Concepção, métodos construtivos e dimensionamento de terminais para contêineres. / Conception, construction methods and design of containers terminal.

O presente trabalho pretende mostrar, as principais etapas de projeto de uma obra portuária, mais especificamente, um terminal de contêineres (cais e retroárea). Para tanto, foi feita uma pesquisa dos tipos estruturais existentes para entender melhor a evolução das estruturas ao longo do tempo e quais tipos adequam-se melhor em cada situação. Para cada função a que o porto se destina, existe um conjunto de ações e combinações muito particulares a considerar. Assim, um terminal de contêineres é muito diferente de um terminal petroleiro. Portanto, procurou-se mostrar os tipos de carregamentos mais importantes num terminal de contêineres, que é objeto deste trabalho, levando-se em conta critérios das Normas NBR-9782 e NBR-8681. Outro fator de grande peso nos projetos portuários e que não poderia ficar de fora, está ligado às fundações, principalmente aos problemas geotécnicos envolvidos. Para tanto, procurou-se dar uma visão geral dos principais problemas até hoje encontrados e comentar algumas soluções em cada caso. Por fim, foi proposto um método de modelagem simplificado para um tipo estrutural muito utilizado hoje em dia (sistema de lajes e vigas premoldadas solidarizadas apoiadas sobre estacas) através de um conjunto de modelos planos e um modelo espacial de checagem. / The present paper intends to show the main stages of port design, particularly, a container terminal (quay and back area). Therefore, it was done a research about the current structures types so it can be better to understand the development process of these structures during the years and which types are more appropriate to each condition. For each function of the port there are particulars actions and combinations to consider. However, a container terminal is very different from a petroleum one. Therefore, it intended to show the most important types from a container terminal, which is the aim of this research, considering the NBR-9782 and NBR-8681 rules recommendations. Another important fact, which has to be mentioned, is about the foundations, especially those related to geotechnical issues involved. Therefore, it was intended to present a general view of the main problems found so far and discuss some solutions for each case. Finally, it was suggested a method to model a kind of structure very used nowadays (a marble slab system and premolded beams settled over piles) through a group of model plans a checking space model.

O presente trabalho centra seu estudo na possível utilização de documentos fiscais eletrônicos no trabalho de rastreio de cargas, com o objetivo de propor metodologia de rastreamento apoiada em seu emprego, tendo em vista o processo em curso de mudança de paradigma no relacionamento fisco-contribuinte e a necessidade de evitar fraudes e sonegação que tanto atrapalham o crescimento do país. A metodologia de pesquisa, a partir da conceituação de documento eletrônico, baseou-se no levantamento, análise e avaliação dos principais documentos fiscais eletrônicos já envolvidos na operação com mercadorias e seu transporte (ou em desenvolvimento), num trabalho comparativo, bem como de algumas tecnologias de rastreamento existentes, atentando para os aspectos de validade jurídica e de legalidade de um e de outro, dentro do contexto tributário brasileiro. Os documentos em questão foram: a Nota Fiscal Eletrônica, o Conhecimento de Transporte Eletrônico e o Manifesto Econômico de Documentos Fiscais. Quanto ao rastreamento e sua legislação, as tecnologias contempladas em sua consonância com o objetivo do trabalho foram: tecnologia OCR (Optical Character Recognition), Weigh-in-Motion (WIN), RFID no veículo, RFID no produto, lacre eletrônico, scanners de carga, rastreadores e georreferenciamento. A conclusão do estudo apontou a possibilidade técnica de integração das informações fiscais com tecnologias de rastreamento de cargas e veículos, atualmente restrita, neste formato, ao rastreio de veículos. As possíveis soluções, embasadas juridicamente, implicam em diferentes decursos de prazos, custos e precisão nas informações prestadas. Há, porém, um caminho longo ainda a ser trilhado para que esta integração e adoção sejam plenas. / This essay focuses its study on the possible usage of electronic invoices for tracking freight, in order to propose tracking methodology, in view of shift the relationship paradigm government-taxpayer and the need to avoid fraud and tax evasion that both hinder the country´s growth. The research methodology, from the conceptualization of an electronic invoice, is based on the collection, analysis and evaluation of the main electronic tax documents already involved in the operation with goods and their transport (or in development), a comparative analysis, as well as some existing tracking technologies, noting aspects of legal validity and legality of one and other, within the context of the Brazilian tax. The observed documents are: goods electronic invoice and freights electronic invoice. As for tracking and its legislation, the technologies covered in line with the objective of the study are: OCR technology (Optical Character Recognition), Weigh-in-Motion (WIN), the RFID vehicle RFID product, electronic seal, scanners, cargo tracking and georeferencing. The conclusion of the study showed the technical feasibility of integrating information technology with tax tracking of cargo and vehicles, currently restricted, in this format, to the screening of vehicles. Possible solutions, supported legally, imply different timing, cost and accuracy of supplied information. However, there is still a long path to tread for this full integration and adoption.

A caracterização do sistema de transporte de carga na Região Metropolitana de São Paulo, apresentada neste trabalho, apoia-se nas diversas formas de organização das atividades e serviços de transporte de carga, pondo em evidência os novos espaços e fluxos que foram gerados nas últimas duas décadas. A questão da circulação e transporte de carga na Região Metropolitana de São Paulo e suas relações com as transformações na organização produtiva são abordadas, entendidas como aspectos importantes da dinâmica sócioespacial da metrópole paulistana. Admite-se que as transformações na organização produtiva da metrópole implicaram o crescimento dos fluxos de bens e mercadorias que, ao afetar a organização da circulação geral e dos serviços de transporte de carga, podem ter repercutido na organização espacial urbana. Tais transformações, também decorrentes de inovações na organização dos próprios serviços de transporte de carga, vêm sendo caracterizadas como atividade logística e visam ganhos globais no processo de produção. Assim sendo, alguns dos novos elementos do sistema de transportes de carga podem estar relacionados a processos sócio-espaciais característicos da formação metropolitana contemporânea, que se expressam em diferentes formas de distribuição espacial das atividades econômicas. Partindo-se desse pressuposto são descritos alguns aspectos da organização da produção e das atividades de comércio e serviços que têm resultado em mudanças no sistema urbano de cargas, argumentando-se que tais mudanças fazem parte das dinâmicas de transformação do espaço metropolitano e confirmam a força de polarização e articulação concentrada na RMSP. / The characterization of load transport system in Metropolitan Region of São Paulo, exposed in this work, holds on several ways of activities and duties organization for cargo transport, calling the attention to the new spaces and flows that were engendered on the last decades. The issue of cargo traffic and transport in São Paulo Metropolitan region and its relation with the transformation of the productive organization are broached, understood as important aspects of social spatial dynamic of São Paulo Metropolis. It is admitted that the transformation of productive organization of the metropolis implied in the increase of the goods and merchandises flows, that when affecting the organization of the general circulation and cargo transport duties, may have rebounded in urban spatial organization. Such transformations, also decurrent from innovations in load transport duties own organization, are becoming characterized as logistics activities and aim global profits in the production process. Thus, some of the new elements of the load transport system may be related to social spatial processes characteristic of the contemporary metropolitan formation, which express itself in different ways of spatial distribution of the economical activities. Starting from this presupposition some aspects of the production organization and of the trading and duties activities are described, which have turned out to changes in the urban load system, arguing that such changes constitute the dynamics transformations of the metropolitan spaces and confirm the polarization strength and the concentrated articulation in RMSP (São Paulo Metropolitan Region).

Emphasizes the importance of logistics in the road, because it is composed it is composed of between who supplies, distributes and uses, focusing also on processes, which begins in demand, with order processing, customer and product information, storage, transport, system back in their packaging and logistics process as all the flows of information that put the products or services on the move, the purpose of providing adequate service levels to customers more quickly and at reasonable costs. The logistics for the delivery of services defines the main activities and support, in which the main ones are: transportation, inventory maintenance and order processing. The focus in the transport activity is to be hanling both raw materials and the finished product and considered by many in the business life, due to its importance in logistics costs, absorbs about two thirds of these costs. Based on this premise, this study sought to point out the economic potential and the challenges encountered in the new air terminal in Goiânia, Goiás state, opened in the year 2016. In addition, addressed the relationship between logistics and the means of transport, particularly air transport, the importance of airport infrastructure efficiently to the dynamics of the market, as well as the relationship between the airport of Goiânia and regional development. It was verified that: even with the inauguration of the new terminal, there was no significant increase in the volume of cargo transported; the base of the economy in Goiás depends fundamentally on the road and rail modes, with the exception of pharmaceutical and automotive industries, which use the air service; and the proximity to Brasília hinders investment and development in the sector, as it is not economically viable to two poles nearby. / Analisar meios de transporte é fundamental para a compreensão acerca da formação dos grandes centros urbanos e o desenvolvimento econômico local, regional e nacional. Com base nessa premissa, o presente estudo buscou apontar as potencialidades econômicas e os desafios encontrados no novo terminal aéreo de Goiânia, estado de Goiás, inaugurado no ano de 2016. Diante disso, abordou-se a relação entre a logística e os meios de transporte, em especial o aéreo, a importância de uma infraestrutura aeroportuária eficiente para a dinâmica de mercado, bem como a relação entre o aeroporto de Goiânia e o desenvolvimento regional. Constatou-se que: mesmo com a inauguração do novo terminal, não houve aumento significativo no volume de cargas transportadas; a base da economia em Goiás depende fundamentalmente dos modais rodoviário e ferroviário, com exceção das indústrias farmacêutica e automobilística, que utilizam o serviço aéreo; e a proximidade com Brasília dificulta investimentos e desenvolvimento no setor, visto não ser economicamente viável dois polos próximos

Characterization and optimization of the in vitro motility assay for fundamental studies of myosin II

Myosin II is the molecular motor responsible for muscle contraction. It transforms the chemical energy in ATP into mechanical work while interacting with actin filaments in so called cross-bridge cycles. Myosin II or its proteolytic fragments e.g., heavy meromyosin (HMM) can be adsorbed to moderately hydrophobic surfaces in vitro, while maintaining their ability to translocate actin filaments. This enables observation of myosin-induced actin filament sliding in a microscope. This “in vitro motility assay” (IVMA) is readily used in fundamental studies of actomyosin, including studies of muscle contraction. The degree of correlation of the myosin II function in the IVMA with its function in muscle depends on how the myosin molecules are arranged on the surface. Therefore a multi-technique approach, including total internal reflection spectroscopy, fluorescence interference contrast microscopy and quartz crystal microbalance with dissipation, was applied to characterize the HMM surface configurations. Several configurations with varying distributions were identified depending on the surface property. The most favorable HMM configurations for actin binding were observed on moderately hydrophobic surfaces.   The effects on actomyosin function of different cargo sizes and amount of cargo loaded on an actin filament were also investigated. No difference in sliding velocities could be observed, independent of cargo size indicating that diffusional processive runs of myosin II along an actin filament are not crucial for actomyosin function in muscle. Furthermore, a tool for accurate velocity measurements appropriate for IVMAs at low [MgATP] was developed by utilizing the actin filament capping protein CapZ. These improvements allowed an investigation of the [MgATP]-velocity relationship to study possible processivity in fast skeletal muscle myosin II.  It is shown that the [MgATP]–velocity relationship is well described by a Michaelis-Menten hyperbola.  In addition, statistical cross-bridge modeling showed that the experimental results are in good agreement with recent findings of actomyosin cross-bridge properties, e.g., non-linear cross-bridge elasticity. However, no effect of inter-head cooperativity could be observed.   In conclusion, the described results have contributed to in-depth understanding of the actomyosin cross-bridge cycle in muscle contraction.

Service Level Management : Equalize unloadingencrustation at Stora Enso – Skoghalls bruk

The report is part of the university´engineer program within Industrial Organization and Economics with specialization in Logistics and Managment at Jönköping´s university. Skoghall´s mill underlies a large supply chain, which means there should be a lot of areas in need of small improvement measures. This report illustrates a small part of one of these areas and is focused on the loading distribution services in the terminal Vänern. At the moment there´s a very unequal load of the load carriers SECU box and Container at the terminal. Skoghall´s mill has the desire to equalize the high and low load levels of the 2 loading carriers named previously. One of the options to solve the problem is to set a limit of how much the carriers can load per week or per day. The mill also wants it to be investigated if the proposition would influence the customers in any way and if both the company and its customers could handle a lower level of delivery services in case the proposition was to be applied. The goal to be achieved with this thesis is to provide balance to the unequall loading of the 2 load carriers, SECU and Container, and investigate whether or not the Skoghall´s mill´s customers, could manage the lower degree of delivery service that the proposition would imply. For this goal to be achieved, random sample investigations were taken from orders of a customer that is provided with SECU load carriers and the same was made with a Container´s customer. The orders were analysed from production to consumption on the investigation. As a result of random sample investigations and customers´s evaluations, the thesis shows that it is possible to lower the resources at Vänerterminalen. This means that there is a full possibility to limitate the loadingprocess without causing any major effects on Stora Enso´s customers. Further investigation should be made by the company, such as random sample investigations of more customers, to generate a better picture of what the proposition would mean in reality. In case that investigation turn out to be not in accordance with the thesis, the authors worked on other two solution propositions. Both of them also would contribute to a more even torrent of unloading at Vänerterminalen.

Identifying these virulence components and how they help in establishing an environment conducive for bacterial growth is central to understanding chlamydial pathogenesis. This is experimentally challenging due to a lack of tools to perform molecular genetic studies. In the absence of genetic tools, we developed a yeast model system to identify and characterize chlamydial proteins involved in virulence mechanisms. In this study we describe the identification of twenty-eight proteins potentially involved in modulating host cellular functions and the secretion of virulence factors into the host. Since the delivery of virulence proteins by a type three secretion (T3S) system is a critical step for Chlamydia, we identified the proteins that interacted with the T3S apparatus by yeast two-hybrid analysis. We discovered several novel interactions between and determined that the C. trachomatis T3S apparatus displayed a similar architecture to that of other T3S systems. Furthermore with these approaches we identified networks of proteins that interacted with the secretion apparatus including a novel secretion chaperone protein. We characterized Ct260/Mcsc one of the putative secretion and demonstrated that it represents a novel class 1B secretion chaperone protein. Unlike other known chaperones, Mcsc directly interact with a conserved component of the T3S apparatus cytoplasmic domain, CdsQ. These finding represents a novel mechanism by which the secretion chaperone protein Ct260 may increase the secretion efficiency of its effector cargo and may reveal new facets of secretory cargo recognition by T3S systems.</p> / Dissertation
