugerem que: a) a MIII é um importante componente do veneno para a geração de hiperalgesia, b) a bradicinina e os derivados da lipoxigenase são mediadores da dor acarretada pelo VBa e pela MIII, c) histamina e serotonina participam também da hiperalgesia induzida pela miotoxina e d) a histamina é mediador do edema induzido pelo VBa e pela MIII. Com relação à hiperalgesia induzida pelo VBj, somente o tratamento com Hoe-140 diminuiu este fenômeno, indicando a participação da bradicinina. Por outro lado, este tratamento não foi capaz de interferir com o edema induzido por este veneno. Cabe ressaltar que TEIXEIRA et al. (1994) demonstraram a participação de eicosanóides e PAF na hiperalgesia induzida pelo VBj. Os dados em conjunto sugerem ainda, dissociação entre os fenômenos de dor e edema acarretados por ambos os venenos e pela miotoxina. / Bothrops venoms cause pronounced local tissue-damage characterized by hemorrhage, myonecrosis, edema and pain. Venom-induced pain has been poorly investigated, despite its clinical relevance. Furthermore, the ability of antivenom to neutralize hyperalgesia induced by these venoms is not known. In the present study the hyperalgesia and edema induced by Bothrops jararaca (BjV) and Bothrops asper (BaV) venom and by myotoxin III-MIII (Asp49- phospholipase A2), a toxin isolated from BaV, were investigated. The chemical mediators involved in these phenomena and the ability of the antivenom to neutralize the hyperalgesia and edema induced by these venoms were also investigated. Pain threshold was assessed before and at several intervals after venom injection, using the rat paw pressure test. Edema of paw was measured phethysmographically at the same periods of time. The intraplantar injection of BjV (5µg/paw), BaV (15µg/paw) or MIII (10µg/paw) caused hyperalgesia and edema, whose peak were observed at the 1st (BjV, MIII) or 2nd (BaV) hours after venom/toxin administration, decreasing thereafter. For neutralization studies, the antivenoms produced either at Instituto Butantan from Brazil (AVIB) or Instituto Clodomiro Picado from Costa Rica (AVCP) were administered intravenously 15 min prior to, or immediately before, or 15 min after venoms injection. When the antivenom from Instituto Butantan was injected 15 min. before BjV, the hyperalgesia and edema were abolished. Furthermore, partial inhibition of edema was also observed when the antivenom was injected together with BjV. On the other hand, hyperalgesia and edema induced by BaV were not modified by AVCP. Incubation of BjV and BaV, for 30 min. at 37oC, with the antivenoms in vitro, abolished the hyperalgesia and edema. The inability of the in vivo treatment with antivenom in abolishing hyperalgesia and edema induced by BaV seems not to be related to the lack of neutralizing antibodies in antivenom, because neutralization was achieved in pre-incubation experiments. In order to investigate the chemical mediation of hyperalgesia and edema induced by the venoms or toxin, animals were treated with several drugs. Pretreatment with Hoe-140, dexamethasone and NDGA blocked the hyperalgesia induced by BaV, whereas only promethazine reduced the edema induced by this venom. The MIII-induced hyperalgesia was blocked by promethazine, methysergide, Hoe-140, dexamethasone and NDGA, whereas the edema was reduced only by promethazine and dexamethasone. These results suggest that: a) MIII may contribute to the BaV-induced hyperalgesia, b) bradykinin and leukotrienes mediate the BaV- and MIII-induced pain and MIII; c) histamine and serotonin also participate in the myotoxin-induced hyperalgesia and d) the edema induced by BaV and MIII is mediated by histamine. Pre-treatment of the animals with Hoe-140 abolished BjV-induced hyperalgesia, suggesting that bradykinin may mediate the venom-induced hyperalgesia. However, this treatment did not modify the BjV-induced edema. It is important to stress that previous studies have shown that BjV-induced hyperalgesia is mediated, at least partially, by eicosanoids and PAF (TEIXEIRA et al.,1994). The data presented herein also suggest that distinct mechanisms may be involved in the development of hyperalgesia and edema induced by both venoms and myotoxin III.

Régulation de la transcription bidirectionnelle chez le Virus de l'Immunodéficience Humaine de type 1 / Bidirectional transcription regulation in Human Immunodeficiency Virus type 1

Le génome des rétrovirus existe sous deux formes différentes : sous forme d'ARN simple brin, qui est traduit ou encapsidé, ou sous forme d'ADN double brin intégré dans le génome de la cellule hôte infectée. Cette dernière forme, l'ADN proviral, est indispensable à la production de tous les ARNm viraux nécessaires à la synthèse des protéines virales, qui en retour agissent sur la région promotrice située au niveau du LTR 5'. Cependant, l'ADN proviral possède un second LTR à son extrémité 3', capable de réguler une transcription antisens, orientée dans la direction opposée à celle contrôlée par le LTR 5'. L'ADN proviral a donc deux brins codants, ce qui offre au virus un plus grand potentiel de synthèse protéique. Dans le cas du Virus de l'immunodéficience Humaine de type 1 (VIH-1), la transcription antisens permet la production d'une protéine, appelée ASP (Antisense Protein). Dans ce manuscrit, nous démontrons que cette activité transcriptionnelle antisens s'exprime préférentiellement dans les cellules d'origine monocytaire, en particulier les cellules dendritiques ; une localisation membranaire de la protéine ASP a par ailleurs été mise en évidence dans ce type cellulaire. Nos résultats suggèrent également que la transcription antisens du VIH-1 est indépendante de la protéine Tat, et que par ailleurs les deux types de transcriptions ne sont pas exprimés simultanément au sein d'une même cellule. En outre, nos données soulignent que la séquence codante de la protéine ASP est très fortement conservée parmi les différents isolats viraux. Sur la base de l'ensemble de ces résultats, notre hypothèse est que la protéine ASP du VIH-1 possède des fonctions cruciales dans le cycle réplicatif des rétrovirus, indépendantes de la production virale. / Genome of retroviruses exists in two different forms: as single-stranded RNA that is translated or packaged, or as double-stranded DNA integrated into the genome of the infected host cell. The latter form, the proviral DNA, is essential for the production of all viral mRNAs required for the synthesis of viral proteins, which in turn act on the promoter region located at the 5 '-LTR. However, the proviral DNA has a second LTR at its 3 '-end, capable of regulating antisense transcription oriented in the opposite direction to that controlled by the 5'-LTR. The proviral DNA has then two coding strands, which gives the virus a greater potential for protein synthesis. In the case of the Human Immunodeficiency Virus type 1 (HIV-1), antisense transcription allows the production of a protein called ASP (Antisense Protein). In this manuscript, we demonstrate that this antisense transcriptional activity is preferentially expressed in cells of the monocyte lineage, in particular dendritic cells; a membrane localization of the ASP protein was also observed in this cell type. Our results also suggest that the antisense transcription of HIV-1 is Tat-independent, and what's more that the two types of transcription are not expressed simultaneously within the same cell. In addition, our data highlight that the ASP protein coding sequence is highly conserved among different viral isolates. Based on these results, our hypothesis is that the ASP protein of HIV-1 has critical functions in the replicative cycle of retroviruses, distinct from viral production.

A proposta do modelo ASP é a comercialização de aplicativos de software como serviço. A satisfação dos clientes está diretamente relacionada à qualidade do serviço prestado. Para garantir que sejam fornecidos serviços com qualidade, é fundamental a adoção dos Acordos dos Níveis de Serviços (SLA - Service Levei Agreements) como mecanismo de controle. No entanto, são escassas as informações formalizadas que venham a contribuir com a especificação desses acordos. Problematika prostřihování otvoru dvouvrstvým polotovarem / Hole punching problematics of two layered sheet metal

The thesis is focussed on project of a tool – a hot working clipping punch, where the hole is punched in the upsetting ends of stabilizer. We talk about punching of a two layered semi-finished product, in which burr is formed at the layer interface, which is undesirable from the point of the customer requirement. The stabilizer is mounted in the car chassis, where it is fastened to the axle with screws. The literature study is focused on the theory and problems of punching. In the practical part, several design solutions of cutter faces are discussed, from which three types of face shape were selected. This was associated with a change in the design of the ejector. Two tool materials were selected for comparison. The cutters were tested on an automatic forming centre. Finally, after summarizing and evaluating all the practical experiments, an optimal solution of a tool designed for punching a hole with a two layered semi-finished product is proposed.

The adoption of Antimicrobial Stewardship Programmes in Ministry of Health hospitals in Saudi Arabia

Aim: This thesis aims to explore and investigate the level and process of adoption of Antimicrobial Stewardship Programmes (ASPs) and factors influencing their implementation in Saudi Ministry of Health (MOH) hospitals. The findings of this study will provide hospitals and policy makers with evidence-based recommendations on how barriers to ASPs adoption can be overcome, which will ultimately improve antimicrobial use and reduce antimicrobial resistance (AMR). Method: A mixed method approach was carried out using both qualitative and quantitative research methods. Semi-structured interviews were conducted with healthcare professionals in three Saudi hospitals to explore the enablers and barriers to their adoption of ASPs. A survey was then developed based on these findings to investigate the level of hospitals' adoption of ASPs and factors influencing their implementation at a national level. Further, a case study using in-depth interviews was utilised to understand the process of ASP adoption in a Saudi hospital, and how adoption challenges were addressed. Finally, a self-administered questionnaire was used to examine patients' knowledge and perceptions of antimicrobial use and resistance, and to evaluate the institutional role of patient education on antimicrobial use in two Saudi hospitals. The overall methodology of the research is summarised in Figure I. Results: Despite the introduction of a national ASP strategy, adoption of ASPs in Saudi MOH hospitals remains low. Organisational barriers such as the lack of senior management support, lack of supportive IT infrastructure and the shortage of ASP team members hinder hospitals' efforts to adopt ASPs. Further barriers relate to the lack of formal enforcement by MOH and the physicians fears of patients' complications and clinical liability. Patients admitted to Saudi hospitals lack knowledge and perceptions of AMR, and the adoption of ASPs may improve hospitals' role in patients' education. Conclusions: Despite the established benefits of ASPs, their adoption in Saudi MOH hospitals remains low. Urgent action is needed to address the strategies priorities associated with AMR, including access to antimicrobials, antimicrobial stewardship and education and research. Policy makers are urged to consider making ASPs adoption in hospitals a regulatory requirement supported by national guidelines and surveillance programmes. It is essential to increase the provision of ID and infection control residency and training programmes to meet the extreme shortage of ID physicians, pharmacists, microbiologists and infection control practitioners. Higher education institutions and teaching hospitals are required to introduce antimicrobial prescribing and stewardship competencies into undergraduate Medical, Pharmacy, Dental, Nursing and Veterinary curriculum, as well as introduction of AMR topics in order to increase knowledge and awareness of ASPs and AMR. Collaboration between ASPs adopting and non-adopting hospitals is essential to share implementation experience, strategies and solutions to overcome barriers. Healthcare specialised associations are needed to be part of AMR conversation and guide healthcare professionals' training and accreditation. Multiple stakeholders should be actively part of the conversations around tacking AMR. Primary care, secondary care, community pharmacies and policy makers should strive to create a shared culture of responsibility among all healthcare partners to improve antimicrobial therapy and reduce risks of AMR.

The Determinants of Customer Perceptions in a Dynamic Business Environment: An Exploratory Analysis of the ASP Business Model

Outsourcing attracted much attention in 1989 when Kodak outsourced its data center operation to IBM (International Business Machines Corp.). Nowadays, this strategy has become more popular. At the beginning of this century, the ASP (Application Service Provider) model was considered one of the typical solutions of Internet-based IT (Information Technology) outsourcing. Although this model has been transformed and renamed (e.g. SaaS - Software as a Service), the principle concept of providing IT service through the Internet or wide area network is still there. This study attempts to explore the determinants of customer perception of Internet-based IT outsourcing by obtaining a comprehensive understanding of the ASP model. The research dimensions not only include factors affecting users' perception of service quality but also ASP business position (i.e. the firm origin of ASP and its provider type) and services utilized by the customers. Through the study of firm history, two important theoretical themes of this research - path-dependence and Ansoff's product/ market growth matrix - are taken account of in exploring the influence of the determinants. Web-based questionnaire survey research is conducted together with a documentation study to collect data. Targeting the customers of the top 50 ASPs selected by ASPnews.com during the period 2001-2004, the researcher contacted 597 potential respondents, and 196 responses were returned. The valid sample consisted of 175 responses, and 124 of them not only provided full information for satisfaction evaluation but also the information for tracking their ASP vendors' business position. The GLM (General Linear Model) and the Pearson correlation coefficient were the major statistical approaches used to evaluate the survey data for developing a structural model. The research findings indicated that the factors associated with service competitiveness, such as capability and performance, reliability and trustworthiness, affordability, integration and customization, have positive effects on customer perceived satisfaction; whereas lock-in has a negative effect. More specifically, the origin of the ASP firm has a direct effect on capacity and performance, and also directly influences the use of IT adoption services. Based on this finding, a descriptive analysis and qualitative research shows that two mechanisms for path-dependence - existing expertise and perceived expertise - can affect the satisfaction level of capacity and performance of ASP services. On the other hand, provider type has a direct effect on affordability and also directly influences the use of facility supporting services. On this basis, another two mechanisms for path-dependence - transaction cost and standardization - can indirectly impact customer's perception of this business model via affordability. In addition to those major findings, some other determinants (e.g. software applications, brand of applications, and intensity of service used) were also identified in this study. The study result can be used for theoretical understanding about the determinants of ASP customer's perception. It not only indicates a new perspective to enhance the current body of research on this topic, but can also be more broadly applied to any fast-growth firm, rapid-change business, or technology intensive industry. Simulation et conception de services déportés sur grappes

Les grappes et grilles de machines connaissent, à l'heure actuelle, un succès grandissant. Les avantages offerts par ces supports d'exécution sont multiples : faible coût d'installation, facilité d'adaptation à la charge, facilité de portage des codes de calculs pré-existants ... De par la démocratisation de ces supports d'exécution, l'informatique évolue vers un modèle déporté où la puissance de traitement est fournie sous forme de services. Cette thèse étudie et propose des solutions à la problématique de mise en place de services déportés sur des grappes de machines. Les problématiques d'observation de l'état des machines, de haute disponibilité, de prise en compte de l'aspect dynamique d'une grappe de machines et de gestion d'accès personnalisés aux grappes de machines ont été étudiées. Le gestionnaire de ressources AROMA (scAlable ResOurces Manager and wAtcher), développé durant cette thèse permet d'apporter des réponses concrètes à ces problématiques. L'originalité de cet outils réside dans la prise en compte de l'aspect dynamique des grappes de machines, et dans la précision des informations d'état collectés sur les machines. La précision de ces informations permet notamment la prise de décisions de placement intégrant la notion de qualité de service. La problématique du dimensionnement des grappes de machines déportées est ensuite abordée. Le comportement des différents éléments ayant une influence sur les performances du système est étudié : les clients, les applications, les machines, les systèmes d'exploitation et la couche réseau. Différents modèles de simulations de ces éléments ont été intégrés au simulateur DHS (Distributed Hybride Simulator). Les résultats obtenus par ce simulateur sont enfin comparés à des modèles analytiques et à des mesures d'exécutions réelles afin de valider leur pertinence et d'évaluer les performances de la simulation. L'originalité de ce travail réside dans la simulation de l'ensemble des éléments ayant une influen ce sur les performances d'une grappe de machines. <p>Vi har i vår studie tittat på dynamisk webbprogrammering med fokus på Microsofts senaste teknik ASP.NET. Studien syftar till att ta reda på varför systemutvecklande organisationer infört ASP.NET, vilka förväntningar de har på tekniken och vilka utvecklingsverktyg som används. Vi har arbetat enligt ett hermeneutiskt synsätt för att få kvalitativ förståelse och ett helhetsperspektiv. Vi har valt semistrukturerade intervjuer för datainsamling och för analysen har vi valt en anpassad variant av Grundad teori. </p><p>Vårt resultat visar att några av anledningarna till införandet av ASP.NET hos organisationerna i studien beror dels på att de vill ligga långt fram i användandet av nya tekniker, dels upplever de att krav ställs på dem att de ska tillhandahålla och behärska de senaste utvecklingsteknologierna. Få uttalade förväntningar fanns på ASP.NET vid införandet och som utvecklingsverktyg används uteslutande Microsoft Visual Studio.NET som anses vara ett kraftfullt verktyg. Microsofts licensavtal och dominans på marknaden har också påverkat. </p><p>Några av slutsatserna blir att konkurrensfördelar kan erhållas genom en övergång till ASP.NET. Systemintegration blir enklare, vilket kan attrahera nya kundgrupper. Bättre möjligheter till struktur i programmeringen gör projekt lättare att planera och system lättare att underhålla. </p> / <p>In our research we have studied dynamical Web programming with the latest technology from Microsoft, ASP.NET. The purpose of the study is to find out why system developing organizations have introduced ASP.NET, what expectations of the technology they have and which development tools they use. We have worked according to a hermeneutical approach in order to get a qualitative understanding and a comprehensive view. We have chosen semi structured interviews for data acquisition and for analyze we have chosen an adapted variant of Grounded Theory. </p><p>Our result shows that some reasons to why the organizations in the study have introduced ASP.NET are that they will be in the frontline regarding the use of new technologies. They also feel that there is an expectation of them to use and know the latest of development technologies. There weren’t much explicit expectations of ASP.NET during the introduction. All of the organizations in the research used Microsoft Visual Studio.NET as development tool, because it is a powerful tool. The license agreement with Microsoft and its dominance on the market has also influenced them. </p><p>Some of our conclusions are that the organizations could have some competitive advantages if they introduce ASP.NET. System integration will be easier, which can attract new groups of customers. The possibilities to structure in the programming will make projects easier to plan and the maintaining of systems will be simpler.</p>

<p>This thesis will show you how to build and construct a interactive website with a web shop and a web based administration system. The website is developed for a company called Varett. Varett will use the website to reach out for a larger group of customers and also for promoting new products. I have develop this site mainly using ASP, MS Access and HTML. </p><p>I have also developed a graphic profile for the company to use at the homepage and at other commercial products. You can find the current version of the website at http://www.varett.se.</p> / <p>Detta examensarbete visar hur man bygger upp och konstruerar en interaktiv hemsida med en webbshop och ett webbaserat administrationssystem. Hemsidan är utvecklad åt ett företag som heter Varett. Varett var i behov av en hemsida för att kunna nå ut till en större kundgrupp och för att kunna göra reklam för nya produkter de själva utvecklat. </p><p>Jag har utvecklat hemsidan mestadels med hjälp av ASP, MS Access och HTML. Jaghar också tagit fram en grafisk profil åt företaget. Den grafiska profilen används inte bara på hemsidan utan också för reklamblad produktbroschyrer mm. Hemsidan finns just nu tillgänglig på http://www.varett.se.</p>
