Tolypocladium cylindrosporum IP419 and IP425, Lecanicillium psalliotae IP456, IP491 and IP492 were isolated from sentinel larvae. Conidiobolus macrosporus IP396 and L. psalliotae IP455 isolated from adult culicíneos. At 5 and 15 days, depending on the fungus, conidia were scraped from sporulated culture plates and adjusted in suspensions. In 30 eggs/ replicate, 50 μL of suspension of the isolates IP419, IP425, IP455, IP456, IP491 and IP492 were applied in five concentrations (3.3 × 103 to 3.3 × 105 conidia/cm2). For IP 396, 2.5 × 104 conidia/cm2 was applied. In experiments with larvae (10 L3/repeat), five concentrations were applied (3.3 × 105 to 3.3 × 107 conidia/ml) for IP419, IP425, IP455, IP456, IP491 and IP492, and (103 to 105 conidia/ ml) for IP396. The controls were treated with distilled water. Adults were exposed to five concentrations (3.3 × 104 to 3.3 × 106 conidia/cm2) of conidia of IP419 and IP425. For IP396 and IP455, IP456, IP491 and IP492, adults were exposed to the sporulated culture. Adult controls were not exposed to fungi. Ten adults were exposed in 4 replicates. There was no significant activity of the isolates tested against eggs. Mortality of larvae treated with T. cylindrosporum was ≤ 90% at the lowest concentration (3.3 × 105 conidia/ml) and 100% at the highest concentration (3.3 × 107 conidia/ml). The LC50 and LC90 were 3.5 × 104 conidia/ml and 6.8 × 106 conidia/ml for IP419 and 5.9 × 105 and 1.3 × 107 conidia/ml for IP425. The TL50 and TL90 for IP419 at the highest concentration (3 × 107 conidia/ml) was ≤ 7 hours. The concentrations applied had a significant effect (P <0.001). In larvae treated with IP396, mortality of L3 increased with conidia concentration, in the highest concentration (105 conidia /ml), was ≤ 57% in 48 h. There was no growth of the fungus on L3 exposed in agar medium for IP396, for T. cylindrosporum the fungal development was ≤ 70%. Mortality of larvae in controls was ≤ 15%. L. psalliotae had no larvicidal activity. Mortality of adults treated with T. cylindrosporum was ≤ 75%. There was fungal development on corpses ≤ 65%, the concentrations applied had a significant effect (P <0.001). The cumulative mortality of IP396 treated adults reached 100% after 5 days, and ≥ 80% over 10 days for L. psalliotae. The results show that T. cylindrosporum and C. macrosporus have potential for integrated control of larvae and adults of A. aegypti. / Microrganismos são agentes de controle biológico, dentre eles, destacam-se os fungos entomopatogênicos. Tolypocladium cylindrosporum IP419 e IP425, Lecanicillium psalliotae IP456, IP491 e IP492 foram isolados de larvas sentinela. Conidiobolus macrosporus IP396 e L. psalliotae IP455 isolados de culicíneos adultos. Em 5 e 15 dias, dependendo do fungo, conídios foram raspados de placas com cultura esporulada e ajustados em suspensões. Em ovos (30 ovos/repetição), foram aplicados 50 μL de suspensão dos isolados IP419, IP425, IP455, IP456, IP491 e IP492 em cinco concentrações (3,3 × 103 a 3,3 × 105 conídios/cm2). Para IP396, aplicou-se 2,5 × 104 conídios/cm2. Em testes com larvas (10 L3/repetição), cinco concentrações foram aplicadas (3,3 × 105 a 3,3 × 107 conídios/ml) para IP419, IP425, IP455, IP456, IP491 e IP492, e (103 a 105 conídios/ml) para IP396. Os controles foram tratados com água destilada. Adultos foram expostos a cinco concentrações (3,3 × 104 a 3,3 × 106 conídios/cm2) de conídios de IP419 e IP425. Para IP396 e IP455, IP456, IP491 e IP492, adultos foram expostos à cultura esporulada. Adultos controle não foram expostos aos fungos. Foram expostos 10 adultos, em 4 repetições. Não houve atividade significante dos isolados testados contra ovos. A mortalidade de larvas tratadas com T. cylindrosporum, foi ≤ 90% na menor concentração (3,3 × 105 conídios/ml) e 100% na maior concentração (3,3 × 107 conídios/ml). A CL50 e CL90 foi de 3.5 × 104 conídios/ml e 6.8 × 106 conídios/ml para IP419 e de 5.9 × 105 e 1.3 × 107 conídios/ml para IP425. O TL50 e TL90 para IP419, na maior concentração (3 × 107 conídios/ml) foi ≤ 7 horas. As concentrações aplicadas tiveram efeito significativo (P < 0,001). Em larvas tratadas com IP396, a mortalidade de L3 aumentou com a concentração de conídios, na maior concentração (105 conídios/ml), foi ≤ 57% em 48 h. Não houve crescimento do fungo sobre L3 expostas em meio ágar para IP396, para T. cylindrosporum o desenvolvimento fúngico foi ≤ 70%. A mortalidade de larvas nos controles foi ≤ 15%. L. psalliotae não teve atividade larvicida. A mortalidade de adultos tratados com T. cylindrosporum foi ≤ 75%. Houve desenvolvimento fúngico sobre cadáveres ≤ 65%, as concentrações aplicadas tiveram efeito significativo (P < 0,001). A mortalidade acumulada de adultos tratados com IP396 chegou a 100% após 5 dias, e ≥ 80% em dez dias para L. psalliotae. Os resultados mostram que T. cylindrosporum e C. macrosporus têm potencial para controle integrado de larvas e adultos de A. aegypti.

The knowledge in plague control, using entomopathogenic fungi, has been growing year by year, but few works have been done to control S. calcitrans. The aims of this study were rearing a colony of S. calcitrans to make a bioassay of microbial control, verify the pathogenic potential of Metarhizium anisopliae in eggs, larvae and pupae and access three methods of egg s exposition to the studied fungi. After methodologies adaptation of rearing S. calcitrans that are described in literature, the colony was maintained in the Laborat?rio de D?pteros Hemat?fagos - UFRRJ. The bioassays were done in the Laborat?rio de Controle Microbiano de Atr?podes de Import?ncia Veterin?ria - UFRRJ, using the strain 959 of M. anisopliae in a water suspension. In the first method, eggs were immersed in the suspension (2.2 ? 108 conidia /ml and dilutions) and transferred to Petri plates with hydrophilic cotton. In the second methods (2.3 ? 108 conidia/ml and dilutions), eggs were immersed and transferred to the larval rearding medium. In the third method (2.1 ? 108 conidia/ml and dilutions), eggs were put in the larval rearding medium surface and one aliquot of fungus suspension were added. Larvae with nine days of development were immersed in suspension (2.1 ? 108 conidia/ml and dilutions) and after that were transferred to the larval rearding medium while pupae with 16 days of development were immersed in suspension (2.3 ? 108 conidia/ml and dilutions) and were held in Petri plates. Experiments were carried out with 107, 106 and 105 concentrations of conidia/ml and positive and negative controls. The mortality was assessed on the 10th day after fungus exposure. It was verified high mortality in all treated and control groups in the first method of eggs exposure. In the other two methods the eggs exposed to 108 conidia/ml concentration were 100% unviable. In the second method the eggs was 92.5% unviable and the third method was 53.33% both in 107 conidia/ml concentration. The subsequent concentrations did not show significant difference when compared to controls, neither between two methods of fungi exposure. Larvae and pupae exposure did not show statistic difference between treated and controls groups. Metarhizium anisopliae suspensions in high concentrations were capable to make unfeasible eggs of S. calcitrans; however, it did not happen in larvae and pupae experiments, in which fungi did not prevent their development. The second method was the best one to control immature forms of S. calcitrans but the third one resembles the nature conditions, in which has significant result only in the highest fungi concentration. Experimental methodologies that simulate nature conditions may optimize the utilization of fungi in arthropods control, in a way that it does not modify its action when applied in the field. / Stomoxys calcitrans ? uma mosca hemat?faga associada ? redu??o da produtividade, devido ao estresse causado aos animais e a dissemina??o de pat?genos. O conhecimento do controle de pragas, utilizando fungos entomopatog?nicos, vem crescendo no decorrer dos anos, por?m poucos trabalhos t?m sido realizados visando o controle de S. calcitrans. O objetivo deste estudo foi estabelecer uma col?nia de S. calcitrans direcionada ao bioensaio de controle microbiano, verificar o potencial patog?nico de Metarhizium anisopliae sobre ovos, larvas e pupas, e ainda, avaliar tr?s m?todos de exposi??o dos ovos ao fungo estudado. Ap?s adapta??o das metodologias de cria??o de S. calcitrans descritas na literatura, a col?nia foi mantida no Laborat?rio de D?pteros Hemat?fagos UFRRJ. Os bioensaios foram realizados no Laborat?rio de Controle Microbiano de Artr?podes de Import?ncia Veterin?ria UFRRJ, onde foi utilizada a cepa 959 de M. anisopliae para elabora??o das suspens?es aquosas. No primeiro m?todo, os ovos foram imersos na suspens?o (2,2 x 108 con?dios/ml e dilui??es) e transferidos para placa de Petri contendo algod?o hidr?filo. No segundo m?todo (2,3 x 108 con?dios/ml e dilui??es) os ovos foram imersos e transferidos para o meio de desenvolvimento larval. No terceiro m?todo (2,1 x 108 con?dios/ml e dilui??es), os ovos foram depositados sobre a superf?cie do meio de desenvolvimento larval e uma al?quota da suspens?o f?ngica foi adicionada. As larvas com nove dias de desenvolvimento foram imersas na suspens?o (2,1 x 108 con?dios/ml e dilui??es) e transferidas para o meio de desenvolvimento larval, enquanto que as pupas de 16 dias de desenvolvimento foram imersas na suspens?o (2,3 x 108 con?dios/ml e dilui??es) e depositadas em placas de Petri. Foram utilizadas dilui??es na concentra??o de 107, 106 e 105 con?dios/ml e controle positivo e negativo. A mortalidade foi avaliada no d?cimo dia ap?s exposi??o, onde foi observado se ocorria mudan?a de est?gio. Foi verificada alta mortalidade em todos os grupos (tratados e controles) no primeiro m?todo de exposi??o de ovos. Nos outros dois m?todos, foram obtidos 100% de inviabilidade dos ovos expostos ? concentra??o de 108 con?dios/ml. No segundo m?todo, houve 92,5% de inviabilidade dos ovos, enquanto que no terceiro m?todo, a inviabilidade foi de 53,33%, ambos na concentra??o de 107 con?dios/ml. As concentra??es subseq?entes n?o apresentaram diferen?a significativa frente aos controles e nem entre os dois m?todos de exposi??o f?ngica. Nas exposi??es de larvas e pupas n?o ocorreu diferen?a estat?stica entre os tratamentos e os controles. Suspens?es de M. anisopliae em altas concentra??es s?o capazes de inviabilizar ovos de S. calcitrans, entretanto, isto n?o ocorreu em larvas e pupas, pois o fungo n?o impediu o desenvolvimento destes. O m?todo mais efetivo para o controle das formas imaturas de S.calcitrans foi o segundo m?todo, entretanto, o que mais se assemelha ?s condi??es naturais seria o terceiro, onde foi obtido resultado significativo apenas na maior concentra??o f?ngica. A ado??o de metodologias experimentais que simulem condi??es naturais pode otimizar a utiliza??o de fungos no controle de artr?podes, de forma que sua a??o n?o seja comprometida, quando aplicados no campo.

Were conducted two experiments in vitro, where were made a germination test of Colletotrichum lindemuthianum conidium after the treatment with Trichoderma spp. filtered, and the direct confrontation test of 21 cultures isolated from Trichoderma spp. agains this pathogen were also conducted two experiments in vivo: on the first imposed treatments in base of Trichoderma in the beans inoculated via seed; on the second experiment the treatments aplication were made through foliar spray of filtered medium of Trichoderma spp. For the experiments in vivo were realized sheets collects for biochemical analysis, in which there were two collects for the experimente that were treated via seed (15 days after the foliar emergency and three days after the pathogen inoculation). Whereas for the experiment which the treatments were via foliar spray of filtered medium of Trichoderma, were realized three collectings, (the first without the treatment aplication, the second three days after the filtered pulverization and the third collect realized three days after the pathogen inoculation). Were determined the polyphenoloxidase activity (PFO), peroxidase (POX) and phenylalanine ammonia lyase (FAL), and were evaluated the total protein content, total phenols, lignin and the severity of anthracnose after treatments in base of Trichoderma for both experiments. For the germination test of C. lindemuthianum, conidium the filtered from the isolated TM1, TI1 and TOD1 reduced the germination in, 33%, 38% and 61% respectivily. In direct confrontation were realized that the isolated inhibited pathogen growth. For the PFO activity in via seed treatment, the isolated TLB12, TOD1 and TOD2B caused an increase thereof after challenge with the pathogen, while the treatment via foliar spray of Trichoderma filtered were not observed significant difference. The POX activity in beans treated at sowing, for the time 1 weren‟t observed difference between the treatments, whereas in the time 2 some isolated tended to suppress the enzymatic activity, while for plants treated via foliar pulverization notes the tendency to enzymatic suppression of POX in time 2 caused by some filtered in relation to the positive control. The FAL enzymatic activity and the total soluble protein contents both in the aplication of the treatments via seed and via foliar pulverization in beans didn‟t result in significant difference. For the total phenol contents and lignin the treatments in base of Trichoderma via sowing and via foliar pulverization didn‟t promote an icrease of the same in beans. The severity in treated beans presented in general low, making these inaccurate data / O objetivo do trabalho foi avaliar a ação do Trichoderma spp. e seus filtrados como agente de biocontrole da antracnose e indutor de resistência em feijoeiro. Conduziram-se dois experimentos in vitro, onde foi realizado teste de germinação de conídios de Colletotrichum lindemuthianum após tratamento com filtrados de Trichoderma spp., e o teste de confronto direto de culturas de 21 isolados de Trichoderma spp. frente a esse patógeno também conduziu-se dois experimentos in vivo: no primeiro se impôs os tratamentos a base de Trichoderma no feijoeiro inoculado via semente; já no segundo experimento a aplicação dos tratamentos se deu por meio de pulverização foliar de filtrados de Trichoderma spp. Para os experimentos in vivo realizaram-se coletas de folhas para análises bioquímicas, no qual houve duas coletas para o experimento cujos tratamentos foram via semente (aos 15 dias após a emergência foliar e três dias após a inoculação do patógeno). Já para o experimento aos quais os tratamentos foram via pulverização foliar de filtrados de Trichoderma, realizaram-se três coletas, (a primeira sem a aplicação do tratamento, a segunda realizada três dias após a pulverização dos filtrados e a terceira coleta realizada três dias após a inoculação do patógeno). Determinou-se a atividade de polifenoloxidase (PFO), peroxidase (POX) e fenilalanina amônia-liase (FAL), e foram avaliados os teores de proteínas totais, fenóis totais, lignina e a severidade da antracnose após os tratamentos a base de Trichoderma para ambos os experimentos. Para o teste de germinação de conídios de C. lindemuthianum, os filtrados dos isolados TM1, TI1 e TOD1 reduziram a germinação em, 33%, 38% e 61% respectivamente. No confronto direto percebeu-se que os isolados inibiram o crescimento do patógeno. Para a atividade de PFO em tratamentos via semente, os isolados TLB12, TOD1 e TOD2B causaram aumento da mesma após o desafio com o patógeno, enquanto que no tratamento via pulverização foliar de filtrados de Trichoderma não foi observada diferença significativa. A atividade de POX em feijoeiro tratado na semeadura, para o tempo 1 não observou-se diferença entre os tratamentos, já no tempo 2 alguns isolados tenderam a suprimir a atividade enzimática, enquanto que para as plantas tratadas via pulverização foliar nota-se tendência de supressão enzimática da POX no tempo 2 ocasionada por alguns filtrados em relação ao controle positivo. A atividade enzimática de FAL e os teores de proteínas solúveis totais tanto na aplicação dos tratamentos via semente quanto via pulverização foliar em feijoeiro não acarretou em diferença significativa. Para os teores de fenóis totais e de lignina os tratamentos a base de Trichoderma via semeadura e via pulverização foliar não promoveram incremento dos mesmos em feijoeiro. Já a severidade em feijoeiro tratado apresentou-se no geral baixa, tornando estes dados imprecisos

Extinctions et recouvrements de coraux au cours de la crise Pliensbachien - Toarcien / Extinctions and recovery of corals during the Pliensbachian – Toarcian crisis

Le passage entre les étages géologiques du Pliensbachien et du Toarcien au Jurassique inférieur est suivi au Toarcien inférieur par un évènement anoxique d’importance planétaire (TOAE pour Toarcian Oceanic Anoxic Event). Dans les archives de l’histoire de la terre, ce moment correspond en bien des endroits à un temps de fossilisation massive de matière organique donnant ici où là des roches mères d’intérêt pétrolier. Si, en termes de fluctuation du niveau eustatique, ces évènements sont souvent situés dans une zone d’inondation maximale de 2ème ordre, ils sont aussi interprétés comme une perturbation globale du cycle du carbone avec acidification des océans, liée à la mise en place des provinces ignées du Karroo-Ferrar en Afrique du Sud et en Amérique du Sud, contrées jointives à ce moment-là. Il s’agit d’une période de réchauffement global faisant suite à une période particulièrement froide au Pliensbachien. Cette perturbation écologique globale a certainement affecté la biosphère dans une mesure qui reste aujourd’hui très mal connue. Nous connaissons des études par exemple sur des ammonites ou des bivalves dont la définition stratigraphique permet rarement de distinguer l’impact du passage de la limite entre les deux étages du passage au Toarcien inférieur de la TOAE et la seule étude disponible sur les coraux est une étude purement bibliographique (Lathuilière et Marchal 2009) qui laisse supposer une extinction significative sur la même période. Les coraux sont très généralement des marqueurs particulièrement précieux des perturbations écologiques majeures comme on peut le voir pour les cinq grandes extinctions et même pour la sixième extinction en cours. Cette extinction au Pliensbachien-Toarcien pour l’instant considérée de second ordre mérite donc d’être analysée de ce point de vue. Dans le cadre de cette thèse, des collectes de coraux ont été effectuées sur le terrain au Maroc et en Italie dans un contexte sédimentologique défini de part et d’autre de la limite Pliensbachien - Toarcien. Les prélèvements ont été réalisés de façon à pouvoir évaluer la diversité ainsi que, dans la mesure du possible, la variabilité intraspécifique. Le traitement statistique des données a été réalisé dans le souci d’assurer une bonne connaissance de la variabilité pour fournir une taxinomie fiable dans l’optique de comparer des populations comparables en termes d’analyse de la diversité, des extinctions et des apparitions de taxons. Au total, 107 espèces ont été décrites (dont au moins 19 nouvelles espèces) réparties parmi 60 genres (dont 5 nouveaux) dans au moins 22 familles (dont 1 nouvelle). Cette étude nous révèle pour les eaux froides et chargées en matière organique du Pliensbachien des assemblages de coraux présentant une grande part d’affinité avec les faunes du Trias (en termes de genres et familles). Ils partagent une partie de leurs niches écologiques sur les plateformes internes de l’ouest téthysien avec des bivalves constructeurs de récifs connaissant alors un succès évolutif fulgurant : les lithiotidés. Les données récoltées témoignent de conditions ayant contraint les coraux à s’adapter et à se diversifier pour se maintenir lors de cette période peu propice, au profit des formes solitaires et phacéloïdes et au détriment des formes de plus hauts niveaux d’intégration. Au Toarcien inférieur, le réchauffement global des eaux couplé à la disparition quasi-totale des concurrents des coraux a permis une explosion de diversité déterminante pour l’avenir de ce groupe avec la mise en place de faunes d’assemblages très similaires à celles du Jurassique moyen et supérieur (en terme de genres, familles et morphologies coloniales). Il s’agit d’un évènement d’extinction biphasé dont la première phase se produit au passage entre Pliensbachien et Toarcien, la seconde phase correspondant au passage du TOAE. L’évènement pris dans son ensemble amène à l’extinction de près de 97% des espèces de coraux présentes au Pliensbachien / The transition from the Pliensbachian to the Toarcian geological stages in Lower Jurassic is followed by a global oceanic anoxic event during the lower Toarcian (TOAE for Toarcian Oceanic Anoxic Event). This moment corresponds to a massive fossilization time for organic matter in the worldwide geological record that produced here and there source rocks of petroleum interest. Concerning the eustatic fluctuations, these events are associated to a second-order maximal flooding zone. They are also interpreted as the remains of a global carbon-cycle perturbation case associated with oceanic acidification and correlated with the setup of Karoo-Ferrar igneous provinces in the southern Pangea territories corresponding to present-day Southern Africa and Southern America. It corresponds to a period of global warming that directly follow a global cooling at the Pliensbachian. This worldwide ecological perturbation probably disturbed the biosphere in a degree that is still poorly quantified. For example, studies about ammonites and bivalvs are known but the stratigraphic definition generally do not allow to distinguish the Pliensbachian-Toarcian boundary itself to the TOAE effect on these fauna. One single available study about corals is purely bibliographic (Lathuiliere and Marchal 2009) and suggests a significant extinction event for this group at the same period. Corals are generally precious indicators for major ecological disruptions as it is testified in the case of the big-five major crisis and the current sixth one. The Pliensbachian-Toarcian extinction has been considered up-to-now as a second-order crisis event and require to be studied as such. In the context of this thesis, fossilized corals have been collected in the field in Morocco and Italy in a well-defined sedimentological context before and after the Pliensbachian-Toarcian boundary. Sampling has been in view to quantify the diversity and as far as possible, the intraspecific variability. The statistical treatment of the data has been realized in view to provide a satisfying quantification of the variability in order to supply a strong taxonomy and be able to compare comparable populations in analyses of diversity, extinctions and apparitions of taxons. In total, 107 species have been describes (including at least 19 new species) as part of 60 genera (including 5 new ones) among 22 families (including a new one). According to this study, the cold and highly concentrated in organic matter oceanic waters of the Pliensbachian contained corals faunas with an important affinity with Triassic faunas (in terms of genera and families). They shared the ecological niches of the western tethysian inner platforms with the lithiotids, a group of aberrants reefal bivalvs that demonstrated a stupendous evolutive success during the same period. Collected data attest of conditions that constrained corals to adapt and diversify in view to maintain during these unfavourable times, for the benefit of the solitary and phaceloid form but at the expense of the highly integrated forms. During the Lower Toarcian, global warming of oceanic waters associated with the almost total disparition of the competitors premised an explosion of diversity that is determinant for the future of the corals, with the apparition of faunal assemblages very similar to the typical Middle and Upper Jurassic ones (in terms of genera, families and colonial morphologies). It is a two-folded extinction event with a first phase during the Pliensbachian –Toarcian transition and a second phase at the onset of the TOAE. In the end, this pulsed event led to the extinction of approximately 97% of the Pliensbachian coral species

Avaliação das alterações dos tecidos ao redor de implantes inseridos em diferentes níveis em relação à crista óssea : estudo clínico, radiográfico, e histométrico em cães /

Resumo: O objetivo do presente estudo foi avaliar alterações ao redor de implantes inseridos em diferentes níveis em relação à crista óssea, e sob diferentes protocolos de restauração. Trinta e seis implantes foram inseridos nas mandíbulas edêntulas de 6 cães. Três implantes foram instalados por hemi-mandíbula, cada qual representativo de um grupo experimental. Estes foram determinados de acordo com a distância entre a junção implante-conector protético (JIC) e a crista óssea: Ao Nível (ao nível da crista óssea), Menos 1 (1mm apical à crista óssea), ou Menos 2 (2mm apical à crista óssea). Cada hemi-mandíbula foi submetida a um protocolo de restauração: convencional (prótese instalada 120 dias após a implantação) ou imediata (prótese instalada 24 horas após a implantação). Parâmetros clínicos, radiográficos, e histométricos foram avaliados após 90 dias de restauração, e os dados foram analisados estatisticamente (alfa=5%). A posição da margem do tecido mole (PTM) não foi influenciada pelo posicionamento apical da JIC; entretanto, sítios submetidos à restauração imediata tiveram a PTM significantemente mais coronal que os submetidos à restauração convencional (p=0,02, análise clínica). A reabsorção do rebordo não foi estatisticamente diferente entre os grupos (p>0,05, análise radiográfica). Menores quantidades de Perda Óssea Lateral (POL) foram observadas nos sítios restaurados imediatamente em comparação com os restaurados convencionamente (p=0,006, análise histométrica). Os presentes achados sugerem que o posicionamento apical da JIC não põe em risco a altura de tecidos periimplantares moles ou duros. Além disto, a restauração imediata foi benéfica para manter a PTM, e minimizar a POL. / Abstract: The aim of the present study was to evaluate changes that occur around dental implants inserted in different levels in relation to crestal bone, under different restoration conditions. Thirty-six implants were inserted in the edentulous mandible of six dogs. Three implants were inserted per hemimandible, each one representing an experimental group according to the distance from the implant-abutment junction (IAJ) to the crestal bone: Bone Level (at crestal bone level), Minus 1 (1mm below crestal bone), or Minus 2 group (2mm below crestal bone). Each hemimandible was submitted to a restoration protocol: conventional (prothesis installed 120 days after implant placement) or immediate (prosthesis installed 24 hours after implant placement). Clinical, radiographic, and histometric parameters were evaluated after 90 days of restoration, and data was analyzed statistically (alpha=5%). The position of soft tissue margin (PSTM) was not influenced by the apical positioning of the IAJ; however, sites submitted to immediate restoration had the PSTM displayed significantly more coronally than those submitted to conventional restoration (p=0.02, clinical analysis). Ridge Loss was not statistically different among groups (p>0.05, radiographic analysis). Greater amounts of Lateral Bone Loss (LBL) were observed for conventionally than for immediately restored sites (p=0.006, histometric analysis). These findings suggest that the apical positioning of IAJ did not jeopardize the height of peri-implantar soft and hard tissues. Plant-Made Biologics: Human Butyrylcholinesterase Mutants for the Treatment of Cocaine Addiction-Related Diseases

abstract: Cocaine abuse affects millions of people with disastrous medical and societal consequences. Despite this, there is still no FDA-approved treatment to decrease the likelihood of relapse in rehabilitated addicts, and acute cocaine toxicity (overdose) is only symptomatically treated. Studies have demonstrated a promising potential treatment option with the help of the human serum enzyme butyrylcholinesterase (BChE), an enzyme capable of breaking down cocaine into biologically inactive side products. This activity of wild-type BChE, however, is relatively low. This prompted the design of variants of BChE which exhibit significantly improved catalytic activity against cocaine. Plants were used as a sustainable, scalable, affordable platform system to produce large amounts of human biologics such as these cocaine hydrolase variants of BChE. Using a tobacco relative, Nicotiana benthamiana, recombinant enzymes can be produced at quantities relevant to clinical use with desired kinetic properties. Next, the ability of the most promising plant-produced cocaine super hydrolase, pCocSH, to counter the lethal effects of cocaine overdose in vivo was tested. These studies revealed that this plant-produced enzyme can protect mice from an otherwise lethal dose of cocaine. Most excitingly, it was found that pCocSH can rescue mice from overdose when given immediately after the onset of cocaine-induced seizures. The flora study, ethnobotanic stands out for understanding of the interaction of society whit plants. In this view, teacher Dr. Vanessa Pasqualotto research group’s has been carrying out ethnobotanic survey in collaboration with the Coqueiros community, with the objective of recovering and valuing the popular knowledge about medicinal plants, among which the Kielmeyera coriacea species, the focus of this study, stands out. In this group, 37 members were interviewed, of which 76% indicate K. coriacea for the treatment of infections, rheumatism, leukemia, tooth pain, among others. Thus, the chemical study of ethanolic extract of roots (EER) of this species, aiming at to characterize its secondary metabolites, since there are no chemical reports in the literature for this part of the plant. In addition antioxidant, antimicrobial and antitumor potential of K. coriacea were evaluated. From the liquid-liquid extraction of EER the hexane (FH), ethyl acetate (FAE) and hydroalcoholic (FHA) fractions were obtained; by the fractionation of FAE, the substance 3-hydroxy-2,4- dimethoxyxanthone was isolated. Additionally, a fingerprint study was performed by LC-MS technique, identifying major compounds such as δ-tocotrienol, prenylated acylphoroglucinol, 2-hydroxy-1-methoxyxanthone, quercitrin. EER, FH, FAE, FHA, its subfractions and the isolated substance were submitted to biological tests. FHA presented antioxidant action with EC50 de 201,53 μg mL-1. EER and FH-10 subfraction from FH fractionation, inhibited the bacterial growth of Streptococcus pyogenes and S. pneumoniae (Minimum Inhibitory Concentration of 6.25 and 1.56 μg mL-1, respectively); EER also inhibited the fungus Candida glabrata in 7.81 μg mL-1. Finally, FH, FAE and FH-10 presented an antitumor effect against the HeLa cells, which are related to cervical cancer (IC50 of 2.5 μg mL-1, 2.5 μg mL-1 and 0.89 μg mL-1, respectively). When correlating the chemical and biological data, it is possible that the FAE-4.7.3 subfractions, from the fractionation of FAE, and FH-10 are constituted by substances such as 4-hydroxy-2,3-methylenedioxy xanthone, 3-hydroxy-1,2-dimethoxy xanthone, lupeol, prenylated acylphoroglucinol and quercitrin, which could be associated with the biological potential found. Therefore, the knowledge acquired in this study contributed to the expansion of the chemical-biological knowledge of K. coriacea. / O Cerrado é uma região cuja riqueza biológica é singular, sendo formado por cerca de 10 mil plantas. No estudo da flora, a etnobotânica se destaca por compreeder a interação da sociedade com as plantas. Nesse contexto, o grupo de pesquisa da Profa. Dra. Vanessa Pasqualotto vem realizando estudos etnobotânicos em colaboração com a comunidade Coqueiros, visando o resgate e a valorização dos saberes populares sobre plantas medicinais, dentre as quais se destaca a espécie Kielmeyera coriacea, foco deste estudo. Neste estudo foram entrevistados 37 membros, em que 76% indicam K. coriacea para o tratamento de infecções, reumatismo, leucemia, dor no dente, dentre outras. Assim, foi realizado o estudo químico do extrato etanólico da raiz (EER) desta espécie, objetivando caracterizar seus metabólitos secundários, uma vez que não há relatos químicos na literatura para esta parte da planta. Ademais, foi avaliado os potenciais antioxidante, antimicrobiano e antitumoral de K. coriacea. A partir da extração líquido-líquido de EER foram obtidas as frações hexano (FH), acetato de etila (FAE) e hidroalcoólica (FHA); pelo fracionamento da FAE foi isolada a substância 3-hidroxi-2,4-dimetoxixantona. O estudo do perfil químico foi realizado por CLAE-EM, identificando-se compostos majoritários como δ-tocotrienol, acilforoglucinol prenilado, 2-hidroxi-1-metoxixantona e quercitrina. EER, FH, FAE, FHA e suas subfrações, bem como a substância isolada foram submetidos aos ensaios biológicos. FHA apresentou ação antioxidante com EC50 de 201,53 μg mL-1. EER e a subfração FH-10, proveniente do fracionamento de FH, inibiram o crescimento bacteriano de Streptococcus pyogenes e S. pneumoniae (Concentração Inibitória Mínima de 6,25 e 1,56 μg mL-1, respectivamente); EER também inibiu o fungo Candida glabrata na 7,81 μg mL-1. Por fim, FH, FAE e FH-10 apresentaram efeito antitumoral frente à célula HeLa, a qual está relacionada ao câncer do cólon do útero (IC50 de 2,5, 2,5 e 0,89 μg mL-1, respectivamente). Ao correlacionar os dados químicos com biológicos, tem-se que as subfrações FAE-4.7.3, proveniente do fracionamento de FAE, e FH-10 são constituídas por substâncias como 4-hidroxi-2,3-metilenodioxixantona, 3-hidroxi-1,2-dimetoxixantona, lupeol, acilforoglucinol prenilado e a quercitrina, as quais poderiam estar associadas ao potencial biológico encontrado. Portanto, o conhecimento adquirido neste estudo contribuiu para a ampliação dos saberes químico-biológicos de K. coriacea.

Significant advances in the recognition of the pathophysiology of disease and injury mechanisms allowed the development of new treatment options with drugs that block inflammatory mediators involved in the mechanism of the disease, known as biologics. These drugs are expensive and of continuous use. They are complex to use due care and management of storage and the risk of immediate and delayed adverse reactions. These therapies are available at the Health System for 12 years, is necessary to evaluate the effectiveness of this drug group in the Brazilian public health system in the treatment of rheumatoid arthritis. OBJECTIVES: a) analyze the effectiveness of biological therapies in patients with rheumatoid arthritis of the State of Mato Grosso, through the assessment of disease activity index DAS28-ESR b) Review the laboratory characteristics of these patients; c) examine the safe use these therapies and to describe adverse events. d) analyze the efficiency of the program dispensing of biological therapies in the State of Mato Grosso, Brazil. METHODS: Two groups of patients with rheumatoid arthritis users Pharmacy High Cost of the State Secretariat of Health of Mato Grosso (FAC / SES) or the University General Hospital. Group I included 68 patients who were already in use of biological therapies. These were assessed for disease activity at the time of inclusion in the study and asked about the historical use of biological drugs, satisfaction with access to treatment and adverse events. Laboratory tests prior to treatment were obtained retrospectively. In group II were enrolled and followed prospectively for 24 weeks, 30 patients with early use of biologic therapies. They were evaluated at baseline and at weeks 4, 8, 12 and 24, the disease activity by DAS28-ESR, laboratory tests, satisfaction with access to treatment and adverse events. RESULTS: The groups were similar in demographic characteristics and laboratory related to rheumatoid arthritis. In group I, 80.6% of patients expressed satisfaction with the care of the FAC / SES. The average use of biological was 12.3 months, and 20.9% (14/68) reported an adverse event, all non-serious. The first medication was discontinued in 26.5% (18/68) of patients, 55.6% (10/18) for loss of efficacy and 22.2% (4/18) adverse events. The mean value of DAS28-ESR was 3.8, and 41.2% were in remission or low disease activity, 39,7% in moderate activity and 19,1% in intense activity. In group II, 91.7% of patients expressed satisfaction with the care provided by FAC / SES. Laboratory evaluation was significant change in the conversion of ANA from 13.3% to 23.3% and anti-DNA (ds), 0 to 6.7%, but no clinical significance. The anti-CCP (2) remained positive and stable at 63.3%. There was an adverse reaction in 23% (7/30) of patients. The only suspension of the first plants were by no severe adverse event (hypersensitivity). Following the DAS28-ESR was a significant reduction in the initial mean value of 4.3 to 3.3 (p = 0.002) after 24 weeks of treatment, 60% of patients were in remission or low disease activity, 23, 3% in moderate activity and 16.7% in intense activity. The average value of ESR-DAS28 after use of the biologic group II was significantly lower than in group I (p= 0.042). CONCLUSIONS: 1) Treatment with biologic therapies in the State of Mato Grosso was effective in prospective group. The response in DAS28-ESR was significantly better in the group where the monitoring and control has been programmed with a routine test and calculate the index of disease activity were used. 2) Among the laboratory characteristics were significant conversion in the profile of autoantibodies, confirming the potential immunogenicity of these drugs inducing the formation of autoantibodies, but no clinical significance in the study group. The anti-CCP remained stable in 63% of patients (group II). 3) Biological therapies were safe. There were no serious adverse events reported. There were no reports of infection or death. The most common reason for discontinuation of the first was the loss of biological effectiveness. 4) Patients with rheumatoid arthritis in the use of biologic therapies in the State of Mato Grosso showed satisfaction with the services offered by SES / MT and the majority of users in the metropolitan area of Cuiabá. The program dispensing biological therapies showed higher efficiency when linked to a reference center for this particular type of treatment. / A artrite reumatoide é doença autoimune, debilitante, com graves consequências físicas, econômicas e emocionais, que afeta cerca de 0,5 a 1% da população adulta mundial. Avanços significativos no reconhecimento da fisiopatologia da doença e mecanismos de lesão permitiram desenvolvimento de novas opções de tratamento através de medicamentos que bloqueiam mediadores inflamatórios envolvidos no mecanismo da doença, denominados biológicos. Esses medicamentos são de alto custo e de uso contínuo. São de uso complexo devido aos cuidados de armazenagem e administração além do risco de reações adversas imediatas e tardias. Essas terapias estão disponíveis no Sistema Único de Saúde há 12 anos, sendo necessário avaliar a efetividade deste grupo de medicamentos no sistema de saúde público brasileiro no tratamento da artrite reumatoide. OBJETIVOS: a) analisar a eficácia das terapias biológicas em pacientes com artrite reumatoide do Estado de Mato Grosso, através da avaliação do índice de atividade da doença DAS28-VHS; b) analisar as características laboratoriais destes pacientes; c) analisar a segurança no uso destas terapias e descrever os eventos adversos. d) analisar a eficiência do programa de dispensação das terapias biológicas no Estado de Mato Grosso-Brasil. MÉTODOS: Utilizou-se dois grupos de pacientes com artrite reumatoide usuários da Farmácia de Alto Custo da Secretaria de Estado de Saúde de Mato Grosso (FAC/SES) ou do Hospital Geral Universitário. No grupo I foram incluídos 68 pacientes já em uso de terapias biológicas. Esses foram avaliados quanto à atividade da doenca no momento de inclusão no estudo e questionados sobre o histórico de uso das medicações biológicas, satisfação com o acesso ao tratamento e eventos adversos. Os exames laboratoriais prévios ao tratamento foram obtidos de forma retrospectiva. No grupo II foram admitidos e acompanhados de forma prospectiva por 24 semanas, 30 pacientes em início de uso de terapias biológicas. Esses foram avaliados no início do tratamento e nas semanas 4, 8, 12 e 24, quanto à atividade da doença através do DAS28-VHS, exames laboratoriais, satisfação com o acesso ao tratamento e eventos adversos. RESULTADOS: O grupos estudados foram semelhantes em características demográficas e laboratoriais relacionados a doença reumatoide. No grupo I 80,6% dos pacientes demonstraram satisfação com o atendimento da FAC/SES. O tempo médio de uso de biológicos foi de 12,3 meses, sendo que 20,9% (14/68) relataram algum evento adverso, todos não graves. A primeira medicação foi suspensa em 26,5% (18/68) dos pacientes, sendo 55,6% (10/18) destes por perda de eficácia e 22,2% (4/18) por eventos adversos. O valor médio do DAS28-VHS foi 3,8, sendo que 41,2% estavam em remissão ou baixa atividade da doença, 39,7% em atividade moderada e 19,1% em atividade intensa. No grupo II, 91,7% dos pacientes demonstraram satisfação com o atendimento prestado pela FAC/SES. Na avaliação laboratorial houve alteração significativa na conversão do FAN de 13,3% para 23,3 % e do anti-DNA(ds), de 0 para 6,7%, porém sem repercussão clínica. O anti-CCP(2) manteve-se positivo e estável em 63,3%. Houve reação adversa em 23% (7/30) dos pacientes. A única suspensão do primeiro biológico foi por evento adverso não grave (hipersensibilidade cutânea). No seguimento do DAS28-VHS houve uma redução significativa do valor médio inicial de 4,3 para 3,3 (p:0,002) após 24 semanas de tratamento, sendo que 60% dos pacientes estavam em remissão ou baixa atividade da doença, 23,3% em atividade moderada e 16,7% em atividade intensa. O valor médio do DAS28-VHS após uso de biológicos no grupo II foi significativamente menor do que no grupo I (p:0,042). CONCLUSÕES: 1) O tratamento com terapias biológicas no Estado de Mato Grosso foi eficaz no grupo prospectivo. A resposta no DAS28-VHS foi significativamente melhor no grupo onde o seguimento foi programado e o controle com exames de rotina e cálculo do índice de atividade da doença foram utilizados. 2) Dentre as características laboratoriais houve conversão significativa no perfil de auto-anticorpos, confirmando o potencial de imunogenicidade destas drogas induzindo a formação de auto-anticorpos, porém sem repercussão clínica no grupo estudado. O anti-CCP manteve-se estável em 63% dos pacientes. 3) As terapias biológicas foram seguras. Foram relatados eventos adversos não sérios. Não houve relato de infecção ou óbito. O motivo mais frequente de suspensão do primeiro biológico foi a perda de eficácia. 4) Os pacientes com artrite reumatoide em uso de terapias biológicas no Estado de Mato Grosso demonstraram satisfação com a prestação de serviços oferecida pela SES/MT sendo a maioria dos usuários da região metropolitana de Cuiabá. O programa de dispensação de terapias biológicas mostrou maior eficiência quando vinculado à um centro de referência específico para esse tipo de tratamento.
