Effects of the components of the Get pathway on prion propagation

Yeast prions e.g. [PSI+], [PIN+] and [URE3] are similar to mammalian amyloids that cause neurodegenerative diseases. [PSI+] is the aggregated self-perpetuating (prion) isoform of Sup35, a translation termination factor. The molecular chaperone Hsp104 plays a crucial role in the maintenance and propagation of [PSI+]. Deletion of the GET2 gene has been shown to cause a [PSI+] curing defect by excess Hsp104 and [PSI+] instability on synthetic medium (S. Muller, J. Patterson and Y. Chernoff, unpublished data; and J. Patterson Honors Thesis). Get2 is a membrane protein working in a complex with Get1 and Get3 proteins. This complex, called GET (Golgi-to-ER Traffic), is known to retrieve resident ER proteins from Golgi. In this particular study we provide further evidence for the connection between the GET pathway and yeast prions. The get2 deletion also leads to a detectable loss of [PIN+] prion on synthetic medium. The role of the other two members of the Get complex in prion propagation is also explored. The levels and the activity of Hsp104 in the get2 mutants is analyzed. The size of [PSI+] aggregates in the get2Δ strain is compared to that found in wild type. Finally, other possible mechanisms for the effect of get2 on prion maintenance and propagation are addressed.

Kry (“get”)-passives in Afrikaans : a minimalist analysis

Thesis (MA)--Stellenbosch University, 2013. / ENGLISH ABSTRACT: This study focuses on the grammatical properties and syntactic derivation of KRY (“get”)-passives in Afrikaans. Analyses of the corresponding phenomenon in other West-Germanic languages are examined and used as background for the study. The aim of the study is to provide a minimalist analysis of the internal structure of Afrikaans KRY-passives; no such analysis has to date been attempted within the framework of Minimalist Syntax. As an introduction to the phenomenon in Afrikaans, some grammatical background about kry and its syntactic distribution is provided, with special attention paid to the use of this verb in passive constructions. A new analysis of Afrikaans KRY-passives is put forward involving a small clause structure and a light verb component. The main hypothesis of the analysis is that eventive and stative KRY-passives are derived in basically the same manner, the important difference being the presence of a [process] and a [stative] feature, respectively, on the small clause light verb. This hypothesis provides support for the idea that the eventual eventive or stative interpretation of the structure is a consequence of the derivational process rather than being based on lexical features that are already present at the start of the derivation. / AFRIKAANSE OPSOMMING: Hierdie studie fokus op die grammatikale eienskappe en sintaktiese afleiding van KRY-passiewe in Afrikaans. Analises van die ooreenstemmende verskynsel in ander Wes-Germaanse tale word ondersoek en as vertrekpunt vir die studie gebruik. Die doel van die studie is om ‟n minimalistiese analise van die interne struktuur van Afrikaanse KRY-passiewe te ontwikkel; so ‟n analise is nog nie tevore binne die raamwerk van Minimalistiese Sintaksis aangebied nie. As algemene inleiding tot die verskynsel in Afrikaans, word enkele aspekte van kry se grammatikale eienskappe en sintaktiese verspreiding beskryf, met besondere klem op die gebruik van hierdie werkwoord in passiefkonstruksies. ‟n Nuwe analise van KRY-passiewe in Afrikaans word voorgestel, een wat gebruik maak van ‟n beknopte sin (“small clause”)-struktuur en ‟n ligte werkwoord (“light verb”)-komponent. Die vernaamste hipotese van die analise is dat gebeurtenis (“eventive”) en toestand (oftewel statief, “stative”) KRY-passiewe wesenlik op dieselfde manier afgelei word, met een belangrike verskil, naamlik die teenwoordigheid van, onderskeidelik, ‟n [proses]- en ‟n [statief]-kenmerk by die beknopte sin se ligte werkwoord. Dié hipotese verleen steun aan die idee dat die uiteindelike gebeurtenis- of toestand-interpretasie van die struktuur die gevolg is van die afleidingsproses, en dus nie gebaseer is op leksikale kenmerke wat reeds teenwoordig is by die aanvang van die afleiding nie.

This means that companies are looking for someone with similar psychological traits as entrepreneurs who are capable of influence positively in firm’s innovation. Therefore, the research question is 'What is the CEMS intrapreneurial level?' The contribution of this thesis is to check whether CEMS students have high intrapreneurship level or not. In order to find out, the General measure Enterprising Tendency (GET) test was sent to these students. The main finding is that CEMS students have score which can located them in the Medium range, meaning they have an average level of intrapreneurship. Thus, it is possible to raise awareness on the intrapreneurial potential that they can achieve. Furthermore, this thesis hopes to grow a concern on this topic so that universities give more emphasis on it.

An investigation of the relationship between bullying and physical activity at one high school in the Western Cape

Magister Educationis - Med / According to Morrison (2007), bullying is considered a critical issue in the Western Cape Schools as well as in South African schools in general. This phenomenon of bullying can affect learners in schools and may lead to several problems such as low esteem, poor academic performance, depression, absenteeism and eventually dropping out of school. In addition, numerous studies in psychology have revealed that physical activity has positive effects on learners, (Ajay, 2011; Shaffer, 2002). This is because it increases self-esteem in learners, boosts confidence and builds the physical, social, emotional, moral and intellectual composition in an individual. This study will aim (1) to determine the relationship between bullying and physical activity among South African learners in one high school in the Western Cape, (2) to measure the frequency of bullying at that high school, (3) to measure the frequency of bully-victimization in the school among South African learners in that high school in the Western Cape, and (4) to establish the level of physical activity by the learners in the high school. This study used a quantitative method and a descriptive survey as research design. The participants were mostly black South African adolescent learners, aged between 14 and 20 years.

En este proyecto se manifiesta una idea de negocio planteada por un grupo de estudiantes de la UPC. Los conocimientos aprendidos a lo largo de la carrera de cada uno han sido expuestos en el siguiente Trabajo. En primer lugar, hemos identificado el problema donde el usuario se ve en la necesidad de quedarse en casa tras el temor a la inseguridad ciudadana, en donde los altos índices de delincuencia han hecho que las personas opten por permanecer en casa o un lugar privado para pasar un buen momento. Por esta razón, creamos un producto para que las personas disfruten de un momento ameno sin exponerse. Siendo nuestro público objetivo personas de 18 a 60 años de edad de los NSE A - B de las zonas 7 y 8 de la ciudad limeña. Por esta razón, fundamos Tantay, una salsa de ají con finas hierbas 100% natural, elaborada con ingredientes de calidad de procedencia peruana y bajo un proceso artesanal, siendo una opción de acompañamiento para snacks u otros alimentos. Una combinación de sabores y texturas es lo que representa nuestra salsa, gracias a la amplia variedad de insumos nacionales que proporcionarán frescura y naturalidad al producto. Para lograr todo lo mencionado, hemos desarrollado un análisis donde se desarrollará planes estratégicos que abarquen Plan de Marketing, Plan operacional, Plan de Recursos Humanos, Plan de Responsabilidad Social y el Plan Financiero para poder evaluar la empresa y desarrollar el proyecto con éxito.

Hur ljud marknadsför en kommande långfilm : En multimodal analys av audiovisuella val i filmtrailers

Film har en stor efterfrågan och en sak som kan avgöra vilken film en person väljer att tittapå, är filmtrailern. Syftet med den här studien är att via multimodalitet analysera fyra olikafilmtrailers: Män som hatar kvinnor (2009) (svensk och amerikansk trailerversion), Get Out(2016) och Tenet (2019). Studien undersöker hur ljudelementen används inom utvaldafilmtrailers, för att se hur ljud och bild samverkar för att skapa ett intresse till att vilja se detillhörande långfilmerna. Analys visar att rytmisk bakgrundsmusik, ljudeffekter och dialogverkar ha stor betydelse i filmtrailern för Get Out (2016) och Tenet (2019), medan det somsticker ut för båda trailerversioner av Män som hatar kvinnor (2009) är avsaknaden av dialog.Studiens resultat visar att det inom skräckfilm verkar som att ljudeffekter har mer betydelseän genren kriminalfilm, men att det i actionfilm framför allt är rytmisk bakgrundsmusik somverkar utgöra det mest unika.

This research serves two main purposes: to create an analyzed edition of Ray Brown's bass lines from the Oscar Peterson Trio's 1964 recording We Get Requests, and to better understand Brown's lines through the lens of Ray Brown's Bass Method. This comparative analysis identifies significant events in the recorded music that closely relate to or resemble exercises from the book. By analyzing the music from the lens of Ray Brown's Bass Method, performers, students, and educators will gain a stronger understanding of the application of select technical devices provided by Brown in his book. The most prominent techniques discussed include scales and intervals, major triads, minor triads, and chords, exercises in tenths, rhythm patterns with drops, and diminished patterns. These evidence-based conclusions have significant applications in jazz bass pedagogy by revealing potential relationships between technical ideals and practical use. Although these conclusions may seem of concern only to jazz bassists, it should in fact concern anyone who cares about the connection between pedagogy and performance.

Les couples mariés qui ont des convictions religieuses fortes doivent respecter deux ordres juridiques parallèles: l’ordre étatique et l’ordre religieux. Des conflits peuvent surgir entre ces deux ordres, notamment lors du divorce des parties. Une brève présentation du droit québécois du mariage et du divorce ainsi que des droits religieux catholique, musulman et juif permet d’effectuer des comparaisons entre ces droits et d’identifier un certain nombre de points de jonctions normatifs. Des voies permettant d’harmoniser les deux univers normatifs sont explorées, dans une perspective de droit préventif: l’adaptation du contrat de mariage civil et du processus de la médiation familiale et la rédaction des ententes entre conjoints. / Married couples with strong religious beliefs must respect two parallel legal orders : the state order and the religious order. Conflicts can arise between these two orders, in particular when the parties divorce. A brief presentation of Quebec’s marriage and divorce laws and catholic, muslim and jewish religious law allows comparison between these systems of law and reveal some normative jonction points. In a preventive law perspective, ways to harmonize these two normative universes are explored : arrangement of the civil marriage contract, of the family mediation’s processus and of the writing of the agreements between partners.
