Estudo ultraestrutural e imunohistoquímico do estroma uterino durante a gestação de camundongos deficientes em decorim. / Ultrastructural and immunohistochemical studies of the uterine stroma in the decorin-deficient mice during pregnancy.

No presente estudo realizamos uma análise ultraestrutural e imuhistoquímica da organização das fibrilas de colágeno no endométrio durante a gestação de camundongos silvestres e deficientes em decorim. Os resultados mostraram que as fibrilas de colágeno em ambos os genótipos sofrem grande variação de forma e tamanho. Observou-se variação significante, na percentagem de distribuição dos diâmetros das fibrilas de colágeno existentes na região decidualizada em ambos os genótipos, porém foi maior nos animais Dcn-/-. Estes animais também apresentam maior percentagem de fibrilas finas quando comparados aos animais Dcn+/+. Observamos ainda que biglicam é expresso no endométrio não decidualizado dos animais Dcn-/-, no 3º dia de gestação. A expressão de lumicam mostrou-se nítida no estroma decidualizado e não decidualizado nos animais Dcn-/- no 7º dia de gestação e foi ausente nos animais Dcn-/-. Estes resultados mostraram que a ausência do decorim promove distúrbios no processo de agregação lateral das fibrilas espessas de colágeno. / The present study is an ultrastructural investigation into the organization of collagen fibrils in the pregnant endometrium of wild-type and decorin-deficient mice. Our results showed that collagen fibrils from both genotypes present a great variability of shape and size in cross section. Significant variation in the diameter of collagen fibrils exists in the decidualized endometrium from both groups of animals. In the decidualized endometrium, the diameter of collagen fibrils increases in both genotypes were higher in Dcn-/- than in Dcn+/+ animals. In the Dcn-/- animals the percentage of thin fibrils with diameter is also higher, when compared with Dcn+/+ animals. We also showed that Bgn is expressed in the non decidualized endometrium in the Dcn-/- animals, on day 3 of pregnancy. The expression of lumican showed a very sharp labeling in the decidualized stroma from day 7, and in the non decidualized estroma from Dcn-/- animals. These results suggest that the deficience of decorin may play a role on collagen fibrillogenesis in different stages of pregnancy.

INTRODUÇÃO: A matriz extracelular no endométrio fornece vasta gama de sinais envolvidos em diferentes processos celulares, tais como morte celular e proliferação. Neste sentido, os glicosaminoglicanos e os proteoglicanos, juntamente com os fatores de crescimento, modulam várias etapas da angiogênese, proliferação celular e remodelação do estroma, o que pode ser importante para o fluxo menstrual regular e a redução dos processos proliferativos. Além disso, são importantes para a adequada interação maternofetal. OBJETIVO: Avaliar a concentração de ácido hialurônico, das enzimas de biossíntese do ácido hialurônico - hialurônico sintases (HAS1, HAS2 e HAS3) e dos pequenos proteoglicanos ricos em leucina (decorim, biglicam, lumicam e fibromodulina) no endométrio de pacientes com síndrome dos ovários policísticos (SOP) e de mulheres eumenorreicas. MÉTODOS: Foram analisadas 20 amostras de endométrio, 10 provenientes de pacientes com SOP e 10 de mulheres com ciclos menstruais regulares na fase proliferativa do ciclo, com idade variando entre 20 e 35 anos, atendidas na Divisão de Clínica Ginecológica do Hospital das Clínicas da FMUSP (HC-FMUSP). A determinação do perfil e da concentração do ácido hialurônico foi efetuada por método bioquímico de ensaio fluorimétrico (ELISA-like). A sua localização no tecido endometrial, assim como as dosagens das enzimas sintases (HAS1, HAS2 e HAS3) e dos pequenos proteoglicanos ricos em leucina (decorim, biglicam, lumicam e fibromodulina e), foi feita por imunoistoquímica e \"western blotting\". Para a análise dos resultados foi utilizado o teste t de student (p<=0,05). Os cálculos foram realizados com auxílio do programa SPSS versão13 (SPSS, Chicago, IL). RESUTADOS: Houve maior concentração de ácido hialurônico no endométrio de mulheres eumenorreicas na fase proliferativa do ciclo menstrual do que no das com síndrome dos ovários policísticos. Com relação às sintases do ácido hialurônico, observou-se maior reatividade de HAS1 e HAS2 e menor reatividade de HAS3 no endométrio de mulheres com SOP em relação às mulheres com ciclos menstruais regulares na fase proliferativa. Quanto aos pequenos proteoglicanos ricos em leucina notou-se maior imunorreatividade de decorim e lumicam no endométrio de pacientes com SOP. CONCLUSÕES: As pacientes com SOP apresentam menor concentração de ácido hialurônico e maior reatividade ao decorim e lumicam em relação às mulheres com ciclos regulares na fase proliferativa. Additionally, it is important for proper fetal-maternal interactions. OBJECTIVE: Evaluate hyaluronic acid concentration, the enzymes of hyaluronic acid synthases (HAS1, HAS2 and HAS3) and small leucine-rich proteoglycans (decorin, lumican, fibromodulim and biglycan) in the endometrium of patients with polycystic ovary syndrome (PCOS) and eumenorrheic women. METHODS: A total of 20 endometrial samples from 10 patients with PCOS and 10 women with regular menstrual cycles in the proliferative phase, with ages ranging between 20 and 35 years, attended at Gynecology Division of Clinical Hospital of the FMUSP (HC-USP). Profile determination and the concentration of hyaluronic acid were performed by biochemical method of fluorimetric assay (ELISA-like). Its location in the endometrial tissue as well as the dosage of enzymes synthases (HAS1, HAS2 and HAS3) and small leucine-rich proteoglycans (decorin, lumican, fibromodulim and biglycan) was done by immunohistochemistry and western blotting. To analyze the results Student t test was used (p < 0.05). The calculations were performed with software SPSS version 13. RESULTS: A higher concentration of hyaluronic acid in eumenorrheic women endometrium in proliferative phase when compared with polycystic ovary syndrome. Regarding hyaluronic acid synthases, there was a higher HAS1 and HAS2 reactivity and lower HAS3 reactivity in the PCOS endometrium compared to women with regular menstrual cycles in the proliferative phase. Decorin and lumican showed higher immunoreactivity in PCOS endometrium. CONCLUSIONS: PCOS patients have a lower concentration of hyaluronic acid and greater reactivity to decorin and lumican than eumenorrheic women in proliferative phase. These data suggest that in patients with PCOS, the endometrium would be less receptive and have mechanisms to prevent excessive proliferation

Expressão gênica diferencial em tecido endometrial tópico e lesões endometrióticas / Genes differentially expressed in topic endometrium and endometriotic lesions.

Perdas gestacionais podem ser determinadas por vários fatores, dentre eles a endometriose, doença ginecológica comum caracterizada pela presença e crescimento de glândulas e estroma endometrial fora da cavidade uterina. Os principais sintomas da doença envolvem, além de problemas de fertilidade, dismenorréia, dispaneuria, dor pélvica crônica e irregularidades menstruais. Os locais mais freqüentes das lesões endometrióticas são o peritônio e os órgãos pélvicos, principalmente ovários. A incidência da endometriose é difícil de ser determinada devido à grande variabilidade de sintomas e à dificuldade para confirmação diagnóstica, que requer método cirúrgico. Estima-se que 15% da população feminina em idade reprodutiva seja afetada pela doença, sendo sua patogênese pouco conhecida. A hipótese mais aceita é a da menstruação retrógrada, onde os fragmentos endometriais descamados durante a fase menstrual seriam transportados através das tubas uterinas até a cavidade peritonial com implantação, crescimento e invasão local e de órgãos adjacentes. No entanto, apenas o refluxo não é suficiente para o estabelecimento da doença, sendo necessário que as células endometriais possuam certas características moleculares que favoreçam o aparecimento e a progressão da implantação ectópica. Vários estudos na literatura evidenciam que as principais divergências moleculares entre portadoras e não portadoras de endometriose estão relacionadas a processos envolvidos na apoptose, adesão celular, angiogênese, biosíntese de estrógeno, sistema imune, além de fatores de crescimento e metaloproteinases. Sendo assim, as pesquisas buscam a investigação de genes que se expressem diferencialmente (maior ou menor expressão) nas células de lesões endometrióticas por meio de várias técnicas. A hibridação subtrativa é uma metodologia de screening gênico que compara dois grupos celulares distintos, permitindo isolar moléculas de cDNA representantes do genoma de somente um dos dois grupos, pois remove as seqüências comuns entre eles. A técnica de hibridação subtrativa rápida (RaSH) simplifica e torna mais eficiente esse processo de subtração, identificando grande quantidade de seqüências diferencialmente expressas. Com objetivo de determinar possíveis marcadores moleculares para diagnóstico e tratamento da doença, aplicamos a técnica de RaSH para identificação de genes com expressão diferencial em 11 amostras de lesões endometrióticas (cinco de origem ovariana e seis de origem peritonial) e em 11 amostras de tecido endometrial tópico de mulheres sem a doença. Após análise dos dados referentes a 166 sequências de referência, os genes HTRA1, LOXL1, SPARC e SSAT foram selecionados para validação por meio da técnica de RT-PCR quantitativa em tempo real, sendo que apenas os dois primeiros mostraram diferença significativa entre os dois grupos estudados. Os genes HTRA1 e LOXL1 apresentaram expressão aumentada em lesões endometrióticas de pacientes afetadas, quando comparada aos níveis de expressão no tecido endometrial de mulheres não afetadas. Os resultados obtidos sugerem que os genes HTRA1 e LOXL1 podem ser considerados candidatos a marcadores moleculares para o diagnóstico. / Gestational losses can be determined by a number of factors including endometriosis, a common gynecological disease characterized by endometrial tissue found outside the uterine cavity. The main symptoms of the disease involve dysmenorrhea, dyspaneuria, chronic pelvic pain and menstrual irregularities, besides fertility problems. Endometriotic lesions are more frequently found in the peritoneum and in pelvic organs, mainly ovaries. Endometriosis incidence is difficult to be determined due to the wide variability of the symptoms and the difficult diagnostic confirmation, which requires a surgical method. Nevertheless, it is believed that around 15% of the female population in reproductive age is affected by the disease, although its pathogenesis remains unclear. The most accepted hypothesis is the retrograde menstruation, where endometrial fragments from the menstrual phase are transported through the uterine tubes to the peritoneal cavity, where they undergo implantation, growth, and adjacent tissues invasion. However, only reflux is not enough for the disease establishment, and it is necessary that endometrial cells present molecular characteristics favoring rise and progression of ectopic implantation. In the literature, a number of studies highlight that the main molecular divergences between women with and without endometriosis are related to processes involved in apoptosis, cellular adhesion, angiogenesis, estrogen biosynthesis, immunological system, growth factors and metalloproteinases. In doing so, a number of researches seek for the investigation of genes differently expressed (up or down regulated) in endometriotic lesions cells using a variety of methodologies. The subtractive hybridization is a gene screening methodology that compares two distinct cellular groups, allowing the isolation of cDNA molecules representative from the genome of only one of the two groups, because it removes common sequences among them. The rapid subtractive hybridization methodology (RaSH) makes the process of subtraction easier and more efficient, identifying a large amount of differently expressed sequences. In this study, RaSH was used to identify differently expressed genes in 11 samples of endometriotic lesions (five from ovarian origin and six from peritoneum origin), and in 11 samples from topic endometrial tissue from women without the disease to determine possible molecular markers for diagnosis and treatment. After data analysis related to 166 reference sequences, genes HTRA1, LOXL1, SPARC and SSAT were selected for validation using real time quantitative RT-PCR. Only HTRA1 and LOXL1 presented significant difference between the two studied groups. Genes HTRA1 and LOXL1 were up regulated in endometriotic lesions from affected patients when compared with the expression levels in the endometrial tissue of non-affected women. Our results suggest that the genes HTRA1 and LOXL1 can be considered candidate molecular markers for the diagnosis of endometriosis.

Efeito do ambiente endócrino peri-ovulatório na qualidade do ambiente uterino em vacas de corte: regulação no metabolismo das poliaminas, estresse oxidativo e proliferação celular / Effect of the periovulatory endocrine milieu on the uterine environment quality in beef cows: regulation on the polyamines metabolism, oxidative stress and cellular proliferation

O objetivo do presente trabalho foi comparar os efeitos dos distintos ambientes endócrinos peri-ovulatórios na proliferação celular, no metabolismo das poliaminas e no ambiente redox do útero bovino durante o diestro inicial. Para isso, controlou-se farmacologicamente o crescimento do folículo objetivando induzir a ovulação de folículos de maior diâmetro (grupo folículo grande-CL grande, FG-CLG) ou de menor diâmetro (grupo folículo pequeno-CL pequeno, FP-CLP). Trinta vacas multíparas Nelore, foram pré-sincronizadas, metade destes animais foram destinados para o grupo FG-CLG e receberam uma dose de prostaglandina F2&#945; (PGF) e um dispositivo de progesterona, juntamente com benzoato de estradiol no D10. No momento da retirada dos dispositivos de progesterona (entre D1,75 e D2,5) todos os animas receberam uma dose de PGF. A ovulação foi induzida com acetato de buserelina (D0). O que diferiu entre os tratamentos foi que os animais do grupo FP-CLP não receberam uma dose de PGF no D10 e o momento da retirada dos dispositivos foi entre D1,25 e o D1,5. No D7 um subgrupo dos animais foi abatido e amostras de endométrio e lavado uterino foram coletadas para as análises laboratoriais. Os animais do grupo FG-CLG apresentaram no D0 um folículo 1,2 vezes maior e estradiol 2,3 vezes maior em relação ao FP-CLP. Consequentemente, as vacas do FG-CLG desenvolveram CL maiores (1,6 vezes) e mais pesados (1,4 vezes) o que proporcionou no D7 concentrações de P4 1,5 vezes maior em relação ao FP-CLP. Primeiramente, uma análise do transcriptoma endometrial foi realizada e indicou que processos celulares como proliferação, composição de matrix extracelular, processos metabólicos e processos de oxirredução foram afetados pelo modelo experimental. Para avaliar a condição proliferativa e/ou apoptótica endometrial análises de imunomarcação foram realizadas (Ki-67, marcador de proliferação e caspase 3 ativada, marcador de apoptose). O número de células positivas para Ki-67 no epitélio luminal foi 4,1 vezes maior no FP-CLP, porém não houve diferença na marcação da caspase 3 ativada. No epitélio glandular tanto o ki-67 (1,4 vezes) como a caspase 3 ativada (2,6) estavam aumentados no grupo FG-CLG. Em relação à síntese das poliaminas não houve diferença entre os grupos nas concentrações das poliaminas e abundância dos transcritos e proteína das enzimas de síntese. Por outro lado, foi detectada uma forte associação entre a expressão do gene SAT1 com o gene ODC1 (r2: 0,73; P<0,01) e também entre os genes AZIN1 e AMD1 (r2: 0,61; P<0,01). Quanto ao ambiente redox não houve diferença entre os tratamentos no lavado uterino mas no endométrio, no grupo FP-CLP, houve redução da atividade das enzimas Catalase (0,5 vs 0,79 U/mg proteína; P<0,01), glutationa peroxidase (2,0 vs 2,43 nmol NADPH/min/mg proteína; P<0,01), aumento da atividade da Superóxido dismutase (44,77 vs 37,76 U; P=0,04) e da peroxidação lipídica (28,5 vs 17,43 nmol/MDA/mg proteína; P<0,001), mas sem alterar as quantidades da espécies reativas. De acordo com os resultados obtidos, pode-se concluir que o ambiente endócrino peri-ovulatório regula a proliferação endometrial mas sem alterar a via de síntese das poliaminas e modula também o ambiente redox uterino / This work aimed to compare the effects of the different periovulatory endocrine milieu on the several molecular pathways: as cellular proliferation, polyamine metabolism and redox environment of the bovine uterus during the early diestrus. For this, we controlled pharmacologically the follicular growth to induce the ovulation of larger follicle (Large Follicle-Large Corpus lutem group LF-LCL) or smaller follicle (Small follicle-Small Corpus luteum group SF-SCL). Thirty multiparous Nelore cows were synchronized. Fifteen cows were assigned for each group. On the LF-LCL, the cows received a dose of prostaglandin F2&#945; (PGF), a progesterone device and an estradiol benzoate on D10. On the time of progesterone device was withdraw (between D1.75 e D2.5) all the animals received a dose of PGF. The ovulation was induced with buserelin acetate (D0). The unique difference between the groups was that animals of SF-SCL did not received a dose of PGF on D10 and the progesterone device withdraw was between D1.25 e o D1.5. On D7 a subset of animals was slaughtered and the endometrial and uterine washings samples were collected for laboratorial analyses. On D0 the animals of LF-LCL had a 1.2 times larger follicles and 2.3 times higher levels of estradiol than animals of SF-SCL. Firstly, an endometrial transcriptome analyses was realized and suggested that several cellular processes as proliferation, metabolic processes and oxi-reduction processes were affected by experimental model. To evaluate the proliferative and/or apoptotic condition, endometrial immunohistochemistry analyses were realized (ki-67, proliferation marker and caspase 3 activated, apoptose marker). The number of Ki-67 positive cells in the luminal epithelium was 4.1 times higher on SF-SCL. However, there was no difference in caspase 3 activated analyses. On the glandular epithelium both ki-67 (1.4) and caspase 3 activated (2.6) were increased on LF-LCL. About polyamine synthesis pathway there was no difference in polyamines levels and gene and protein expression of synthesis enzymes between the groups. In other hand, it was detected a strong association between SAT1 gene expression and ODC1 gene expression (r2: 0.73; P<0.01) and also between AZIN1 and AMD1 gene expression (r2: 0.61; P<0.01). On the redox environment analysis there was no difference between the groups on the uterine washings but the SF-SCL group had lower endometrial catalase (0.5 vs. 0.79 U/mg protein, P<0.001) and glutathione peroxidase (GPx; 2.0 vs. 2.43 nmol NADPH/min/mg protein, P=0.04) activity, as well as higher lipid peroxidation (28.5 vs. 17.43 nmol MDA/mg of protein, P < 0.001) and superoxide dismutase (SOD) activity (44.77 vs. 37.76 U; P=0.04). There were no differences in the endometrial reactive species (RS) between the groups. In conclusion, the periovulatory endocrine milieu regulates the endometrial proliferation but without alterations in polyamine metabolism and also modulates the uterine redox environment in beef cattle

O mecanismo pelo qual a obesidade promove o aparecimento destas doenças ainda não está totalmente esclarecido. No entanto, sugere-se que a produção de estrogênios, decorrentes da conversão dos andrógenos no tecido adiposo e os efeitos metabólicos relacionados à hiperinsulinemia, mediados pelo IGF-1, possam estar relacionados. Embora se espere que o endométrio após a menopausa esteja atrófico, existe uma proporção que mantém padrão proliferativo fraco, provavelmente como resposta ao baixo e constante estímulo estrogênico. O objetivo deste estudo é avaliar a associação existente entre a expressão de receptores esteróides, IGF-1, Bcl-2 e Ki-67 no endométrio, correlacionando-os com índice de massa corporal e resistência tecidual insulínica. Sujeitos e Métodos: Foi realizado estudo de corte transversal, sendo selecionadas 85 mulheres após a menopausa, do Centro de Atenção Integral à Saúde da Mulher (Caism - Unicamp) e do Hospital Dr. José de Carvalho Florence (São José dos Campos-SP). Foram avaliados os receptores endometriais para estrogênio (RE), progesterona (RP), fator de crescimento insulinóide 1 (IGF-1), Ki-67 e Bcl-2 na amostra endometrial obtida correlacionando-os com a resistência tecidual insulínica (RI) e índice de massa corporal (IMC). Os dados foram analisados através dos testes de Pearson e de Spearman, considerando-se o valor ? de 0,05. Resultados: Obtivemos 36,5% de mulheres com sobrepeso e 40% com obesidade. Não foi encontrada correlação estatística entre IMC com variável estudada. Observou-se expressão moderada e intensa para os RE (94,1%), RP (80%) e IGF-1R (78,8%) nas glândulas endometriais. A expressão do Ki-67 foi moderada e intensa em 75,3% e Bcl-2 em 65,9% das amostras. O índice HOMA-IR correlacionou-se com os RE (p=0,0402) e RP (p=0,0034). Os IGF-IR e RP se correlacionaram com CA (p=0,0189 e 0,0170) respectivamente. Observou-se correlação entre RE, RP e Ki-67 e Bcl-2. Conclusão: Mulheres após a menopausa apresentam grande expressão endometrial dos receptores estrogênicos, progestogênicos e IGF-1, independentemente do seu peso corporal ou do índice de massa corporal. Houve correlação entre os RE, RP e IGF-1R com marcadores de resitência tecidual insulínica / Abstract: Introduction: After menopause, weight gain and the distribution of fat deposits may be altered with a predominance of abdominal fat, favoring the development of metabolic diseases and cancer. The mechanism by which obesity promotes the onset of these diseases is not fully understood. However it is suggested that the production of estrogens, resulting from the conversion of androgens in adipose tissue and metabolic effects associated with hyperinsulinemia, mediated by IGF-1, may be related. While is hoped that the postmenopausal endometrium is atrophic, there is a proportion that maintains low proliferative index, probably in response to low estrogen and constant encouragement. The aim of this study is to evaluate the association between the expression of steroid receptors, IGF-1, Bcl-2 and Ki-67 in the endometrium, correlating them with body mass index and insulin resistance tissue. Subjects and Methods: Was conducted cross-sectional study, were 85 selected women after menopause, in the Center for Integral Attention to Women's Health (Caism - Unicamp) and the Hospital Dr. José de Carvalho Florence (São José dos Campos-SP). We evaluated the endometrial receptors for estrogen (ER), progesterone (PR), IGF-1, Ki-67 and Bcl-2 in endometrial sample obtained by correlating them with tissue insulin resistance (IR) and body mass index (BMI). Data were analyzed using Pearson tests and Spearman, considering ? value of 0.05. Results: We obtained 36.5% of overweight women and 40% were obese. We don't found statistical correlation between body mass index with no variable studied. We observed moderate to intense expression for ER (94.1%) PR (80%) and IGF-1R (78.8%) in the endometrial glands. The expression of Ki-67 was moderate and severe in 75.3% and Bcl-2 in 65.9% of samples. The index of HOMA-IR was correlated with the ER (p = 0.0402) and PR (p = 0.0034). The IGF-IR and PR were correlated with CA (p = 0.0189 and 0.0170) respectively. There was a correlation between ER, PR and Ki-67 and Bcl-2. Conclusion: Women after menopause have a great expression of endometrial estrogen, progestin and IGF-1 receptors, independent of their BMI. Sujeitos e métodos: Realizou-se um estudo de corte transversal no qual 193 mulheres voluntárias do Ambulatório de Reprodução Humana, Departamento de Tocoginecologia, Faculdade de Ciências Médicas, CAISM - Unicamp foram avaliadas com biópsia de endométrio com Pipelle de Cornier. Os achados anatomopatológicos foram catalogados como endométrio normal, hiperplasia ou câncer de endométrio e estes resultados foram comparados com o ìndice de massa corporal (IMC; kg/m2). Para a análise dos dados, as mulheres foram divididas em dois grupos: em idade reprodutiva e na pós-menopausa e pelo IMC (kg/m2) em sobrepeso ou obesidade. Resultados: A prevalência de hiperplasia na amostra total foi de 7,3%, na idade reprodutiva de 5,8% e na pós-menopausa de 12,1%. A prevalência de adenocarcinoma de endométrio na amostra total foi de 1,5%, na idade reprodutiva de 1% e na pósmenopausa de 3%. A regressão logística multivariada mostrou que o risco de apresentar hiperplasia e câncer de endométrio foi de 1,19 (IC 95%; 0,36¿3,90) para as mulheres na pós-menopausa, de 1,58 (IC 95%; 0,30-8,23) para as que tinham obesidade severa e de 2,72 (IC 95%; 0,65¿11,5) para as que tinham obesidade morbida. Estar na idade reprodutiva, com sobrepeso ou obesidade moderada não alterou o risco. Conclusões: No grupo avaliado, mulheres assintomaticas na pós-menopausa com obesidade severa ou mórbida apresentaram risco aumentado de apresentar hiperplasia ou câncer de endométrio. O mesmo não ocorreu com mulheres em idade reprodutiva / Abstract: Objetives: To determine the prevalence of endometrial cancer and hyperplasia in non-symptomatic women with overweight or obesity. Subjects and methods: A cross-sectional study was carried-out at the Obstetrics and Gynecology Departament, School of Medicine, Unicamp, with 193 women. The women were evaluated through endometrial biopsy with a Pipelle de Cornier. The hystophatologic findings were classified as normal endometrium, hyperplasia and cancer and those results were compared to the body mass index (BMI; kg/m2). For the analysis pourpose the women were divided in two groups: at reproductive age and at the post-menopause and by the BMI (kg/m2) in overweigth and obesity. Results: The prevalence of hyperplasia in the total sample was 7.3%, among women at reproductive age of 5.8% and at the post-menopause it was of 12.1%. The prevalence of endometrial cancer at the total sample was 1.5%, among women at reproductive age it was 1% and at the post-menopause of 3%. The logistic regression showed that the risk of endometrial hyperplasia or cancer was of 1.19 (95% CI: 0.36¿3.90) for women at the post-menopause, 1.58 (95% CI: 0.30¿8.23) for those who presented severe obesity, and of 2.72 (95% CI: 0.65¿11.5) for those women with morbid obesity. Be at the reproductive age, with overweight or moderate obesity did not modify the risk. Conclusions: At the evaluated group of women, non-symptomatic women at the post-menopause with severe or morbid obesity presented a high risk to have endometrial hyperplasia or cancer.
