Cross-Talk Between Estrogen and Thyroid Hormones During Amphibian Development

Duarte Guterman, Paula January 2011 (has links)
It is generally thought that in amphibians, thyroid hormones (THs) regulate metamorphosis, while sex steroids (estrogens and androgens) regulate gonadal differentiation. However, inhibition of TH synthesis in frogs alters gonadal differentiation, suggesting instead that these two endocrine axes interact during development. Specifically, THs may be involved in male development, while estrogens may inhibit tadpole metamorphosis. However, we do not currently know the mechanisms that account for these interactions, let alone how such mechanisms may differ between species. To develop and test new hypotheses on the roles of sex steroids and THs, I first examined transcriptional profiles (mRNA) of enzymes and receptors related to sex steroids and THs during embryogenesis and metamorphosis in Silurana tropicalis. Tadpoles were exposed to either an estrogen synthesis inhibitor (fadrozole) or TH (triiodothyronine, T3) during early larval or tadpole development. Acute exposures of S. tropicalis to fadrozole or T3 during early development resulted in increased expression of androgen- and TH-related genes in whole body larvae, while chronic exposure to fadrozole during metamorphosis affected gonadal differentiation but did not affect tadpole development. On the other hand, acute exposure to T3 during metamorphosis increased the expression of androgen-related transcripts both in the brain and gonad. In S. tropicalis, the results suggested that cross-talk is primarily in one direction (i.e., effect of THs on the reproductive axis) with a strong relationship between TH and androgen status. Lastly, I established developmental transcript profiles and investigated T3 regulation of brain and gonad transcripts in Engystomops pustulosus. I then compared these results with S. tropicalis and an earlier study in Lithobates pipiens. While each species developed with similar profiles, they differed in their response to T3. Exposure to T3 resulted in either an increase in androgen-related genes (S. tropicalis) or a decrease in estrogen-related genes (E. pustulosus and L. pipiens). In conclusion, these data demonstrated that cross-talk mechanisms differ among these three evolutionary separate species, but in all cases, T3 appears to affect the balance of sex steroids, stimulating the androgen system and providing potential mechanisms of the masculinising effects of THs. These results will contribute to understanding the mechanisms of hormone interactions and their evolutionary basis in frogs.

To affirm or inhibit children's sexual development : Preschool teacher's experience and approach to children's sexuality

Karlsson, Jessica January 2021 (has links)
The importance of children being treated in a way that conveys the message that their sexuality is something positive and enjoyable has been emphasized by the WHO. The aim of the study was therefore to investigate how experienced preschool teachers experience and respond to children's sexuality. Focus group discussions were used as a method of gathering teachers' knowledge and experience. The results showed that the interviewed preschool teachers agreed that children's sexuality is something natural and important to consider. According to the informants, further training within the subject was also unusual and discussions with colleagues and parents were usually conducted only in the event of incidents. The importance of confirming and responding to children's sexuality in a positive way was analyzed and it was discussed whether children's sexuality is distracted too often and if the focus on negative sexuality prevails. One conclusion was that children's sexuality should be normalized so that adults' fears do not harm children's sexual development.

Análise do gene MAMLD1(CXorf6) em pacientes com distúrbios do desenvolvimento sexual 46,XY de origem indeterminada / Analysis of the MAMLD1 (CXorf6) gene in patients with undetermined 46,XY disorders of sexual development

Brandão, Maíra Pontual 30 August 2011 (has links)
O papel desta variante protéica do MAMDL1 na etiologia DDS 46,XY deve ser melhor determinada através de estudos in vivo / Introduction: MAMLD1 has been shown to be implicated in the etiology of penoscrotal hypospadias. To date, 3 loss-of-function nonsense mutations have been identified in Japanese patients with hypospadias. Objective: To screen MAMLD1 for mutations in a large cohort of Brazilian patients with undetermined 46,XY DSD. Patients and Methods: We evaluated 50 patients in which defects of testosterone synthesis, 5 alfa-reductase 2 deficiency and partial androgen insensitivity were previously excluded. The entire coding region and the flanking splicing sites of MAMLD1 were amplified and sequenced from genomic DNA. Six microsatellite markers flanking the gene were used to analyze founder effect of new allelic variant. Transactivation function of MAMLD1 was analyzed by a luciferase assays. COS-1 cells seeded in 12-well dishes were transiently transfected with luciferase reporter vector (p-Hes1, p-Hes3 and p-Hes5), expression vector for MAMLD1 (WT and allelic variant) and pRL-CMV vector as an internal control. All experiments were performed in triplicates and repeated 3 times. Results: We identified an allelic variant, the p.H347Q on exon 3 in 4 unrelated patients (3 sporadic and 1 familial cases). Two patients underwent gonadectomy in infancy and were raised as girls. Pelvic ultrasound showed a uterus in one of them. The other two male patients had micropenis, cryptorchidism and perineal hypospadias. This allelic variant was found in 1 out of 250 Brazilian controls frequency 0,4%. The transactivation activities of the variant protein were 2.0 folds higher than the WT with p-Hes3. No founder effect was demonstrated in these families. Discussion: The allelic variant is located in a highly conserved region of MAMDL1 which is essential for male genitalia development. HES is a family of transcriptional repressors of Notch signaling. Hes gene expression pattern is tissue and cell specific and control the timing of biological events. In gonads, their expression pattern remained to be elucidated. In somitogenesis, Hirata showed that both loss of expression or persistent expression of Hes genes result in the same defects. Therefore, overexpression of Hes genes should impair the normal male differentiation by continuous suppression of downstream genes involved in male sex development. Conclusion: This is the first report of an allelic variant of MAMLD1 determining a gain of function in its protein in patients with 46,XY DSD. The role of this variant protein of MAMDL1 in the etiology of 46, XY DDS should be better determined by in vivo studies

Novas perspectivas no estudo genético do hipogonadismo hipogonadotrófico isolado (HHI) por meio da técnica de sequenciamento paralelo em larga escala / New perspectives in the genetic study of congenital isolated hypogonadotrophic hypogonadism (IHH) using targeted next-generation sequencing

Amato, Lorena Guimarães Lima 03 August 2018 (has links)
Congenital IHH is more prevalent in men and about 50% to 60% of affected individuals present with associated anosmia or hyposmia, characterizing Kallmann\'s syndrome. Several genes have already been associated with the pathogenesis of congenital IHH, but most cases still remain without a molecular diagnosis. Until recently, identification of the genetic causes of IHH was performed by sequencing candidate genes using the Sanger technique. However, with the growing number of genes and the genetic complexity of IHH, it has become almost impossible to keep the screening of all candidate genes updated using the traditional techniques. The advent of next-generation sequencing (NGS) has allowed the simultaneous genotyping of several regions, faster and with lower relative cost. The present project was developed with the objective of tracking candidate genes in patients with congenital IHH using large-scale parallel sequencing, in aiming to increase the genetic knowledge of this rare condition. A total of 130 unrelated patients with IHH was studied by targeted NGS, using a panel containing 36 IHH associated genes. Initially, 104 potentially pathogenic variants were identified in 77 patients (59.2%). However, after an individualized analysis of each variant, the number of patients considered to carry pathogenic or probably pathogenic variants dropped to 41 (31.5%). The genes KAL1, FGFR1, CHD7 and GNRHR were the most frequently affected and these results confirm the importance of genes classically associated with IHH. It is noteworthy the high prevalence of variants in CHD7 (10.8%), a rather extensive gene, leading to technical difficulty of sequencing by traditional methods, which had not been studied in this cohort. CHD7 is the causative gene of CHARGE syndrome, however it has been recently identified in a growing number of congenital IHH patients with or without additional features of the syndrome. Among the results, we emphasize a novel mutation in the GNRH1 gene, a rare cause of IHH, and the identification of deleterious variants in the IGSF10 gene, recently associated with pubertal delay but without a clear role in the IHH phenotype, in two patients with reversible hypogonadism. Probably pathogenic variants in genes with few descriptions or even no reports of association with the IHH phenotype (SPRY4, FLRT3, IGSF1, NSMF, SOX10 and OTX2) were also identified in this cohort, increasing the genetic knowledge of IHH. Oligogenicity, previously described with a prevalence of 2.5% to 7%, was observed in 22% of our patients, demonstrating an increase in oligogenicity cases when compared to previous studies using only the Sanger sequencing. In conclusion, targeted NGS was able to increase the percentage of patients with molecular diagnosis from 22% to 31.5% in our cohort when compared to the previous data using the Sanger sequencing, and has been shown to be a fast, reliable and effective tool in the molecular diagnosis of congenital IHH

Management of children with sexual behaviour problems, between the ages of five to nine years old, by educators and social workers

Philander, Rochelle January 2018 (has links)
Magister Artium - MA (Child and Family Studies) / The Management of children, younger than twelve years of age, who pose a risk to other children, remains complex and confusing. When their behaviour includes sexual aggression towards other children, ignorance about how to manage them becomes even more challenging. Society has an expectation that any sexual aggressor should be punished, however, when the aggressor is younger than ten years old, different responses are necessary. The aim of this current study was to explore the management of learners with sexual behaviour problems, within the primary school setting. The main question underpinning this study was: How are children, aged five-to-nine-years, with sexual behaviour problems, managed by social workers and educators? A qualitative methodology, with focus group discussions and semi-structured interviews were used to conduct this study. Educators from primary schools, as well as social workers from the Western Cape Education Department, were purposively selected to form the sample for this study. Focus group discussions were conducted with the educators, while semi-structured interviews were conducted with the social workers.

Conclusion: The accelerated sexual maturation was associated with changes in bioelectrical parameters, body composition, greater increase in BMI and obesity in children and adolescents
