Regulation of cholesterol intake by the corpus luteumMiranda, Leonor 04 1900 (has links)
Résumé
L’approvisionnement en cholestérol est un facteur limitant la stéroïdogenèse ovarienne. Pour cette raison, la majorité du cholestérol requis pour la synthèse des stéroïdes est importé de la circulation via les récepteurs des lipoprotéines de haute (HDL) et de basse densité (LDL) nommés scavenger receptor (SR-BI) et low-density lipoprotein receptor (LDLr). L’ARN messager de SR-BI est exprimé dans les ovaires de porcs durant toutes les étapes de la folliculogenèse ainsi que dans le corps jaune (CL). L’expression de la protéine SR-BI a également été détectée dans les follicules de souris lors du cycle œstral. Chez les deux espèces, l’expression est concentrée dans le cytoplasme et en périphérie des cellules du follicule. Les gonadotrophines induisent l'expression de SR-BI dans les cellules de la granulosa porcines, avec une expression cytoplasmique qui augmente durant la période périovulatoire, et avec une migration aux périphéries cellulaires durant la maturation du CL. Une conformation de 82 kDa de SR-BI est fortement exprimée dans le CL porcin, avec une conformation moins abondante de 57 kDa. Les différences entre les conformations sont attribuables à la glycosylation. La culture in vitro de follicules porcins avec des gonatrophines chorioniques humaines (hCG) a induit une hausse de régulation dépendante du temps du SR-BI de 82 kDa dans les cellules du granulosa. SR-BI et LDLr ont été exprimés réciproquement, avec LDLr étant le plus élévé dans les cellules folliculaires du granulosa et diminuant précipitamment avec la formation du CL. Pour explorer plus en détail les mécanismes d’approvisionnement en cholestérol de la stéroïdogenèse ovarienne, nous avons examiné des souris soumis à un traitement de désaccouplement de l'ovulation, et des souris portant la mutation nulle du gène Scarb1 (SR-BI-/-). Les résultats ont démontré que des ovocytes enfermés dans des structures lutéinisées expriment SR-BI. Les souris SR-BI-/ - présentaient de petits CLs, et de large follicules avec des cellules de thèque hypertrophiées et des kystes folliculaires avec des cavités remplies de sang et une diminution de 50% du niveau de progestérone dans le sérum. Les souris SR-BI-/ - traitées avec une combinaison de 20 g / g de mevinoline et 100 g / g de chloroquine ont démontré une diminution de 43% du niveau de progestérone sérique chez le type sauvage et de 30% chez les souris SR-BI-/ -. L’expression protéique de l’enzyme limitant pour la synthèse du cholestérol, 3-hydroxy-3-methyl-glutaryl-CoA reductase (HMGR), a augmenté chez les souris SR-BI-/-. Nous avons présenté des preuves démontrant que les cellules des follicules expriment le SR-BI durant la stéroïdogenèse et que la lutéinisation augmente l’expression de SR-BI. La maturation post-transcriptionelle est caractérisée par la glycosylation. Sous des conditions normales, l’expression de LDLr est arrêtée durant la lutéinisation. Ainsi SR-BI devient le facteur principal pour l’importation du cholestérol extracellulaire. En plus, la perturbation extracellulaire du cholestérol synthétisé de novo et l’absorption par les LDLr chez les souris SR-BI-/- diminuent la fonction lutéal. L’homéostasie du cholestérol ovarien est très importante pour une lutéinisation adéquate et sa perturbation mène à une réduction, mais non à un blocage complet, de la fonction lutéal. En conclusion, l’expression de SR-BI est un facteur important, mais non essentiel, pour maintenir l’homéostasie du cholestérol ovarien et la synthèse des stéroïdes, et la lutéinisation. Un réseau de mécanismes complémentaires et compensatoires d’approvisionnement en cholestérol agit en concert pour assurer la synthèse des stéroïdes ovariens. / Abstract
Ovarian cholesterol supply is rate limiting to ovarian steroidogenesis. For this reason, the majority of cholesterol required for steroid synthesis is imported via scavenger receptor-BI (SR-BI) and the low-density lipoprotein (LDL) receptor from circulating HDL and LDL. SR-BI mRNA is expressed in pig ovaries at all stages of folliculogenesis and in the corpus luteum (CL). SR-BI protein expression in mouse ovary during estrous cycle was also detected. In both species, expression is concentrated in cytoplasm and periphery of follicular cells. Gonadotropins induce SR-BI expression in pig granulosa cells, with cytoplasmic expression increasing through the periovulatory period, with migration to the cell periphery as the CL matured. An 82-kDa form of SR-BI is strongly expressed in the pig CL, with the less abundant 57-kDa form, differences between forms are attributable to glycosylation. In vitro culture of pig follicles with human chorionic gonadotropin (hCG) induced time-dependent upregulation of 82-kDa SR-BI in granulosa cells. SR-BI and LDL receptor were reciprocally expressed, with the latter highest in follicular granulosa cells, declining precipitously with CL formation. To further explore mechanisms of cholesterol supply to ovarian steroidogenesis, we examined mice treated to uncouple ovulation and mice bearing null mutation of the Scarb1 gene (SR-BI-/-). Results show entrapped oocytes in luteinized structures expressed SR-BI. SR-BI-/- mice displayed small corpora lutea, large follicles with theca cells hypertrophied, follicular cysts with blood filled cavities and 50% decreased in plasma progesterone. In SR-BI-/- mice, treatment with a combination of 20 g/g of mevinolin and 100 g/g of chloroquine (CHLORO) was employed to disturbed cholesterol sources. Serum progesterone was reduced by 43% in wild type and 30% in SR-BI-/- mice. The protein expression of the rate-limiting enzyme for cholesterol synthesis, 3-hydroxy-3-methyl-glutaryl-CoA reductase (HMGR) increased in SR-BI-/- mice. It was concluded that follicular cells express SR-BI during follicle development and luteinization causes upregulation of SR-BI expression. Posttranslational maturation is characterized by glycosylation. Under normal conditions expression of the LDLr (low density lipoprotein recepors) is extinguished during luteinization such that SR-BI becomes the principal means of importation of extracellular cholesterol. Further, perturbation of cholesterol de novo synthesis and uptake from LDLr in SR-BI-/- mice leads to a reduction of luteal function. Ovarian cholesterol homeostasis is central to adequate luteinization, and its perturbation leads to reduction, but not to complete impairment, of luteal function. We conclude that SR-BI expression is an important but not essential factor in maintaining ovarian cholesterol homeostasis, steroid synthesis and luteinization. A network of complementary and compensatory cholesterol supply mechanisms act in concert to assure ovarian steroid synthesis. / Studies were funded by Colegio de Postgraduados, México. 台灣高科技公司一直以來,在OEM/ODM領域有卓越的表現,隨著毛利逐步被壓縮之際,越來越多的我國廠商終於認知到,透過自有品牌(OBM)的建立是提升企業價值很重要的方式。但是,若要由OEM/ODM 跨入OBM,台灣廠商最缺乏的莫過於對「市場知識」的了解。本研究藉由對神達公司(近幾年內自有品牌Mio掌上型電腦/PDA上快速發展至全球前三大)的深入個案研究,探討「市場知識」的產生與能力建構之議題,並得到以下結論:
1、市場知識的來源似乎是很多元的;而對於剛採自有品牌策略之廠商的初期階段來說,似乎是較不完整的。
2、從事代工為主要業務型態的廠商,轉向發展自有品牌初期時,影響其市場知識來源的因素,受到內部因素(組織既有核心能力)影響及牽制的成分較多。
3、從事代工為主要業務型態的廠商,轉向發展自有品牌初期時,對其在取得完整市場知識的思考與運作上有所阻礙。儘管如此,組織既有核心能力對其所需要的部份市場知識仍有所助益。
4、企業在發展自有品牌的過程中,在取得市場知識與建構市場知識能力上似乎展現出「學習」的歷程。而在市場知識能力與行銷能力的建構上似乎又與高階主管的態度以及組織相關的管理有關。
5、除了本研究將「市場知識」定義為「關於顧客與競爭者的有系統、有意義且有用之知識」。從個案中發現,市場知識除了顧客與競爭者之外,其他環境因素也相當重要,例如政府的管制、社會文化的趨勢、科技的改變的趨勢等。
6、台灣資訊硬體製造能力,若能結合數位內容的創意,透過軟體與硬體的結合,似乎能創造更有價值和差異化的產品並創造更大的競爭優勢。 / Most of the high-tech corporations of Taiwan have remarkable performances in the OEM/ODM fields. Due to the compressing gross profit, more and more corporations realize that building own brand is a very important way to promote the enterprise value. However, if they want to make strides from OEM/ODM to OBM, the most deficient thing is the understanding of “Market Knowledge”. Through the case study of “MITAC international corp.”, we discuss the issue of the creation and the construction of ability of “Market Knowledge” and obtain the following conclusions.
1、The sources of “Market Knowledge” are diversified and they can be obtained by many different ways. For the new entered corporations in the OBM field, their sources of “Market Knowledge” are less complete at the initial period stage.
2、At the initial period stage of the OEM/ODM corporations in transformation to OBM, the factors which affect the sources of “Market Knowledge” are more affected and limited by the internal factors: the “Organizational Core Capabilities”.
3、At the initial period stage of the OEM/ODM corporations in transformation to OBM, they have the hindrance of the thinking and operation of obtaining the complete “Market Knowledge”. But, the “Organizational Core Capabilities” still can give some help in getting partial “Market Knowledge”.
4、In the process of obtaining “Market Knowledge” and establishing “Market Knowledge Capabilities”, there seems to be having some processes of “Learning”. And at the part of establishing “Market Knowledge Capabilities” and “Marketing Capabilities”, there seems to be having relationships with the senior managers’ attitudes and organizational management.
5、In this research, we define the meaning of “Market Knowledge” as the systematical, meaningful and useful knowledge which is about the customers and competitors. But from the case study, we find that “Market Knowledge” is not only about the customers and competitors, other environment reasons also have important impacts on it: the restrain of the government, the tendency of the society culture and the changes of technology.
6、If the OEM/ODM corporations can combine their hardware manufacturing capability with some contents of software, it seems a good way to them to create more values, more diversified products and more competition advantages.

The Sustainability Journey : An exploration into small and medium-sized enterprises' quest for legitimacy: the B Corp casevan Eck, Wiep, Kelly, Daniel January 2018 (has links)
This qualitative study contributes to the research field on legitimacy theory and creating shared value (CSV) in small and medium-sized enterprises (SMEs), regarding helping entrepreneurs and business owners better understand the journey of engaging with social and environmental issues. Recent years have seen a growing number of organisations engage in CSV, which builds on identifying societal needs and approaching these as business opportunities. A standard that purports to uphold these values and is comprised of for-profit companies committed to sustainability-related initiatives is the B Corp label. We consider B Corp certification and explore how sustainably oriented SMEs engage with their wider community in a way that fosters corporate credibility and legitimacy. Empirical data about five SMEs organisational narratives working with CSV, implementation of social and environmental activities, and effects of B Corp certification were collected through in-depth semi-structured interviews with company representatives and a range of secondary data materials. Analysing the data from these five interviewed companies established them to be engaging with the notion of CSV and regard addressing societal or environmental issues as the purpose of their business. Furthermore, findings suggested that values and beliefs incorporated by the SMEs supersede the generally adopted values in society, thereby representing a paradox with legitimacy theory. As a result, these businesses aim to close the perceived legitimacy gap by trying to raise awareness and engage the public with the appropriateness of their business, thereby slowly providing citizens with a more conscious mindset.

Seminario para optar al título de Ingeniero Comercial, Mención Administración / La inversión de empresas en mercados exteriores ha encontrado un escenario particularmente dinámico en el panorama mundial reciente. Avances de conectividad y globalización se han desarrollado en un período intermedio en la historia de expansión internacional de compañías, entre ellas conviven líderes, seguidoras, beneficiadas y afectadas por este contexto. Dentro del ambiente latinoamericano se encuentra Chile, representante de una valiosa experiencia e historia de empresas que proyectaron sus actividades a nuevos países, cuyos antecedentes muestran el porqué de la internacionalización, el dónde y cómo se refleja a través de sus decisiones. La balanza de IED de Chile a su vez transmite el efecto de estos desarrollos y que regionalmente pueden conectarse con la historia local.
Este trabajo se basa en el estudio de un grupo de empresas chilenas consideradas como grandes, catalogadas por expertos como exitosas o sobresalientes dentro del marco nacional-regional, con el interés de comprender y analizar su historia, pasado y factores que definen su presente, las formas de internacionalización que produjeron estos resultados, las semejanzas y diferencias que se pueden apreciar entre ellas y qué se puede aprender del fracaso y éxito al internacionalizarse desde Chile. Se seleccionó siete grandes empresas chilenas que comparten evaluaciones destacadas en la reseña y opinión especializada, así como rankings y artículos. Consecuentemente, los hallazgos mostraron puntos en común a priori por sus estrategias interpretadas y rubros, se pudo observar semejanzas antes de abordar el panorama internacional.
Para comprender el trasfondo de las decisiones de expansión, se realizó una comparación de la evidencia de empresas respecto a la teoría, esto con el fin de verificar el posible ajuste de similitud entre modelos de internacionalización y la realidad, que, de cumplirse ese ajuste, entonces se podría inferir la aplicación de esquemas para alcanzar éxito futuro. Los resultados de este trabajo indican que; en primer lugar, parte de los modelos propuestos y de conceptos poseen alto grado de ajuste con la forma de expansión de las empresas de la muestra; los hallazgos reflejaron alto nivel de correlación entre ingresos de actividades ordinarias de filiales extranjeras con los ingresos de actividades ordinarias de la empresa consolidada, retail presentó un mayor peso relativo de las ventas de filiales de mercados de Baja Distancia sobre sus ventas consolidadas, existieron diferencias estadísticamente significativas entre los ingresos generados por filiales de Baja Distancia comparado a las de Alta Distancia, la parte que más se asemejó a la realidad del modelo Uppsala es el concepto de distancia psíquica y proceso de destino. Se logró percibir la existencia de las fuerzas motoras de expansión internacional del modelo de motivación de entrada, fue posible clasificar las compañías bajo las definiciones existentes de cada estrategia internacional, se reconoció existencia de diferencias e imperfecciones de mercado, las compañías multinacionales de la muestra presentaron ventajas competitivas únicas e internalizadas que proveen un escenario beneficioso para enfrentar demanda y competencia en el exterior, y el modelo de marco de distancia tuvo un gran grado de ajuste a la realidad de la muestra.
En segundo lugar, existen otros factores cualitativos e internalizados a la compañía que condicionaron el éxito, a partir de los objetivos empresariales y estrategia es posible apreciar que el desempeño pudo ser propio bajo aquellas condiciones y no necesariamente explicarse completamente por la teoría disponible. En relación con lo anterior, se plantea que los resultados son propios de la muestra, la realidad de todas las grandes empresas chilenas requerirá de un mayor campo de investigación, las conclusiones sobre términos conceptuales como distancia psíquica que podría ser explicativa se remite a lo estudiado en este trabajo. Un tema interesante del estudio es considerar casos de híbridos en modelos, empresas que no pudieron calzar estrictamente con una definición teórica, sino más bien se acercaron más a un concepto que otro, por lo tanto, en la realidad las empresas observarían un comportamiento no restrictivo ni excluyente a modelos y definiciones teóricas.

Previous issue date: 2011-06-22 / I propose with this paper a reflection on the experiences contained in the creation of the body-
in-art (FERRACINI, 2006a, b) that originated the show Rosmaninhos... This process was
developed within the coletivo UZUME teatro from Jo?o Pessoa PB, through recreations and
resignifications of the corporeity and physicality contained in the steps, loas, aboios, songs
and choreography observed in the manners that Mestre Zequinha plays in his group of Cavalo
Marinho (Sea Horse), resident in the city of Bayeux - PB, and starting from the appropriation
of the text Hamlet of William Shakespeare. The body-in-art is understood in this work as a
vectorial body that dilates its daily functionality, recognizing a potential learning area capable
to generate creative escape lines that destabilize the "subject centered in an individuality and
identity" (FOUCAULT apud FERRACINI, 2006b, p.14), being open to the differentiation of
itself, indicating the possible existence of an itself-other and of the exchange-in-art space.
This process of construction of the body-in-art based on Master Zequinha s ways of playing
the Cavalo Marinho was methodically guided by the appropriation of the coletivo UZUME
teatro of the stages of Observation, Codification and Theatricalization contained in the
technique of corporal mimeses proposed by the LUME Teatro (Campinas - SP). That use
resulted in two phases: Active Observation and Composition of the body-in-art. Through the
repetition of these aesthetic matrixes of the Cavalo Marinho, the actors discovered actions
that when, codified and organized, can configure their body-in-art, which created a vectorial
exchange-in-art space to what was found in the Cavalo Marinho party. This search proposed
the means of potentiating the actors' work when it comes to a preparation that allowed to
dilate the scenic presence and stimulated the production of actions, which culminated in the
mounting of the show Rosmaninhos... / Proponho nesta disserta??o refletir sobre as experi?ncias contidas na cria??o dos corpos-em-arte (FERRACINI, 2006a, b) que originaram o espet?culo rosmaninhos.... 自工業革命以降,資本主義驅動著經濟與社會不斷前進,市場經濟所創造的交易效率幾乎滲透了人們生活的方方面面。但是使人類引以為傲的資本主義卻也創造了日益嚴重的貧富差距與自然環境的破壞,廠商犧牲了利害關係人的利益,極大化股東的利益,這促使我們開始思考是否有不同的方式可以同時創造合理的利潤,也同時善待組織的利害關係人?Fauchart 和 Gruber (2011) 所提出的使命型創業家很可能是一個新解,使命型創業家以改變社會為己任,期待藉由自身組織來改變社會與環境問題。然而,是什麼樣的原因促使使命型創業家出現?且使命型創業家是如何透過商業模式在使組織賺錢的同時增進利害關係人之效益?為了回答這個問題,本研究欲透過Shane (2000) 的前期經驗理論、使命型創業家之自我認同、利社會動機理論,與 Porter 和 Kramer (2011) 的共享價值理論來檢視個案公司。本研究選擇綠然能源作為研究個案,綠然能源是全台灣第一間由美國B Lab認證的B型企業 (B Corp),成立已有四年的時間,且已經開始獲利,此外,綠然能源之共同創辦人再加入此事業前,皆擁有豐富的商業經驗與獨特對社會與環境的生命體驗。研究者透過質性研究方式,以半結構式訪談、實地觀察法和次級資料搜集進行研究,以探究前期經驗、自我認同與利社會動機對個案公司創辦人之影響,以及個案公司如何發展出具有共享價值之價值主張。最後本研究提出以下發現,使命型創業家之自我認同、利社會動機與前期經驗是其創業行為的核心,並且這將影響組織發展共享價值之價值主張,而團隊成員豐富的前期經驗也有助於組織辨識機會,發展具共享價值之商業模式。 / After Industrial Revolution, Capitalism drives human society and economy forward constantly. The efficiency that market economy creates, has almost penetrated into every level of our daily lives. However, the powerful Capitalism has also done severe damage to environment, enlarging the gap between the rich and the poor. These phenomena cause us to think is there any possible a firm can earn a reasonable profit and treat its stakeholders well at the same time? The research on missionary entrepreneur of Fauchart and Gruber (2011) may become a solution to the problem we are facing. The missionary entrepreneur considers contributing to a better world is critical to them and it is their responsibility to do act for the social & environmental cause. Yet what makes one become a missionary entrepreneur? And, how does missionary entrepreneur develop a business model to solve the social & environmental problem and make profit as well? To answer these questions, the study aims to analyze the case company with the prior knowledge theory of Shane (2000), the self-identity of missionary entrepreneur (Fauchart & Gruber, 2011), prosocial motivation, and creating shared value (Porter & Kramer, 2011). The study takes DOMI as the case company, because DOMI is the first one certified B Corp in Taiwan. 4 years after DOMI was founded, now the company has already made profit from its business. Additionally, the co-founders all have diverse and multiple business and entrepreneurial experience. Plus, all of them had unique experience of social & environmental problems. The research is done by qualitative research method, and the research data is collected through semi-structured interview, field observation and secondary data analysis. In the end, the study shows that diverse prior knowledge of organization members will help organization recognizes opportunity. Besides, prior knowledge, self-identity, and prosocial motivation influence the missionary entrepreneur behavior, which can also enhance organization to create shared value in its value proposition.
