A serological survey to determine the prevalence of Brucella Canis infection in dogs within the Nelson Mandela Bay metropolitan in the Eastern Cape, South Africa

The prevalence of Brucella canis in South Africa is unknown and suspected to be under-detected. The majority of dogs in South Africa are not tested for Brucella canis, not only because of the level of awareness of Brucella canis in South Africa, but also because of the lack of clinical suspicion. It is not known how the infection entered South Africa. Brucella canis, a zoonotic organism that causes canine brucellosis in dogs, is a significant cause of reproductive failure in dogs worldwide. Canine brucellosis is a chronic infectious zoonotic disease whose main etiological agent, the Brucella canis bacterium, are rough, intracellular proteobacteria in the Brucellaceae family. Clinical signs in bitches are mainly infertility and abortion, while in males, epididymitis and orchitis occur. However, discospondylitis may develop in both sexes. A serological survey was conducted to determine the prevalence of Brucella canis infection in dogs from the Nelson Mandela Bay Metropolitan (NMBM) Port Elizabeth (PE) area. A total of 400 samples were collected, 350 of which were collected in seven different townships and 50 were collected in the three different welfare organisations in the study area. Of the 400 serum samples collected, 39 (9.75%) tested serologically positive by using the Tube Agglutination Test (TAT), the 2-Mercaptoethanol-TAT (2ME-TAT) or the Compliment Fixation Test (CFT). The results of the CFT showed that nine of the 39 positive samples had a maximum antibody titre of 784 IU/ml. The prevalence rate varied tremendously between the samples from the townships and those from the welfare organisations. The prevalence rate of seropositive animals in PE ranged between 5% and 16% in the study area. No positive cases were found in KwaMagxaki and the Animal Welfare Society of PE, but both were surrounded by areas that had positive cases of Brucella canis infection. The female dogs in the study area had a higher sero-prevalence of only 0.0169 (95% CI, 0.0631 to 0.1489) differences in proportion, and were thus not significant (p > 0.05). However, the female spayed dogs had a much higher significant difference of 0.1898 (95% iv CI, 0.1058 to 0.2738) in proportion to the male neutered dogs and were thus statistically significant (p < 0.05). Despite those results, out of all the dogs positive for Brucella canis only two were neutered males and five were spayed females, and the rest were all intact. In conclusion, according to the results, Brucella canis antibodies were detected in sera of dogs mostly from the townships surveyed. Preventive measures against this contagion should be taken into consideration to eliminate Brucella canis infection from the entire dog population. Reservoir dogs and actively infected dogs either should be kept in quarantine or should be euthanized, because not only can they spread the disease and end the reproductive life of any breeding animal, but they are also a risk to human health. Even though this is the first survey conducted in the Eastern Cape, the results are still high dogs in the study area had a higher sero-prevalence of only 0.0169 (95% CI, 0.0631 to 0.1489) differences in proportion, and were thus not significant (p > 0.05). La Guerre Civile libanaise (1975-1990) fut notamment marquée par l’enlèvement de militaires mais aussi de civils. Depuis la loi d’Amnistie de 1991, nous examinerons précisément les répercussions sur la santé psychique de familles d’enlevés, grâce à des entretiens non directifs et la collecte de récits poétiques comme épistolaires, selon la lecture du TAT. La justice transitionnelle est une prospective de reconstruction du Liban. Elle permettrait de lever le voile d’ignorance sur le sort des disparus, pour faciliter le processus de deuil. Mais pour l’heure, les familles souffrent du silence de l’Etat qui enterre l’histoire du Liban, ceci tant que le sort des disparus demeurera inidentifiable. En dépit de ce silence qui annihile toute entreprise de séparation psychique entre les familles et les disparus, condamnant ainsi les mères libanaises à la répétition infinie du trauma, à l’instar de Pénélope tissant et détissant sans cesse les liens du linceul de Télémaque, celles-ci peuvent réaccoucher d’elles-mêmes ; et se tourner, en pleine conscience, vers un avenir, une destinée propre, via le processus du « deuil paradoxal ». Ce concept, loin d’abonder dans le sens de l’évitement ou du déni des disparus, consiste en une réapparition du moi des mères, dans l’opération psychosomatique de procéder au deuil, non de leurs proches, mais du traumatisme qui les avait de prime abord anéanties, jusqu’au vide dépressif. C’est, paradoxalement, grâce à leur fonctionnement limite que les mères des disparus libanais vont pouvoir opérer ce « retour » à leur moi qu’elles imaginaient perdu. / The Lebanese Civil War (1975 - 1990) has been mainly remarkable as far as the kidnapping of soldiers as much as of civilians is concerned. Up to the 1991 Amnesty Law, we will precisely examine the consequential effects on psychic health on rapted families thanks to non guiding interwiews, and the gathering poetical or written accounts, according to the reading of TAT.The transitional justice is a prospective for the rebuild of Lebanon. It could help to clear the mist about the missing's fate, to help people to go out of mourning. But at the present time, the families are enduring the silence of the State, that is burying and forgetting the history of Lebanon, that the missing's fate will remain unestablished for ever. In spite of this silence that destroys any attempt of psychic breaking up between the families and the missings, blocking up that way the Lebanese mothers in the perpetual repetition of the trauma, like Penelope doing and undoing her work that consisted in weawing the shroud of Telemaque's father- in - law, they can deliver of herselves; and decide to look at a future, an own destiny, through the process of "paradoxal nourning".This concept,far from avoiding or denying the missings, consists in a new coming out of the mother's ego, in the psychosomatic way to initiate mourning not of the next of kin, but the traumatism that had prostrated them first up to the depressive emptyness. Paradoxically, bringing the mothers of Lebanese missings to their extreme limits will next allow them to get back to their ego that they imagined as lost for ever at first.

An exploration of the intrapsychic development and personality structure of serial killers through the use of psychometric testing

The mystery surrounding serial homicide is the apparent lack of motive for the murder. No extrinsic motive such as robbery, financial gain, passion or revenge exists, as there usually is in the case of other murders. Serial homicide is a serious, worldwide problem that has received a large amount of media attention, but only a relatively small amount of scientifically based research exists on this phenomenon. Since the 1970s various models such as the psychosocial theory model, learning theory, the motivational, fantasy, neurological theory, psychiatric, post-modern, feminist and the paranormal/demonological models were used to explain the phenomenon of serial homicide. The researcher, however, states that these models do not satisfactorily address the intrapsychic/object relation development of the serial killers personality. The structure of the research project consists of analysis of the background information of two serial killers which was gathered from one semi-structured personal interview, psychometric testing (Thematic Apperception Test, Test of Object Relations and the Picture Test of Separation and Individuation) as well as from relevant literary sources. The information was interpreted using the selected psychoanalytic theory of Sigmund Freund and the object relation theory of Melanie Klein and the data was then analysed, interpreted and tested against the following research questions: “What is the intrapsychic origin of serial homicide?”; “What is the parent-child relationship like?”; “How does the parent-child relationship influence the object relations of the serial killer?”; “How does the parental relationship influence the serial killer’s interpersonal relationships?”. These questions determine the specific internalised factors that may have contributed to the eventual development of a serial killer’s personality. The research method that is employed is a qualitative, exploratory case study method. A qualitative study was selected due to the fact that there are currently not enough incarcerated serial killers in South Africa and from those incarcerated serial killers only two were willing to participate in the research. The case study method was selected because it deals with contemporary events, multiple data sources may be used and the findings can be generalised to other case studies. Data gathering was done by psychometric testing (TAT, PTSI and TOR), a semi-structured interview and other biographical information on the subjects. The data was analysed by the descriptive-dialogic case study method. The data integration method that was selected is the data integration method of the descriptive-dialogic case study method. The data was discussed in relation to the already developed theories. This implies that parallels were drawn between the processed information and the theories. HIV-1 Gene Expression: Transcriptional Regulation and RNA Interference Studies: a Dissertation

Gene expression of human immunodeficiency virus type-1 (HIV-1), which causes Acquired Immunodeficiency Syndrome (AIDS), is regulated at the transcriptional level, where negative factors can block elongation that is overcome by HIV Tat protein and P-TEFb. P-TEFb, a positive elongation transcription factor with two subunits, CDK9 and Cyclin T1 (CycT1), catalyzes Tat-dependent phosphorylation of Ser-5 in the Pol II C-terminal domain (CTD), allowing production of longer mRNAs. Ser-5 phosphorylation enables the CTD to recruit mammalian mRNA capping enzyme (Mce1) and stimulate its guanylyltransferase activity. This dissertation demonstrates that stable binding of Mce1 and cap methyltransferase to template-engaged Pol II depends on CTD phosphorylation, but not on nascent RNA. Capping and methylation doesn't occur until nascent pre-mRNA become 19-22 nucleotides long. A second and novel pathway for recruiting and activating Mce1 involved direct physical interaction between the CTD, Tat and Mce1. Tat stimulated the guanylyltransferase and triphosphatase activities of Mce1, thereby enhancing the otherwise low efficiency of cotranscriptional capping of HIV mRNA. These findings imply that multiple mechanisms exist for coupling transcription elongation and mRNA processing at a checkpoint critical to HIV gene expression. To elucidate P-TEFb's function in human (HeLa) cells, RNA interference (RNAi) was used to degrade mRNA for hCycT1 or CDK9. Down-regulation of P-TEFb expression by RNAi can be achieved without causing major toxic or lethal effects and can control Tat transactivation and HIV replication in host cells. High-density oligonucleotide arrays were used to determine the effect of P-TEFb knockdown on global gene expression. Of 44,928 human genes analyzed, 25 were down-regulated and known or likely to be involved in cell proliferation and differentiation. These results provide new insight into P-TEFb function, its potent role in early embryonic development and strong evidence that P-TEFb is a new target for developing AIDS and cancer therapies. To fulfill the promise of RNAi for treating infectious and human genetic diseases, structural and functional mechanisms underlying RNAi in human cells were studied. The status of the 5' hydroxyl terminus of the antisense strand of short interfering RNA (siRNA) duplexes determined RNAi activity, while a 3' terminus block was tolerated in vivo. A perfect A-form helix in siRNA was not required for RNAi, but was required for antisense-target RNA duplexes. Strikingly, crosslinking siRNA duplexes with psoralen did not completely block RNAi, indicating that complete unwinding of the siRNA helix is not necessary for RNAi in vivo. These results suggest that RNA amplification by RNA-dependent RNA polymerase is not essential for RNAi in human cells.

In oligopoly prerequisites are likely to prevail inducing a 'normal' behaviour in accordance with profit maximization. This leads to a price combination located at an exactly definable line section. Finally some consequences upon economic policy are outlined. / Dieser Beitrag versucht zu zeigen, daß das Oligopolproblem wesentlich determinierter ist als gemeinhin angenommen. Im Oligopol liegen die Voraussetzungen besonders günstig, daß durch ein aus dem Streben nach Gewinnmaximierung abgeleitetes, "normales" Verhalten eine Preiskombination auf einem exakt abgrenzbaren Kurvenabschnitt zustande kommt. Abschließend werden wirtschaftspolitische Konsequenzen angedeutet.

Expression of human α-N-Acetylglucosaminidase in Sf9 insect cells: effect of cryptic splice site removal and native secretion-signaling peptide addition.

Human α-N-Acetylglucosaminidase (Naglu) is a lysosomal acid hydrolase implicated in tthe rare metabolic storage disorder known as mucopolysaccharidosis type IIIB (MPS IIIB; also Sanfilippo syndrome B). Absence of this enzyme results in cytotoxic accumulation of heparan sulphate in the central nervous system, causing mental retardation and a shortened lifespan. Enzyme replacement therapy is not currently effective to treat neurological symptoms due to the inability of exogenous Naglu to access the brain. This laboratory uses a Spodoptera frugiperda (Sf9) insect cell system to express Naglu fused to a synthetic protein transduction domain with the intent to facilitate delivery of Naglu across the blood-brain barrier. The project described herein may be broken down into three main sections. Firstly, the impact of two cryptic splice sites on Naglu expression levels was analyzed in both transiently expressing Sf9 cultures and stably selected cell lines. Secondly, the effectiveness of the native Naglu secretion-signaling peptide in the Sf9 system was examined. Finally, purification of a Naglu fusion protein from suspension culture medium was performed using hydrophobic interaction chromatographic techniques. The ultimate goal of this research is to develop an efficient system for economical, large-scale production of a human recombinant Naglu fusion protein that has the potential to be successfully used for enzyme replacement therapy to treat MPS IIIB. / Graduate
