Analysis of the IgE network : inhibition of CD23-mediated IgE upregulation and CD21/C3d interaction

Allergic reactions are mainly mediated by the interactions between the IgE and its ligands, amongst them CD23 and CD21 in what is termed the IgE network. CD23 is involved in upregulating IgE expression by forming a trimolecular complex with CD21 and IgE on the B-cell surface, resulting in the specific activation of IgE-positive B cells. CD21 also interacts with C3d and is a bridge between the innate and the immune system. A crystal structure of the interaction has been solved (Szakonyi et al., 2001) but was controversial because it contradicted previous biochemical analyses. The aims of this thesis were to use various biophysical techniques to study the interactions between the molecules in the IgE network and its possible inhibition. Part 1: Characterisation of a phage display-derived peptide that inhibit IgE binding to CD23 A peptide was previously derived using phage display technology and tested for binding ability to CD23 using SPR and ITC. Subsequent NMR experiments were performed to identify the binding site, followed by characterization of its derivatives. Crystallisation of CD23 with the peptide and soaking with its truncated tripeptide, NWP, were also attempted. Part 2: Characterisation of CD23 and its interaction with its ligands X-ray crystallography was undertaken to solve the structure of derCD23 in complex with a phage display-derived peptide (Part1) followed by crystal soaking with a truncated tripeptide, NWP. However, a reproducible, high-resolution wild type derCD23 structure was determined at 1.9 Å. A comparison of the binding behaviour between the monomeric derCD23 and a trimeric CD23 construct was carried out in order to see the effect of oligomerisation upon IgE binding. Using the known interaction map as well as a crystal structure, the possible interacting residues between CD23 and IgE were examined. The characterisation of the CD23/CD21 interaction was continued from previous efforts in order to confirm that the binding epitope of CD23 for CD21 lies within the C-terminus of CD23. Characterisation of the interactions of CD23/IgE/FcεRI was performed to examine these multimolecular interactions and possible regulatory mechanisms in mast cell degranulation. It was shown that CD23 can form multimeric complexes with IgE-Fc that bind to FcεRI with higher apparent affinity than IgE-Fc alone, which may lead to increases in mast cell degranulation. It was also found that the IgE bound on FcεRI still binds to CD23 although with a lower binding capacity, presumably due allosteric changes. The binding of CD23 with a monoclonal antibody IDEC-152 was also characterised using SPR and NMR spectroscopy. It was proposed that IDEC-152 might interfere with the trimerisation site of CD23 thus reducing its affinity for IgE. A thermofluor assay was developed and optimised for potential screening of compounds that bind to derCD23 using a qPCR machine, which may be useful to screen compounds that bind to CD23 as part of future drug discovery project. Crystallisation of the derCD23/CD21 and IgE/triCD23/CD21 complexes was also attempted as part of ongoing crystallisation projects. Part 3: The interaction between C3d and CD21 The interaction between C3d and CD21 is believed to be a bridge between the innate and adaptive immune response, and is thought to be pivotal in the initiation of autoimmune disease. Following from previous studies on this interaction, further characterisations were performed using NMR and ITC to confirm the involved sites on CD21 (SCR1-2) in binding to C3d. Several potential salt bridges have been identified so far, allowing a high-resolution docked structure of the C3d/CD21 complex.

Reports and cooperation on an "Asthma, eczema, diabetes, flu" project were used to detect the suffering children. Both, healthy and sick children cooperate to achieve set goals within the project. Lastly, the matter of this thesis also includes prevention - recommendation to parents, whose children suffer from the diseases mentioned above. Theoretical observations having reference to chronic diseases, structure of reports, methods of evaluating the effects, preparation of the project and recommendation to parents whose children suffer from chronic disease are applied to completed report examination, project implementation, and discussion with parents.

The increasing incidence of children suffering from allergic diseases could be caused by sensitization of immature immune system during the intrauterine development. Several important scientific papers have demonstrated the ability of cord blood cells to respond by elevated proliferation activity after stimulation by common allergens. Following these findings, present study follows the production of cytokines which play a role in the pro- and anti-allergenic tuning of the immune system. Umbilical cord blood cells were stimulated with polyclonal activators (phytohaemagglutinin) and common allergens (ovalbumin, timothy grass, birch, mite). Subsequently, cytokine production was monitored using selected methods that reflect different stages of cell activation - at the level of mRNA by quantitative real time PCR (qRT-PCR), by flow cytometry detection of the presence of intracellular cytokines in different cell subpopulations and by ELISA measurement of cytokines in CBMC culture supernatants. The results obtained point to a very weak ability of these common allergens (timothy grass, birch, mite, ovalbumin) to stimulate CBMC to produce cytokines observed by all of these methodological procedures. This diploma thesis was focused on the study of lysozyme and lysozyme-like proteins, either of similar function (antibacterial) or molecular weight (14 - 17 kDa), of synanthropic acaroid mites. In general, animals utilize lysozymes for defensive (antimicrobial) or digestive purposes but also as a digestive enzyme. Some chitinases or other enzymes that act similarly to lysozyme can be utilized for similar purposes. Chitinases belong to house dust mite allergens. One of major mite are historically named lysozyme-like proteins which name relates to their size similar to lysozyme. Bacteriolytic activity has also 14.5 kDa (UniprotKB Q8MWR6) protein. The species selected for the study were domestic mites Dermatophagoides farinae, D. pteronyssinus and Lepidoglyphus destructor. Presence of lysozyme was detected by direct detection with polyclonal antibody using immunohistochemistry and dot blots. Immunohistochemistry proved presence of lysozyme epitopes in the feces of D. farinae, D pteronyssinus a L. destructor. Dot blot analysis demonstrated the presence of imunoreactivity of antibody in spent growth medium extracts (SGME) of all three species. This implies that lysozyme is synthesized in the midgut. Title: Occurrence and Prevention of Asthma in Youth Division Cross-country Skiers Objectives: The aim of this thesis is to determine the incidence of asthma in youth division cross-country skiers and its prevention. Methods: This thesis will comprise the method of survey and content analysis of documents. Hypotheses: No.1 Assume that asthma will have lower prevalence in youth division cross- country skiers than in adult cross-country skiers. No.2 Assume that the majority of the surveyed cross-country skiing coaches met with asthmatic symptoms in their athletes. Results: We found that 12% of youth division athletes suffer from asthma and 24% from allergies. La régulation de l'homéostasie intestinale est de la plus haute importance en raison de la constante exposition de l'intestin aux antigènes alimentaires et à la flore commensale. La perturbation de la flore intestinale est souvent associée à diverses maladies telles que l'allergie, l'obésité et certaines maladies inflammatoires. La plupart des individus sont tolérant aux antigènes alimentaires et ne développe pas de réponse immunitaire sauf en cas de prédisposition génétique ou d'exposition à un environnement défavorable. La réponse allergique se caractérise par la production d'IgE stimulé par les lymphocytes Th2. Les symptômes allergiques sont très variés et affectent plusieurs parties de l'organisme. La plupart des travaux de recherche se sont focalisé jusqu'à présent sur le rôle des cellules de l'immunité adaptative dans le développement de l'allergie en sous-estimant le rôle majeur des cellules épithéliales et des cellules de l'immunité innée. L'objectif de ce projet est de comprendre comment les cellules épithéliales intestinales modulent la réponse immunitaire à distance vers la muqueuse pulmonaire ou la peau après stimulation allergique. L'ingestion de l'antigène associé à l'adjuvant de la toxine cholérique permet d'étudier la réponse allergique chez l'animal. Nous avons démontré sur ce modèle animal que l'absence de la kinase inhibitrice IKKb dans la voie de signalisation du facteur de transcription NF-kB altère la composition de la flore intestinal d'une part et transforme la réponse immunitaire inflammatoire au niveau pulmonaire et de la peau grâce à la présence d'IgA et de lymphocyte Th17 d'autre part. En adéquation avec les observations cliniques rapportées chez les patients allergiques (allergies alimentaires, asthme, dermatite atopique), nos résultats identifient IKKb dans la cellule épithéliale intestinale comme cible potentielle pour traiter les allergies alimentaires. De futurs efforts devront être faits pour développer de nouvelles stratégies thérapeutiques qui considèrent la muqueuse intestinale, la production d'IgA et l'importance des bactéries commensales dans le traitement des allergies. / Immune homeostasis is of paramount importance in the gastrointestinal tract, which is constantly exposed to ingested antigens and commensal microbiota. The gut microbiota can be perturbed by endogenous or exogenous factors and it is now established that microbial dysbiosis is associated with allergy, obesity, and inflammatory diseases. Ingestion of food antigens generally fails to promote brisk immune responses but rather results in a state of immune tolerance. However, aberrant immune responses can develop in individuals with a genetic predisposition. Food allergies are generally regarded as pathologic responses to food antigens mediated by excessive Th2 responses and antigen-specific IgE antibody responses. Clinical manifestations of food allergies are very broad and symptoms can affect different organs. While past research on allergy focused on the role of cells and molecules involved in adaptive immunity, epithelial cells lining the sites of antigen entry and innate immune responses have recently emerged as important players in allergy. This project was undertaken to understand the mechanisms employed by intestinal epithelial cells (IECs) to shape immune responses to allergens and influence allergic manifestations in distant mucosal sites such as the airways or the skin. Oral administration of food antigen with cholera toxin as adjuvant in experimental animals is a well-accepted model to study allergic sensitization to food antigens. Using this model, we show that a localized impairment of the canonical NF-κB pathway through deletion of IkB kinase (IKKβ) in IECs alters the gut microbiota during oral allergic sensitization and regulates the magnitude of allergic inflammatory responses at distant sites of the airway and the skin through enhancement of IgA Abs and Th17 responses. Consistent with the clinical observations linking atopic diseases (food allergy, allergic asthma, atopic dermatitis), our results identify IKKβ in IECs as a potential therapeutic target for treatment of food allergies and subsequent disease. They also suggest that future efforts for controlling allergic responses in the airways and the skin could include strategies that use the gut microbiota and promote IgA Ab responses and prevent IL-17 responses.

Para elucidar os mecanismos envolvidos na inibição da hipersensibilidade do tipo I em proles murinas mediada pela imunização materna com Ovalbumina (OVA), as proles foram avaliadas quanto aos linfócitos Tγδ produtores de IL-17 (CD27-). A imunização materna com OVA reduziu a expressão de CD27, o que também se refletiu nos pulmões, e a produção de IL-17 por linfócitos Tγδ das proles. A redução da expressão de CD27 também foi evidenciada em linfócitos Tγδ intratímicos das proles após a transferência passiva de anticorpos IgG alérgeno específicos para fêmeas gestantes não imunizadas e in vitro em resposta a estes mesmos anticorpos, efeito que parece envolver a expressão de receptores para IgG expressos por outras células presentes no timo. As evidências obtidas indicam que a imunização materna influi na maturação tímica de linfócitos inibindo a população que colaborara com a inflamação alérgica. Aparentemente, os anticorpos IgG maternos são responsáveis por este fenômeno e a população estudada está envolvida na inibição da inflamação alérgica observada nas proles. / To elucidate the mechanisms involved in inhibition of type I hypersensitivity in murine offsprring mediated by maternal immunization with Ovalbumin (OVA), the offspring were evaluated as IL-17-producing γδT cells (CD27-). Maternal immunization with OVA reduced the expression of CD27, which was also reflected in the lungs, and the production of IL-17 by γδT cells of offspring. Reduction of CD27 expression was also evidenced in intrathymic γδT cells of offspring after passive transfer of allergen-specific IgG antibodies to non-immunized pregnant females and in vitro in response to these same antibodies, effect that seems to involve the expression of IgG receptors expressed by other cells present in the thymus. Evidence obtained indicates that maternal immunization influences the thymic maturation of lymphocytes by inhibiting the population that collaborate with allergic inflammation. Apparently, maternal IgG antibodies are responsible for this phenomenon and the population studied is involved in the inhibition of allergic inflammation observed in offspring.

Efeitos do treinamento físico aeróbio sobre a inflamação pulmonar alérgica crônica em camundongos / Effects of aerobic physical training on lung allergic lung inflammation in mice

A asma é uma doença inflamatória crônica, predominantemente das vias aéreas, mas também envolve o sistema vascular e parênquima pulmonar, na qual as células inflamatórias, a musculatura lisa e o epitélio brônquico têm um papel fundamental na fase inicial, progressão e perpetuação da doença. O treinamento físico aeróbio tem sido indicado como uma relevante forma de auxílio no tratamento de pacientes asmáticos por melhorar a qualidade de vida e reduzir sintomas e o uso de medicamentos. No entanto, não existe um consenso a respeito sobre a intensidade de treinamento ideal para esses pacientes assim como também existem pouquíssimos estudos a respeito dos possíveis mecanismos da atividade física aeróbia para esses pacientes. Por esses motivos, os objetivos do presente estudo foram avaliar os efeitos de duas intensidades de atividade física aeróbia (leve e moderada) sobre um modelo de inflamação pulmonar alérgica crônica em camundongos. Os animais foram sensibilizados com ovalbumina por 51 dias. A atividade física aeróbia teve início no dia 21 e perdurou por 30 dias. Os resultados demonstraram que ambas as intensidades de atividade aeróbia inibiram o desenvolvimento da inflamação predominantemente eosinofílica no lavado broncoalveolar, nos compartimentos peribrônquico, perivascular e no parênquima pulmonar, a expressão de interleucina 4 e 5 pelas células inflamatórias nestes três compartimentos pulmonares. Ambas intensidades de atividade física aeróbia também inibiram significativamente a deposição de fibras colágenas e elásticas (nas vias aéreas e vasos pulmonares) e também o espessamento da musculatura lisa brônquica e vascular assim como da camada epitelial brônquica. Por outro lado, ambas intensidades de atividade física aeróbia não inibiram a síntese de anticorpos anafiláticos IgE e IgG1 e a hiperresponsividade brônquica. Portanto, concluímos que a atividade física aeróbia de intensidade leve e moderada são capazes de inibir o desenvolvimento da inflamação e do remodelamento pulmonar, mas não a hiperresponsividade num modelo experimental de inflamação pulmonar alérgica crônica em camundongos. / Asthma is a chronic inflammatory disease, predominantly involving the airways, but also involving the pulmonary vessels and parenchyma, in which the inflammatory cells, bronchial smooth muscle and epithelium have a central role in the initial phase, progression and perpetuation of the disease. The low and moderate intensity of aerobic physical training have been indicated as a relevant mean to help in the treatment of asthmatic patients, improving life quality, decreasing symptoms and the use of medicines. However, there is not a consensus about the best intensity of training to these patients and also there are few studies about the possible mechanisms of aerobic physical training for asthmatic patients. Therefore, the aims of this study was to evaluate the effects of two intensities of aerobic physical training (low and moderate) on an experimental model of chronic allergic lung inflamattion in mice. The animals were sensitizes with ovalbuim during 51 days. The aerobic physical training started to 21st day and endures for 30 days. The results showed that as low as moderate intensities of aerobic physical training inhibited the eosinophilic inflammation in the bronchoalveolar lavage, peribronchial, perivascular and in the pulmonary parenchyma, as well as the expression of interleukin 4 and 5 by inflammatory cells in these three pulmonary compartments. Both intensities of aerobic physical training also inhibited the collagen and elastic fibers deposition (in the airways and in the pulmonary vessels) and also the thickness of smooth muscle in the airways and vessels, as well as of the airway epithelial layer. On other hand, both intensities of aerobic physical training did not inhibit the synthesis of anaphylactic antibodies IgE and IgG1 and the hyperresponsiveness. Therefore, we conclude that aerobic physical training, to both intensities, were capable of inhibit the development of pulmonary inflammation and remodeling, but not of hyperresponsiveness in an experimental model of allergic lung inflammation in mice.

Determinação dos componentes alergênicos da proteina isolada da soja / Determination of allergenic components isolated soy protein

O objetivo deste trabalho foi esclarecer qual das frações protéicas da soja é mais alergênica, avaliando-se sua imunogenicidade e alergenicidade por imunoensaios e teste de anafilaxia cutânea passiva. Além desse propósito, este estudo visou a produção de anticorpos monoclonais para utilizá-lo como ferramenta na padronização de um ensaio imunoenzimático com a finalidade de detectar proteínas da soja, em produtos comercializados. A purificação das frações 2S,7S e 11S da soja foi realizada com base em estudos prévios, padronizando-se a metodologia dentro das condições experimentais do laboratório. A constatação da pureza foi realizada por eletroforese em gel de poliacrilamida (gel de empilhamento a 5% e de separação em gradiente de 7 a 15%), onde se verificou as bandas protéicas características de cada fração. Evidenciou-se as subunidades α (63,17 KDa) α\' (58,06 KDa) e β (42,09 KDa), correspondentes à fração 7S da soja e as subunidades ácida (38,8 KDa) e básica (21,04 KDa), correspondentes à fração 11S da soja. A fração 2S da soja mostrou uma banda protéica de 20 KDa, nesse ensaio. Os resultados da cinética demonstraram que as frações 7S e 11S da soja são muito imunogênicas, visto que induziram uma grande produção de anticorpos das classes IgM e IgG, em camundongos BALB/c. Por outro lado, a fração 2S da soja induziu uma pequena produção de anticorpos IgG, principalmente no 30º dia após a imunização. No teste de anafilaxia cutânea passiva, quando se avaliou a capacidade das frações protéicas da soja de induzirem a produção de anticorpos IgE, em camundongos BALB/c, verificou-se que a fração 7S da soja é alergênica, nesse modelo experimental. Entretanto, as frações 2S e 11S não estimularam o sistema imunológico desses animais a produzir essa classe de anticorpo. Quando se investigou a presença de anticorpos IgE dirigidos contra as frações protéicas 2S, 7S e 11S da soja, no soro de pacientes alérgicos, obteve-se resultado negativo com todas as três proteínas estudadas. Entretanto, na análise da presença de IgG, tanto o soro dos pacientes alérgicos como o dos controles mostraram-se reativos às frações 2S e 7S da soja, sugerindo a presença do anticorpo IgG4 anafilático no soro teste. Os quatro anticorpos monoclonais reativos às frações 7S e 11S da soja, obtidos neste estudo, mostraram-se reativos até as diluições 1/8000 (2A8 e 1H4) e 1/10000 (1F9) para a fração 7S e 1/12000 (3F2) para a fração 11S da soja. A identificação dos determinantes antigênicos reconhecidos pelos anticorpos monoclonais foi realizado por Imunotransferência. Observou-se que os anticorpos monoclonais anti-fração protéica 7S da soja (1H4, 2A8 e 1F9) reconheceram as subunidades α, α e β (1F9), α\' e β (1H4) e β (2A8), além de outras proteínas. Por outro lado, o anticorpo monoclonal anti-fração protéica 11S da soja (3F2) reagiu apenas com a subunidade básica dessa proteína. A padronização do ensaio imunoenzimático com o objetivo de detectar proteínas de soja, em produtos comercializados, mostrou que os anticorpos monoclonais dirigidos contra as frações protéicas 7S e 11S reconhecem, apenas, as proteínas da soja. Em conclusão, esses resultados sugerem que as frações protéicas 7S e 11S da soja são imunogênicas em camundongos, enquanto que na espécie humana, essa resposta foi observada com as frações 2S e 7S da soja. No teste de anafilaxia cutânea passiva, a fração protéica 7S da soja mostrou a capacidade de induzir anticorpos da classe IgE em camundongos BALB/c, sugerindo que essa fração é alergênica nesse ensaio. Por outro lado, os anticorpos monoclonais reativos às frações 7S e 11S detectaram as proteínas da soja nos produtos comercializados testados. / The goal of this study was to elucidate which of the soy protein fractions is more allergenic by using enzyme immunoassays and passive cutaneous anaphylactic activity test. Furthermore, monoclonal antibodies were obtained in order to standardize immunoassays to detect soy protein fractions in commercial soy-derived products. Purification of 2S, 7S and 11S fractions from soy was based on previous studies which were adapted to our experimental conditions. Purity of the isolated fractions was determined by polyacrilamide gel electrophoresis (7-15%). The subunits α (63,17 KDa) α\' (58,06 KDa) and β (42,09 KDa), corresponding to 7S fraction as well as the acid (38,8 KDa) and basic (21,04 Kda) subunits of 11S fraction were visualized. The 2S fraction showed a 20 KDa band only. Data showed that the 7S and 11S soy fractions are immunogenic as they elicited IgM and IgG antibodies in BALB/c mice. In contrast, the 2S fraction was not immunogenic in this specie. The passive cutaneous anaphylactic activity test showed that 7S fraction is allergenic as it elicited IgE production in BALC mice. However, this test was negative for 2S and 7S fractions. No IgE reactive against the 2S, 7S e 11S was found in sera of patients (n=5) presenting previous food allergy symptoms. However, either these patients or controls (n=5) showed IgG reactive against 2S and 7S fractions in their sera. The four monoclonal antibodies obtained in this study were reactive up to 1/8000 (2A8 and 1H4) and 1/10000 (1F9) against 7S fraction, and up to 1/12000 (3F2) for 11S fraction. Immunoblot analysis showed that the monoclonal antibodies anti-7S fraction (1H4, 2A8 e 1F9) recognized the α α\' and β (1F9), α\' and β (1H4) and α (2A8) subunits. The monoclonal antibody anti-11S fraction (3F2) reacted with the basic subunit of this fraction only. By using ELISA and the monoclonal antibodies anti-7S and anti-11S it was possible to detect especifically these soy protein fractions in commercial soy-derived products. In conclusion, data suggest that 7S and 11S soy protein fractions are immunogenic in mice while 2S and 7S fraction are immunogenic in humans. The IgE response to 7S fraction in BALB/c mice showed by the passive cutaneous anaphylactic activity test indicates that this soy protein fraction is allergenic in this experimental model. A hipersensibilidade a alimentos é comum em crianças e adultos. O camarão, apesar de muito apreciado, é um alimento potencialmente capaz de desencadear reações alérgicas em indivíduos geneticamente predispostos. O principal alérgeno em camarão é a tropomiosina. O objetivo desse experimento foi avaliar os efeitos da radiação gama na reatividade IgE a tropomiosina de camarão da espécie Litopenaeus vannamei e avaliar as modificações físico-químicas e sensoriais no produto, induzidas por esse processo. As amostras foram constituídas de camarões sem o cefalotórax, descongelados e liofilizados, submetidos a diferentes doses de radiação gama: 0 (test.), 5, 10 e 15 kGy. Utilizou-se para a irradiação um irradiador multipropósito, semi-industrial de 60 Co. A taxa de dose empregada foi de 7,5 kGy/hora. Determinou-se a reatividade IgE utilizando as técnicas: ELISA e \"Imunobloting\". Foram realizados testes físico-químicos e sensoriais, para verificar a viabilidade do produto após a irradiação. Os dados obtidos foram submetidos à análise de variância (ANOVA) e teste de Tukey (p<=0,05). As doses de radiação gama aplicadas nesse experimento não alteraram de forma significativa a reatividade IgE, a cor, o colesterol, teor de ácidos graxos e a aceitação dos camarões. Concluiu-se que a radiação gama nessas doses, apesar de não causar modificações que comprometam a qualidade do produto, não pode ser indicada para o controle de alérgenos em camarões, por ter apresentado baixo impacto na modificação da estrutura protéica alergênica / The hypersensitivity to foods is common in children and adults. The shrimp in spite of very appreciated is a food potentially capable to unchain allergic reactions genetically in individuals per disposed. The major allergen in shrimp is the tropomiosin. The aim of this study was to evaluate the effects of gamma irradiation on IgE reactivity to shrimp tropomyosin (Litopenaeus vannamei) and evaluate the physical and chemical changes and sensory product, induced by this process. The samples will be constituted of thawed and freeze-dried shrimp submitted to different radiation doses: 0 (test.), 5, 10 and 15 kGy. Was used for irradiating an irradiation multipurpose semi-industrial sector. 60 Co. The dose rate used was 7.5 kGy / hr. The samples, later to the irradiation, they will go by analyses for allergens determination using the techniques: ELISA and Imunobloting. Acceptance test will be done to verify physic-chemical and sensorial attributes after irradiation. Datas will be evaluated by ANOVA and Tukey test (p<=0.05). The gamma radiation doses applied in this experiment did not change significantly IgE reactivity, color, cholesterol, fatty acid content and the acceptance of shrimp studied. It was concluded that the gamma radiation in these doses, although not causing changes that compromise the quality of the product cannot be indicated for control of allergens in shrimp because it presented a low impact on allergenic protein structure modification
