Tuberculosis, caused by the intracellular pathogen Mycobacterium tuberculosis, is one of the most prevalent infectious diseases in our world today. In order to survive within the host the bacteria need to sense and respond to changes in the environment; however, signal transduction in this bacterium is poorly understood. PknB is a serine/threonine kinase essential for the in vitro survival of M. tuberculosis and therefore a potential drug target against the bacteria. It is the goal of the current study to elucidate downstream substrates of PknB. We have found that PknB shares in vitro substrates with another serine/threonine kinase, PknH, implying the potential complexity of the signaling pathways in the bacteria. We have also provided the first description of the coupling between serine/threonine kinases PknB and PknH with a two-component system response regulator DevR, and further proposed Ser/Thr phosphorylation as the negative regulator of DevR transcription activator activity based on LC-MS/MS analysis. Finally, we have identified a previously unknown phosphoprotein glyceraldehyde 3-phosphate dehydrogenase encoded by the ORF Rv1436, which demonstrates autophosphorylation activity and which phosphorylation is independent of PknB. The increased use of computer networks has lead to the adoption of Internet-based solutions for reducing telephony costs. This has proved to be a boon to callers who can reach the other party directly via the Internet. Unfortunately numerous business persons still need to call to and from mobile phones which are currently a domain where the customers are generally tightly bound to their operators. To provide a simple solution to this problem for companies, Opticall AB has designed an integrated system called the Dial over Data solution, coupling a mobile interface with a low-rate communication channel, which allows calls to be originated remotely at the best price, exploiting the customer company's existing network. This scheme allows the customer company to easily control telecommunications costs, to monitor their employees' efficiency, and more generally speaking to claim a central role in the communications of their employees. The proposed solution allows distant callers (usually employees of the customer company) to benefit from the company's internal network, which is usually more cost effective and offering connectivity to more networks than a cell phone. The Dial over Data solution enables communication between any phone accessible from the customer company's telephony network (such as SIP clients, landline phones, and mobile phones) at a lower cost.</p> This thesis project analyzes existing technologies and compares them to the pre-existing prototype to ascertain the validity of the method and of the components used. This project also explains the improvements brought to the features offered by the DoD solution: the initial prototype has been developed into a stable and functional product, and has been tested internally. Prompted by a need for scalability and additional features, the replacement of Asterisk for the handling of SIP calls by other SIP servers has also been considered and tested. / Den nuvarande ökningen av datanätverk har lett till adoptionen av Internetbaserade lösningar för att förminskar kostnader inom telefoni. Tyvärr behöver åtskilliga affärsmän fortfarande ringa till och ifrån mobiler som återstår som ett område där kunderna är fastkedjade till deras operatörer. För att tillföra en enkel lösning till detta problem för kundföretag har Opticall AB planlagt ett integrerat system som kallas Dial over Data som kopplar ihop ett mobilt gränssnitt, med en billig kommunikationsmedel, som tillåter telefonsamtal påbörjas avlägset på det billigaste priset tack vare företagets nätverk. Det ger möjligheten till kundföretaget att vara centralt för sina personals kommunikationer. Det medger ett enkelt sätt att kontrollera kostnader samt övervaka personalens effektivitet. Den Dial over Data lösningen är en lösning som tillåter avlägsen besökarna med kundföretagets inre nätverk kommer att dra nytta av eftersom det är mer kostnadseffektiv och flexibel än en blott mobiltelefon. Konnektiviteten till företagets inre nätverk samt en låg besvärlighetsnivå är garanterade respektive genom konfigurationsförmågan av produkten och ett praktiskt gränssnitt som är redo för korporativkontaktlistor och visar alla informationen som är relevanta till förbrukarnas erfarenhet.</p> Det här avhandlingsprojektet analyserar existerande teknologier och sätter dem i relation till den sedan tidigare framtagna prototypen för att utröna validiteten hos metoden och beståndsdelar. Projektet förklarar även de förbättringar som gjorts till de egenskaper som erbjuds av DOD-lösningen: prototypen har utvecklats till en stabil och funktionell produkt och har testats internt. [pt] O presente trabalho visa apresentar as possíveis relações e implicações entre os atributos do Ser parmenídico e os pressupostos da Teoria das Ideias platônicas, relações e implicações exploradas, particularmente, por meio do exame das aporias da participação dos entes sensíveis nas Ideias realizado no diálogo Parmênides. Para apresentar essas possíveis implicações, torna-se necessário fazer uma apresentação das teses sobre o Ser, apresentadas por Parmênides em seu poema, Sobre a natureza, analisando, sobretudo, o que se reconhece como Via da verdade, mais precisamente, naquele que se reconhece como o fragmento 8 desse poema. Logo em seguida é necessário examinar passagens de diálogos consideradas, por alguns comentadores, como contendo traços do desenvolvimento do que é comumente admitida como a Teoria das Ideias. Assim, passamos, brevemente, por determinados diálogos da fase inicial, bem como alguns diálogos da fase de maturidade, detendo-nos, mais detalhadamente, aos diálogos médios como Fédon e Banquete, nos quais essa teoria é estabelecida em seu formato mais tradicional. Buscamos ainda, examinar a terminologia associada à Ideia nesses diálogos. Para que, ao final, se estabeleça uma relação entre a crítica do diálogo Parmênides com as teses presentes no poema de Parmênides. / [en] This thesis aims to present the possible relationships and implications between the attributes of the Parmenidean Being and important assumptions of Plato s Theory of Ideas. I will explore relationships and implications particularly through the examination of the aporias of participation of sensitive beings in the Ideas, such as carried out in the dialogue Parmenides. To undertake this line of arguments, it is necessary to tackle the theses about Being, presented by Parmenides in his poem, On Nature, analyzing. Above all, it is necessary to investigate what is known as the Way of Truth, specially the fragment 8 of Parmenides poem. Next, I will examine relevant passages of Plato s dialogues, namely those considered relevant for the development of the principal tenets of Plato s Theory of Ideas. Thus, I will briefly go through certain dialogues from Plato s initial phase, as well as some mature dialogues, focusing in more detail on the medium dialogues such as Phaedo and Banquet, in which this theory is established in its more traditional format. I will also seek to examine the terminology associated with the Idea in these dialogues, so that, in the end, a relationship is established between the criticism of the Parmenides dialogue with the theses presented in Parmenides poem.

Drought is a major environmental stress factor that poses a serious threat to food security. The effects of drought on early reproductive tissue at 1-2 DAP (days after pollination) is irreversible in nature and leads to embryo abortion, directly affecting the grain yield production. We developed a working RNA-Seq pipeline to study maize (Zea mays) drought transcriptome sequenced by Illumina GSIIx technology to compare drought treated and well- watered fertilized ovary (1-2DAP) and basal leaf meristem tissue. The pipeline also identified novel splice junctions - splice variants of previously known gene models and potential novel transcription units. An attempt was also made to exploit the data to understand the drought mediated transcriptional events (e.g. alternative splicing). Gene Ontology (GO) enrichment analysis revealed massive down-regulation of cell division and cell cycle genes in the drought stressed ovary only. Among GO categories related to carbohydrate metabolism, changes in starch and sucrose metabolism-related genes occurred in the ovary, consistent with a decrease in starch levels, and in sucrose transporter function, with no comparable changes occurring in the leaf meristem. ABA-related processes responded positively, but only in the ovaries. GO enrichment analysis also suggested differential responses to drought between the two tissues in categories such as oxidative stress-related and cell cycle events. The data are discussed in the context of the susceptibility of maize kernel to drought stress leading to embryo abortion, and the relative robustness of actively dividing vegetative tissue taken at the same time from the same plant subjected to the same conditions. A hypothesis is formulated, proposing drought-mediated intersecting effects on the expression of invertase genes, glucose signaling (hexokinase 1-dependent and independent), ABA-dependent and independent signaling, antioxidant responses, PCD, phospholipase C effects, and cell cycle related processes. [pt] Esta pesquisa tem como objetivo compreender o papel privilegiado da angústia na existência a partira da obra ser e tempo da Martin Heidegger. O foco deste estudo esta justamente no privilégio deste papel para aquele que pode se angustiar, ou seja, o ser-aí humano. Tal prerrogativaestá apoiada na possibilidade de uam transformação existencial. Uma vez que a angústia suspende as determinações a partir das quais nós somos e estamos. Esta suspensão abre espaço para que outors modos de ser aconteçam, ou seja, para que outra relação com o espaço existencial possa surgir. Em meio ao automatismo cotidiano pouco espaço se dá para que algo diverso apareça. Na verdade pouco espaço se dá para o próprio espaço no qual a existência se constitui. Esta abertura faz surgir justamente quando nós angustiamos. Isso significa que a angústia traz um espaço, mas não garante que algo diverso surja daí, e modo que, o automatismo sempre pode ser retomado. O papel privilegiado almejado nesta pesquisa dá margem para pensarmos a possibilidade de duas vozes de angústia: a primeira esta relacionada a essa quebra de automatismos, e a segunda apronta para uma prontidão ao angustiar-se. É exatmente a partir da escuta destas das vozes que ocorre uma trasnformação existencial, ou seja, uma apreensão e transparência com relação aos modos de ser mais própios. / [en] This research aims to comprehend the privileged role of angst in the existential analytics based on Martin Heidegger’s work Being and Time. The focus of this study lies precisely on the privilege of this role for those who can be anguished, ie, the human Dasein. This prerogative is supported by the possibility of an existential transformation. Once the anguish suspends determinations that we are. This suspension makes room/space for other ways of being, which means that another relation with the existential space may arise. In the average everydayness there is a narrow space for something different appears. In fact, in this quotidian way of being a narrow space is given to the space where the existence itself is constituted. This disclosure will come just when we are anguished. This means that the angst brings a space, but does not guarantee that something different can emerge from there, which means that automatism can always be retaken. The privileged role pursued in this research raises from the possibility of two anguish’s voices: the first one is related to break of the automatism, while the second one points to an acceptance of angst as an existential possibility. It is just when this two voices are listened that can occurs an existential transformation, which means an apprehension and a transparency related to our authentic way of being.

The theme of this research is Integral Education and the conceptions of the teachers on Integral Education and on the “Projeto Ser Pleno in 2014. Highlighting propositions from authors like Cavaliere, Coelho, Libâneo, Moll, Paro, Limonta, among others, and concerning the analysis and understanding of the theme, the research problem was formulated: what do the teachers of the schools of the “Projeto Ser Pleno” think about Integral Education and, in particular, about “Being Full”. The research aims were: to identify the “Projeto Ser Pleno Goiânia/GO teachers conception of Integral Education and “Being Full”; to outline the historic route of the Integral Education framing in the Brazilian context; establish the collation between teacher’s perspective about Integral Education and the conception of “Being Full”. To investigate the problem, a qualitative research was carried out involving bibliographical and field research. The review covered the period between 2010 and 2016 focusing on relevant works about the studied subject. The field research involved four public primary schools from the “Projeto Ser Pleno”. The data collection included interviews and documentary analysis. For the documentary analysis, it was tried to raise in the educational legislation the main materials that regulate the integral education, analyzing documents like the Federal Constitution of 1988, the Law of Directives and Bases of the National Education of 1996, the National Plan of Education (2014-2024), the “To be full Project” and other national documents, besides the Pedagogical Political Project of the schools in which the research was carried out. The data analyses were based on the Bogdan & Biklen (1994) guidelines for data categorization and systematization, outlining the following categories: Integral Education, “To be full” and Expectations regarding schools future. The results reveal the teacher’s understanding about the “Projeto Ser Pleno” and Integral Education Project, namely, the state's lack of responsibility for public education, even though the student's stay in school produces the apparent perception that the State is increasing their performance; emphasis on the issue of the social risk of children and adolescents, so that the social assistance function is incorporated into the responsibilities of the school institution; distinction between integral school and integral education, based on the idea that it is not enough to offer a full-time school, but it is necessary to offer an integral education that goes beyond the school environment. The research reveals the need to understand full-time school as a right to a school education that promotes intellectual development / O presente estudo insere-se na linha de pesquisa Teorias Educacionais e Processos Pedagógicos, do Programa de Pós-Graduação em Educação da Pontifícia Universidade Católica de Goiás (PUC). Tem como objeto o Projeto experimental de educação em tempo integral Ser Pleno elaborado pela Secretaria de Estado de Educação, Cultura e Esporte (SEDUCE) em parceria com o Instituo Jaime Câmara, implantado em dez escolas da rede de ensino estadual do Estado de Goiás a partir de 2014. O problema que se buscou esclarecer foi quais as concepções dos professores das escolas que participaram do Projeto Ser Pleno sobre educação integral e, em particular, sobre o Projeto Ser Pleno e o conceito de Ser Pleno? Privilegiando as proposições dos autores Cavaliere, Coelho, Libâneo, Moll, Paro, Limonta, entre outros, a pesquisa. Teve como objetivos: identificar a concepção de Educação Integral e Ser Pleno dos professores do Projeto Ser Pleno – Goiânia/Go; delinear o percurso histórico da concepção de educação integral no contexto brasileiro; estabelecer o cotejamento entre a visão dos professores sobre educação integral e a concepção de ser pleno. Para investigar o problema realizou-se uma pesquisa qualitativa envolvendo pesquisa bibliográfica e pesquisa de campo. A pesquisa bibliográfica abrangeu o período de 2010 a 2016 com foco em obras pertinentes ao tema estudado. A pesquisa de campo envolveu quatro escolas públicas da Educação Básica do Projeto Ser Pleno – Goiânia/Go vinculadas à Secretaria Estadual de Educação e teve como sujeitos os professores do Projeto Ser Pleno. A coleta de dados incluiu entrevistas e análise documental. Para a análise documental, buscou-se levantar na legislação educacional os principais materiais que regulamentam a educação integral, analisando-se documentos como a Constituição Federal de 1988, a Lei de Diretrizes e Bases da Educação Nacional de 1996, o Plano Nacional de Educação (2014-2024), o Projeto Ser Pleno e outros documentos nacionais, além do Projeto Político Pedagógico das escolas em que se desenvolveu a pesquisa. Na análise dos dados foram adotadas as orientações de Bogdan & Biklen (1994) para a categorização e sistematização dos dados, delineandose às seguintes categorias: Educação Integral, Ser Pleno e Expectativas quanto ao futuro das escolas. Os resultados obtidos revelam a compreensão dos professores acerca do Projeto Ser Pleno e de Educação Integral, quais sejam, a desresponsabilização do Estado pela educação pública, ainda que a permanência do aluno na escola produza a aparente percepção de que o Estado está aumentando sua atuação, ênfase na questão do risco social de crianças e adolescentes, de modo que a função de assistência social passa a ser incorporada às responsabilidades da instituição escolar, distinção entre escola integral e educação integral, com base na ideia de que não basta oferecer uma escola de tempo integral, é preciso ofertar uma educação integral, que extrapola o ambiente escolar. Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / The attendant work advances as a central goal the analysis of the problem of logical principles applicability to the normative ambit, according to the conceptions of the Austrian philosopher of Law, Hans Kelsen. This problem doesn t receive the same approach by the author along his several works. Thus, the problem will be in this work inquired by distinguishing three phases in its treatment by Kelsen. The arguments that correspond to the three phases lie, respectively, in the works General Theory of Law and State (1945), the second edition of the Pure Theory of Law (1960) and the General Theory of Norms (1979), the main study objects in this work. Each chapter will hence approach the treatment of one of these phases, in order to, along them, analyse not only the main theses and concepts of Kelsen about the relation between Logic and Law, but also the motivations and purposes that leaded the author to leave old theses in behalf of new ones. It s intended that such approach will be able to elucidate the already traditional problem about the possibility of a relation between the principles of Logic mainly the principle of non-contradiction and the rule of inference and Moral and Legal norms. In order to achieve that, through the presentation and analysis of each one of Kelsen s postures about the problem, is searched a systematic regard of the different arguments given for its treatment, which represent a growing refuse of Kelsen for the logical elements insertion in the normative field. Nevertheless, until end up refusing this application, Kelsen s inquiry crosses over less extreme approaches, in which are noticed the limitations of some concepts and theses inside his theory, that will be, some of them, modified or abandoned, in behalf of one ultimate approach, severely commited to the positivists and purists ideals maintained by the author. / O presente trabalho tem por objetivo analisar o problema da aplicabilidade de princípios lógicos ao âmbito normativo segundo a concepção do jusfilósofo austríaco Hans Kelsen. Este problema não é abordado da mesma forma pelo autor em suas diversas obras. Portanto, o problema será investigado nesta dissertação distinguindo-se três fases do seu tratamento por Kelsen. Os argumentos que correspondem às três fases encontram-se, respectivamente, nas obras Teoria Geral do Direito e do Estado (1945), na segunda edição da Teoria Pura do Direito (1960) e na Teoria Geral das Normas (1979), os principais objetos de estudo deste trabalho. Cada um dos capítulos da dissertação irá deter-se, deste modo, ao tratamento de uma destas fases, de modo que, ao longo delas, serão analisados não somente as teses e conceitos kelsenianos sobre a relação da Lógica com o Direito, mas também as motivações e propósitos que levaram o autor a abandonar teses antigas em favor de novas teses. Pretende-se que tal abordagem elucide o já tradicional problema da possibilidade de relação entre os princípios da Lógica especialmente o princípio da não-contradição e a regra de inferência às normas do Direito e da Moral. Para isso, através da presente apresentação e análise de cada uma das posições de Kelsen, busca-se uma sistematização dos diferentes argumentos fornecidos para o seu tratamento, que representam uma crescente recusa pela inserção de elementos da Lógica no plano normativo. No entanto, até culminar em uma recusa final, a investigação de Kelsen passa por posições menos extremadas, nas quais se notam as limitações de certos conceitos e teses dentro de sua Teoria, que serão, alguns, modificados ou abandonados, em favor de uma última abordagem rigidamente comprometida com os ideais positivistas e puristas sustentados pelo autor.

FundaÃÃo Cearense de Apoio ao Desenvolvimento Cientifico e TecnolÃgico / Essa dissertaÃÃo pretende investigar como o Quem dera ser um peixe (QDSP) â grupo que se constituiu com o objetivo de se colocar contra a construÃÃo de um oceanÃrio milionÃrio na orla de Fortaleza â utilizou as redes sociais em sua prÃtica comunicativa, a fim de mobilizar os usuÃrios da internet em torno da questÃo do Acquario CearÃ, instituindo, no percurso, novos processos de resistÃncia e novos paradigmas de aÃÃo coletiva em nosso estado. Para tanto, estabelecemos como corpus da pesquisa a fan page âQuem dera ser um peixeâ e o perfil âPeixuxa Acquarioâ, ambas do site Facebook â a plataforma mais frequentada pelo Quem dera ser um peixe. Escolhemos limitar nossa pesquisa a 2012, perÃodo de maior incidÃncia do grupo na internet, ano que contou com um grande nÃmero de simpatizantes produzindo conteÃdo e auxiliando a amplificar as denÃncias contra a obra. A metodologia usada para abordar essa questÃo, alÃm da descriÃÃo da prÃtica comunicativa do Quem dera ser um peixe, tanto no espaÃo virtual, como no espaÃo real, foi a AnÃlise de MobilizaÃÃo de quadros, a qual busca compreender como se dà o alinhamento entre o enquadramento de uma instituiÃÃo ou movimento social, por exemplo, e os atores sociais que a compÃe, elemento essencial para se entender o processo de mobilizaÃÃo polÃtica. AlÃm disso, recorremos a quatro entrevistas com diferentes membros do Quem dera ser um peixe, a fim de compreender suas posiÃÃes acerca de questÃes centrais para o grupo, como o papel da informaÃÃo e da comunicaÃÃo para o tipo de ativismo que pratica; e tambÃm de questÃes ligadas a sua forma de organizaÃÃo. Esse aspecto, em particular, à de grande importÃncia e procuramos abordÃ-lo no trabalho, embora sem a pretensÃo de esgotar o assunto, por suas implicaÃÃes na prÃpria prÃtica comunicativa do grupo. Partindo de um pequeno histÃrico da internet, delineamos como se deu sua passagem de tecnologia militar para suporte material da sociedade em rede, enfocando os usos que o QDSP fazia e faz dela. O acompanhamento de suas atividades fora do espaÃo virtual, em reuniÃes, aÃÃes, articulaÃÃes foi de grande importÃncia para dar materialidade aos dados coletados das plataformas de interaÃÃo, fornecendo mais elementos capazes de dar conta da complexidade do fenÃmeno que representa o QDSP. Um aspecto que salta aos olhos, por exemplo, o qual sà poderia ter sido observado por meio da ida a campo diz respeito à complexa relaÃÃo estabelecida entre os integrantes do grupo e o PoÃo da Draga, comunidade popular que se localiza a poucos metros do Acquario. Em conclusÃo, podemos afirmar que o QDSP foi bem sucedido na conduÃÃo de um ativismo virtual (o qual era subsidiado por extensas investigaÃÃes e pelo uso inventivo e dinÃmico da comunicaÃÃo, borrando os limites entre ciberespaÃo e espaÃo real) que conseguiu nÃo sà barrar por diversas vezes a obra, como tambÃm ampliar e popularizar muitas questÃes ligadas ao oceanÃrio, a ponto de, hoje em dia, o projeto estar longe de ser uma unanimidade entre a populaÃÃo de Fortaleza.

The purpose of this thesis is to make a study of the third and fourth series of the Episodios nacionales. The study will deal with the plot structure, the characterization, and the historicity of the twenty novels which make up the two series.
