Situated social cognition

In der vorliegenden Dissertation werden vier Studien vorgestellt, in denen untersucht wurde, wie altrozentrische (Mimikry) und egozentrische (Selbstprojektion) Prozesse der sozialen Kognition in Abhängigkeit vom sozialen Kontext und persönlichen Dispositionen reguliert werden. Studie 1 zeigte, dass die Tendenz, fröhliche Gesichtsausdrücke anderer nachzuahmen abhängig von dem mit der beobachteten Person assoziierten Belohnungswert ist. Die Auswirkung der Belohnung ging jedoch weder in die vorhergesagte Richtung, noch konnten wir einen Einfluss von Oxytocin, einem Hormon, das der Neurobiologie der sozialen Anpassung zugrunde liegt, finden. Studie 2 zeigte, im Vergleich zu vorherigen Studien, keine allgemeine Verbesserung der automatischen Nachahmung nach direktem Blickkontakt im Vergleich zum abgewandten Blick. Wir konnten jedoch potenzielle dispositionelle Faktoren (z.B. autistische Eigenschaften) identifizieren, denen unterschiedlichen Mimikry-Reaktionen auf den Blickkontakt zugrunde liegen könnten. Studie 3 kombinierte kurze Phasen der Emotionsinduktion mit psychophysischen Messungen der Emotionswahrnehmung. Es zeigte sich, dass emotionale Gesichtsausdrücke tendenziell als fröhlicher beurteilt werden, wenn Personen angeben, dass sie sich fröhlich im Vergleich zu traurig fühlen. Emotionale egozentrische Verzerrungen wurden in Studie 4 erneut untersucht. Im Gegensatz zu unseren Vorhersagen fanden wir jedoch keine stärkeren egozentrischen Verzerrungen, wenn die Teilnehmenden emotionale Gesichtsausdrücke von ähnlichen im Vergleich zu unähnlichen Personen beurteilten. In allen Studien fanden wir Hinweise für den kontextabhängigen Charakter der sozialen Kognition. Allerdings konnten wir einige der in der Literatur berichteten Phänomene nicht replizieren. Diese Ergebnisse unterstreichen die Notwendigkeit, die Robustheit und Generalisierbarkeit früherer Befunde systematisch neu zu bewerten. / This dissertation presents four studies that investigated how altercentric (mimicry) and egocentric (self-projection) processes of social cognition are regulated according to the social context and personal dispositions. Study 1 showed that the tendency to mimic others’ happy facial expressions depends on the reward value associated with the observed agent. However, the effects of reward were not in the hypothesised direction, nor could we detect an influence of oxytocin treatment, a hormone involved in the neurobiology of social adaptation. Study 2 could not detect a general enhancement of the tendency to automatically imitate others’ hand actions following direct gaze compared to averted gaze, in contrast to previous studies. However, we could identify dispositional factors (e.g., autistic traits) that might underlie different mimicry responses to gaze cues. Combining brief emotion induction blocks with psychophysical measures of emotion perception, Study 3 showed that facial emotional expressions tend to be judged as happier when individuals feel happy than when they feel sad. Emotional egocentric biases were replicated in Study 4. But contrary to our predictions, we did not find stronger egocentric biases when participants judged emotional facial expressions of similar compared to dissimilar others. Across all studies, we found evidence supporting the contextual nature of social cognition. However, we could not replicate some of the phenomena reported in the literature. These results highlight the need to systematically re-evaluate the robustness and generalizability of prior findings.

Modulation of Oxytocin Receptors in Right Ventricular Hypertrophy

L’hypertension pulmonaire (HP) est une maladie dont l’étiologie est inconnue et qui entraîne ultimement une défaillance du ventricule droit (VD) et le décès. L’HP peut être induite chez le rat par la la monocrotaline (MCT), un alcaloïde pyrrolizidique extrait de la plante Crotalaria Spectabilis, causant des lésions à l’endothélium des artères pulmonaires, menant à un épaississement de ces dernières et à une augmentation de la résistance vasculaire. Ceci à pour conséquence de causer une hypertrophie du VD, de l’inflammation, une dysfonction endothéliale NO-dépendante des artères coronariennes et une augmentation des peptides natriurétiques circulants. Objectif: Nous avons testé l’hypothèse selon laquelle l’étiopathologie de l’HP impliquerait le récepteur à ocytocine (OTR) dû à son implication fonctionnelle avec les cytokines inflammatoires et la libération du peptide natriurétique atrial (ANP) et du NO. Méthodes: Des rats mâles Sprague-Dawley pesant 220-250g reçurent une seule injection sous-cutanée de MCT (60 mg/kg). 6 à 7 semaines (46±1 jours) suivant l’injection, les rats furent sacrifiés et l’expression génique et protéique fut déterminée par PCR en temps réel et par western blot, respectivement, dans le VD et le ventricule gauche (VG) Résultats: Les rats traités au MCT démontrèrent une augmentation significative du VD. Une hypertrophie du VD était évidente puisque le ratio du VD sur le VG ainsi que le poids du septum étaient près de 77% plus élevés chez les rats traités au MCT que chez les rats contrôles. Le traitement au MCT augmenta l’expression génique d’ANP (3.7-fois dans le VG et 8-fois dans le VD) ainisi que le NP du cerveau (2.7-fois dans le VG et 10-fois dans le VD). Les transcrits de trois récepteurs de NP augmentèrent significativement (0.3-2 fois) seulement dans le VD. L’expression protéique de la NO synthase (iNOS) fut également augmentée de façon sélective dans le VD. Par contre, les transcripts de NOS endothéliale et de NOS neuronale étaient plus élevés (0.5-2 fold) dans le VG. L’ARNm et l’expression protéique d’OTR furent diminués de 50% dans le VD, tandis qu’une augmentation de l’expression des cytokines IL-1β and IL-6 fut observée. L’ARNm de Nab1, un marqueur d’hypertrophie pathologique, fut augmentée de deux-fois dans le VD. Conclusion: L’augmentation d’expression génique de NP dans le VD des rats traités au MCT est associée à une augmentation des transcripts du récepteur NP, suggérant une action locale de NP dans le VD durant l’HP. L’expression d’OTR est atténuée dans le VD, possiblement par des cytokines inflammatoires puisque le promoteur du gène de l’OTR contient de multiples éléments de réponse aux interleukines. Diminuer l’expression d’OTR dans le VD durant l’hypertension pulmonaire pourrait influencer de manière positive la fonction cardiaque car l’OTR régule la contractilité et le rythme cardiaque. PH can be induced in the rat with monocrotaline (MCT), a pyrrolizidine alkaloid from the plant Crotolaria spectabilis that damages the pulmonary artery endothelium leading to thickening of the pulmonary arteries and increased vascular resistance. This subsequently results in RV hypertrophy, inflammation, nitric oxide (NO)-associated coronary endothelial dysfunction and increment of natriuretic peptides (NP) in the circulation. Objective: We verified hypothesis that the etiopathogenesis of PH involves the oxytocin receptor (OTR) because of its functional association with inflammatory cytokines and release of atrial natriuretic peptide (ANP) and NO. Methods: Male Sprague-Dawley rats weighing 220-250g received a single subcutaneous injection of 60 mg/kg of MCT. Six to 7 weeks (46±1 days) following the injection, rats were sacrificed and gene and protein expression were detected by real-time PCR and western-blot analysis, respectively, in the RV and LV (left ventricle). Results: MCT-treated rats displayed significant increases in RV weight. RV hypertrophy was evident as the ratio of the RV to LV plus septum weight was nearly 77% higher in MCT-treated rats compared to control rats. MCT treatment increased transcripts of ANP (3.7-fold in the LV and 8-fold in RV) and brain NP (2.7-fold in the LV and 10-fold in RV). Transcripts for three NP receptors significantly increased (0.3-2 fold) only in the RV. iNOS (inducible NO synthase) protein expression also increased selectively in the RV. In contrast, the endothelial NOS and neural NOS transcripts heightened (0.5-2 fold) in the LV. Both OTR mRNA and protein were decreased by 50% in the RV, whereas an up-regulation of cytokines IL-1β and IL-6 was observed. Nab1 mRNA, a marker of pathological hypertrophy, increased two-fold in the RV. Conclusion: Increased gene expression of NP in the RV of the MCT-treated rat correlates with upregulation of NP receptor transcripts indicating local NP action in the RV during PH. OTR expression is decreased in the RV possibly by inflammatory cytokines, IL-1 and IL-6 because OTR promoter region contains multiple putative interleukin-response elements. Lowering OTR in RV during pulmonary hypertension can influence cardiac function since OT regulates heart rate and cardiac contractility and is linked with cardioprotective system ANP and NO. Evaluation of oxytocin pharmacokinetic : pharmacodynamic profile and establishment of its cardiomyogenic potential in swine

La thérapie cellulaire est une avenue pleine de promesses pour la régénération myocardique, par le remplacement du tissu nécrosé, ou en prévenant l'apoptose du myocarde survivant, ou encore par l'amélioration de la néovascularisation. Les cellules souches de la moelle osseuse (CSMO) expriment des marqueurs cardiaques in vitro quand elles sont exposées à des inducteurs. Pour cette raison, elles ont été utilisées dans la thérapie cellulaire de l'infarctus au myocarde dans des études pre-cliniques et cliniques. Récemment, il a été soulevé de possibles effets bénéfiques de l'ocytocine (OT) lors d’infarctus. Ainsi, l’OT est un inducteur de différenciation cardiaque des cellules souches embryonnaires, et cette différenciation est véhiculée par la voie de signalisation du monoxyde d’azote (NO)-guanylyl cyclase soluble. Toutefois, des données pharmacocinétiques de l’OT lui attribue un profil non linéaire et celui-ci pourrait expliquer les effets pharmacodynamiques controversés, rapportés dans la lttérature. Les objectifs de ce programme doctoral étaient les suivants : 1) Caractériser le profil pharmacocinétique de différents schémas posologiques d'OT chez le porc, en développant une modélisation pharmacocinétique / pharmacodynamique plus adaptée à intégrer les effets biologiques (rénaux, cardiovasculaires) observés. 2) Isoler, différencier et trouver le temps optimal d’induction de la différenciation pour les CSMO porcines (CSMOp), sur la base de l'expression des facteurs de transcription et des protéines structurales cardiaques retrouvées aux différents passages. 3) Induire et quantifier la différenciation cardiaque par l’OT sur les CSMOp. 4) Vérifier le rôle du NO dans cette différenciation cardiaque sur les CSMOp. Nous avons constaté que le profil pharmacocinétique de l’OT est mieux expliqué par le modèle connu comme target-mediated drug disposition (TMDD), parce que la durée du séjour de l’OT dans l’organisme dépend de sa capacité de liaison à son récepteur, ainsi que de son élimination (métabolisme). D'ailleurs, nous avons constaté que la différenciation cardiomyogénique des CSMOp médiée par l’OT devrait être induite pendant les premiers passages, parce que le nombre de passages modifie le profile phénotypique des CSMOp, ainsi que leur potentiel de différenciation. Nous avons observé que l’OT est un inducteur de la différenciation cardiomyogénique des CSMOp, parce que les cellules induites par l’OT expriment des marqueurs cardiaques, et l'expression de protéines cardiaques spécifiques a été plus abondante dans les cellules traitées à l’OT en comparaison aux cellules traitées avec la 5-azacytidine, qui a été largement utilisée comme inducteur de différenciation cardiaque des cellules souches adultes. Aussi, l’OT a causé la prolifération des CMSOp. Finalement, nous avons observé que l'inhibition de la voie de signalisation du NO affecte de manière significative l'expression des protéines cardiaques spécifiques. En conclusion, ces études précisent un potentiel certain de l’OT dans le cadre de la thérapie cellulaire cardiomyogénique à base de cellules souches adultes, mais soulignent que son utilisation requerra de la prudence et un approfondissement des connaissances. / Cell therapy has been suggested as a promising treatment for myocardial regeneration through cardiomyocyte replacement or by preventing apoptosis of surviving myocardium and/or improving neovascularisation. Bone marrow stem cells (BMSCs) express cardiac markers in vitro upon stimulation with different inducers. The BMSCs have been used as cell therapy after myocardial infarction (MI) in pre-clinical and clinical studies. Recent reports have uncovered the potential beneficial effects of oxytocin (OT) after MI. Particularly, OT is an inducer of cardiomyogenic differentiation of embryonic stem cells and this differentiation is mediated by the nitric oxide (NO)-soluble guanylyl cyclase pathway. However, some studies have shown that OT exhibits nonlinear pharmacokinetics and that this could explain the previously described controversial hemodynamic alterations. Therefore the objectives of the present work were to: 1) Characterize the pharmacokinetic profile of different dosing regimens of OT in swine, by using a more suitable pharmacokinetic / pharmacodynamic modelization that could explain the time-course of cardiovascular and renal effects observed following OT administration. 2) To isolate, differentiate and find the optimum time of porcine BMSC (pBMSC) differentiation based on the expression of cardiac related transcription factors and structural proteins expressed at different passages. 3) To induce and quantify the OT-mediated cardiomyogenic differentiation of pBMSCs. 4) To document the role of the NO pathway in the OT-mediated cardiomyogenic differentiation of pBMSCs. We found that OT pharmacokinetics are better explained by target-mediated drug disposition (TMDD) kinetics, because the time-course of plasma OT concentration depends on the binding capacity to its receptor, as well as OT elimination (metabolism). Also, we found that OT-mediated cardiomyogenic differentiation of pBMSCs should be induced during the first passages, because passaging affects the phenotypic profile of pBMSCs, as well as the differentiation potential of pBMSCs. We observed that OT induces cardiomyogenic differentiation of pBMSCs, because OT-induced cells expressed cardiac markers, and the expression of cardiac specific proteins was more abundant in OT-treated cells vs. 5-azacytidine-treated cells, which has been used widely as a cardiomyogenic differentiation inducer of adult stem cells. Moreover, OT improved proliferation of pBMSCs. Finally, we observed that the inhibition of the NO pathway significantly affects the expression of cardiac specific proteins. To conclude, these studies demonstrate some interesting potential in cardiomyogenic differentiation of adult stem cells for OT, but its precise role in cell therapy will need prudence and further investigations.

Evolution of Vertebrate Endocrine and Neuronal Gene Families : Focus on Pituitary and Retina

The duplication of genes followed by selection is perhaps the most prominent way in which molecular biological systems gain multiplicity, diversity and functional complexity in evolution. Whole genome duplications (WGDs) therefore have the potential of generating an extraordinary amount of evolutionary innovation. It is now accepted that the vertebrate lineage has gone through two rounds of WGD in its early stages, after the divergence of invertebrate chordates and before the emergence of jawed vertebrates. These basal vertebrate WGDs are called 2R for two rounds of whole genome duplication. An additional WGD called 3R occurred early in the evolution of teleost fishes, before the radiation of this species-rich group. This thesis describes the evolution of several endocrine and neuronal gene families in relation to the vertebrate WGDs, through a comparative genomic approach including both phylogenetic analyses and chromosomal location data across a wide range of vertebrate taxa. These results show that numerous endocrine gene families have expanded in 2R and in several cases also in 3R. These include the gene families of oxytocin and vasopressin receptors (OT/VP-R), somatostatin receptors (SSTR) and insulin-like growth factor binding proteins (IGFBP). For the OT/VP-R and SSTR families, previously undescribed subtypes were identified. The protein hormone family that includes growth hormone (GH), prolactin (PRL) and somatolactin (SL) acquired a new PRL gene in 2R, however the origins of GH, PRL and SL likely predate 2R. The corresponding family of receptors diversified during different time periods through a combination of local duplications and 3R. Neuronal gene families of the visual system have also expanded in 2R and 3R. The results presented here demonstrate that the vertebrate repertoire of visual opsin genes arose in 2R as part of chromosomal blocks that also include the OT/VP-R genes. The gene families including the transducin alpha, beta and gamma subunits also arose in 2R, hinting at the importance of these events in the diversification and specialization of phototransduction cascades for rods and cones. Thus, the whole genome duplications have been important contributors to the evolution of both vision and endocrine regulation in the vertebrates.

Les objectifs de ce programme doctoral étaient les suivants : 1) Caractériser le profil pharmacocinétique de différents schémas posologiques d'OT chez le porc, en développant une modélisation pharmacocinétique / pharmacodynamique plus adaptée à intégrer les effets biologiques (rénaux, cardiovasculaires) observés. 2) Isoler, différencier et trouver le temps optimal d’induction de la différenciation pour les CSMO porcines (CSMOp), sur la base de l'expression des facteurs de transcription et des protéines structurales cardiaques retrouvées aux différents passages. 3) Induire et quantifier la différenciation cardiaque par l’OT sur les CSMOp. 4) Vérifier le rôle du NO dans cette différenciation cardiaque sur les CSMOp. Nous avons constaté que le profil pharmacocinétique de l’OT est mieux expliqué par le modèle connu comme target-mediated drug disposition (TMDD), parce que la durée du séjour de l’OT dans l’organisme dépend de sa capacité de liaison à son récepteur, ainsi que de son élimination (métabolisme). D'ailleurs, nous avons constaté que la différenciation cardiomyogénique des CSMOp médiée par l’OT devrait être induite pendant les premiers passages, parce que le nombre de passages modifie le profile phénotypique des CSMOp, ainsi que leur potentiel de différenciation. Nous avons observé que l’OT est un inducteur de la différenciation cardiomyogénique des CSMOp, parce que les cellules induites par l’OT expriment des marqueurs cardiaques, et l'expression de protéines cardiaques spécifiques a été plus abondante dans les cellules traitées à l’OT en comparaison aux cellules traitées avec la 5-azacytidine, qui a été largement utilisée comme inducteur de différenciation cardiaque des cellules souches adultes. Aussi, l’OT a causé la prolifération des CMSOp. Finalement, nous avons observé que l'inhibition de la voie de signalisation du NO affecte de manière significative l'expression des protéines cardiaques spécifiques. En conclusion, ces études précisent un potentiel certain de l’OT dans le cadre de la thérapie cellulaire cardiomyogénique à base de cellules souches adultes, mais soulignent que son utilisation requerra de la prudence et un approfondissement des connaissances. / Cell therapy has been suggested as a promising treatment for myocardial regeneration through cardiomyocyte replacement or by preventing apoptosis of surviving myocardium and/or improving neovascularisation. Bone marrow stem cells (BMSCs) express cardiac markers in vitro upon stimulation with different inducers. The BMSCs have been used as cell therapy after myocardial infarction (MI) in pre-clinical and clinical studies. Recent reports have uncovered the potential beneficial effects of oxytocin (OT) after MI. Particularly, OT is an inducer of cardiomyogenic differentiation of embryonic stem cells and this differentiation is mediated by the nitric oxide (NO)-soluble guanylyl cyclase pathway. However, some studies have shown that OT exhibits nonlinear pharmacokinetics and that this could explain the previously described controversial hemodynamic alterations. Therefore the objectives of the present work were to: 1) Characterize the pharmacokinetic profile of different dosing regimens of OT in swine, by using a more suitable pharmacokinetic / pharmacodynamic modelization that could explain the time-course of cardiovascular and renal effects observed following OT administration. 2) To isolate, differentiate and find the optimum time of porcine BMSC (pBMSC) differentiation based on the expression of cardiac related transcription factors and structural proteins expressed at different passages. 3) To induce and quantify the OT-mediated cardiomyogenic differentiation of pBMSCs. 4) To document the role of the NO pathway in the OT-mediated cardiomyogenic differentiation of pBMSCs. We found that OT pharmacokinetics are better explained by target-mediated drug disposition (TMDD) kinetics, because the time-course of plasma OT concentration depends on the binding capacity to its receptor, as well as OT elimination (metabolism). Also, we found that OT-mediated cardiomyogenic differentiation of pBMSCs should be induced during the first passages, because passaging affects the phenotypic profile of pBMSCs, as well as the differentiation potential of pBMSCs. We observed that OT induces cardiomyogenic differentiation of pBMSCs, because OT-induced cells expressed cardiac markers, and the expression of cardiac specific proteins was more abundant in OT-treated cells vs. 5-azacytidine-treated cells, which has been used widely as a cardiomyogenic differentiation inducer of adult stem cells. Moreover, OT improved proliferation of pBMSCs. Finally, we observed that the inhibition of the NO pathway significantly affects the expression of cardiac specific proteins. To conclude, these studies demonstrate some interesting potential in cardiomyogenic differentiation of adult stem cells for OT, but its precise role in cell therapy will need prudence and further investigations.

Efeitos a longo prazo de diferentes separações dos filhotes no período neonatal sobre as genitoras

Esse estudo foi realizado para verificar se a exposição a separações repetidas (por diferentes intervalos de tempo) de mães dos seus filhotes no período neonatal poderiam ser classificadas como indutoras de um estado do tipo deprimido em genitoras. Sessenta ratas Wistar prenhes foram divididas em 3 grupos: controle, separação por 10 minutos e separação por 3 horas. As intervenções neonatais foram realizadas nos dias 1-10 pós parto. Após o desmame as genitoras foram submetidas ao teste do nado forçado, ao teste do labirinto em cruz elevado e ao teste do odor de predador. Também foi avaliado o comportamento alimentar e os padrões de reatividade a um sabor doce e a um sabor amargo. Foi medido os níveis de ocitocina no líquido cefaloraquidiano, corticosterona plasmática e atividade hipocampal Na+, K+-ATPase, assim como a atividade das enzimas antioxidantes catalase, glutationa peroxidase, superoxido dismutase, produção de radicais livres, e a produção de óxido nítrico, além dos níveis dos receptores A2A de adenosina e D2 de dopamina no estriado dorsoventral e no hipocampo. Foi observado que somente a separação por 3 h induziu um aumento significativo no tempo de imobilidade no teste do nado forçado, o que é consistente com estudos prévios. A atividade hipocampal da Na+, K+-ATPase se mostrou diminuída no grupo separado por 10 minutos e mais significativamente diminuída nas genitoras submetidas a separação por 3 horas de seus filhotes. Adicionalmente, os níveis de ocitocina no líquido cefaloraquidiano se encontravam aumentados no grupo separado por 10 minutos, o que pode estar relacionado a um aumento no cuidado materno, induzido por esta manipulação dos filhotes, por parte das genitoras, como reportado na literatura. Uma redução nos níveis de óxido nítrico no hipocampo das genitoras separadas por 3 horas foi observado Nessas genitoras também foi verificado um aumento no comportamento de risco, uma diminuição no sentimento prazeroso frente a uma solução doce e aumento na sensibilidade a uma solução aversiva, o que é congruente a um perfil de estado do tipo deprimido. Além disso, nós verificamos uma diminuição na quantidade do receptor de dopamina D2 no estriado das mães submetidas a separação por 3 horas dos filhotes, o que poderia ser relacionado com uma diminuição no prazer (anedonia) que acontece na depressão. Conclui-se que a retirada dos filhotes das mães por longos períodos tornam essas mães mais susceptíveis ao desenvolvimento de características depressivas. / This study was carried out to ascertain whether exposure to repeated separations (different times) of mothers from their pups in the neonatal period could be classified as an induction of a depressive-like state in dams. Sixty Wistar rats were divided into 3 groups: control, brief separation and long-term separation. The neonatal interventions were done on postpartum days 1-10. After weaning, the dams were subjected to the forced swimming test, elevated plus maze and predator odor test. It was also evaluated the feeding behavior and the taste reactivity patterns to a sweet and to a bitter solution. It was mesaured cerebral spinal fluid oxytocin, plasma corticosterone, and hippocampal Na+, K+-ATPase activity, as well as the activity of the antioxidant enzymes catalase, glutathione peroxidase, superoxide dismutase, free radicals production, and the production of nitric oxide and the levels of adenosine A2A and dopamine D2 receptors in the dorsoventral striatum and hippocampus. It was observed that only the 3 h separation induced a significant increase in the immobility time of rats in the forced swimming test, which is consistent with previous studies. Hippocampal Na+, K+-ATPase activity was decreased in the brief separated group and more significantly decreased in dams subjected to 3h separation from their pups. Additionally, cerebral spinal fluid oxytocin was increased in dams of the brief separated group, which may be related to the increased handled-induced maternal care, as reported in the literature. A reduction in nitric oxide levels in the hippocampus in dams of the long separated group was also observed. It was also verify an increase in the risk-taking behavior by the 3h separated mothers. The 3h separated mother also demonstrated a diminished feeling of pleasure with a sucrose solution and a increased sensibility to a aversive solution, wich is congruent with a depressive like state profile. Thus, the LPBN deactivation could modify the activity of hypothalamic oxytocinergic pathways or the gastric emptying control system, another inhibitory system, as well as facilitatory areas and the reward system. The aim of this study was to investigate the effect of LPBN injections of methysergide (4 μg/0.2 μl, serotonergic antagonist) in cell dehydrated animals on: activity of brain areas involved in ingestive behavior by measuring c-Fos protein immunoreactivity and tissue levels of dopamine, serotonin and metabolites or plasma hormone levels; pre-systemic satiety involving gastric emptying; selectivity of paradoxical sodium intake. The effect of disinhibition of the natriorexigenesis on the reward system was tested by repeated deactivations of the LPBN with muscimol (2 nmol/0.2 μl; GABAA agonist) and its effect on ingestive behavior sensitization and water deprivation with partial rehydration followed by sodium access (WD-PR protocol) on the lateral hypothalamus self-stimulation (LHSS). Holtzman or Sprague-Dawley rats (280-320 g), intacts or operated (femoral vein cannulation and/or guide cannulas implanted in direction to the LPBN or bipolar electrode implanted in the hypothalamus), were used in the experiments. Animals treated with methysergide and hyperosmotic by gavage of 2 M NaCl (2 ml) compared to the control treatment (vehicle) showed: (a) increase in ir-Fos in the area postrema and intermediate nucleus of the solitary tract, subfornical organ and non-oxytocinergic neurons of the ventral portion of the paraventricular nucleus of the hypothalamus; (b) increase in tissue levels of dopamine in the amygdala, but not in the accumbens; (c) unchanged activity of oxytocinergic system (ir-Fos in oxytocinergic neurons and oxytocin plasma levels similar to control group). Hyperosmotic rats (iv infusion of 2 ml of 2 M NaCl) treated with methysergide and a gavage of 0.3 M NaCl (3 ml) showed a hypertonic gastric and intestinal content similar to the control group (vehicle) after gavage. In hydrated animals with a history of two previous treatments of muscimol into the LPBN, the hypertonic NaCl intake induced by muscimol was higher than the control animals pretreated with vehicle. The LHSS was not altered at any stage of WD-PR protocol, as well as in cell dehydrated animals in comparison with hydrated control group. The results demonstrate that the deactivation of the LPBN enhances specifically the intake of solutions containing sodium and suggest the involvement of the brain stem and the amygdala during the appetitive phase of the paradoxical sodium intake, while the deactivation of other inhibitory mechanisms (oxytocin and gastric retention) seems not to be essential. Furthermore, the repetition of LPBN deactivation sensitizes hydrated animals for sodium intake. Removing the inhibition of sodium appetite by partial rehydration in WD-PR does not change the LHSS reward. / A ingestão de sódio é induzida por sinais facilitadores, como a angiotensina II e a aldosterona. Apesar de classicamente considerada antinatriorexigênica, a desidratação celular também se mostra facilitadora da ingestão paradoxal de sódio quando a inibição da ingestão de sódio pelo mecanismo ocitocinérgico do hipotálamo ou pelo núcleo parabraquial lateral (NPBL) está desativada. A desativação do NPBL poderia, então, atuar sobre o mecanismo ocitocinérgico ou de controle do esvaziamento gástrico, ambos inibidores, ou ainda modificar a atividade de áreas facilitadoras ou envolvidas na recompensa. O objetivo deste estudo foi investigar o efeito da injeção de metisergida (4 μg/0,2 μl, antagonista serotonérgico) no NPBL em animais com desidratação celular sobre a: atividade de áreas encefálicas envolvidas no comportamento ingestivo, medindo-se imunorreatividade para proteína c-Fos e dosagem tecidual de dopamina, serotonina e metabólitos em áreas de interesse ou dosagem hormonal; saciedade pré-sistêmica envolvendo esvaziamento gástrico; seletividade da ingestão paradoxal de sódio. Também foi avaliado o possível efeito da desinibição da natriorexigênese sobre o sistema de recompensa através da sensibilização por desativações repetidas do NPBL pelo muscimol (2 nmol/0,2 μl; agonista GABAA) e na privação hídrica com reidratação parcial e posterior acesso ao sódio (modelo PH-RP) sobre a autoestimulação elétrica do hipotálamo lateral (AEHL). Ratos (280-320 g) Holtzman ou Sprague-Dawley, intactos ou operados (canulação da veia femoral e/ou cânulas-guia direcionadas ao NPBL ou eletrodo bipolar no hipotálamo), foram utilizados nos experimentos. Animais tratados com metisergida e hiperosmóticos por gavagem de NaCl 2 M (2 ml) apresentaram, em relação ao tratamento controle (veículo): (a) aumento da ir-Fos na área postrema e núcleo do trato solitário intermediário, no órgão subfornical e em neurônios não ocitocinérgicos da porção ventral do núcleo paraventricular do hipotálamo; (b) aumento da concentração tecidual de dopamina na amígdala, mas não no accumbens; (c) atividade do sistema ocitocinérgico inalterada (ir-Fos em neurônios ocitocinérgicos e ocitocina plasmática semelhantes ao controle). Ratos tratados com metisergida e hiperosmóticos por infusão iv de NaCl 2 M (1,5 ml), que receberam uma gavagem de NaCl 0,3 M (3 ml) apresentaram conteúdo gástrico e intestinal hipertônico semelhantes ao controle (veículo) após a gavagem. Em animais hidratados com histórico de dois tratamentos prévios de muscimol no NPBL, a ingestão de NaCl hipertônico induzida por muscimol foi superior aos animais controle tratados previamente com veículo. A AEHL não foi alterada em nenhuma fase do protocolo PH-RP, assim como em animais com desidratação celular, em relação aos controles hidratados. Os resultados demonstram que a desativação do NPBL potencializa especificamente a ingestão de solução contendo sódio e sugerem a participação do tronco encefálico e da amígdala na fase apetitiva da ingestão paradoxal de sódio, enquanto a desativação de outros mecanismos inibidores (ocitocina e retenção gástrica) parece não ser essencial. Ainda, a repetição da desativação do NPBL sensibiliza a ingestão de sódio em animais hidratados. A remoção da inibição ao apetite ao sódio pela reidratação parcial na PHRP não altera a recompensa na AEHL.
