The main goal of this diploma thesis is the proposal to use agile development procedures of Scrum framework with UML modeling language applicable for creating websites of small scale. The emphasis is especially put on creating a practically usable type model of the websites. The theoretical part of the thesis has two main chapters. In the "Agile methodology" chapter are listed principles of agile approach to creating software. The next part of this chapter is focus on agile methodologies (Scrum, XP, ASD, FDD, DSDM, LD and Crystal). In the "UML" chapter is introduced modeling language UML and its principles. The next part of this chapter is focus on UML diagrams used in practical part of this thesis (use case diagram, class diagram, sequence diagram, activity diagram and deployment diagram). The practical part of this thesis focuses on the design type model for creating websites of small scale. The type model is based both on the theoretical part of this work and practical experience of the author. In the first part is introduced scheme of model, where is shown the mutual relationship of individual phases. In the next part of this chapter are discussed the individual phases where is used the principle of agile development framework Scrum and examples of use of the UML diagrams.

Theoretical Study of Voltage-driven Capture and Translocation Through a Nanopore : From Particles to Long Flexible Polymers

Voltage-driven translocation, the core concept of nanopore sensing for biomolecules, has been extensively studied in silico and in vitro over the past two decades. However, the theories of analyte capture are still not complete due to the complex dynamics resulting from the coupling of multiple physical processes such as di usion, electrophoresis, and electroosmotic flow. In this thesis, I build and design translocation simulations for analytes ranging from point-like particles to rod-like molecules and long flexible polymers. The primary goal is to test, clarify and complete the existing capture theories. For example, we revisit and revise the existing definitions of the capture radius, clarify the concept of depletion zones, and investigate the impacts of the flat field near the pore. Earlier theories of translocation underestimate the importance of the electric field out- side the nanopore. In our work, we analyze the non-equilibrium dynamics during the cap- ture process originating from the converging field lines, i.e., rod orientation and polymer deformation. We characterize the rod orientation and quantify its impact on capture time both with and without Electrohydrodynamic interactions. We investigate the polymer chain deformation and calculate the translocation time by taking the electric field outside the nanopore into account as opposed to the conventional simulation approaches. Besides nanopore sensing, there are many undiscovered possibilities for nanopore trans- location technologies. We test two proof-of-concept ideas in which we suggest to use capture and translocation to separate molecules of di erent physical properties. For example, we show how one could selectively capture particles sharing the same mobility but di erent di usion coe cients using a pulsed field. Moreover, we demonstrate that it is possible to build a ratchet using pulsed fields and a nanopore to change the concentration ratios of a polymer mixture of different sized polyelectrolytes.

This dissertation is a historical evaluation of the movement of the English courts from the doctrine of pith and marrow to the Catnic test in the determination of non-textual infringement of patents. It considers how and why the doctrine was replaced with the Catnic test. It concludes that this movement occurred as a result of the adoption by a group of judges of literalism in the construction of patents while another group dissented and maintained the correct application of the doctrine. Although the Court of Appeal and the House of Lords initially approved the literalist approach, they, after realising its untennability, adopted the dissenters’ approach, but, ultimately, adopted the Catnic test in which features of the dissenters’ approach were included. Genetische Analysen für eine markergestützte Verbesserung der Trockenstresstoleranz von Winterackerbohnen / Genetic analysis for marker assisted improvement of drought tolerance in autumn sown Faba Bean

In dieser Arbeit zu genetischen Analysen für die Vorbereitung eine markergestützten Selek-tion auf Trockenstresstoleranz bei der Winterackerbohne wurden 196 Winterackerbohnen-Inzuchtlinien und vier Sommerackerbohnen-Inzuchtlinien genotypisiert. Diese Inzuchtlinien wurden außerdem hinsichtlich der Physiologie-Merkmale Spad-Wert, Membranstabilitäts-index, Blattwassergehalt, Gesamtgehalt löslicher Zucker sowie Prolin- und Glycinbetainge-halt in je einer Kontroll- und einer Stressbehandlung phänotypisiert. Anhand eines Verifika-tionssatzes von 40 der 196 Winterackerbohnen-Inzuchtlinien wurden korrelative Verbin-dungen zwischen den physiologischen Merkmalen sowie feldbasierten und züchterisch relevanten Merkmalen wie bspw. Ertrag gesucht. Diese feldbasierten Merkmale wurden mit Hilfe von Rain-Out-Sheltern an den Standorten Göttingen und Groß Lüsewitz in den Jahren 2010/2011, 2012 und 2012/2013 erfasst. Ferner wurden die Möglichkeiten einer Simulation von Trockenstressreaktionen anhand dieses Verifikationssatzes durch Sikkationsversuche mit Kaliumjodidapplikation untersucht. Es konnten keine eindeutigen Beziehungen zwi-schen der Stressreaktion induziert durch Wassermangel und durch Kaliumjodidapplikation ermittelt werden. Außerdem wurden keine eindeutigen Beziehungen der physiologischen Merkmale zu den feldbasierten Trockenstressresultaten gefunden. Mittels einer Kartierungspopulation von 101 RIL wurde eine genetische Karte der Acker-bohne mit zwölf Kopplungsgruppen bestehend aus insgesamt 1451 Markern und einer Län-ge von 1633,2 cM erstellt. Fünf dieser Kopplungsgruppen konnten als Fragmente identifi-ziert werden. Die verbleibenden sieben Kopplungsgruppen wurden mit den verwendeten SNP-Markern mittelbar den sechs Chromosomen der Ackerbohne zugeordnet. Hierbei stel-len z. B. die erste und vierte Kopplungsgruppe gemeinsam eine Kopplungsgruppe dar. Die so kartierten Marker wurden hinsichtlich ihres Spaltungsverhältnisses innerhalb des A-Satzes – bestehend aus 189 der 196 phänotypisierten Winterackerbohnen-Inzuchtlinien – überprüft und für eine Assoziationsanalyse mit den Physiologiemerkmalen ausgewählt. Das Gametenphasenungleichgewicht zwischen 323 610 Markerpaaren wurde ihrer jeweiligen Distanz auf der genetischen Karte gegenübergestellt. Es konnte gezeigt werden, dass in der Entstehungsgeschichte des untersuchten Materials das Gametenphasenungleichgewicht durch Rekombination stark abgebaut wurde. In die Assoziationsanalyse flossen insgesamt 1322 Marker ein. Mittels dieser molekularen Marker konnten insgesamt sechs QTL für Physiologie-Merkmale identifiziert werden. Dabei entfiel je ein QTL auf die Merkmale absolute Differenz im Glycinbetaingehalt zwischen Stress- und Kontrollbehandlung und Glycinbetaingehalt in der Kontrollbehandlung. Vier QTL konnten für die absolute Differenz zwischen dem Prolingehalt in der Stress- und Kontroll-behandlung identifiziert werden. Die gefundenen QTL können anhand der vorliegenden feldbasierten Verifikationsdaten nicht als markergestützte Selektionsmöglichkeit auf Tro-ckenstresstoleranz empfohlen werden. Der nächste Schritt ist demzufolge, mittels feldba-sierter Prüfungen der Inzuchtlinien in realen, relevanten Trockenstresslagen über ausrei-chend viele Orte und Jahre die Bedeutung der physiologischen Merkmale weiter zu prüfen.

Usage of Natural Gas in Modern Steel-making : A Financial and Environmental Evaluation of Available Steel-making Technology in Sweden

A method of producing steel that is not used in Sweden today is direct reduction using natural gas, a method which could has enormous potential in the future of steelmaking. Historically, steelmaking prioritized productivity and profitability. However, other aspects such as safety, sustainability, and environmentally friendliness have become increasingly important to Swedish steelmakers. This study evaluates the usage of natural gas to  directly reduce iron ore into the porous form known as iron sponge, then finally processing that sponge into crude steel. The technology available today is assessed through a literature review, then two calculative data-based model: using DRI sponge in a traditional integrated steelmaking line or replacing steel scrap with it in an electric arc furnace. While this technology is used in many regions where natural gas is plentiful, it has not yet been used in Sweden. Now, when the Swedish natural gas network is expanding and the vehicular transportation of liquid natural gas is becoming more and more viable, it is a prime situation to evaluate direct reduction in Sweden. While models require assumptions and estimations, they suggest that DRI will absolutely be a viable option in the years to come. Integrated plant operators can reduce their immense carbon emissions for a reasonable price, while DRI sponge melting in an electrical arc furnace can either help steelmakers escape the volatile scrap market or be used to replace the blast furnace as a whole in the future. / En metod för att producera stål som inte används i Sverige idag är direkt reduktion med hjälp av naturgas, en metod som har en enorm potential i framtida ståltillverkning. Historiskt har ståltillverkningen prioriterat produktivitet och vinst, men säkerhet, hållbarhet, och miljövänlighet har blivit områden mer och mer viktiga för svenska ståltillverkare. Den här studien utvärderar användningen av naturgas för att direkt reducera järnmalm till den porösa formen känd som järnsvamp, och sen bearbeta den till primärt stål. Teknologin som flnns idag utvärderas genom en litteraturstudie, vars data beräknas till två scenariomodeller: användningen av järnsvamp i ett traditionellt integrerat stålverk eller genom att ersätta stålskrot med järnsvamp i ljusbågsungen. Den här teknologin används redan idag i regioner där naturgas är lättillgängligt, men har hittills inte använts i Sverige. Men nu när Sveriges naturgasnät byggs ut och fordonstransporterad flytande naturgas blir mer och mer kostnadseffektiv så är det lämpligt att utvärdera direkt reduktion i Sverige. Modellerna kräver antaganden och uppskattningar, men de pekar på att direkt reducering av järnmalm kommer att vara en genomförbar metod för ståltillverkning i en nära framtid. Integrerade masugnslinjer kan minska sina enorma koldioxidutsläpp till ett rimligt pris, och järnsvampssmältning i ljusbågsugn kan hjälpa ståltillverkare att undanfly den instabila stålskrotsmarknaden eller användas för att helt ersätta masugnsproduktion i framtiden.

The fate of nitrogen in lactose-depleted dairy factory effluent irrigated onto land

A two-year lysimeter study was undertaken to compare the environmental effects (e.g. nitrate leaching and nitrous oxide emissions) of soil applied lactose-depleted dairy factory effluent (LD-DFE) with lactose-rich DFE. The aim of this experiment was to determine the fate of nitrogen from LD-DFE and dairy cow urine applied to a Templeton fine sandy loam soil (Udic Ustrochrept), supporting a herbage cover of ryegrass (Lolium perenne) and white clover (Trifolium repens). Measurements were carried out on the amount of nitrogen lost from the soil via leaching, lost by denitrification, removed by the pasture plants, and immobilized within the soil organic fraction. Further, a comparison between the fate of nitrogen in LD-DFE irrigated onto land under a "cut and carry" system, as opposed to a "grazed" pasture system was undertaken. Lactose-depleted dairy factory effluent was applied at three-weekly intervals during the summer months at rates of 25 and 50 mm, until nitrogen loading targets of 300 and 600 kg N ha⁻¹ yr⁻¹ had been achieved. Measured leaching losses of nitrogen averaged 2 and 7 kg N ha⁻¹ yr⁻¹ for Control 25 and Control 50 treatments; 21, 20 and 58 kg N ha⁻¹ yr⁻¹ for 25 and 50 mm "cut and carry" treatments respectively; and 96 kg N ha⁻¹ yr⁻¹ for the 25 mm "grazed" treatment. The range of nitrate-N leaching loss from LD-DFE plus urine is no different from the lactose-rich DFE nitrate leaching loss. Uptake of nitrogen by the growing pasture averaged 153, 184,340,352,483, and 415 kg N ha⁻¹ yr⁻¹ for Control 25, Control 50, LD-DFE 25 and LD-DFE 50 mm "cut and carry" treatments, and the LD-DFE 25 mm "grazed" treatment, respectively. Denitrification losses were 0.06, 4.4, 1.69, 19.70, and 7.4 kg N ha⁻¹ yr⁻¹ for Control 25, the LD-DFE 25 "cut and carry" treatments, the LD-DFE 25 mm "grazed" treatment, and calculated "paddock losses", respectively. Isotopic nitrogen studies found that 29.4 and 25.8% of applied LD-DFE nitrogen was immobilised in the LD-DFE 25 and LD-DFE 50 "cut and carry" treatments. The results of this experiment confirm the findings of the previous lactose-rich DFE study, in that the effects of grazing stock are of greater environmental concern than the removal of lactose from the effluent waste stream.

Non-canonical subjects and subject positions / locative inversion, V2-violations, and feature inheritance

Diese Dissertation untersucht die Syntax nichtkanonischer Subjekte und Subjektpositionen - insbesondere zwei Phänomene: Lokativinversion (LI) in Englisch, Französisch, Italienisch und Hebräisch; und Verbdrittverletzungen der Verbzweitregel (V2) in Kiezdeutsch - eine urbane Kontaktvarietät des Deutschen. In LI besetzt eine Lokativ-XP die präverbale Stelle, aber das kanonische DP-Subjekt in Nominativ taucht postverbal auf. Sprachübergreifend vergleiche ich a) die Verteilung unterschiedlicher Null- und overten Argumente in LI und b) die Verfügbarkeit von LI in Matrix- und Nebensätzen. Die zweite Fallstudie befasst sich mit kiezdeutschen V2-Verletzungen, denn sie folgen einer regelmäßigen Reihenfolge: [Rahmensetzer > Subjekt > finites Verb]; dies ist bemerkenswert aufgrund der Verletzung der ansonsten strengen Verbzweitregel und auch, weil es auf die Innovation einer Art Subjektposition hindeutet, die im Standarddeutschen fehlt. Anhand einer Korpusstudie komme ich zu der Erkenntnis, dass die scheinbare Subjektvoraussetzung auch für resumptive V3-Dislozierungsphänomene gilt. Dennoch wird gezeigt, dass die Subjektvoraussetzung mit dem nominativischen DP-Subjekten verbundenen EPP nicht ganz übereinstimmt. Ich entwickele eine Theorie von Subjektvoraussetzungen, die sowohl die Breite der untersuchten Variation in LI als auch die An/Abwesenheit von Subjektvorausetzungen im Vorfeld bei V3-Sätzen im Standarddeutschen und Kiezdeutschen erklären kann. Schließlich lassen sich diese Phänomene durch unterschiedliche Verteilungen und Vererbungsoperationen von D-, ϕ- und informationsstrukturellen δ-Merkmalen (cf. Miyagawa 2017) zwischen dem Phasenkopf C und T erklären. Die Anwesenheit nichtkanonischer Subjekte in LI und kanonischer Subjekte in einer nichtkanonischen Subjektposition im Kiezdeutschen werden durch Variation in der Verteilung eines für das Prädikatssubjekt spezifizierten δ-Merkmal abgeleitet. Dieses Merkmal ist aber unabhänging von den üblichen EPP-Voraussetzungen. / This dissertation addresses syntactic structures involving non-canonical subjects and non-canonical subject positions, investigating two phenomena: Locative Inversion (LI) in English, French, Italian, and Hebrew; and verb-third violations of the verb-second (V2) rule in Kiezdeutsch, an urban contact variety of German. In LI a spatio-deictic XP appears in the preverbal canonical subject position, while the canonical nominative subject DP surfaces postverbally. I compare the distribution of different covert and overt arguments participating in LI and the availability of LI in embedded and matrix contexts crosslinguistically. The second case study concentrates on Kiezdeutsch V2 violations, as they follow a regular order of [frame-setting adverb > Subject > finite verb]; this is remarkable because it both violates an otherwise strict V2 requirement and also indicates the innovation of a subject position lacking in Standard German. I carry out a corpus study and find that an apparent subject requirement extends to other verb-third resumptive-dislocation phenomena, yet we cannot understand this requirement in the sense of an EPP position associated with nominative DP subjects. From a theoretical perspective, this dissertation develops a theory of subject requirements capable of accounting for the breadth of investigated crosslinguistic variation in LI and the presence or absence of a high clausal subject requirement in Kiezdeutsch V2-violations and more standard varieties of German. Ultimately, I make use of finite differences across C and T in the distribution of D, ϕ, and discourse-related δ-features (cf. Miyagawa 2017) via different inheritance options from the phase head. The presence of non-canonical subjects in LI and the presence of canonical subjects in a seemingly non-canonical subject position in Kiezdeutsch are both derivable via variation in the placement of a δ-feature with a specification for Subject of Predication orthogonal to typical EPP requirements.
