Dietary soy isoflavones have been shown to ameliorate insulin resistance and Type 2 diabetes. However, many in vitro studies used supra-physiological concentrations of individual isoflavones that make it difficult to interpret the results as potential mechanisms in vivo. Since the insulin-sensitizing effects of thiazolidinediones, anti-diabetic drugs, have been shown to be mediated through activation of peroxisome proliferators-activated receptor gamma (PPARγ), the key transcription factor for adipocyte differentiation, we examined the effects of the two main soy isoflavones genistein and daidzein either as individual compound or combined on adipocyte differentiation and PPARγ expression, as well as whether the Wnt/β-catenin signaling pathway is the underlying molecular mechanism. In 3T3-L1 cells, genistein and daidzein significantly enhanced adipocyte differentiation. Similarly the expression of PPARγ increased particularly at 20 µmol/L. The stimulatory effect is greater when the two isoflavones are combined, indicating a synergistic effect. Genistein and daidzein also increased the relative abundance of insulin-responsive glucose transporter 4 (GLUT4) mRNA with a greater effect when combined. Wnt10b expression was not affected by soy isoflavones treatments, while Wnt5b expression was only increased by the combination of genistein and daidzein. Our results suggest, that the combination of soy isoflavones has a greater effect in increasing the newly formation of adipocytes that are highly insulin-sensitive via an increase in PPARγ expression as well as increasing the expression of GLUT4. However, genistein and daidzein actions on Wnt signaling remain unclear. These data further support the epidemiological findings for the beneficial effect of soy consumption on insulin sensitivity.

Characterization of antioxidant activities of soybeans and assessment of their bioaccessibility after in vitro digestion

Nine Virginia soybeans grown in a single location were compared for their antioxidant properties and isoflavone profiles. The extracts were evaluated for their total phenolic contents (TPC), Oxygen Radical Absorbance Capacity (ORAC), and DPPH™ radical scavenging activities. In order to evaluate efficient preparation methods for soybean antioxidants, three Virginia-grown soybeans were extracted using different extraction strategies. The extraction techniques included soxhlet extraction, conventional solvent extraction, and ultrasonic-assisted extraction (UAE) with 5 different common solvent systems including 50% and 80% aqueous acetone, 50 and 70% aqueous ethanol, and 80% aqueous methanol. The TPC in the soybean extracts and isoflavone compositions were significantly different among cultivars. Malonylgenistin was the major isoflavone in all soybean seeds, accounting for 75-83% of the total measured isoflavones. The V01-4937 variety had the highest total isoflavone and malonylgenistin contents, followed by V03-5794. The antioxidant activities of the soybean extracts were also significantly different. Overall, the V01-4937 soybean was the variety that stood out from the other tested Virginia soybeans because it had the highest TPC, ORAC value, and isoflavone contents as well as the second highest DPPH™ scavenging activity. Ultrasonic treatment improved the extraction of soybean phenolics by more than 50% compared to solvent alone. The UAE with 50% aqueous acetone was the most efficient for extraction of phenolic compounds in the soybean seeds. The conventional and UAE with 70% aqueous ethanol extracts had the highest ORAC values, while the soxhlet methanol extracts had the highest DPPH™ radical scavenging activities. Our results suggest that different extraction technologies have a remarkable effect on soybean antioxidant estimation and the UAE is more appropriate for soybean phenolic extraction because it is less time and solvent consuming than the conventional solvent and soxhlet extractions. The V01-4937 soybean with the highest TPC was evaluated for its antioxidant activity and isoflavone contents in an in vitro digestive system. After gastrointestinal digestion, soybean extracts contained higher TPC and ORAC values than cooked soybean (before digestion) but they were relatively low in DPPH™ radical scavenging capacity. The glucosides, daidzin, genistin, and malonylgenistin showed stability during simulated digestion with 83.3 %, 59.4 %, and 10.7 % recovery, respectively. Aglycones, including daidzein and genistein, were recovered at 37 % and 73.7 %, respectively, after in vitro digestion. In this study, daidzin was the most stable and bioaccessible isoflavone determined using the in vitro digestive system. Among the aglycones, genistein was more stable and bioaccessible than daidzein after digestion. In conclusion, soybean antioxidant activities were different among cultivars and efficient extraction for TPC was found using UAE with 50% aqueous acetone. Furthermore, antioxidant activities were stable during digestion and genistein, within aglycones tested, was the most stable and bioaccessible compound following in vitro digestion. This information may provide manufacturers or researchers information required to develop food or nutraceutical products processed for better bioaccessibility of soybean bioactive components. / Ph. L’estil de vida que durant les últimes dècades s’està imposant a la nostra societat afavoreix el desequilibri del balanç energètic i en conseqüència l’aparició de la síndrome metabòlica, amb les implicacions que suposa no només en salut pública sinó també a nivell socioeconòmic. Malgrat que existeixen treballs que evidencien afectes beneficiosos sobre paràmetres de la síndrome metabòlica atribuïts al consum de soja i te verd, les vies moleculars d’actuació dels seus principals biocompostos no han estat encara totalment caracteritzades. En aquesta tesi s’ha descrit que les vies d’actuació i els mecanismes en els que participen les isoflavones genisteïna, daidzeïna i el seu metabòlit equol, contribuirien a una millora de la hiperglucèmia, la hiperlipidèmia, la inflamació i l’esteatosi hepàtica, importants components de la fisiopatologia de la síndrome metabòlica en humans. Per altra banda, s’ha demostrat que la principal catequina del te verd, el gal•lat d’epigal•locatequina, actua mitjançant la seva autooxidació, com a senyalitzador cel•lular, induint un desacoblament mitocondrial compensat per un augment de l’activació d’AMPK. Aquestes troballes contribueixen a una descripció dels mecanismes d’acció que podrien explicar els efectes beneficiosos sobre les alteracions del metabolisme energètic associats al consum de soja i te verd. / El estilo de vida que durante las últimas décadas se está imponiendo en nuestra sociedad favorece el desequilibrio del balance energético y con ello el desarrollo del síndrome metabólico, cuyas patologías asociadas son la principal causa de muerte en los países desarrollados. Aunque existen trabajos que evidencian efectos beneficiosos sobre parámetros del síndrome metabólico atribuidos al consumo de soja y té verde, las vías moleculares de actuación de sus principales biocompuestos no han sido aún caracterizadas. En esta tesis se ha descrito no sólo las vías sino también los mecanismos mediados por las isoflavonas genisteína, daidzeína y su metabolito equol, en modelos in vitro insulinorresistentes de los principales tejidos diana de la acción insulínica. Por su parte, se ha demostrado que la principal catequina del té verde, el galato de epigalocatequina, actúa mediante su autooxidación, como señalizador celular, induciendo un desacoplamiento mitocondrial, que se compensa por un incremento de la activación de AMPK. Estos hallazgos contribuyen a la descripción más de los mecanismos de acción que podrían explicar los efectos beneficiosos sobre los desajustes del metabolismo energético asociados al consumo de soja y té verde / Changes in Life style during the last decades in our society has lead to a shift of the energy balance directly associated to the development of metabolic syndrome that nowadays are the main cause of death in developed countries. Although beneficial effects of soy and green tea ingestion have been described previously, the molecular mechanisms of their main biocompounds have not been completely characterized yet. The aim of this thesis is to provide knowledge to help dilucidate them. In this context, the mechanism of action of the isoflavones genistein, daidzein and its metabolite equol have been described with potential interest to improve hyperglycemia, hyperlipidemia, inflammation and hepatic steatosis. On the other hand, it has been demonstrated that the main catechin of green tea, epigallocatequin gallate, through its autooxidation products, modifies cell signalling, leading to a mitochondrial uncoupling compensated by activation of AMPK. In conclusion, these findings offer a more detailed description of the action mechanisms that justify part of the beneficial effects of soy and green tea ingestion on alterations of energy metabolism.

Com o aumento da população mundial, são necessárias estratégias criativas para aumentar a produção de alimentos. Para superar esse imenso desafio, novos cultivares vem sendo desenvolvidos, através de diferentes técnicas, e com diferentes características, para alcançar uma maior produção de alimentos. Para conservação dessa produção vários métodos podem ser empregados, e a irradiação com cobalto constitui um método seguro e eficaz para o aumento no tempo de vida útil dos alimentos. Devido à importância comercial e nutricional da soja, possíveis alterações promovidas devem ser estudadas. O presente trabalho visa determinação dessas possíveis alterações causadas pela irradiação (com doses de 2, 4 e 8 kGy) em dois diferentes cultivares de soja (BRS 213 e Embrapa48) realizando análises químicas de perfil lipídico, isoflavonas, polifenóis totais, livres e ligados a proteínas, análise sensorial descritiva quantitativa, ensaio biológico para determinação da qualidade proteíca e ensaio com insetos para determinação de quebra de ressitência dos grãos e desinfestação. Os dois cultivares apresentaram perfil lipídico semelhante, e a irradiação promoveu pequena redução nas quantidades de ácidos linolênico e linoléico. Com relação as isoflavonas, o tratamento de radiação promoveu aumento nas quantidades das glicosiladas com dose de 2 kGy e diminuição com doses de 4 e 8 kGy enquanto aumentou a capacidade antioxidante e reduziu as proteínas ligadas à polifenóis. As alterações nas qualidades sensoriais dos grãos foram com relação a aparência e textura. Foi comprovada a eficiência da irradiação na desinfestação dos grãos e perda da resistência natural. As amostras irradiadas com 4 e 8 kGy se aproximaram dos índices de qualidade protéica aparente do padrão caseína. Os resultados obtidos indicam que o tratamento de radiação se oferece como um processo efetivo de desinfestação, sem prejudicar a qualidade da valiosa proteína de soja e com mínima alteração sensorial / With the increasement of the world population, criative strategies will be necessary to increase food prodution. To achieve this chalenge, new cultivars have been development, though different tecniques and with characteristics to improve the production. To maintain food conservation, several methods can be used. The use of Cobalto-60 radiation is a secure and useful method to increase the life time of foods. Due to the commercial and nutritional importance of soybean, some alterations must be studied. This study has the objective to determinate this alterations caused by irradiation (with doses of 2, 4 and 8 kGy) in grain of two different cultivars of soybean (BRS 213 e Embrapa 48), this study includes chemical analysis of lipid profile, isoflavones, total, free and protein-bound polyphenols, analysis sensory descriptive quantitative, biological assay for the determination of protein quality and insects assay for determination of break of resistance in the grains and desinfestation. Two cultivars presented similar lipid profile, and the irradiation promoted small reduction in the amounts of linoleic and linolenic acids. Regard to the isoflavones, the radiation treatment promoted increase in the amounts of the glycosides with dose of 2 kGy and decrease with doses of 4 and 8 kGy promoted increase in the antioxidant capacity and reduced protein-bound polyphenols. The alterations in the sensory qualities of the grains were regarding appearance and texture. It was proven the efficiency of the irradiation in the desinfestation of the grains and loss of the natural resistance. Samples irradiated with 4 and 8 kGy approached of the indices for apparent protein quality to the pattern casein. The results of this study indicating that radiation treatment of soybean offers considerable promise as an effective desinfestations process, without impairing the quality of the valuable soybean protein and with low sensory alteration

Porém, essa vantagem desaparece na pós-menopausa, sugerindo que os hormônios sexuais femininos exercem algum efeito cardioprotetor. Um dos mecanismos propostos para explicar essa proteção é o fato dos estrógenos promoverem a produção de importantes fatores vasoativos pelo endotélio vascular, entre eles o óxido nítrico e a prostaglandina I2. Com a diminuição da quantidade de estrógenos circulante, as mulheres na pós-menopausa, estão mais suscetíveis à disfunção endotelial e a doenças cardiovasculares. Estudos têm demonstrado que a terapia de reposição hormonal (TRH) utilizada por mulheres na pós-menopausa combate os sintomas deste período, melhora o quadro de disfunção endotelial e o perfil lipídico e aumenta a síntese de fatores vasoativos, que auxiliam na prevenção da DCV. Entretanto a TRH vem sendo questionada por grandes estudos como o WHI (Writing Group for the Women\'s Health Initiative Investigators) e o HERS (Heart and Estrogen/Progestin Replacement Study), que mostraram um risco aumentado de desenvolvimento de câncer de mama e endometrial em mulheres fazendo uso da TRH. Embora a soja em grão seja pouco consumida pela população brasileira, seus derivados protéicos são utilizados como ingredientes em diversos alimentos e a tendência é um aumento significativo do seu uso com a aprovação pela FDA e pela ANVISA da alegação funcional referente ao consumo de proteína de soja. Em paralelo, um número crescente de pesquisas sobre as isoflavonas, fitoestrógenos presentes em quantidades significativas na soja, vem demonstrando diversos efeitos benéficos destes compostos, entre os quais a sua ação antioxidante, anticarcinogênica e hipocolesterolêmica. O objetivo foi estudar as interações entre isoflavonas e proteínas da soja, seu efeito na biodisponibilidade in vitro e in vivo e o status antioxidante das isoflavonas. Os resultados sugerem que a presença da proteína reduz a quantidade e leva a um retardo no tempo de absorção das isoflavonas em relação à administração na forma livre. O efeito sobre a capacidade antioxidante do plasma e sobre a atividade e expressão gênica das enzimas CAT, GPx e SOD divergiu para a suplementação de isoflavonas ou proteínas separadamente ou em associação. / Although soybeans are not commonly consumed by the Brazilian population, soybean products are used as ingredients by the food industry and there is an increased trend in their consumption due to the approval of FDA and ANVISA on the claim regarding the health benefits of soy proteins. Therefore, many studies involving isoflavones, the phytoestrogens found in significant quantities in soybeans, have demonstrated several health benefits of these compounds, such as antioxidant capacity, and anticarcinogenic and hypocholesterolemic effects. The objective of the present study was to evaluate the interaction between isoflavones and soy proteins as well as the effect of this interaction on the in vitro and in vivo bioavailability of isoflavones and on the antioxidant status. The results indicated that the association with protein decreased the rate and proportion of absorption of isoflavones when compared to the free form. The effect of isoflavones on the plasma antioxidant capacity and on the enzymatic activity and gene expression levels of CAT, GPx and SOD was different when isoflavones were combined with soy protein compared to isoflavones alone.

Resumo: O crescimento da demanda por alimentos saudáveis está estimulando a inovação e o desenvolvimento de novos produtos na indústria alimentícia. Os alimentos saudáveis são chamados de alimentos funcionais, que são alimentos consumidos como parte de uma dieta usual que produzem efeitos metabólicos ou fisiológicos, com capacidade de reduzirem o risco de doenças crônico - degenerativas, além de suas funções nutricionais básicas. Entre os alimentos funcionais, o presente estudo destaca os produtos derivados da soja e os probióticos. As isoflavonas da soja parecem ser os componentes principais no combate ao câncer, por apresentarem, atividade antioxidante, aumento da apoptose, inibição da angiogênese e modulação da resposta imune. O produto fermentado por bactérias láticas, induz resposta imune inata e adaptativa. Os mecanismos pelos quais o produto fermentado pode inibir o crescimento do tumor são diminuição da resposta inflamatória induzida pelo carcinógeno, aumento da IgA, aumento da apoptose celular e aumento da taxa de TNF-a . Considerando o efeito benéfico dos fitoestrogênios da soja em relação à inibição do câncer e considerando, também, o fato de que determinadas bactérias láticas apresentam atividade anti-tumoral, parece-nos plenamente justificável a realização do estudo para verificar se os alimentos: produto de soja não fermentado (PSNF); iogurte de soja (IS) e iogurte de soja suplementado com isoflavonas (ISS), e Ração (R) exercem efeito no desenvolvimento do câncer de mama e verificar os níveis das citocinas, bem como a concentração de óxido nítrico liberado por macrófagos, e IFN-g em cultura de células esplênicas, indução da apoptose/necrose em macrófagos e angiogêneses no tumor. / Abstract: The growing demand for healthy foods is encouraging the development of new products by the food industry. Functional foods are normally part of a regular diet, but besides their nutritional properties, they also elicit metabolic and physiologic responses that may reduce the risk of chronic degenerative diseases. This study focuses on probiotics and soy-derived products. Soy isoflavones seem to play the most important role in the fight against cancer because, besides their anti-oxidant effects, they stimulate apoptosis, inhibit angiogenesis and modulate the immune response. Probiotics may inhibit tumor growth by diminishing the inflammatory response provoked by the carcinogen and by increasing apoptosis and TNF-_ and IgA levels. No. of bitstreams: 0 Previous issue date: 2011-01-26 / Introdução: É sabido que tanto as isoflavonas da soja (ISO) e a vibração mecânica de baixa intensidade e alta freqüência (VIB) possuem efeitos protetores no tecido ósseo de ratas ooforectomizadas (OVX). Objetivos: estudar os efeitos dos tratamentos com ISO e VIB isolados e combinados no tecido ósseo de ratas OVX. Métodos: 50 ratas Wistar com 6 meses de idade foram SHAM operadas ou OVX e após três meses, divididas em 5 grupos: SHAM; OVX; OVX tratadas diariamente por gavage com 200mg/kg de ISO por 90 dias consecutivos (OVX-ISO); OVX tratadas com VIB, 20 min./dia, 30 Hz, 0.6g, 5 dias por semana por 90 dias (OVX-VIB) e OVX tratadas com ISO e VIB (OVX-ISO+VIB). Após o tratamento, os fêmures e tíbias foram coletados e seus comprimento e espessura foram medidos. Os fêmures foram processados para histomorfometria e aos métodos histoquímicos, bioquímico e do TUNEL. As tíbias foram congeladas e destinadas para testes físicos e biomecânicos. Resultados: Os tratamentos não exerceram efeitos significantes no peso corporal e nas medidas de comprimento e espessura, exceto na espessura das tíbias dos grupos OVX-ISO e OVX-ISO+VIB que foi significantemente maior comparado ao grupo SHAM. Os testes físicos mostraram que o peso imerso, a porcentagem de material orgânico e a densidade óssea foram maiores nos grupos SHAM, OVX-VIB e OVX-ISO+VIB quando comparados aos outros grupos, enquanto que o volume ósseo, a densidade mineral óssea, porc entagem de material mineral e o peso das cinzas foram maiores nos grupos OVX e OVX-ISO do que nos outros grupos e não houve diferenças significantes nos parâmetros biomecânicos analisados entre os grupos. Nos fêmures, os tratamentos não exerceram efeitos significantes no volume ósseo trabecular, porém, o tratamento combinado exerceu efeitos positivos no osso cortical. Os tratamentos tiveram efeitos positivos em relação à quantidade e na organização das fibras colágenas e esse efeito foi mais expressivo no grupo OVX-ISO+VIB. O método do TUNEL não detectou diferenças nas células TUNEL-positivas entre os grupos. Conclusão: Esses resultados demonstram que o tratamento combinado (ISO+VIB) exerce maior efeito nos constituintes do tecido ósseo de ratas ooforectomizadas osteopênicas, quando comparado aos tratamentos isolados. / Introduction: It has been shown that both soy isoflavones (ISO) and low intensity high frequency mechanical vibration (VIB) have protective effects in bone tissue of ovariectomized (OVX) rats. Objective: We investigated whether combined treatments with ISO and VIB would exhibit cooperative effects in bone tissue of OVX rats. Methods: Fifty female Wistar rats at the age of 6 months were either sham-operated or OVX. After operation, the rats were divided in five groups: SHAM; OVX; OVX treated daily by gavage with 200mg/kg/body weight of ISO for 90 days (OVX-ISO); OVX treated with VIB, 20 min./day, at 60 Hz, 0.5g, 5 days/ week for 90 days (OVX-VIB) and OVX treated with ISO and VIB (OVX-ISO+VIB). After treatment, the femurs and tibias were collected and its length and thickness were measured. The femurs were then processed for histomorphometry, histochemistry and biochemical assay. The tibias were frozen for subsequent physical and mechanical tests. Results: Treatments did not have significant effects on body weight and in the bone measurements, except in the tibial thickness of the OVX-ISO and OVX-ISO+VIB groups that was significant higher than the SHAM group. Physical tests showed that the immersed weight, content of organic material and bone density were higher in the SHAM, OVX-VIB and OVX-ISO+VIB groups than the other groups, while the bone volume, bone mineral density, mineral material and the ash weight were higher in the OVX and OVX-ISO groups than the other groups and there were no significant differences in the mechanical tests among the groups. Treatment did not have significant effects in the trabecular bone volume, but it was not detected cortical bone loss in the group treated with the combined therapy. The treatments showed positive effects in the content of glycosaminoglycans, as well as in the organization of collagen fibers and these effects was higher in the OVX-ISO+VIB group. The TUNEL-positive cell profile was similar among the groups. Conclusion: These results demonstrate that the combined treatment (ISO+VIB) have better effects in the bone tissue constituents of ovariectomized osteopenic rats, when compared with the treatments isolated. / TEDE / BV UNIFESP: Teses e dissertações
