Membrane Properties Involved in Calcium-Stimulated Microparticle Release from the Plasma Membranes of S49 Lymphoma Cells

The mechanism of microparticle shedding from the plasma membrane of calcium-loaded cells has been investigated in erythrocytes and platelets. Recent studies have revealed the physiological and clinical importance of microparticle release from nucleated cells such as lymphocytes and endothelium. The experiments of this study were designed to address whether simple mechanisms discovered in platelets and erythrocytes also apply to the more complex nucleated cells. Four such mechanisms were addressed: potassium efflux, transbilayer phosphatidylserine migration, cytoskeleton degradation, and membrane lipid order. The rate and amount of microparticle release in the presence of a calcium ionophore, ionomycin, was assayed by light scatter at 500 nm. To inhibit the calcium-activated potassium current, cells were exposed to 1 mM quinine or a high-potassium buffer. Both interventions substantially attenuated microparticle shedding induced by ionomycin. Microparticle release was also greatly reduced in a lymphocyte cell line deficient in the expression of scramblase, the enzyme responsible for calcium-stimulated phosphatidylserine migration to the cell surface. This result indicated that such phosphatidylserine exposure is also required for microparticle shedding. The importance of cytoskeletal rearrangement was evaluated through the use of E64-d, a calpain inhibitor, which appeared to have no affect on release. Thus, if cytoskeleton degradation is important for microparticle release, a different enzyme or protein must be involved. Finally, the effect of membrane physical properties was addressed by varying the experimental temperature (32–42 °C). A significant positive trend in the rate of microparticle release as a function of temperature was observed. Fluorescence experiments with trimethylammoniumdiphenylhexatriene and patman revealed significant differences in the level of apparent membrane order along that temperature range. Ionomycin treatment appeared to cause further disordering of the membrane, although the magnitude of this change was minimally temperature-sensitive. Thus, it was concluded that microparticle release depends more on the initial level of membrane order than on the change imposed by calcium uptake. In general, mechanisms involved in particle release from platelets and erythrocytes appeared relevant tolymphocytes with the exception of the hydrolytic enzyme involved in cytoskeletal degradation.

Laserablation mit induktiv gekoppelter Plasma-Massenspektrometrie für die medizinische Diagnostik

In dieser Arbeit wurde eine neue Markierungsstrategie von Antikörpern mit dem Markierungsreagenz MeCAT (Metal Coded Tag) unter physiologischen Reaktionsbedingungen, sowie deren Anwendung in einem 8-fach Multiplex-Immunoassay von in Formalin-fixierten und in Paraffin-eingebetteten Gewebeschnitten entwickelt. Für eine aussagekräftige LA-ICP-MS Detektion von MeCAT-modifizierten Antikörpern, wurde eine Standardisierung für biologische Proben auf NC-Membranen, basierend auf einer homogenen Aufbringung eines internen Standards und Kalibrierstandards durch einen kommerziell verfügbaren Tintenstrahldrucker entwickelt und mit der ICP-MS Analyse von Lösungen evaluiert. Die LA-ICP-MS wurde in zwei 8-fach Multiplex-Immunoassays von Tissue Micro Arrays vom Prostatakarzinom und in Maushirngewebeschnitten zur Einschätzung von neurogenerative Erkrankungen erfolgreich eingesetzt werden. Es konnte hierbei gezeigt werden, dass das Nachweisvermögen, der hier entwickelten Methode bereits ausreicht, um die gängigen klinischen Biomarker mit guter Ortsauflösung nachzuweisen. / In this work a new tagging strategy of antibodies with the tagging reagent MeCAT (Metal Coded Tag) was developed under physiological reaction conditions. Their application was proved in an 8-fold multiplex immunoassay of formalin-fixed and paraffin-embedded tissue sections. For a significant LA-ICP-MS detection of MeCAT tagged antibodies standardization for biological samples owere developed. The standardization based on a homogeneous deposition onto the NC membrane via conventional CD-ink-jet printer was validated in addition with the ICP-MS analysis of solutions. The internal standardization of LA-ICP-MS was successfully applied in two 8-fold multiplex immunoassays for Tissue Micro Arrays (TMA) of prostate cancer and for detection of biomarkers for neurodegenerative diseases in mouse brain tissue sections. In both examples it could be shown that the detection capability of the new tagging strategy in combination with the printing standardization allows the detection of the clinical biomarker with good spatial resolutions.

The purpose of this research was to investigate the influence of height, crosssection and metal alloy in the mechanical act of teardrop loops used in incisors retraction and in en masse retraction of incisors and canines, aiming analyze the mechanical behavior of these loops in relation to optimum forces presented in the literature. Two hundred and forty loops were submitted to a tensile load on the testing machine considered heights of 6mm, 8mm and 10mm; cross-sections of 0.018in.x 0.025in and 0.019in.x 0.025in and wire materials of stainless steel and beta-titanium alloys. It was obtained magnitudes of horizontal force (N) and load-deflection (N/mm) for each loop across activations of 0.5mm, 1mm, 1.5mm, 2mm and 2.5mm. The results showed that height (p<0.01), cross-section and metal alloy (p<0.05) were considered significant factors with reference to horizontal force and load-deflection. There was no influence of one factor but a combination of these trying to achieve very light forces. This study suggest that is necessary a large orthodontic biomechanics knowledge with respect to loops design used in incisors retraction and in en masse retraction of incisors and canines, selecting adequate height, crosssection and metal alloy. / O objetivo desta pesquisa foi investigar os efeitos da altura, secção transversal e tipo de liga no desempenho mecânico de alças confeccionadas em forma de gota, indicadas para a retração de incisivos e para a retração em massa de incisivos e caninos, buscando avaliar o comportamento das mesmas em relação às magnitudes de força ótima existentes na literatura. Foram submetidas a um ensaio mecânico 240 alças, considerando as alturas de 6mm, 8mm e 10mm; as seções transversais de 0.018 x 0.025 e de 0.019 x 0.025 e as ligas de aço inoxidável e beta-titânio (TMA). Foram obtidos valores de força horizontal (N) e carga-deflexão (N/mm) para cada alça mediante ativações de 0.5mm, 1mm, 1.5mm, 2mm e 2.5mm. Os resultados mostraram que a altura (p<0.01), a secção transversal e a liga (p<0.05) foram consideradas fatores significativos na força horizontal e cargadeflexão gerada. Não houve influência de um fator sobre os demais, mas sim, uma combinação destes, buscando atingir forças muito leves. Este estudo sugere que é necessário amplo conhecimento da biomecânica ortodôntica na confecção de alças direcionadas para a retração dos dentes anteriores, selecionando adequadamente a altura, a secção transversal e a liga.

Ab-initio studies of reactions to functionalize carbon nanotubes

Since the rediscovery of carbon nanotubes (CNTs) due to the publication of Sumio Iijima's article Helical microtubules of graphitic carbon in the magazine Nature in 1991 the interest in carbon nanotubes has rapidly increased. This bachelor thesis also deals with this popular material with the aim to functionalize CNTs for further uses in the microelectronic industry. A promising approach is the functionalization of the CNTs with metal nanoparticles or metal films. To achieve this, one can perform an atomic layer deposition (ALD) on CNTs. In the present work the Trimethylaluminum (TMA) ALD is the chosen process for the functionalization of the CNTs, which will be studied here. Since the available knowledge on the CNT-functionalization by gas phase reactions is very limited, a theoretical study of possible reaction pathways is necessary. Those studies are carried out with two modern quantumchemical programs, Turbomole and DMol³, which are described together with an introduction into Density Functional Theory, as well as an introduction of CNTs and the ALD process. A basic model of a CNT with a Single Vacancy defect, which had been selected according to the demands of the studies, is introduced. Because the TMA ALD process requires hydroxyl groups as its starting point, not only is the performance of a TMA ALD cycle on a CNT studied, but also reactions which result in the CNTs owning of hydroxyl groups. Consequently, this bachelor thesis will focus on two di erent aspects: The performance of one TMA ALD cycle and the study of possible educts for the TMA ALD process. Tense, mood, and aspect expressions in Nafsan (South Efate) from a typological perspective

In dieser Arbeit untersuche ich aus einer typologischen Perspektive die Bedeutung von Tempus, Modalität und Aspekt (TMA) in Nafsan (South Efate), einer ozeanischen Sprache Vanuatus. Ich konzentriere mich auf die Bedeutung des perfektiven Aspekts und der Realis/Irrealis-Modalität in Nafsan und anderen ozeanischen Sprachen, als Fallstudien zur Untersuchung der sprach-übergreifenden Merkmale dieser TMA-Kategorien. Um ihre Bedeutungen in Nafsan zu analysieren, untersuche ich die Grammatik von Nafsan (Thieberger, 2006) und den Korpus von Thieberger (1995–2018), gefolgt von meiner Feldarbeit (Krajinovic, 2017b). Meine Analysen zeigen, dass Perfekt in Nafsan alle Funktionen hat, die für das Perfekt im Englischen typisch sind, mit Ausnahme der zusätzlichen Bedeutung von Zustandsänderungen. Die Verwendung des Nafsan-Perfekts liefert einen Beitrag zu der Debatte über die sprachübergreifende Gültigkeit von Iamitive, definiert durch die Bedeutung von Zustandsänderungen (Olsson, 2013). Basierend auf den Daten aus Nafsan und anderen ozeanischen Sprachen zeige ich, dass die von Klein (1994) vorgeschlagene semantische Definition des Perfekts ausreichend ist, um zusätzliche Funktionen des Perfekts zu berücksichtigen, ohne eine neue Iamitive-Kategorie zu etablieren. Was die Unterscheidung zwischen Realis und Irrealis betrifft, so habe ich festgestellt, dass die Kategorie Realis in Nafsan semantisch unterbewertet ist, wie sie in Irrealis-Kontexten auftreten kann, die mit der Bedeutung von Realis unvereinbar sein sollten. Ich schlage vor, dass “Realis” gelegentlich Realis-Bedeutungen durch pragmatischen Wettbewerb mit Irrealis erhaltet. Indem ich das “branching-times’’ Modell annehme, das den Ausdruck von Modalität und zeitlichem Bezug vereint (Prince, 2018), zeige ich, dass Nafsan und mehrere andere ozeanische Sprachen Beweise dafür liefern, dass Irrealis als Modalitätskategorie, die sich auf nicht-aktuelle Welten bezieht, eine semantisch sinnvolle Kategorie ist. / In this thesis I study the meaning of tense, mood, and aspect (TMA) expressions in Nafsan (South Efate), an Oceanic language of Vanuatu, from a typological perspective. I focus on the meanings of the perfect aspect and realis/irrealis mood in Nafsan and other Oceanic languages, as case studies for investigating the cross-linguistic features of these TMA categories, frequently disputed in the literature. In order to analyze their meanings in Nafsan, I studied the Nafsan grammar (Thieberger, 2006) and corpus by Thieberger (1995–2018), followed by storyboard and questionnaire elicitation in my fieldwork (Krajinovic, 2017b). I found that the Nafsan perfect has all the functions considered to be typical of the English-style perfect, except for the additional meaning of change of state. I place the analysis of the Nafsan perfect in the debate about the cross-linguistic validity of the newly proposed category of iamitives, defined by the meaning of change of state akin to `already' and lacking experiential and universal perfect functions (Olsson, 2013). Based on the data from Nafsan and other Oceanic languages, I show that, when language-internal processes are considered, the semantic definition of perfect proposed by Klein (1994) is sufficient to account for additional perfect functions, without the need to posit the new iamitive category. Regarding the realis/irrealis distinction, I have found that the “realis” category is semantically underspecified in Nafsan, as it can occur in irrealis contexts that should be incompatible with realis meanings. I propose that “realis” in Nafsan only occasionally receives realis meanings through pragmatic competition with the irrealis category. By adopting a branching-times model that unites the expression of modality and temporal reference (Prince, 2018), I also show that Nafsan and several other Oceanic languages provide evidence that irrealis as a mood category referring to non-actual worlds is a semantically meaningful category.

Quantum Chemical Feasibility Study of Methylamines as Nitrogen Precursors in Chemical Vapor Deposition

The possibility of using methylamines instead of ammonia as a nitrogen precursor for the CVD of nitrides is studied using quantum chemical computations of reaction energies: reaction electronic energy (Δ𝑟𝐸𝑒𝑙𝑒𝑐) reaction enthalpy (Δ𝑟𝐻) and reaction free energy (Δ𝑟𝐺). The reaction energies were calculated for three types of reactions: Uni- and bimolecular decomposition to more reactive nitrogen species, adduct forming with trimethylgallium (TMG) and trimethylaluminum (TMA) followed by a release of methane or ethane and surface adsorption to gallium nitride for both the unreacted ammonia or methylamines or the decomposition products. The calculations for the reaction entropy and free energy were made at both STP and CVD conditions (300°C-1300°C and 50 mbar). The ab inito Gaussian 4 (G4) theory were used for the calculations of the decomposition and adduct reactions while the surface adsorptions were calculated using the Density Functional Theory method B3LYP. From the reactions energies it can be concluded that the decomposition was facilitated by the increasing number of methyl groups on the nitrogen. The adducts with mono- and dimethylamine were more favorable than ammonia and trimethylamine. 𝑁𝐻2 was found to be most readily to adsorb to 𝐺𝑎𝑁 while the undecomposed ammonia and methylamines was not willingly to adsorb.

Dissection moléculaire de la sénescence cellulaire induite par le stress et la thérapie dans le cancer de l’ovaire et son impact sur la réponse des patientes

Le cancer de l’ovaire (COv) est le cancer gynécologique le plus létal chez la femme et les traitements existants, chirurgie et chimiothérapie, ont peu évolué au cours des dernières décennies. Nous proposons que la compréhension des différents destins cellulaires tels que la sénescence que peuvent choisir les cellules du cancer de l’ovaire en réponse à la chimiothérapie pourrait conduire à de nouvelles opportunités thérapeutiques. La sénescence cellulaire a été largement associée à l’activité de la protéine TP53, qui est mutée dans plus de 90% des cas de cancer de l’ovaire séreux de haut grade (COv-SHG), la forme la plus commune de la maladie. Dans nos travaux, à partir d’échantillons dérivés de patientes, nous montrons que les cultures primaires du cancer de l’ovaire séreux de haut grade exposées au stress ou à des drogues utilisées en chimiothérapie entrent en senescence grâce à l’activité d’un isoforme du gène CDKN2A (p16INK4A). Dans ces cellules, nous avons évalué les caractéristiques fondamentales de la sénescence cellulaire tels que les altérations morphologiques, l’activité béta galactosidase associée à la sénescence, les dommages à l’ADN, l’arrêt du cycle cellulaire et le phénotype sécrétoire associé à la sénescence. En utilisant des micromatrices tissulaires construites à partir d’échantillons humains de COv-SHG pré- et post-chimiothérapie, accompagnées de leurs données cliniques, nous avons quantifié des marqueurs de sénescence incluant une diminution de la prolifération cellulaire quelques semaines après chimiothérapie. De façon intéressante, l’expression de p16INK4A dans les échantillons de COv-SHG prétraitement corrèle avec une survie prolongée des patientes suite au traitement. Ceci suggère ainsi pour la première fois un impact biologique bénéfique pour la présence de cellules cancéreuses qui sont capable d’activer la sénescence, particulièrement pour le traitement du cancer de l’ovaire. Dans le but de complémenter les thérapies actuelles avec des approches de manipulation pharmacologique de la sénescence, nos résultats suggèrent qu’il serait important de déterminer l’impact positif ou négatif de la sénescence induite par la thérapie sur la progression de la maladie et la survie, pour chaque type de cancer de façon indépendante. / Human ovarian cancer (OvCa) is the deadliest gynecologic malignancy and existing surgical/chemotherapeutic treatment options have been relatively static for decades. We propose that understanding OvCa cell fate decisions taken in response to chemotherapy could guide new therapeutic opportunities. Damage-induced cellular senescence is often associated with TP53 activity, which is heavily mutated in high grade serous (HGS) OvCa (>90%), the most common form of this disease. Here, using patient derived tissues, we show that primary HGS-OvCa cultures predominantly trigger CDKN2A- associated (p16INK4A isoform) senescence following exposure to stress or chemotherapy. Key senescence hallmarks including altered morphology, senescence-associated-Betagalactosidase, DNA damage, cell cycle arrest and the senescence-associated secretory phenotype were evaluated and detected in damaged cells. Using tissue microarrays built from pre- and post-treatment human HGS-OvC tissue samples with accompanying clinical data, we quantified post-treatment hallmarks of senescence including reduced cell proliferation weeks after chemotherapy. Importantly, p16INK4A expression in pretreatment HGS-OvC samples correlated with increased patients survival, suggesting for the first time that senescence-competence in human cancer cells may have a beneficial impact on treatment outcomes for patients. In order to guide the potential improvement of existing human therapies via pharmacological senescence manipulation, our results suggests that it is important to determine for many types of human cancer whether treatment-induced senescence positively or negatively impacts disease progression and patient survival.
