Such methodologies must also be applied in the so-called Virtual Learning Environments. The aim of this work is to present a planning methodology based on known pedagogical theories which contributes to the incorporation of assessment in the process of teaching and learning. With this in mind, the pertinent literature was reviewed in order to identify the key pedagogical concepts needed to the definition of this methodology and a descriptive approach was used to establish current relations between this conceptual framework and distance education. As a result of this procedure, the Contents Map and the Dependence Map were specified and implemented, two teaching tools that promote the planning of a course by taking into account assessment still in this early stage. Inserted on Moodle, the developed tools were tested in a course of distance learning for practical observation of the involved concepts. It could be verified that the methodology proposed by the above-mentioned tools is in fact helpful in course planning and in strengthening educational assessment, placing the student as central element in the process of teaching and learning / Com a expans?o da Educa??o a Dist?ncia, promovida principalmente pelos novos recursos tecnol?gicos, existe atualmente uma procura por ferramentas e metodologias que possibilitem trabalhar o ensino-aprendizagem, de acordo com teorias pedag?gicas consolidadas e que possam ser inseridas nos Ambientes Virtuais de Aprendizagem. This study is a qualitative research project classified as practical and participatory action research designs, which has as its aim the creation of an instructional design, used as a learning metric for Domus Software - Procel Edification, which integrates concept maps and the revised Bloom taxonomy into an intelligent web assessment system. This metric aligns curriculum concept maps along with conceptual and procedural knowledge of the Domus software - Procel edifies the retaining cognitive processes, provides understanding and application, through offering a model of instructional design, which assigns weights to those cognitive processes attained by students and identifies principles to be used in its applicability for the evaluation of distance learning. The authors also present the results for alignment, inferring weights as well as an outline of the logical sequence along with steps for the implementation of the intelligent system through the association of some exemplification slides. / O presente trabalho é uma pesquisa qualitativa e classificada como practical and participatory action research designs (desenho de pesquisa de prática e ação participativa), que tem como intuito criar um desenho instrucional para uma métrica da aprendizagem do Software Domus Procel Edifica, integrando mapas conceituais à taxonomia revisada de Bloom em um sistema inteligente de avaliação na Web. Essa métrica alinha mapas conceituais curriculares, conhecimentos procedimentais e conceituais do software Domus − Procel Edifica aos processos cognitivos de retenção, entendimento e aplicação, oferecendo um modelo de desenho instrucional, que atribui pesos aos processos cognitivos alcançados pelos estudantes e identifica alguns princípios para sua aplicabilidade na avaliação da aprendizagem a distância. Divided into two parts, this study includes first, an approach around the literary criticism, which has been focusing on the relationship between the biblical text and the universe of literary studies. Of the number of experts who have devoted their lives to studying it, we have selected two important names of the literary criticism international in the 20th century, that have stood on this interaction in perspective that we can say of complementary and rhizomatic as the postulates of epistemology proposed by Gilles Deleuz and Félix Guattari. They are the Canadian Northrop Frye and the American Harold Bloom. Of both, the latter is the one that more intensely established the dialog between the Jewish-Christian book and the Literature. From the intriguing conclusions of these theorists, we opened for a second time that establishes the relationship between the thoughts of these experts, with emphasis in Bloom, and the lusophone fiction of three Romanesque clippings: A mulher que escreveu a Bíblia, by Moacyr Scliar; Caim, by José Saramago and Grande sertão: veredas, by João Guimarães Rosa. These works are analyzed in inter-textual dialogs with other literary representations national and international, that include the relationship between the Bible and the Literature in an approach of hypertextuality of conceptualization by Gerard Genette, that in turn converses with considerations of Linda Hutcheon alluding to studies on the parody. As a methodology, we are employing the bibliographic research relating to the object of research relevant to the thesis. As a result of our research, we conclude that there is a plentiful cartography, that this work is an example, which contradict the existence of biblical-literary rhizome, which leads to the proposition that the Bible, one of the most important artistic codes of Western culture, it is a work that can be seen as a literary piece as advocates Harold Bloom, without prejudice of the rhizome that links it to the religiosity, working as one of the most intrigu ing hypo-texts of Western Literature, which can also be researched on a level of high erudition in his hypertextuality. / A presente tese objetiva apresentar, numa perspectiva epist emológica e ficcional, a relação entre a Bíblia e a Literatura. Dividido em duas partes, este estudo contempla primeiramente uma abordagem em torno da crítica literária que tem se debruçado sobr e a relação entre o texto biblista e o universo dos estudos literários. Do conjunto de estudiosos que se dedicaram a estudá-la, pinçamos dois importantes nomes da crítica literária internacional no século XX que se detiveram sobre esta interação em perspectiva que se pode dizer complementar e rizomática, conforme os postulados da epistemologia proposta por Gilles Deleuze e Félix Guattari. São eles o canadense Northrop Frye e o norte-americano Harold Bloom. De ambos, Harold Bloom é o que mais intensamente estabeleceu o diálogo entre o livro judaico -cristão e a Literatura. A partir das instigantes ilações destes teóricos, abrimo s para um segundo momento que estabelece a relação entre o pensamento desses estudiosos, com ênfase em Bloom, e a ficcionalidade lusófona de três recortes romanescos: A mulher que escreveu a Bíblia, de Moacyr Scliar; Caim, de José Saramago e Grande sertão: veredas, de João Guimarães Rosa. Estas obras são analisadas em diálogos intertextuais com outras representações literárias nac ionais e internacionais que contemplam a relação entre a Bíblia e a Literatura numa abordagem de hipertextualidade da conceituação de Gérard Genette, que por sua vez dialoga com as considerações de Linda Hutcheon alusivas aos estudos sobre a paródia. Como metodologia, servimo -nos da pesquisa bibliográfica relativa ao objeto de pesquisa pertinente à tese. Como resultado de nossa pesquisa, chegamos à conclusão de que há uma farta cartografia, de que este trabalho constitui um exemplo, que referenda a existência do rizoma bíblico-literário, o que conduz à proposição de que a Bíblia, um dos mais importantes códigos artísticos da cultura Ocidental, é uma obra que pode ser vista como uma peça literária, como advoga Harold Bloom, sem prejuízo algum do rizoma que a vincula à religiosidade, funcionando como um dos mais instigantes hipotextos da Literatura no Ocidente, passível de ser também pesquisado em um nível de alta erudição em sua hipertextualidade.

Estudo do transcriptoma na síndrome de Bloom / Transcriptome study in Bloom\'s syndrome

A síndrome de Bloom (SB) é uma síndrome de instabilidade cromossômica rara, transmitida por herança autossômica recessiva. Caracteriza-se por deficiência de crescimento pré e pós-natal, microcefalia, hipoplasia malar, eritema telangiectásico em face e comprometimento do sistema imunológico, entre outras manifestações clínicas. Os pacientes com SB apresentam predisposição aumentada para o desenvolvimento de neoplasias em idade precoce, sendo esta, a principal causa de óbito. Ao estudo citogenético observa-se aumento de quebras cromossômicas espontâneas e trocas entre cromátides irmãs (TCI), que são utilizadas como marcador diagnóstico para a SB. Além disso, a literatura mostra que a maioria dos pacientes também apresenta mutações no gene BLM, que estão relacionadas a defeitos no mecanismo de reparo do DNA. No entanto, os mecanismos fisiopatológicos não são completamente compreendidos. Nesse sentido estudamos o transcriptoma de duas pacientes portadoras da síndrome de Bloom e de três controles utilizando a metodologia RNA-seq (Illumina, Inc., San Diego, CA). A análise de expressão diferencial revelou 216 genes diferencialmente expressos relacionados a vias relacionadas à resposta imune como replicação negativa da regulação da replicação do genoma viral, regulação positiva da proliferação de células B, via de sinalização mediada por interferon gama, ativação de células B, resposta a vírus, resposta imune adaptativa e processo efetor imune, e nenhuma diferença da expressão em genes de reparo de DNA. Concomitantemente, observamos a hiperexpressão do gene BLM para ambas as pacientes, contribuindo para a desestabilização de genes envolvidos em vias imunológicas, fenômeno também observado em alguns tumores. Dessa forma, sugerimos que a combinação de defeitos de proliferação linfocitária e defeitos de sinalização celular somados a outros, como perda celular e expressão alterada do gene BLM, podem contribuir diretamente para as principais características observadas na síndrome de Bloom, como a deficiência de crescimento e o elevado risco de câncer. Futuramente, o estudo do transcriptoma, aplicado a outros portadores da SB e outras síndromes de instabilidade, possibilitará uma análise mais acurada das interações gênicas relevantes para a desestabilização do genoma / Bloom Syndrome (BS) is a rare chromosome instability syndrome, with recessive autosomal inheritance. The main clinical manifestations are pre and postnatal growth deficiency, microcephaly, malar hypoplasia, telangiectasic facial erythema and compromised immune system, among others. Patients with BS present increased risk to the development of neoplasias at an early age, which is the main cause of death. Cytogenetic test is used as a diagnostic marker for BS since the patient\'s cells present increase in spontaneous chromosomal breaks and sister chromatid exchange (SCE). In addition, the literature reveals that most patients also present mutations in the BLM gene, which are related to defects in the DNA repair mechanism; however, it is still not completely understood. In this sense, we studied the transcriptome of two patients with Bloom\'s syndrome and three controls using the RNA-seq methodology (Illumina, Inc., San Diego, CA). Differential expression analysis revealed 216 differentially expressed genes related to immunological pathways such as: negative replication of the regulation of the viral genome replication, positive regulation of B cells proliferation, gama-interferon mediated signalization pathway, B cells activation, virus response, adaptive immune response and immune effector process, and absence of difference of DNA repair genes expression. At the same time, we observed the hyperexpression of the BLM gene for both patients contributing for the destabilization of genes involved in immunological pathways, a phenomenon also observed in some tumors. Thus, we suggest that the combination of lymphoid proliferation defects and cell signaling defects added to others such as cell loss and altered expression of the BLM gene may contribute directly to the main characteristics observed in Bloom\'s syndrome, such as growth failure and high risk of cancer. In the future, the study of the transcriptome applied to other BS carriers and other instability syndromes, will allow a more accurate analysis of the relevant gene interactions to the destabilization of the genome

Approximate Distributed Set Reconciliation with Defined Accuracy

Mit aktuell vorhandenen Mitteln ist es schwierig, objektiv approximative Algorithmen zum Mengenabgleich gegenüberzustellen und zu vergleichen. Jeder Algorithmus kann durch unterschiedliche Wahl seiner jeweiligen Parameter an ein gegebenes Szenario angepasst werden und so zum Beispiel Bandbreiten- oder CPU-optimiert werden. Änderungen an den Parametern gehen jedoch meistens auch mit Änderungen an der Genauigkeit bei der Erkennung von Differenzen in den teilnehmenden Mengen einher und behindern somit objektive Vergleiche, die auf derselben Genauigkeit basieren. In dieser Arbeit wird eine Methodik entwickelt, die einen fairen Vergleich von approximativen Algorithmen zum Mengenabgleich erlaubt. Dabei wird eine feste Zielgenauigkeit definiert und im Weiteren alle die Genauigkeit beeinflussenden Parameter entsprechend gesetzt. Diese Methode ist universell genug, um für eine breite Masse an Algorithmen eingesetzt zu werden. In der Arbeit wird sie auf zwei triviale hashbasierte Algorithmen, einem basierend auf Bloom Filtern und einem basierend auf Merkle Trees angewandt, um dies zu untermauern. Im Vergleich zu vorherigen Arbeiten zu Merkle Trees wird vorgeschlagen, die Größe der Hashsummen dynamisch im Baum zu wählen und so den Bandbreitenbedarf an die gewünschte Zielgenauigkeit anzupassen. Dabei entsteht eine neue Variante des Mengenabgleichs mit Merkle Trees, bei der sich erstmalig die Genauigkeit konfigurieren lässt. Eine umfassende Evaluation eines jeden der vier unter dem Genauigkeitsmodell angepassten Algorithmen bestätigt die Anwendbarkeit der entwickelten Methodik und nimmt eine Neubewertung dieser Algorithmen vor. Die vorliegenden Ergebnisse erlauben die Auswahl eines effizienten Algorithmus für unterschiedliche praktische Szenarien basierend auf einer gewünschten Zielgenauigkeit. Die präsentierte Methodik zur Bestimmung passender Parameter, um für unterschiedliche Algorithmen die gleiche Genauigkeit zu erreichen, kann auch auf weitere Algorithmen zum Mengenabgleich angewandt werden und erlaubt eine objektive, allgemeingültige Einordnung ihrer Leistung unter verschiedenen Metriken. Der in der Arbeit entstandene neue approximative Mengenabgleich mit Merkle Trees erweitert die Anwendbarkeit von Merkle Trees und wirft ein neues Licht auf dessen Effektivität. / The objective comparison of approximate versioned set reconciliation algorithms is challenging. Each algorithm's behaviour can be tuned for a given use case, e.g. low bandwidth or computational overhead, using different sets of parameters. Changes of these parameters, however, often also influence the algorithm's accuracy in recognising differences between participating sets and thus hinder objective comparisons based on the same level of accuracy. We develop a method to fairly compare approximate set reconciliation algorithms by enforcing a fixed accuracy and deriving accuracy-influencing parameters accordingly. We show this method's universal applicability by adopting two trivial hash-based algorithms as well as set reconciliation with Bloom filters and Merkle trees. Compared to previous research on Merkle trees, we propose to use dynamic hash sizes to align the transfer overhead with the desired accuracy and create a new Merkle tree reconciliation algorithm with an adjustable accuracy target. An extensive evaluation of each algorithm under this accuracy model verifies its feasibility and ranks these four algorithms. Our results allow to easily choose an efficient algorithm for practical set reconciliation tasks based on the required level of accuracy. Our way to find configuration parameters for different, yet equally accurate, algorithms can also be adopted to other set reconciliation algorithms and allows to rate their respective performance in an objective manner. The resultant new approximate Merkle tree reconciliation broadens the applicability of Merkle trees and sheds some new light on its effectiveness.

XSiena: The Content-Based Publish/Subscribe System

Just as packet switched networks constituted a major breakthrough in our perception of the information exchange in computer networks so have the decoupling properties of publish/subscribe systems revolutionized the way we look at networking in the context of large scale distributed systems. The decoupling of the components of publish/subscribe systems in time, space and synchronization has created an appealing platform for the asynchronous information exchange among anonymous information producers and consumers. Moreover, the content-based nature of publish/subscribe systems provides a great degree of flexibility and expressiveness as far as construction of data flows is considered. However, a number of challenges and not yet addressed issued still exists in the area of the publish/subscribe systems. One active area of research is directed toward the problem of the efficient content delivery in the content-based publish/subscribe networks. Routing of the information based on the information itself, instead of the explicit source and destination addresses poses challenges as far as efficiency and processing times are concerned. Simultaneously, due to their decoupled nature, publish/subscribe systems introduce new challenges with respect to issues related to dependability and fail-awareness. This thesis seeks to advance the field of research in both directions. First it shows the design and implementation of routing algorithms based on the end-to-end systems design principle. Proposed routing algorithms obsolete the need to perform content-based routing within the publish/subscribe network, pushing this task to the edge of the system. Moreover, this thesis presents a fail-aware approach towards construction of the content-based publish/subscribe system along with its application to the creation of the soft state publish/subscribe system. A soft state publish/subscribe system exposes the self stabilizing behavior as far as transient timing, link and node failures are concerned. The result of this thesis is a family of the XSiena content-based publish/subscribe systems, implementing the proposed concepts and algorithms. The family of the XSiena content-based publish/subscribe systems has been a subject to rigorous evaluation, which confirms the claims made in this thesis.

The dissertation "Modern Israeli short story" focuses on the form of the current short prose studied on the work of four of the renowned Israeli contemporary authors. Employing the method of text comparison we try to illustrate the specifics of the modern Israeli short story. The dissertation contains both the theoretical overview as well as analytical material. The authors were selected according to several criteria: two male and two female writers which enables us to determine whether "gender" plays a role - meaning if each author favors heroes of a specific gender or if they focus only on "female" or "male" contents. We also chose two native Israeli writers and two authors of foreign descent that immigrated to Israel which enabled us to study the linguistic angle and to realize whether the writer's background is reflected in his/her work.
