Avaliação do efeito osteogênico por diferentes fitoestrógenos em cultura de osteoblastos derivados de células tronco mesenquimais / Evaluation of the osteogenic effect of different phytoestrogens in osteoblasts culture derived from mesenchymal stem cells

A menopausa é provocada pela falência da produção de hormônios ovarianos e tem como consequências alterações desfavoráveis no metabolismo e perda de massa óssea. O declínio da produção de estrógeno é considerado um grande fator de risco para o desenvolvimento da osteoporose em mulheres e como tratamento faz-se o uso da Terapia de Reposição Hormonal. No entanto, esta terapia tem trazido riscos á saúde de alguns grupos de mulheres. Como alternativa ao tratamento tradicional, tem-se os fitoestrógenos, e com eles as isoflavonas, encontradas principalmente na soja, Trifolium pratense e Cimicifuga racemosa. Este estudo teve como objetivo comparar a capacidade de estimular a osteogênese in vitro, a partir de cultura de osteoblastos derivados de células tronco mesenquimais, em duas preparações de fitoestrógenos: O extrato de soja biotransformado pelo fungo Aspergillus awamori (ESBF), e o Menoflavon® 40mg (Melbrosin International) composto pela isoflavona Trifolium pratense. Para este objetivo foram realizadas: a) Avaliação do crescimento e proliferação celular b) Viabilidade e crescimento das culturas de osteoblastos. c) Dosagem de proteína total das culturas de osteoblastos. d) Determinação da atividade específica da enzima fosfatase alcalina. e) Formação da matriz mineralizada. O Menoflavon® foi testado nas concentrações de 28,75 nM de daidzeína (D) + 7,5 nM de genisteína (G) (0,5 ?g/mL de Menoflavon®); 57,5 nM de D + 15 nM de G (1 ?g/mL de Menoflavon®); e 230 nM de D + 60 nM de G (4 ?g/mL de Menoflavon®); controle padrão de 57,5 nM de D + 15 nM de G comercial e controle de dimetilsulfóxido (DMSO). O ESBF foi testado nas concentrações de 1,181 nM de D + 0,922 nM de G (0,5 ?g/mL de ESBF); 2,361 nM de D + 1,845 nM de G (1 ?g/mL de ESBF); 9,445 nM de D + 7,379 nM de G (4 ?g/mL de ESBF); controle padrão de 2,361 nM de D + 1,845 nM de G comercial e controle de DMSO. Com a metodologia do MTT (3[4,5-dimetiltiazol-2-il]-2,5-brometo difenil tetrazolium) e da Resazurina comprovamos que não houve morte celular nas concentrações testadas com as duas formulações. A dosagem de proteínas totais manteve-se constante com as duas formulações e a formação de matriz mineralizada também manteve-se constante em relação ao controle para ambos. A atividade específica da fosfatase alcalina teve um decréscimo significativo ao 14º dia com todas as concentrações testadas e ao 21º dia com algumas concentrações de Menoflavon e com o ESBF decresceu em alguns dias, no entanto manteve-se estável no restante do teste. O ESBF mostrou ser melhor que o Menoflavon®, já que obtivemos resultados semelhantes e sua concentração é 24 vezes menor. No entanto, o estudo realizado mostrou que tanto o ESBF quanto o Menoflavon® não são capazes de estimular a osteogênese in vitro, a partir de cultura de osteoblastos derivados de células tronco mesenquimais. / Menopause is caused by failure in the production of ovarian hormones and its consequences are unfavorable changes in metabolism and bone loss. The decline in estrogen production is considered a major risk factor for the development of osteoporosis in women, and the Hormone Replacement Therapy is used as treatment. However, this therapy has brought some risks to the health of some groups of women. As an alternative to the traditional treatment, phytoestrogens as isoflavones, found mainly in soy, Trifolium pratense, Cimicifuga racemosa and rye can be used. This study aimed to compare the ability of two preparations of phytoestrogens to stimulate osteogenesis in vitro (from cultures of osteoblasts derived from mesenchymal stem cells): soy extract biotransformed by the fungus Aspergillus awamori (ESBF), and Menoflavon® 40mg (Melbrosin International) composed by the isoflavone Trifolium pratense. With this objective, were used: a) Evaluation of cell growth and proliferation; b) Viability of the cultures of osteoblasts; c) Determination of total protein from the cultures of osteoblasts; d) Determination of the specific activity of the enzyme alkaline phosphatase; e) Formation of mineralized matrix. Menoflavon® was tested at the concentrations of 28.75 nM of daidzein (D) + 7.5 nM of genistein (G) (Menoflavon® 0.5 ?g/mL); 57.5 nM D + 15 nM G (Menoflavon® 1 ?g/mL); and 230 nM D + 60 nM G (Menoflavon® 4 ?g/mL); standard control of commercial 57.5 nM D + 15 nM G and DMSO control. With the MTT and Resazurin methods we verified that there was no cell death for all concentrations tested with the two formulations. The amount of total protein remained constant with the two formulations, and the formation of mineralized matrix also were the same as the control. The specific activity of alkaline phosphatase decreased significantly on day 14 for all concentrations tested, and at day 21 for some concentrations of Menoflavon®, with the ESBF decreased some days, however remained constant in the other tests. Therefore, ESBF proved better than Menoflavon®, since we obtained similar results for both, but the concentration of ESBF is 24 times smaller than the concentration of Menoflavon®. However, both the ESBF and the Menoflavon® were not capable of stimulating osteogenesis in vitro from cultures of osteoblasts derived from mesenchymal stem cells.

Estudo das condições de processamento para obtenção de isolado protéico de soja com teor aumentado de isoflavonas / Study of conditions the processing to production of isoflavone-rich soy protein isolates

Os isolados protéicos de soja são utilizados como ingredientes em diversos alimentos e sua utilização vêm aumentando juntamente com o aumento das pesquisas sobre os metabólitos secundários da soja, as isoflavonas. Alguns efeitos benéficos vem sendo associados às isoflavonas, entre estes a sua ação antioxidante, a redução ao risco de câncer, doenças cardiovasculares e osteoporose. O objetivo deste estudo foi o de otimizar as condições de extração das isoflavonas e de suas formas conjugadas a partir da farinha desengordurada de soja, visando o preparo de isolado protéico de soja. Os resultados mostraram que a obtenção de isolados protéicos de soja com teor aumentado de isoflavonas depende da utilização de condições brandas de centrifugação para a separação do precipitado isoelétrico, assim como da utilização de água acidificada na sua lavagem. A presença de isoflavonas no isolado resulta de três fatores, o primeiro referindo-se à associação entre isoflavonas e proteínas através de interações hidrofóbicas, eletrostáticas, e pontes de hidrogênio; o segundo à menor solubilidade das isoflavonas presentes na farinha desengordurada de soja no pH isoelétrico; e o último ao processo de carreamento (físico) das isoflavonas pelas proteínas insolubilizadas. / Soy protein isolates are used as ingredients in several food products and their use is increasing together with the increase of the researches on the secondary metabolites of soy, the isoflavones. Some beneficial effects have been associated to the isoflavones, among these their antioxidant action, prevention of cancer, cardiovascular diseases and osteoporosis. The objective of this study was to optimize the extraction conditions of the isoflavones from the defatted soy flour, seeking the preparation of soy protein isolates. The results showed that the obtention of soy protein isolates with increased content of isoflavones depends on the use of mild conditions of centrifugation for the separation of the isoelectric precipitate, as well as on the use of water acidified in the washing step. The presence of isoflavones in the isolates resulted from three factors, the first refers to the association between isoflavones and proteins through hydrophobic; and electrostatic interactions, and hydrogen bonding; the second to the decreased solubility of the isoflavones extracted from the defatted soy flour in the isoelectric pH; and the last to the carrying process (physical) of isoflavones by the precipitating proteins.

Determinação de isoflavonas e capacidade antioxidante de alimentos industrializados à base de soja e/ou produtos derivados consumidos no Brasil / Determination of isoflavones content and antioxidant capacity of industrialized foods containing soy and soy products consumed in Brazil

A soja contém alto conteúdo protéico e de lipídeos poliinsaturados e representa a principal fonte de isoflavonas. Por este grão não fazer parte da dieta brasileira, produtos contendo soja surgiram no mercado como fontes de isoflavonas, adaptados ao paladar da nossa população. Os condimentos também têm sido incorporados aos ditos \"alimentos funcionais\" pela sua relação com efeitos benéficos à saúde. Este trabalho objetivou determinar o teor de isoflavonas dos produtos contendo soja e/ou derivados, o teor de flavonóides de diferentes condimentos e marcas, e a capacidade antioxidante destas amostras. Os resultados mostram que o quibe vegetariano destaca-se como fonte de isoflavonas, e o chocolate pela capacidade antioxidante. Entre os condimentos, houve diferença significativa entre tipos e marcas. O orégano destacou-se quanto à capacidade antioxidante e a salsa quanto ao teor de flavonóides. Alimentos contendo soja ou derivados e os condimentos podem representar fontes de compostos bioativos para a população brasileira. / Soy contains high protein and polyunsaturated lipids content and represents the main source of isoflavones. This grain is not part of the Brazilian diet, so some products containing soy emerged as sources of isoflavones, adapted to the taste of our population. Spices have also been incorporated into \"functional foods\" by their relation to health benefits. This study aimed to determine the isoflavone content of products containing soy and/or derivatives, flavonoids content in different brands and condiments, and antioxidant capacity of these samples. The results showed that vegetarian \"quibe\" is a good source of isoflavones, and chocolate had the highest antioxidant capacity. Among spices, there were significant differences between the types and brands. Oregano had the highest antioxidant capacity and parsley had the highest flavonoids content. Foods containing soy or derivatives and condiments may represent sources of bioactive compounds for Brazilian population.

Combating Stress: The Use of Isoflavones as Nutraceuticals to Improve Immunity and Growth in Nile Tilapia (<i>Oreochromis niloticus</i>)

Stressors in the aquaculture environment can lead to negative impacts on growth and immune health, resulting in susceptibility to infectious diseases. These stressors are expected to increase as the growth of aquaculture continues to rise to meet demands for quality fish protein. Isoflavones, as a crude extract or as a pure isolate, may be effective in modulating the stress response, promoting growth and immunity. The objective of these studies was to examine the effect of various pure isoflavone isolates and crude isoflavone extracts on stress, growth, and immunity. Nile tilapia (Oreochromis niloticus) were stressed by adding hydrocortisone to the feed. In a 7-week study, pure isoflavone isolates of genistein and puerarin were evaluated to determine their respective effects on stress, growth, and immunity. A separate 10-day physiological and 6-week growth study focused on crude isoflavone extracts from kudzu (Pueraria lobata), red clover (Trifolium pratense), and soybean (Glycine max) was performed to determine their respective effects on stress, growth, and immunity. Numerous physiological parameters of the fish were measured (serum cortisol concentration, blood glucose concentration, hematocrit, hepatosomatic index, plasma protein concentration, lysozyme activity, and spleensomatic<br>index) to determine the effects of these pure isoflavone and crude isoflavone extracts on the modulation of stress and immunity. Many growth parameters were examined (length, weight, condition factor, weight gain, specific growth rate, feed intake, feed conversion ratio, and protein efficiency ratio) as well to determine the effects of these pure isoflavones and isoflavone extracts on growth. The addition of isoflavone and crude isoflavone extracts to the diet of Nile tilapia ameliorated some of the negative consequences of stress. Compared to stressed fish fed commercial feed, genistein and puerarin added to the diet appeared to improve serum cortisol concentrations, which resulted in increased plasma protein, albeit at different durations of stress. Puerarin, as well as all three crude isoflavone extracts, significantly increased spleen-somatic index compared to non-supplemented stressed fish, although the crude isoflavone extracts did not appear to improve serum cortisol concentrations. Crude isoflavone extracts also showed overall increases in lysozyme activity compared to non-supplemented stressed fish, although this was not significant. Genistein, puerarin, and red clover showed increased growth rates, feed conversion ratio, and protein efficiency. Overall, pure isolates of isoflavone appear to be more effective in modulating stress, immunity, and growth than the crude isoflavone extracts, although red clover extract showed promises in the ability to modulate the stress response and improve growth and immunity. There are likely substantial interactions between the isoflavones in the crude extracts that cannot be fully understood by measuring the effects of single isoflavones. Regardless, isoflavone supplementation (pure or crude) appeared to generally have an overall positive impact on stressed Nile tilapia, requiring more research to better understand the effects and mechanisms behind these isoflavones.

The material for study was collected in the campus of Embrapa in Bel?m, Par? state, and identified by Dra Silvane Taveres Rodrigues. The compounds described in this phytochemistry investigation were isolated by the solvents partition and chromatographyc techniques of the extracts obtained by maceration at room temperature with hexane, ethyl acetate and methanol. The structures were determined through analysis of data provided by IR, 1H and 13C NMR (1D an 2D techniques), mass spectrometry including GCMS and HPLC-MS of natural compounds and some derivatives. From the stem dichloromethane extract friedelin, friedelinol, sitosterol, stigmasterol, campesterol, 3-b-O-Dglucopyranosyl- stigmasterol, 2,6-dimethoxy?benzoquinine, 2,6-dimethoxy-hydroquinone, the isoflavones: 5,4'-dihydroxy-7,3',5'-trimethoxy-isoflavone, 5-hydroxy-7,3',4',5'-tetramethoxyisoflavone, 5,4'-dihydroxy-7,3'-dimethoxy-isoflavone, 7,5-dihydroxy-3',4',5'-trimethoxyisoflavone, 7,5,4?-trihydroxy-3?,5?-dimethoxy-isoflavone, and ferulic and syringic aldehyde were isolated. From the dichloromethane partition of the methanol extract of the stem vanillic acid, 4 ((1E)-3-hydroxy-1-propenyl)-2-methoxyphenol and 3,5-dimethoxy-4-hydroxydihydrocinamaldehyde were isolated. From hexane fraction of methanol extract from the leaves lupeone was isolated, and from the dichloromethane methanol partition were identified the biflavonoids amentoflavone and 7-methyl-amentoflavone, known as sequoiaflavone, along with syringic acid. From the ethyl acetate of the methanol extracts partition the epicatechin which absolute configuration was defined by circular dichroism spectral analysis was isolated. The sequioflanove is been identified in Ochnaceae for the first time. From the polar fraction the total phenol were determined by adapted Folin-Denis and precipitation with casein methods and by NMR spectral analysis. / Este trabalho descreve o estudo fitoqu?mico de caule e folhas da esp?cie vegetal Ouratea ferruginea Engl, Ochnaceae. O material para estudo foi coletado no campus da Embrapa em Bel?m do Par? e identificado pela Dra Silvane Tavares Rodrigues. As subst?ncias descritas nesta investiga??o fitoqu?mica foram isoladas atrav?s de parti??o com solventes e t?cnicas cromatogr?ficos de extratos obtidos atrav?s de macera??o a frio com hexano, acetato de etila e metanol. Da parti??o em diclorometano do extrato metan?lico do caule foram isolados ?cido van?lico, 4((1E)-3-hidroxi-1-propenil)-2- metoxifenol e 3,5-dimetoxi-4-hidroxi-diidrocinamalde?do. Das folhas foi isolada a lupeona na parti??o em hexano do extrato metan?lico; e na parti??o em diclorometano foram identificados os biflavon?ides amentoflavona e 7-metil-amentoflavona, conhecida como sequoiaflavona, e o ?cido sir?ngico. Na parti??o em acetato de etila foi isolado a epicatequina cuja configura??o absoluta foi definida com an?lise do espectro de dicro?smo circular. A sequioflavona est? sendo registrada pela primeira vez em Ochnaceae. Das fra??es polares foram determinados o teor de fen?is totais e taninos por m?todos de Folin-Denis e precipita??o com case?na, adaptados, al?m de an?lise com espectros de RMN.

Der Einfluss von diätetisch verabreichten Sojaisoflavonen auf den Homocysteinmetabolismus und die Endothelfunktion bei gesunden, postmenopausalen Frauen / The impact of soy isoflavones on homocysteine metabolism and endothelial function in healthy postmenopausal women

Homocystein (tHcy) gilt als unabhängiger kardiovaskulärer Risikofaktor und korreliert eng mit einer endothelialen Dysfunktion, welche nichtinvasiv mittels der flussinduzierten Vasodilatation (FMD) messbar ist. Experimentelle Hyperhomocysteinämie ist mit einer reduzierten Bioverfügbarkeit von endothelialen Stickstoffmonoxid (NO) bei gleichzeitig erhöhten Spiegeln des kompetetiven Inhibitors der NO-Biosynthese asymmetrisches Dimethylarginin (ADMA) assoziiert. In-vivo senkt eine Östrogenbehandlung neben tHcy auch die ADMA-Spiegel und verbessert signifikant die Endothelfunktion. Hinsichtlich ihrer Wirkung als selektive Östrogenrezeptormodulatoren wird angenommen, dass Phytoöstrogene, speziell Sojaisoflavone, ähnliche Effekte hervorrufen.<br><br> Innerhalb einer europäischen, multizentrischen, doppelblinden Interventionsstudie an 89 gesunden, postmenopausalen Frauen wurde der Einfluss von Sojaisoflavonen auf den Homocysteinmetabolismus, den Blutdruck und die in-vivo Endothelfunktion untersucht. Die cross-over Studie umfasste zwei achtwöchige Interventionsperioden, die von einer gleichlangen Wash-out-Phase unterbrochen waren. Die Zuteilung zum Isoflavon- (50 mg/d) oder Plazeboregime für die erste Interventionsphase erfolgte randomisiert. Endpunkterhebungen fanden jeweils in den Wochen 0 und 8 der Interventionsperioden statt.<br><br> Die renale Ausscheidung von Genistein, Daidzein und Equol war während der Isoflavonintervention signifikant erhöht (P>0,001). Die Phyoöstrogene hatten weder einen Effekt auf die tHcy-Konzentration (P=0,286), noch auf ADMA, Erythrozytenfolat und Vitamin B-12 (P>0,05) im Plasma. Während die Summe aus Nitrat und Nitrit (NOx), welche die NO-Bioverfügbarkeit reflektiert, im Verlaufe der Plazebobehandlung abfiel, wurde ein leichter Anstieg bei der Isoflavonsupplementation beobachtet (Delta Wo8-Wo0: -2,60 [-8,75; 2,25] vs. 1,00 [-6,65; 7,85] µmol/L P<0,001), was zu einem signifikanten Behandlungseffekt führte. Weiterhin wurde eine positive Korrelation zwischen ADMA und Vitamin B-12 gefunden (R=0,252; P=0,018). Die flussinduzierte Vasodilatation (P=0,716), ein Maß für die Endothelfunktion, blieb durch die Isoflavonbehandlung unbeeinflusst, obwohl sich diese über die Zeit insgesamt verbesserte (P>0,001). Bis auf einen marginalen Anstieg des systolischen Wertes (P=0,032) im Vergleich zur Plazebobehandlung blieb der Blutdruck während der Isoflavonintervention unverändert.<br><br> Im Gegensatz zu Östrogen übten Sojaisoflavone weder einen Einfluss auf die in-vivo Endothelfunktion noch auf die traditionellen und neuen kardiovaskulären Risikofaktoren den Blutdruck, tHcy und ADMA aus. Demzufolge ist der gesundheitliche Nutzen isolierter Isoflavone hinsichtlich einer Prävention hormonmangelbedingter Erkrankungen in gesunden postmenopausalen Frauen fraglich. / Homocysteine (tHcy) is a strong and independent risk factor for cardiovascular disease. Hyperhomocysteinemia contributes to endothelial dysfunction as assessed by flow-mediated vasodilation (FMD). The mechanisms by which homocysteine generates endothelial dysfunction remain incompletely understood although a growing body of data suggests that the bioavailability of nitric oxide (NO) is reduced. The principal competitive inhibitor of endothelial NO-synthase asymmetric dimethylarginine (ADMA) may play a central role in homocysteine related dysfunction as it is derived from homocysteine metabolism. Cardiovascular risk factor modification has suggested beneficial effects of estrogen on endothelial function by lowering homocysteine and ADMA levels. We hypothesize that phytoestrogens particular isoflavones act in a similar manner.<br><br> The effects of soy isoflavones on homocysteine metabolism and endothelial function were investigated within a multi-centre, double blind, cross-over intervention trial in 89 European postmenopausal women. Subjects consumed either fruit cereal bars with or without soy isoflavones (50 mg/d) for 8 weeks each with a 8 weeks washout period in between. Endpoint measurements were during both treatment phases at baseline and weeks 8, respectively. <br><br> Urinary phytoestrogens increased significantly after isoflavone intervention (P<0.001). Isoflavone supplementation did affect neither plasma total homocysteine (P=0.286) nor ADMA, vitamin B-12 or folate (P<0.05). Isoflavones had a favorable effect on NO-metabolism assessed by analysis of NO-metabolites (NOx) nitrite and nitrate. While NOx concentration significantly decreased during placebo there was a slight increase after isoflavone supplementation leading to a significant treatment difference (delta wk8-wk0: -2.60 [-8.75; 2.25] vs. 1.00 [-6.65; 7.85] µmol/L P<0.001). There was no association between total homocysteine and ADMA whereas a positive correlation was found for ADMA and vitamin B-12 (R=0.252; P=0.018). The endothelial function model did not demonstrate any difference between either treatment regime (P=0.716), although endothelial function assessed by flow-mediated vasodilation improved in general (P<0.001). A potential adverse effect was noted, with an elevation in systolic blood pressure (P=0.032) whereas diastolic blood pressure and mean arterial pressure remained unaffected.<br><br> Soy isoflavones did not have beneficial effects on endothelial function as well as on traditional and novel cardiovascular risk factors like plasma homocysteine, blood pressure and ADMA as observed for estrogen treatment. The health benefit of isolated isoflavones in healthy postmenopausal women is questionable.

Six isoflavones derived from Millettia griffoniana namely, 4’-methoxy-7-O-[(E)-3-methyl-7hydroxymethyl-2,6 octadienyl]isoflavone (7-O-DHF), Griffonianone C (Griff C), 7-O-geranylformononetin (7-O-GF), 3’,4’-dihydroxy-7-O-[(E)-3,7-dimethyl-2,6-octadienyl]isoflavone (7-O-GISO), Griffonianone E (Griff E), 4’-O-geranylisoliquiritigenin (4-O-GIQ) were tested for potential oestrogenic activities in three different oestrogen receptor alpha (ERα) dependent assays, namely a recombinant yeast assay, a reporter gene assay based on stably transfected MCF-7 cells (MVLN cells) and the induction of alkaline phosphatase in Ishikawa cells. The oestrogenic activities of isoflavones from Millettia griffoniana could be completely suppressed by the pure oestrogen antagonist, fulvestrant. The expression of Ki-67, proliferating cell nuclear antigen (PCNA) and cyclin D1 (CD1) mRNA used as indicator of cell proliferation in MCF-7 cells was assayed. Based on these in vitro results, Griff C was further tested in vivo. The main objective of this part of the work was to study the mechanistic basis of the oestrogenicity Three different doses of Griff C (2, 10, or 20 mg/kg BW) of Griff C in ovariectomised Wistar rats. 17β-oestradiol (E2: 10 µg/kg BW) was used as positive control. They were treated daily for three consecutive days and sacrificed 24 hours after receiving the last dose. The whole uterus was removed and weight. Liver and vena cava fragments were also collected and stored together with uteri in liquid nitrogen for subsequent real-time PCR to evaluate the effects of Griff C on the regulation of some relevant oestrogen–responsive genes in the uterus, the liver and the vena cava. The role of Griff C in apoptosis or in cell survival, through mediation of the phosphatidylinositol 3 kinase-Akt (PI3K-Akt) signaling pathway, was also investigated. Western blot analysis revealed that Griff C slightly increased the phosphorylation of Akt at its serine 473 residue. In this work, oestrogenic properties of the isoflavones derived from Millettia griffoniana are described using reporter gene assays and the oestrogen-inducible alkaline phosphatase Ishikawa model for the first time. These in vitro data were verified in vivo showing the regulation of the expression of various relevant oestrogen-responsive genes by Griff C. The spectrum of its activity was clearly similar to that of 17β-oestradiol on uterine hepatic and vena cava tissues of ovariectomised rats except for the proliferative response. However Griff C remained 100 to 1000 times less effective than oestradiol. These findings confirmed that some of the biological effects attributed to Millettia griffoniana are closely related to oestrogen-mediated action.

Produção de farinha de kinako a partir de variedade de soja BRS 257 e desenvolvimento e caracterização de pão de forma com kinako e chia (Salvia hispânica)

O kinako é uma farinha integral obtida a partir do grão de soja torrada e moída. Esta farinha tem elevada qualidade nutricional em função do conteúdo de proteínas, fibras, ácidos graxos insaturados e compostos bioativos como as isoflavonas. As isoflavonas estão presentes na soja principalmente nas formas conjugadas (β-glicosídicas, acetil e malonil) e, em menor proporção, nas formas livres (agliconas). Diversos estudos comprovam a eficiência das isoflavonas agliconas na prevenção de doenças crônicas. Algumas tecnologias têm sido empregadas para a conversão das isoflavonas β-glicosídicas em agliconas através de altas temperaturas. Neste contexto, o presente trabalho objetivou a conversão de isoflavonas contidas no kinako utilizando o processo de torra, buscando melhorar a qualidade nutricional e as propriedades biológicas para posterior uso como ingrediente em produtos panificados. A torra foi conduzida através de delineamento fatorial 22,com 4 pontos axiais e 4 repetições no ponto central, totalizando assim 12 corridas, para verificar os efeitos das variáveis tempo em minutos e temperatura em graus celcius sobre o teor de isoflavonas no kinako. O processo contribuiu para melhorar o perfil de isoflavonas especialmente no tratamento 8 (200 °C por 31 min), convertendo as isoflavonas conjugadas em agliconas. A partir do kinako obtido no tratamento 8, juntamente com 2 % de chia, semente rica em ácidos graxos insaturados, foram elaboradas quatro formulações de pães variando a quantidade de kinako (0, 10, 20 e 30 %). Mediante análise sensorial não se observou diferença significativa (p>0,05) para a formulação padrão e com 10 % de kinako acrescida de 2 % de semente de chia. A conversão das isoflavonas do kinako por processo de torra mostrou ser uma estratégia promissora para agregar valor nutricional juntamente com a semente de chia em pães. / The kinako is an integral flour obtained from the seed of roasted and ground soybean. This flour has high nutritional quality according to the content of protein, fiber, unsaturated fatty acids and bioactive compounds such as isoflavones. The isoflavones in soy are present mainly in conjugated forms (β-glucosides, acetyl and malonyl) and, to a lesser extent, in the free form (aglycone). Several studies have demonstrated the efficacy of isoflavone aglycone in the prevention of chronic diseases. Some technologies have been employed for the conversion of β-glucosides isoflavones in aglycones atravéz high temperatures. In this context, this paper aims at conversion of isoflavones present in kinako using the roasting process, seeking to improve the nutritional quality and biological properties for later use as an ingredient in baked goods. Roasting was performed using a factorial design with 22, with the variables Temperature (°C) and time (min) of exposure. The process contributed to increased levels of protein, lipid and crude fiber besides improving the profile of isoflavones especially in treatment 8 (200 °C for 31 min), converting conjugated isoflavones in aglycones. From the kinako 8 obtained in the treatment along with 2% chia seed rich in unsaturated fatty acids breads four formulations varying the amount of kinako (0, 10, 20 and 30%) were prepared. By sensory analysis no significant difference (p> 0.05) for the standard formulation with 10% plus 2% kinako chia seed was observed. The conversion of isoflavones from kinako by the roasting process proved to be a promising strategy for adding nutritional value along with the chia seed bread.

O presente trabalho teve como objetivo desenvolver uma metodologia para produção de extratos de soja enriquecidos em isoflavonas agliconas e avaliar suas atividades na síntese e na degradação de óxido nítrico (NO) e na proliferação de células de adenocarcinoma de mama (MCF-7) cultivadas in vitro. O doador de óxido nítrico (NO), nitroprussiato de sódio (SNP), foi utilizado como agente indutor para o acúmulo de isoflavonas em sementes de soja, que foram identificadas e quantificadas por cromatografia líquida de alta eficiência e espectrometria de massas. O tratamento com SNP aumentou a síntese das isoflavonas agliconas de forma dosedependente. A produção relativa de formas agliconas foi de aproximadamente 75% nos exsudatos (solução indutora), em comparação com a extração feita apenas a partir somente dos tecidos (34%). Após tratamento com SNP, o exsudato do eixo embrionário acumulou a maior quantidade de isoflavonas livres (1,61 µg/mg) em relação ao cotilédone (1,1 µg/mg), enquanto que nos controles a produção total de isoflavonas foi de 0,3 µg/mg, sendo 95% nas formas conjugadas. Os extratos (350 µg) do eixo embrionário ou cotilédone enriquecido em agliconas, mas não os controles, estimularam a redução ácida do nitrito (determinada com eletrodo específico) levando à produção de 1,4 e 0,9 ?mol de NO em pH 2,0, valores que foram 8 vezes e 5,1 vezes maiores quando comparados à redução apenas na presença de 50 µM de nitrito. A atividade redutora de nitrito foi diminuída com a elevação do pH e estes resultados foram validados através da detecção de NO por quimiluminescência. As principais isoflavonas, livres ou conjugadas, identificadas nos extratos não estimularam a redução ácida do nitrito. Entretanto, foi observado que o tratamento com SNP também provocou o acúmulo de outros fenólicos, como os ácidos gálico, caféico e p-coumárico, que estimularam a redução ácida do nitrito. Em pH neutro os extratos apresentaram atividade sequestrante de NO sendo o de embrião novamente o mais ativo (0,85 ?mol de NO degradado/min), quando comparado com o cotilédone (0,24 ?mol/min) e com a degradação espontânea do controle (0,1 ?mol/min). Os extratos do eixo embrionário e do cotilédone apresentaram efeito bifásico dose-dependente na viabilidade e proliferação de células MCF-7. Nas doses menores (250-500 µg/mL) os extratos promoveram (10-25%) e em concentrações mais elevadas (750-1000 µg/mL) inibiram (15%) a proliferação celular. Na concentração de 500 µg/mL, o efeito proliferativo dos extratos foi revertido por nitrito (100 µM) em 34-36%. As doses mais elevadas dos extratos também estimularam a emissão de NO (detectado com um indicador fluorescente). O efeito bifásico foi atribuído à ação das isoflavonas agliconas em ativar os receptores estrogênicos e estimular a produção de NO, o que foi potencializado pelo nitrito. O conjunto dos resultados sugere que os extratos de soja enriquecidos em compostos bioativos interferem no metabolismo do NO e na proliferação de células de adenocarcinoma de mama / Abstract: Although soybean isoflavones naturally accumulate in their glyco- and malonylconjugated forms, the beneficial effects that soybean containing foods have on human health have been credited to their aglycones. This study aimed to develop a methodology for producing soybean extracts enriched with isoflavones aglycones and evaluate their activities in the synthesis and degradation of nitric oxide (NO) and proliferation of breast carcinoma cells (MCF-7) cultured in vitro. The nitric oxide (NO) donor, sodium nitroprusside (SNP), was used as a promoter for the accumulation of isoflavones in soybean seeds, which were identified and quantified by high performance liquid chromatography and mass spectrometry. SNP treatment increased the synthesis of the aglycone isoflavones in a dose-dependent manner. The relative production of aglycone forms was approximately 75% in the exudates (promoter solution), compared to the extraction made from only the tissues (34%). After treatment with SNP, exudate from the embrionary axis accumulated the largest amount of free isoflavones (1.61 µg/mg) compared to cotyledon (1.1 µg/mg), whereas in controls the total production of isoflavones was 0.3 µg/mg, with 95 % in conjugated forms. The extracts (350 µg) of embrionary axis or cotyledon enriched in aglycones, but not the controls, stimulated the acid reduction of nitrite (measured with specific electrode) leading to production of 1.4 and 0.9 ?mol of NO at pH 2, 0, values that were 8 times and 5.1 times higher when compared to the reduction only in the presence of 50 µM nitrite. The nitrite reduction activity was decreased with increasing pH and these results were validated by detection of NO by chemiluminescence. The main isoflavones, free or conjugated, identified in the extracts did not stimulate the acid reduction of nitrite. However, we observed that treatment with SNP also caused the accumulation of other phenolic compounds such as gallic, caffeic and p-coumaric acids, which stimulated the acid reduction of nitrite. At neutral pH extracts showed scavenging activity of NO and the embryo again was the most active (0.85 ?mol NO degraded/min) when compared to the cotyledon (0.24 ?mol/min) and to the spontaneous degradation of the control (0.1 ?mol/min). The embryo and cotyledon extracts showed dose-dependent biphasic effects on viability and proliferation of MCF-7 cells. At lower doses (250-500 µg/mL) extracts promoted (10-25%) and at higher concentrations (750-1000 µg/mL) inhibited (15%) cell proliferation. At 500 µg/mL, the proliferative effect of the extracts was reversed by nitrite (100 µM) in 34-36%. The higher doses of the extracts also stimulated emission of NO (detected with a fluorescent indicator). The biphasic effect was attributed to the action of isoflavone aglycones in activating estrogen receptors and stimulating the production of nitric oxide, which was increased by nitrite. Avaliação do efeito osteogênico por diferentes fitoestrógenos em cultura de osteoblastos derivados de células tronco mesenquimais / Evaluation of the osteogenic effect of different phytoestrogens in osteoblasts culture derived from mesenchymal stem cells

A menopausa é provocada pela falência da produção de hormônios ovarianos e tem como consequências alterações desfavoráveis no metabolismo e perda de massa óssea. O declínio da produção de estrógeno é considerado um grande fator de risco para o desenvolvimento da osteoporose em mulheres e como tratamento faz-se o uso da Terapia de Reposição Hormonal. No entanto, esta terapia tem trazido riscos á saúde de alguns grupos de mulheres. Como alternativa ao tratamento tradicional, tem-se os fitoestrógenos, e com eles as isoflavonas, encontradas principalmente na soja, Trifolium pratense e Cimicifuga racemosa. Este estudo teve como objetivo comparar a capacidade de estimular a osteogênese in vitro, a partir de cultura de osteoblastos derivados de células tronco mesenquimais, em duas preparações de fitoestrógenos: O extrato de soja biotransformado pelo fungo Aspergillus awamori (ESBF), e o Menoflavon® 40mg (Melbrosin International) composto pela isoflavona Trifolium pratense. Para este objetivo foram realizadas: a) Avaliação do crescimento e proliferação celular b) Viabilidade e crescimento das culturas de osteoblastos. c) Dosagem de proteína total das culturas de osteoblastos. d) Determinação da atividade específica da enzima fosfatase alcalina. e) Formação da matriz mineralizada. O Menoflavon® foi testado nas concentrações de 28,75 nM de daidzeína (D) + 7,5 nM de genisteína (G) (0,5 ?g/mL de Menoflavon®); 57,5 nM de D + 15 nM de G (1 ?g/mL de Menoflavon®); e 230 nM de D + 60 nM de G (4 ?g/mL de Menoflavon®); controle padrão de 57,5 nM de D + 15 nM de G comercial e controle de dimetilsulfóxido (DMSO). O ESBF foi testado nas concentrações de 1,181 nM de D + 0,922 nM de G (0,5 ?g/mL de ESBF); 2,361 nM de D + 1,845 nM de G (1 ?g/mL de ESBF); 9,445 nM de D + 7,379 nM de G (4 ?g/mL de ESBF); controle padrão de 2,361 nM de D + 1,845 nM de G comercial e controle de DMSO. Com a metodologia do MTT (3[4,5-dimetiltiazol-2-il]-2,5-brometo difenil tetrazolium) e da Resazurina comprovamos que não houve morte celular nas concentrações testadas com as duas formulações. A dosagem de proteínas totais manteve-se constante com as duas formulações e a formação de matriz mineralizada também manteve-se constante em relação ao controle para ambos. A atividade específica da fosfatase alcalina teve um decréscimo significativo ao 14º dia com todas as concentrações testadas e ao 21º dia com algumas concentrações de Menoflavon e com o ESBF decresceu em alguns dias, no entanto manteve-se estável no restante do teste. O ESBF mostrou ser melhor que o Menoflavon®, já que obtivemos resultados semelhantes e sua concentração é 24 vezes menor. No entanto, o estudo realizado mostrou que tanto o ESBF quanto o Menoflavon® não são capazes de estimular a osteogênese in vitro, a partir de cultura de osteoblastos derivados de células tronco mesenquimais. / Menopause is caused by failure in the production of ovarian hormones and its consequences are unfavorable changes in metabolism and bone loss. The decline in estrogen production is considered a major risk factor for the development of osteoporosis in women, and the Hormone Replacement Therapy is used as treatment. However, this therapy has brought some risks to the health of some groups of women. As an alternative to the traditional treatment, phytoestrogens as isoflavones, found mainly in soy, Trifolium pratense, Cimicifuga racemosa and rye can be used. This study aimed to compare the ability of two preparations of phytoestrogens to stimulate osteogenesis in vitro (from cultures of osteoblasts derived from mesenchymal stem cells): soy extract biotransformed by the fungus Aspergillus awamori (ESBF), and Menoflavon® 40mg (Melbrosin International) composed by the isoflavone Trifolium pratense. With this objective, were used: a) Evaluation of cell growth and proliferation; b) Viability of the cultures of osteoblasts; c) Determination of total protein from the cultures of osteoblasts; d) Determination of the specific activity of the enzyme alkaline phosphatase; e) Formation of mineralized matrix. Menoflavon® was tested at the concentrations of 28.75 nM of daidzein (D) + 7.5 nM of genistein (G) (Menoflavon® 0.5 ?g/mL); 57.5 nM D + 15 nM G (Menoflavon® 1 ?g/mL); and 230 nM D + 60 nM G (Menoflavon® 4 ?g/mL); standard control of commercial 57.5 nM D + 15 nM G and DMSO control. ESBF was tested at the concentrations of 1.181 nM D + 0.922 nM G (ESBF 0.5 ?g/mL); 2.361 nM D + 1.845 nM G (ESBF 1 ?g/mL); 9.445 nM D + 7.379 nM G (ESBF 4 ?g/mL); standard control of commercial 2.361 nM D + 1.845 nM G and dimetilsulfoxide (DMSO) control. With the MTT and Resazurin methods we verified that there was no cell death for all concentrations tested with the two formulations. The amount of total protein remained constant with the two formulations, and the formation of mineralized matrix also were the same as the control. The specific activity of alkaline phosphatase decreased significantly on day 14 for all concentrations tested, and at day 21 for some concentrations of Menoflavon®, with the ESBF decreased some days, however remained constant in the other tests. Therefore, ESBF proved better than Menoflavon®, since we obtained similar results for both, but the concentration of ESBF is 24 times smaller than the concentration of Menoflavon®. However, both the ESBF and the Menoflavon® were not capable of stimulating osteogenesis in vitro from cultures of osteoblasts derived from mesenchymal stem cells.
