The Use of Lignin Derivatives to Improve Selected Paper Properties

<p>Ved består huvudsakligen av tre typer av polymerer, cellulosa, hemicellulosa och lignin. Lignin bildas i naturen genom enzymatiskt initierad oxidativ koppling av tre olika typer av fenylpropan-enheter. Dessa bygger genom olika kol-kol- och kol-syre-bindningar upp en amorf tredimensionell polymer. När kemisk massa tillverkas bryts lignin ner och löses ut i kokluten. Luten innehåller de förbrukade kokkemikalierna och bränns generellt i en sodapanna för att regenerera kemikalierna och producera ånga. Sodapannan är emellertid dyr. Därför har den blivit produktionsbegränsande på många massabruk. Att avlägsna en del av ligninet från avluten vore därför önskvärt och att finna ekonomiskt intressanta produkter baserade på lignin från svartlut är därför ett viktigt forskningsområde .</p><p>Ett lämpligt område för ligninprodukter vore som tillsatts i oblekt massa. Oblekt massa används till stor del för tillverkning av kraftliner, topp- och bottenskikten på wellpapp. När lådor av wellpapp lagras i containrar som färdas över haven, förändras den relativa luftfuktigheten. Detta gör att lådorna kollapsar lättare än om de skulle ha lagrats vid konstant luftfuktighet, även en hög sådan. Detta är på grund av det så kallade mekanosorptiva- eller accelererade krypfenomenet. Genom tillsatts av våtstyrkemedel till kraftliner eller behandla den med hydrofoba ämnen, finns indikatoner på att mekanosorptiva effekten skulle kunna minska.</p><p>För att försöka minska den effekten har ett lågmolekylärt kraftlignin, som utvunnits med hjälp av tvärsflödesfiltrering av svartlut och svavelsyrafällning, använts. Genom derivatisering av detta lignin med linolja erhölls ett hydrofobt ligninderivat som uppvisar strukturella likheter med biopolymeren suberin. När detta suberinlika ligninderivat tillsätts till massa verkar det mekanosorptiva krypet minska. När lågmolekylärt lignin används tillsammans med ligninradikalinitiatorerna lackas eller mangan(III) i kraftlinermassa erhålls dessutom en våtstyrka på ca 5% av torrstyrkan. Efter aminering av detta lignin gav en tillsatts till kraftlinermassan en våtstyrka på upp till 10% av torrstyrkan. Det finns indikationer på att det mekanosorptiva krypet samtidigt minskar när dessa behandlingar görs som ger upphov till ökad våtstyrka.</p> / <p>Wood consists mainly of three types of polymers; cellulose, hemi cellulose and lignin. Lignin is formed in nature through enzymatic initiated oxidative coupling of three different kinds of phenyl propane units. These form by various carbon-carbon and carbon-oxygen bonds, an amorphous three-dimensional polymer. As chemical pulp is produced, lignin is degraded and dissolved into pulping liquors. These liquors contain the spent cooking chemicals and are generally burnt in a recovery boiler to regenerate cooking chemicals and produce steam. However, the recovery boiler is expensive. Hence, it has become the bottleneck for production in many pulp mills. Removal of some lignin from the spent cooking liquor would, for that reason, be desired and valuable products based on lignin from cooking liquors are searched for.</p><p>One suitable area for lignin products would be as additive in unbleached pulp. A major product from unbleached pulp is kraftliner, the top and bottom layers of corrugated board. When boxes of corrugated board are stored in containers travelling overseas the relative humidity is varying. This makes the boxes collapse more easily than if they were stored at constant humidity, even a high one. This is due to the so called mechano-sorptive or accelerated creep phenomenon. By addition of wet strength additive to kraftliner or treating it with hydrophobic compounds there are indications on that the mechano-sorptive effect would decrease.</p><p>Trying to decrease this effect, low molecular weight kraft lignin has been used. It was obtained by cross-flow filtration of black liquor and precipitation by sulphuric acid. By derivatisation of this lignin by linseed oil, a hydrophobic lignin derivative was obtained, similar in structure to units in the biopolymer suberin. As this suberin-like lignin-derivative was added to pulp the mechano-sorptive creep seemed to be lowered. Furthermore, when the low molecular weight lignin was used together with the lignin radical initiators laccase or manganese(III) in kraftliner pulp, a wet strength of about 5% of dry strength was obtained. An amination treatment of this lignin and addition to kraftliner pulp resulted in a wet strength of up to 10% of dry strength. There are indications of that the mechano-sorptive creep also decreases as these treatments, resulting in increased wet strength, are made.</p>

<p>Emma Eriksson and Christina Henriksson: Mirror mirror on the wall: A study on how ballad texts reflect women as carriers of tradition in nineteenth century Sweden, Växjö: Musikvetenskap C, 10 p 2005.</p><p>The aim of this essay is to analyze relations between ballad texts and the lives of female carriers of tradition in nineteenth century Sweden. Four carriers of tradition have been chosen, connected by being informants to Gunnar Olof Hyltén Cavallius, a collector of medieval ballads in Sweden around 1850. The four women are Anna Ehrenström (1786-1857), Catarina Andersdotter (1792-1857), Johanna Gustafva Angel (1791-1869) och Wendela Hebbe (1808-1899). Their lives are seen from a women’s historical perspective.</p><p>Although there are uncertainties regarding to what extent the collected repertoire of the four women reflects their repertoire as a whole, some conclusions can be drawn regarding relations between remaining ballads and their lives as women. These relations are traced in textual themes and in textual form (e.g. repetition of words and phrases). The closeness between life situation, and choice and shape of ballad texts, leads us to conclude that these occurrences are due to deliberate choices, and that the ballad text thus mirror the lives of these women.</p>

Wir schlagen einen allgemein anwendbaren Algorithmus vor, der unter Verwendung des Skalarprodukts von Kraft und Weg zum richtigen Vorzeichen in den Gleichungen für die Arbeit und die Potentielle Energie bei reversiblen Prozessen (Druck-Volumen-Änderung, Dehnung, Elektrostatische Wechselwirkung, Hub)führt. Wir zeigen, dass es dabei möglich ist, systemimmanente oder externe Kräfte zu benutzen. Wir zeigen, dass bei Verwendung von systemimmanenten Kräften das Skalarprodukt mit negativem Vorzeichen anzusetzen ist. Zudem ist es sehr wichtig, nötige Vorzeichenwechsel bei den einzelnen Schritten zu beachten. Wir betonen dies, weil gelegentlich übersehen wird, dass ein Vorzeichenwechsel nötig ist, wenn das Wegdifferential ds durch das Höhendifferential dh beziehungsweise durch das Abstandsdifferential dx oder dr ersetzt werden muss.

Short-term regulating capacity and operational patterns of The Lule River with large wind power penetration

The growing share of installed wind power in the Swedish electricity system has caused concerns whether the available regulating power will be sufficient. Several studies have examined the need of regulating power using both statistical and modelling approaches. However, there is a risk that some aspects of the short-term regulation of hydropower might have been missed. By using one of Vattenfall’s hydropower planning tools, the short-term operation of The Lule River has been simulated with an increasing penetration of wind power. The tool includes detailed models of reservoirs, generating units including efficiency curves and start/stop costs. By introducing a day-by-day simulation with a seven-day window price forecast, updated with a new wind forecast for each iteration, a 21-days scenario has been simulated. Transmission limits are disregarded and the thermal production is reduced with the average wind production. To quantify and compare the regulation capacity, the regulation factor is introduced. It reflects the ability to utilise high-price hours and considering that the need of regulating power for the short-term perspective is reflected in the price it will also reflect the regulation capacity. It is shown that the regulating factor is correlated to the discharge factor,whichis the relation between the maximum discharge to the average statistical discharge for a plant. A high discharge factor provides the flexibility to utilise the fluctuations in price. The discharge factor is adapted to the plants placement in the reach, accounting for both reservoirs located upstream and downstream, especially for The Lule River which has been designed to regulate for the fluctuations in the load. The flexibility required by the rest of the Nordic rivers is quantified for future studies. It is concluded that The Lule River is able to meet some of the fluctuations of wind power production due to the overcapacity ininstalled power. The production can, at the expense of decreased efficiency of the generating units, alter the production to suit a more fluctuating price.It is important to emphasise that The Lule River alone cannot balance a large penetration of wind power. To fully take into account the effects of a large penetration of wind power the study must be expanded to include more scenarios. The study should include different types of hydrological prerequisites and the seasonal variations in power production as well as additional rivers.

Bakgrunn og hensikt: For mennesker som blir akutt/kritisk syk blir livet oftest endret på kort tid fra å værevelfungerende i hverdagen til innleggelse i intensivavdeling hvor selveste livet kan stå på spill. Tradisjonelt settblir intensivpasienter tatt vare på gjennom medisinsk kunnskap og et naturvitenskapelig kunnskapsparadigme. Åvære akutt/kritisk syk innebærer å få spesielle opplevelser og erfaringer som går ut over hverdagserfaringene, oghvordan disse erfaringene trer fram i situasjonen. Denne kunnskapen pasientene bærer i seg er ikke bareindividuell og privat kunnskap, den representerer også en form for viten som trenger å forskes fram og løftes inni det offentlige rom både folkehelsevitenskapelig og sykepleievitenskaplig, nettopp fordi den omhandler viktigeforutsetninger for at pasientene skal bli sett og møtt også eksistensielt. Denne kunnskapen representerer en annenontologi og epistemologi enn den naturvitenskapelige kunnskapen og representerer derfor et annetkunnskapsperspektiv, også når det gjelder kunnskapsformidlingen i en klinisk veiledningssammenheng. Avhandlingens overordnede mål er å øke forståelsen av det å være alvorlig syk og hvordan kunnskap ut fraerfaringer har betydning i kunnskapsformidling i en intensivkontekst. Data og metoder: Avhandlingen har en kvalitativ tilnærming. I delstudie I ble det gjort en review avartikler for å se på stress relatert til kropp, rom og relasjon. I delstudie II ble det gjennomført intervju medtidligere intensivpasienter om opplevelsen av kropp, kraft og bevegelse under kritisk sykdom. I delstudie III bletidligere intensivpasienter intervjuet angående deres opplevelse av kroppsnærværet til pårørende under kritisksykdom. I delstudie IV ble det gjort observasjoner med påfølgende intervjuer av intensivsykepleiere om deresformidling av erfart og fortolket kunnskap i en klinisk veiledningssituasjon. I alle fire delstudiene benyttes detfenomenologisk-hermeneutiske analyser. Funn: I delstudie I kommer det fram at pasientene opplever stress knyttet til kropp, rom og relasjon.Kroppen påvirkes gjennom søvnavbrudd, smerter, angst og pasientene mister kontrollen over kropp og situasjon.Stress knyttet til rom framkommer gjennom at rommet blir et speil av situasjonen ved at den både representererlivet, men også redselen for døden. Gjennom det horisontale leie opplever pasientene at situasjonen utgjør enform for makt og de selv settes i avmakt. Relasjonelt stress framkommer oftest knyttet til sammenhenger hvordet utføres observasjoner, stell og behandling. Travelhet og organisering i avdelingen påvirker det relasjonellesamværet i form av stress. I delstudie II opplever intensivpasientene at kropp kraft og bevegelse trer fram bådegjennom ”tapet” av kroppen, hvordan de handterer situasjonen og hvordan de gjenerobrer kropp, kraft ogbevegelse. De opplever kraftløshet og bevegelsesbergrensninger. Dette innvirker på avhengigheten til andre ogskaper konflikter til pårørende. Pasientene handterer det kroppslige tapet gjennom mobilisering av familiengjennom å involvere dem. Samtidig blir de selvbeskyttende og utestenger familiesammenhengene. Kampen forlivet håndterer de gjennom galgenhumoren og skriket om hjelp som siste nødrop. Gjenerobringen av kroppen erknyttet til små framskritt, gjennom personalets motivasjon og gjennom drømmen om å utrette noe i livet. Idelstudie III opplever pasientene konflikt mellom nærheten og distanse til pårørende. De kjenner seg utestengt,samtidig som de bekreftes med gaver. Pasientene opplever det konfliktfylt når de sammenligner pårørendesreaksjoner og egen situasjon. De er redd at fellesskapet med pårørende skal opphøre, samtidig må de begrensesamværet. Å ikke kunne kommunisere med pårørende i kroppsnærværet oppleves motsetningsfullt. I delstudieIV formidler sykepleierne kunnskap til intensivstudentene gjennom meningsskapende kunnskapsbevegelser. Deformidler inntrykk fra situasjonen for å skape oversikt, de formidler kunnskap om fenomenene som framtrer ogde formidler hvordan de kan hjelpe kroppens egne prosesser. De vurderer og formidler også pasienteneskroppslige uttrykk og hvordan disse kan forstås og imøtekommes. Sykepleierne er også opptatt å formidlekunnskap for at studentene skal utvikle og få egne erfaringer i kroppen. Konklusjon: Gjennom fire delstudier løfter avhandlingen fram kunnskap som står i klar kontrast til dennaturvitenskapelige kunnskapen. Pasientene utsettes for store opplevelsesmessige belastninger i situasjonen somakutt/kritisk syke som setter de i en avmaktssituasjon ved at sykepleierne ikke fanger opp og handterer kroppensegen kunnskap i situasjonen. Pasientene befinner seg ofte i en eksistensiell situasjon som er preget avmarginalitet og eksistensiell væren. Pasientene viser at de har forutsetninger for å mestre situasjonen, menhjelpes lite til slik mestring. Intensivsykepleierne formidler ulikt faglig innhold til studentene for å skapeerfaringer og forståelse i situasjonen. Erfaringskunnskapen fra intensivkonteksten må derfor sees som et viktigkunnskapssupplement til folkehelsearbeidet. / Background and purpose: Acute/critically ill patients have had their life-conditions severely altered from awell-functioning everyday situation to a situation in which they struggle to survive. Traditionally the knowledgeparadigmfrom natural sciences together with medical expertise constitutes the framework for treatment. To beacute/critically ill involves experiences and events and phenomena typical to the situation. The patients’embodied knowledge is not limited to the personal and private sphere, it refers to highly relevant preconditionsfor visibility and reception, also on an existential level; it represents knowledge which needs to be researchedand discussed openly within the public health and nursing sciences. This knowledge represents a differentontology and epistemology from the natural sciences and thus a different perspective, also with regards toclinical tutoring. The dissertation’s primary target is to increase the knowledge about how critical illness isexperienced and how experience based knowledge is significant to teaching in an intensive care context. Data and methods: The dissertation is based on qualitative methods. Sub-study I includes reviews ofarticles on stress in a perspective of body, space and relationship. Sub-study II contains interviews with formerICU patients relating to their experience of body, strength and movement during critical illness. Sub-study III isbased on former ICU patients’ experience of the physical presence of their significant others during criticalillness. Sub-study IV contains observations and interviews of ICU nurses on how they teach experience basedand interpretation based knowledge in clinical tutoring. Phenomenological-hermeneutic analysis method isapplied to all four sub-studies. Findings: Sub-study I describes how patients experience stress in a perspective of body, space andrelation. The body is influenced by sleep deprivation, pain and anxiety. Patients experience loss of control withtheir bodies as well as with the situation. The room mirrors the situation patients are in, creating stress by itsrepresentation of life as well as fear of dying. The patient’s horizontal position reflects disproportionate powerinducing a sense of disempowerment. Relational stress situations usually appear in contexts involvingobservation, care or treatment. The organizational structure and activity on the ward affects stress levels. In substudyII the ICU patients experience how power and movement is affected by a sense of loss of body, how theymanage to respond to the situation and how they eventually reclaim the body, power and movement. Theyexperience disempowerment and limitations to movement. This causes dependency and creates conflicts withtheir significant others, especially with regards to proximity and distance. The patients address their loss of bodyby drawing on resources from their significant others, yet refrain from allowing themselves involvement infamily matters. The struggle for survival is characterised by gallows humour and the cry for help as their finalcry. Reclaiming the body happens stepwise with the aid of staff combined with a need to achieve. Sub-study IIIdescribes problems related to proximity to, and distance from, the significant others. A sense of exclusion iscontradicted by being showered with gifts. It is unsettling for patients to have their situation compared to theirsignificant others’. They fear the loss of community with loved ones, yet they need to limit visits fromsignificant others. The lack of ability to communicate when others are physically present is unsettling. Sub-studyIV describes how ICU nurses tutor students in practise by demonstrating knowledge in meaningful actions. Theyconvey their impressions of situations to provide an overview, convey knowledge of observable phenomena, andteach skills to assist the body’s own processes. They also assess and evaluate physical expressions in the patientand how these can be read and addressed. Nurses are also concerned with conveying embodied knowledge totheir students. Conclusion: In four sub-studies this dissertation highlights a form of knowledge which clearly contraststhat of the natural sciences. During acute critical illness patients experience severe stress and disempowermentcaused by a lack of response and action from nurses who are unable to interpret the body’s own knowledge inthe situation. Patients are often facing life and death questions characterised by marginality and existentiality.Patients demonstrate ability to cope with the situation, but are rarely assisted with coping. Intensive care nursesdisseminate a variety of professional skills and knowledge to their students in order to provide comprehensionand understanding of situations. Experience based knowledge from an intensive care context thus becomes asignificant supplementary factor in the public health perspective

The Planck Constant and the Origin of Mass due to a Higher Order Casimir Effect

The Planck constant is one of the most important constants in nature, as it describes the world governed by quantum mechanics. However, it cannot be derived from other natural constants. We present a model from which it is possible to derive this constant without any free parameters. This is done utilizing the force between two oscillating electric dipoles described by an extension of Weber electrodynamics, based on a gravitational model by Assis. This leads not only to gravitational forces between the particles but also to a newly found Casimir-type attraction. We can use these forces to calculate the maximum point mass of this model which is equal to the Planck mass and derive the quantum of action. The result hints to a connection of quantum effects like the Casimir force and the Planck constant with gravitational ones and the origin of mass itself.

Test-retest reliability and construct validity of three golf specific rotational power tests in 1080 Quantum : and a presentation of the power-force-velocity profiles of elite level golfers

Background: Evaluation of power-force-velocity profiles in ballistic push-offs and in sprint movements are beneficial to enhance sport performance. There have only been a few studies investigating force-velocity profiles in rotational movements and there have not been any studies conducted on golf specific rotational movements. There is a lack of isoinertial power assessment protocols which can relate to golf swing performance. For this reason, 1080 Quantum was used for testing which is a machine able to measure power, force and velocity in several different resistance modes. Aim: The aim was to study the reliability of three golf specific rotational tests and to assess the reliable test’s validity. The secondary aim was to study the isoinertial force-velocity and power relationship in tests that assessed acceptable test-retest reliability and acceptable construct validity. Methods: Twelve high level golfers (handicap -1.5±1.2), 8 men and 4 women performed the tests with five different loads; 2 kg, 6 kg, 10 kg, 14 kg and 18kg with three golf specific rotational movements in a motorized cable machine (1080 Quantum, Sweden). The three new tests utilized were: full body rotational test (FBRT), thorax rotational test (TRT) and pelvis rotational test (PRT). Data gathered was used to study golfer’s individual force-velocity profile and to assess construct validity of the reliable tests against previously recorded normal-swing driver clubhead speed (CHSnormal), and maximum-swing driver clubhead speed (CHSmax). Construct validity was based on á priori stated hypothesizes, and data were analyzed with Spearman’s correlation coefficient (rs). Intraclass correlation coefficient (ICC) and coefficient of variation (CV) was used to assess test-retest reliability of FBRT, TRT and PRT. Results: In the reliability assessment, the highest peak power (PP) (ICC, 0.968, 95% CI (0.889- 0.991)), corresponding peak force (PF) (ICC, 0.993, 95% CI (0.96- 0.998)) and corresponding peak velocity (PV) (ICC 0.773, 95% CI (0.17- 0.936)) was only found to be acceptable in FBRT. Construct validity assessment of FBRT was not found acceptable in either of the resistances. The calculated average slopes indicated a velocity dominant force-velocity profile for women and a force dominant for men. Conclusion: The study demonstrated that only the FBRT assessed acceptable test-retest reliability for measuring force, velocity and power. Based on our hypothesizes FBRT is not a valid indicator of golf performance on either resistance, however it had a significant moderate to good correlation with CHSmax and with CHSnormal at all resistances apart from 2 kg. FBRT should not be applied as a test, nor to be used for the calculation of the force- velocity profile since it is not valid, even though it is reliable. since it is not valid, even though it is reliable. As a result, FBRT could only be used for exercise and needs further development to improve its validity. / <p>While only my name is featured on the title page of this thesis, it reached its final stage with the guidance and support of many others. Die Proteinfaltung ist ein Prozess der molekularen Selbstorganisation, bei dem sich eine lineare Kette von Aminosäuren zu einer definierten, funktionellen dreidimensionalen Struktur zusammensetzt. Der Prozess der Faltung ist ein thermisch getriebener diffusiver Prozess durch eine Gibbs-Energie-Landschaft im Konformationsraum für die Struktur der minimalen Energie. Während dieses Prozesses zeigt die freie Enthalpie des Systems nicht immer eine monotone Abnahme; stattdessen führt eine suboptimale Kompensation der Enthalpie- und der Entropieänderung während jedes Faltungsschrittes zur Bildung von Freien-Enthalpie-Faltungsbarrieren. Diese Barrieren und damit verbundenen hochenergetischen Übergangszustände, die wichtige Informationen über Mechanismen der Proteinfaltung enthalten, sind jedoch kinetisch unzugänglich. Um den Prozess der Barrierebildung und die strukturellen Merkmale von Übergangszuständen aufzudecken, werden Proteine genutzt, die über barrierefreie Pfade falten – so genannte “downhill folder“. Aufgrund der geringen Faltungsbarrieren werden wichtige Interaktionen der Faltung zugänglich und erlauben Einblicke in die ratenbegrenzenden Faltungsvorgänge. In dieser Arbeit vergleichen wir die Faltungsdynamiken von drei verschiedenen Varianten eines Lambda-Repressor-Fragments, bestehend aus den Aminosäuren 6 bis 85: ein Zwei-Zustands-Falter λWT (Y22W) und zwei downhill-folder-artige Varianten, λYA (Y22W/Q33Y/ G46,48A) und λHA (Y22W/Q33H/G46,48A). Um auf die Kinetik und die strukturelle Dynamik zu greifen zu können, werden Einzelmolekülkraftspektroskopische Experimente mit optische Pinzetten mit Submillisekunden- und Nanometer-Auflösung verwendet. Ich fand, dass die niedrige denaturierende Kraft die Mikrosekunden Faltungskinetik von downhill foldern auf eine Millisekunden-Zeitskala verlangsamt, sodass das System für Einzelmolekülstudien gut zugänglich ist. Interessanterweise zeigten sich unter Krafteinwirkung die downhill-folder-artigen Varianten des Lambda-Repressors als kooperative Zwei-Zustands-Falter mit deutlich unterschiedlicher Faltungskinetik und Kraftabhängigkeit. Drei Varianten des Proteins zeigten ein hoch konformes Verhalten unter Last. Die modellfreie Rekonstruktion von Freien-Enthalpie-Landschaften ermöglichte es uns, die feinen Details der Transformation des Zwei-Zustands-Faltungspfad direkt in einen downhill-artigen Pfad aufzulösen. Die Auswirkungen von einzelnen Mutationen auf die Proteinstabilität, Bildung der Übergangszustände und die konformationelle Heterogenität der Faltungs- und Entfaltungszustände konnten beobachtet werden. Interessanterweise zeigen unsere Ergebnisse, dass sich die untersuchten Varianten trotz der ultraschnellen Faltungszeit im Bereich von 2 μs in einem kooperativen Prozess über verbleibende Energiebarrieren falten und entfalten, was darauf hindeutet, dass wesentlich schnellere Faltungsraten notwendig sind um ein downhill Limit vollständig zu erreichen. / Protein folding is a process of molecular self-assembly in which a linear chain of amino acids assembles into a defined, functional three-dimensional structure. The process of folding is a thermally driven diffusive search on a free-energy landscape in the conformational space for the minimal-energy structure. During that process, the free energy of the system does not always show a monotonic decrease; instead, sub-optimal compensation of enthalpy and entropy change during each folding step leads to formation of folding free-energy barriers. However, these barriers, and associated high-energy transition states, that contain key information about mechanisms of protein folding, are kinetically inaccessible. To reveal the barrier-formation process and structural characteristics of transition states, proteins are employed that fold via barrierless paths – so-called downhill folders. Due to the low folding barriers, the key folding interactions become accessible, yielding insights about the rate-limiting folding events. Here, I compared the folding dynamics of three different variants of a lambda repressor fragment, containing amino acids 6 to 85: a two-state folder λWT (Y22W) and two downhill-like folding variants, λYA (Y22W/Q33Y/G46,48A) and λHA (Y22W/Q33H/G46,48A). To access the kinetics and structural dynamics, single-molecule optical tweezers with submillisecond and nanometer resolution are used. I found that force perturbation slowed down the microsecond kinetics of downhill folders to a millisecond time-scale, making it accessible to single-molecule studies. Interestingly, under load, the downhill-like variants of lambda repressor appeared as cooperative two-state folders with significantly different folding kinetics and force dependence. The three protein variants displayed a highly compliant behaviour under load. Model-free reconstruction of free-energy landscapes allowed us to directly resolve the fine details of the transformation of the two-state folding path into a downhill-like path. The effect of single mutations on protein stability, transition state formation and conformational heterogeneity of folding and unfolding states was observed. Noteworthy, our results demonstrate, that despite the ultrafast folding time in a range of 2 µs, the studied variants fold and unfold in a cooperative process via residual barriers, suggesting that much faster folding rate constants are required to reach the full-downhill limit.

Varje kommun ska ha en byggnadsnämnd. Byggnadsnämnden består av förtroendevalda poli-tiker som bland annat ansvarar för lovärendeprocessen. Tjänstemännen som jobbar med lovä-rendeprocessen är anställda av byggnadsnämnden. Tjänstemännens huvuduppgift är att ta be-slut i lovärenden via delegation från byggnadsnämnden. I lovärenden där delegation saknas ska tjänstemännen bereda lovärendet och skriva ett förslag till beslut. Förslaget ska sedan redovisas inför byggnadsnämnden, som därefter tar det slutliga beslutet. I denna uppsats har enbart lovä-renden där byggnadsnämnden och tjänstemännen har haft olika bedömningar granskats. Det har även granskats vilken laghänvisning och bedömning som ligger till grund för deras förslag eller beslut. Kommunen ska ge berörda grannar och sakägare tillfälle att yttra sig när det är en avvikelse från en detaljplan, områdesbestämmelser eller utanför detaljplan. Vid ett positivt lovbeslut ska kommunerna kungöra lovärendet i Post och Inrikes Tidningar. Båda dessa uppgifter måste ge-nomföras på rätt sätt för att hanteringen ska vara rättssäker och kvalitetssäker samt för att lo-värendet ska vinna laga kraft. Detta arbete ska undersöka hur kommunerna hanterar rättssäkerheten och kvalitetssäkerheten i lovprocessen för att ett lovärende ska vinna laga kraft. För att få vidare rådgivning, kunskap och information angående lagtolkning gällande granne-hörande och kungörelse togs kontakt med Ulrika Nolåker, Byggutbildarna, Ulf Jensen, profes-sor på Högskolan Väst och Eidar Lindgren universitetsadjunkt på Kungliga Tekniska Högsko-lan. I undersökningen studerades fyra kommuner i Västra Götalands län. Undersökningen delades upp i tre delar som bestod av intervju dels av tjänstemän och sakkunniga samt granskning av lovärenden. I kommunerna var det 1386 lovärenden som kom in och togs beslut under 2015. Av dessa var det 23 lovärenden där byggnadsnämnden inte tog beslut i enlighet med tjänste-männens förslag. Kommunernas lovärendeprocess saknar regelbundet arbetssätt både när det gäller grannehöran och kungörelse av lovärenden. Kommunerna använder en så kallad "tyst accept" vid granne-höran, vilket innebär att grannarna förlorar sin demokratiska rättighet att påverka åtgärden. Genom att hänvisa lagparagraf i ett beslut kvalitetssäkras beslutsunderlag. Att hänvisa till en lagparagraf är något som varken byggnadsnämnden eller tjänstemännen gör regelbundet. Kungörelsen i Post- och Inrikes Tidning gjordes mellan 12-28 dagar efter beslutet togs samt att det förkom lovärenden som inte var kungjorda överhuvudtaget. Endast tre av de 23 grans-kade lovärendena anses ha vunnit laga kraft. / Each municipality in Sweden must have a local building committee. The committee consists of elected politicians responsible for managing the permit process related to construction, dem-olition and ground processing. The committee manages the permit process by delegating the entire task or parts of it to employed officials. If there is no delegation the officials will write a suggested decision to be reviewed and decided by the committee. This essay investigates construction-, demolition- and ground processing permit where the of-ficials and the local building committee differ in their decisions and the assessment which led to these decisions. The examination included studies of four municipalities in the county of Västra Götaland: two smaller, Färgelanda and Munkedal municipalities, and two larger, Trollhättan and Uddevalla. The investigation consisted of two parts. One examination of the permit protocols, and a series of interviews with officials. During 2015 there were 1386 cases of permits within the four municipalities. 23 of these were cases of the committee not deciding in accordance with the proposition of the officials. A number of errors were also found, such as there not being a stated reason for the decision, or lack of any clear legal reference. This raises the question of whether or not the rule of law is followed during these circumstances. After consulting with Ulrika Nolåker from Byggutbildarna, the issues were expanded further when it was discovered that more errors occur during the municipal management of the con-cerned parties and the announcements of the cases. In order to receive further guidance, knowledge and information on how to interpret the laws handling concerned parties and an-nouncements, contact was made with Ulf Jensen and Eidar Lindgren, professor in real estate science at Högskolan Väst, and lecturer of real estate sciences at Kungliga Tekniska Högsko-lan, respectively. The conclusions of the examination is that the building committee puts a lot of responsibility on the officials through delegations. The cases when there is no delegation and the case is decided by the committee are few. The reasons why the proposition and the final decision not always matches is due to different factors such as different knowledge or different assessments. Neither the committee nor the officials are consistent with the law referencing in the decisions. The municipality cannot guarantee that every concerned party have been given the chance to give their opinion, when some of them use what’s called "silent agreement". Only three of the cases has the municipality received and opinion from every concerned party. None of the cases has been announced in the correct time and two has not been announced at all.
