The project aim is to research what characterized Splet’s work with sound effects, if it represented innovation in film language and, if so, how it compared to previous films that had also presented sound effect superposition. Another goal is to verify the applicability of the concepts of polyphony and counterpoint to the sound construction of the film. / A obra do sound designer Alan Splet (1939-1994) e a importância da edição de efeitos sonoros estudadas através da análise do filme Veludo azul (Blue velvet, EUA, 1986), dirigido por David Lynch. O intuito do projeto é pesquisar o que caracterizava o trabalho com efeitos sonoros de Splet, se ele representou inovação em termos de linguagem cinematográfica e, em caso afirmativo, como isso se deu em comparação a obras anteriores que também tenham apresentado sobreposição de efeitos sonoros. Outro objetivo é verificar ainda a aplicabilidade dos conceitos de polifonia e contraponto em relação às construções sonoras do filme.

Besides, some discussions will be redeemed on cinematic apparatus, since its conceptual proposition by Baudry, through updates as proposed by Metz and some recent notions such as those presented by André Parente. The outlook on the cinematic apparatus are related to the concepts of mise en abyme, operated by André Gide and subsequently developed by Dällenbach, and applied to the films of David Lynch by the work of Fatima Chinita; and the metacinematographic reflectivity and metafilmic reflectivity, presented by Metz, as well. It will present another issues that orbit the filmography of the director and researches on the topic oftenly, namely: the dispossession of artistic languages, conflicts between a genre cinema and an independent film and the surrealistic art movement. These discussions help us to understand some secondary traits of our research that pervade both the trajectory of the author as the films under our observation. The analyze of both movies are based on the routes listed above and, where possible, articulated with transdisciplinary thoughts that allow us to explore specific aspects, especially with cyberculture, studies of film theory and psychoanalysis. The relationship of these different theoretical perspectives endow the artifices to deepen narrative, technical and aesthetic instances of Mulholland Drive and Inland Empire. The management of those aspects apparently fragmented, in theory and in the works of the north-american filmmaker, are reflected in the structure of this work, with noncumulative and non-sequential chapters for each treated subject. Finally, the conceptual operations proposed to analyze the chosen corpus of moving images allow us to defend that reflectivity is a structural element in the David Lynch film works and, in general, in some parts of contemporary cinema. / Este trabalho se propõe a analisar os modos de agenciamento do cinema reflexivo proposto pelo artista estadunidense David Lynch, com atenção maior para as obras Mulholland Drive e Inland Empire. Para isso, serão resgatadas discussões sobre o dispositivo cinematográfico, desde a proposição por Baudry, passando por atualizações como as propostas por Metz e noções mais recentes como as apresentadas por André Parente. As perspectivas sobre o dispositivo são relacionadas com o conceito de mise en abyme; operacionalizado por André Gide, posteriormente desenvolvido por Dällenbach, e aplicados aos filmes de David Lynch por Fátima Chinita; bem como os de reflexividade metacinematográfica e reflexividade metafílmica, apresentados por Metz. São ainda trabalhadas algumas questões que costumam orbitar a filmografia do cineasta e as pesquisas referentes ao tema, sendo estes: a desterritorialização das linguagens artísticas, os conflitos entre um cinema de gênero e um cinema independente e o surrealismo. Estas discussões nos ajudam a compreender traços secundários de nossa pesquisa que perpassam tanto a trajetória do autor quanto os filmes em observação. As análises dos filmes são fundamentadas nos percursos apresentados acima e, quando possível, articuladas com pensamentos transdisciplinares que nos possibilitem explorar aspectos específicos, principalmente com estudos da cibercultura, das teorias cinematográficas e da psicanálise. A relação dessas diferentes correntes teóricas nos munem de artifícios para aprofundar instâncias narrativas, técnicas e estéticas de Mulholland Drive e Inland Empire. O ordenamento desses aspectos aparentemente fragmentados, na teoria e nas obras do cineasta estadunidense, traduzem-se na própria estrutura do trabalho, com capítulos referentes a cada tema e não-cumulativos e sequenciais. Por fim, as operações conceituais propostas à análise das imagens em movimento do corpus escolhido nos permitem defender que a reflexividade é um elemento estruturante nos trabalhos do cinema de David Lynch e, de modo geral, em partes do cinema contemporâneo.

Le syndrome de Lynch est une affection héréditaire autosomique dominante due à des mutations constitutionnelles des gènes du système de réparation de l'ADN (MLH1, MSH2 et MSH6). Depuis 20 ans le réseau français des laboratoires impliqués dans le syndrome de Lynch a identifié un total de 6687 variants. Sept cent sept d'entre eux, essentiellement des variants faux sens, restent encore des variants de signification inconnue (VSI), sans utilité pour le conseil génétique. Le but de notre étude était de développer un algorithme permettant de classer les variants de signification inconnue. Les critères utilisés étaient les données des analyses in silico, phénotypiques (ségrégation, critères d'Amsterdam), l'état de la fonction MMR (MisMatch Repair) dans les cellules tumorales, les tests fonctionnels et d'épissage, ainsi que les données publiées. Cet algorithme a été appliqué à l'ensemble des VSI de la base de données française et nous a permis de caractériser 370 variants . Les données ont été intégrées dans la base de données française UMD des gènes MMR afin d'être disponibles pour la communauté scientifique. Grace aux données collectées par le réseau, nous avons également pu caractériser le phénotype du syndrome de Lynch. Nous avons ainsi confirmé que le cancer du sein ne fait pas partie du spectre du syndrome de Lynch et que les formes de ce syndrome associées à une mutation du gène EPCAM n'entrainent qu'un risque très faible de cancers de l'endomètre, permettant ainsi d'adapter les recommandations de suivi dans cette situation.de l'endomètre, permettant ainsi d'adapter les recommandations de suivi dans cette situation. / Lynch syndrome is a frequent cancer predisposition with an autosomal dominant mode of inheritance and caused by heterozygous germ line mutations in one of the major DNA mismatch repair (MMR) genes (MLH1, MSH2 and MSH6). For 20 years, the French laboratories network involved in Lynch syndrome identified a total of 6687 variations. Among them, 707, mainly missense variations, remained variants of uncertain significance (VUS), thus could not be used for reliable genetic counseling. The aim of our study was to develop an algorithm able to classify VUS, according to the international consensus (IARC). This algorithm was constructed based on criteria usually required for genetic characterization such as in silico analysis, phenotypical data (segregation, Amsterdam criteria's), MMR status in tumor cells, functional assays, splicing analyses and published data. Data were registered in the French database. As a result of this work, we were able to classify 370 variants of the 707 (52,3%). As part of this work, we also analyzed phenotypical data of patients with Lynch syndrome and showed that breast cancer can definitively be excluded from the spectrum of Lynch-related cancers, and that EPCAM mutations, which may lead to Lynch syndrome, are associated with a very low incidence of endometrial cancer and have probably to be considered as an allelic disease with specific clinical recommendations.

Many small municipalities, in contrast to larger centers, struggle to offer natural social venues in their neighborhoods. As people choose larger cities for social gatherings, smaller city centers becoming depopulated and in turn they lose their necessary demand to sustain social meeting places. The purpose of this work is to see what small towns can do to make social spaces more attractive and vibrant and thereby encourage people to participated more in the public realm. This study will focus on Smedjebacken (Sweden) municipality's urban development and present proposals for measures that the municipality can take to become more socially sustainable from a planning perspective. The work is much based on Kevin Lynch and Jan Gehl's theories. Where Lynch's scientific strategies see the great perspective of how the city is built and Gehl's ideologies are at a more detailed level where the residents meetings in the city are the primary. A case study has been conducted to gather information about past and future strategies concerning Smedjebacken's city center, as well as documents, ideas and views of municipal official and knowledgeable persons through interviews. A Lynch-based observation analysis of central parts of the city was also used. The method is based on theories in Lynch’s The image of the city, Lynch argues that people see five critical elements in cities: paths, edges, districts, nodes and landmarks. This analysis was conducted during two days in Smedjebacken’s city center. The results show that Smedjebacken has several improvement points in order to increase the social sustainability in the city center. The city is divided into three major districts and has an incoherent character, which means that the few shops and social venues in the city are dispersed and do not benefit from one another as a result. In order to fuse the core of the city, recommendations based on Jan Gehl's theories are advanced. The key strategy recommended is to create social meetings that encourage residents to visit and participate in their community center. / Många mindre kommuner har idag svårt att bibehålla de naturliga sociala mötesplatserna i sina tätorter till följd av att större städer har mer att erbjuda. Att allt fler människor väljer större städer för olika sociala sammankomster leder ofta till att de mindre stadskärnorna blir folktomma och i sin tur försvinner den efterfrågan som behövs för att upprätthålla sociala mötesplatser. Syftet med det här arbetet är att se vad småkommuner kan göra för att uppmuntra sin befolkning att lämna hemmen och göra de sociala rummen mer attraktiva och levande. Arbetet kommer att ha fokus på Smedjebacken kommuns stadsutveckling och presentera förslag på åtgärder som kommunen kan vidta för att bli mer socialt hållbara ur ett planeringsperspektiv. Arbetet grundar sig mycket i Kevin Lynch och Jan Gehls teorier. Där Lynch vetenskapliga strategier ser det stora perspektivet av hur staden är uppbyggd och Gehls ideologier är på en mer detaljerad nivår där invånarnas möten inom staden är det primära. En fallstudie har genomförts för att samla in information om tidigare och framtida strategier för Smedjebackens stadskärna, även olika dokumentationer från kommunen har använts och insatta personer har intervjuats. En Lynchbaserad observationsmetod över de centrala områdena inom staden har tillämpats. Metoden grundar sig i Lynch teorier från boken The image of the city där Lynch hävdar att människor ser och upplever staden utifrån de fem kritiska elementen: stråk, gränser, distrikt, noder och landmärken. Analysen genomfördes under två dagar i centrala Smedjebacken. Resultatet visar att Smedjebacken har ett flertalet olika punkter som skulle kunna förbättras för att främja den sociala hållbarheten inom centrum. Staden är uppdelad i tre större distrikt och har en osammanhängande karaktär vilket leder till att de få affärerna och sociala mötesplatserna i staden är utspridda och inte gynnas av varandra till följd av det. För att knyta samman stadskärnan har stadsplaneraren Jan Gehls teorier applicerats där han menar att människor måste uppmuntras till sociala möten i staden för att vilja vistas där.

Vid cancerdiagnostisering av Lynch syndrom används immunohistokemisk färgning för undersökning av eventuell defekt hos mismatch repair-generna MLH1, MSH2, MSH6 och PMS2. Färgningen använder sig av antikroppar som undersöker om cellkärnor i vävnad har intakta eller förlorade proteinuttryck hos respektive gen. Syftet med studien var att utföra en antikroppsvalidering för att granska om de nuvarande Dako Link Ready-to-Use-antikropparna (Link RTU) och koncentrerade antikroppen Dako PMS2 1:40 kunde bytas ut till Dako Omnis Ready-to-Use-antikroppar (Omnis RTU). Omnis RTU-antikropparna är optimerade för det automatiserade analysinstrumentet Dako Omnis som används vid patologavdelning på Universitetssjukhuset Örebro. Tolv vävnader med colorektalcancer och två externa positiva kontroller färgades med samtliga antikroppar för respektive gen och en jämförelse mellan Link RTU samt Dako PMS2 1:40 gjordes mot Omnis RTU-antikropparna. De nya färgningarna med Omnis RTU bedömdes som sämre, likvärdig eller bättre infärgad. Om antikroppen färgade rätt målceller och med stark färgintensitet bedömdes den och motsvarande protokoll som godkänd. Resultatet visade att Omnis RTU för MLH1 gav en förbättrad färgning. Omnis RTU för MSH2 gav i majoritet en förbättrad färgning med något mer bakgrundsfärg än önskvärt. Omnis RTU för MSH6 gav en svagare infärgning i majoriteten av fallen och Omnis RTU för PMS2 gav i majoritet en förbättrad färgning, men med för mycket bakgrundsfärg. Slutsatsen var att Omnis RTU för MSH6 och PMS2 kräver vidare optimering av protokoll innan de används i laboratoriets rutinfärgning. Protokollet för Omnis RTU för MSH2 kan behöva en mindre justering för minskad bakgrundsfärg och Omnis RTU för MLH1 fungerar väl med det nuvarande protokollet. / Immunohistochemistry staining is used in diagnostics of Lynch Syndrome to investigate possible defects in the mismatch repair genes MLH1, MSH2, MSH6 and PMS2. Antibodies is used to examine whether cell nuclei in cancer tissue have intact or loss of protein expression in said genes. The aim of the study was to perform an antibody validation to examine if the current Dako Link Ready-to-Use antibodies (Link RTU) and the concentrated PMS2 antibody can be replaced with Dako Omnis Ready-to-Use antibodies (Omnis RTU) that are optimized for the analytical instrument Dako Omnis, which is used in Universitetssjukhuset Örebro. Twelve tissues with colorectal cancer and two external positive controls was stained with all antibodies for each gene. A comparison was done between the old antibodies and Omnis RTU, where Omnis RTU was judged to be worse, equivalent or better. The antibody and used protocol were approved of laboratory use if it stained the target cells with a strong intensity. The results showed that Omnis RTU MLH1 had an improved staining. Omnis RTU MSH2 had in majority an improved staining, but with slightly more background staining than preferred. Omnis RTU MSH6 had in majority a weak staining while Omnis RTU PMS2 had an improved staining but with too much background staining. The conclusion was that the protocols for Omnis RTU MSH6 and PMS2 needed further optimization before laboratory use. The protocol for Omnis RTU MSH2 may need a minor adjustment to reduce background staining, while Omnis RTU MLH1 works well with the current protocol.
