Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / Most of the features are displayed by complex organisms results of multifactorial inheritance, such as skin color in humans. This study aimed to investigate the high school students conceptions about multifactorial inheritance and, based on the information collected, develop educational materials that promote the application of genetic concepts in everyday matters. Participated in this study 48 students from two high school classes, enrolled in two public schools in Santa Maria / RS and São Vicente do Sul / RS. The survey was conducted in three stages called application of pre-test, didactic approach and implementation of the post-test. We develop a game, a didactic model and a hypertext is recommended for high school classes. The game and the teaching model was applied by the researcher and indicate their use for classes that have studied basics of monogenic and multifactorial inheritance. The applied materials were evaluated as motivation and use of genetic concepts. The evaluation by students and teachers indicated that the proposal is challenging and useful for genetic education with good applicability to review concepts. Hypertext was developed in three versions, according to the student's reality, being a material useful for situations where you have little time to teaching content to be a practical and easy handling means. To make it more interesting and interactive activity is recommended to be done beyond reading a discussion so that students and teachers can share ideas on the subject under study. Students from both groups showed an increase in the proportion of correct answers after the application of the didactic sequence, this improvement was most notable for the students in the class had a low rate of correct answers in the pre-test which had not studied the subject multifactorial inheritance ( class 2). / A maioria das características apresentadas pelos organismos complexos são resultados de heranças multifatoriais, como por exemplo, a cor da pele nos seres humanos. Este trabalho teve como objetivo investigar as concepções de alunos do Ensino Médio sobre herança multifatorial e, com base nas informações coletadas, desenvolver materiais didáticos que favoreçam as aplicações de conceitos de genética em assuntos do cotidiano. Participaram deste trabalho 48 alunos de duas turmas de Ensino Médio, matriculados em duas escolas públicas localizadas em Santa Maria/RS e São Vicente do Sul/RS. A pesquisa foi realizada em três etapas denominadas aplicação do pré-teste, abordagem didática e aplicação do pós-teste. Desenvolvemos um jogo, um modelo didático e um hipertexto sendo recomendados para turmas de Ensino Médio. O jogo e o modelo didático foram aplicados pela pesquisadora e indicamos a sua utilização para turmas que já estudaram conceitos básicos de herança monogênica e multifatorial. Os materiais aplicados foram avaliados conforme motivação e utilização de conceitos de genética. A avaliação por alunos e professores indicou que a proposta é desafiadora e útil para o ensino de genética, com boa aplicabilidade para revisar conceitos. O hipertexto foi desenvolvido em três versões, de acordo com a realidade do aluno, sendo um material útil para situações em que se tenha pouco tempo para o ensino do conteúdo por ser um meio prático e de fácil manipulação. Para tornar a atividade mais interessante e interativa recomenda-se que seja feita além da leitura uma discussão para que os alunos e professor possam compartilhar ideias sobre o tema em estudo. Os alunos de ambas as turmas apresentaram aumento na proporção de respostas corretas após a aplicação da sequência didática, essa melhora foi mais notável para os alunos da turma que teve baixo índice de acertos no pré-teste a qual não havia estudado o tema herança multifatorial (turma 2).

Vliv akné na každodenní život žáků vybrané střední školy / The impact of acne on everyday life of students in a selected high school

This diploma thesis entitled "The impact of acne on everyday life of students in a selected high school" is divided into two main parts. The first theoretical part is focused on the characteristics of the disease and on the impact of this disease on the quality of life of patients. There is a brief characterization of skin structure, it provides information about pathogenesis of acne and triggering factors, its clinical manifestations and clinical picture. In this part we can also find the diagnosis and treatment options for acne and an overview of other selected facial dermatoses. This part includes the effect of acne on patients' quality of life and an overview of the most common myths about acne. The second part is practically oriented. I used an online questionnaire to find out the impact of acne on everyday life of students in a selected high school. The results of my questionnaire survey show that 12 students (37,5 %) out of 32 respondents suffering from acne (100 %) reached the limit set by me, which determined whether their lives are affected by this disease or not. The first partial goal of the work was to determine the students' knowledge of this disease. Le créole afro-portugais de Casamance reste encore méconnu. La présente étude constitue la première description des pratiques langagières des créolophones casamançais. Elle est fondée sur un corpus de première main recueilli lors de discussions spontanées entre des locuteurs créolophones dans des villes multilingues : Dakar, Thiès et Ziguinchor. Dans ce corpus, les principales langues en contact sont essentiellement le créole casamançais, le français et le wolof. Cette thèse décrit les changements induits par ce contact dans le corpus, notamment le repérage temporel et les phénomènes touchant spécifiquement les groupes nominaux (déterminants et génitifs). L’étude de ces champs m’a permis de constater que des éléments grammaticaux et lexicaux sont fournis à la fois par l’ensemble de ces langues. Ce cas de figure n'est pas souvent pris en compte dans les approches théoriques du contact de langues, lesquelles proposent souvent une séparation fonctionnelle des langues fondée sur une dichotomie entre langue matrice et langue insérée, et tendent à ignorer les situations de contact impliquant plus de deux langues. Cette thèse constitue une contribution à l’étude du contact de langues et permet en particulier de porter un regard neuf sur une situation de contact trilingue, impliquant une langue créole et deux autres langues qui en sont typologiquement éloignées. La prise en compte d’une telle configuration revêt un caractère particulièrement novateur dans le domaine des études créoles, où les chercheurs intéressés par le contact de langues se concentrent surtout sur des situations de contact entre les créoles et leurs langues lexificatrices respectives. / Casamancese Afro-Portuguese Creole is still largely unknown. The study of these domains has led me to realize that all three languages involved both grammatical and lexical elements are provided by all these languages.Such a scenario is generally not taken into consideration in the theoretical approaches to language contact, which often posit a functional separation of the languages involved based on a dichotomy between matrix language and embedded language, and tend to ignore contact situations involving more than two languages.This dissertation is therefore a contribution to the study of language contact; more specifically, it allows for the possibility to take a fresh look at a trilingual contact situation involving a Creole language and two other languages typologically distant from the former. The inclusion of such a configuration is particularly innovative in the domain of Creole studies, where researchers interested in contact languages focus mainly on situations of contact between Creoles and their respective lexifier languages

O Transtorno de Oposição e Desafio (TOD) é definido por um padrão recorrente de comportamento desafiante, desobediente e hostil com início na infância e adolescência e caracteriza-se por uma alta taxa de comorbidades. Estudos longitudinais apontam o TOD na infância como um dos principais preditores de psicopatologia na idade adulta. Uma possível explicação para a grande heterogeneidade de comorbidades e trajetórias longitudinais é de que o diagnóstico de TOD abrange distintas dimensões de sintomas, cada qual com seu desfecho. O primeiro objetivo desta tese foi a validação das distintas dimensões do TOD em uma amostra comunitária Brasileira composta de 2512 sujeitos. Através de análise fatorial confirmatória, demonstramos que o modelo que melhor representa a heterogeneidade do TOD é composto por três dimensões: a dimensão \"argumentative/defiant\" que está associada com transtorno de déficit de atenção/hiperatividade (TDAH); a dimensão \"vindictiveness\" que possui associação com transtorno de conduta (TC); e a dimensão \"angry/irritable mood\" onde predominam as associações com transtornos depressivos e de ansiedade. O objetivo seguinte foi investigar o papel da dimensão irritável na classificação nosológica dos transtornos mentais na infância e adolescência. A apresentação da irritabilidade é um aspecto crucial: irritabilidade crônica caracterizada por baixa tolerância à frustração e frequentes explosões de raiva, que é distinta da apresentação episódica, associada ao diagnóstico Transtorno de Humor Bipolar (TB). \"Severe mood dysregulation\", \"disruptive mood dysregulation disorder\", ou dimensão irritável do TOD são formas distintas de classificar o fenótipo de irritabilidade crônica. Entretanto, independente da classificação utilizada, a alta taxa de comorbidades é invariavelmente o denominador comum em estudos sobre irritabilidade. Neste sentido, examinamos o impacto da irritabilidade como uma dimensão subjacente a vários transtornos. Para tanto, avaliamos o impacto da dimensão irritável do TOD através de vários cenários: indivíduos sem diagnóstico, indivíduos com TDAH e sujeitos com transtornos emocionais. Esta 9 investigação foi realizada em duas amostras, uma brasileira constituída por 2.512 sujeitos e uma amostra britânica composta de 7.977 sujeitos. Os resultados demonstram que a irritabilidade está associada ao aumento do prejuízo funcional independente do diagnóstico comórbido concomitante. Seguindo esta linha, investigamos a influência genética na etiologia da irritabilidade. Para tanto, criamos um escore poligênico que incluiu polimorfismos associados à baixa tolerância à frustração, raiva, agressividade reativa e labilidade emocional. O escore poligênico foi altamente preditivo dos níveis de irritabilidade em 350 sujeitos da amostra brasileira. A associação foi específica para irritabilidade e não foi significativa para TDAH, TOD ou medidas contínuas de sintomas. Além disso, a influência genética se manteve mesmo quando fatores ambientais foram incluídos no modelo estatístico. Por fim, quando testada em diferentes ambientes, a influência genética na etiologia da irritabilidade foi mais importante em ambientes de alto risco, sugerindo uma correlação gene-ambiente (rGE). Concluindo, nossos resultados sugerem que a irritabilidade se caracteriza como um traço dimensional subjacente a inúmeros transtornos na infância e adolescência e agregando impacto e prejuízo funcional. Longitudinal studies support ODD as a predictor of psychopathology in adulthood. A potential explanation for such heterogeneity of comorbidities and longitudinal trajectories is that ODD diagnosis encompasses distinct clusters of symptoms, each with its outcome. The first aim of this work was the validation of ODD dimensions in a Brazilian community sample of 2512 subjects. Confirmatory factorial analysis showed that the best model for ODD comprised three dimensions: an \"argumentative/defiant\" dimension, which associates with attention deficit/hyperactivity disorder (ADHD); a \"vindictiveness\" dimension, which associates with conduct disorder (CD); and an \"angry/irritable\" dimension where emotional disorders such as depression and anxiety are the most common associations. The next step was the investigation of the role of the irritable dimension of oppositionality in diagnostic classifications of childhood mental disorders. The pattern of irritability is a crucial point: its chronic presentation as easy annoyance and frequent temper outbursts should be differentiated from the episodic course of irritability associated with the specific diagnosis of Bipolar Disorder (BD). \"Severe mood dysregulation\", \"disruptive mood dysregulation disorder\", and the irritable dimension of oppositionality are different ways to classify the chronic irritability phenotype. However, regardless of the classification, the high rate of comorbidities is invariably the common denominator in studies of irritability. Therefore, we examined the impact of irritability as a dimension cutting across multiple settings: individuals without any diagnosis, subjects with ADHD, and also those with emotional disorders. For that we used two samples, one from Brazil, with 2.512 subjects, and one from the UK, with 7.977 individuals. Results showed that irritability associates with increased functional impairment regardless of concurrent comorbid status. We then investigated the genetic influence on the etiology of irritability. A polygenic score was generated encompassing polymorphisms previously associated with anger, emotional lability and reactive aggression. The polygenic score significantly predicted irritability in 350 subjects in the brazilian sample, yet failed to predict ADHD, ODD, CD and continuous measures of symptoms. Moreover, the association between the polygenic score and irritability remained significant even after taking into account environmental factors. Finally, when stratified across diverse levels of environmental risk, genetic influence upon the etiology of irritability appears to be stronger in high-risk environments. Taken together, our results suggest that irritability is characterized as a dimensional trait that underlies multiple disorders, adding functional impairment. Thus, the construct of irritability fits well within the concept of Research Domain Criteria (RDoC) that suggests that mental disorders should be understood through dimensions underlying diagnostic categories

Case variation in nominative object constructions in the history of Russian

Diese Arbeit legt eine diachrone korpuslinguistische Untersuchung der Kasusvariation in sog. Nominativobjekt-Konstruktion in der Geschichte der russischen Sprache. Bei der Nominativobjekt-Markierung handelt es sich um eines der in der Geschichte der russischen Sprache erscheinenden syntaktischen Merkmale, das auch in mehreren ostslawischen und ostbaltischen Dialekten sowie den westfinnischen Sprachen zu finden ist. Diese Art von Konstruktion lässt sich in der frühen russischen Schriftsprache bis zum 12. Jahrhundert nachweisen und wurde allgemein, wenn auch nicht durchgehend, in der russischen Kanzleisprache des 16. und 17. Jahrhunderts verwendet, wonach sie aus der Schriftsprache verschwand und z.T. nur noch in heutigen nordrussischen Dialekten mundartlich wurde. Ziel der Analyse ist es das Auftreten des Nominativs im Vergleich zum ebenfalls auftretenden und (aus der Perspektive des heutigen Standradrussischen) erwartbaren Akkusativ anhand von korpuslinguistische Studie zu beschreiben und zu erklären. Die Arbeit liefert eine multifaktorielle diachronische Analyse von sprachlichen und außersprachlichen Faktoren (wie Dialekt- oder Registervariation), die diese Kasusvariation auslösen, um die Strategien der Kasuswahl und mögliche Mikroverschiebungen zu rekonstruieren. Ein zentrales Ergebnis der Studie ist, dass bereits in den ältesten Dokumenten beide Konstruktionen mit einem Nominativ und einem Akkusativ zu finden sind, die auch noch nicht als völlig austauschbare Varianten in der Sprache existierten. Außerdem konnte gezeigt werden, dass die Kasuswahl im Altrussischen nicht rein syntaktisch geregelt wurde, wie es im Mittelrussischen oft der Fall war. Einige oben genannte Fakten erlauben es zwei ursprünglich voneinander unabhängige Konstruktionen anzunehmen, die jedoch unterschiedliche Initialkonstruktionen hatten und deswegen die Überlappungsfälle ermöglichten. / This work presents a diachronic study on case variation in so called “nominative object constructions” attested in Old and Middle Russian, and in some modern North Russian dialects. The origin of those type of constructions in Russian and the syntactic status of the argument in nominative has been widely discussed and different explanations have been suggested in various historical and typological works on this phenomenon. In this study, I account for the differences in the paralleled use of nominative and accusative object constructions. This interchangeability is often explained as complementary use (distribution) of two variants of one construction or as a simply stylistic variation, but in fact, this is the expression of two independent competing rules (in means of “competing grammar approach”). The goal of this study is to present the results of a first extensive quantitative corpus-based analysis on theoretical considerations about development and micro-changes in these types of constructions with special focus on the different factors determining the case choice. It will be shown in this study that constructional change at the morphological level does not happen in complete isolation from developments at other levels. In addition, each constructional change can have repercussions on other constructions. In the case of nominative object construction and the case variation with accusative, a variety of linguistic and non-linguisitic factors can be assumed to interact. Hence, the diachronic change of different constructional variants can also be interpreted as a case of functional re-organization in that non-canonical object marking constructions.

La demande mondiale de nourriture et d’energie est en constante augmentation. L’accès à de nouvelles terres arables étant limité, les agriculteurs devront produire environ 70% de plus par hectare d'ici 2050. L'optimisation et la rationalisation des systèmes de production agricole sont essentielles pour assurer la sécurité alimentaire des populations dans des conditions durables. La communauté agricole serait en mesure d'optimiser les performances des cultures en ayant accès à une meilleure compréhension des systèmes de production, notamment concernant leur potentiel et principaux facteurs limitants. Dans ce contexte, la caractérisation des conditions de croissance des cultures prenant en compte leurs impacts sur le potentiel de production et sur les pertes de rendement est importante pour identifier les marges de progrès et proposer des systèmes agricoles améliorés.Dans cette étude une approche est proposée pour caractériser l'environnement de grandes zones géographiques prenant en compte les impacts des ressources clés au développement des plantes (température, rayonnement, eau et azote) sur la performance des cultures. Le niveau de disponibilité de ces ressources a été estimé en utilisant des approches de modélisation de culture, intégrant données météorologiques et caractéristiques de sol. L'efficacité d'utilisation des ressources disponibles a été caractérisée par les pratiques culturales ainsi que la stratégie et l’environnement technique des agriculteurs. Une procédure générique d’intégration de la donnée a été établie et utilisée pour décrire la production de maïs grain en Amérique du Nord de 1991 à 2013. Le jeu de données résultant de cette intégration couvre 84% des superficies plantées en maïs, décrites par 1 558 unités spatiales élémentaires, pour un total de 28 303 scénarios indépendants. Cette méthode combine et harmonise les observations de rendement issues des statistiques agricoles avec un large ensemble de descripteurs pertinents de conditions de croissance.Un sous-ensemble de 21 facteurs limitants a été identifié par une analyse de sélection de variables pour expliquer 66% de la variabilité des pertes de rendement observées. Compte tenu du nombre de dimensions du jeu de donnée, la méthode relaxed lasso a été choisie comme un compromis intéressant entre capacité de prédiction et d’interprétation. Les descripteurs sélectionnés montrent la contribution de la gestion des cultures à la variabilité des pertes de rendement, en particulier les niveaux d'intensification de l’agriculture et l’environnement technique des agriculteurs. Le troisième facteur principal est la disponibilité en eau et la sécheresse qui en résulte. L’utilisation des connaissances en physiologie dans la conception de descripteurs a considérablement amélioré la capacité d’interprétation de la proposition et la confiance des utilisateurs finaux dans l'approche.La structure des facteurs limitant le rendement a été utilisée pour identifier les Environnements Types les plus fréquents. Deux approches ont été proposes : l’une axée uniquement sur les facteurs physiologiques et l’autre intégrant également l’identification de marchés de taille homogène. 11 Environnements Types ont été identifiés en fonction des facteurs physiologiques et 8 en combinant facteurs physiologiques et contraintes de taille de marché. Les Environnements Types basés sur les facteurs physiologiques sont très informatifs sur l'évolution historique des pratiques culturales et les changements de stratégie des agriculteurs au cours du temps. Une telle catégorisation des conditions de croissance a montré des capacités à reproduire l'expertise de terrain et à soutenir l'évaluation de génotypes. Trois cas d'utilisation ont servi à illustrer l'intérêt de l'approche pour (i) décrire l'historique environnemental d'un marché, (ii) définir des populations d’Environnements cibles (TPE) et (iii) évaluer la pertinence de l'échantillonnage environnemental de réseaux multi-lieux (MET). / The global demand for food and energy is constantly increasing. As access to spare arable land is limited, growers will have to produce about 70% more per hectare by 2050. Optimization and rationalization of the agricultural production systems is then critical to ensure food security under sustainable conditions. The agricultural community would be in a position to optimize crop performance by better understanding the cropping systems, especially regarding potential production and limiting factors. In this context, the ability to characterize crop growing conditions in regards to their impacts on potential yield and yield gap is important in order to identify margins of progress and propose improved farming systems.An approach is proposed to characterize the crop environment of large geographical areas taking into consideration the impact on crop performance of the key resources for plant development (temperature, solar radiation, water and nitrogen). The level of availability of each resource was estimated by using crop modeling approaches integrating weather data and soil characteristics as inputs. The efficiency of use of the available resources was characterized by crop practices, grower strategy and grower technical environment. A generic integration procedure was established and used to describe corn production for grain in North America from 1991 to 2013. The resulting dataset covers 84% of the total corn planted area, deciphered in 1,558 elementary spatial units, for a total of 28,303 independent scenarios. Such a method combines and harmonizes, at scale, yield observations from agricultural statistics with a large set of relevant descriptors of growing conditions.A subset of 21 main limiting factors was identified through variable-selection analysis to explain 66% of the observed yield-gap variability. The relaxed lasso method resulted in an interesting compromise between interpretability and prediction ability. The selected descriptors highlighted the contribution of crop management in yield-gap variability, especially regarding levels of crop intensification and the technical environment of growers. The third main factor is water availability and resulting drought. The integration of knowledge in crop physiology into descriptor design significantly improved the interpretability of the proposal and the confidence of end-users in the approach.Yield-gap patterns were used to identify the set of most frequent Environment Types. Two approaches were proposed: one focused on crop physiology drivers and another also considered market-size homogeneity among Environment Types. This resulted in 11 Environment Types based on crop physiology drivers and 8 Environment Types when combining crop physiology drivers and market constraints. Crop-physiology Environment Types are very informative on the historical evolution of cultural practices and the changes in grower strategies over the studied period. Such categorization of growing conditions demonstrated the ability to reproduce field expertise and support genotype evaluation. Three business Use Cases were used to illustrate the interest of the approach in (i) describing the environmental history of a market, (ii) defining relevant Target Population Environments (TPE) and (iii) evaluating the environmental sampling relevance of Multi-Environment Trials (MET).

Tipificação do HLA nos fenótipos alérgico e não alérgico da asma / HLA typing in allergic and non-allergic asthma phenotypes

A asma é uma doença heterogênea caracterizada por um processo inflamatório crônico das vias aéreas inferiores que está associado ao desenvolvimento da hiperresponsividade brônquica e remodelamento da via aérea. Atualmente, a asma é considerada uma síndrome, ou ao menos uma doença com diversos fenótipos. Tradicionalmente, dois fenótipos são bem definidos pela clínica e exames subsidiários: asma alérgica e asma não alérgica. Eles são diferentes quanto á idade de início, apresentação clínica, história pessoal e familiar de atopia e resposta ao tratamento. Ao contrário da asma alérgica, cuja fisiopatologia está bem caracterizada, a etiologia e mecanismos envolvidos na asma não alérgica não estão bem elucidados. Algumas possibilidades incluem alergia desencadeada por antígenos desconhecidos (fungos), infecção persistente (Chlamydia trachomatis, Mycoplasma sp) e auto-imunidade. Estudos têm descrito em diferentes populações associações entre a asma e alelos/antígenos HLA classe I e II, mas os resultados têm sido inconclusivos. O objetivo deste estudo foi identificar possíveis associações do antígeno leucocitário humano (HLA) classe I (A, B, C) e II (DR, DQ, DP) em pacientes brasileiros com asma alérgica e não alérgica. Um total de 109 pacientes com o diagnóstico de asma (56 com asma alérgica e 53 com asma não alérgica) que estavam em acompanhamento no Serviço de Imunologia Clínica e Alergia do Hospital das Clínicas da Faculdade de Medicina da Universidade de São Paulo, e 297 controles (doadores falecidos de órgãos sólidos) tiveram seu sistema HLA classe I (A, B e C) e II (DR, DQ e DP) tipificado. Os pacientes também realizaram espirometria e coletaram sangue para a quantificação da imunoglobulina E (IgE) sérica total e nível sérico de eosinófilos. Além disso, foram avaliados quanto à IgE específica para aeroalergenos através do teste cutâneo de puntura e a pesquisa da IgE sérica específica (ImmunoCAP). O grupo com asma alérgica foi constituído por pacientes que apresentavam resultado positivo para a pesquisa da IgE específica em ambos teste cutâneo de puntura e na investigação in vitro. E o grupo com asma não alérgica apresentava resultados negativos nos dois testes. A comparação do HLA classe I nos grupos estudados identificou frequência significativamente maior do HLA-B*42 e HLA-C*17 no grupo com asma alérgica, enquanto o HLA-B*48 estava estatisticamente associado com o fenótipo não alérgico. Na análise do HLA classe II, o HLA-DPA1*03 e HLA-DPB1*105 apresentou associação com os pacientes com asma alérgica. Concluindo, o estudo observou diferentes associações dos alelos HLA classe I e II com asma alérgica e não alérgica na população brasileira, a qual é caracterizada pela diversidade de origens e miscigenação. Porém, a predisposição genética para asma é poligênica e novos estudos em grandes populações são necessários para confirmar a associação do HLA como fator protetor ou causador da doença / Asthma is a heterogeneous chronic inflammatory disease of lower airways associated with the development of bronchial hyperresponsiveness and airway remodeling. Currently, asthma is regarded as a syndrome or at least a disease with several phenotypes.Traditionally, two phenotypes of asthma have been defined according to clinical and laboratory features: allergic and non-allergic asthma. Each of them has distint age of onset, clinical presentation, personal and family history of allergy and response to therapy. In contrast to allergic asthma, which pathophysiology is well characterized, the etiology and mechanisms involved in non-allergic asthma remain unclear. Some possibilities include allergy triggered by unknow antigens (fungi), persistent infection (Chlamydia trachomatis, Mycoplasma sp) and autoimmunity. Studies have reported associations between asthma and HLA class I and II alleles/antigens in different populations, but the results have been inconclusive. The objective of this study was to identify possible associations of the human leukocyte antigens (HLA) class I (A, B and C) and II (DR, DQ and DP) in Brazilian patients with allergic and non-allergic asthma. A total of 109 patients with asthma (56 with allergic asthma and 53 with non-allergic asthma), who were being followed at the Service of Clinical Immunology and Allergy of the Hospital das Clínicas of the University of São Paulo Medical School, and 297 controls (deceased solid organ donors) had their HLA class I (A,B and C) and II (DR, DQ and DP) typing. Patients performed spirometry and had their blood drawn to measure total serum immunoglobulin E (IgE) levels and eosinophil count. Furthermore, they were assessed for specific IgE to aeroallergens with skin prick test and serum tests (ImmunoCAP). The allergic asthma group was composed of patient presenting positive results for specific IgE in both skin prick test and in vitro assay. And the non-allergic asthma group had negative results in both tests. There were significantly higher frequencies of HLA-B*42 and HLA-C*17 in the allergic asthma group, whereas the HLA-B*48 was associated with the non-allergic group. Regarding HLA class II analysis, HLA-DPA1*03 and HLA DPB1*105 were associated with allergic asthma patients. In conclusion, the study identified different associations of HLA class I and II with allergic and non-allergic asthma in the Brazilian population, which is characterized by diversity of origins and miscegenation. However, the genetic predisposition of asthma is polygenic and new studies on large populations are needed to confirm the role of HLA as a protective or predisposing factor of disease

Les micelles de caséines, composées de caséines, minéraux et eau, sont en équilibres dynamiques, elles échangent en permanence de leur matière avec leur environnement. Les micelles de caséines possèdent d’intéressantes fonctionnalités pour la fabrication de produits laitiers, telles que leur capacité à stabiliser des émulsions et à former des gels sous l’action de la présure. Des changements environnementaux, variations de pH, additions de sels ou d’agents chélatants, affectent les équilibres des micelles et induisent des modifications de leurs compositions et propriétés colloïdales. Ces changements modifient également leurs propriétés fonctionnelles, bien que cet aspect soit peu décri. Le but de ce projet était de comprendre les relations liant l’environnement, l’équilibre minéral, les propriétés colloïdales et fonctionnelles des micelles de caséines. L’impact de cinq facteurs (pH, Na3Cit, NaCl, CaCl2, MgCl2) modifiant les micelles fut étudié en focalisant sur leurs propriétés émulsifL’acidification et l’addition de Na3Cit ont causé les modifications minérales les plus importantes en solubilisant le phosphate de calcium micellaire. Le premier conduisit au « gonflement » des micelles alors que l’agent chélatant causa leur dissociation en petits agrégats. L’ajout de NaCl n’eut aucun impact sur le contenu minéral des micelles mais provoqua aussi la libération d’agrégats, révélée par cryo microscopie électronique et diffusion de rayons-X aux petits angles. Le rôle du phosphate de calcium sur les fonctionnalités fut confirmé et l’étude révéla l’importance de contrôler la libé / Caseins micelles, composed of caseins, minerals and water, are under dynamic equilibria, they constantly exchange materials with their surrounding environments. In addition, casein micelles possess valuable functionalities in regards to the formation of dairy products, such as the ability to stabilize emulsions or to form rennet gels. Environmental changes, such as variations in pH, additions of salts or chelating agents, affect the casein micelles equilibria and lead to modifications in their compositional and colloidal properties. Such changes also modify their functional properties, although this aspect is poorly described in the literature. This project aimed to understand the relationships that link the environmental modifications, the mineral balance, the colloidal and functional properties of the casein micelles. The impact of five modifying factors (pH, Na3Cit, NaCl, CaCl2, MgCl2) of the casein micelles were studied and the focus was placed on emulsion and rennet coagulation fuThe former only induce the swelling of casein micelles while the latter led to their disruption into smaller aggregates. NaCl had no impact on the micellar mineral content but also caused the release of small aggregates, as revealed by electronic microscopy and small angle X-ray scattering analyses. The decisive role of micellar calcium phosphate on the functionalities was confirmed and this study highlighted the importance of monitoring the release of small aggregates, as they strongly affected emulsions stability and gels firmness.
