Modélisation et optimisation de la production de cellulases par Trichoderma reesei pour les bioraffineries lignocellulosiques

Dans le contexte énergétique et climatique mondial, le coût élevé des enzymes Cellulolytiques (cellulases) freine le développement des bioraffineries lignocellulosiques, pour produire des biocarburants et composés chimiques à partir d'une matière première végétale renouvelable. L'objectif de ce travail est de caractériser et de modéliser le métabolisme du micro-organisme Trichoderma reesei, afin d'optimiser le protocole industriel de production de cellulases. Cette étude a été réalisée sur des milieux modèles représentatifs de ceux attendus à l'échelle industrielle. Tout d'abord, la stoechiométrie des réactions de croissance et de production a été établie, puis une étude cinétique a été menée pour mesurer précisément le comportement du micro-organisme à forte induction de la production de cellulases. Le modèle résultant a été utilisé pour optimiser le protocole industriel de production. Ensuite, l'intégration de cette étape dans une bioraffinerie lignocellulosique a été étudiée, avec l'effet sur le métabolisme i) des mélanges de sucres disponibles, ii) des composés inhibiteurs issus de la dégradation de la lignocellulose, et iii) du changement d'échelle. Ces travaux ont fait progresser de façon substantielle les connaissances du métabolisme de T. reesei en ce qui concerne la production de cellulases, et les modèles développés sont des outils d'aide rationnelle à la définition d'un procédé de production de cellulases intégré dans une bioraffinerie lignocellulosique. / In the global energetic and climatic context, the high cost of the cellulolytic enzymes (cellulases) postpones the development of lignocellulosic biorefineries, dedicated to produce biofuels and chemical compounds from renewable vegetable feedstocks. The aim of this work was to measure and model the metabolism of the micro-organism Trichoderma reesei, in order to optimize the industrial protocol for the production of cellulase. This study was carried out using synthetic media representative of industrial ones. First, the stoichiometries of growth and protein production reactions were determined. Then, a kinetic study was conducted to precisely measure the specific rates of T. reesei at high induction of cellulase production. The resulting model was used to optimize the industrial production protocol. Finally the integration of this step in a lignocellulosic biorefinery was studied by determining the impacts on the metabolism of i) available sugar mixtures, ii) inhibitory compounds from lignocellulosic biomass degradation, and iii) scale-up. These results significantly contributed to improve the knowledge of T. reesei metabolism on cellulase production. The developed models are rational tools for the optimization of a cellulase production protocol suited to lignocellulosic biorefineries.

Indicators for Minimizing Energy Consumption and GHG Emissions at Wastewater Treatment Facilities

Wastewater treatment facilities around the world use significant amount of energy which contributes to large quantities of greenhouse gas (GHG) emissions. According to the U.S.EPA, nearly 3% of the USA's energy is used to treat wastewater. This consumption is increasing at faster rates with increase in population and regulations. Wastewater facilities use large number of pumps in their transfer stations, treatment plants, and effluent pump stations. All these pumps consume considerable amounts of energy. This study presents a preliminary energy inspection of two facilities from Louisiana. This audit provides an inventory of the energy consumed for various activities like pumping, treatment, and discharge. This analysis helps the operators to identify the potential power consuming areas and optimize by adopting several energy conservation measures (ECMs). This study also involves the quantification of GHG emissions based on the energy consumption. The benefits of the study include minimizing energy and GHG emission.

The removal of color from the galvanostatic electrolysis of synthetic wastewater MR and BN, and the actual sample has been observed under different conditions (different current densities and temperature variation). The investigation of these parameters was performed in order to establish the best conditions for removal of color and chemical oxygen demand (BOD). According to the results obtained in this study, the electrochemical oxidation processes suitable for the degradation process of color and COD in wastewater containing such textile dyes, because the electrocatalytic properties of Pt and BDD anodes consumption energy during the electrochemical oxidation of synthetic solutions AN and MR and real sample, mainly depend on the operating parameters of operation, for example, the synthetic sample of MR, energy consumption rose from 42,00kWhm-3 in 40 mAcm-2 and 25 C to 17,50 kWhm-3 in 40mAcm-2 and 40 C, from the BN went 17,83 kWhm-3 in 40mAcm and 40?C to 14,04 kWhm- 3 in 40mAcm-2 and 40 C (data estimated by the volume of treated effluent). These results clearly indicate the applicability of electrochemical treatment for removing dyes from synthetic solutions and real industrial effluents / Neste trabalho, a tecnologia eletroqu?mica foi utilizada no tratamento de efluentes sint?ticos, contendo Vermelho de Metila (VM) e Azul de Novacron (AN), atrav?s da oxida??o an?dica utilizando ?nodos de Ti recoberto com platina (Ti/Pt). Optimisation of sludge pretreatment by low frequency sonication under pressure / Optimisation du prétraitement de boues par ultrasons à très basses fréquences et sous pression

L'objectif de ce travail est d'optimiser le prétraitement de boues par des ultrasons de puissance (US) à basses fréquences, et en particulier d‘étudier pour la première fois des améliorations possibles en modifiant la pression hydrostatique, et la fréquence jusqu’à l’audible. Après un examen préliminaire des conditions du procédé (conditionnement des boues, type de boues, alcalinisation préalable, contrôle de la température), les effets des paramètres ultrasonores (puissance, intensité, énergie spécifique, fréquence) et de la pression hydrostatique ont été spécifiquement étudiés, séparément et simultanément, d’abord à température constante (28°C), puis sans refroidissement. On a ainsi vérifié que l’énergie spécifique joue un rôle clé dans la désintégration des boues sous US (i.e. solubilisation de la matière organique) et que l'élévation de température pendant la sonication adiabatique est bénéfique grâce aux effets combinés d’hydrolyse thermique et de cavitation. Pour une énergie spécifique donnée, une faible fréquence (12 kHz contre 20 kHz) et une haute puissance améliorent la solubilisation de la matière organique grâce à une cavitation plus violente, tandis qu’on observe un optimum de pression hydrostatique en raison de ses effets opposés sur le seuil et l'intensité de la cavitation. Un résultat important est que la pression optimale dépend de l’intensité ultrasonore et du profil de température, mais pas de l’énergie spécifique, ni de la fréquence, ni du type de boues. Après avoir fixé les conditions les plus favorables (soit 12 kHz, 360 W, 28 gTS/L et conditions adiabatiques), l‘optimisation finale a fourni la pression de travail (3,25 bar) et les paramètres du mode séquentiel (US ON/OFF, permettant d‘éviter de hautes températures qui amortissement l‘intensité de la cavitation et peuvent endommager le transducteur). Ces conditions ont permis d‘atteindre un rendement d’extraction de la DCO très élevé, mais n’améliorent que faiblement le rendement ultérieur de méthanisation. / The objective of this work is to optimize high-power low-frequency sonication (US) pretreatment of sludge, and especially to investigate for the first time possible improvements by higher pressure and audible frequency. After a preliminary examination of regular process conditions (sludge conditioning, sludge type, prior alkalization, temperature control, etc.), effects of US parameters (power -PUS, intensity -IUS, specific energy input -ES, frequency -FS, etc.) and of hydrostatic pressure (Ph) were specifically looked into, separately and in combination, first under cooling at constant temperature (28°C), then under the progressive temperature rise provoked by sonication. First, it was confirmed that specific energy input (ES) plays a key role in sludge US disintegration (i.e. solubilisation of organic matter) and that temperature rise during adiabatic-like sonication is beneficial through additional effects of thermal hydrolysis and cavitation. At a given ES value, low FS (12 kHz vs. 20 kHz) and high PUS enhance soluble chemical oxygen demand (SCOD) due to more violent cavitation, while hydrostatic pressure gives rise to an optimum value due to its opposite effects on cavitation threshold and intensity. One major result is that optimal pressure depends on IUS (PUS) as well as temperature profile, but not on ES, FS, nor sludge type. Setting the other parameters at the most favorable conditions expected, i.e. 12 kHz, 360 W , 28 gTS/L, and adiabatic conditions, final optimization was achieved by searching for this pressure optimum and examining sequential procedure to avoid too high temperature dampening cavitation intensity and damaging the transducer. Such conditions with sequential mode and Ph of 3.25 bar being selected succeeded in achieving very high SCOD, but only marginally improved subsequent methanization yield.

Oxigênio dissolvido e demanda bioquímica de oxigênio no Rio Uberabinha: um estudo da poluição orgânica biodegradável

O rio Uberabinha, localizado na região do Triângulo Mineiro, em Minas Gerais, compõe a sub-bacia do rio Araguari, afluente da bacia do rio Paranaíba, na região hidrográfica do rio Paraná. Mede, da sua nascente no município de Uberaba à foz no rio Araguari, aproximadamente 150 km, percorrendo cerca de 135 km no município de Uberlândia. Drena uma bacia de cerca de 2.190 km2, onde se localizam as captações da água que abastece uma população que hoje se aproxima de meio milhão de habitantes. Os esgotos produzidos pela cidade são também todos descarregados no rio. O trabalho consistiu na análises de pH, temperatura, oxigênio dissolvido e demanda bioquímica de oxigênio em amostras coletadas em cinco pontos do rio Uberabinha, a partir de uma seção a montante da cidade, até cerca de 20,5 km a jusante do último ponto de lançamento de esgoto, abrangendo uma extensão de aproximadamente 42 km. Neste trecho, o relevo passa de medianamente dissecado para intensamente dissecado, com presença de inúmeras corredeiras e cachoeiras. As amostras foram coletadas de junho de 1999 a maio de 2000 e a vazão também foi medida nos dois pontos extremos de coleta. Os resultados mostram que os teores de OD sofrem uma redução ao receber a carga de poluição e gradativamente tendem a recuperar sua taxa de saturação. Em contrapartida, a DBO segue uma curva inversa, denotando a atuação do fenômeno da autodepuração, embora dentro do trecho estudado não se verifique ainda a completa remoção da carga orgânica. / The Uberabinha River, located in the region of Triângulo Mineiro, in the state of Minas Gerais, Brazil, is part of the Araguari River sub-basin. From its head in the city of Uberaba to its mouth in the Araguari River, it measures approximately 150 km, spanning about 135 km in the municipality of Uberlândia. The river drains a watershed of about 2.190 km2, from which water is collected to supply a population that approaches half a million inhabitants. Later, the sewage produced by the city is also discharged directly into the river. This research consisted in the analyses of pH, temperature, dissolved oxygen and biochemical oxygen demand of samples collected in five points of the Uberabinha River, from a section starting upstream of the urban area down to 20.5 km downstream the last point of sewage discharge, encompassing approximately 42 km. In this river section, the landforms go from low to medium hills with many rapids and waterfalls. The samples were collected from June 1999 to May 2000 and the water flow was measured on the two extremes of the studied river section. The results show that the dissolved oxygen values are reduced when receiving the sewage and it tends to slowly return to its normal saturation rate. On the other hand, the biochemical oxygen demand follows a reverse path, as a result of the natural phenomenon of self-restoration even though the organic material is not yet completely removed from the studied river section. / Dissertação (Mestrado)

The constantly tightening wastewater treatment legislation require high-quality runoff from small-scale producers of pollution who are typically represented by small municipalities up to 500 inhabitants or decentralized parts of larger municipalities. Small municipalities are faced with high investment-intensity associated with the acquisition of new wastewater treatment plant. After several years of development, today natural technologies of wastewater treatment are in competition with mechanical-biological treatment plants. Natural technologies can even achieve better results than popular biological treatment plants. However, natural technology does not eliminate the main investment limit connected with the acquisition of wastewater treatment plant. If there was cheaper solution connected with using alternative filter materials (in our case recycled construction and demolition waste), small municipalities could take constructed wetland without large subsides. At the same time, the use of recycled construction and demolition wastes will lead to benefits in the field of sustainable urban development and replacement of non-renewable materials. Despite these benefits, recycled aggregates have never been considered as alternative filter materials for natural wastewater treatment plants. For that reason, the aim of diploma thesis is to assess the possibility of applying recycled aggregates for wastewater treatment which will include evaluation of real trial operation.

Effect of substrate types on fermentation of mixed cultures to produce volatile fatty acids / Effekt av substrattyper för fermentering av blandade kulturer för att producera flyktiga fettsyror

Innovativa teknologier, produkter och processer krävs på grund av marknadens ökande behov av bioprodukter med låga koldioxidavtryck. Idén att producera flyktiga fettsyror via biologiska processer, främst genom anaerob fermentering med blandkulturer, har återupplivats på grund av den omfattande exploateringen av oljeresurser. Målet för denna studie var att utvärdera kvaliteten och livskraften för produktion av flyktiga fettsyror från enkla substrat. Flyktiga fettsyror-generering undersöktes i ett satsvis syrafermenteringssystem för tre enkla substrat (olja, stärkelse, och gelatin). Vi visade att ett initialt surt pH var fördelaktigt för acidogenes och den totala genereringen av flyktiga fettsyror för alla tre substraten. Den högsta produktionseffektiviteten av flyktiga fettsyror uppnåddes och flyktiga fettsyror genomgick evolutionär utveckling med blandad kulturfermentering som kördes vid ett initialt surt pH på 5,0. 7933,08 mg COD/L smörsyra producerades från stärkelse vid pH 3,87 på dag 15, vilket var den högsta nivån. Smörsyra och ättiksyra var de dominerande produkterna i det initiala sura pH-värdet 5. Alla tre substraten i batch-reaktorer visade högt produktionsutbyte av ättiksyra och smörsyra. Vid lågt initialt pH var propionsyra den vanligaste syratypen i olja. / Innovative technology, products, and processes are required because of the market's rising need for bioproducts with low carbon footprints. The concept of producing volatile fatty acids (VFAs) via biological processes, primarily through anaerobic mixed culture fermentation, has been revived because of the extensive exploitation of oil resources. The goal of this study was to evaluate the quantity and efficiency of VFA production from simple substrates. The generation of VFA was examined using a batch type acid fermentation system for three simple substrates (oil, starch, and gelatin). We showed that an initial acidic pH was advantageous for acidogenesis and the total generation of VFAs for all three substrates. The VFA production efficiency was achieved and underwent evolutionary development with mixed culture fermentation (MCF)  tests running at an initial acidic pH of 5.0. 7933.08 mgCOD/L butyric acid were produced from starch at pH 3.87 at day 15, which was the highest level. Butyric and acetic acids were the predominant products in the initial acidic pH 5. All the three substrates in reactors showed high VFA production yield of acetic acid and butyric acid. At low initial pH, propionic acid was the most prevalent acid type in oil.

Soil aquifer treatment of artificial wastewater under saturated conditions

A 2000 mm long saturated laboratory soil column was used to simulate soil aquifer treatment under saturated conditions to assess the removal of chemical and biochemical oxygen demand (COD and BOD), dissolved organic carbon (DOC), nitrogen and phosphate, using high strength artificial wastewater. The removal rates were determined under a combination of constant hydraulic loading rates (HLR) and variable COD concentrations as well as variable HLR under a constant COD. Within the range of COD concentrations considered (42 mg L(-)(1)-135 mg L(-)(1)) it was found that at fixed hydraulic loading rate, a decrease in the influent concentrations of dissolved organic carbon (DOC), biochemical oxygen demand (BOD), total nitrogen and phosphate improved their removal efficiencies. At the high COD concentrations applied residence times influenced the redox conditions in the soil column. Long residence times were detrimental to the removal process for COD, BOD and DOC as anoxic processes and sulphate reduction played an important role as electron acceptors. It was found that total COD mass loading within the range of 911 mg d(-)(1)-1780 mg d(-)(1) applied as low COD wastewater infiltrated coupled with short residence times would provide better effluent quality than the same mass applied as a COD with higher concentration at long residence times. The opposite was true for organic nitrogen where relatively high concentrations coupled with long residence time gave better removal efficiency.

Effect of a Grass and Soil Filter on Tucson Urban Runoff: A Preliminary Evaluation

From the Proceedings of the 1972 Meetings of the Arizona Section - American Water Resources Assn. and the Hydrology Section - Arizona Academy of Science - May 5-6, 1972, Prescott, Arizona / Storm runoff from the Tucson metropolitan area is unsuitable for most uses without processing. A lysimeter comprised of a grass and soil filter was constructed and is being evaluated as a water-quality treatment facility. The lysimeter is 200 feet long, 4 feet wide and 5 feet deep, and contains homogeneous calcareous loam covered by common grasses. Experimental apparatus was installed to divert less than a cubic foot per second of runoff from urbanized Arcadia Watershed. Runoff flows by gravity over the lysimeter, where surface inflow, surface outflow and subsurface outflow are measured and sampled. Four trials, each associated with a discrete runoff event, were conducted in the fall of 1971. Water samples were analyzed for inorganic chemical constituents, chemical oxygen demand (COD), coliforms, turbidity and sediment contents. Subsurface-outflow samples from initial trials were high in COD and total dissolved solids, representing soil flushing or leaching. Concentrations of inorganics reached a maximum value within a few hours of initial seepage, and then decreased. The peaking represents a salt build-up between trials. Concentrations of COD, coliforms, turbidity and sediment in subsurface-outflow samples decreased significantly during each trial. Surface-outflow samples had lower turbidity, COD, bacteria and sediment contents than surface-inflow samples. Turbidity, suspended and volatile solids, coliforms and COD in runoff samples may be reduced by grass and soil filtration. Increased grass development and soil settling work to produce a better quality effluent. Quantification of the lysimeter's effectiveness will be useful for urban watershed management.
