Targeted content - a case study on increasing online user engagement

The data generated from the user's activities and surfing online can have a significant impact when used to improve the content of web pages. The user has little time to scan the content of a web page and the motivation to save time is high. Recommendations and personalized content are becoming more and more common and both businesses and consumers seem to benefit from more targeted content. In order to know what recommendations to present to each individual, information about the user is needed and there are many tools to analyze the behaviours of the users online. In this study, we examined if relevant content on the front-page of Visma, a business software provider, can increase the interaction on the front-page and what metrics can be used to measure the interaction. Together with experts in the field we investigated the eAccounting software users’ preferences for targeted recommendations. By having a pop up survey for users of Visma, conducting an A/B test and using a heatmap for the eAccounting users we found out that a more customized front-page makes the user interact more with the page.

Leadership in times of terrorism threat : An Analysis of George W. Bush’s and Barack Obama’s leadership qualities concerning counterterrorism

While the US-presidency comprises the most powerful office of leadership in the world and has also been investigated at various angles in personal, biographical approaches as well as comparative studies, this research paper specifically lays focus and compares the two presidencies of President Bush and President Obama in regard of their leadership qualities and counter-terrorism strategies. Because counterterrorism seems to be a good example of how presidents perform crises-management and because the subject by itself is critical in the perspective of human rights and security concerns, the research paper has chosen to focus on President Bush and President Obama who are known to have chosen different routes and strategies in countering terrorism. However, when comparing the two presidents’ leadership qualities regarding their counter-terrorism strategies, this paper concludes that on this matter the strategies of the two presidents were not quite so different as expected and a possible path of continuity runs through this realm of US-foreign affairs and presidents’ leadership decisions.

Differential Responses of MET Activations to MET kinase Inhibitor and Neutralizing Antibody

Background: Aberrant MET tyrosine kinase signaling is known to cause cancer initiation and progression. While MET inhibitors are in clinical trials against several cancer types, the clinical efficacies are controversial and the molecular mechanisms toward sensitivity remain elusive. Methods: With the goal to investigate the molecular basis of MET amplification (MET amp ) and hepatocyte growth factor (HGF) autocrine-driven tumors in response to MET tyrosine kinase inhibitors (TKI) and neutralizing antibodies, we compared cancer cells harboring MET amp (MKN45 and MHCCH97H) or HGF-autocrine (JHH5 and U87) for their sensitivity and downstream biological responses to a MET-TKI (INC280) and an anti-MET monoclonal antibody (MetMab) in vitro, and for tumor inhibition in vivo. Results: We find that cancer cells driven by MET amp are more sensitive to INC280 than are those driven by HGF-autocrine activation. In MET amp cells, INC280 induced a DNA damage response with activation of repair through the p53BP1/ATM signaling pathway. Although MetMab failed to inhibit MET amp cell proliferation and tumor growth, both INC280 and MetMab reduced HGF-autocrine tumor growth. In addition, we also show that HGF stimulation promoted human HUVEC cell tube formation via the Src pathway, which was inhibited by either INC280 or MetMab. These observations suggest that in HGF-autocrine tumors, the endothelial cells are the secondary targets MET inhibitors. Conclusions: Our results demonstrate that MET amp and HGF-autocrine activation favor different molecular mechanisms. While combining MET TKIs and ATM inhibitors may enhance the efficacy for treating tumors harboring MET amp , a combined inhibition of MET and angiogenesis pathways may improve the therapeutic efficacy against HGF-autocrine tumors.

Répercussions psychosociales des symptômes dermatologiques induits par les thérapies ciblées anticancéreuses / Psychosocial impact of dermatological toxicities induced by targeted therapies in cancer

Contexte. Synonymes de progrès thérapeutique, les thérapies ciblées anticancéreuses ne sont pas sans effet secondaire, en particulier dermatologique. Très peu de données sont disponibles quant à leur impact sur la qualité de vie. Le service de dermatologie de Gustave Roussy a consacré une partie de son activité de recherche à cette thématique. Inscrite dans ces travaux, notre thèse avait pour objectif principal de décrire les changements observés du point de vue de l'état émotionnel, de l'image corporelle et des interactions sociales avec l'apparition des atteintes cutanées, en s'intéressant à la place des représentations associées au traitement dans le processus d'ajustement des patients.Méthode. L'étude prospective comprenait quatre temps d'évaluation (initiation du traitement, un, deux et trois mois après) et associait deux modes d'évaluation : quantitatif (questionnaires) et qualitatif (entretiens semi-directifs). L'inclusion était proposée par les oncologues aux patients allant débuter une thérapie ciblée. Les analyses statistiques ont été menées avec le logiciel SPSS 14.0 ; l'analyse des entretiens a combiné méthode thématique et méthode par questionnement analytique, en recourant au modèle théorique de Pedinielli. Résultats. Sur 82 patients inclus dans la recherche biomédicale, 62 ont accepté de participer à l'étude psychologique. La partie quantitative a été complétée par 33 patients, pour moitié hommes (âge moyen 56 ans) soignés pour un cancer métastatique ; 84% a développé au moins un des symptômes suivants: rash cutané, syndrome main-pied, alopécie ou photosensibilité. Les changements observés ont été un inconfort physique et une gêne à la réalisation des activités du quotidien. Aucun signe de perturbation de la sphère émotionnelle, de l'image du corps et des relations sociales n'a été mis en évidence. La représentation d'un médicament contrôlant la maladie a émergé comme un des facteurs significativement associés aux variations de l'impact des toxicités cutanées sur la qualité de vie. La partie qualitative a concerné 41 patients (caractéristiques médicales et sociodémographiques très similaires à celles de l'échantillon quantitatif). Pour une majorité les symptômes dermatologiques ont été « gênants », voire « perturbants » lorsqu'ils entraînaient douleurs, difficultés à la mobilité ou troubles du sommeil, mais sont restés « gérables, supportables ». Les représentations associées au traitement, très positives, sont apparues comme un élément soutenant dans l'ajustement des patients. Du discours des patients en souffrance psychologique sont ressorties une défiance vis-à-vis du regard d'autrui et une impossibilité d'amorcer le travail de renoncement nécessaire à l'intégration des transformations liées au cancer et à ses traitements. L'origine de cette souffrance serait un débordement des défenses psychiques par une angoisse de mort : la difficulté pour restaurer l'état d'équilibre psychique antérieur provenant de l'activation concurrentielle de deux dynamiques, l'une surnommée « substantielle », l'autre « identitaire ».Discussion. Ces résultats rejoignent les données de la littérature en concluant à un impact d'intensité faible à modérée des toxicités cutanées sur la qualité de vie pour une majorité de patients. Contrairement à ce qui était attendu, il n'a pas été observé de changement sur le plan de l'état émotionnel, de l'image corporelle et des interactions sociales. L'investissement positif du traitement, la réappréciation des valeurs, le très bon état général des patients et l'optimisme ont été évoqués l'absence de perturbation. L'importance de l'encadrement médico-soignant a aussi été soulignée.Conclusion. Le développement croissant des thérapies ciblées appelle à une consolidation des mesures de prévention et de prise en charge des symptômes dermatologiques, ce qui nécessite un renforcement des actions de formation et de sensibilisation des acteurs de soin à cette problématique. / Background. Considered as a significant therapeutic progress in cancer, targeted agents are not without side effects, particularly dermatological ones. Very little information is presently available about their consequences on patients' quality of life, that is why the dermatological team of Gustave Roussy has developed a prospective research. Integrated into this work, our thesis aimed to describe the psychosocial changes occurring with cutaneous toxicities and to explore the implication of treatment representations in the patient's adjustment process.Methods. The study included four phases of evaluation (treatment initiation, one, two and three months after) and used two methods: a quantitative one (questionnaires) and a qualitative one (semi-structured interviews). The inclusion was proposed by oncologists to patients who were about to start a targeted therapy. Statistical analyzes were conducted with SPSS 14.0 software; analyzes interviews combined thematic approach and analytical questioning methods and referred to the Pedinielli's theoretical model.Results. Eighty- two patients gave their consent for biomedical research, 62 of them agreed to participate to the psychological study. The quantitative part was completed by 33 patients (50% men, mean age 56 years) treated for a metastatic cancer; 84% developed at least one of the following symptoms: skin rash, hand-foot syndrome, alopecia or photosensitivity. The observed changes were characterized by a physical discomfort and difficulties in the activities of daily life. No sign of disturbance was noted in emotional domain, body image or social relations. The representation of a drug controlling the disease was significantly associated with a lower impact of skin problems on the quality of life. The qualitative part included 41 patients (medical and sociodemographic characteristics very similar to those of the quantitative sample). For most people, dermatological side effects were "boring", "uncomfortable", sometimes "impressive" or "disturbing" when they were associated with pain, mobility difficulties or sleeping troubles, but remained "manageable, bearable". The very positive treatment representations appeared as a supporting element in patients' adjustment. Psychological distress seemed appear when patients feared being stared by others and failed to engage themselves in the renouncement work which is needed to adjust oneself to the transformations related to cancer and its treatments. In such situation psychological distress was supposed to come from an overflow of the psychic defences by a fear of death; the difficulty to restore mental balance would be explained by the activation of two competitive dynamics, which struggle for the organism and the identity survival.Discussion. These results are consistent with the literature data. The skin toxicities impact on quality of life is mild to moderate for a majority of patients. Contrary to our expectations, there was no evidence of change in the domains of emotions, body image and social interactions. The positive investment of treatment, a reassessment of values, the very good physical state of patients and the influence of optimism in patients state of mind have been cited to explain not only the absence of disturbance, but also the very low levels of anxiety, sadness and fatigue generally reported. The importance of the caregiving provided by health professionals was also highlighted: anticipating difficulties, giving information about side effects and effectively managing problems secure patients.Conclusions. The growing development of targeted therapies implies strengthening prevention and management of dermatological side effects. Moreover, it requires to aware and to train more health professionals to this problem, recalling the singular dimension of each patient which can not being reduced to the medical observable.

Development of a functional assay for CHD7, a protein involved in CHARGE syndrome / Mise au point d'un test fonctionnel pour la protéine CHD7 impliquée dans le syndrome CHARGE

Le syndrome CHARGE (CS) est une maladie génétique rare caractérisée par de nombreuses anomalies congénitales, majoritairement causées par des altérations de novo du gène CHD7. Celui-ci code pour une protéine à chromodomaines, impliquée dans le remodelage ATP-dépendant de la chromatine. La grande majorité des altérations de CHD7 consiste en allèles nuls tels que des délétions, des substitutions non-sens ou des décalages du cadre de lecture. Nous avons réalisé le premier diagnostic moléculaire d’un patient Indonésien atteint du CS, en étudiant un panel de gènes (CHD7, EFTUD2, et HOXA1) par NGS (next-generation sequencing). Nous avons identifié une nouvelle mutation non-sens hétérozygote dans l’exon 34 du gène CHD7 (c.7234G>T ou p.Glu2412Ter). Par ailleurs, il n'existe pas d’analyse fonctionnelle qui permettrait de caractériser la pathogénicité des variants de la protéine CHD7 rencontrés chez des patients. C’est pourquoi l’objectif de ce travail est de mettre au point un test fonctionnel de la protéine CHD7, sous forme sauvage ou mutée. Pour cela, nous avons généré par mutagénèse dirigée des vecteurs codant pour trois variants faux-sens de CHD7 et le variant présentant une insertion de cinq acides aminés. Ensuite, les protéines CHD7, sous forme sauvage ou variante, ont été surexprimées dans la lignée HeLa. L’expression des protéines a été mise en évidence par western blot et par immunofluorescence. Pour étudier la fonctionnalité de CHD7, nous avons quantifié par RT-qPCR les transcrits de cinq gènes (l’ADNr 45S, SOX4, SOX10, MYRF, et ID2), dont la transcription est selon le littérature régulée par CHD7. Nous avons observé que l’expression de CHD7 sauvage entraînait une diminution significative et reproductible des quantités de transcrits correspondant à tous les gènes rapporteurs. Par contre, l’expression des quatre allèles variants de CHD7 n’avait aucun impact, ce qui suggère que ces variants ne sont pas fonctionnels. Par ailleurs, nous avons appliqué notre test biologique dans des cellules de la lignée SH-SY5Y, pour lesquelles nous avons introduit une mutation faux-sens dans le génome en utilisant la technique CRISPR/Cas9. Lorsque ce variant était exprimé, les niveaux de transcription des cinq gènes rapporteurs n’étaient pas significativement différents de ceux observés dans les cellules où les deux allèles de CHD7 avaient été invalidés. Par conséquent, les variants étudiés peuvent être répertoriés comme résultant de mutations causales du CS. / CHARGE syndrome (CS) is a rare genetic disease characterized by numerous congenital abnormalities, mainly caused by de novo alterations of the CHD7 gene. It encodes a chromodomain protein, involved in the ATP-dependent remodeling of chromatin. The vast majority of CHD7 alterations consists in null alleles like deletions, non-sense substitutions or frameshift-causing variations. We report the first molecular diagnosis of an Indonesian CS patient by a targeted NGS (next-generation sequencing) gene panel (CHD7, EFTUD2, and HOXA1). We identified a novel heterozygous nonsense mutation in exon 34 of CHD7 (c.7234G>T or p.Glu2412Ter). Functional analyses to confirm the pathogenicity of CHD7 variants are lacking and urgently needed. Therefore, the aim of this study was to establish a functional test for wild-type (WT) or variants of CHD7 protein found in CS patients. Using an expression vector encoding CHD7, three variants harboring an amino acid substitution and one variant with a five-amino acid insertion were generated via site-directed mutagenesis. Then CHD7 proteins, either wild-type (WT) or variants, were overexpressed in HeLa cell line. Protein expression was highlighted by western blot and immunofluorescence. We then used real-time RT-PCR to study CHD7 functionality by evaluating the transcript amounts of five genes whose expression is regulated by CHD7 according to the literature. These reporter genes are 45S rDNA, SOX4, SOX10, ID2, and MYRF. We observed that, upon WT-CHD7 expression, the reporter gene transcriptions were downregulated, whereas the four variant alleles of CHD7 had no impact. This suggests that these alleles are not polymorphisms because the variant proteins appeared non-functional. Furthermore, we applied our biological assay in SH-SY5Y cell line in which endogenous CHD7 gene was mutated using the CRISPR/Cas9 technique. Then, we observed that when a CHD7 missense variant was expressed, the transcription levels of the five reporter genes were non-significantly different, compared with the cells in which both CHD7 alleles were knocked-out. Therefore, the studied variants can be considered as disease-causing of CS.

The Effects of Unidirectional Peer Tutoring on Mathematics Outcomes for Students with Learning Disabilities in an Inclusive, Secondary Setting

Students with disabilities included in general education settings often require varying levels of supports and services to execute classroom routines effectively while maximizing their learning opportunities. One support that holds potential to increase outcomes for students with disabilities in general education settings is peer tutoring. Therefore, the purpose of this study was to determine the effects that a targeted, unidirectional tutoring intervention had on the math outcomes for students with learning disabilities in a Secondary Mathematics I class. High school students that had advanced skills in mathematics served as interventionists (i.e., peer tutors) and were trained to deliver the unidirectional tutoring intervention to students with learning disabilities who were identified as low achievers in mathematics. All three tutees increased their criterion and normative (i.e., as compared to their peers) performance on teacher-developed weekly mathematics quizzes when receiving the tutoring intervention. Furthermore, all three tutees increased their quantity and quality of mathematics problems completed as well as academic engagement during the tutoring intervention. Finally, tutees, tutors, and a general education teacher indicated that they perceive the unidirectional tutoring intervention as effective and socially desirable.

Assessing Educators’ School Safety and Security Preparedness at a New Jersey K-12 Nonpublic School

School shootings and emergencies have created the need for educators to be proficient in emergency response procedures; yet they do not always receive the requisite training. The lack of an established delineated training program for New Jersey, kindergarten to Grade 12 institutions has created a situation where educator preparedness varies immensely at schools. Numerous national events of targeted school violence have exemplified the need for quick and proper responses by educators to mitigate the tragic results until first responders arrive. The purpose and goal of this study was to assess educators’ perceptions and to determine the best practices in creating a comprehensive safety and security training program to prepare educators for school crises in order to offer a model for stakeholders to follow or gain ideas to improve their institution’s specific school safety and security emergency plans. Guided by the U.S. Department of Education’s best practices in developing high-quality school safety plans, this study analyzed the perceptions of 60 educators in one New Jersey kindergarten to Grade 12 school on the effectiveness of training. A mixed-methods approach, using a survey questionnaire and interviews, measured changes in the perceptions of these educators after the 15-week program. Data results included a revelation of the implementation of a comprehensive school safety and security plan with related training program resulted in a statistically significant increase in the perceptions of educators’ knowledge and abilities to respond effectively to school targeted violence and emergencies. These findings support the concept that best practices in the field of school safety and security management include appropriate and comprehensive school safety and security plans and training for educators to combat and mitigate school targeted violence and emergent events.

(English) The R&D management system was examined in terms of legislation, managing authorities, types of support, support providers, budget and strategic documents in this thesis. In the area of strategic documents, persistent shortcomings were revealed and manifested in non-specific interconnections or overlaps. At the same time, a qualitative shift in the quality of the documents was identified. Furthermore, it was examined whether the strategic measures for the development in the field of molecular biology were fulfilled in more detail. The research was carried out on data related to targeted support, which were obtained from the R&D Information System. Two periods in the years 1995-2014 were analyzed. The structure of the second period from 2005 to 2014 was related to the structure of the first period from 1995 to 2004, which served as a reference. In relation to strategic measures, research has shown that resources have been optimized, capacities have been strengthened and infrastructure has been massively developed.
