A diarréia por rotavírus tem sido umas das principais causas de mortalidade infantil. Pesquisas, que relatam a presença de IgA no leite e colostro humanos reativos com o rotavírus. Nosso objetivo foi avaliar a presença de anticorpos IgA anti-G3P[2] em amostras de leite e colostro e anti-G9P[8] em amostras de leite pela técnica de ELISA, verificar sua capacidade neutralizante e analisar a reatividade pelo Immunoblotting. Todas as amostras apresentaram anticorpos detectados pelo ELISA e neutralização do vírus G3P[2] e G9P[8]. Não foi observada correlação entre os títulos de ELISA e os neutralizantes para G3P[2], mas para o sorotipo G9P[8] foi possível estabelecer uma correlação significante. As amostras reconheceram frações do antígeno viral no ensaio de Immunoblotting, mas não foi possível estabelecer uma correlação entre altos títulos e alguma fração antigênica específica. Este trabalho fornece subsídios para avaliações das estratégias de vacinação. / The rotavirus diarrhea is one of the main causes of infant mortality. Studies have shown the presence of IgA in milk and colostrum reactive with human rotavirus. Our aim was to evaluate the presence of IgA anti-G3P[2] in milk and colostrum samples and anti-G9P[8] in milk ones, evaluate the IgA reactivity by ELISA and evaluate the IgA reactivity by immunoblotting. In addition, this work aimed to verify the neutralizing all titles of the samples. All of them had antibodies reactive with G3P[2] and G9P[8] and showed varied neutralization titles. There was a significant correlation between anti-G9P[8] IgA titers and the neutralizing ones, but the same was not observed for serotype G3P[2]. All samples recognized some viral antigenic fraction in immunoblotting test, but it was not possible establish a correlation between high antibodies levels and some specific antigenic fraction. This approach may be important for studies concerning vaccination strategies.

Padronização de uma nova técnica para detecção de anticorpos neutralizantes anti-dengue baseada na RT-PCR em tempo real / Standardization of a new technique for anti-dengue neutralizing antibodies detection based on Real Time RT-PCR

Por representar a mais importante arbovirose em nível mundial, as infecções causadas pelos vírus da dengue são de grande importância em nosso país, apresentando uma ampla variedade de sintomas clínicos que vão desde infecção assintomática até formas mais graves da doença. O título de anticorpos neutralizantes produzidos frente à infecção por dengue parece ser determinante na forma de apresentação da doença no paciente. Atualmente, a forma com que o teste de neutralização é realizado demanda tempo para sua realização. O objetivo deste trabalho foi padronizar um ensaio de neutralização viral por RT-PCR em tempo real em cepas virais dos quatro sorotipos dengue para, posteriormente, ser empregada na detecção rápida e em grande escala de anticorpos em soro de pacientes e de candidatos vacinais. Para isso, foram construídas curvas padrão, para cada sorotipo viral, por meio da transcrição in vitro do RNA viral. O ensaio de neutralização padronizado nesse estudo reduziu o número de dias de detecção da neutralização em 48 horas quando comparada com a técnica de neutralização tradicional (PRNT), além de utilizar técnicas moleculares sensíveis e específicas para detecção como a RT-PCR em tempo real que garantem maior aplicabilidade do teste. / Because Dengue virus is the most important arboviral disease worldwide, infections caused by this pathogen are of great importance in Brazil, producing a wide variety of clinical symptoms ranging from asymptomatic infection to more serious forms of the disease. The title of neutralizing antibodies produced against the dengue infection appears to be determinant in the outcome of the disease. Currently, neutralization tests that have been performed take time for its execution. The aim of this study was to standardize a viral neutralization assay by real-time RT-PCR of viral strains of the four Dengue serotypes to subsequently be used for rapid detection and large-scale using antibodies of patients and for vaccine candidates. For this matter, standard curves were constructed for each Dengue serotype, by in vitro transcription of viral RNA. The neutralization assay in this study reduced the period of neutralization to 48 hours, compared to traditional neutralization test (PRNT), and it uses a more sensitive and specific molecular technique for detection of neutralizing antibodies, such as real time RT-PCR, to ensure greater applicability of the test

Role of Middle Managers in Mitigating Employee Cyberloafing in the Workplace

Companies in the United States are concerned about the indeterminate effectiveness of corporate cyberloafing mitigation efforts leading to the persistence of employee cyberloafing behavior. Although middle managers are the driving force behind the transformational influences that guide employee productivity and could proffer practical solutions, a lack of clarity surrounds the middle manager's role in the overall cyberloafing mitigation efforts within organizations. The central research question for this transcendental phenomenological research study explored the lived experiences of middle managers regarding their roles in mitigating employee cyberloafing at higher education institutions in Florida. This study used a social constructivist-interpretive framework that draws from the multiple realities constructed through social interactions and lived experiences. Participants included 7 middle managers with experience mitigating cyberloafing at higher education institutions in Florida. Four major themes emerged from an inductive analysis of the data, including managing employee performance, proximity matters, cyberloafing interventions, and understanding employee online technology use. The results and recommendations of this study provide implications for social change. Business organizations may modify cyberloafing mitigation strategies and policies from a better understanding of manager/employee interactions, transformational managerial influences used to mitigate employee cyberloafing, and managerial knowledge of employee appropriation of online technology.

Cette étude de l’image dans l’œuvre de Tournier vise à déchiffrer la conception qu’a l’écrivain de l’image, la spécificité de son monde imaginaire ainsi que sa passion à créer la nouveauté, à l’instar des artistes. Ainsi, les images, photographiques et peintes, tantôt maléfiques tantôt bénéfiques, révèlent l’imaginaire spécifique de Michel Tournier : l’ambiguïté d’une imaginaire « bifrons ». Une image n’est jamais limpide ni durablement stable. Le caractère dual et le plus souvent antithétique des images illustre bien le projet esthétique de Tournier : l’ambiguïté, la contradiction, la polysémie. Ce projet esthétique lance un défi à la pensée et aux habitudes du lecteur. Une image n’est pas telle qu’elle semble l’être ; un mot ne renvoie pas non plus à ce qu’il représente dans l’œuvre littéraire ; le monde n’est pas tel qu’il est perçu. Toutes les valeurs, les règles imposées par la société, par l’homme demandent à être inversées et interprétées. Analysant les images chez Tournier, cette recherche permet spécifiquement de réfléchir aux possibilités de la représentation, à leur valeur et donc à la création littéraire, considérée comme une « re-présentation ». / This study of image in Tournier's work aims to decipher the writer’s conception of image, the specificity of his imaginary world, his passion to create « new » like artists do. Thus, images, photographic and painted, either evil or beneficial, reveal the specific imagination of Michel Tournier – the ambiguity of an imaginary « bifrons ». An image is neither clear nor stable. The dual and often antithetical nature of image illustrates the aesthetic project of the writer: ambiguity, contradiction and polysemy. This aesthetic project is a challenge to the reader’s usual thoughts and habits. An image is not like it seems to be, a word is not changes his meanings in literary works, the world is not like it is perceived. All values, rules imposed by society, by human beings have to be inverted and interpreted. By analyzing images in the works of Tournier, this research specifically invites us to reflect on the possibilities of representation, and on their value and literary creation, considered as « re-presentation ».

Reaction dynamics on highly excited states

In this thesis I have performed theoretical studies on the reaction dynamics in few-atom molecules. In particular, I have looked at reaction processes in which highly excited resonant states are involved. When highly excited states are formed, the dynamics becomes complicated and approximations normally used in chemical reaction studies are no longer applicable.To calculate the potential energy curve for some of these states as a function of internuclear distance, a combination of structure calculations and scattering calculations have to be performed, and the reaction dynamics on the potentials has been studied using both time-independent and time-dependent methods.The processes that have been studied and which are discussed in this thesis are ion-pair formation in electron recombination with H3+, dissociative recombination and ion-pair formation of HF+, mutual neutralization in H++F- collisions and dissociative recombination of BeH+. Isotope effects in these reactions have also been investigated. Our calculated cross sections are compared with experimentally measured cross sections for these reactions.

Adenovirus-mediated Gene Therapy of Prostate Cancer

Adenovirus-mediated gene therapy is a potential complement to standard cancer treatments. Advantages are that vectors can be used to target tumors and that replicating viruses lead to increased therapeutic dosage. In this thesis, an oncolytic serotype 5 adenovirus (Ad5), Ad[i/PPT-E1A, E3], was developed where viral replication is controlled by the insulator-shielded (i) prostate-specific PPT promoter. The adenoviral E3 region was inserted for its immune regulatory and lysis functions. Ad[i/PPT-E1A, E3] had improved cytotoxic abilities both in vitro and in a prostate cancer xenograft mouse model compared to a virus lacking the E3 region. To further improve adenoviral vectors, the histone deacetylase inhibitor (HDACi) FK228 was studied. FK228 has been proposed to enhance the effect of adenoviral therapy by upregulation of CAR, the primary receptor for Ad5 infection. In the present study, we observed that FK228 promotes transgene expression even better when administered after viral transduction, indicating a post-transductional enhancement of transgene expression. Another interesting finding was that FK228 reduced transgene expression from the PPT promoter in the prostate cancer cell line LNCaP. This is explained by the fact that different HDACi have the ability to provoke a neuroendocrine phenotype of LNCaP. A potential drawback with adenoviral gene therapy is the rapid clearance of the virus from the circulation. Viral particles have been coated with polyethylene glycol (PEG) to evade immune recognition, a strategy that works well in mouse models. However, less is known about the effects of adenoviral PEGylation in human blood. We have studied cell interactions and immune responses to PEGylated and uncoated Ad5 vectors in human whole blood using a blood loop model with constant blood flow. Limited effects of PEGylation were observed in human blood, which were associated with the neutralizing ability of the donor blood. An important finding that donors with high neutralizing ability in whole blood do not necessarily have neutralizing antibodies against the virus strongly implies that neutralization should be measured in whole blood.

Investigation Of 8-year-long Composition Record In The Eastern Mediterranean Precipitation

Measurement of chemical composition of precipitation is important both to understand acidification of terrestrial and aquatic ecosystems and neutralization process in the atmosphere. Such data are scarce in the Mediterranean region. In this study, chemical composition of daily, wet-only, 387 number of rain water samples collected between 1991 and 1999 were investigated to determine levels, temporal variation and long-term trends in concentrations of major ions and trace elements between 1991 and 1999. Samples had already been collected and some of the analysis had been completed. The anions SO42-, NO3- and Cl- were analyzed by HPLC coupled with UV-VIS detector, NH4+ was analyzed by colorimetry and H+ ion was analyzed by pH meter. The major ions and trace metals were analyzed by using Atomic Absorption Spectrometry (AAS) and Graphite Furnace Atomic Absorption Spectrometry (GFAAS). In this study complete data set were generated by analyzing samples that had not been previously analyzed for major ions and trace elements with Inductively Coupled Plasma with Optical Emission Spectrometry (ICP-OES). Statistical tools were used to determine the distribution of the pollutants. The rain water data tends to be log-normally distributed since data show large variations due to meteorological conditions, physical and chemical transformations and air mass transport patterns. The median pH of the rain water was found to be 5.29, which indicates that the rain water is not strongly acidic. This case is not a result of lacking of acidic compounds but rather indicates extended neutralization process in rain water. Eastern Mediterranean atmosphere is under the influence of three general source types: (1) anthropogenic sources, which are located to the north and northwest of the basin brings low pH values to the region (SO42-, NO3- ions) / (2) a strong crustal source, which is dried and suspended local soil and air masses transported from North Africa transport which have high pH values (Ca2+, Al, Fe ions) and (3) a marine source, which is the Mediterranean Sea itself (Na+, Cl- ions). In the region, the main acid forming compounds are H2SO4 and HNO3 whereas / CaCO3 and NH3 are responsible for the neutralization process. To describe the level of pollutant concentrations and the factors that affect their variations in rain water / ion compositions, neutralization of acidity, short and long-term variability of ions and elements, their time trend analysis and wet deposition fluxes were investigated briefly. Positive matrix factorization (PMF) was used to determine components of ionic mass in the precipitation. In Antalya Station the rain water has five factors: free acidity factor, crustal factor, marine factor, NO3- factor and SO42- factor. Potential Source Contribution Function (PSCF) and trajectory statistics were used to determine source regions generating these components. NO3- has potential source regions of Western Mediterranean countries and North Africa, whereas SO42- has additional southeasterly trajectory components of Israel and south east of Turkey.

Rôle des anticorps neutralisants autologues dans la guérison spontanée lors d'une infection par le virus de l'hépatite C / Role of autologous neutralizing antibodies in spontaneous recovery during infection by hepatitis C virus

Environ 30% des patients infectés par le virus de l’hépatite C guérissent spontanément. Le but de ce travailétait (i) d’examiner l’évolution des protéines d’enveloppe en cas de guérison spontanée (ii) de comparerl’infectivité des variants présents aux stades précoces (iii) d’explorer la capacité neutralisante des anticorps(Ac) vis-à-vis des variants majoritaires et minoritaires. Nous avons sélectionné 2 patients avec une hépatiteaigue C suivie d’une guérison très rapide. Pour explorer la capacité d’entrée de ces variants et leur aptitude àêtre neutralisés, nous avons produit des pseudo-particules rétrovirales portant les enveloppes de différentsvariants. Pour le 1er patient, une réponse neutralisante autologue était détectable précocement, avec unmaximum entre le 2ème et le 3èmemois suivant la cytolyse. Elle était encore détectable au 30ème mois. Pourle 2ème patient, des Ac vis-à-vis du variant majoritaire étaient détectés dans le sérum prélevé 4 jours après lacytolyse et dans les sérums plus tardifs. Le titre des Ac était maximum au 5ème mois. La réponse neutralisanteest d’apparition précoce et persiste même après l’élimination virale. Ces observations nous questionnent parrapport au rôle éventuel joué par ces Ac dans les cas de recontamination. / Only 30% of Hepatitis C virus infected individuals recover spontaneously. We investigated the mechanismsleading to early HCV clearance. The purpose of this work was: (i) to explore the diversity and the early geneticevolution of the HCV envelope glycoproteins, and the infectivity spectrum of isolated variants and (ii) toanalyze the ability of the autologous neutralizing response to control these variants. We selected two patientswho developed an acute HCV infection. To explore the impact of mutations on infectivity and neutralization,retroviral pseudoparticules were produced with representative E1 and E2 sequences. For the first case, themaximum neutralizing activity was observed in the serum collected between 2 and 3 months post ALT peak,the activity was still detectable after 30 months. For the second case, autologous neutralizing activity wasdetected in every serum collected between 4 days and 13 months after. A gradual increase of neutralizationactivity was observed over time with a maximum 5 to 6 months. We have shown that the neutralizing responsewas detectable at early stages of primoinfection and was sustained beyond the time at which the virus wascleared. These observations raise interesting questions about the role of such antibodies in case of re-exposure.

Análises sorológicas e filogenéticas de amostras de herpesvírus bovino tipos 1 e 5 / Serological and phylogenetic analysis of bovine herpesvirus types 1 and 5 isolates

O presente estudo foi conduzido com o objetivo de determinar se a sensibilidade do teste de Soroneutralização (SN) seria afetada quando utilizados distintos subtipos de herpesvírus bovino tipos 1 (BoHV-1) e 5 (BoHV-5). Dessa forma, soros de bovinos, coletados randomicamente (n= 287) foram testados por SN frente a três amostras de BoHV-1 (BoHV-1.1: EVI123/98 e Los Angeles (LA); BoHV-1.2a: SV265/96) e três amostras de BoHV-5 (BoHV-5a: EVI88/95; BoHV-5b: A663 e BoHV-5c: ISO 97/95), utilizando um período de incubação soro-vírus de 24 horas. A sensibilidade da SN variou significativamente dependendo da amostra viral utilizada. Esta variação foi de 77% (80/104 soros positivos) até 91% (95/104) com as amostras ISO 97/95 e LA, respectivamente, quando cada vírus foi considerado individualmente. A sensibilidade máxima (104/104) foi obtida quando os resultados positivos de uma combinação particular de quatro vírus (LA + EVI123 + SV265 + A663), algumas combinações de cinco vírus, ou ainda, todos os seis vírus foram adicionados. Estes resultados evidenciaram que quando a SN é realizada frente a uma única amostra viral, a sensibilidade pode variar significativamente, podendo comprometer programas de controle e erradicação das infecções por estes agentes. Além disso, a realização de SN frente a diferentes isolados virais mostrou aumentar significativamente a sensibilidade do teste. Com isso, a caracterização de novos isolados de campo pode favorecer futuras avaliações de diferentes subtipos de BoHV-1 e BoHV-5 e contribuir com a escolha de amostra e/ou combinação de amostras mais sensíveis. Deste modo, buscou-se caracterizar isolados de campo de BoHV-1 e BoHV-5 com base na análise molecular da região carboxi-terminal do gene que codifica a glicoproteína C (gC) e na análise com enzima de restrição (REA) do genoma viral. Para tanto, a multiplicação de 24 isolados da América do Sul foi realizada em células CRIB para posterior extração do DNA viral, PCR e sequenciamento. As seqüências foram alinhadas utilizando o programa ClustalX2 para uma inferência filogenética pelo método de Neighbor-Joining (Mega 4.0), Kimura 2-parâmetros. O alinhamento das seqüências de nucleotídeos revelou níveis de identidade variando de 70 a 99,6% entre os isolados de BoHV-1; de 66,9 a 100% entre os isolados de BoHV-5 e de 62,9 a 92,8% entre os isolados BoHV-1 e BoHV-5. A árvore filogenética mostrou o agrupamento dos vírus de acordo com o tipo (BoHV-1 e BoHV-5) e subtipo de BoHV-1 (BoHV-1.1 e BoHV-1.2). No entanto, essa técnica não permitiu a diferenciação dos isolados em diferentes subtipos de BoHV-5. Do mesmo modo, a análise por restrição enzimática não proporcionou uma clara diferenciação dos isolados em subtipos devido à presença de variações nos padrões de restrição. Somente um isolado (ISO 94/232) pode ser diferenciado como subtipo 5a. Todavia, este estudo mostrou que a análise filogenética utilizada representa uma potencial ferramenta para a diferenciação e classificação dos vírus em BoHV-1.1, BoHV-1.2 e BoHV-5. No entanto, ambas as técnicas podem ser empregadas, de maneira complementar, quando maiores informações sobre estes vírus forem requeridas. Além disso, com o presente estudo, foi possível expandir o número de amostras caracterizadas, fornecendo subsídios para estudos futuros. / This study was carried out to determine whether the sensitivity of serum neutralization (SN) tests would be affected by the use of distinct subtypes of bovine herpesvirus 1 (BoHV- 1) and 5 (BoHV-5) as test challenge viruses. Bovine sera collected from a randomized sample (n = 287) were tested in a 24 hour incubation SN against three type 1 viruses (BoHV-1.1 strains „„Los Angeles‟‟ (LA) and „„EVI 123‟‟; BoHV-1.2a strain „„SV 265‟‟) and three type 5 viruses (BoHV-5a strain „„EVI 88‟‟; BoHV-5b strain „„A 663‟‟ and BoHV-5c „„ISO 97‟‟). SN sensitivity varied greatly depending on the challenge virus used in the test, particularly when results against each virus were considered individually, where it ranged from 77% (detecting 80 out of 104 antibody-positive sera) to 91% (95/104) with ISO 97/95 and LA strain, respectively. Maximum sensitivity (104/104) was achieved when positive results to a particular combination of four of the challenge viruses (LA + EVI 123 + SV 265 + A 663) or some combinations of five viruses (or all six viruses) were added cumulatively. These results clearly shown that when SN is performed with single test challenge viruses, sensitivity could vary significantly that might compromise control or eradication efforts. Performing SN against a number of different viruses demonstrated to improve significantly the test‟s sensitivity. Thus, news field isolates characterization could aid future evaluations of different BoHV-1 and BoHV-5 subtypes and also provide the better strain and/or strains combination choice. In such case, this study aimed to characterize field isolates of BoHV-1 and BoHV-5 by molecular analyses of glycoprotein C (gC) carboxy-terminal region and viral genome restriction enzymatic analysis (REA). The 24 isolates from South America were propagated in CRIB cells for viral DNA extraction, PCR and sequencing. The sequences were aligned in ClustalX2 to perform a distance–based phylogenetic analysis by Neighbor-Joining method in MEGA 4.0 software under de Kimura 2-parameter. The nucleotide sequence alignments revealed levels of genomic similarity ranging from 70% to 99.6% between BoHV-1 isolates; from 67% to 100% among BoHV-5 isolates and from 63% to 93% between BoHV-1 and BoHV-5 isolates. The phylogenetic tree clustered the viruses according to types (BoHV-1 and BoHV-5) and BoHV-1 subtypes (BoHV-1.1, -1.2). However, this method did not allow clearly differentiated in BoHV-5 subtypes. Likewise, REA did not clear showed differentiation in subtypes due presence variation in restriction pattern. Only one isolate (ISO 94/232) could be differentiated in subtype 5a. The results suggest that the phylogenetic analysis performed could be an important tool for differentiation and classification of such viruses in BoHV-1.1, BoHV-1.2, BoHV-5. However, when adiccional informations were resquested both techniques should be performed. Furthermore, with this work it was possible to expand the number of samples characterized to support future investigation of these viruses.

Ao falar de qualidade na indústria farmacêutica, fala-se, sobretudo da qualidade físico-química e microbiológica tanto das matérias-primas empregadas na fabricação como do produto terminado obtido após as diversas etapas produtivas. Com objetivo de alcançar esta qualidade este projeto buscou verificar as metodologias empregadas no dia a dia da Divisão de Controle de Especificação do Laboratório Farmacêutico da Marinha (LFM). As técnicas empregadas no controle microbiológico, em sua maioria, são as mesmas, consistindo na realização do ensaio limite (contagem de micro-organismos) e na pesquisa e identificação de patógenos. No entanto deve-se verificar se o produto analisado não afeta o crescimento microbiano, inibindo-o e fazendo com que se tenha resultados falso-negativos, o que acarretaria em desvios da qualidade podendo levar a danos para a saúde do usuário e prejuízos para a imagem da Instituição. Elegeram-se as seguintes formulações, todas de uso oral, para serem avaliadas: aciclovir 200 mg comprimidos, isoniazida 100 mg comprimidos, pirazinamida 500 mg comprimidos, ofloxacino 400 mg comprimidos, pirazinamida suspensão 3%, bromexina xarope, prednisona 5 mg comprimidos e complexo vitamínico e minerais comprimidos. Estas formulações foram avaliadas frente às cepas de referência, conforme procedimentos preconizados pela Farmacopeia Brasileira 5ª Ed. Durante o ensaio verificou-se que todas as formulações, exceto o aciclovir e a pirazinamida suspensão, inibiram o crescimento das cepas testadas. Foram testados métodos de neutralização da atividade inibitória do crescimento microbiano, tendo sido selecionados os seguintes procedimentos: diluição para as formulações de isoniazida 100 mg, prednisona 5 mg e complexo vitamínico, uso de neutralizante, polissorbato 80, para bromexina xarope e filtração por membrana para pirazinamida 500 mg comprimidos, logrando-se êxito para o ensaio do limite. Somente para o ofloxacino não foi possível recuperar os micro-organismos e no caso da pirazinamida comprimidos, a mesma não foi susceptível à pesquisa de Escherichia coli, nas condições preconizadas pela Farmacopeia Brasileira 5ª Ed. O presente trabalho identificou que das oito formulações testadas, seis inibem o crescimento microbiano e, portanto interferem na análise. Após adequações, os métodos responderam aos testes e foram desta forma considerados validados, podendo ser utilizados de forma segura na rotina do laboratório. / Nowadays it is remarkable the development of the Quality Management, together with it the Good Manufacturing Practices and Control, which are tools that help to afford products with high quality. Concerning to quality issues in the Pharmaceutical Industry, the main objective is to achieve high physical-chemical and microbiological quality of the raw materials used for manufacturing, as well of the final product after the several processing steps. Aiming to succeed in attaining this quality parameter, this project has attempted to verify the methodologies daily employed by the Specification Control Division of Brazilian Navy Pharmacautical Laboratory (LFM). The techniques employed for the microbiological quality control are usually the same, and consist of performing the limit assay (microorganism counting) and the search and identification of pathogens. Nevertheless, it must be checked if the product under analysis does not affect the microbial growth, causing an inhibition and leading to false-negative results, which might drive to quality deviance, ultimately harming the final user and bringing damage to the institution image. In this project, the following oral formulations have been selected to be evaluated: aciclovir 200 mg tablets, isoniazid 100 mg tablets, pyrazinamide 500mg tablets, ofloxacin 400 mg tablets, pyrazinamide 3% suspension, bromexine syrup, prednisone 5 mg tablets and tablets containing vitamin complex and minerals. These formulations have been evaluated against the growth of reference strains, in accordance to the procedures described in the Brazilian Pharmacopeia 5th Edition. During the assay, it has been found all formulations, exemption given to aciclovir and pyrazinamide suspension, inhibited the growth of the assayed strains. Methods for neutralizing the microbial growth inhibition activity have been assessed and the selected procedures were: diluition for isoniazid 100mg, prednisone 5 mg and vitamin complex; use of the neutralizer polysorbate 80 for bromexine syrup; and membrane filtration for pyrazinamide 500 mg tablets, which allowed the recovery of all microorganisms. Only for ofloxacin it was not possible to recover the microorganisms and for pyrazinamide tablets, the formulation was not susceptible to the Escherichia coli assay, pursuant to the Brazilian Pharmacopeia 5th Edition. The present study has identified that six out of the eight assayed formulations have inhibited microbial growth, therefore interfering in the analysis outcome . After adjustments, the methods have trustworthy worked and were thus considered validated, and now can be applied safely in the laboratory's routine.
