<p>Food allergy is an IgE-mediated immunological disease, which affects almost 4% of the adult population and up to 6% of children. Proteins from milk, egg, peanuts, soybean, wheat, fish and nuts are the main cause of food allergies. A less common allergen is pea protein. The National Food Administration analyses undeclared pea protein and contaminations of pea protein in foods using rocket immunoelectrophoresis and immunodiffusion. For both methods an antiserum against pea protein is needed. The aim of this study has been to characterize a newly developed rabbit-antiserum against pea protein. It is important to know if the antiserum is specific against peas, the detection as well as the quantification limits before it can be taken into use. The results of the study show that the antiserum was not absolutely specific, since it cross-reacted with chickpeas, fenugreek and lenses. However there is an "in-house" established PCR-method that can distinguish between chickpeas, fenugreek and peas and that method can be used as a complement to the rocket immunoelectrophoresis. The PCR-method cannot be used alone because it is not quantitative. Rocket immunoelectro¬phoresis detects 0,003% pea protein with purified IgG-antibodies from the antiserum.</p>

O manejo da solução nutritiva leva em consideração as concentrações e as relações entre os nutrientes para a obtenção de elevada produtividade e qualidade das hortaliças cultivadas em sistemas hidropônicos. A relação adequada entre K e Ca é fundamental para tornar máxima a absorção desses elementos, uma vez que o excesso de K pode reduzir a absorção de Ca e viceversa. Além dos elementos considerados essenciais, os benéficos como o silício pode estimular a absorção de outros nutrientes, dentre eles o cálcio, aumentar a produtividade e a durabilidade pós-colheita dos produtos. Esse trabalho teve por objetivo avaliar a influência das relações K:Ca 0,75:1,0; 1,25:1,0 e 1,5:1,0 e da aplicação de silício (0 e 56 mg L-1) via solução nutritiva, no crescimento, na produtividade, na marcha de absorção de nutrientes aos 7, 14, 21 e 28 dias após o transplante das mudas (DAT), e na da qualidade pós-colheita do coentro Português Pacífico, cultivado em sistema hidropônico NFT. O experimento foi conduzido em blocos ao acaso, em um fatorial 3x2 (três relações K:Ca: 0,75:1,0; 1,25:1,0 e 1,50:1,0 e dois níveis de Si: 0 e 56 mg L-1), em esquema de parcelas subdivididas no tempo, avaliadas em quatro épocas (7, 14, 21 e 28 dias após transplante das mudas DAT). As relações K:Ca de 1,25:1,0 e 1,50:1,0 originaram plantas mais vigorosas quanto à altura, massa fresca e seca da parte aérea e das raízes, área foliar e número de folhas. O Si aumentou a massa fresca da parte aérea e das raízes e o número de folhas aos 28 DAT. A relação K:Ca 0,75:1,0 na presença de Si proporcionou maior número de estômatos na face abaxial da folha. A maior espessura do parênquima lacunoso foi obtida na K:Ca 1,5:1,0. Na presença de Si, a relação K:Ca 1,5:1,0 foi a que gerou maior espessura do parênquima paliçádico e do mesófilo. A ordem de extração tanto na parte aérea como nas raízes foi: K > N > Ca > P > Mg > S. O acúmulo de N, P, K, Ca, Mg, S e Si pela parte aérea representou 85%, 79,9%, 89,2%, 86,4%, 77,8%, 82,3% e 58,1%, respectivamente, em relação ao total. Nos três dias de armazenamento do coentro ocorreu menor perda de massa nos tratamentos que receberam silício em solução nutritiva. / The management of nutrient solution takes into account nutrient concentrations and relationships between nutrients to achieve high productivity and quality of vegetables grown in hydroponic systems. The appropriate ratio between K and Ca is essential for maximum absorption of these elements, once an excessive amount of K may reduce Ca absorption, and vice versa. Besides the elements considered essential, benefic elements such as silicon may not only stimulate absorption of other nutrients, including Ca, but also increase productivity and longevity of postharvest products. This work aimed to study the influence of relations K: Ca 0,75:1,0; 1,25:1,0 and 1.5:1.0 and application of silicon (0 to 56 mg L-1) nutrient solutions on growth, productivity, nutrient uptake at 7, 14, 21 and 28 days after transplanting (DAT), and postharvest quality coriander Português Pacific, grown in hydroponic system NFT. The K: Ca 1,50:1,0 and 1,25:1,0 ratio originated more vigorous plants for plant height, fresh and dry weight of shoots and roots, leaf area and leaf number. Si increased in fresh weight of shoots and roots and number of leaves at 28 DAT. The K: Ca 0,75:1,0 ratio in Si the presence of a greater number of stomata under the leaf. The thick spongy parenchyma was obtained with K: Ca 1.5:1.0. In the presence of Si, the ratio K: Ca 1.5:1.0 was the one that created a greater thickness of the palisade and mesophyll. The order of uptake, both in shoots and roots was: K> N> Ca> P> Mg> S. The accumulation of N, P, K, Ca, Mg, S and Si in shooting accounted for 85%, 79.9%, 89.2%, 86.4%, 77.8%, 82.3% and 58.1%, respectively, compared to the total. In three days of coriander storage less weight loss occurred in treatments with silicon in the nutrient solution.

A busca por embalagens flexíveis rapidamente degradáveis tem sido intensa nos anos mais recentes. Amidos de variadas fontes têm sido avaliados para a produção de filmes, sendo que a maioria destes apresenta boas características mecânicas e de barreira ao oxigênio, entretanto, uma série de limitações é observada com relação às suas características hidrofílicas e permeabilidade ao vapor d´água. O amido de ervilha normalmente apresenta teores mais elevados de amilose, o que pode melhorar a obtenção da rede formadora de filmes. Além disto, as associações amido-goma podem modificar a gelatinização e retrogradação do amido, processos que interferem na formação de filmes. O acréscimo de plastificantes também tem se mostrado de importância para melhorar as propriedades mecânicas dos filmes. Assim, o objetivo do presente trabalho foi avaliar o efeito da adição de goma xantana e de glicerol ao amido de ervilha verde de alto teor de amilose (cv. Utrillo) na elaboração de filmes e em suas características. Soluções filmogênicas (SF) com diferentes teores de amido de ervilha (3, 4 e 5%), de goma xantana (0, 0,05 e 0,1%) e de glicerol (proporção glicerol-amido de 1:5 p/p) foram estudadas. As SF foram obtidas por ebulição (5 min.), seguida de autoclavagem por 1h. a 120°C. Os filmes foram prepa rados segundo a técnica de casting. O aumento da concentração de amido na solução filmogênica aumentou a espessura, fator de ruptura, resistência à perfuração e solubilidade dos filmes em água. O aumento da concentração da goma xantana interferiu apenas nas propriedades associadas à cor dos filmes, aumentando os valores da tonalidade e decrescendo o croma a* e a transparência. O aumento da concentração do glicerol interferiu nas propriedades mecânicas dos filmes, gerando redução dos valores de resistência máxima à tração, tensão na ruptura, resistência à perfuração e fator de ruptura. O plastificante também gerou aumento do alongamento na ruptura, redução da transparência e aumento da espessura e da solubilidade do filme em água. / In the past few years, the attempt to obtain degradable flexible packaging has increased considerably. In this sense, starches obtained from different botanical sources have been evaluated to produce films. Most of these films present good mechanical characteristics and provide a good barrier to oxygen. Nevertheless, some limitations have been observed in relation to their hydrophilic properties and permeability to water vapor. Studies using starch from green peas with high levels of amylose to produce films were not found in the literature. The higher levels of amylose can improve the achievement of the matrix that forms films. In addition, starch-gum associations can modify starch gelatinization and retrogradation, processes which interfere with film formation. The addition of plasticizers has also been considered important to improve the mechanical properties of films. The aim of this study was to evaluate the effect of the addition of xanthan gum and glycerol to the starch of green peas with high content of amylose (cv. Utrillo) in the preparation of films and in their features. Filmogenic solution (FS) with different levels of pea starch (3, 4 and 5%), xanthan gum (0, 0.05 and 0.1%) and glycerol (glycerol-starch ratio of 1:5 p/p) were tested. The FS was obtained by boiling (5 min.), followed by autoclaving for 1h. at 120°C. The films were prepared by casting. The increased concentration of starch in the solution increased the thickness, factor of rupture, puncture force and solubility of films in water. Increasing amounts of xanthan affected only the properties associated with the color of the films, increasing the values of hue angle and decreasing the chroma a* and transparency. The increased concentration of glycerol interfered in the mechanical properties of the films, causing reduction of the maximum values of tensile strength, strain at break, puncture force and factor of rupture. The plasticizer also caused an increase of elongation at break, reduced transparency and increased thickness of the film and the solubility in water.

The objective of this study was to estimate genetic parameters of resistance to coriander and characterization of six cultivars depending on the reaction to M. incognita (races 1 and 3) and M. javanica. The experiment was conducted in a greenhouse on the premises of HortiAgro® Seeds Ltda. In the municipality of Ijaci, located in southern Minas Gerais State, during the period October to December 2011. We evaluated six cultivars of coriander in randomized complete block design with six replications and 16 plants per plot were used inocula of M. incognita race 1, M. incognita race 3 and M. javanica whose eggs were added to the substrate trays in a proportion of 1.200 eggs. ml-1 solution. With the data obtained, we carried out analyzes of variance and the variances were also estimated phenotypic, genetic and environmental, the broad-sense heritability at the level of the cultivar means, the coefficient of genetic variation, the ratio between the coefficients of genetic variation and environmental variation for the characteristics: incidence of galls, number of galls, number of egg mass and egg number. Regarding the characterization of the cultivars were evaluated characteristics: number of egg mass, number of eggs, reproductive factors and index playback. The results indicate that there is genetic variability within and among cultivars and, based on multiplication of nematode, the coriander cultivars showed resistant reaction to M. incognita race 3 and M. javanica. The characteristics, number of egg masses and the number of eggs can be used for the evaluation of higher levels of resistance. The cultivars showed variability within families for resistance to nematode galls, which may be used in breeding programs aimed at developing cultivars resistant to Meloidogyne spp. / A cultura do coentro vem sendo atacada por nematoides. É necessário trabalhos que explorem a variabilidade genética existente para se obter cultivares resistentes a esse fitopatógeno. Portanto o objetivo do trabalho foi estimar os parâmetros genéticos da resistência do coentro, bem como a caracterização de seis cultivares em função da reação a Meloidogyne incognita (raças 1 e 3) e Meloidogyne javanica. O experimento foi conduzido em estufa nas dependências da HortiAgro® Sementes Ltda., no município de Ijací, localizado na região Sul do Estado de Minas Gerais, no período de outubro a dezembro de 2011. Foram avaliadas seis cultivares de coentro no delineamento experimental de blocos casualizados, com seis repetições e 16 plantas por parcela Utilizaram-se inóculos de M. incognita raça1, M. incognita raça 3 e M. javanica cujos ovos foram adicionados ao substrato das bandejas na proporção de 1.200 ovos. ml-1 de solução. Com os dados obtidos, realizaram-se as análises de variância e também foram estimadas as variâncias fenotípica, genética e ambiental; a herdabilidade no sentido amplo ao nível das médias das cultivares; o coeficiente de variação genética; razão entre os coeficientes de variação genética e variação ambiental para as características: incidência de galhas, número de galhas, número de massa de ovos e número de ovos. Com relação à caracterização das cultivares foram avaliadas as características: número de massa de ovos, número de ovos, fator de reprodução e índice de reprodução. Os resultados indicam que existe variabilidade genética entre e dentro das cultivares e, baseado na multiplicação do nematoide, as cultivares de coentro apresentaram reação de resistência para M. incognita raça 3 e M. javanica. As características, número de massas de ovos e número de ovos podem ser utilizadas para avaliações de níveis superiores de resistência. As cultivares apresentaram variabilidade dentro das famílias para resistência ao nematoide das galhas, podendo estas serem utilizadas em programas de melhoramento que visem o desenvolvimento de cultivares resistentes à Meloidogyne ssp.

<p>This investigation addresses how photosynthetic efficiency is affected when pea (Pisum sativum) plants are restricted to a sole nitrogen source (i.e. ammonium or nitrate). The pea plants were watered with different nutrient solutions without NO3- or NH4+ for different time-periods in order to assay for nitrogen content. The soluble ammonium and nitrate content was measured throughout the entire growth period. No major differences were observed in nitrogen content during the starvation period up to 25 days. For technical reasons, cultivation of plants could not be extended beyond this time. The chloroplasts and thylakoids were isolated after 25 days and assayed for chlorophyll contents and photosynthetic activity.</p><p>The outcome of these tests indicates a small but unambiguous decrease in the photosynthesis activity for all treatments, relative the control.</p>

<p>The reproductive health of individuals is severely compromised by HIV infection, with candidiasis being the most prevalent oral complication in patients. Although not usually associated with severe morbidity, oropharyngeal candidiasis can be clinically significant, as it can interfere with the administration of medications and adequate nutritional intake, and may spread to the esophagus. Azole antifungal agents are commonly prescribed for the treatment and prophylaxis of candidal infections. However, the emergence of drug resistant strains and dose limiting toxic effects have complicated the treatment of candidiasis. Consequently, safe and effective and affordable medicine is required to combat this fungus. Commercial garlic (Allium sativum) has been used time since immemorial as a natural antibiotic, however very little is known about the antifungal properties of two indigenous South African species of garlic, namely Tulbaghia alliacea and Tulbaghia violacea, that are used as folk medicines for a variety of infections. This study compares the in vitro anti-candidal activity of Tulbaghia alliacea, Tulbaghia violacea and Allium sativum extracts. It was found that the greatest concentrations of inhibitory components were extracted by chloroform or water. The IC50 concentrations of Tulbaghia alliacea were between 0.007 &ndash / 0.038% (w/v). Assays using S. cerevisiae revealed that the T. alliacea extract was fungicidal, with a killing half-life of approximately 2 hours. This inhibitory effect of the T. alliacea extracts was observed via TLC, and may be due to an active compound called Marasmicin, that was identified using NMR. This investigation confirms that extracts of T.alliacea exhibit anti-infective activity against candida species in vitro.</p>

Root Border Cell Development and Functions of Extracellular Proteins and DNA in Fungal Resistance at the Root TipWen, Fushi January 2009 (has links)
Soilborne plant pathogens are responsible for many of the major crop diseases worldwide. However, plant root tips are generally resistant to pathogen infections. The goal of this dissertation research is to understand the mechanism of this natural resistance by testing the hypothesis that root caps and root border cells control the rhizosphere community through the biological products which they deliver to the soil. Specific objectives of this dissertation project are 1) identifying, isolating, and characterizing the genes important for border cell development and for root exudates delivery, and 2) analyzing the function of extracellular macromolecules in root exudates in root tip-fungal pathogen interaction. The expression of a primary cell wall synthesis gene, PsFut1, encoding Pisum sativum fucosyltransferase, was characterized during border cell production, and the impact of silencing this gene on border cell development was examined. Another gene, BRDgal1, encoding β-galactosidase, was identified and characterized in Pisum sativum during this study. It was shown that this β-galactosidase is specifically produced in and secreted from root border cells. The microarray transcriptional profiling in M. truncatula and mRNA differential display analysis in pea plants were carried out following the induction of border cell production to gain a broader understanding of the genes which potentially influence border cell development. In order to study the commonality of border cell production across different plant species, the expression of rcpme1, the marker gene for border cell production, was compared between the garden pea and a gymnosperm species, the Norway spruce (Picea abies). To accomplish the second objective, the focus of this study was shifted from border cell development to mucilaginous root exudates excreted by border cells and root cap cells. This resulted in a breakthrough in the understanding of the mechanisms of root tip resistance. The presence of extracellular DNA in the root mucilage was discovered and its requirement for root tip resistance to fungal infection was demonstrated. Extracellular proteins in the root mucilage were identified and they were shown to be also required for the root tip resistance to fungal infection. This work provided new insights into understanding plant defense mechanisms.
