A biossÃntese dos lipÃdeos dos cloroplastos pode ser importante para a tolerÃncia ao estresse hÃdrico e para a recuperaÃÃo apÃs reidrataÃÃo. Nesse trabalho nos estudamos o metabolismo dos cloroplastos, monogalactosil-diacilglicerol (MGDG), digalactosil-diacilglicerol (DGDG), sulfoquinovosil-diacilglicerol (SQDG), phosphatidil-glicerol (PG), no Ãmbito do dÃficit hÃdrico e da reidrataÃÃo apÃs o fim do estresse. Com este intuito, nos medimos o teor dos lipÃdeos da folhas, acompanhamos a incorporaÃÃo do precursor 14C-acetato nos lipÃdeos e analisamos a expressÃo dos genes codificadores das enzimas de sÃntese chave dos lipÃdeos (MGD1, MGD2, DGD1, DGD2, SQD2 e PGP1) durante o estresse hÃdrico e apÃs a reidrataÃÃo. Visando de uma melhor compreensÃo da relaÃÃo entre o metabolismo destes lipÃdeos e a tolerÃncia a seca, nos trabalhamos com duas cultivares de Vigna unguiculata L. Walp, uma tolerante (cv. EPACE) e outra sensÃvel (cv. 1183) a seca. Por meio de varredura diferencial de um biblioteca de cDNA de V.unguiculata, foram obtidas as seqÃÃncias completas dos cDNA dos genes VuMGD1, VuMGD2, VuDGD1, VuDGD2, VuSQD2 e VuPGP1. Os resultados mostram que em condiÃÃes de estresse hÃdrico a cultivar tolerante, alÃm de preservar seu teor de lipÃdeos durante a seca, à igualmente capaz de estimular a biossÃntese do DGDG aumentando significativamente a relaÃÃo DGDG:MGDG de suas membranas. NÃs sugerimos que o DGDG acumulado em condiÃÃo de seca à transportado para as membranas externas ao cloroplasto e que isso contribui para a tolerÃncia à seca. Os efeitos da desidrataÃÃo celular sobre as membranas tÃm conseqÃÃncias diretas sobre a capacidade das plantas a se recuperarem apÃs reidrataÃÃo. 48 horas apÃs a rega, a cv. sensÃvel 1183 nÃo à capaz de se recuperar em termos de teor de galactolipÃdeos e incorporaÃÃo do precursor. Na cv. tolerante, no entanto, o teor de DGDG permanece elevado, mesmo apÃs a reidrataÃÃo. Em conclusÃo, nossos resultados sugerem a importÃncia dos lipÃdeos membranares na tolerÃncia/sensibilidade das plantas ao dÃficit hÃdrico, em particular o balanÃo entre as classes lipÃdicas de propriedades fÃsico-quÃmicas diferentes (SQDG versus PG e DGDG versus MGDG) que poderiam afetar a estrutura e o funcionamento das membranas. / Membranes are main targets of degradation by reactive oxygen species and hydrolytic activities induced by drought. Chloroplasts lipid biosynthesis, especially galactolipids monogalactosyl-diacylglycerol (MGDG) and digalactosyl-diacylglycerol (DGDG) are important for plant tolerance to water deficit and for recovery after rehydration. In this thesis, we studied the metabolism of the chloroplast membrane lipids, MGDG, DGDG, sulphoquinovosyl-diacylglycerol (SQDG), phosphatidyl-glycerol (PG) under drought and during recovery from drought. Aiming this, we measured leaf lipids content, followed 14C-acÃtate incorporation and expression of genes coding for chloroplast membrane lipid synthases (MGD1, MGD2, DGD1, DGD2, SQD2 and PGP1) during drought and recovery. In order to better understand the relationship between drought tolerance and lipid metabolism, two cultivars of Vigna unguiculata L. Walp, one drought tolerant (cv. EPACE) the other drought susceptible (cv. 1183) were compared. The cDNA complete sequences for VuMGD1, VuMGD2, VuDGD1, VuDGD2, VuSQD2 and VuPGP1 were obtained from screening of a V.unguiculata cDNA library. The results showed that under water stress conditions, the tolerant cultivar, besides its ability to preserve its lipids pool despites drought, is able to strongly stimulate the DGDG biosynthesis, increasing the DGDG:MGDG ratio in its membranes. We suggest that DGDG accumulated under drought condition, when phosphate is deficient, is exported for extrachloroplastic membranes, and thus contributes to plant drought tolerance. Effects of loss of water on cell membranes have direct consequences on plant capacity to recover from stress. 48 hours after rewatering, the susceptible cv. 1183 was not able to fully recover from a moderate stress in terms of leaf galactolipid content and acetate incorporation into MGDG. In EPACE-1, MGDG leaf content remained unchanged after rehydration and DGDG remained higher than in the control plants. In conclusion, our results highlight the importance of membrane lipids in plant adaptation to water deficit and in their capacity to recover from stress. Of particular importance is the balance between lipid classes with various physico-chemical properties (SQDG versus PG, DGDG versus MGDG), since they most likely have a profound influence on membrane structure and function.

Selektering och urvalsprocessen inom elitidrott, hur upplevs den i praktiken av elever/spelare : en kvalitativ studie inom fotboll och ishockey / Selection process in elite sports, how is it experienced in reality by students/players : a qualitative study in football and ice hockey

Syfte och frågeställning Syftet med studien är att utforska hur elever/spelare resonerar kring sina upplevelser om urval och selekteringsprocessen inom NIU-gymnasium samt elitidrott (fotboll och ishockey) på manliga ungdomssidan från 16 år och uppåt. Syftet är vidare att belysa vilka egenskaper elever/spelare upplever är viktigast vid uttagningsprocessen. Frågeställningar: - Hur upplevs selektering och urvalsprocessen i praktiken av elever/spelare? - I vilken mån upplever elever/spelare att rekryteringsansvariga/skolan tar hänsyn till kriterierna varje NIU-gymnasium ska göra inför urval och selektering? Metod Studien har använt sig av en kvalitativ ansats. Detta då studien avsett att undersöka elever/spelares tankar och åsikter kring urval och selekteringsprocessen. En semistrukturerad intervjuguide har använts vid intervjuerna. Sex intervjuer genomfördes, tre personer verksamma inom fotbollen och tre personer verksamma inom ishockeyn. Deltagarna i studien var mellan 16–20 år gamla och har gått eller går på ett NIU gymnasium samt är aktiva inom sin idrott.  Resultat Vad gäller resultatet framkommer det att kontakter inom idrotten är av stor vikt för att ta sig vidare i selekteringsprocessen, även egenskaper som spelförståelse, drivkraft och inställning är viktigt. När det gäller metoder för uttagningsprocessen så nämns tryouter som en väldigt central del men även scouting förekommer. Flertalet av deltagarna upplever orättvisor vad gäller selekteringsprocessen och kopplar oftast det till kontakterna inom sporten. Sett till användandet av de fem NIU kriterierna är upplevelsen bland majoriteten att man oftast bara tar hänsyn till det första kriteriet, idrottsspecifika färdigheter och förutsättningar. De andra kriterierna hamnar oftast i skymundan och man hoppas kunna lösa dessa under resans gång. Slutsats Slutsatsen vi kan dra utifrån studien är att selekteringsprocessen är ett komplext område som väcker mycket diskussioner. Respondenternas upplevelser varierar angående om det sker en rättvis eller orättvis bedömning vid olika urval. Utifrån respondenternas upplevelser lyder slutsatsen att man tagit sig långt i arbetet med selekteringsprocessen, den kritik som dyker upp är att alla spelare bör få fler chanser och inte selekteras bort redan efter en uttagning. / Aim The purpose of the study was to investigate how students/players reason about their experiences of selection and the selection process within NIU high school and elite sports, football and icehockey on the male youth side from 16 years and up. The purpose was also to illustrate whatattributes that are most important in the selection process. The study is based on the following questions: - How is the selection and selection process experienced in practice by students/players?- In what way do students/players feel that the person in charge of recruitment/the school takes consideration to the criteria that each NIU high school must make before selection? Method The study has used a qualitative method. This is because the study is intended to examine students/players thoughts and opinions about the selection and selection process. A semistructured interviewguide has been used in the interviews. Six interviews were conducted, three people were active in football and three people were active in ice hockey. The participants in the study were between 16–20 years old and have attended or are attending a NIU high school and are active in their sport. Results  It appears that contacts in the sport are of great importance to move forward in the selection process, also attributs such as game understanding, drive and attitude are important. When it comes to methods for the selection process, tryouts are mentioned as a very central part, but scouting also occurs. Most of the participants experience injustices regarding to the selection process and usually link it to the contacts within the sport. In terms of the use of the five NIU criteria, the experience among the majority is that they usually only take consideration into the first criterion, sports-specific skills and conditions. The other criteria usually end up in the shadows and the persons who is responsible hopes to be able to solve these during the journey. Conclusion The selection process is a complex area that raises a lot of discussions. The respondents experiences vary regarding whether there is a fair or unfair assessment in different selections. Based on the respondents experiences, the conclusion is that they have come a long way in the work with the selection process, the criticism that arises is that all players should get more chances and not be selected away already after one selection.

Colour and semantic change : a corpus-based comparison of English green and Polish zielony

The purpose of my research is to investigate the processes and mechanisms of semantic change in two basic colour terms: green in English and zielony in Polish. My research methodology focuses on existing English and Polish corpora, namely the British National Corpus, the Corpus of Contemporary American English and the National Corpus of Polish. I analyze my data both synchronically and diachronically (comparing two periods of time: 1985-1994, 2001-2010). My study also evaluates the use of corpus evidence for the purpose of investigating the processes of semantic change. Various factors have caused the Basic Colour Terms (BCTs) green and zielony to form metaphorical and metonymical meanings that have been conventionalised in English and Polish respectively. These processes have long played an important role in our understanding of the surrounding world. Investigating semantic changes in these two colour terms and two periods of time is key to my cross-cultural research, and this entails answering the questions: Why do green and zielony develop different senses? What are the similarities and differences between these two colour terms? How have these two terms developed and might they develop new senses in future? Are metonymy and metaphor the only mechanisms of semantic change in green and zielony? The semantic change of each colour term is shown through a network of meanings, where all the different meanings of green and zielony are presented together with their stages of development in the form of codes. Additionally each stage is a separate prototype. The aim of the network is to show the etymological prototype and various senses (new prototypes) developing from this original sense. Moreover the number of occurrences of each prototype might indicate which meaning or meanings are most common or even central in a given language at a certain point in time. The network of meanings is a visual representation of semantic change and processes involved in it. A very detailed analysis of corpus examples provides an insight into the uses of green and zielony in English and Polish respectively. The data are analyzed both qualitatively and quantitatively. Such an approach offers a thorough analysis of the two terms in question.

A linguistic ethnographic perspective on Kazakhstan's trinity of languages : language ideologies and identities in a multilingual university community

This thesis presents a linguistic ethnographic study of language ideologies and identities in a multilingual, university community in Kazakhstan: a university aspiring to put Kazakhstan’s ‘Trinity of Languages’ project, aimed at developing societal tri-lingualism in Kazakh, Russian and English, into practice. Data was collected at a Kazakhstani university from 2012 to 2013, combining participant-observation and fieldnotes, audio recordings and interviews. Drawing on the concept of heteroglossia (Bakhtin 1981), the research investigates how young people draw on ideologies of separate and flexible multilingualism (Blackledge and Creese 2010) and on the often contested indexicalities of Kazakh, Russian and English linguistic resources to negotiate identities as multilingual people in Kazakhstan, particularly in contexts of performance, and stance-taking. Consideration of these ideological and linguistic resources also sheds light on Kazakhstan’s wider ‘processes of ideological transformation’ (Smagulova 2008:195) and their real-life implications for multilingual people. Furthermore, the analysis highlights how participants construct stances towards translanguaging (Garcia 2009) and suggests that acts of contextualisation, which frame interactions as being more or less ‘on-stage’ or ‘off-stage’, shape the way that speakers draw on linguistic resources and their indexical meanings, and how these contexts can afford or constrain speaker agency in the negotiation of identities.

Design, techno-economic and environmental risk assessment of aero-derivative industrial gas turbine

Increased availability of natural gas has boosted research and development efforts to further increase gas turbine performance. Performance has been increased remarkably and unit cost reduced due to achievements gained in improving thermodynamic cycles and cooling technologies. However, increased complexity in power industry regulations and fluctuations in fuel price have indicated that all the aforementioned improvements in gas turbine performance could not cope with the increased competition in the gas turbine industrial market. Innovation within the aero-derivative concept has enabled further significant improvement in the performance of industrial gas turbines. It allows a more beneficial approach than developing new designs of industrial gas turbines owing to reduced designing time and cost. Objectives in this project focus on developing a methodology of design and assessing aeroderivative gas turbine engines derived from a 130-seat aircraft engine. Developed methodology includes techno-economic and environmental assessment, conducted through further developments of models based on Techno-economic and Environmental Risk Assessment (TERA) philosophy, to be applied in further industrial applications. Tools used in this investigation include a significant literature research on the development of aero-derivative gas turbine technologies, including thermodynamic cycles and its land-based applications. Turbomatch is a homebased code developed in Cranfield University, used in calculating design point and predicting off-design performance of parent aero-engine and the aeroderivative engines developed. Excel and FORTRAN code are also used in calculating engine’s design parameters, and creating a model of life estimation Creep. Moreover, FORTRAN code is used for building emission and economic models for power generation and combined heat and power applications. Finally, MATLAP code is used in creating a small model for generating performance TXT files, and running marine integrated models platform. All models needed to develop the methodology have been created, and calculations of an engine’s performance and assessment were conducted based on this developed methodology. Sensible results are generated from the investigated methodology and they show acceptable designs of aero-derivative engines on different thermodynamic cycles. Based on the acceptable level of technology and material thermal barriers, all design and off-design performance limitations of new developed aero-derivative engines have been determined for a wide range of ambient conditions. Techno-economic and environmental assessment performed through implementing the developed aero-derivative engines on power generation and marine applications under different operating scenarios. Results of operating the engines on power generation and marine applications have been investigated and compared. It is observed that engines respond differently when operating under different environmental profiles, depending on the number of units engaged and their thermodynamic cycle as well as mechanical configurations. Also, the selected specific gas turbine engine can be the best economical choice for operating on determined scenario, while it cannot be when operating in different scenarios. Assessment of developed engines on the investigated application shows how the lowest specific cost (small engine size) can constitute important criteria in engine selection.

Feasibility of using 100% recycled asphalt pavement mixtures for road construction

Recycled Asphalt Pavement (RAP) is the largest recycled good in the United States and 80 million tons are recycled yearly, saving taxpayers about $1.5 billion dollars. This paper explores the possibility of utilizing 100% RAP materials in asphalt pavement. Asphalt mixtures are produced at 135°C in a typical asphalt plant. However, at 135°C, not all binder from RAP materials may not become effective for coating aggregates. The main objective of the study is to determine the amount of effective binder available from RAP in the asphalt plant. The 100% RAP mixes have aged binder that can alter mix designs and interaction with virgin binder. In this study, to determine low temperature cracking resistance and fatigue performance, samples were prepared using a 100% RAP mix with no virgin binder and a 100% RAP mix with virgin asphalt binder to achieve the optimum binder content of the mix. Second, to determine the effectiveness of binder from RAP materials, compaction tests were performed by heating RAP materials at various temperatures. It was found that 100% RAP mixes cannot be feasible for field use if additional virgin binder is added to reach the optimum asphalt content. Based on limited test results, the low temperature grade was not within proper limits but the beam fatigue testing results were acceptable. Based on compaction test results, additional heating is needed to increase the effectiveness of asphalt binder from RAP materials.

<p>Weight loss and other nutritional problems are common in cancer patients. The problems are of importance for response to treatment and survival and the well-being of the patients.</p><p>Nutritional support can be carried out in different ways. The efforts considered in this thesis are; assessment of nutritional status to find the patients who are at risk to become or already are malnourished, assessment of dietary intake, dietary advice, information and support to the families, information and education to the caregivers, and supplementation with drugs that possibly could influence the weight development. The Swedish version of the Patient Generated Subjective Global assessment of nutritional status, PG-SGA, is useful in assessment of nutritional status in cancer patients. Dietary advice and support to patients and their families combined with information and education to the staff, at the hospital and in the home care, turned out to have a positive influence at the weight development and other parameters related to nutrition. The effects were seen in consecutive patients with small cell lung cancer in comparison with a historical control group, and in patients in a randomised trial. Fish oil and melatonin could stabilise weight development in patients with advanced gastrointestinal cancer, but had no marked influence on factors reflecting cachexia. Problems with nutrition in cancer patients are possible to recognise and various interventions may be beneficial.</p>

Screening Of Tomato Seeds For Genetic Modification And Identification Of Genetically Modified Ripening Delayed Tomato Seeds

Tomato has been genetically modified for providing properties such as insect-resistance or delayed-ripening. Tomato seeds purchased from several bazaars and markets were screened for the presence of genetic modification by targeting NptII kanamycin resistance, Nos terminator, and 35S promoter gene regions which are the most commonly transformed gene regions in transgenic plants, and then ripening-delayed tomato seeds were tried to be identified in this study. F type truncated-PG gene and Sam-k gene were selected as the indicator of genetically modified ripening delayed tomatoes. DNAs of 25 seed samples were isolated by CTAB method and examined with several primer pairs, and the primer sets that provided consistent results were selected to conduct routine testing by PCR analysis of the samples. In screening analysis via conventional PCR, 4 samples were amplified with 35S, Nos and NptII primer sets. Among other samples, 3 of them were amplified with 35S and Nos primer sets and 2 of them were amplified only with 35S primer set. The amplification was observed with Nos, NptII and Sam-k primers in one sample and this sample was identified as 35 1 N, since the sequence result of the PCR product amplified with Sam-k primers showed high homology with the Samase gene of T3 Coliphage. F type truncated PG gene was not observed in any of the samples. Although this study demonstrates the presence of commonly used gene regions in genetically modified tomatoes, further analysis of the genetically modified ripening delayed tomato seeds via construct specificor event specific PCR techniques is needed for confirmation.

Bakhtin, carnival and comic theory

In Rabelais and His World, Mikhail Bakhtin presents us both with a theory of carnival, and with an account of the historical decline of the carnivalesque since the Renaissance. This thesis uses Bakhtin's work as a point of departure for an analysis of particular moments in the history of post-Renaissance comic theory. It is argued both Bakhtin's account of carnivalesque decline provides us with a potent framework within which to perform such an analysis, and that this in turn facilitates a thorough interrogation of, and engagement with, Bakhtin's theory of carnival. Chapter One outlines Bakhtin's theory, identifying its historical and utopian dimensions, and exploring some of the problems which it generates. Chapter Two addresses some of the methodological issues relating to a historical analysis of comic theory, and situates Bakhtin's theory of carnival in relation to recent work in the area of comic theory. The remaining chapters focus on particular comic theory texts in the light of Bakhtin's thesis. Chapter Three contrasts Kant's analysis of humour with Schopenhauer's theory, relating the former to its Enlightenment context and the latter to its Romantic context. Chapter Four explores Bergson's discussion of laughter, situating it in relation to modernism, while Chapter Five reviews Freud's theory of jokes, examining the proximity between the structures of carnival and the structures of the Freudian joke. Chapter Six focuses on a Brechtian theory of comedy, assessing its relationship with the carnivalesque tradition, while Chapter Seven attempts to update Bakhtin's thesis in relation to contemporary configurations by exploring recent arguments concerning the comic credentials of postmodern culture. It is argued in conclusion that, if post-Renaissance culture has witnessed a decline in the significance of the carnivalesque, then the trajectory of that decline has undergone' a complex series of historical shifts and reversals.
