Organoteluretos na preparação de substâncias bioativas / Organotellurides in the preparation of Bioactive compounds

O presente trabalho teve como foco principal, a preparação one pot de compostos orgânicos alquílicos e vinílicos contendo o grupamento butiltelurenila em sua estrutura, e o estudo do comportamento desses em variados tipos de reações. Uma delas foi a reação de troca Te/Li. Assim, teluretos foram utilizados como equivalentes sintéticos de vários reagentes organometálicos. Os teluretos foram submetidos à reação de troca Te/Li, e os respectivos compostos organolítio gerados, transformados em reagentes organometálicos de zinco, cobre e cério através de reações de transmetalação. Esses reagentes gerados in situ foram submetidos à reação com variados eletrófilos. Os reagentes organometálicos de zinco e cobre gerados foram submetidos à reação de acilação frente a cloreto de ácidos, resultando em 1,4-hidroxicetonas. Um exemplo dessas hidroxicetonas foi obtido em sua forma enantiopura utilizando um telureto quiral, e esse utilizado na síntese formal e enantiosseletiva da (-)-Pirenoforina. Diferentes estequiometrias de cupratos e outros reagentes organometálicos oriundos de um telureto também foram preparados, e esses testados em reações de substituição de tosilatos e abertura de epóxidos. Os substratos gerados dessas reações foram empregados na síntese das moléculas bioativas Endo-Brevicomina e Frontalina. Apresentaremos também, o resultado obtido na reação direta entre uma espécie di-litiada quiral na presença de benzonitrila. A hidroxicetona assim produzida é um análogo do Ipomeanol, um composto com atividade contra câncer de pulmão. Será apresentado também, o comportamento de entidades di-aniônicas contendo grupamentos silila em reações catalisadas por CeCl3, em que a captura desses com eletrófilos como aldeídos, forneceram 1,4-enedióis em bons rendimentos químicos e alta diastereosseletividade. Será apresentado também o estudo do comportamento de teluretos com hibridização sp2 em reações pericíclicas. A reação pericíclica estudada compreende compostos contendo telúrio (II) e (IV) em reações de Diels-Alder. / One of the main purposes of this work was to develop a new direct methodology to prepare functionalized organic tellurides, to be submitted to a series of different reactions, as for example the Te/Li exchange reaction, aimed to afford synthetic equivalents of several organometallic reagents. Zinc, copper and cerium organometallic compounds were generated via a transmetalation reaction of the lithium species, and were reacted with several electrophiles. As a result, 1,4-hydroxyketones could be successfully prepared. According to this methodology, an enantioenriched 1,4-hydroxyketone could be obtained, using a chiral telluride as starting material. This chiral nonracemic intermediate was employed in an enantioselective formal synthesis of (-)-Pyrenophorin. Cuprates and other organometallic reagents, prepared from a specific telluride and using the above described methodology, were the choosen nucleophiles for performing some aliphatic substitution reactions and, in particular, for the ring opening of epoxides. The resulting products were employed as building blocks for the synthesis of the bioactive cyclic compounds Endo-Brevicomin and Frontalin. The direct reaction of the chiral dilithiated specie with benzonitrile afforded a 1,4-hydroxyketone as an analogue of Ipomeanol, a bioactive compound for cancer therapy. Bis-anionic sililated species, prepared from a telluride, were submitted to reaction with a series of aldehydes. Such reactions, in which CeCl3 was employed as catalyst, showed to be highly diastereoselectives, affording (E)-1,4-enediols in good yields. The pericyclic reaction of insaturated tellurides was also investigated. It is worth mentioning that the same Diels-Alder reaction could be performed either with tellurium II or tellurium IV species.

Dentre esses materiais encontram-se titanosillenitas com diferentes dopantes e os materiais fotorrefrativos do grupo II-VI como o telureto de c'admio (CdTe) e telureto de zinco (ZnTe), também dopados, e cujo interesse está no fato de terem uma banda proibida menor que as sillenitas, e serem bem mais rápidos que as sillenitas. O modelo matemático que descreve o efeito foto-fem com um padrão de "speckle" oscilante e de grande amplitude foi desenvolvido, tornando-se o modelo mais completo até hoje existente. O novo modelo prêve o aparecimento de um máximo no sinal de speckle-foto-fem para um determinado valor da amplitude normalizada sobre o tamanho do speckle d , em conformidade com os resultados experimentais. Verificou-se também, que a posição deste máximo depende fortemente da relação entre a condutividade no escuro e fo-tocondutividade (Rd). Através da técnica Fotocondutividade Resolvida por Comprimento de Onda foi estudado as amostras de BTO puro e dopado, CdTe e ZnTe dopados, onde foi possível identificar alguns estados localizados dentro da banda proibida destes materiais. A partir disto, alguns cristais, como o BTO, Cdte:V e CdTe:Ge foram selecionados para os experimentos de medidas de vibrações transversais utilizando a técnica speckle-foto-fem. Os experimentos de speckle-foto-fem foram, pela primeira vez, utilizados para caracterizar materiais fotorrefrativos, a partir da determinação do tempo de resposta e da estimativa da condutividade no escuro. / Abstract: A study of new photorefractive materials was performed through the techniques of wa-velength resolved photoconductivity and Speckle-photo-fem. Among this materials are the titanosillenites with different dopants and the group II-IV Photorefractive materials like the Cadmium telluride (CdTe) and the Zinc telluride (ZnTe), also doped, which are interesting for having a band gap smaller than that of the sillenites, and for being a lot faster than the sillenites. The mathematical model that describes the Photo-emf effect with an oscillating and high amplitude speckle pattern was developed, becoming the most complete model in existence. The new model predicts the presence of a maximum in the signal of speckle-photo-emf for a determined value of the amplitude normalized over the size of the speckle d , in conformity with the experimental results. Was also verified that the position of this maximum is strongly depen-dent on the relation between the dark conductivity and photoconductivity (Rd). Through the technique of wavelength resolved photoconductivity the samples of pure and doped BTO, CdTe and ZnTe were studied and it was possible to identify some localized states inside the band gap of those materials. From that, some crystals like the BTO, CdTe:V and CdTe:Ge were se-lected for the experiments on transversal vibration measurement using the Speckle-photo-emf technique. The speckle-photo-emf experiments were, for the first time, used to characterize photorefractive materials, from the determination of the response time and the estimate of their dark conductivity. / Doutorado / Ótica / Doutora em Ciências

Room temperature CdZnTe X- and gamma-ray detectors for nuclear physics applications

Gamma-ray spectroscopy is undoubtedly the most effective tool for understanding the structure of the nucleus. In common with many other problems however, there is more information available that can be readily measured by standard experimental facilities. Therefore, this thesis investigates the potential for the use of a new detector material, CdZnTe, in nuclear physics applications. To evaluate the requirements of detection systems for nuclear physics applications, a y-ray spectroscopy experiment was performed to investigate neutron alignments in 100Mo, 104Ru and 108Pd using deep-inelastic reactions. This showed that a detector capable of detecting low energy (< 100 keV) X- rays without compromising y-ray detection efficiency could have significant benefit. A room temperature CdZnTe semiconductor detector could reasonably form part of a standard escape suppressed spectrometer. However, there is a substantially higher leakage current associated with room temperature semiconductor devices than standard cryogenically cooled semiconductor detectors. CdZnTe suffers from significant charge trapping, and therefore the rise time of the radiation induced pulses forms an important part of the signal analysis from such detectors. These two problems have implications on the design of preamplifier systems for CdZnTe detectors. For this reason, this thesis describes the design of optimised electronic systems for use with room-temperature operated CdZnTe detectors. Here, the focus is on the preamplifier design, and on practical ways of analysing noise performance of the preamplifier. A new preamplifier configuration with digital output has been developed, and a detailed signal-to-noise analysis performed. Such a circuit facilitates simultaneous measurement of both energy and pulse shape information.

Derivados aromáticos de selênio e telúrio: aplicação da biocatálise na preparação de selenetos e teluretos aromáticos enantiomericamente enriquecidos / Aromatic compounds containing Selenium and Tellurium: application of biocatalysis for preparation of enatiomerically enriched aromatic selenides and tellurides

A primeira parte desta tese consiste na preparação de compostos aromáticos contendo átomos de selênio e de telúrio. Quatro metodologias foram utilizadas para essa finalidade, a saber: orto-metalação de compostos contendo oxigênio, troca metal-halogênio em haletos aromáticos substituídos, reação de substituição eletrofílica aromática entre tetracloreto de telúrio e compostos aromáticos ativados e reação de sais de diazônio com disselenetos e diteluretos orgânicos. Alguns dos teluretos preparados foram usados em reações de acoplamento com alcinos terminais catalisada por Paládio, em reações de troca telúrio-lítio e em reações de oxidação de Te (II) a Te(IV). A segunda parte da tese consiste no uso de calcogenetos de arila em reações biocatalisadas. Inicialmente foram estudadas biotransformações em substratos não contendo átomo de selênio e de telúrio. Reações de redução de carbonila e de desracemização de álcoois foram observadas por ação de fungos e de raízes de plantas. Meta e para organosseleno acetofenonas foram reduzidas aos organosseleno feniletanóis correspondentes por ação de fermento de pão, células íntegras de fungos e por Daucus carota com conversões e excessos enantioméricos que chegaram a >99%. Orto organosseleno e metiltio feniletanóis foram resolvidos em seus isômeros R e S por ação de lipase imobilizada (Novozyme 435) em presença de acetato de vinila com excessos enantioméricos acima de 99% . / The first part of this thesis shows the preparation of organic compounds containing selenium or tellurium. For this purpose, four methodologies were applied: ortho-metallation of 1-phenylethanol, metal-halogen exchange involving aromatic halides, electrophilic aromatic substitution using tellurium tetrachloride and reactions with aryldiazonium salts and diselenides or ditellurides. Next, the aryl tellurides were applied in Palladium catalyzed coupling reactions with terminal alkynes, tellurium-lithium exchange reactions and oxidation of Te(II) to Te(IV). The second part of this thesis consists in the inverstigation of biocatalyzed reactions. Biotransformations of substrates without Se or Te atoms were initially investigated. Carbonyl reduction reactions and deracemization of secondary alcohols were observed by means of whole fungal cells and plants. Meta and para organoseleno acetophenones were then reduced with baker´s yeast, whole fungal cells and Daucus carota, yielding the corresponding organoseleno phenylethanols optically pure with enantiomeric excess up to >99%. Ortho organoseleno e methylthio phenylethanols were resolved in both enantiomeric forms by reacting them with immobilized lipase (Novozyme 435) and vinyl acetate in hexane. High values of enantiomeric excess (>99%) were obtained.
