Evaluation of oil cakes from Amarula (Sclerocarya birrea), Macadamia (Integrifolia) and Baobab (Adansonia digitate L.) as protein supplements for ruminant diets

The current research was done to evaluate the nutritive values and the ruminal degradation of dry matter (DM) and crude protein (CP) from three non-conventional oil cakes, viz: amarula (Sclerocarya birrea) (AOC), macadamia (Integrifolia) (MOC) and baobab (Adansonia digitate L.) (BOC). The oil cakes were collected from biodiesel producers in Limpopo Province, transported to the ARC-Animal Production campus, where proximate and ruminal nutrient degradation analysis were conducted. Triplicates samples from each oil cake were analyzed for the nutritive values, mineral and amino acids contents. Three rumen cannulated mid-lactating (days in milk; DIM: 180±5) Holstein cows weighing 667±43 kg body weight were allocated to determine the in situ ruminal dry matter (DM) and crude protein (CP) degradation. The cows were offered a totally mixed ration (TMR) (60 concentrate: 40 forage ratio) that was compounded according to their daily nutrient requirements, and were milking was done twice per day at 12 hrs intervals. The three oil cake samples were ground using a 2-mm screen after which sub-samples (6.5 g) were put in 10 x 20 cm; 50 μm pore size polyester bags to achieve 15 mg/cm² (ratio of the sample size to surface area). The bags were then fistulated in each cow’s rumen in triplicate for a period of 2, 4, 8, 16, 24, or 48 hrs. After being incubated, the bags were removed from the rumen and washed with cold (4°C) water in 20-L buckets. Following immersing in cold water, the bags were machine washed until clean water was obtained. The bags were then dried at 60 °C in an oven for 48 hrs. The dried bags were individually weighed, and the content of each bag were removed and stored into glass vial until analysis. The remaining two duplicate sets of each sample were rinsed using cold water in order to determine solubility at 0 hrs. The AOC had higher (P<0.05) ether extract (EE) and CP content than both BOC and MOC. Macadamia oilcake (MOC) and BOC had higher (P<0.05) fractions of fibre (NDF, ADF and ADL) compared to the AOC. The AOC had greater (P<0.05) content of essential amino acids than in the BOC and MOC. Additionally, AOC had a high (P<0.05) phosphorus, but low calcium and potassium concentration. While AOC had high effective degradability of DM, it also had high water soluble as well as DM and CP rapidly degradable fractions. Effective degradation of CP was higher in AOC and BOC than in MOC. However, BOC had a high insoluble but degradable fraction of CP. Oilseed meals as dietary protein sources for juvenile Nile tilapia (Oreochromis niloticus L.)

One of the major problems facing aquaculture in Ghana is the non-availability of quality and affordable fish feeds. The present study investigated the nutritional suitability and cost-effectiveness of some Ghanaian oilseed by-products, soybean meal (Glycine spp), cottonseed meal (Gossypium spp), groundnut cake (Arachis hypogaea L.) and groundnut husk, as alternative protein sources to fishmeal (FM) in the diet of Nile tilapia (Oreochromis niloticus L.). The oilseed meals were used individually, as mixtures, as mixtures enriched with methionine and mixtures detoxified by heat processing (autoclaving) and/or addition of supplements (viz. phytase and ferrous sulphate) intended to reduce levels of the most important antinutritional factors (ANFs). Diets, containing the oilseed meals at inclusion levels from 25% to 75% dietary protein, were formulated to be isonitrogenous (320 g.kg-1), isolipidic (100 g.kg-1) and isoenergetic (18 KJ.g-1) and fed to juvenile Nile tilapia at 4-10% of their body weight for a period of eight weeks. Proximate analysis showed that soybean meal (SBM), cottonseed meal (CSM), groundnut cake (GNC) and groundnut husk (GNH) had 500.3, 441.4, 430.5 and 205.6 g.kg-1 crude protein, 38.2, 89.5, 12.8 and 89.2 g.kg-1 crude fibre and 20.19, 19.61, 23.17 and 22.18 kJ.g-1 gross energy respectively. Generally the oilseed meals had good essential amino acid (EAA) profiles with the exception of GNH. The EAA profile of SBM compared very well with FM but methionine and threonine were low (0.73 and 1.50 % of protein respectively) and the same was true for CSM and GNC with even lower levels. Analyzed ANFs in SBM, CSM, GNC and GNH were 17.54, 31.64, 14.86 and 3.99 g.kg-1 phytic acid, 14.09, 1.24 and 2.34 g.kg-1 trypsin inhibitors and 5.80, 6.50, 8.01 and 10.08 g.kg-1 saponin respectively and in CSM 5.6 g.kg-1 gossypol. Nutrient digestibility of these oilseed proteins suggested that Nile tilapia may be able to utilize SBM, CSM and GNC efficiently as dietary protein sources due to high apparent protein digestibility of 94.50%, 84.93% and 90.01% respectively. However, GNH may not be suitable because of very low apparent protein digestibility (27.67%). These protein sources when used individually were shown to cause depressed growth and feed efficiency when substituting more than 50% of the FM protein in diets. This may be attributed to high levels of ANFs, high fibre content and poor EAA profile. However, the use of mixtures of these meals was found to be marginally more effective than that of single sources. This may have been as a result of lower levels of ANFs and improvement in essential amino acid profile due to mixing. Supplementing the mixtures with methionine led to improvement in feed utilization but without significantly improving the nutritive value compared with FM. Heat processing was effective in reducing heat labile trypsin inhibitors in SBM, CSM and GNC by almost 80%, but not phytic acid and saponins, which remained virtually unaffected. Use of meals detoxified by heat processing with/without supplements at 50% inclusion improved growth and feed utilization compared to the unprocessed meals and performance was generally not significantly different from FM. Cost effectiveness analysis revealed that diets containing single feedstuffs or mixtures, particularly those containing equal proportions of oilseed meals and higher proportion of CSM replacing between 50% - 75% FM protein, were more profitable than FM diet. Similarly, the use of heat processed meals at 50% replacement of FM protein yielded greater profit than all other diets including the FM diet. However, essential amino acid supplementation of the meals was less profitable compared to the control. Generally, fish fed diets with oilseed meals would take longer to attain harvest size compared with FM and this could lead to an increase in production costs or a decrease in the number of production cycles which could be achieved within a year. It can be concluded that there is nutritional and economic justification for using SBM, CSM and GNC as partial replacement for FM in diets of Nile tilapia. Based on growth performance, nutrient utilization and economic benefits the diet with heat processed oilseed meal mixtures (containing equal proportions of 16.67% each) at 50% inclusion has the best prospects for replacing FM protein in diets of O. niloticus.

The injected water displaces oil to the producer wells and avoid reservoir pressure decline. However, suspended particles in the injected water may cause plugging of pore throats causing formation damage (permeability reduction) and injectivity decline during waterflooding. When injectivity decline occurs it is necessary to increase the injection pressure in order to maintain water flow injection. Therefore, a reliable prediction of injectivity decline is essential in waterflooding projects. In this dissertation, a simulator based on the traditional porous medium filtration model (including deep bed filtration and external filter cake formation) was developed and applied to predict injectivity decline in perforated wells (this prediction was made from history data). Experimental modeling and injectivity decline in open-hole wells is also discussed. Despite of the obstacles related to its use in the animal feed, there are evidences that this residue has great potential for biotechnology applications due to its arsenal of molecules. Thus, the present study aimed to purify and characterize a trypsin inhibitor from jatropha cake, in order to make a better use of this residue. The centesimal composition analysis showed to be the jatropha cake mainly composed of proteins (37.21%), fiber (34.26%) and lipids (19.16%). The crude extract obtained from the jatropha cake under alkaline condition (100 mM sodium borate buffer, pH 10.0) showed the presence of lectin (319.76 HU/gF), papain inhibitor (1,287.38 IU/gF), protease (2.69 AU/gF) and, especially, trypsin inhibitor (1,649.7 IU/gF). The trypsin inhibitor, named JcTI, was purified by fractionation of the crude extract with trichloroacetic acid (2.5%) followed by affinity chromatography (trypsin-sepharose-4B) and molecular exclusion (sephacryl S-200). JcTI is a glycoprotein (6.4% carbohydrates) with molecular mass in the range of 20-21 kDa, pI of 6.6, NH2-terminal sequence (VRDICKKEAERQDLSSCENYITQRRGY) showing identity around 60% with plant albumins and highly stable to heat, pH and salinity. JcTI (500 µg/mL) slowed the growth of important phytopthogenic fungi, including Colletotrichum gloeosporioides, Colletotrichum lindemuthianum, Fusarium oxysporum and Fusarium solani. This inhibitor also presented antibacterial activity against human pathogenic bacteria such as Bacillus subtilis, Salmonella choleraesuis and Staphylococcus aureus, with minimum inhibitory concentration less than 5 µg/mL. The results demonstrate the potential of JcTI for biotechnological application as a new defense protein against phytopthogenic fungi and human pathogenic bacteria. / O resíduo (torta) obtido após a extração de óleo das sementes de pinhão-manso (Jatropha curcas) para a produção de biodiesel é tóxico. Apesar dos entraves relacionados à sua utilização na alimentação animal, existem evidências de que esse resíduo tem grande potencial de utilização biotecnológica graças ao arsenal de moléculas que o constitui. Assim, o presente trabalho teve como foco a purificação e caracterização de um inibidor de tripsina da torta de pinhão-manso, vislumbrando um maior aproveitamento desse resíduo. A análise da composição centesimal revelou ser a torta de pinhão-manso composta principalmente por proteínas (37,21%), fibras (34,26%) e lipídios (19,16%). O extrato bruto obtido a partir da torta sob condição alcalina (tampão borato de sódio 100 mM, pH 10,0) mostrou a presença de lectina (319,76 UH/gF), inibidor de papaína (1.287,38 UI/gF), protease (2,69 UA/gF) e, principalmente, de inibidor de tripsina (1.649,7 UI/gF). O inibidor de tripsina, denominado JcTI, foi purificado do extrato bruto por fracionamento com ácido tricloroacético (2,5%) seguido de cromatografias de afinidade (tripsina-sepharose-4B) e de exclusão molecular (sephacryl S-200). JcTI é uma glicoproteína (6,4% de carboidratos) com massa molecular na faixa de 20-21 kDa, pI de 6,6, sequência NH2-terminal (VRDICKKEAERQDLSSCENYITQRRGY) exibindo similaridade em torno de 60% com albuminas vegetais e altamente estável ao calor, salinidade e pH. JcTI (500 µg/mL) retardou o crescimento de vários fungos fitopatogênicos de importância agrícola, incluindo Colletotrichum gloeosporioides, Colletotrichum lindemuthianum, Fusarium oxysporum e Fusarium solani. Esse inibidor também mostrou atividade antibacteriana frente a bactérias patogênicas ao homem, tais como Bacillus subtilis, Salmonella choleraesuis e Staphylococcus aureus, com concentração inibitória mínima inferior a 5 µg/mL. Os resultados obtidos demonstram o potencial do JcTI para aplicação biotecnológica como uma nova proteína de defesa contra fungos fitopatogênicos e bactérias patogênicas ao homem.

Resíduos agroindustriais como fonte de silício para a cultura do milho / Agroindustrial subproducts as silicon source to corn crop

The objective of this work to assess the supply of silicon for organic waste agribusiness, and to evaluate the influence of this nutrient in the accumulation of nitrogen (N), phosphorus (P), potassium (K), calcium (Ca), magnesium (Mg ) and sulfur (S) in maize. We analyzed the accumulation of silicon in the leaves and stems of the plant and the effect of these sources in stem diameter, the height and mass of dry corn plant, as well as the accumulation of N, P, K, Ca, Mg and S in corn leaves provided by agro-industrial residues and by levels of silicon used. The experiment was conducted in a greenhouse in a completely randomized design (4x3) + 1. We evaluated three sources organic agribusiness (bagasse ash from sugar cane filter cake rotary sugarcane and rice hulls), and for the supply of silicon also evaluated the mineral source (calcium silicate PA); three strengths of SiO2 (175 kg ha-1, 350 kg ha-1 and 700 kg ha-1) and additional treatment (control), which received no SiO2. The sources and doses of SiO2 did not affect the mass of dried corn plants. However, the addition of SiO2 dose provided an increase in the diameter of stem of the maize plant. The rice hulls was more efficient among all treatments in Si supply to the corn crop. The supply of Si favors the absorption and consequent accumulation of potassium and phosphorus from the corn plant. There was no relationship between the supply of Si and Mg and S uptake by corn plant. Finally, there was no relationship between the accumulation of Si and the reduction of leaf N in corn plants. / Os resíduos agroindustriais possuem elevado potencial para fornecimento de silício (Si) e outros nutrientes para as culturas agrícolas. Objetivou-se, com esse trabalho, avaliar o fornecimento de silício por resíduos orgânicos agroindustriais, e avaliar a influência desse nutriente no acúmulo de nitrogênio (N), fósforo (P), potássio (K), cálcio (Ca), magnésio (Mg) e enxofre(S) na planta de milho. Foi analisado o acúmulo de silício nas folhas e nos colmos da planta e o efeito dessas fontes no diâmetro do colmo, na altura e na massa da planta de milho seca, assim como o acúmulo de N, P, K, Ca, Mg e S nas folhas de milho proporcionado pelos resíduos agroindustriais e pelas doses de silício utilizadas. O experimento foi instalado em casa de vegetação no delineamento experimental inteiramente casualizado (4x3) + 1. Foram avaliadas três fontes orgânicas agroindustriais (cinza de bagaço de cana-de-açúcar, torta de filtro rotativo de cana-de-açúcar e casca-de-arroz carbonizada), e para o fornecimento de silício também avaliou-se a fonte mineral (silicato de cálcio P.A.); três dosagens de SiO2(175 Kg ha-1, 350 Kg ha-1 e 700 Kg ha1) e o tratamento adicional (testemunha), o qual não recebeu SiO2. As fontes e doses de SiO2 não influenciaram a massa das plantas secas de milho. Entretanto, o acréscimo das doses de SiO2 proporcionou o aumento no diâmetro de colmo da planta de milho. A casca-de-arroz carbonizada mostrou-se mais eficiente entre todos os tratamentos no fornecimento de Si para a cultura do milho. O fornecimento de Si favoreceu a absorção e consequente acúmulo de P e K pela planta de milho. Não se observou relação entre o fornecimento de Si e a absorção de Mg e S pela planta de milho. Por fim, não houve relação entre o acúmulo de Si e a redução do N foliar nas plantas de milho.

Para avaliação do híbrido Débora Victory, os tratamentos foram constituídos por nove épocas de amostragem, aos 2, 32, 46, 60, 77, 91, 109, 122 e 137 dias após o transplante (DAT) realizado em 01/04/2014. As plantas atingiram acúmulo estimado de massa seca e fresca total na última coleta de 550,2 e 9.528,2 g planta-1, respectivamente. A ordem decrescente do acúmulo total de nutrientes foi K > N > Ca > P > S > Mg, com valores estimados de 22,6; 10,4; 5,0; 2,3; 2,1 e 1,6 g planta-1, respectivamente. Na última coleta, o maior acúmulo de N, P, K ocorreu nos frutos e de Ca, S e Mg na parte vegetativa (caule+folhas+ inflorescências). O estudo da composição dos biofertilizantes foi constituído por seis biofertilizantes, elaborados com a mesma base para a fermentação (composta de 1 kg de farelo de arroz, 250 mL de melaço, 250 mL de microorganismos, marca EMBIOTIC, misturados em 15 litros de água), diferindo entre si pelo acréscimo de um ingrediente: T1 = torta de mamona (2 kg); T2 = farinha de sangue (2 kg); T3 = pó de casco e chifres (2 kg); T4 = farinha de ossos (2 kg); T5 = farinha de sangue (1 kg); T6 = farinha de ossos (1 kg) e cinco momentos de avaliação, aos 3, 7, 15, 22 e 30 dias após o preparo (DAP) dos biofertilizantes. Em cada momento foram analisados os teores de N, P e K e a condutividade elétrica. O biofertilizante a base de pó de casco e chifres apresentou maior teor de N do que os outros, alcançando o teor de 6,8 g L-1 aos 30 DAP. Os biofertilizantes a base de torta de mamona e pó de casco e chifres apresentaram maior teor de P, com médias de 3,2 e 2,6 g L-1, respectivamente, do que os de farinha de ossos (1 e 2 kg). O biofertilizante a base de torta de mamona apresentou maior teor de K, média de 2,5 g L-1, do que todos os demais, que foram iguais entre si. Independente do tipo de biofertilizante, os teores de K não se alteraram, os de N e a condutividade elétrica aumentaram e os de P reduziram ao longo do tempo de fermentação até os 30 DAP. / The objective of this research was to study the growth curve and absorption of nutrients in tomato 'Debora Victory', grown in organic system field in Itápolis-SP and to study the contents of N, P and K of six biofertilizers during the process fermentation. To evaluate the Débora Victory hybrid, the treatments consisted of nine sampling times at 2, 32, 46, 60, 77, 91, 109, 122 and 137 days after transplanting (DAT) held on 01.04.2014. The plants had estimated total dry and fresh weight, in the last sampling, of 550.2 and 9528.2 g plant-1, respectively. The descending order of total nutrient accumulation was K> N> Ca> P> S> Mg, with estimates of 22.6; 10.4; 5.0; 2.3; 2.1 and 1.6 g plant-1, respectively. In the last sampling, the greater accumulation of N, P, K occurred in fruits and Ca, Mg and S in the vegetative parts (stem + leaves + inflorescences). The experiment consisted of six biofertilizers, prepared on the same basis for fermentation (composed of 1 kg of rice bran, 250 ml of molasses, 250 ml of microorganisms, brand EMBIOTIC, mixed in 15 liters of water), differing from each other by an ingredient addition: T1 = castor bean cake (2 kg), T2 = blood meal (2 kg); T3 = hoof and horn powder (2 kg), T4 = bone meal (2 kg), T5 = blood meal (1 kg); T6 = bone meal (1 kg)) and five dates of evaluation: 3, 7, 15, 22 and 30 days after preparation (DAP) of biofertilizers. The following variables were analyzed: contents of N, P and K and the electrical conductivity. The hoof and horn powder biofertilizer showed higher N content than the others, reaching a maximum of 6.8 g L-1 at 30 DAP. The castor bean cake and hoof and horn powder, showed higher P contents, with average of 3.2 and 2.6 g L-1, respectively, than bone meal (1 and 2 kg) biofertilizer. The castor bean cake biofertilizer showed higher K content, average of 2.5 g L-1, than the others that did not differ each other. Regardless of the type of biofertilizer, the K content did not change, the N content and the electrical conductivity increased and the P content reduced during the time of fermentation until 30 DAP.

Several mechanisms, including straining, are responsible for particle retention and pore blocking causing formation damage and injectivity decline. Predicting formation damage and injectivity decline is essential in waterflooding projects. The Classic Model (CM), which incorporates filtration coefficients and formation damage functions, has been widely used to predict injectivity decline. However, various authors have reported significant discrepancies between Classical Model and experimental results, motivating the development of deep bed filtration models considering multiple particle retention mechanisms (Santos & Barros, 2010; SBM). In this dissertation, inverse problem solution was studied and a software for experimental data treatment was developed. Finally, experimental data were fitted using both the CM and SBM. This study had the objectives of: 1) screening plant parasitic nematodes in Zingiberales (Etlingera elatior, Zingiber spectabilis, Alpinia purpurata, Musa coccinea, Tapeinoquilos ananassae e Heliconia spp.) producing areas of Pernambuco, 2) predicting number of samples for nematodes monitoring at different acceptable error levels, 3) studying the effect of resistance abiotic inductors on nematode comunity associated with plant roots and soil, 4) evaluating enzymes activity in E. elatior, and 5) evaluating enzymes production in A. purpurata infected with Meloidogyne incognita. Samples (soil and roots of nematode parasited plants) were collected in producing areas from 2002 to 2004. Populational density of plant parasitic nematodes were evaluated and used to predict number of samples for monitoring. The study of induction resistance was carried out in one and two years old plants of a commercial area, fromSeptember to December in 2005. Acibenzolar-S-methyl (ASM), potassium silicate and neem cake were used as inductors, being analyzed free living and plant parasitic nematodes population density, area under nematode population density curve (AUNPDC), reproduction factor (RF), diameter of plant stem, β-1,3-glucanase and peroxidase activity. Enzymatic activity of esterase, phosphatase and peroxydase was evaluated in A. purpurata infected with M. incognita. Pratylenhus sp., Rotylenchulus sp., Meloidogyne spp., Helicotylenchus sp. and Criconemella sp. were frequents in most of samples. Symptoms of root-knot nematodes were verified in 100 % of the growing areas, being levels of Meloidogyne spp. considered high, up to 375 nematodes per 300 cm3 soil and 6090 nematodes per 20 g of roots, considering different areas and hosts. Among Meloidogyne species, M. incognita, M. arenaria and M. javanica were detected. Data analysis indicatedthat at least 11, 20, 10, 18 and 10 samples per area are recommended for monitoring the genus Pratylenchus, Rotylenchulus, Meloidogyne, Helicotylenchus and Criconemella, respectively, at 20 % error. The associations M. arenaria with E. elatior, Rotylenchulus sp. with A. purpurata, Heliconia spp. and M. coccinia, and Criconemella sp. with A. purpurata consisted of the first occurrence report in Pernambuco, Brazil. ASM was the best resistance inductor, reducing M. incognita population density, AUNPDC and FR with results similar to the nematicide control, but with no influence on free living nematodes in soil. Fitotoxic symptoms were not verified in any treatment. The action of ASM seems to be more related with peroxidases production than the β-1,3-glucanases activity in the plant. Among all of the inductors evaluated, ASM was the most effective inducing resistance against M. incognita in E. elatior. Enzymatic production was higher in A. purpurataparasited than in the controls. The acid phosphatase activity was high in contrast with the low esterase and peroxydase activity expressed in parasited plants. / O aumento expressivo na área plantada com ornamentais tropicais em Pernambuco reflete o crescente interesse pela floricultura no Estado. Este estudo teve por objetivos: 1) realizar levantamento de fitonematóides em áreas produtoras de Zingiberales em Pernambuco, 2) estimar o número de amostras recomendado para monitoramento, 3) avaliar os efeitos de indutores abióticos de resistência sobre a nematofauna associada à rizosfera, 4) avaliar a atividade de enzimas em plantas de bastão do imperador (Etlingera elatior), 5) analisar a produção de enzimas em plantas de alpínia (Alpinia purpurata) sob parasitismo de Meloidogyne incognita. No período de 2002 a 2004, foram visitadas 10 áreas produtoras de Zingiberales ornamentais (Etlingera elatior, Zingiber spectabilis, Alpinia purpurata, Musa coccinea, Tapeinochilos ananassae e Heliconia spp.), no estado de Pernambuco. Em cada propriedade, foram coletadas raízes de plantas parasitadas por fitonematóides e solo da rizosfera. Após processamento das amostras, foram determinadas as densidades populacionais dos gênerosde nematóides presentes e estimado o número recomendado de amostras para monitoramento, considerando diferentes níveis de erro aceitável. O estudo com indutores foi desenvolvido em plantio comercial, com plantas cultivadas por um e dois anos, no período de setembro a dezembro de 2005. Acibenzolar-S-metil (ASM), silicato de potássio e torta de nim foram utilizados como indutores. Foram analisadas a densidade populacional de nematóides parasitos e de vida livre, curva de densidade populacional dos nematóides, área abaixo da curva da densidade populacional(AACDP), fator de reprodução (FR), diâmetro do pecíolo e atividade das enzimas β-1,3-glucanase e peroxidase. No estudo de enzimas em plantas de alpínia, sob parasitismo de M. incognita foram determinadas as atividades da esterase, fosfatase ácida e peroxidase. No levantamento, constatou-se ocorrência freqüente dos gêneros: Pratylenhus, Rotylenchulus,Meloidogyne, Helicotylenchus e Criconemella. Sintomas da meloidoginose foram constatados em 100 % das áreas de plantio, com os níveis populacionais médios de Meloidogyne spp. elevados, alcançando 375 espécimes/300 cm3 de solo e 6090 espécimes/20 g de raízes, considerando diferentes hospedeiros e áreas. Entre as espécies de Meloidogyne foram detectadas M. incognita, M. arenaria e M. javanica. As estimativas do número de amostras indicaram que pelo menos 11, 20, 10, 18 e 10 amostras por área são recomendadas para monitoramento dos gêneros Pratylenchus, Rotylenchulus, Meloidogyne, Helicotylenchus e Criconemella, respectivamente, considerando um nível de erro de 20 %. As associações M. arenaria com E. elatior, Rotylenchulus sp. com A. purpurata, Heliconia spp. e M. coccinia, e Criconemella sp. com A. purpurata constituíram os primeiros relatos da ocorrência em Pernambuco. ASM reduziu a densidade populacional, AACDP e FR de M. incognita, apresentando resultados semelhantes aos obtidos pelo nematicida, destacando-se dos demais tratamentos, sem no entanto influenciar negativamente as populações de outros nematóides presentes no solo. Não foram verificados sintomas de fitotoxidez em nenhum dos tratamentos realizados. A ação do ASM mostrou-se mais relacionada com a produção de peroxidases do que com a atividade de β-1,3-glucanases em plantas de bastão do imperador. A produção de enzimas foi maior em A. purpurata parasitada do que na testemunha. A maior atividade foi verificada pela enzima fosfatase ácida, enquanto esterase e peroxidase expressaram fraca atividade em plantas parasitadas.

Foram realizados ensaios para determinação das isotermas de rendimento global, as curvas globais de extração (cinéticas de extração) e a identificação da composição química dos extratos obtidos. Na determinação da isoterma de rendimento global foram estudadas as pressões no intervalo de 100 a 350 bar com variações de 50 bar para as temperaturas de 30 e 50 ºC inicialmente para a matéria prima A. A quantidade utilizada de matéria prima (matéria prima A) foi de 2,5 gramas e uma vazão de CO2 de 0,249 ± 0,002 kg/h. Todos os ensaios foram realizados em duplicata e o tempo de extração de 60 min.. Para obtenção das curvas globais de extração foram utilizadas matérias primas distintas provenientes de 3 regiões do estado de São Paulo (matérias primas A, B e C). A temperatura de 50 ºC foi escolhida inicialmente para a matéria prima A, com as pressões de 200 bar e 350 bar respectivamente. Em seguida foi utilizado o binômio: temperatura e pressão, de 60 ºC e 200 bar para a matéria prima A, B e C sendo a temperatura escolhida pela característica do extrato de alta viscosidade (sólido à temperatura ambiente). A quantidade de matéria prima utilizada em cada ensaio foi de 25 gramas, uma vazão de 0,249 ± 0,003 kg/h de CO2, que fornece uma relação de 15 kg de CO2/kg de matéria prima. O tempo de extração utilizado para cada ensaio foi de 360 minutos. A partir do rendimento de extração, do tempo de extração do período CER (Constant Extraction Rate) e das características físicas da matéria prima (densidade, porosidade, etc.), foi estimada a capacidade dos extratores tomando-se o cuidado de dimensionar um extrator compatível com a capacidade de extratores comerciais existentes. Esta estimativa de capacidade permitiu a obtenção do custo do produto, a partir de uma análise econômica. A análise de sensibilidade do custo de produção para variações no custo de investimento, custo de insumos, custo de energia, custo de mão de obra e rendimento de extração, mostrou que o mesmo é influenciado principalmente pela variação no custo de investimento e pela variação do rendimento de extração. O custo de produção do extrato de cera foi estimado em US$ 13.626,93/tonelada / Abstract: The purpose of this work was to determine the manufacturing cost for the wax extract obtained from residues of the sugar cane and ethanol industry (filter cake), using SFE (Supercritical Fluid Extraction). Experiments were accomplished to determine the global yield isotherms, the overall extraction curves (kinetic of extraction) and to identification chemical composition of the extracts. The global yield isotherms were determined at the pressures of 100, 150, 200, 250, 300 and 350 bar and temperatures of 50 ºC and 30 ºC The amount of raw material (raw material A) used in each trial was 2.5 grams and a CO2 flowrate of 0.249 ± 0.002 kg/h. The experiments were duplicated. In order to determine the overall extraction curves (OEC), distinct raw materials were used from three different regions of São Paulo State (raw materials A, B and C). The OEC for the raw material A was initially determined at 50 ºC, with 200 bar and 350 bar of pressure respectively. In spite of 50 ºC be chosen initially, the experiments were accomplished in the SFE (Supercritical Fluid Extraction) equipment at 60 ºC and 200 bar for the raw material A, B and C being the temperature chosen due to characteristics of extract high viscosity (solid state at the ambient or room temperature). A 25 grams raw material sample was used in each trial with an outflow of 0,249 ± 0,003 kg/h of CO2 (a relation of 15 kg of CO2/kg of raw material). The extractors capacity was estimated from the extraction yield, the extraction time of Constant Extraction Rate period (CER) and the physical characteristics of the raw material (density, porosity, etc.). Extractors sizing considered the compatibility with existent extractors commercial capacity. This capacity estimation allowed the attainment of the product cost, from an economic analysis where investment cost, equipment cost, cost of operating labor, cost of operating supplies, etc. have been determined, composing the total extraction cost. The cost sensitivity was analyzed for variations in the investment cost, cost of operating supplies, energy cost, cost of operating labor and extraction income, showing that the production cost is influenced mainly by the variation in the investment cost and by the variation of the extraction yield. Valorisation chimique de la biomasse oléagineuse d’origine béninoise : Lophira lanceolata et Carapa procera / Chemical enhancement of the oleaginous biomass from Benin : Lophira lanceolata and Carapa procera

Lophira lanceolata (Ll) et Carapa procera (Cp) sont des plantes oléagineuses, peu étudiées. Au Bénin, elles sont pourtant utilisées à des fins alimentaires, cosmétiques et thérapeutiques. Cette étude vise la caractérisation de leurs graines, coques et bois. Les huiles végétales de Ll ont été obtenues par différentes méthodes dont une aqueuse traditionnelle tandis que celle de Cp l’a été par utilisation d’hexane. De façon générale, les huiles de Ll montrent un profil nutritionnel riche en acides gras polyinsaturés (>50% m/m: masse pour masse). Outre ses propriétés chimiques meilleures, celle obtenue par le procédé traditionnel est plus riche en acides gras essentiels, en composés phytostéroliques comme le lupéol et en tocols. La torréfaction et l’utilisation d’enzymes ont également permis d’évaluer l’impact de ces méthodes sur la composition chimique des graines de Ll. Quant aux graines de Carapa p., elles présentent un profil plutôt abondant en acides gras monoinsaturés, en tocotriénols (85,56% m/m) et en lanostérols (28,03%, m/m). Les tourteaux, coques et bois des deux espèces montrent une variabilité chimique en composés pariétaux (extractibles, hémicelluloses, celluloses et lignines). Une caractérisation in fine des hémicelluloses de ces parties des deux plantes a permis de montrer qu’elles sont essentiellement de type glucuronoxylanes. Les extractibles de ces plantes ont également offert une large gamme de composés à connotations industrielles et pharmaceutiques positives. Enfin, les conditions optimales de la biosorbption du bleu de méthylène sur les coques de Lophira ont également été évaluées. Cette évaluation a permis de mettre en exergue la potentielle utilisation de ces résidus agroforestiers pour rendre potables les eaux usées industrielles / The chemical composition of wild oilseeds, such as Lophira lanceolata (Ll) and Carapa procera (Cp) of Benin is mostly unknown. Yet they undergo crafted transformations for food, cosmetic and therapeutic purposes. This study aims to characterize their seeds, hulls and woods. From these crops, different oils have been extracted. One of them has been produced in rural area according to aqueous ancestral method. On the whole, oils of Ll have presented an interesting nutritional profile. They are rich in polyunsaturated fatty acids (> 50% m/m: mass for mass), especially that extracted by artisanal process. Beyond its good chemical properties, it provides essential fatty acids, phytosterols such as lupeol and more tocols compounds. Roasting and the use of enzymes have also assessed the impact of these methods on the chemical composition of LI seeds. Differently, Cp oil’s has an abundant presence of MUFA, tocotrienols (85.56% w/w) and the richest composition in lanosterol (28.03%, m/m). The seeds cakes, hulls and wood of both species showed various distributions on chemical components (extractives, hemicellulose, cellulose and lignin). The characterization of hemicelluloses from different parts of plants has shown that they are essentially glucuronoxylans type. Extractives also offered a wide range of compounds mostly appreciated for industrial and pharmaceutical purposes. The chemical composition of the shells of Lophira was rich in organic compounds such as lignin (32.13%, dry weight) so their biosorbent capacity was evaluated. They showed methylene blue good adsorption capacity in aqueous solution, which highlighted their potential use in the purification of wastewater
