Apple scab, the most serious disease of apple is caused by fungal pathogen Venturia inaequalis. Knowledge about the apple response to apple scab attack on the cellular and tissue level is insufficient. For studies of Malus-Venturia interaction on the cellular and tissue level, the establishment of methods for cell structures visualization in apple leaves is necessary. In this work, the experimental plant material grown in vitro and ex vitro was successfully established and the method of apple infection by conidia of V. inaequalis was optimized. Various methods of cell components visualization such as vital staining, in situ immunolocalization, transformation, environmental scanning electron microscopy and confocal microscopy, were tested. Cell structures, such as the cytoskeleton, the cell wall and the cuticle were visualized in apple leaves. Preliminary experiments following specific the changes of cell wall structures induced by V. inaequalis attack were performed. Polyomaviruses are small unenvelope DNA viruses, whose replication take place in cell nucleus. Despite its small genome size, these viruses can cause significant changes in the host cell, one of the most significant is cell transformation. Most studies of human pathogens from this family is the focus of clinical research, but do not provide enough information about the individual events of the life cycle of viruses. This thesis mainly aims to determining the exact time when the creation of the individual viral products and generate a timeline of events during natural infection in cells that are targets for BKV in the human body. It was found that the time course of the life cycle of BKV is very similar to those for model polyomaviruses MPyV and SV40 and in permissive cells takes about 40 - 50 hours.

This study was focused on vector-pathogen-host interaction. Questing ticks from field were tested for presence of Borrelia burgdorferi s.l. and host DNA. Small rodents were trapped, ticks were collected from them, infestation patterns were estimated regarding the species and stage of ticks and species, sex and body weight of the host. Ticks aquired from hosts were tested for presence of Borrellia burgdorferi s.l. and tick-borne encephalitis virus. Both results from identification of hosts and from detection of pathogens were compared to be able to investigate interactions between host, vector and pathogen.

Bakgrund Forskning visar att delar av samhället anser att idrott och hälsa (IDH) är ett redskap, vars syfte är att eliminera fetma och andra former av sjukdomar. Vilket tyder på att vissa anser att IDH bör förhålla sig till det patogena hälsoperspektivet. Detta däremot, ställer sig flera forskare kritiska till, då de anser att IDH bör förhålla sig till vad som främjar och utvecklas hälsa, vilket innefattar ett salutogent hälsoperspektiv. Syftet med studien Syftet med detta arbete är att undersöka hur lärare undervisar hälsa och dess dimensioner, samt hur undervisningen förhåller sig till salutogent- och patogent hälsoperspektiv. Metod Empirin samlades genom fyra semi-strukturerade kvalitativa intervjuer, ungefär 40 minuter vardera. Dessa intervjuer genomfördes via konferensprogrammet ”Zoom”. Resultat och diskussion Det framkommer att fysiska aktiviteter är grunden till all hälsoundervisning, vilket tillför möjligheten för lärare att sätta icke-fysiska aspekter av hälsa i kontext. Dessutom framkommer det att lärare nästan forcerar samarbete hos eleverna, vilket ska utveckla eleverna förmåga att bygga relationer, minimera exklusion och förmågan att samverka. Lärare för det mesta implementerar flertalet lärdomar inom en aktivitet. Eleverna ges möjligheten attreflektera kring aktivitetens övriga effekter på hälsa, bortsett från de fysiska aspekterna. Det visade sig att det patogena- och det salutogena hälsoperspektivet framkommer i hälsoundervisningen. Detta för att de både kompletterar varandra och tillför viktiga aspekter till både lärares undervisning metoder och kunskap till eleverna. / Background Research suggests that parts of society view physical education (PE) as a tool, the purpose of which is to eliminate obesity and other kinds of illness. The result is that some argue that PE should have a pathogenic view on health. This view however is criticized by some researchers who suggest that PE should focus on what improves health, in other words a salutogenic view on health. Aim of this study The aim of this study is to investigate how teachers educate health and its dimensions, and how health education adheres to the salutogenic- and pathogenic health perspectives. Method Empirical data were collected from four semi-structured qualitative interviews, approximately 40 minutes each. Interviews were conducted with the conference program “Zoom”. Result and discussion Participants argue that physical activities are the foundation of all health education, this provides teachers with an opportunity to put the non-physical aspects of health into context. These activities of which can be used to enhance and develop psychological and social health aspects. Through almost forced co-operations, students improve their relation building competence, reinforces inclusion and ability to co-operate. Moreover, teachers mostly implement several teachings within one activity. They do this by making the students reflect on “spin-off” effect from the activity, where other health developments other than physical aspects within an activity are required. It is revealed that both pathogenic and salutogenic perspective is present in the health education, and that they complement each other and bring different important aspects to teachers’ methods and knowledge to students.

Vad är hälsa för högstadieelever? : En intervjustudie om flickor och pojkar och deras uppfattning om begreppet hälsa i årskurs 9Almali, Leyli January 2019 (has links)
Aim The purpose of this survey is to investigate what students in grade 9 thinks what health is. How they relate to health in the subject physical education and what image they have on the subject. Questions What do students in grade 9 think about what they learn about the notion of health. How do the teachers convey their knowledge to the students about health. Do the students have a pathogen or a salutogenic perspective of health. Method To answer the questions the survey used a qulitative method with interviews. Six students from grade 9 and 3 teachers in physical education participated in the study. The interviews were semistructured because the content and questions were decided before hand but the follow up questions were adapted to the people that participated. Results The students perceived the notion of health in the subject differently. Some students connects the notion of health to different physical activities whilst some students thinks about nutrition as an important part of health. All the participating teachers used theory sessions to educate about health. However, the content could differ between the teachers. The image of what the students think about health is mainly from a salutogenic perspective. 5 of 6 students said they interpreted that they had a salutogenic perspective and only one person thought about health from a pathogen perspective. Conclusions The students interpreted the notion of health differently. The teachers used theory sessions when they educated about health. Both students and teachers thought about health from mainly a salutogenic perspective. / Syfte Syftet med denna studie är att undersöka vad elever i årskurs 9 anser vara hälsa. Hur de relaterar hälsa till undervisningen i ämnet idrott och hälsa, samt vilken bild av hälsa de har. Frågeställningar: Vad anser eleverna i årskurs 9 att de lär sig om begreppet hälsa i ämnet idrott och hälsa? Hur förmedlar lärarna kunskap om hälsa till eleverna? Har eleverna en patogen eller salutogen bild av begreppet hälsa? Metod För att svara på studiens frågeställningar har en kvalitativ metod använts där intervjuer gjorts på sex olika elever i årskurs 9 och tre idrottslärare. Intervjuerna var semistrukturerade då innehåll och följd på intervjufrågor förutbestämde synsättet. Studiens teoretiska utgångpunkt tas i begreppen salutogen och patogen. Det salutogena tar fokus i människans främjande mot en bättre hälsa än vad denne har idag medan det patogena synsättet fokuserar på själva sjukdomen i sig, varför man blir sjuk. Resultat Eleverna har olika tolkningar kring begreppet hälsa i ämnet idrott och hälsa. Några elever kopplar begreppet hälsa till olika typer av fysiska aktiviteter medan andra tar in kosten som en viktig del i begreppet. Samtliga lärare använder teoriundervisning när de vill förmedla kunskap kring begreppet hälsa. Undervisningens innehåll däremot skiljer sig mellan lärarna. Eleverna har huvudsakligen en salutogen bild av hälsa. Slutsats Elevernas bild av hälsa är huvudsakligen salutogen, det vill säga man har ett hälsofrämjande synsätt i motsats till att fokusera på vad som orsakar sjukdom. / <p>Studiegång Idrott, fritidskultur och hälsa. A diversidade genética de Crinipellis perniciosa foi avaliada quanto à capacidade adaptativa de diferentes genótipos à dispersão geográfica e à associação preferencial ou diferencial com hospedeiros. Isolados polispóricos de C. perniciosa provenientes de diferentes Estados brasileiros (AM, RO, MG, BA, SP, MT e PA) e diferentes hospedeiros (Theobroma cacao L., Solanum cernuum, Solanum paniculatum, Solanum grandiflorum L., Solanum e lianas), foram analisados pelas técnicas de compatibilidade somática (SCG - Somatic Compatibility Group), através de reações de compatibilidade e/ou incompatibilidade, o uso de gel de eletroforese de proteínas totais, velocidade de crescimento, ELISA e Western Blot. Os diversos isolados foram agrupados com base em conjuntos de dados sobre suas características genéticas, identificando-se agrupamentos de acordo com a dispersão geográfica e hospedeiros de onde foram isolados. Foram construídos dois dendogramas pelo teste de compatibilidade permitindo classificar os isolados em três grupos: i)grupo formado pelos isolados de Rondônia e Pará e ii) grupo formado pelos isolados de Rondônia, Pará e Amazonas e o iii) grupo formado pelo isolado de Mato Grosso; o segundo teste separou os isolados em dois grandes grupos: i) grupo formado pelos isolados da Bahia, Minas Gerais, São Paulo, Mato Grosso e Rondônia e ii) grupo formado pelo isolado do Pará. O dendograma criado pelo perfil protéico permitiu agrupar os isolados em três grupos, de acordo com o hospedeiro e região de origem: (1) grupo formado pelo isolado de Theobroma cacao de Ouro Preto DOeste; (2) grupo constituído pelos isolados de Theobroma cacao da Bahia e os diversos isolados de Solanaceae do Estado de Minas Gerais, Bahia e São Paulo e grupo (3) formado pelo isolado de Lianas (cipós). De forma geral, conclui-se que há ampla diversidade genética entre os isolados de C. perniciosa provenientes das diferentes regiões brasileiras e em relação aos hospedeiros, comprovando-se haver elevada capacidade adaptativa caracterizada pela prevalência em ambientes diversos e pela disseminação a longas distâncias geográficas. Há também indicativos de que a velocidade de crescimento do fungo pode ser um dos fatores importantes para assegurar a colonização, já que é correlacionada com a dispersão geográfica, e também, para a seleção de biótipos patogênicos e para a variabilidade genética do fungo. / The genetic diversity of Crinipellis perniciosa was evaluated how much to the capacity of adaptation of different genotypes to the geographic dispersion and the preferential or distinguishing association with hosts. Of general form, it is concluded that it has ample genetic diversity enters the isolated ones of C. perniciosa proceeding from the different Brazilian regions and in relation to the hosts, proving to have high capacity of adaptation characterized by the prevalence in diverse environments and the dissemination the long geographic distances. It has also indicative of that the growth speed can be one of the factors important to assure the correlated settling since with the geographic dispersion, favors new biotypes, important factor for the genetic variability of fungus.

Functional characterization of AWR affector proteins from the phytopathogen "R. solanacearum" (Caracterització funcional de les proteïnes efectores AWR del fitopatogen "R. solanacearum")Solé Castellví, Montserrat 25 November 2011 (has links)
"R. solanacearum" is a devastating bacterial pathogen that infects "Solanaceae" spp. such as tomato, eggplant or banana. A functional T3SS is required for virulence and more than 70 putative effectors have been described, although only few have been studied. This thesis focuses on a five-member gene family of effectors named "awr".
We demonstrated that awr gene family is extremely conserved among R. solanacearum strains but also present in other plant pathogens such as Acidovorax or Burkholderia spp. and even present in the human pathogen B. pseudomallei. Virulence of a Ralstonia mutant strain devoid of all awr genes was tested on tomato, eggplant and Aradidopsis. Plant growth of quintuple mutant strain was considerably reduced in natural hosts, indicating a role in virulence, but remained unchanged in Arabidopsis. Col-0 infection with Pseudomonas syringae DC3000 heterologously expressing each AWR was also performed. While presence of some AWRs in Pseudomonas did not have an effect on plant growth, others (like AWR5) dramatically reduced the pathogen multiplication, pointing out a possible plant detection. In order to unravel the functions of AWR proteins, they were transiently expressed by means of Agrobacterium in non-host Nicotiana spp. Upon AWR expression, necroses took place to different extents on the plant leaves. AWR5 induced the strongest necrosis, resembling an HR phenotype which was later confirmed by TB/DAB staining and by RT-PCR of specific HR marker genes. Furthermore, a strong reduction in yeast cells was experimented upon several AWR protein expressions which indicate that the mechanisms that might be altered by these effector proteins is conserved among eukaryotes and hence reinforces their role in virulence. AWR4 appeared not to be toxic in this model organism and for that reason we sought to decipher some of the plant targets of this AWR protein as a start point. Out of more than 60 interacting clones were sequenced after a yeast-two hybrid screening with Arabidopsis root cDNA from R. solanacearum challenged plants. Among them, several defense-related proteins were found: phenylalanine ammonia-lyase, MPK6, DMR6 or KIN10. In order to find other key genes for AWR activity, the AWRs that displayed a strong yeast toxicity were heterologously produced in both E. coli and R. solanacearum to be ready to be employed as a bait for plant protein complexes that will be analysed by mass spectrometry.
In summary, AWR are highly conserved effectors that play an important role in both pathogenesis and plant recognition as they reduce P. syringae virulence and trigger an HR-like phenotype in non-host plants. Deciphering effector function will open promising avenues towards the design of new strategies to control R. solanacearum. / R. solanacearum és un patogen bacterià capaç d’infectar diferents solanàcies com ara la tomaquera, la patatera, l’alberginiera o el plataner. Aquest fitopatogen injecta més de 70 proteïnes efectores en la cèl•lula vegetal hoste, tot i que només algunes han sigut ja estudiades. Aquesta tesi es centra en una família multigènica d’efectors: els AWRs.
Els estudis científics duts a terme durant aquesta tesi van demostrar que
la família de AWR no només estava altament conservada en el llinatge de R. solanacearum sinó que també es trobava present en altres fitopatògens o inclús en el patogen humà Burkholderia pseudomallei. A més a més, diferents assajos de patogenicitat en tomaquera i alberginiera van provar que els gens awr presentaven un paper clar en virulència per aquests hostes. Contràriament, la presència d’aquestes proteïnes en la planta model Arabidopsis thaliana produïen una disminució en la capacitat infectiva/multiplicativa. Això indicaria una dualitat dels efectors AWR depenent del context que ens trobem, ja sigui contribuint a la patogenicitat del bacteri o bé éssent reconeguts per la planta i així disminuint la patogenicitat bacteriana.
Per tal de desentranyar les funcions de les proteïnes AWR, es van expressar de forma transitòria a la planta model no-hoste Nicotiana spp. L’expressió d’algunes proteïnes AWR va provocar una forta necrosi de les fulles que s’assemblaria a una resposta hipersensible. Mitjançant diferents tincions i assajos de PCR en temps real es va corroborar que l’AWR5 presentava aquest tipus de mort cel•lular programada. L’elevada toxicitat d’algunes AWRs es va demostrar també en llevat.
En el transcurs d’aquesta tesi també s’ha realitzat un crivellatge en doble híbrid per tal de buscar proteïnes dianes de la planta per a l’AWR4 (la menys tòxica). A més a més, es va posar a punt l’expressió dels AWRs a E. coli o bé a R. solanacearum per tal d’abordar altres tècniques que permetin una millor cerca d’interactors en el futur.

Access to clean drinking water is one of the most important resources for any society and its citizens. Without access to clean drinking water, health and living quality will be affected when pathogens will make people ill.In arid countries such as Ethiopia with seasonal drought, efforts to manage the water resources to last longer can be made by constructing dams and artificial irrigation systems. One example of this is the Koga dam in the Merawi region where a dam has been constructed to provide water for artificial irrigation. By irrigating an area one changes the hydrological conditions which might change the chemical composition of groundwater.This report aims to try and collect data that support the theory of the hydrological conductivity between irrigation water and groundwater in the area. Samples of groundwater and surface water was collected and compared to see if any conclu-sions supporting this theory could be made. Also general drinking water parameters were included such as presence of biological pathogens. The number of samples collected is on its own not enough to draw conclusions from but when combined with previous field work in the area and studying literature it supports the assumption of irrigation affecting the groundwater composition. / Tillgång till rent dricksvatten är en av de viktigaste resurserna för samhället såväl som varje enskild människa. Utan tillgång till rent dricksvatten kan hälsa och livs-kvalitet komma att påverkas av vattenburna patogener.I torra länder som Etiopien med periodisk torka har man gjort insatser för att hushålla med vattnet utöver regnperioderna. Ett exempel på detta är Kogadammen i Merawi, där en fördämning har konstruerats för att förse ett område med konst-bevattning. Genom bevattning ett område ändrar man de hydrologiska förhållanden som råder vilket gör att grundvattnets sammansättning kan komma att ändras.Denna rapport syftar till att försöka samla in data som stödjer teorin att konst-bevattningen i området påverkar grundvattnets sammansättning. Prover av grund-vatten och ytvatten samlades och jämfördes för att se om några slutsatser kunde dras. Också allmänna dricksvatten parametrar ingick såsom förekomsten av bio-logiska patogener. Antalet prover som tagits är ensamt inte tillräckligt för att dra slutsatser, men när de kombineras med data från tidigare fältarbete i området och litteraturstudier stödjer de antagandet om att konstbevattningen i området påverkar grundvattensammansättningen.

The subject of my thesis was to genetically modify a potato for increased resistance against its pathogens and pests. In developing a resistant plant, it is quite common to use the same type of molecules that plants use themselves in their defense reactions. In this work I used the gene SPI-2 originating from a honeycomb moth (Galleria mellonella). The protein SPI-2 is a member of serine protease inhibitors. Since the previous attempts of the team to detect the protein in transformed plants haven't been successful, the basic form of the gene was modified by adding a Kozak sequence near the start codon, which should have increased the translation initiation and hence increase the level of the protein. Two constructs were prepared for the transformation: SPI-2-T a SPI-2-Y. They differ by one amino acid, which slightly changes their inhibitory activity. First, the construct SPI-2-T was used for a transient transformation of tobacco Nicotiana benthamiana by agroinfiltration of its leaves. Then both constructs were used for a stable transformation of Solanum tuberosum cv. Desireé. The detection of the protein has not been successful, although the inserted gene was transcribed and his sequence was verified by sequencing.
