Cytotoxicity of in vitro exposure of polystyrene latex bead nanoparticles to human keratinocyte (HaCaT) cells and human cervical cancer (HeLa)cells

Yes / Nanoparticles are increasingly used in industry and medicine due to their unique physiochemical properties such as their small size, charge, shape, chemical architecture, large surface area, surface reactivity and media interactions, etc [1-5]. However, very little is still known on the interactions between nanoparticles and the biological system. This study aims to evaluate the cytotoxicity of polystyrene latex bead nanoparticles on HaCat and HeLa cell lines. Carboxyl-modified 20 nm polystyrene NPs core labelled with fluorophore were from Invitogen. We chose to use polystyrene NPs because this specific type of NP is being increasingly characterized for use in nanosensors and drug nanocarrier investigations. 1x 104 cells/100 μl of cell culture medium were plated into 96-well plates in triplicate, measuring activity post 24 hours at concentrations of 10, 50, 100 μg/ml of polystyrene NPs exposure. The extracellular lactate dehydrogenase release was measured by using a colorimetric CytoTox 96 nonradioactive assay kit from Promega and the absorbance were recorded at 450nm (FLUO-star) with Elisa micro plate reader. The MTT assay was used to evaluate mitochondrial activity. This was performed by inserting a premixed optimized dye solution in the culture wells. The Absorbance was recorded at 570 nm, from the recorded absorbance is directly proportional to the number of live cells. The cell lines were kept in a plastic T-75cm2 tissue culture flasks grown in DMEM. We found that cytotoxicity of polystyrene NPs on both cells was concentration dependent. For Hela cells, with exporesure of polystyrene NPs at concentrations of 10, 50, 100 μg/ml for 24 hrs, the percentage cytotoxicity of positive control for LDH assay was 35.9%, 49.5% and 73.4% respectively. With the MTT cell viability assay the percentage MTT reduction of negative control was 88.9%, 42.9% and 26.4% respectively. Cell toxicity increased with increasing polystyrene NPs concentration. For HaCaT cells, the cytotoxic effect is less significant than those on Hela cells. With MTT assay, when compared to HaCaT cells exposed to a negative control containing only PBS, the cell viability decreased as the concentrations of NPs increased. Cells exposed to 100μg/ml of polystyrene NPs for a period of 24 hours compared to those exposed to a positive control (100% cell viability) had an average cell viability of 49%, with those numbers decreasing from 59% for cells exposed to 10μg/ml of polystyrene NPs to 57% for cells exposed to 50μg/ml of polystyrene NPs. Our results indicated that polystyrene NPs acted differently in two different cell types and that cautions should be taken about its cytotoxicity. Further understanding of the mechanism involving the ROS generation could provide more information on how polystyrene NPs increase cytotoxicity.

Stabilisation of acrylic latexes containing silica nanoparticles for dirt repellent coating applications

Yes / This study examines the feasibility of using colloidal silica nanoparticles as active agents in high concentration waterborne polymer latex formulations. We showed that distributing the silica throughout the waterborne emulsion formed a composite coating material with a hydrophilic surface that consequently reduced exterior dirt pickup. Two grades of silica nanoparticles were studied, one using sodium stabilisation and another using epoxysilane modification to introduce glycidox-ypropyltrimethoxysilane surface functionality. Rheological study of the waterborne latex on mixing showed that there was an immediate pH responsive interaction between the silica sols and the polymer latex. Once loading of sodium charge stabilised silica NPs exceeded the volume required for heteroflocculation to occur the mixture demonstrated the potential to gel on standing – a process which took weeks, or months, to occur depending on the pH and relative concentrations used. At least fifty percent silane modification to the NP surface was found to be necessary to maintain a stable colloidal dispersion for long term storage of the waterborne latex. Despite this both grades of silica were found to imbue reductions in dirt pickup when applied to exterior masonry concrete studies over a 3-month weathering test / This work was supported by the Royal Society of Chemistry [E21-8346952505].

Isolation and characterization of latex-specific promoters from Papaver somniferum L.

The pharmacologically important alkaloids morphine and codeine are found in latex of opium poppy (Papaver somniferum). Latex is harbored in laticifers, a specialized vascular cell-type. Isolation and characterization of latex-specific genes may provide a useful tool to metabolically engineer increased alkaloid production. Previous research in the Nessler laboratory identified genes that exhibit latex-specific gene expression. Latex-specific genes were an 2-oxoglutarate-dioxygense (DIOX), involved in hydroxylation, desaturation and epoxidation reactions, and two of the major latex proteins, MLP146 and MLP149. MLP-like proteins function in fruit ripening in various species that do not have the laticifer cell type. The latex-specific promoters (LSPs) for the three genes were sequenced. The 2.5 kb DIOX promoter was fused to the reporter gene Β-glucuronidase (GUS) to characterize its expression pattern. To assess the functional sites within the DIOX promoter, deletions were made 1.5 kb and 0.14 kb upstream of the ATG start codon, fused to GUS, and transformed into opium poppy, Arabidopsis thaliana, and tobacco (Nicotiana tabacum). The 2.5 kb DIOX:GUS and 1.5 kb EcoRIDIOX:GUS reporter gene constructs showed vascular specific expression in opium poppy, Arabidopsis, and tobacco. The 0.14 kb SpeIDIOX promoter deletion construct showed no activity in opium poppy, and limited expression in the shoot apical meristem and root hypocotyl axis in Arabidopsis. These results indicate that the minimum active DIOX promoter is greater than 0.14 kb. Over 1 kb of the LSPs were sequenced and analyzed for regulatory elements using the Plant cis-acting regulatory DNA elements database, PLACE (http://www.dna.affrc.go.jp/PLACE). Knowledge of the cis-elements and regulatory regions of LSPs would serve as a tool for metabolic engineering of poppy alkaloids. Sixty-five elements were conserved among 2 of the 3 LSPs. Among the cis-elements identified, some are associated with basic functions such as: light regulation, carbon metabolism and plant defense. Other elements include: WRKY elements that are binding sites of transcription factors known for signaling plant defense genes, a vascular cis-element, and a fruit specific element. The presence of plant defense and vascular cis-elements in the LSPs, correlate with the concept that latex is a protective defense mechanism found in the vascular system. The latex-specific promoters isolated and cis-elements identified in this research are potential tools for driving increased alkaloid production in opium poppy. / Master of Science

The plant is wildly distribuited from Guianas until Southeast Brazil. Latex obtained by cutting its stem bark is mixed with water and extensively sold in public markets. People use Janaguba milk (“Leite de Janaguba”- LJ) to treat or prevent different inflammatory disorders such as gastritis, ulcers as well as cancer, among other uses. Despite, there is not enough scientific data confirming these pharmacological properties. In this work, Janaguba milk was preliminarily investigated in an attempt to validate its anti- inflammatory activity. Its major protein fraction (HdLP) was assessed for the presence of anti- inflammatory, anti-nociceptive and toxicological effects. LJ inhibited the inflammatory response induced by carrageenan in rats (98%, p < 0.05), whereas HdLP did it independent of the route of administration. HdLP significantly reduced the infiltration of neutrophils into the peritoneal cavity (96%, p < 0.05) concomitant with increased nitric oxide synthesis in plasma and decrease of pro-inflammatory cytokines IL- 1 and TNF-α in peritoneal fluid. This fraction also prevented vacuolization and Kupfer cell hyperplasia caused by carrageenan in liver. Further, the anti-inflammatory properties were shown to be associated with the protein fraction of LJ. HdLP exhibited anti-inflammation, even after heat-treatment (100 C, 30 min) or proteolysis. Moreover, a pro-inflammatory effect was observed after HdLP treatment. It also suppressed abdominal constrictions in acetic acid-treated mice and reduced paw licking induced by formalin, both in a dose-dependent manner. An acute toxicological evaluation demonstrated no toxic effects after 14 days from LJ and HdLP administration by oral route even when high doses were tested. A qualitative phytochemical analysis showed the absence of lupeol and presence of saponinas, tannins and free steroids in HdLP. However, pharmacological properties are probably not related to them. It is therefore concluded that the latex of Himatanthus drasticus exhibits both the anti-inflammatory and anti-nociceptive activities claimed by its users. The protein fraction of the latex is an important contributor to the pharmacological properties of LJ. Resistance to heat and proteolytic treatment can explain the effectiveness of HdLP even when administered orally. The absence of toxic effects by oral route confirms the potential use of this plant as a phytotherapic agent. / Himatanthus drasticus (Apocynaceae) é uma espécie tradicionalmente utilizada na medicina popular da região nordeste deste país. Sua distribuição ocorre das Guianas ao sudeste do Brasil. O látex desta planta é obtido após uma injúria no caule e amplamente comercializado em mercados públicos. As pessoas ingerem o látex, conhecido como Leite de Janaguba (LJ), para o tratamento e/ou prevenção de diferentes doenças inflamatórias, gastrite, úlcera, câncer, entre outras. Apesar das diversas informações oriundas da medicina popular, não existem relatos científicos suficientes confirmando tais propriedades farmacológicas. Neste trabalho, LJ foi investigado com o propósito de validar sua tão popularmente relatada atividade anti-inflamatória. A fração proteica deste látex (HdLP) também foi investigada quanto à presença de atividades anti-inflamatória, antinociceptiva, além de seus efeitos toxicológicos. LJ inibiu a resposta inflamatória induzida por carragenina em ratos (98%, p < 0.05), assim como o fez a fração HdLP, independente da rota de administração. HdLP reduziu a infiltração de neutrófilos na cavidade peritoneal (96%, p < 0.05). Paralelamente a isto, ocorreu o aumento da síntese de óxido nítrico no plasma e a redução de citocinas pró-inflamatórias (IL-1 e TNF-α) no fluido peritonal. Esta fração também preveniu a vacuolização e hiperplasia das células de Kupfer, causadas pelo efeito da carragenina no fígado. Atividade anti-inflamatória está provavelmente associada com a fração proteica de LJ. HdLP exibiu um efeito anti-inflamatório mesmo após aquecimento (100 C, 30 minutos) ou proteólise. Além desta atividade, um efeito pró-inflamatório foi observado após o tratamento com HdLP. Esta também suprimiu contrações abdominais induzidas por ácido acético e reduziu a lambedura de pata induzida por formalina, ambos de maneira dose-dependente. A avaliação da toxicidade aguda demonstrou a ausência de efeitos tóxicos após tratamento com LJ e HdLP (v.o.) mesmo utilizando doses 5000 vezes maiores que as recomendadas. Uma dosagem qualitativa de fitoquímicos em HdLP mostrou a ausência de lupeol e presença de saponinas, taninos e esteróides livres. Entretanto, as propriedades farmacológicas provavelmente não estão relacionadas com estas moléculas. Desta forma, concluímos que o látex de H. drasticus exibe duas das atividades farmacológicas relatadas por seus usuários e a fração proteica deste látex é um importante contribuinte para tais propriedades medicinais. A resistência ao aquecimento e à proteólise pode explicar a efetividade de HdLP quando administrada oralmente. A ausência de efeitos tóxicos por via oral torna esta planta uma potencial candidata a fitoterápico.

L'objectif de ce travail de thèse est de préparer des latex nanocomposites à base d’argile, la Laponite RD, en émulsion aqueuse, à l'aide de la polymérisation radicalaire contrôlée par transfert de chaîne réversible par addition-fragmentation (RAFT). Les plaquettes de Laponite ont été choisies comme charge inorganique surtout pour leur anisotropie de forme, ce qui pourrait permettre l’elaboration de films nanostructurés, mais aussi pour leurs propriétés thermiques et mécaniques, leur pureté chimique élevée et la distribution uniforme en taille des plaquettes. Des polymères hydrophiles (macroRAFT) à base de polyéthylène glycol (PEG), d’acide acrylique (AA) ou de méthacrylate de N,N- diméthylaminoéthyle (DMAEMA) et comportant des unités hydrophobes d’acrylate de n-butyle (ABu) (dans certains cas) et un groupe trithiocarbonate terminal, ont été tout d'abord synthétisés. Ensuite, l'interaction entre les macroRAFTs et l’argile a été étudiée à travers le tracé des isothermes d'adsorption. En agissant comme des agents de couplage et des stabilisants, ces macroRAFTs ont eté utilisés dans la copolymérisation en émulsion du (méth)acrylate de méthyle et de l’ABu en mode semi-continu en presence d’argile. Des particules de latex hybrides de différentes morphologies ont été obtenues et les morphologies ont été reliées à la nature et à la concentration de l’agent macroRAFT, au pH de la dispersion macroRAFT/Laponite, à la température de transition vitreuse du copolymère final (fonction de la composition du mélange de monomères hydrophobes) et aux conditions de polymérisation. Les analyses par cryo-MET indiquent des plaquettes de Laponite décorées par des particules de polymère (plusieurs particules de latex en surface des plaquettes d'argile), des particules ‘haltère’, janus, ‘carapace’ (particules de latex décorées en surface par les plaquettes de Laponite) ou encore des particules multi-encapsulées (plusieurs plaquettes encapsulées dans chaque particule de latex). Les propriétés mécaniques des films de polymère/Laponite ont été étudiées par spectrométrie mécanique dynamique et corrélées à la morphologie des particules et à la microstructure des films / The aim of this work is to prepare Laponite RD-based nanocomposite latexes by aqueous emulsion polymerization, using the reversible addition-fragmentation chain transfer (RAFT) polymerization. Laponite platelets were selected as the inorganic filler due, especially, to their anisotropic shape, which allows the production of nanostructured films, but also for their thermal and mechanical properties, their high chemical purity and the uniform dispersity of the platelets. Hydrophilic polymers (macroRAFT) composed of poly(ethylene glycol) (PEG), acrylic acid (AA) or N,N-dimethylaminoethyl methacrylate (DMAEMA) and comprising hydrophobic n-butyl acrylate (BA) units (in some cases) and trithiocarbonate terminal group were initially synthesized. Then, the interaction between the macroRAFTs and the clay was studied through the plot of adsorption isotherms. By acting as coupling agents and stabilizers, the macroRAFT agents were used in the emulsion copolymerization of methyl (meth)acrylate and BA by semi-continuous process in the presence of the clay. Hybrid latex particles with different morphologies were obtained and the results were associated to the nature and concentration of the RAFT (co)polymers, to the pH of the macroRAFT/Laponite dispersion, the glass transition temperature of the final copolymer (function of the composition of the hydrophobic monomers mixture) and to the polymerization conditions. The cryo-TEM images indicate the formation of polymerdecorated Laponite platelets (several latex particles located at the surface of the platelets), dumbbell-like, janus, Laponite-decorated (armored) latex particles, and multiple encapsulated particles (several platelets inside each latex particle). The mechanical properties of polymer/Laponite films were studied by dynamic mechanical analysis and correlated with the particles morphology and the films microstructure / Este trabalho de tese tem como objetivo a preparação de látices nanocompósitos à base da argila Laponita RD em emulsão aquosa, via polimerização radicalar controlada por transferência de cadeia via adição-fragmentação reversível (RAFT). A Laponita foi escolhida como carga inorgânica devido principalmente à forma anisotrópica de suas lamelas, o que permite a elaboração de filmes nanoestruturados, mas também por suas propriedades térmicas e mecânicas, por sua alta pureza química e pela distribuição uniforme, em termos de tamanho, de suas partículas. Inicialmente, polímeros hidrofílicos (macroRAFT) à base de poli(etileno glicol) (PEG), de ácido acrílico (AA) ou de metacrilato de N,N-dimetilaminoetila (DMAEMA) que contêm unidades hidrofóbicas de acrilato de nbutila (ABu) (em alguns casos) e um grupo tritiocarbonílico terminal foram sintetizados. Em seguida, a interação entre os macroagentes de controle (macroRAFTs) e a argila foi estudada através de isotermas de adsorção. Atuando como agentes de acoplamento e estabilizantes, esses macroRAFTs foram então utilizados na copolimerização em emulsão do (met)acrilato de metila e do ABu em processo semicontínuo na presença da argila Laponita. Partículas de látex híbrido de diferentes morfologias foram obtidas e os resultados foram correlacionados à natureza e à concentração dos macroRAFTs, ao pH da dispersão macroRAFT/Laponita, à temperatura de transição vítrea do copolímero final (função da composição da mistura de monômeros hidrofóbicos) e às condições de polimerização. As análises de cryo-TEM indicam a formação de lamelas de Laponita decoradas com partículas de polímero (várias partículas de látex localizadas na superfície das lamelas), de partículas do tipo dumbbell, janus, blindadas (partículas de látex decoradas com lamelas de argila em sua superfície) ou ainda de partículas multiencapsuladas (diversas lamelas encapsuladas dentro de uma única partícula de látex). La synthèse de macroalcoxyamines hydrosolubles à partir d’acide méthacrylique et d’un faible pourcentage de styrène sulfonate de sodium a été étudiée en solution aqueuse. Ces macroalcoxyamines ont été utilisées comme macroamorceurs pour la polymérisation en émulsion sanstensioactif du méthacrylate de méthyle et du méthacrylate de n-butyle avec un faible pourcentage destyrène. Les copolymères diblocs amphiphiles produits s’auto-assemblent in situ et conduisent, en fonction de leur taux d’hydrophilie, à l’obtention de nano-objets présentant des morphologies variées(sphères, fibres, vésicules). La plus intéressante est la morphologie fibrillaire car les suspensions correspondantes présentent des propriétés rhéofluidifiantes à basse concentration et possèdent un comportement viscoélastique à haute concentration. La synthèse de ces latex a été étudiée et développée dans des conditions monotopes les plus proches possibles d’une production industrielle. / Water-soluble, SG1-based macroalkoxyamines composed of methacrylic acid with a low percentage of sodium 4-styrene sulfonate were synthesized in aqueous solution. These macroalkoxyamines were used as macroinitiators in the surfactant-free emulsion polymerization ofmethyl methacrylate or n-butyl methacrylate with a low percentage of styrene. The polymerization induced self-assembly of the so-formed amphiphilic block copolymers leads to nanoobjects with various morphologies (spheres, fibers, vesicles), depending on their hydrophilic/hydrophobic balance. The most interesting morphology is the nanofiber type of organization as the suspensions presentshear-thinning properties at low concentration and viscoelastic behavior at high concentration. The synthesis of these latexes was investigated and developed in one-pot conditions in order to allow further production at the industrial scale.

Compréhension du suivi de polymérisation d’un système réactif phénolique fortement dilué en présence d’une phase latex pour l'élaboration d’un adhésif sans formaldéhyde / Understanding of polymerization monitoring of a highly diluted phenolic reactive system in the presence of a latex phase for the preparation of a formaldehyde-free adhesive

Un nouveau système réactif de type phénolique plus respectueux de l’environnement, c’est-à-dire sans phénol, résorcinol ou formaldéhyde a été caractérisé lors de ces travaux de thèse. Ce système implique la copolymérisation du phloroglucinol et du téréphthalaldéhyde (TPA) en solution aqueuse très diluée. Dans un premier temps, une recherche bibliographique a permis de sélectionner les techniques expérimentales pertinentes pour suivre l’avancement de ces systèmes évolutifs complexes en solution. La combinaison d’analyses chromatographiques et spectroscopiques a été nécessaire pour la caractérisation de la partie soluble du système phloroglucinol-TPA. Un mécanisme de polymérisation conduisant à des espèces linéaires a été proposé pour des temps courts de réaction. L’augmentation de la fraction insoluble lors de la réaction dans les analyses nécessitant une étape de solubilisation met en évidence un second mécanisme de réaction des oligomères entre eux. Une méthode de suivi de l’état du système par mesures rhéologiques a été également développée pour le contrôle de production. Dans un deuxième temps, ce système a été utilisé pour la conception d’un adhésif Phloroglucinol-TPA-Latex. Celui-ci a été étudié à l’état liquide lors du ‘mûrissement’ et à l’état solide après réticulation sous la forme de films libres pour différentes situations modèles. La stabilité de l’adhésif lors du ‘mûrissement’ a été vérifiée même si des réactions de condensation au sein du système Phloroglucinol-TPA se poursuivent. Après réticulation, la morphologie à deux phases des films observée est spécifique à la nature de la phase latex. Il a été conclu que le nouveau système phloroglucinol-TPA a un comportement similaire à celui du système résorcinol-formaldéhyde en termes de mécanismes de réaction et lors de son utilisation en présence de latex. Celui-ci a donc la capacité de remplacer le système initial dans l’élaboration d’un nouvel adhésif plus respectueux de l’environnement. / A new phenolic-type reactive system more environment friendly, namely phenol-, resorcinol- or formaldehyde-free, was characterized during this Ph.D. This system consists in the copolymerization of Phloroglucinol and Terephthalaldehyde (TPA) in highly diluted water solution conditions. First, a literature study allowed a selection of relevant experimental technics in order to follow the advancement of these complex systems in solution. The combination of chromatographic and spectroscopic analyses is required to characterize the soluble part of the Phloroglucinol-TPA based system. A mechanism of reaction to synthesize linear oligomers was proposed for short reaction times. The increase of the non-soluble part during the reaction in the analyses made after a solubilization step leaded to the conclusion of another type of mechanism where the oligomers are involved. A monitoring method to follow the state of the system by rheological measurement was also developed for the production process. Secondly, this system was used in the preparation of an adhesive based on Phloroglucinol, TPA and Latex. This adhesive was studied in a liquid state during it ‘maturation’ and in a solid state after curing resulting in films from several model systems. The stability of the system during maturation was checked even if condensation reactions of the Phloroglucinol-TPA still took place. After curing, a two-phase morphology of the films was observed which was found to be specific to the nature of the latex phase. It was concluded that the new Phloroglucinol-TPA system has a similar behavior to the one of Resorcinol-Formaldehyde in terms of mechanism and use in presence of latex. It showed the capability to replace the initial system in the preparation of a new adhesive more environment friendly.

Utilisation de composés biosourcés pour la conception de papiers à haute résistance à l'eau et à propriétés antifongiques / Utilization of biobased compounds to design water resistant and antifungal papers

Ce travail de thèse a été conduit dans le but d’élaborer des papiers résistants à l’eau liquide et ayant des propriétés antifongiques, ce par utilisation de composés biosourcés. Deux approches ont été proposées : soit une modification en surface par enduction, soit une modification des fibres de cellulose. Dans le cas de la modification en surface, deux formulations d’enductions ont été étudiées. La première, composée de chitosane et de carbonate de calcium précipité (PCC) modifié, a été utilisée pour limiter le développement de moisissures et améliorer la résistance des papiers à l’eau. La formulation incorporant 20 % de PCC modifié a montré un retard de croissance significatif de la souche fongique sélectionnée en tant que souche cible. La seconde formulation, destinée à améliorer la résistance à l’eau des papiers, par enduction d'un latex prévulcanisé d’hévéa, a montré de très bons résultats, conduisant à une réduction de près de 95 % de l’absorption d’eau liquide. Toutefois, une perte d’opacité des papiers a été observée après pénétration de l’eau dans le matériau. Par conséquent, une seconde approche a été proposée, basée sur la modification physico-chimique des fibres de cellulose. Un procédé d’hybridation a été utilisé, permettant l’adsorption d’acide stéarique sur le PCC synthétisé in situ à la surface des fibres de cellulose. Un compromis entre la rétention des charges, la résistance à l’eau et les propriétés mécaniques a pu être proposé. Après détermination des conditions optimales de mise à l’échelle industrielle du procédé d’hybridation, un essai industriel a pu être réalisé et a conduit à des résultats prometteurs. / The present work investigates the creation of water-resistant and antifungal papers by using biobased compounds. Two approaches were developed: A surface modification by coating or a cellulose fibre modification. For the coating approach, two formulations were studied. The first one, consisting in chitosan and modified precipitated calcium carbonate (PCC) was mainly used to limit the development of molds while improving paper water resistance. The formulation incorporating 20 % of modified PCC showed a significant increase of the lag phase of the target fungal strain. The second coating formulation, especially designed to improve papers water resistance by a coating layer of prevulcanized natural rubber latex, showed very positive results, leading to 95 % reduction of liquid water absorption but with a negative impact on the material opacity after water penetration. As a consequence, physico-chemical modification of cellulose fibre was investigated in a second approach. An hybridization process was used, leading to stearic acid adsorption on in situ PCC synthesized at the surface of cellulosic fibres. A compromise between fillers retention, water resistance and mechanical properties was found. After determination of optimal conditions carried out to an industrial scale up, an industrial pilot was performed and gave promising results.
