Development of control strategies for Francisella noatunensis subsp. orientalis in Nile tilapia, Oreochromis niloticus

Nile tilapia, Oreochromis niloticus, is one of the most important farmed fish globally. One of the most serious bacterial diseases constraining global tilapia production is Francisellosis caused by Francisella noatunensis subsp. orientalis (Fno). Although outbreaks of Fno are increasing worldwide, there are no licenced commercial vaccines to prevent the disease for use on tilapia farms. Thus, the current treatment of choice is the use of antibiotics combined with increasing water temperature up to 30°C. Studies investigating the diversity of circulating Fno isolates and the immune response of tilapia elicited by vaccination against piscine francisellosis are lacking. In addition, the current conventional and molecular tools used for detection of Fno have many drawbacks, making detection of Fno a challenging process. In this study, five clinical isolates of Fno from diverse geographical locations (UK, Costa Rica, Mexico, Japan and Austria), previously characterised by morphology, genotype, antimicrobial susceptibility and virulence, were used in a proteomic study. The whole proteomic cell profile of the five isolates were homogenous by one-dimension sodium dodecyl polyacrylamide gel electrophoresis (1D-SDS-PAGE), while minor differences in the intensity of 15 proteins between the strains were observed by two-dimension SDS-PAGE (2DE), including some important virulence related proteins. The UK isolate was the most significantly different isolate when compared to the other Fno isolates in the current study. The Fno UK isolate had significantly higher abundance of 10/15 of the significantly expressed proteins including four of the essential pathogenicity and virulence related proteins (IglC, GroEL, DnaK, ClpB) compared to the other used Fno isolates. The antigenic profiles of the five Fno isolates were studied by 1D western blotting using tilapia hyper immune sera which recognised an immunodominant band of a molecular weight of ~ 17-28 kDa in all tested Fno isolates. Liquid chromatography-electrospray ionization-tandem mass spectrometry (LC/ESI/MS/MS) identified 47 proteins in this antigenic band. Some of the identified proteins are associated with Fno pathogenicity. 2D western blot analysis of the vaccine isolate (Fno UK) revealed differential antigen recognition between sera from vaccinated and non-vaccinated fish following experimental challenge (26 antigenic spots recognised by sera from vaccinated fish; 31 antigenic spots recognised by sera from vaccinated and challenged fish and 30 antigenic spots recognised by non-vaccinated and challenged fish). The identity of these proteins was determined by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS) and some of them are known Francisella virulence related proteins. Bioinformatics analyses revealed diverse categories of proteins with high biological functions, however the vast majority of these proteins are involved in energy production and metabolic pathways of the bacteria. This detailed analysis will facilitate the development of cross-strain protective subunit Fno vaccines and antigen-targeted Fno diagnostics. The outer membrane proteins (OMPs) of the same five Fno isolates were extracted using the ionic detergent sarkosyl. The OMP fraction of the different isolates were separated via 1D-SDS PAGE and the digested peptides of the UK isolate were analysed by LC/ESI/MS/MS. High degree of similarity was observed in the OMP profile of the five Fno isolates with an abundant protein band at 17-28 kDa, which was found to be antigenic by 1D western blot using convalescent tilapia sera. LC/ESI/MS/MS analysis of the OMPs of the Fno UK isolate identified 239 proteins, including 44 proteins in the antigenic band (17-28 kDa). Comparison between the proteins identified in the immunogenic band of whole cell lysate and OMP fraction of the Fno UK isolate showed 30 common proteins between the two preparations, 17 proteins were identified only in the whole cell extract and 14 were identified only in OMP fraction. Outer membrane proteins (e.g. Omp-A), virulence related proteins such (e.g. IglC) and other stress related proteins (e.g. AhpC/TSA family peroxiredoxin) were more abundant in the OMP fraction than the whole cell lysate. In silico analysis enabled prediction of the function and location of the OMPs identified by Mass-spectrometry. The findings of this study provide preliminary data on bacterial surface proteins that exist in direct contact with the host immune defence during infection and offering an insight into their potential role as novel targets for Fno diagnostics and vaccine development. The efficacy of an injectable whole cell oil-adjuvanted vaccine was evaluated against challenge with heterologous Fno isolates in Nile tilapia, Oreochromis niloticus. Three duplicate groups of 130 healthy Nile tilapia (~15 g) were intraperitoneally (i.p.) injected with the vaccine, adjuvant-alone or PBS followed by an i.p. challenge with three Fno isolates from geographically distinct locations. The vaccine provided significant protection to all immunised tilapia groups with a significantly higher relative percent survival (RPS) of 82.3% against homologous challenge, compared to 69.8% and 65.9% after heterologous challenge. Protection correlated with significantly elevated specific antibody responses and western blot analysis demonstrated cross-isolate antigenicity with sera from fish post-vaccination and post-challenge. Moreover, a significantly lower bacterial burden was detected by quantitative real-time polymerase chain reaction (qPCR) in conjunction with significantly greater expression of IgM, IL-1β, TNF-a and MHCII 72 hours post-vaccination (hpv) in spleen samples from vaccinated tilapia compared to those of adjuvant-alone and control fish. The latter results suggested stimulation of protective immune responses following vaccination. In addition, a whole cell formalin killed autogenous immersion vaccine against Fno was developed using the same isolate used for the injectable vaccine. Duplicate tanks of 35 tilapia fry were immersed in the vaccine or in sterile Modified Muller Hinton broth (MMHB) diluted in tank water (1:10 dilution) for 30 s and at 30 days post-vaccination (dpv), all fish groups were immersion challenged with the homologous Fno isolate and monitored for 21 days. A moderate RPS of 43.7% was provided by the vaccine. Serum IgM levels were below the threshold in 30 % of the vaccinated fry 30 dpv. Also, the IgM levels of the vaccinated fry were not significantly different from control fry 21 days-post challenge. A recombinase polymerase amplification (RPA) assay was developed and validated for rapid detection of Fno. The RPA reaction was performed at a constant temperature of 42°C for 20 min. The RPA assay was performed using a quantitative plasmid standard containing a unique Fno gene sequence. Validation of the assay was performed not only by using DNA from Fno, closely related Francisella species and other common bacterial pathogens in fish farms, but also by screening 78 Nile tilapia and 5 water samples collected from UK and Thailand. All results were compared with those obtained by previously established real-time qPCR. The developed RPA showed high specificity in detection of Fno with no cross-detection of either the closely related Francisella spp. or the other species of bacteria tested. The Fno-RPA performance was highly comparable to the published qPCR with detection limits at 15 and 11 DNA molecules detected, respectively. The Fno-RPA was rapid, giving results in approximately 6 min in contrast to the qPCR that required approximately 90 min to reach the same detection limits. Moreover, the RPA was more tolerant to reaction inhibitors than qPCR when tested with field samples. The fast reaction, simplicity, cost-effectiveness, sensitivity and specificity make the RPA an attractive diagnostic tool that will contribute to control the infection through prompt on-site detection of Fno. The overall results of this study indicated that Fno isolates from different origins share a high degree of homology in their proteomic and antigenic profile. Proteomic characterisation data of Fno isolates has contributed to understanding the diversity of Fno isolates and assisted in identifying suitable candidates for developing an effective Fno vaccine. / Moreover, this study has proven the efficacy of a cross protective Fno injection vaccine in tilapia fingerlings, with further optimisation needed for immersion vaccination of fry, and given insights into the immune response of tilapia to vaccination against francisellosis. In addition, it provided a rapid, sensitive, specific and robust molecular tool for detection of Fno that can assist surveillance and control of piscine francisellosis on tilapia farms.

How do prizes induce innovation? learning from the Google Lunar X-prize

Inducement prizes-where cash rewards are given to motivate the attainment of targets--have been long used to encourage scientific research, develop technological innovations, or stimulate individuals, groups, and communities to accomplish diverse goals. Lately, prizes have increasingly attracted the attention of policy-makers, among others, due to their potential to induce path-breaking innovations and accomplish related goals. Academic research, however, has barely investigated these prizes in spite of their long history, recent popularity, and notable potential. This research investigates prizes and the means by which they induce innovation. It uses an empirical, multiple case-study methodology, a new model of innovation applied to prizes, and multiple data sources to investigate three cases of recent aerospace technology prizes: a main case study, the Google Lunar X Prize (GLXP) for robotic Moon exploration; and two pilot cases, the Ansari X Prize (AXP) for the first private reusable manned spacecraft and the Northrop Grumman Lunar Lander Challenge (NGLLC) for flights of reusable rocket-powered vehicles. The investigation unveils the dynamics of prizes and contributes a better understanding of their potential and disadvantages in a context in which more traditional mechanisms are used to induce innovation. This research shows that prizes are a more complex mechanism and their investigation requires analyzing entrant- and context-level factors generally not considered by the literature. Prizes complement and not replace patents and other incentive mechanisms. The incentives offered by prizes attract entrants with diverse characteristics, including unconventional entrants--individuals and organizations generally not involved with the prize technologies. Entrants are generally attracted by the non-monetary benefits of participation and the potential market value of the technologies involved in competitions. Many more volunteers, collaborators, and partners also participate indirectly and support official entries as they also perceive opportunities to accomplish their personal and organizational goals. The monetary reward is important to position the competition in the media and disseminate the idea of the prize. Prizes can induce increasing R&D activities and re-direct industry projects to target diverse technological goals, yet the evolution of prize competitions and quality of the technological outputs is generally difficult to anticipate. The overall organization of prize R&D activities and their outputs depend on entrant-level factors and can only be indirectly influenced by setting specific competition rules. The most remarkable characteristic of prize R&D activities is their interaction with fundraising efforts which, in some circumstances, may constrain the activities of entrants. Prizes can also induce innovation over and above what would have occurred anyway, yet their overall effect depends significantly on the characteristics of the prize entrants and the evolution of the context of the competition. The ability of prizes to induce innovation is larger when there are larger prize incentives, more significant technology gaps implicit in the prize challenge, and open-ended challenge definitions. To successfully induce technological breakthroughs, prizes may require complementary incentives (e.g. commitments to purchase technology) or support (e.g. seed funding.) Prizes are particularly appropriate to, for example, explore new, experimental methods and technologies that imply high-risk R&D; induce technological development to break critical technological barriers; accelerate technological development to achieve higher performance standards; and, accelerate diffusion, adoption, and/or commercialization of technologies. They involve, however, higher programmatic risks than other more traditional mechanisms and their routine use, and/or challenge definitions that overlap, can weaken the incentive power of the mechanism. Successful implementation of competitions requires many parameters to be properly set.

Om utmaning i förskolan : Hur är man rättvis och inkluderande som pedagog?

In my essay, I will start with a story that represents my dilemma from my own experience. In the story, dealing with children that challenges teachers and other children in preschool, I wonder how I as a teacher can be fair and inclusive when I feel insufficient on several occasions. I reflect on my own approach because it can be difficult when we encounter children with different needs and abilities in preschool. I write the essay in experience-based essay form. The essays form makes it possible for me to reflect on my own experience and by using selected theoretical perspectives processing my dilemma. In my essay, I explore my own approach, what is meant by fairness and how the concept can be interpreted and how I can in the best way include all children in activities. I conclude that fairness from a perspective can be seen from the five criteria similarity, need, performance, compensation and rights. The teachers should reflect on their approach and take into account all the needs of every children based on their conditions. Fronesis, practical knowledge, is required to, among other things, make good judgement and to make wise decisions in preschool.

Les casual games : définition à l’aide du savoir professionnel des designers de jeux

Le jeu casual est un phénomène majeur de l’industrie vidéoludique, pourtant il reste peu étudié et mal considéré. Les quelques définitions scientifiques recensées présentent des divergences importantes, ainsi qu’un décalage par rapport aux jeux produits. Les modèles théoriques en design de jeu, centrés sur le gameplay, semblent inadéquats face à ce phénomène. En utilisant le modèle du praticien réflexif de Donald Schön, l’objectif de notre recherche est d’apporter un nouvel éclairage sur le jeu casual grâce au savoir professionnel des designers de jeux. Notre approche est qualitative, d’inspiration phénoménologique : nous avons recueilli l’expérience de huit designers de jeux par l’intermédiaire d’entretiens semi-dirigés proches du récit de pratique. Nos résultats montrent que les concepts utilisés dans les modèles vidéoludiques traditionnels restent pertinents pour définir le jeu casual, mais demandent à être retravaillés afin de rendre compte des mutations de l’expérience de jeu. Ainsi, un challenge dans un jeu casual n’est pas un obstacle mais une opportunité d’action pour le joueur. De plus, la progression se fait par la variation des challenges plutôt que par l’augmentation de la difficulté. Ensuite, certains concepts qui semblaient pertinents pour définir les jeux casual, tels que la fiction positive, ont été rejetés par nos participants. Enfin, notre étude a permis de rassembler des informations sur le rôle du designer de jeu casual et le contexte dans lequel il exerce, ce qui permet d’expliquer certaines causes de la vision péjorative du jeu casual. / Casual games are one of the main phenomena in the video game industry, but remain under-explored and are not highly regarded. Scientific definitions are scarce, inconsistent, and often disconnected from actual games. Further, theoretical models for game design, centered on gameplay, often fall short in defining casual games. The objective of this study is to use Donald Schön's model of the reflective practitioner to shed a new light on casual games through the collection of professional knowledge of game designers. This study follows a qualitative approach with phenomenological inspiration. Eight one-on-one semi-structured interviews were conducted with casual game designers who were asked to provide a narrative of their professional practice. Results show that traditional game design concepts remain relevant for understanding and defining casual games, but need to be reworked in the light of new experiences and contexts in gaming. Namely, challenge is no longer synonymous with obstacle, but should be considered as an opportunity of a player’s action. Progression should be based on the variation of challenges instead of increasing difficulty. In addition, participants rejected some concepts that were previously considered appropriate for defining casual games, like positive fiction. Finally, this study provides insight into the role of casual game designers and the context of their work, which can help explain the often negative perception of casual games.

Étude de l’efficacité de la vaccination à Salmonella Enteritidis chez la poule pondeuse et de la protection contre l’infection

Les infections à Salmonella Enteritidis chez les humains sont associées à la consommation d’œufs ou d’ovoproduits contaminés. La vaccination est un outil utilisé pour diminuer les risques d’infection à SE chez la volaille, mais avec des résultats variables. Au Canada deux bactérines, MBL SE4C et Layermune, sont couramment utilisées pour lutter contre SE. Cependant, leur efficacité n’a pas été complètement déterminée chez les poules pondeuses plus âgées. Par ailleurs, la capacité de ces vaccins à prévenir la transmission verticale et horizontale n’a pas encore été étudiée. L’objectif principal de cette étude était d’évaluer l’effet des deux bactérines sur la réponse immunitaire chez les poules pondeuses, de vérifier la protection conférée par ces vaccins contre l’infection expérimentale à SE, et d’identifier des protéines immunogènes afin de développer un vaccin sous-unitaire. Les oiseaux ont été vaccinés avec deux protocoles d’immunisation en cours d’élevage (soit à 12 et 18, ou à 16 semaines d’âge). Le groupe contrôle a été injecté avec la solution saline. Les oiseaux ont été inoculés per os avec 2 x 109 CFU de la souche SE lysotype 4 à 55 ou à 65 semaines d’âge. Les anticorps (IgG et IgA) ont été mesurés à différents temps avec un ELISA maison en utilisant l’antigène entier de SE. La phagocytose, flambée oxydative, les populations des splénocytes B et T ont été analysées en utilisant la cytométrie en flux. Les signes cliniques, l’excrétion fécale, la contamination des jaunes d’œufs et l’invasion des salmonelles dans les organes ont été étudiés pour évaluer l’efficacité de protection. La transmission horizontale a aussi été étudiée en évaluant l’infection à SE chez les oiseaux mis en contact avec les oiseaux inoculés. Les protéines immunogènes ont été identifiées par SDS-PAGE et Western blot à l’aide d’antisérums prélevés suite à la vaccination et/ou à l’infection expérimentale/naturelle, puis caractérisées par la spectrométrie de masse. Le protocole de vaccination avec deux immunisations a généré un niveau élevé de séroconversion à partir de 3 jusqu’à 32-34 semaines post-vaccination par rapport à celui avec une seule immunisation (p < 0.02), mais il n’y avait plus de différence entre les groupes à 54 et 64 semaines d’âge. Il n’y a pas eu de corrélation entre les niveaux d’IgG et les taux d’isolement des salmonelles dans les organes et des jaunes d’œuf. La production des IgA n’a été observée que chez les oiseaux vaccinés avec 2 injections de MBL SE4C (p ≤ 0.04). Après l’infection expérimentale, la production des IgA a été significativement plus élevée aux jours 1 et 7 p.i dans l’oviducte des oiseaux vaccinés (sauf pour le groupe vacciné avec 2 injections de Layermune) par comparaison avec le groupe contrôle (p ≤ 0.03). Seule la bactérine MBL SE4C a eu un effet protecteur contre la contamination des jaunes d’œuf chez les oiseaux infectés. Ce vaccin réduit partiellement en utilisant deux immunisations, le taux d’excrétion fécale des salmonelles chez les oiseaux inoculés et les oiseaux horizontalement infectés (p ≤ 0.02). Cinq des protéines identifiées par la spectrométrie de masse sont considérées comme des protéines potentiellement candidates pour une étude plus approfondie de leur immonogénicité: Lipoamide dehydrogenase, Enolase (2-phosphoglycerate dehydratase) (2-phospho-D-glycerate hydro-lyase), Elongation factor Tu (EF-Tu), Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) et DNA protection during starvation protein. En général, les bactérines ont induit une immunité humorale (IgG et IgA) chez les poules pondeuses. Cette réponse immunitaire a protégé partiellement les oiseaux quant à l’élimination des salmonelles, la contamination des jaunes d’œuf, ainsi que la transmission horizontale. Dans cette étude, la bactérine MBL SE4C (avec deux immunisations) s’est montrée plus efficace pour protéger les oiseaux que la bactérine Layermune. Nos résultats apportent des informations objectives et complémentaires sur le potentiel de deux bactérines pour lutter contre SE chez les poules pondeuses. Étant donné la protection partielle obtenue en utilisant ces vaccins, l’identification des antigènes immunogènes a permis de sélectionner des protéines spécifiques pour l’élaboration éventuelle d’un vaccin plus efficace contre SE chez les volailles. / Contaminated eggs and egg products have been associated with outbreaks of human Salmonella Enteritidis (SE) infections. Killed bacteria (bacterins) have been used to control Salmonella infections in poultry but variation in the conferred protection has been observed. In Canada the bacterins MBL SE4C and Layermune are currently used to control SE. However, their efficacy in protecting older layers has not been fully determined. Furthermore, the capacity of these bacterins to prevent vertical and horizontal transmissions has not yet been investigated. The main objectives of this study were to evaluate the effect of two available commercial bacterins on the immune response of laying hens, to verify the protection conferred by these vaccines against SE challenge and to identify immunogenic proteins to develop an oral subunit vaccine. Laying hens were vaccinated with two immunization schedules prior to the lay cycle (either at 12 and 18, or 16 weeks of age). The control group was injected with a saline solution. Laying hens were later inoculated per os with 2 x 109 CFU of SE PT4 strain either at 55 or 65 weeks of age. Serum IgG and mucosal IgA antibodies were measured with an in-house SE whole cell antigen ELISA. The phagocytosis, oxidative burst, splenic T and B cells populations were analyzed using flow cytometry. Clinical signs, fecal shedding, egg yolks contamination and organ invasion by SE were assessed to evaluate vaccine protection. Potential horizontal transmission from inoculated laying hens to non-inoculated laying hens, housed in the same isolator unit, was also evaluated. Immunogenic proteins were identified by SDS-PAGE and Western blot with sampled antisera during vaccination and/or infection of poultry with SE and then subjected to mass spectrometry. The vaccination protocol with two immunizations showed a higher seroconversion level than the single vaccination at 3 until 32-34 weeks post vaccination (p < 0.02) but no difference before challenge (54 and 64 old weeks). There was no relationship between high IgG level and SE isolation rates in organs and egg yolks. Only the MBL SE4C vaccine elicited IgA antibody production at 3 weeks post vaccination in both immunization protocols (p ≤ 0.04). Significant higher mucosal IgA levels were observed at day 1 and 7 post challenge in oviduct of vaccinated birds (except for the twice vaccinated Layermune group) compared to the control group (p ≤ 0.03). Humoral efficacy to protect from SE contamination of egg yolk was only observed in MBL SE4C vaccinated group and only this bacterin administered twice reduced SE shedding rate in inoculated birds and their exposed cagemates (p ≤ 0.02). A set of 5 proteins were considered as putative protein candidates to further detailed study on their immunogenicity: Lipoamide dehydrogenase; Enolase (2-phosphoglycerate dehydratase) (2-phospho-D-glycerate hydro-lyase); Elongation factor Tu (EF-Tu); Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) and DNA protection during starvation protein. Overall, the commercial bacterins induced humoral immunity (IgG and IgA antibodies) in laying hens. This immune response partially protected for SE clearance, egg yolks contamination as well as horizontal transmission. In this study, MBL SE4C bacterin appeared to be more efficient in comparison to Layermune for protection of hens with a vaccination protocol comprising two immunizations. Our results provide additional and objective information on the potential of these vaccines for the control of SE in laying hens. Considering the partial protection achieved with the use of these bacterins, the identification of immunogenic antigens could help in the selection of specific proteins to elaborate a more efficient vaccine against SE in poultry.

組織中的建言行為:員工目標導向與主管建言行為之探討 / The Voice Behavior in Organization: The Study of Employee’s Goal Orientation and Supervisor’s Voice Behavior

在現今高度競爭的商業環境,員工建言行為的展現對於組織而言日顯重要。過往許多文獻即顯示員工建言行為對於組織有許多益處,包含增進組織決策品質、組織創新與提升團體績效表現。為了全面性瞭解組織中的建言行為,本研究目的為四:(1)了解不同目標導向員工對於建言行為之影響、(2)從主管、同儕與個體本身之角度,了解建言行為對個體之影響、(3)探討建言行為在不同目標導向與結果間所扮演之中介角色,以及(4)探討主管建言行為對員工建言行為之影響。 本研究採對偶的問卷調查,共蒐集157位主管與242位部屬資料,並透過階層迴歸進行資料分析。研究結果顯示:(1)證明目標導向及逃避目標導向皆與挑戰型建言及支持型建言有顯著正向關係、(2)挑戰型建言與支持型建言分別在證明目標導向與組織導向自尊具有部分中介效果、然而兩種建言對於目標導向與主管評價及目標導向與知覺職場排擠皆無中介效果,以及(3)主管建言行為對於目標導向與建言行為間並無調節效果。 根據研究結果,本研究證實目標導向與建言行為之間的關係,以及建言行為對證明目標導向與組織導向自尊的中介效果。最後,研究者進一步提出本研究之理論貢獻、管理實務意涵、研究限制與未來研究方向,以供業界及組織行為領域之學者參考。 / In today’s hypercompetitive business environment, employees’ voice behavior has become even more important to organizations. Many references also showed that employees’ voice behavior can bring benefits to organizations, such as improved organizational decision quality, innovation, and enhancing team performance. To build toward a comprehensive understanding of voice behavior in organizations, the purpose of this research are as follows: (1) to demonstrate how different goal orientations predict employees’ voice behavior, (2) to explore the results of employees’ voice behavior from the perspectives of the supervisor, co-worker, and individual, (3) to demonstrate the mediator role between different goal orientation and outcome variables, and (4) to demonstrate the influence of supervisors’ voice behavior on employees’ voice behavior. With data collected from 157 supervisors and 242 employees to examine the hypothesized relationships, the present study also utilizes the dyad approach. The results showed that (1) both prove goal orientation and avoid goal orientation are positively related to challenge voice and supportive voice, (2) both challenge voice and supportive voice mediated prove goal orientation and organizational based self-esteem respectively, but didn’t play mediators role neither on goal orientation and employee’s performance from supervisor’s view nor on goal orientation and workplace ostracism, and (3) supervisors’ voice behavior has no moderating effect on goal orientation and employee’s voice behavior. In conclusion, the present study showed that goal orientation have the influence on voice behavior, and both challenge voice and supportive voice mediated prove goal orientation and organizational based self-esteem respectively. Finally, theoretical implications, practical implications, limitations and future directions are discussed for managers’ and researchers’ reference.

The South African mining environment has not been an exception to this phenomenon. The aim of this study was to inform the design of talent retention strategies through determination of turnover intention predictors and exploration of the turnover process experienced by employees. The study hypothesised that job satisfaction and affective organisational commitment mediate the effect of overall justice, centralisation, trust and job challenge on turnover intentions. A cross-sectional survey design with a random sample of trainers and learners from a South African gold mining company was used (n=171 and n=230, respectively). Results indicate that job satisfaction mediates the effect of centralisation on turnover intention for learners. Only job satisfaction directly predicts turnover intention for trainers, and overall justice and trust play a direct role in the prediction of turnover intention for learners. It was also found that job challenge predicts job satisfaction for trainers, whilst job challenge, centralisation and trust predict job satisfaction for learners. Therefore, to retain trainers, the mining company should focus on enhancing those factors that facilitate job challenge perceptions and job satisfaction. 國小新移民學童英語學習成就、學習態度及學習挑戰之研究 / A study on new immigrant children's English learning achievement, learning attitude and learning challenge in elementary school

由於國小新移民學童的逐年增加,研究新移民學童的學習情況有其必要性。本研究主旨在專門調查中台灣一所國民小學新移民學童的英語學習狀況。研究主要在探討:(1)新移民學童的英語學習成就;(2)新移民學童的英語學習態度;(3)新移民學童的英語學習成就與其英語學習態度之關係;(4)新移民學童學習英語所面臨的挑戰;(5)新移民學童對自己英語學習的看法;以及(6)教學現場的英語教師對新移民學童英語學習的看法。 本研究的研究對象為十四名五、六年級的新移民學童,並以六十名台灣學童做為比照。研究比較七十四名學童的英語成績,且所有七十四名學童完成一份英語學習態度量表。十四名新移民學童則參與一次訪談,兩名教學現場的英語教師也經歷一次訪談。研究所收集的資料經統計分析及質性分析後,所得的結果如下:(1)大部分新移民學童的英語學習成就不佳;(2)大部分新移民學童英語學習態度是正面的;(3) 新移民學童的英語學習成就與其英語學習態度並無相關性;(4)新移民學童面臨的英語學習挑戰不少,包括他們對不同英語老師的教學風格的適應能力、他們上英語課時產生的焦慮感及緊張感、他們不易牢記英語生字、他們不會寫英語作業、他們沒有辦法去上英語補習班等;(5)新移民學童大部分對自己的英語學習持樂觀看法;(6)英語教師對新移民學童的英語學習則持較悲觀看法。 根據本研究的發現,研究者在本研究報告最後提出數點建議及未來相關研究的研究方向。 / With the increasing of New Immigrant Children in elementary schools, studies to understand New Immigrant Children’s learning conditions become imperative. The purpose of this study was to investigate New Immigrant Children’s English learning conditions in an elementary school in central Taiwan. The study focused on exploring: (1) New Immigrant Children’s English learning achievement; (2) New Immigrant Children’s English learning attitudes; (3) The relationship between New Immigrant Children’s English learning achievement and their learning attitudes; (4) New Immigrant Children’s English learning challenges; (5) New Immigrant Children’s own perspectives about their English learning; (6) English teachers’ perspectives about New Immigrant Children’s English learning. The participants in this study included 14 New Immigrant Children in fifth and sixth grades, with 60 Taiwanese Children working as comparison. These 74 children’s English scores were compared, and they all responded to an English learning attitude scale. The 14 New Immigrant Children then underwent a group interview. Two English teachers were also interviewed. The data collected were then analyzed with statistical and qualitative analyses. The following results were drawn: (1) Most New Immigrant Children were English low-achievers; (2) New Immigrant Children’s English learning attitudes were generally positive; (3) No correlation was found between New Immigrant Children’s English learning achievement and their learning attitudes; (4) New Immigrant Children faced several challenges in learning English, including their adaption to different English teachers’ teaching styles, their feelings of anxiety and nervosity during English classes, their problems to memorize new words and do English homework, their not being able to go to English cram-schools, etc.; (5) New Immigrant Children in general were optimistic about their English learning; (6) English teachers in general were pessimistic about New Immigrant Children’s English learning. Based on the findings of this study, some implications were provided and several suggestions for further studies were offered at the end of the report.

Objects and objectivity : Alternatives to mathematical realism

This dissertation is centered around a set of apparently conflicting intuitions that we may have about mathematics. On the one hand, we are inclined to believe that the theorems of mathematics are true. Since many of these theorems are existence assertions, it seems that if we accept them as true, we also commit ourselves to the existence of mathematical objects. On the other hand, mathematical objects are usually thought of as abstract objects that are non-spatiotemporal and causally inert. This makes it difficult to understand how we can have knowledge of them and how they can have any relevance for our mathematical theories. I begin by characterizing a realist position in the philosophy of mathematics and discussing two of the most influential arguments for that kind of view. Next, after highlighting some of the difficulties that realism faces, I look at a few alternative approaches that attempt to account for our mathematical practice without making the assumption that there exist abstract mathematical entities. More specifically, I examine the fictionalist views developed by Hartry Field, Mark Balaguer, and Stephen Yablo, respectively. A common feature of these views is that they accept that mathematics interpreted at face value is committed to the existence of abstract objects. In order to avoid this commitment, they claim that mathematics, when taken at face value, is false. I argue that the fictionalist idea of mathematics as consisting of falsehoods is counter-intuitive and that we should aim for an account that can accommodate both the intuition that mathematics is true and the intuition that the causal inertness of abstract mathematical objects makes them irrelevant to mathematical practice and mathematical knowledge. The solution that I propose is based on Rudolf Carnap's distinction between an internal and an external perspective on existence. I argue that the most reasonable interpretation of the notions of mathematical truth and existence is that they are internal to mathematics and, hence, that mathematical truth cannot be used to draw the conclusion that mathematical objects exist in an external/ontological sense.
