A Role for Neuronal Nicotinic Acetylcholine Receptors in Dopamine-Mediated Behaviors and the Hypnotic Response to Anesthetics: A Dissertation

Neuronal nicotinic acetylcholine receptors (nAChRs) are ligand-gated cation channels that most notably influence dopamine (DA) release. In this thesis, I examine the role of nAChRs in mediating DA-related behaviors such as movement and drug dependence. To accomplish this, I utilized a “gain-offunction” knock-in mouse (the Leu9’Ala line) containing agonist-hypersensitive α4* nAChRs (* indicates other nAChR subunits in addition to α4 are within the receptor complex) that renders receptors 50-fold more sensitive to nicotine and acetylcholine than wild-type (WT) receptors. I found that DHβE, a selective antagonist for α4β2* nAChRs, induced reversible and robust motor dysfunction characterized by hypolocomotion, akinesia, catalepsy, tremor, and clasping in Leu9’Ala but not WT mice. Reversal of the phenotype was achieved by targeting dopamine signaling. Blockade of mutant α4* nAChRs elicited activation of brain regions in the basal ganglia including dorsal striatum and substantia nigra pars reticulata indicated by c-Fos immunoreactivity. These data indicate that blocking α4* nAChRs in Leu9’Ala mice activates the indirect motor pathway resulting in a motor deficit. We also determined that α4* nAChRs involved in motor behaviors did not contain the α6 subunit, a nAChR subunit highly expressed in DAergic neurons suggesting that different nAChR subtypes modulating striatal DA release have separate functions in motor output. Conditioned place aversion and hypolocomotion, behaviors elicited during nicotine withdrawal, were also induced by DHβE in nicotine-naïve Leu9’Ala but not WT mice. Together these data suggest that DHβE globally reduces DA release in the CNS. In a separate project, I determined that α4* and α6* nAChRs modulate drug-induced hypnosis. Activation of nAChRs increased sensitivity to ketamine-induced hypnosis; whereas antagonizing nAChRs had the opposite effect. Additionally, α4 knockout (KO) mice were less sensitive to the hypnotic effects of ketamine, but α6 KO were more sensitive. High doses of ethanol induce an anesthesia-like state characterized by immobility, analgesia, and hypnosis. Testing the effects of ethanol hypnosis in α4 KO revealed that α4* nAChR do not play a large role in the acute effects of ethanol-induced hypnosis, but are involved in tolerance to this ethanol-induced behavior. The mechanisms of anesthetic-induced hypnosis are still largely unclear, despite the wide use of anesthesia. Future work on these receptors and their involvement in the anesthetic response will help to define a mechanism for hypnosis and improve the use of anesthetic drugs.

Syftet med studien var att beskriva anestesisjuksköterskors upplevelse av möten med vuxna patienter som är oroliga inför generell anestesi. Bakgrund Att sövas innebär för de flesta patienter någon form av oro. För anestesisjukskö- terskor innebär det att på kort tid skapa ett möte där patienter känner trygghet, tillit och där- med mindre oro. Detta är en utmaning då det idag är ett högt flöde av patienter som ska ope- reras, samtidigt som det ställs höga krav på en säker vård där alla patienter ska mötas utifrån sina egna förutsättningar. Få studier har gjorts angående patienters oro utifrån anestesisjuk- sköterskornas perspektiv och är motiveringen till denna studie. Design Studien genomfördes med en kvalitativ design med induktiv ansats. Metod Deltagarna valdes ut med ett ändamålsenligt urval. Datainsamlingen bestod av semi- strukturerade intervjuer med anestesisjuksköterskor på två sjukhus i norra Sverige. Intervjuer- na transkriberades och analyserades med en kvalitativ innehållsanalys. Resultat Elva intervjuer genomfördes under hösten 2015. Analysen av materialet mynnade ut i ett tema med titeln ”Vara professionell”. Temat bestod av fem kategorier med benämningen ”Att lyckas med ett möte”, ”Att se patienternas oro”, ”Att bemöta patienternas oro”, ”Att på- verkas av mötet” och ”Att samarbeta och utvecklas”. Slutsats Anestesisjuksköterskorna upplevde att det i mötena med vuxna oroliga patienter handlade om att vara professionell i sin yrkesutövning. Att vara professionell inkluderade många områden, bland annat inställning, förhållningssätt och att sträva efter samma mål, detta för att kunna se och bemöta patienternas oro. / The aim of this study was to describe anesthetic nurses experiences, when interacting with adult patients that expresses anxiety in regards to their upcoming general anesthesia. Background: To undergo general anesthesia, is for most people, associated with some anxie- ty. The anesthetic nurse only has a limited time to build the patient ́s trust, and to try to relieve the anxiety and apprehension for the upcoming anesthesia. This is now becoming a bigger challenge, due to an increase in patient flow and stricter patient safety regulations, including requirements regarding patient centerdness. There has only been a limited number of studies looking at patient ́s pre-anesthetic anxiety, from a nursing perspective, hence the purpose of this study. Design: A qualitative research design with an inductive approach. Methods: The participants in this study were chosen based on their work. The data collection consisted of semi-structured interviews with anesthetic nurses at two hospitals in northern Sweden. The interviews were transcribed and analyzed using a qualitative content method. Findings: Eleven interviews were conducted during the autumn of 2015. When analyzing the material, a theme ”Be professional” was established. This was further divided to five catego- ries entitled: “How to succeed with a patient interaction”, “To acknowledge patient ́s anxie- ty”, “To master the patient ́s anxiety”, “To be affected by the interaction” and “To cooperate and develop professionally. Conclusion: The anesthetic nurses emphasized the importance of professionalism when deal- ing with anxious pre-anesthetic patients. This professionalism encompasses a number of dif- ferent attributes such as attitude, approach and to work towards a common goal, to acknowledge and address patients concerns and anxiety.

Background: The majority of the children who undergo surgery experience severe anxiety prior to surgery. Preoperative anxiety should be avoided to reduce the psychological trauma. The nurse anesthetist should create a good contact and help the family cope with the frightening situation. There are both pharmacological and non-pharmacological ways to reduce anxiety. Aims: The purpose of the study is to describe the anesthetic nurses' experiences of anesthetizing children. Method: The study is based on interviews with twelve nurse anesthetists, operating at three of the sections on surgery center at one of Sweden's regional hospital. The interviews were based on semi-structured questions and analyzed using qualitative content analysis. Findings: Six categories emerged: parents, profession and compliance to different situations, the important meeting, premedication and external factors. Conclusion: An optimized premedication was highlighted as the most important factor for a successful anesthetic induction of children. If the child already had an intravenous line upon arrival at the surgical department it simplified the induction significantly. How well the parents handled the situation was also important for how the situation turned out. / Bakgrund: Att ett barn ska genomgå en operation är en händelse som påverkar hela familjen. En stor del av de barn som ska opereras upplever ångest och oro inför operationen. Preoperativ oro bör förebyggas för att minska det psykiska traumat. Anestesisjuksköterskan måste skapa en god kontakt och hjälpa familjen bemästra situationen. Det finns både farmakologiska och icke farmakologiska vägar för att minska oro och ångest. Syfte: Syftet med studien är att beskriva anestesisjuksköterskors erfarenheter av att söva barn. Metod: Studien är baserad på intervjuer med tolv anestesisjuksköterskor, verksamma vid tre av sektionerna på operationscentrum vid ett av Sveriges regionsjukhus. Intervjuerna byggde på semistrukturerade frågor som analyserades med hjälp av kvalitativ innehållsanalys. Resultat: Sex kategorier framkom: föräldrar, yrkesutövandet, följsamhet till unika situationer, det viktiga mötet, premedicinering och yttre faktorer. Slutsats: En optimerad premedicinering framhölls som allra viktigast för en lyckad barnanestesi. Att barnet redan hade en intravenös infart vid ankomst till operationsavdelningen förenklade induktionen betydligt. Hur väl föräldrarna hanterade situationen hade även det stor betydelse för hur situationen föll ut, en nervös förälder kunde förvärra barnets oro medan en trygg förälder kunde hjälpa till att lugna barnet.

Single channel analysis of thiol binding to a putative site of alcohol action on the glycine receptor

An alcohol and anesthetic binding pocket is hypothesized to exist among transmembrane domains of the α1 glycine receptor (GlyR). Prior work has shown that amino acid residue serine-267 plays a significant role in the enhancing effects of alcohol and anesthetics and is theorized to form part of an alcohol and anesthetic binding cavity among subunit transmembrane domains. Propyl methanethiosulfonate (PMTS), an alcohol-like thiol, was previously shown to bind to a cysteine residue introduced at position 267 (S267C) and this resulted in permanent enhancement of GlyR function. If ethanol is binding to residue 267 in wildtype GlyR to potentiate receptor function then we hypothesized that covalent thiol labeling would produce receptor enhancement by the same mechanisms as ethanol. Using outside-out patch single channel electrophysiology we determined the open and closed dwell-times and burst properties of S267C GlyR in the absence and presence of PMTS. The primary consequence of PMTS binding to S267C GlyR was an increase in the lengths of burst durations, paralleling the main effect of ethanol on wildtype GlyR. Our findings thus provide a new line of evidence suggesting that ethanol is exerting its enhancing effects on the GlyR through its interactions with amino acid residue 267 in the second transmembrane domain. / text

Accidentes Hipóxicos en Pacientes del Curso de Cirugía de Animales Menores de la Universidad Ricardo Palma

Una de las principales complicaciones durante la anestesia de un proceso quirúrgico es el monitoreo de los problemas de oxigenación del paciente; la falta de oxigenación desencadena un estado de hipoventilación que consecuentemente produce un estado de hipoxemia, lo cual conllevará a un severo deterioro en la salud del paciente y, en el peor de los casos, a la muerte del mismo. El objetivo de este estudio fue determinar el porcentaje de accidentes hipóxicos en pacientes del curso de Cirugía de Animales Menores de la Universidad Ricardo Palma en la ciudad de Lima. Se evaluaron 45 fichas clínicas de registro anestésico entre Agosto del 2012 a Julio del 2014; con los protocolos de Xilacina - Ketamina (Protocolo 1); Xilacina - Ketoprofeno - Ketamina (Protocolo 2) y Xilacina - Tramadol - Ketamina (Protocolo 3). Las fichas de record anestésico se analizaron mediante los programas SPSS y Microsoft Excel. Los resultados promedio indicaron que del total del tiempo de cirugía en cada paciente, el 55% de dicho tiempo el paciente se encontraba en hipoxemia, con un valor de saturación de oxígeno de 92 ± 4.4. Los resultados por protocolo anestésico mostraron un mayor porcentaje de hipoxemia con el Protocolo 3. Los resultados por American Society of Anesthesiologists (ASA) presento un mayor porcentaje de hipoxemia en los ASA 1. Por otro lado se mostraron un mayor porcentaje de hipoxemia en la especie felina. Asimismo se demostró un mayor porcentaje de hipoxemia en ovariohisterectomías. Se comprobó la existencia de estados hipoxémicos en los procedimientos quirúrgicos de la Sala de Cirugía de Animales Menores recomendándose realizar un monitoreo más exhaustivo de constantes vitales en los diferentes procedimientos quirúrgicos y clasificaciones ASA, empleando las diferentes fichas de monitoreo para cada especie. One of the main complications of anesthesia during a surgical procedure, it´s the monitoring of patient oxygenation problems, the lack of oxygen trigger a state of hypoventilation which consequently produces a state of hypoxia, which it would give rise to a severe deterioration in the health of the patient and in the worst cases, death of it self. The aim of this study was to determine the percentage of hypoxic injury in patients during surgery of small animals in the Ricardo Palma´s University in Lima. 45 anesthesic records registered from August 2012 to July 2014; with the protocols of Xylazine – Ketamine (Protocol 1); Xylazine – Ketoprofen – Ketamine (Protocol 2) and Xylazine – Tramadol – Ketamine (Protocol 3) were analyzed using SPSS and Microsoft excel. The average results indicated that the total operation time for each patient, 55% of the time the patient was in hypoxemia with oxygen saturation value 92 +- 4.4. the results for anesthesic protocol demonstrated higher percentage of hypoxemia with Protocol 3. The results for ASA demonstrated a higher percentage of hypoxemia in the ASA 1. On the other hand a higher percentage of hypoxemia were shown in the feline species. A higher percentage of hypoxemia in ovariohysterectomiy is also demonstrated. The existence of hypoxemic states in surgical procedures for the surgery of small animals recommending more comprehensive monitoring of vital signs in different surgical procedures and ASA classifications using different monitoring records for each species registered

Objetivos específicos: Realizar o mapeamento cruzado entre a classificação de DE da NANDA-International (NANDA-I) e o conteúdo do registro de enfermagem utilizado na SRPA; Elencar os DE após o mapeamento cruzado; Propor para cada DE eleito as IE e respectivas atividades de enfermagem (AE) e RE e respectivos indicadores de resultados (IR) da Classificação das Intervenções de Enfermagem (NIC) e da Classificação dos Resultados de Enfermagem (NOC). Métodos: Trata-se de um estudo descritivo, exploratório e retrospectivo de análise documental, aprovado pelo Comitê de Ética e Pesquisa das instituições participantes, para a construção de um instrumento de registro manual do enfermeiro em uma SRPA de um hospital de referência em tratamento oncológico e realização de transplante de medula óssea no interior do Estado de São Paulo. O universo foi composto por 264 fichas pertencentes aos prontuários físicos com os dados: sinais vitais, necessidades afetas, prescrição médica e anotações de enfermagem. O consumo de medicamentos de qualidade é pré-requisito básico para o reestabelecimento da saúde dos indivíduos. No trabalho de pesquisa desenvolvido na faculdade de ciências farmacêuticas da Universidade de São Paulo(USP), destacou-se o controle físico-químico da qualidade dos medicamentos, as principais metodologias adotadas nos laboratórios de controle de qualidade enfatizando-se a identificação e quantificação das impurezas, e de que forma as mesmas são geradas nos processos de fabricação e armazenamento dos medicamentos. Evidencia-se a importância da atuação de órgãos de vigilância sanitária como a Agência Nacional de Vigilância Sanitária (ANVISA) através da consulta da legislação específica das Boas Práticas de Fabricação. Neste contexto, explorou-se o estudo de um medicamento anestésico injetável utilizadoem ambiente hospitalar que está apresentando diversas notificações de desvios de qualidade, através de queixas técnicas e eventos adversos recebidos no sistema de notificações(Notivisa) da Agência Nacional de Vigilância Sanitária. Para a pesquisa, o mestrando utilizou diferentes métodos de identificação, citando-se a cromatografia de camada delgada, espectroscopia no infravermelho, ponto de fusão, espectroscopia UV-Vis e termoanálise. Para a identificação e quantificação, destacou-se a cromatografia líquida de alta eficiência(CLAE) e eletroforese capilar(EC). Thisresearch, developed in the Laboratory of Physical and Chemical Quality Control of Pharmaceutical Preparations and Cosmetics and Central Analytical Center at theFaculty of PharmaceuticalSciences-University of São Paulo, aims the studyand application ofthe main and newmethodologies usedin qualitycontrol laboratories. It was emphasizedthe identification andquantification ofimpuritiesand howthey can begeneratedin the manufacturing processesand storage of pharmaceutical preparations. It was evidenced the importanceof theroleof health surveillanceagenciessuch as the NationalHealth Surveillance Agency(ANVISA) through theconsultationof the specific legislationof GoodManufacturing Practices (GMP). In this context, the local anesthetic drug, bupivacaine in injectables, was selected for the study. This formulation is very much used in hospitals, and recently, several complaints of quality deviations related to technical complaints and adverse events were made through the ANVISA notification system. Several analytical methods were used for identification, such as, thin layer chromatography, infrared spectroscopy, determination of the melting point, UV-VIS spectrophotometry, and termoanalysis. For identification and quantification of the degradation products, high performance liquid chromatography (HPLC) and capillary electrophoresis (CE) were used in the research.

Análise funcional do bloqueio ciático em ratos com lidocaína associada à hialuronidase em complexação com hidroxipropil -  ß-ciclodextrina e citotoxicidade / Functional analysis of rat sciatic block with lidocaine associated to hyaluronidase in complexation with hydroxypropyl- ß-cyclodextrin and cytotoxicity

O uso da enzima hialuronidase como adjuvante do anestésico local está consagrado em oftalmologia, por melhorar a eficácia anestésica devido ao seu efeito difusor de fármacos, ocasionado pela quebra temporária do hialuronan (ácido hialurônico) que é o principal constituinte de tecidos conectivos. Em odontologia, a utilização da enzima como adjuvante no bloqueio do nervo alveolar inferior não apresentou vantagens quando injetada concomitante ao anestésico local, resultando em dor e trismo. Entretanto, com a utilização de um novo protocolo onde a aplicação da hialuronidase ocorria aos 30 min do início da anestesia, prolongou o efeito anestésico e sem ocorrência de efeitos adversos. Este protocolo oferecia a vantagem de evitar complementação anestésica reduzindo os riscos de toxicidade local e sistêmica, porém ainda havia a desvantagem da necessidade de uma nova puntura para aplicação da enzima. Surgiu a hipótese de se manter os benefícios desse protocolo com o uso da hialuronidase, de prolongar a duração da ação anestésica, injetando-a concomitantemente ao anestésico local, porém estando a enzima em um sistema de liberação lenta. Deste modo, a hialuronidase (75 UTR/ml) foi incorporada a uma nanopartícula, a hidroxipropil- ß-ciclodextrina, que atrasaria sua biodisponibilidade e simularia o protocolo da injeção antes do término da ação anestésica. Assim, este trabalho teve como objetivo verificar a eficácia do complexo de inclusão hialuronidase: hidroxipropil- ß-ciclodextrina injetado concomitantemente ao anestésico local, em prolongar a duração de ação do bloqueio nervoso funcional, a fim de oferecer uma alternativa futura de solução anestésica em procedimentos clínicos demorados ou para o alívio de dor, principalmente, em pacientes com comprometimento sistêmico cuja utilização de vasoconstritor adrenérgico ou anestésico de ação prolongada esteja contraindicada. Também foi avaliada a citotoxicidade do novo sistema carreador, o complexo de inclusão hialuronidase: hidroxipropil- ß-ciclodextrina. O bloqueio funcional sensitivo, motor e proprioceptivo do nervo ciático de ratos foi avaliado através de reflexo de retirada da pata e pinçamento, reflexo extensor tibiotarsal e claudicação, e resposta de salto. A citotoxicidade foi avaliada através do teste contagem por Azul de Tripan em fibroblastos e de hemólise em eritrócitos, ambos de rato. Os grupos onde o complexo de inclusão hialuronidase: hidroxipropil- ß-ciclodextrina foi injetado concomitantemente ao anestésico local apresentaram aumento significativo em comparação a todos os demais. O complexo de inclusão não se apresentou citotóxico, mas quando associado à lidocaína, manteve a citotoxicidade desta. Nenhum efeito hemolítico foi observado nas concentrações utilizadas para enzima, o complexo de inclusão e para a lidocaína. / The use of hyaluronidase as an adjuvant to local anesthetic is established in ophthalmology anesthesia to improve the effect of the drugs due to its spreading effect caused by a temporary break of hyaluronan (hyaluronic acid), which is the main component of connective tissue. In dentistry, the use of the enzyme as adjuvant in inferior alveolar nerve blockade was not advantageous when injected concomitantly with local anesthetic, resulting in pain and trismus. However, when the enzyme was injected before the end of nervous block, the anesthetic effect was extended and had no adverse effects. This protocol offered the advantage of avoiding anesthetic supplementation and reduces the risk of local and systemic toxicity, but there was still the disadvantage of receiving a new puncture to inject the enzyme. It has been hypothesized to maintain the benefits of this protocol with the use of hyaluronidase, to extend the duration of anesthetic action, injecting the hyaluronidase concomitantly with the local anesthetic, but the enzyme should be in a slow release system. Thus, hyaluronidase (75 TRU/ml) was incorporated into a nanoparticle, hydroxypropyl- ß-cyclodextrin, which would delay its bioavailability and simulate the protocol of the injection before the end of anesthetic action. This study aimed to determine the efficacy of inclusion complex hyaluronidase: hydroxypropyl- ß- cyclodextrin injected concomitantly with the local anesthetic to extend the duration of action of functional nervous block, to offer a future alternative of anesthetics to the clinical prolonged procedures or to relieve the pain, mainly in patients with systemic compromising whose the use of adrenergic vasoconstrictor or long-acting anesthetic are contraindicated. The cytotoxicity of the inclusion complex hyaluronidase: hydroxypropyl- -cyclodextrin was also evaluated. The nociceptive, motor and proprioceptive functional blockade of the sciatic nerve in rats was assessed by paw withdrawal reflex and clamping (pinprick), tibiotarsal extensor reflex response and claudication, and jumping response, respectively. The cytotoxicity was assessed through cell viability test by Trypan blue counts of fibroblasts and hemolysis test, both in rat cells. Groups where the inclusion complex hyaluronidase : hydroxypropyl- ß-cyclodextrin was injected concomitantly to the local anesthetic showed a significant increase of duration of anesthetic action compared to the others. The plain inclusion complex showed no cytotoxicity, but when associated with lidocaine, the cytotoxicity of this was maintained. None haemolytic effect was observed for the concentration used to the groups hyaluronidase, inclusion complex and lidocaine.

Desenvolvimento e caracterização físico-química de um sistema de liberação controlada da hialuronidase incluída em hidroxipropil--ciclodextrina / Development and physical-chemical characterization of a controlled delivery system of hyaluronidase included in hydroxypropyl- - cyclodextrin

A hialuronidase é uma enzima que vem sendo utilizada como adjuvante anestésico na oftalmologia há décadas para melhorar a eficácia anestésica, por aumentar a difusão do anestésico para o nervo. Na odontologia, quando foi injetada concomitantemente ao anestésico local em bloqueio pterigomandibular, reduziu a duração da anestesia e causou trismo, pois no momento inicial da anestesia ainda havia provavelmente muito anestésico no sítio da injeção, dispersando-o para tecidos adjacentes. Entretanto, em outro protocolo, a enzima foi injetada em outro momento, antes do término do efeito anestésico e, não induziu efeitos adversos e ainda aumentou a duração de ação, provavelmente, por difundir apenas o anestésico remanescente para o nervo. A fim de evitar uma nova puntura para injetar a hialuronidase, a enzima poderia ser incorporada a um carreador, que modularia a sua liberação tardiamente, simulando o protocolo anterior. O desenvolvimento desse sistema de liberação controlada da enzima, a ser futuramente associada aos anestésicos locais visa ainda diminuir a dose necessária para o bloqueio completo, diminuindo assim o risco de toxicidade sistêmica. Este estudo laboratorial teve por objetivo desenvolver o complexo de inclusão utilizando a como carreador da hialuronidase, uma ciclodextrina, a hidroxipropil--xtrina, formando o complexo de inclusão hialuronidase:hidroxipropil--ciclodextrina, em duas proporções molares, 1:1 e 2:1. Neste estudo ainda foram avaliadas as características físico-químicas dos componentes a fim de verificar se havia interação entre as moléculas. Após o preparo, a análise físico-química das substâncias foi realizada utilizando os seguintes métodos: espectroscopia na região do Infravermelho com transformada de Fourier, análise térmica, ressonância magnética nuclear, além da análise morfológica (por MEV). Mudanças observadas na espectroscopia na região do infravermelho e na calorimetria diferencial exploratória indicam interação molecular entre a hialuronidase e a hidroxipropil --ciclodextrina, em ambas razões molares, como sugerem o aspecto morfológico do complexo. / Hyaluronidase is an enzyme that has been used as an adjuvant anesthetic in ophthalmology for decades to improve the effectiveness of anesthesia by increasing the diffusion of the anesthetic to the nerve. In dentistry, when it was injected concomitantly with local anesthetic in inferior alveolar nerve blockade, reduced the duration of anesthesia and caused trismus, because in the initial moment of anesthesia there was probably a lot of anesthetic in the injection site, spreading it to adjacent tissues. However, in another protocol, the enzyme was injected later, before the regression of the anesthetic effect and did not induce adverse effects, and also increased the duration of action, probably by spreading only the remaining anesthetic to the nerve. In order to avoid another puncture to inject hyaluronidase, the enzyme could be incorporated into a carrier, which modulates its release later, simulating the previous protocol, while maintaining its benefits. The development of such controlled release system of the enzyme, to be in the future associated to local anesthetics, also designed to decrease the dose required to obtain the complete block, thus reducing the risk of systemic toxicity. This laboratory study aimed to develop the inclusion complex using as carrier to hyaluronidase, a cyclodextrin, the hydroxypropyl--cyclodextrin forming hyaluronidase : hydroxypropyl--cyclodextrin inclusion complex, in both molar ratios 1:1 and 2:1. This study also evaluated the physico-chemical characteristics in order to ascertain whether there was interaction between the molecules. After being prepared, the physical-chemical analysis of the substances was performed using the following methods: Fourier transform infrared spectroscopy, thermal analysis, nuclear magnetic resonance (RMR), besides the morphological analysis by scanning electronic microscopy (SEM). Changes observed in infrared spectroscopy and differential scanning calorimetry indicate molecular interaction between hyaluronidase and hydroxypropyl-beta-cyclodextrin, in both molar ratios, as suggested by the morphological aspect of the complex.
