The health of British seamen in the West Indies, 1770-1806Convertito, Coriann January 2011 (has links)
This thesis examines the impact of disease and mortality on the Royal Navy in the West Indies from 1770 to 1806. It also investigates the navy’s medical branch which was established to manage the care of sick seamen. Using an interdisciplinary approach, this thesis produces a cohesive understanding of how disease and mortality affected the navy’s presence in the West Indies and the ways in which the navy attempted to mitigate their impact. This thesis explores various aspects of naval medicine including the history of the Sick and Hurt Board, the diseases which distressed seamen, the medicines distributed by the navy, the key personnel who were integral in generating changes to the medical system and the development of hospital facilities. Largely based on Admiralty records including correspondence and minutes from the Sick and Hurt Board, ships’ muster books and surgeons’ journals, this thesis investigates the most prevalent diseases in the West Indies and the prescribed treatments advocated by the navy. It then examines how these diseases and treatments affected seamen on board ships in that region through a quantitative analysis; then focuses on a number of the integral naval personnel who ushered in sweeping changes to naval medicine; and explores the navy’s increasing desire to transition from hired sick quarters to purpose-built naval hospitals on various West Indies islands. It concludes with a case study of the development of Antigua naval hospital which demonstrates the effectiveness of these facilities in convalescing sick seamen. Through a quantitative analysis of ships’ muster books, this thesis argues that the levels of sickness and mortality in the navy in the West Indies during the late eighteenth century are largely exaggerated in historical studies while also discrediting the myth that those islands were the ‘white man’s graveyard’ for many naval personnel. By surveying over 100,000 seamen on board ships in that region, sickness and mortality figures emerge which indicate that, on average, less than 4 per cent of seamen were on the sick list at any given time and only a small percentage died, meaning that the majority remained on active duty. This thesis then argues that many of the changes to the navy’s medical system that facilitated such low percentages were primarily instigated by surgeons, physicians and captains who identified beneficial medicines and championed their general distribution among the entire fleet. By looking at these aspects of naval medicine through a multidisciplinary lens rather than a purely administrative one, it is possible to understand the true state of health of British seamen in the West Indies during the last quarter of the eighteenth century.

Previous issue date: 2009 / O objetivo da tese é refletir sobre as atividades de saneamento no Estado do Amazonas no período da Primeira República no Brasil. O momento coincide com o auge e o declínio da economia da borracha, fazendo da Região um importante centro de repercussão da cultura e das idéias científicas. Analisamos como as idéias da medicina tropical foram apropriadas por médicos do Amazonas e como foram colocados em prática os princípios daquela disciplina, com o objetivo de realizar a profilaxia da febre amarela e da malária. Essas duas endemias mobilizaram os diversos atores em torno das teorias, que debatiam sobre os mecanismos de transmissão e sobre as formas de combater e controlar os vetores. Diferentes comissões atuaram no Amazonas nesse período, que envolveram: médicos,cientistas e engenheiros de Manaus, da Capital Federal e de Instituições internacionais. A única comissão que realizou o saneamento do interior do Estado foi o Serviço de Saneamento e Profilaxia Rural, que transformou o posto rural em posto itinerante. O saneamento do Amazonas foi proposto e realizado por estas diferentes comissões, que teve como pano de fundo o ambiente e o regime das águas: na capital os igarapés, e no interior a imensa rede de rios e lagos. O Amazonas, nesse período, constituiu-se, portanto, em uma espécie de laboratório para a pesquisa e a experimentação das idéias científicas correntes. A região, em relação à nação, afirmou a sua autonomia e a sua identidade, através da ação dos médicos e cientistas que refletiram sobre a o saneamento do Estado .

causar doença grave e morte. O vírus da FA é um membro da família Flaviviridae do gênero
Flavivírus. No Brasil, todas as regiões já apresentaram casos da doença e o número de casos
fatais tem aumentado. Até hoje, a vacinação é o meio mais eficaz para prevenir e controlar a FA.
A partir de 2011, o quantitativo solicitado tanto pelo Ministério da Saúde quanto para exportação
aumentou, e este é um dos motivos que leva a discussão para a formação de um estoque
estratégico deste produto em Bio-Manguinhos. Portanto, para que seja garantido o suprimento da
vacina febre amarela (atenuada) aos órgãos nacionais e internacionais em momentos de maior
demanda, assim como aumentar a oferta e melhorar o aproveitamento de doses distribuídas,
evitando o desperdício de uma vacina estável, estudos de estabilidade foram realizados visando à
extensão do prazo de validade das vacinas febre amarela (atenuada) 05 e 10 doses de 24 para 36
meses, quando armazenadas a -20ºC e a 2-8ºC. Para aanálise da estabilidade da vacina 10 doses,
foram avaliados dados retrospectivos de potência, termoestabilidade, umidade residual, e outros
parâmetros menos críticos, obtidos a partir de estudos de estabilidade longa duração (-20ºC e 2-8ºC / 48 meses) e pós-reconstituição (2-8ºC / 08 horas), assim como dados obtidos nos períodos
de 2009 e 2010. Para a análise da estabilidade da vacina 05 doses, foram avaliados dados
retrospectivos de potência obtidos de estudos de acompanhamento da estabilidade realizados
entre 2003 e 2009 (-20ºC / 36 meses), além de estudos complementares de acompanhamento da
estabilidade, realizados com lotes produzidos entre2005 e 2006 (-20ºC e 2-8ºC / 48 meses). Os
dados obtidos para ambas as apresentações puderam ser comparados, pois elas são constituídas
pela mesma formulação, diferenciando-se somente no volume de envase. Para a análise estatística
dos dados foram utilizados a regressão linear, a análise de covariância (ANCOVA), além do
cálculo de intervalos de confiança, intervalos preditivos e extrapolação de dados. Os resultados
de potência e termoestabilidade tanto para a vacina05 doses quanto para a 10 doses decaem ao
longo do tempo de armazenamento em ambas as temperaturas, entretanto permanecem de acordo
com as respectivas especificações. Já os resultadosde umidade residual obtidos para a vacina 10
doses revelaram um incremento na umidade ao longo do tempo, mas também se mantêm de
acordo com a respectiva especificação. Os resultados de potência e esterilidade, obtidos para a
vacina 10 doses em estudos pós-reconstituição, também demonstraram a estabilidade deste
produto armazenado por 08 horas a 2-8ºC. Portanto, o prazo de validade das vacinas febre
amarela (atenuada) 05 e 10 doses pode ser estendidopara 36 meses, quando armazenado a -20ºC
ou a 2-8ºC, sem alteração de sua qualidade. / Yellow fever (YF) is an acute viral disease, transmitted by mosquitoes, which can cause serious
illness and death. The YF virus is a member of the Flaviviridaefamily, Flavivirus genus. In
Brazil, all regions have already had cases of the disease and the number of fatalities has
increased. Until now, vaccination is the only effective means to prevent and control YF. From
2011, the amount of vaccine requested by Ministry of Health and for exports increased, and this
is one of the reasons leading the discussion for the formation of a strategic stock of this product
in Bio-Manguinhos. Therefore, to ensure the regularsupply of yellow fever vaccine (attenuated)
to the national and international agencies in timesof high demand, as well as increase supply and
improve the use of doses distributed, avoiding the waste of a stable vaccine, stability studies were
performed to propose the extension of the shelf-life of yellow fever vaccine 05 doses (YFV-05)
and 10 doses (YFV-10) from 24 to 36 months when stored at -20ºC and 2-8°C. For the stability
analysis of YFV-10 were evaluated retrospective potency, thermostability and residual moisture
data, and other parameters less critical, obtained from studies of long-term stability (-20ºC and 2-8ºC / 48 months) and after reconstitution (2-8ºC / 08 hours) as well as data obtained during the
periods of 2009 and 2010. To analyze the stability of YFV-05 were evaluated retrospective
potency data obtained from annual stability studiesconducted between 2003 and 2009 (-20ºC /
36 months) in addition to complementary annual stability studies, carried out with batches
produced between 2005 and 2006 (-20ºC and 2-8ºC / 48 months). The data obtained for both
presentation could be compared because both have the same formulation, differing only in filling
volume. For the statistical analysis were used linear regression, analysis of covariance
(ANCOVA), and the calculation of confidence intervals, predictive intervals and data
extrapolation. The results of potency and thermostability for both YFV-05 and YFV-10 decay
over time at both storage temperatures and remain according to specifications. However, the
results obtained for residual moisture to YFV-10 revealed an increase in moisture over time, but
remaining according to specification. Analysis of potency and sterility obtained for YFV-10 after
reconstitution study also demonstrated the stability of the product stored for 08 hours to 2-8°C.
Therefore, the shelf-life of yellow fever vaccine 05 and 10 doses may be extended for 36 months
when stored at -20 C or 2-8ºC, without changing itsquality.

A Febre Amarela (FA) é uma doença viral infecciosa, não contagiosa que pode se manifestar desde um quadro febril até a forma clássica íctero-hemorrágica, cujo agente etiológico é o vírus da febre amarela (VFA). A doença tem avançando para as regiões Sudeste e Sul com um aumento de 4 vezes no número de vítimas fatais. O presente trabalho teve como objetivos avaliar a possível circulação do vírus da febre amarela silvestre, através da detecção de anticorpos IgM por meio da técnica de MAC-ELISA, e anticorpos IgG, pelas técnicas de Inibição da hemaglutinação e Neutralização; detecção do genoma viral utilizando a técnica de RT-PCR no soro de indivíduos assintomáticos; bem como avaliar a proteção vacinal, por meio da técnica de neutralização em soros de indivíduos das zonas rurais do município de Jacupiranga e Teodoro Sampaio. Um total de 238 soros foram coletados, 152 (Jacupiranga) e 86 (Teodoro Sampaio). Foram detectados um total 15,9% (38/238) de anticorpos IH contra o vírus selvagem e/ou vacinal. Destes 13,16% (5/38) anticorpos IH foram provenientes dos soros de Jacupiranga e 86,84 (33/38) de Teodoro Sampaio. Anticorpos neutralizantes foram detectados em 34% (13N/38IH) dos IH reativos, destes 15,38% (2/13) foram oriundos deJacupiranga e 84,62% (11/13). A detecçção de anticorpos neutralizantes nos indivíduos de Jacupiranga levantam a necessidade de pesquisas futuras na busca do vírus da febre amarela nos seus reservatórios selvagens e vetores. Não houve a detecção do genoma viral por RTPCR nas duas populações, bem como a detecção de anticorpos IgM específicos, por meio das reações de MACELISA, o que evidencia a não circulação recente deste vírus na população estudada. O grau de imunizaçao na população de Teodoro Sampaio, com histórico de vacinação, por meio das reações de IH e Neutralização foi baixo, uma vez que somente 42,30% da população apresentou anticorpos inibidores da hemaglutinação e o grau de proteção pela reação de neutralização foi de apenas 33,33%. Este trabalho aponta não só para a necessidade de monitoramento pós-vacinal em áreas endêmicas de febre amarela, com indicação de vacinação, como também a necessidade de estudos epidemiológicos, uma vez que se detectou reação monotípica no teste do IH, além de anticorpos neutralizantes para o vírus selvagem da febre amarela em um indivíduo residente em município de risco para a febre amarela. / Yellow fever (YF) is an non-contagious infection disease. The clinical signs can be a nonspecific fever or the classical ictero-hemorrhagic form as a result of the Yellow Fever virus (YFV) infection. Considering the recent spread of the disease to the South and Southeast of Brazilian regions and the increased number of fatal cases, the aim of this study was to evaluate the possible circulation of the YFV, using the IgM enzyme-linked immnunosorbent assay (MACELISA) and Reverse transcriptase polymerase chain reaction (RT-PCR), as well as, to conduct a serological inquire and evaluate the protective vaccine response, by the hemagglutination inhibition test (HI) and serum neutralization test (SN), in the rural population of Jacupiranga and Teodoro Sampaio, located in the southern region of Brazil. A total of 238 serum samples were tested, 152 from Jacupirang and 86 from Teodoro Sampaio. Of the serum collected, 15,9% (38/238) were positive by HI, using wild and vaccine strains of YF, and out of these 13,16% (5/38) were from Jacupiranga and 86,84% (33/38) were from Teodoro Sampaio. Neutralizing antibodies were detected in 34% of the HI positive samples (13SN/38HI) and out of these 15,38% (2/13) were from Jacupiranga and 84,62% (11/13) were from Teodoro Sampaio. None of the samples were positive by MACELISA and RT-PCR indicating no evidences of recent virus circulation in the population analyzed in this study. However, the YF neutralizing antibodies detection in samples from Jacupiranga indicates the necessity of further researches in order to detect the YFV in its wild reservoirs and vectors. On the other hand, considering that 42,30% of the Teodoro Sampaio samples were positive by HI and out of these only 33,33% had neutralizing antibodies to the YFV, our results also pointed out to the importance of pos-vaccination protective immunity surveys, in order to evaluate the efficiency of the vaccines in endemic areas.

[pt] Os antibióticos eritromicina e canamicina, pertencentes às
classes dos
macrolídeos e aminoglicosídeos respectivamente, têm sua
importância como
agentes preservativos no processo de preparação e durante
o processo de
utilização da vacina contra a febre amarela - apresentação
de cinco doses -
produzida no Instituto de Tecnologia em Imunobiológicos,
Bio-Manguinhos,
FIOCRUZ, RJ. Este trabalho propõe apresentar duas novas
abordagens para a
determinação espectrofluorimétrica desses antibióticos,
considerando o
desempenho, inclusive com relação à seletividade, na
análise dessa matriz
específica (vacina). Um estudo preliminar do comportamento
luminescente
(fosforescência e fluorescência) dessas substâncias foi
realizado em diferentes
condições experimentais a fim de encontrar as que
permitissem o
desenvolvimento dos métodos analíticos. Para a
eritromicina, que não apresentou
luminescência nativa, foi utilizado um procedimento de
derivação fotoquímica de
soluções do analito preparadas em meio contendo ácido
sulfúrico. Como resultado
obteve-se um foto-produto que apresentou fluorescência em
412/465 nm. As
condições experimentais foram otimizadas visando a
maximização desse sinal
fluorescente. Nesse caso, foram estudados o tempo de
irradiação com UV, o tipo e
a concentração do ácido utilizado, o tempo e a temperatura
de aquecimento. O
método foi parcialmente validado apresentando limites de
detecção e de
quantificação iguais a 0,25 ug mL-1 e 0,85 ug mL-1
respectivamente. A faixa
linear do método estendeu-se até 200 ug mL-1 e os
parâmetros de precisão e
robustez foram bastante satisfatórios. O método foi
aplicado na análise de uma
simulação de vacina contra a febre amarela e na análise de
medicamentos
comerciais com um percentual de recuperação entre 98,2 e
105,2 %. A
metodologia desenvolvida para a canamicina, que também não
apresentou fluorescência natural, baseou-se na reação de
oxirredução deste composto com o
Ce (IV) produzindo o Ce (III), que é uma espécie
fortemente fluorescente
(255/360 nm). As intensidades dos sinais fluorescentes
medidos no par
excitação/emissão do Ce (III) foram diretamente
proporcionais às concentrações
de canamicina. As condições para a realização da reação
foram estudadas,
avaliando a quantidade de Ce (IV), a concentração e tipo
de ácido usado no meio
reacional e o tempo e temperatura de aquecimento. A ampla
faixa linear (até 1000
ug mL-1) foi obtida, com limites de detecção e
quantificação de 1,22 ug mL-1 e
4,08 ug mL-1, respectivamente. O percentual de recuperação
obtido na análise da
vacina diluída foi de 109,8 % e em amostra diluída de
urina enriquecida com o
analito foi de 103,4 %. / [en] The antibiotics erythromycin and kanamycin, members of the
macrolide and
aminoglycoside classes respectively, have their importance
as preservative agents
for the preparation process and during the using of the
yellow fever vaccine, five
doses presentation, produced in Technology on
Immunobiologicals Institute, Bio-
Manguinhos, FIOCRUZ, RJ. In this work two new approaches
for the
spectrofluorimetric determination of these antibiotics are
presented, taking into
consideration their selectivity performance for this
specific matrix (vaccine). A
preliminary study was performed to evaluate the
luminescent behavior
(phosphorescence and fluorescence) of these substances in
different experimental
conditions aiming to find the best ones that would lead to
the development of the
analytical methods. For erythromycin, which does not
present native
luminescence, a photochemical derivatization was employed
using analite
solutions prepared in sulfuric acid. As a result, a photo-
product was obtained,
which presented fluorescence at 412/465 nm. The
experimental conditions were
optimized aiming the maximization of the fluorescent
signal. In this case, the
studied parameters were the UV irradiation time, the type
and concentration of the
acid utilized, and the time and temperature used for the
heating step. The method
was partially validated, indicating limits of detection
and quantification of
0.25 ug mL-1 and 0.85 ug mL-1, respectively. The linear
dynamic range of the
method extended up to 200 ug mL-1 and the parameters
related to precision and
robustness were very satisfactory. The method was applied
in the analysis of a
simulated yellow fever vaccine and in commercial
pharmaceutical formulations.
Recovery percent between 98.2 and 105.2 % were achieved.
The methodology
developed for kanamycin, which also do not presented
natural fluorescence, was
based on the oxirreduction reaction of this compound with
Ce (IV), producing Ce
(III), a strongly fluorescent species (255/360 nm). The
fluorescence intensities, measured in the
excitation/emission pair of Ce (III), was directly
proportional to
the kanamycin concentration. The reaction conditions were
studied by the
evaluation of the amount of Ce (IV), the type and
concentration of the acid
utilized in reactional medium and heating time and
temperature. Large linear
range (up to 1000 ug mL-1) was obtained, with detection
and quantification limits
of 1.22 ug mL-1 and 4.08 ug mL-1, respectively. The
recovery percent obtained in
the analysis of a diluted vaccine was 109.8 % while the
recovery achieved for
diluted spiked urine was 103.4 %.

Epidemiology and Laboratory Diagnostics of Dengue, Yellow Fever, Zika, and Chikungunya Virus Infections in AfricaAdam, Awadalkareem, Jassoy, Christian 08 May 2023 (has links)
Arbovirus infections are widespread, and their disease burden has increased in the past decade. In Africa, arbovirus infections and fever with unknown etiology are common. Due to the lack of well-established epidemiologic surveillance systems and accurate differential diagnosis in most African countries, little is known about the prevalence of human arbovirus infections in Africa. The aim of this review is to summarize the available epidemiological data and diagnostic laboratory tools of infections with dengue, yellow fever, Zika, and chikungunya viruses, all transmitted by Aedes mosquitoes. Studies indicate that these arboviral infections are endemic in most of Africa. Surveillance of the incidence and prevalence of the infections would enable medical doctors to improve the diagnostic accuracy in patients with typical symptoms. If possible, arboviral diagnostic tests should be added to the routine healthcare systems. Healthcare providers should be informed about the prevalent arboviral diseases to identify possible cases.

O vírus da dengue (DENV) e o vírus da febre amarela (YFV) (gênero Flavivirus, família Flaviviridae) representam importantes arbovírus causadores de doenças em humanos que afetam as regiões tropicais e sub-tropicais do planeta, somando milhões de infectados anualmente. Embora exista uma vacina contra o YFV, ainda são notificados muitos casos de febre amarela nas regiões endêmicas das Américas e, principalmente, da África. Recentemente, foi licenciada uma vacina contra o DENV, mas seu uso ainda é bastante restrito. Não existem agentes terapêuticos para tratamento da infecção contra nenhum desses vírus. Portanto, estudos para identificação de fármacos para combaterem a infecção pelos DENV e YFV são de suma importância. Nosso grupo descreveu a ação antiviral da fosfolipase A2 crotoxina B (PLA2-CB) da serpente Crotalus durissus terrificus, a qual tem ação diretamente sobre o envelope de DENV e YFV. A meta principal deste trabalho foi a produção das duas isoformas da PLA2-CB (CB1 e CB2) recombinantes e avaliação de suas atividades antivirais, a fim de contribuir com a prospecção de novas drogas antivirais. Para alcançar tais propósitos, as sequências codificadoras das isoformas foram otimizadas para expressão em sistema procarioto, quimicamente sintetizadas e enzimaticamente inseridas no vetor de expressão pGS-21a. Os plasmídeos gerados foram utilizados para transformar células E. coli das cepas BL21(DE3), Origami B(DE3) e ArcticExpress (DE3). As proteínas recombinantes foram expressas juntamente com uma cauda de polihistidina (6xHis) no extremo C-terminal (CB1+6xHis_opt e CB2+6xHis_opt). Ambas as proteínas foram reconhecidas por anticorpos produzidos contra a PLA2-CB in natura em ensaio de Western Blot. Considerando que as proteínas foram expressas de forma insolúvel, a purificação foi realizada em sistema de cromatografia líquida (FPLC), utilizando coluna com resina de afinidade por níquel sob condições desnaturantes, utilizando ureia como agente solubilizador. As proteínas foram renaturadas na própria coluna de níquel (on-column) durante a purificação, utilizando um gradiente decrescente de ureia (6-0M - 120ml - 0,1ml/min) e o detergente CHAPS como agente estabilizador. As proteínas CB1+6xHis_opt e CB2+6xHis_opt em ensaio colorimétrico de hidrólise de fosfatidilcolina apresentaram atividade fosfolipásica, indicando preservação do sítio catalítico. As proteínas CB1+6xHis_opt e CB2+6xHis_opt podem vir a ser um importante modelo na prospecção de novas drogas antivirais e como ferramenta no estudo do mecanismo de replicação viral. / Dengue virus (DENV) and yellow fever virus (YFV) (genus Flavivirus, family Flaviviridae) are important arboviruses that cause diseases in humans affecting the tropical and subtropical regions of the planet with millions of infected annually. Although there is a vaccine against YFV, many cases of yellow fever are still reported in the endemic regions of the Americas, and especially in Africa. A vaccine against DENV has recently been licensed, but its use is still quite restricted. There are no therapeutic agents to treat the infection against any of these viruses. Therefore, studies to identify drugs to combat DENV and YFV infection are of extreme importance. Our group described the antiviral action of the phospholipase A2 crotoxin B (PLA2-CB) isolated from Crotalus durissus terrificus, which acts directly on the envelope of DENV and YFV. The aim of this study was to produce the two recombinant PLA2-CB (CB1 and CB2) isoforms and evaluate their antiviral activities in order to contribute to the prospection of new antiviral drugs. To achieve such purposes, the coding sequences of the isoforms were optimized for prokaryotic expression system, chemically synthesized and enzymatically inserted into the pGS-21a vector. The plasmids generated were used to transform E. coli cells from BL21 (DE3), Origami B (DE3) and ArcticExpress (DE3) strains. Recombinant proteins were expressed tagged with a polyhistidine (6xHis) tail at the C-terminus (CB1+6xHis_opt and CB2+6xHis_opt). Both proteins were recognized by antibodies raised against native PLA2-CB in Western blot assay. Considering that the proteins were expressed in insoluble form, purification was performed in liquid chromatography system (FPLC) using nickel resin column under denaturing conditions, using urea as the solubilizing agent. The proteins were renatured on-column during purification procedure using a decreasing gradient of urea (6-0M - 120ml - 0,1ml/min) and CHAPS as a stabilizing agent. CB1+6xHis_opt and CB2+6xHis_opt proteins showed phospholipase activity in a colorimetric assay based on phosphatidylcholine hydrolysis, indicating preservation of the catalytic site. In a virucidal assay against DENV-2, YFV and two other enveloped viruses: Chikungunya virus (CHIKV) and Zika virus (ZIKV), the recombinant proteins reduced the plaques of lysis formation exhibiting selectivity indices at the mean of 0.6. The CB1+6xHis_opt and CB2+6xHis_opt proteins could be important models for the prospection of new antiviral drugs and as tools for the study of the mechanism of viral replication.

Filogeografia da febre amarela na América do Sul / Phylogeography of the Yellow Fever in South AmericaSouza, Renato Pereira de 11 April 2013 (has links)
Os Flavivírus são vírus de 40 50 nm de diâmetro, com formas esféricas e RNA de fita simples, com sentido positivo e aproximadamente 11 kb de comprimento. O Vírus da Febre Amarela, protótipo do grupo, é o agente causador da Febre Amarela, uma antiga doença que causou epidemias generalizadas na África, Américas do Norte e do Sul e Europa do século XVII ao início do século XX, e depois ressurgiu nas últimas décadas na África sub- saariana e América do Sul tropical. O presente trabalho busca a reconstrução da transmissão da Febre Amarela na América do Sul, no tempo e espaço, em especial, considerando a provável influência das populações humanas, primatas não humanos e mosquitos, na evolução e distribuição das linhagens genéticas de Febre Amarela, aplicando modelos de inferência Bayesiana para análises filogenéticas e filogeográficas e testando hipóteses de distribuição geográfica com modelagem de nicho ecológico. Os dados dão poucas evidências de que as estratégias de vacinação vigentes tenham efetivamente colaborado para a diminuição da ocorrência de Febre Amarela, indicando possíveis erros na estratégia de vacinação. A partir da análise Coalescente da população viral de Febre Amarela, a população viral apresentou um decréscimo importante iniciado em meados dos anos 90. A análise filogeográfica sugere um padrão geral de transmissibilidade Source-Sink destacando a região amazônica como fonte de diversidade para as outras áreas estudadas, com uma estrutura filogeográfica secundária em ondas. Assim, as introduções do vírus em áreas fora da amazônia tem ocorrência aleatória e podem ser ligadas temporalmente e geograficamente ao norte da America do Sul. Os modelos de distribuição geográfica corroboram esse padrão e indicam uma área possível para circulação da Febre Amarela ampla, englobando diversos ecótonos. Os resultados indicam um possível efeito em longo prazo da vacinação atuando diretamente sobre a evolução e dinâmica filogenética da Febre Amarela e sugere que monitorar a evolução do vírus da Febre Amarela é uma estratégia válida para compreender sua distribuição geográfica e evidenciar mecanismos complexos de transmissão e introdução. Por sua vez, os modelos de Nicho Ecológico mostraram ser ferramentas adequadas para calcular o risco da doença em determinadas áreas, sem sua ocorrência prévia, contribuindo como um modelo preditivos para orgãos de Vigilância prepararem suas estratégias de prevenção e controle no caso de possível introdução de patógenos / The flaviviruses are viruses of 40-50 nm in diameter, with spherical shaped and single-strand RNA with positive sense and approximately 11 kb in length. The Yellow Fever virus is the prototype of the group and the causative agent of Yellow Fever, a disease which caused widespread epidemics in Africa, North America, South America and Europe of the seventeenth century to the early twentieth century. The disease reemerged in recent decades in sub-Saharan Africa and tropical South America. This manuscript aims to reconstruct, in time and space, the transmission of yellow fever in South America, through the applying of a Bayesian inference model, considering the probable influence of human populations, nonhuman primates and mosquitoes on the evolution and distribution of Yellow Fever genetic lineages. Distributional pattern hypothesis will be tested by computational modeling of ecological niche. The data provide little evidence that current vaccination strategies have effectively contributed to reducing the occurrence of Yellow Fever, indicating possible errors in the vaccination strategy. From the analysis of the Yellow Fever population Coalescence, the viral population showed a significant decrease started in the mid-90s. The phylogeographic analysis suggests a general pattern of transmissibility \"Source-Sink\" highlighting the Amazon region as a source of diversity for the other areas studied, with a secondary phylogeographic wave like structure. Thus, the introductions of the virus into areas outside the Amazon has random occurrence and can be linked temporally and geographically to the north of South America The geographical distribution models corroborate this pattern and indicate a broad possible area for Yellow Fever circulation, encompassing many ecotones. The results indicate a possible long-term effect of vaccination acting directly on the evolution and phylogenetic dynamics of Yellow Fever and suggests that monitoring the evolution of the Yellow Fever virus is a valid strategy to understand the geographical distribution and highlight complex transmission mechanisms and spatial movements. In turn Ecological Niche models showed as an appropriate tool to calculate disease risk in certain areas without previous occurrence of the disease, working as a predictive model for Surveillance institutions prepare their strategies for prevention and control in the case of possible pathogen introduction
