Exploring the effects of 5-HT2A and AMPA receptors on brain 5-HT via a mechanism-based pharmacodynamic model

Depression is a common mood disorder. Although major ethical challenges make it nearly impossible to invasively and directly measure serotonin (5-hydroxytryptamine, 5-HT) levels in human brains, neuroimaging technologies have shown macroscopic structural and functional abnormalities in the prefrontal cortex (PFC) of depressed patients. The monoamine hypothesis of depression is based on the neurotransmitter imbalance, such as deceased serotonin brain levels are implicated in the cause of depression. Research has focused on the control mechanisms involved in the dorsal raphé nucleus (DRN) which is the serotonergic control center located in the midbrain. We hypothesized that activation 5-HT 2A receptor in PFC would increase serotonin levels by an AMPA-dependent mechanism in both DRN and PFC. Enhancement of the 5-HT in DRN may inhibit 5-HT level in PFC by 5-HT 1A receptor. This becomes the full feedback loop system. While 5-HT levels in the PFC have been well studied, pathway that modulate this DRN pool through upstream cascade interactions leading to a downstream feedback loop have been difficult to elucidate. Developing a mechanism-based pharmacokinetics (PK) and pharmacodynamics (PD) model to quantitatively describe the effect of 5-HT 2A receptors regulation to serotonin in the DRN and PFC would help us to better understand the complex brain. 5-HT 2A receptor agonist and AMPA receptor agonist and antagonist were used to activate or block the related receptor. Male Wistar rats underwent neurosurgery for implantation of microdialysis (MD) probes. Three to five rats were randomly assigned to experimental arms. Using the MD method, the drug combination was examined to explore the drug effect on time course of 5-HT release in DRN and PFC. Based on the experiment results, a mechanism-based PD model was developed. Phoenix WinNonlin ® and Berkeley Madonna™ were used for model estimation, external validation with secondary data set, and simulation. The result supports the possibility of a 5-HT 2A /AMPA feedback control circuit that originates in the PFC and modulates DRN and PFC 5-HT levels through feedback coupling of 5-HT. The time-course profiles of 5-HT in both DRN and PFC was well modeled and model parameters were estimated with good precision (CV% ranged from 1.37% to 35.03%). The mechanism model was developed to characterize and better understand the neurotransmitter mechanisms, providing estimations of various parameters of the disease related receptor system.

Nutritional Habits of Student Musicians and Their Effects: Consumption of Fats, Fruits, and Vegetables as It Relates to BMI, Mental Health, and Other Factors

This study conducted the first nationwide epidemiological survey to investigate the effects of dietary fat, fruit, and vegetable intake on the health of student musicians in the United States. Despite the recognized importance of nutrition in other performance fields, such as athletics, the relationship between diet and health in musicians has not been rigorously explored until now. The survey assessed dietary intake, body mass index (BMI), mood states via the DASS-21, engagement with healthcare, nutritional knowledge, and mental health history among 641 music majors. Key findings include higher fat intake and BMI predicted higher DASS-21 total scores as well as each separate sub-score for depression, anxiety, and stress; a significantly lower mean BMI in the study group compared to the general U.S. college student population; and mixed results regarding healthcare engagement and its relation to diet and BMI, suggesting the need for a more suitable model for analysis. Notably, sources of nutritional knowledge and barriers to healthy eating significantly influenced diet quality, indicating potential misinformation about fat consumption and the positive impact of accessible healthy foods on diet quality. Additionally, mental health diagnoses were associated with lower fat and fruit/vegetable scores but not BMI. These results underscore the potential negative impact of dietary habits on mental health among music majors and highlight a widespread misunderstanding of what constitutes a healthy diet. Future research should refine dietary assessments and incorporate biometric data, while music education institutions are urged to include nutrition education in their curricula, emphasizing the role of diet in overall musician wellness.

Sprache und Denken / zum zeitlichen Ablauf von Äußerungskonzeptualisierung und syntaktischer Kodierung

Die Studie hat zum Ziel, im Rahmen des Äußerungsproduktionsmodells die Annahme thinking for speaking (Slobin, 1996) experimentell zu überprüfen. Ansatzpunkt ist der Unterschied in der Stellung der Markierung einer Entscheidungsfrage zwischen drei Sprachen. Während der Fragemodus im Deutschen/Polnischen vor dem Frageinhalt markiert wird, geschieht die Modusmarkierung im Chinesischen nach dem Inhalt. Um die entsprechende Satzstruktur aufzubauen, sollte der Fragemodus beim syntaktischen Kodierungsprozess (speaking) im Deutschen/Polnischen vor, im Chinesischen nach dem Inhalt verarbeitet werden. Unter Zugrundelegung der inkrementellen Äußerungsproduktion gehen wir davon aus, dass die Verarbeitungsreihenfolge beim vorangehenden Konzeptualisierungsprozess (thinking) mit der syntaktischen Kodierungsabfolge übereinstimmt: Der Fragemodus wird im Deutschen/ Polnischen vor, im Chinesischen hingegen nach dem Inhalt konzeptualisiert. Um den zeitlichen Ablauf der zwei Konzeptualisierungsprozess zu ermitteln, bedienten wir uns des lateralisierten Bereitschaftspotenzials (lateralized readiness potential, LRP) im binären Wahlreaktions-Go/Nogo-Paradigma. Im Versuch reagierten deutsche, chinesische und polnische Muttersprachler auf dargebotene Bilder mit Tastendrücken und Sprechen. Zu beobachten war das Auftreten eines LRP bei Nogo, das signalisierte, in welcher Reihenfolge sich die Handwahl und die Nogo-Entscheidung realisierten Ein Nogo-LRP trat bei allen drei Sprechergruppen auf. Zudem wurde festgestellt, dass die Nogo-Entscheidung, die erwartungsgemäß mit der sprachlichen Verarbeitung des Fragemodus verbunden wurde, bei den drei Sprechergruppen ungefähr zeitgleich getroffen wurde. Die Befunde legen nahe, dass der Fragemodus in den drei Sprachen zeitlich nicht unterschiedlich, sondern einheitlich geplant wurde. Die Schlussfolgerung wird im Rahmen von thinking for speaking sowie dem Äußerungsproduktionsmodell diskutiert. / Languages differ in the marking of the sentence mood of a polar interrogative (yes/no question). For instance, the interrogative mood is marked at the beginning of the surface structure in Polish, whereas the marker appears at the end in Chinese. In order to generate the corresponding sentence frame, the syntactic specification of the interrogative mood is early in Polish and late in Chinese. In this respect, German belongs to an interesting intermediate class. The yes/no-question is expressed by a shift of the finite verb from its final position in the underlying structure into the utterance initial position, a move affecting, hence, both the sentence final and the sentence initial constituents. The present study aimed to investigate whether during generation of the semantic structure of a polar interrogative, i.e. the processing preceding the grammatical formulation, the interrogative mood is encoded according to its position in the syntactic structure at distinctive time points in Chinese, German, and Polish. In a two-choice go/nogo experimental design, native speakers of the three languages responded to pictures by pressing buttons and producing utterances in their native language while their brain potentials were recorded. The emergence and latency of lateralized readiness potentials (LRP) in nogo conditions, in which speakers asked a yes/no question, should indicate the time point of processing the interrogative mood. The results revealed that Chinese, German, and Polish native speakers did not differ from each other in the electrophysiological indicator. The findings suggest that the semantic encoding of the interrogative mood is temporally consistent across languages despite its disparate syntactic specification. The consistent encoding may be ascribed to economic processing of interrogative moods at various sentential positions of the syntactic structures in languages or, more generally, to the overarching status of sentence mood in the semantic structure.

Examining the maintaining factors of anorexia nervosa

This thesis is a qualitative investigation of the factors which maintain anorexia nervosa (AN) according to the transdiagnostic theory of eating disorders (Fairburn et al., 2003). AN is difficult to treat and continues to evade complete understanding. The present study aimed to promote further understanding of food restriction and physical activity in relation to the constructs of clinical perfectionism, core low self-esteem, mood intolerance, and interpersonal difficulties. Twenty females with self-reported AN were recruited from Guelph, Ontario and participated in semi-structured interviews. Thematic analysis revealed eight major themes for clinical perfectionism, five for core low self-esteem, five for mood intolerance, and six for interpersonal difficulties. The in-depth emotional accounts and details of food restriction and physical activity in relation to the four constructs examined in this study may contribute to further appreciation of AN, informing practitioners and family members, promoting empathy, and improving treatment options.

INTRODUÇÃO: Apesar da importância dos medicamentos antidepressivos no tratamento dos transtornos de humor e de ansiedade, sua ação sobre sistemas cerebrais responsáveis pela expressão emocional ainda não foram claramente estabelecidos. Estudos recentes têm examinando o sinal dependente de nível de oxigenação sanguínea (do inglês, \"BOLD\") durante estímulos emocionais em indivíduos saudáveis sob uso de antidepressivos. Nesse estudo, pretendemos estender essa avaliação às alterações do humor e comportamento emocional devido ao uso prolongado de um antidepressivo bloqueador de serotonina e noradrenalina em pessoas saudáveis. MÉTODOS: foram selecionados dezesseis voluntários, sem antecedentes psiquiátricos pessoais ou familiares, que participaram de um ensaio farmacológico simples-cego de quatro semanas de doses baixas de clomipramina (até 40 mg/dia). Ao final desse período, dez sujeitos foram selecionados como não responsivos, e os restantes seis sujeitos foram selecionados como responsivos por apresentarem claras mudanças em três dos quatro seguintes critérios: tolerância interpessoal, eficiência, bem estar, e mudança substancial em sua auto percepção. O grupo de sujeitos classificados como responsivos foram submetidos a um ensaio controlado duplo-cego confirmatório. A aquisição de imagens cerebrais ocorreu após quatro semanas de uso de medicação (simples cego) e quatro semanas após a sua suspensão, ao final da participação no ensaio farmacológico. O imageamento cerebral foi realizado durante a indução de estados afetivos de felicidade, irritabilidade e neutros por relatos autobiográficos. A resposta emocional desses estados foi obtida por escalas de auto avaliação de ansiedade, irritabilidade e felicidade. As diferenças de sinal entre os estados afetivos foram utilizadas para a análise estatística da interação dos efeitos estado medicamentoso e grupo por testes ANOVA. RESULTADOS: Foi encontrada uma interação significativa entre o efeito de grupo e o estado medicamentoso sobre os estados afetivos de irritabilidade, mas não sobre os de felicidade. Se observou redução na auto avaliação de ansiedade no grupo responsivo com o uso de medicação na diferença entre os estados induzidos de irritabilidade e felicidade, em comparação com o efeito no grupo não responsivo; e também, redução na auto avaliação de felicidade com o uso de clomipramina na totalidade da amostra, na diferença entre o estados induzidos de irritabilidade e neutro. A alteração sobre o efeito BOLD (p < 0,005) foi localizada em regiões adjacentes à junção frontoparietal para a indução de irritabilidade em relação à felicidade e em relação aos estados neutros, no grupo responsivo em relação ao não responsivo, durante o período em uso relativo àquele sem uso de clomipramina, e na junção têmporo-paríeto-occipital, exclusivamente para a diferença irritabilidade-felicidade. Recent studies have examined blood oxygen level dependent (BOLD) signal during emotion stimuli in healthy individuals in antidepressant use. The study intends to extend these measures to the changes over mood and emotional behavior by prolonged use of a serotonin and noradrenaline blocker antidepressant. METHODS: we selected sixteen subjects, with no personal or family history of psychiatric disorders, which participated of a four-week single-blind trial with low doses of clomipramine (up to 40mg/day). After this period, ten subjects were classified as non responsives, the remaining six subjects being classified as responsives because clomipramine provided positive changes in three of four of the following criteria: interpersonal tolerance, self-efficacy, mood and self-perception of a substantial change. Responsives were included in a placebo-controlled confirmatory trial. Imaging sessions occurred at the end of the four-week course of clomipramine and again after a four-week clomipramine washout period, at the end of pharmacological trial. Subjects were scanned during the induction of irritability, happiness and neutral affective states by autobiographical recall. Self-report assessment was performed for each induction by anxiety, irritability and happiness scales. Inter-condition differences (affective induction) were used in the analysis of interaction of medication status and group effects by ANOVA tests. RESULTS: A significant interaction was found between group and treatment during irritability, but not during happiness emotions. It was observed a reduction in the scale of self-evaluation of anxiety in responsive group with the use of medication to the difference between irritability-happiness states, compared to the non responsive group; and a reduction in auto evaluation of happiness, in the totality of the sample in use of clomipramine, in difference irritability-neutral. There was a significant increase of BOLD effect in the responsive group during use relative to the period without use of clomipramine, compared to the effect on non-responsive group (p < 0.005). This effect was located in regions that surround the frontoparietal junction to the irritability relative to happiness and to irritability relative to neutral induction, and in the temporo-parieto-occipital junction, exclusively to the irritability relative to happiness induction. CONCLUSIONS: The favorably changes in healthy subjects who respond to prolonged serotonin and noradrenaline blocker use, may relate to changes in neural processes of autobiographical memory of negative emotions

Pain : psychological measurement and treatment

This research was executed as three separate studies. Study 1 focused on the perception of pain and the semantic aspects of pain. Study 2 focused on the measurement of acute pain and mood states. Study 3 focused on the psychological treatment of cancer pain. In Study 1 a group of 66 Setswana-speaking adults were required to describe what they saw, what happened, and what would be the outcome with respect to three visually presented pain scenes using The Pain Apperception Test (PAT) A qualitative analysis of their responses shows that pain is experienced as an all encompassing experience affecting all aspects of their lives, such as the physical, emotional, social, and economic. This was found to be true, irrespective of gender and age with the exception of economic issues. A qualitative analysis of their responses to the Pain Eliciting Incidents Questionnaire (PEIQ) reveals that the Setswana pain descriptors are classifiable according to the three dimensions of pain namely, the sensory-discriminative, affective-motivational, and cognitive-evaluative. Sludy 2 applied the Profile of Mood States (POMS) preoperatively to a group of 58 female laparotomy (gynaecological) patients. These patients were also tested post-operatively with the Visual Analogue Scale (VAS) and the Wisconsin Brief Pain Questionnaire (WBPQ) as pain measures. The pain measures were taken at no medication and at the peak of medication. Factor analysis could not confirm the validity of the six POMS scales. These scales also did not show correlations with post-operative pain. Correlations between the pain measures showed acceptable reliability and validity of the VAS and the WBPQ. In Study 3 three groups of 15 cancer patients each, suffering from chronic pain, were treated over a period of two weeks with either cognitive behavioural therapy plus medication, reassurance therapy plus medication, or medication only. Comparison of before and after treatment pain measures showed that both cognitive behavioural therapy and reassurance therapy had a beneficial effect. Follow-up results three months later showed that the beneficial effect of reassurance therapy did not persist.
