Propolis is a mixture of vegetable resins, salivary secretions, wax, pollen grains and ground. In this work eight propolis samples from jandaira collected in the backlands of Paraiba, Brazil were analyzed, revealing the presence of 22 different pollen types, among which, Myrcia, Mimosa, Parapiptadenia, Poincianella and Senna, which are present in almost all samples with pollen grain frequency between 3,3 to 50,0%. The quantitative analysis of phenolic compounds were characterized with values ranging from 92,56 to 201,62 mg GAE/g for EtOH extracts from 157,43 to 320,48 mg GAE/g for fractions AcOEt and 128,63 to 322,40 mg GAE/g for phenolic fractions. The ethanol extracts and phenolic fractions were also tested for antinociceptive activity, showing inhibition in writhing induced by acetic acid in mouse ranging from 96,9% to 100% for the extracts and 71,4% and 93,5% for phenolic fractions. Also, they have the ability to protect the DNA plasmid, with antioxidant potential against the DPPH, ABTS and β-carotene/linoleic acid system and showed antifungal activity against strains of yeasts and dermatophytes. The EtOH extracts exhibited a condensed tannin content ranging from 92,56 to 201,62 mg CE/g sample. Phenolic fractions were analyzed by HPLC-PDA-QTOF-MS and MS/MS in negative mode (ESI-) showing 66 peaks, and 36 peaks were identified mainly as phenyl propanoides, flavonoids and tannins. The major compounds were derivatives of galloyl cinnamoyl/coumaroyl glucopyranoside. The antinociceptive activity, antifungal and protection of plasmid DNA of the geopropolis from Melipona subnitida, as long as we know, was never described before in the literature. / Melipona subnitida, conhecida popularmente como jandaíra, é uma abelha nativa, social e sem ferrão que produz mel, geoprópolis e pólen. A geoprópolis é produzida a partir de uma mistura de resinas vegetais, secreções salivares, cera, grãos de pólen e terra. Foram analisadas oito amostras de geoprópolis da jandaíra coletadas no sertão da Paraíba-Brasil. As análises palinológicas das oito amostras revelou a presença de 22 tipos polínicos diferentes, dentre os quais, os tipos Myrcia, Mimosa, Parapiptadenia, Poincianella e Senna, estão presentes em quase todas as amostras com freqüência de grão de pólen entre 3,3 a 50,0%. As análises quantitativas dos compostos fenólicos foram caracterizadas com valores variando entre 92,56 - 201,62 mg EAG/g para os extratos EtOH, 157,43 - 320,48 mg EAG/g para as frações AcOEt e 128,63 - 322,40 mg EAG/g para as frações fenólicas. Os extratos etanólicos e frações fenólicas no ensaio de atividade antinociceptiva, produziram inibição nas contorções abdominais induzidas por ácido acético em camudongos, variando de 96,9% a 100% para os extratos e 71,4% a 93,5% para as frações fenólicas. Além disso, apresentaram capacidade de proteção ao DNA Plasmídeo, potencial antioxidante frente ao DPPH, ABTS e sistema β-caroteno/ácido linoléico e atividade antifúngica contra cepas de leveduras e dermatófitos. Os extratos EtOH exibiram um teor de taninos condensados variando entre 92,56 a 201,62 mg de EC/g de amostra. As frações fenólicas foram analisadas por UHPLC-PDA-qTOF-MS e MS/MS no modo negativo (ESI-). Foram detectados 66 picos, dos quais 36 foram identificados principalmente como fenil propanóides, flavonoides e taninos. Os principais compostos foram os derivados de galoil cinamoil/coumaroil glicopiranosídeo. A atividade antinociceptiva, antifúngica e a proteção ao DNA plasmídeo da geoprópolis de Melipona subnitida, estão sendo descritas pela primeira vez na literatura.

Os objetivos deste trabalho foram obter um processo de pasteurização da polpa de juçara embalada em bolsas plásticas (100 mL) de alta, a fim de minimizar a degradação das antocianinas, mantendo as propriedades nutricionais e organolépticas do produto, com viabilidade técnica e econômica para as comunidades tradicionais e agricultores familiares. Para tanto se avaliou a degradação da antocianina e a atividade enzimática presente na polpa de juçara, submetida à acidificação e pasteurização depois de embalada; armazenada por 60 dias; e monitorou-se a qualidade da polpa mediante análises físicas, químicas e sensoriais. A polpa foi acidificada com ácido cítrico a pH 4,0. A outra parte não sofreu processo de acidificação. Em seguida, a polpa foi embalada em sacos de polipropileno diferenciado. Por fim, as polpas, acidificadas com ácido cítrico a pH abaixo de 4,0 e não acidificada, foram submetidas aos seguintes tratamentos: armazenada sob refrigeração a 6 °C; armazenada sob congelamento a -17 °C; pasteurizada a 80 °C por 5 minutos e armazenada sob refrigeração a 6 °C; pasteurizada a 80 °C por 5 minutos e armazenada sob congelamento a -17 °C; Num segundo momento, após a realização das análises do primeiro processo, a polpa embalada foi pasteurizada em tacho com aquecimento a gás, com água à temperatura de fervura - 97°C. A polpa foi mantida a esta temperatura por um (1) minuto. A acidificação e armazenamento foram realizados da mesma forma que no primeiro processo. A polpa de juçara manteve-se estável por 45 dias de armazenamento refrigerado e congelado para todos os tratamentos. Sensorialmente, não houve diferença significativa entre os tratamentos. Considerando-se a estabilidade da polpa, sua aceitação sensorial e o uso da refrigeração como alternativa ao congelamento, a pasteurização em tacho a 97°C se torna uma possibilidade às comunidades extrativistas. / The objectives of this study were to obtain a pasteurization process the juçara pulp packed in plastic bags (100 mL) high barrier in order to minimize anthocyanin\'s degradation, maintaining the organoleptic and nutritional properties of the product, technical and economic feasibility for the traditional communities and family farmers. Análise in vitro da atividade antioxidante do suco e extrato de maçã em células RINm5f submetidas a diferentes condições de estresse oxidativo

Apple, world’s second largest produced fruit, is classified as having one of the high antioxidant potential due to its content and profile of phenolic compounds. The aim of this research was to evaluate the antioxidant activity of apple and check its effect on cells secreting insulin (RINm5f) subjected to oxidative stress induced by high concentration of glucose (GA) and H2O2. Once prepared juices and extracts of whole fruit, flesh and epicarp of Gala and Fuji cultivars, the following parameters were analyzed: total non-reducing sugars (AT), total phenols content (FT), antioxidant activity (AA) before and after centrifugation and filtration. Cytotoxicity, cell viability (VC) on oxidative stress and production of intracellular superoxide analysis were performed in biological tests. Several dilutions (1:10, 1:50, 1:100, 1:250) and quantities of extracts (5, 10, 50,100 and 250 mg/mL) of apple juice were tested to evaluate cytotoxicity. To evaluate biological effects, juice dilutions of 1:50, 1:100 and 1:250 were used for Gala and Fuji apple. Epicarp extracts concentration of 10, 50 and 100 mg/mL were used for Gala apple and 10 mg/mL for Fuji apple. In the whole fruit and fruit flesh, AT content ranged from 41.69 to 50.80 g/100g and epicarp’s was about 10 times less. Gala and Fuji juice AT content was, respectively, 12.94 g and 10.33 g/100g. Big loss of FT and the AA was observed after centrifugation and especially after filtration. Highest amount of FT was found in epicarp extract of Fuji apple before filtration. After filtration the highest concentration of FT was in extracts of Gala apple. Even presenting less FT than Gala apple after filtering, the epicarp extract of Fuji apple kept higher AA. Gala apple juice presented more FT and AA before and after filtration than Fuji apple juice. During cytotoxicity test, only Fuji apple epicarp extract at concentrations above 10 g/mL was cytotoxic. Under GA induced stress, Gala apple epicarp extract (10 mg/mL), all tested dilutions of Gala apple juice and the Fuji apple juice (1:250), increased the VC. Fuji apple juice showed cytotoxic effect on dilution 1:10. Gala apple apicarp extract (10, 50 and 100 g/mL) Fuji apple juice (1:250), reduced production of intracellular superoxide. Gala and Fuji apple juice (1:50 and 1:100) had cytoprotectant effect under H2O2 induced stress, increasing VC when compared to H2O2 group. However, none of the juices tested was able to significantly reverse the damage caused by H2O2 when cell morphology was evaluated. Significant increase of 50% on average in the secretion of insulin was observed in Gala apple juice (1:100 and 1:250) and Fuji apple juice (1:50). It could be concluded that according to oxidative stress type and extract of epicarp and juice of Gala and Fuji apples concentration, oxidative stress on cells RINm5f may be reduced. / Os compostos fenólicos, encontrados em vegetais e frutas, podem apresentar elevada atividade antioxidante. A maçã, segunda fruta de maior produção mundial, apresenta alto potencial antioxidante devido a seu teor e ao seu perfil de compostos fenólicos. O objetivo dessa pesquisa foi analisar a atividade antioxidante da maçã e estabelecer seu efeito sob células secretoras de insulina (RINm5F) submetidas ao estresse oxidativo induzido por elevada concentração de glucose (GA) e por H2O2. Nos extratos de fruta inteira, epicarpo e mesocarpo e sucos das cultivares Gala e Fuji, foram analisados açúcares redutores totais (AT), teor de fenóis totais (FT), atividade antioxidante (AA) antes e após centrifugação e filtração. Os ensaios biológicos compreenderam análises de citotoxicidade, viabilidade celular (VC) sob estresse oxidativo, produção intracelular de superóxido e análise da funcionalidade celular. Para avaliação da citotoxicidade foram testados diferentes diluições dos sucos de maçãs (1:10, 1:50, 1:100, 1:250) e quantidades de extratos (5, 10, 50 e 100 e 250 μg/mL). Para avaliar os efeitos biológicos foram utilizadas as diluições 1:50, 1:100 e 1:250 e as concentrações de extratos do epicarpo 10, 50 e 100 μg/mL para maçã Gala e 10 μg/mL para maçã Fuji. Na fruta inteira e mesocarpo os teores de AT variaram de 9,88 a 12,10 g/100g e no epicarpo foram na ordem de 3 vezes menos. Suco de maçã Gala e Fuji foi encontrado, respectivamente, 12,94 e 10,33 g/100g de AT. Grande perda de FT e da AA foi observada após centrifugação e em especial após filtração. Maior quantidade de FT foi observada no extrato de epicarpo de maçã Fuji antes da filtração. Após filtração o maior conteúdo de FT estava nos extratos de maçã Gala. Mesmo apresentando menos FT em relação à maçã Gala após a filtração, o extrato de epicarpo de maçã Fuji manteve maior AA. O suco de maçã Gala, em relação ao suco da maçã Fuji apresentou mais FT e AA antes e após a filtração. No ensaio de citotoxicidade, apenas o extrato do epicarpo de maçã Fuji em concentrações acima de 10 μg/mL foi citotóxico. Sob estresse induzido por GA, o extrato de epicarpo de maçã Gala (10 μg/mL), todas as diluições testadas do suco de maçã Gala, assim como o suco da maçã Fuji (1:250), aumentaram a VC. O suco de maçã Fuji apresentou efeito citotóxico na diluição 1:50. Extrato de epicarpo de maçã Gala (10, 50 e 100 μg/mL), assim como o suco da maçã Fuji (1:250), reduziram a produção intracelular de superóxido. Na presença de estresse induzido por H2O2, o suco de maçã Gala e Fuji (1:50 e 1:100) apresentaram efeito citoprotetor, aumentando a VC quando comparados ao grupo H2O2. Entretanto, quando analisada a morfologia celular, nenhum dos sucos testados foi capaz de reverter significativamente os danos causados pelo H2O2. Aumento significativo de 50% em média na secreção de insulina foi observado no suco de maçã Gala (1:100 e 1:250) e suco de maçã Fuji (1:50). Foi possível concluir que de acordo com tipo de estresse oxidativo e das concentrações utilizadas, extrato de epicarpo e suco das maçãs Gala e Fuji, podem diminuir o estresse oxidativo e melhorar a funcionalidade de células RINm5f.

As frutas, principalmente as que apresentam a coloração vermelha/azul, contêm elevadas quantidades de antioxidantes, sendo importantes fontes de compostos fenólicos. Aos compostos antioxidantes tem sido atribuída uma grande gama de efeitos biológicos, incluindo ações de prevenção contra doenças. Acredita-se que o consumo regular destas frutas, aliado a um estilo de vida saudável, incluindo dieta equilibrada e exercícios físicos, pode trazer potenciais benefícios à saúde. Porém, nem sempre as frutas e verduras são consumidas in natura, muitas vezes são consumidas após serem submetidas a algum tipo de processamento, o que pode alterar suas características nutricionais. Com isso se torna muito importante compreender melhor a estabilidade destes compostos em alimentos, frente ao processamento a que são submetidos antes da ingestão. Dessa forma, o objetivo deste trabalho foi o de avaliar o efeito do processamento por diferentes métodos de cocção (sous vide, micro-ondas, fervura e desidratação) sobre as propriedades antioxidantes de frutas vermelhas (amora, mirtilo, cereja, jabuticaba e morango). Para isso, verificou-se o conteúdo de compostos fenólicos e a capacidade antioxidante de cada uma delas in natura e processadas. De uma forma geral, foi possível observar que a jabuticaba foi a fruta que apresentou maior conteúdo de compostos fenólicos e maior atividade antioxidante dentre todas as frutas vermelhas estudadas, em todos os processos avaliados, com exceção da desidratação, na qual amora e morango tiveram os maiores resultados nas condições estudadas. Além disso, foi possível observar também que as frutas em geral processadas por cocção em micro-ondas e desidratação apresentaram maior conteúdo de compostos fenólicos e maior atividade antioxidante do que as submetidas aos demais processos. Os resultados obtidos demonstram haver clara influência do tipo de processo a que a fruta é submetida sobre as propriedades antioxidantes da mesma. Novos estudos, no entanto, se fazem necessários a fim de melhor esclarecer os efeitos do processamento sobre a atividade antioxidante das frutas vermelhas. / There is an increasing interest in the relationship between food and health. Recently the intake of food containing naturally occurring antioxidants has gained increasing evidence. Fruits, specially blue or red ones, contain high amounts of antioxidants and are important sources of total phenolic compounds. A wide range of biological effects, including disease prevention, have already been attributed to antioxidant compounds. It is widely believed that a regular fruit and vegetable consumption, along with a healthy life style including balanced diet and physical exercise, can bring potential benefits to human health. However, fruits and vegetables are not always consumed fresh, they are often consumed after being submitted to some kind of processing, which can alter its nutritional properties. It seems therefore important to study the stability of these compounds in food processed prior to consumption. In this way, the aim of this work was to evaluate the effects of heat processing by different methods (sous vide, microwave, boiling and dehydration) in the antioxidant properties of red fruit (blackberry, bilberry, cherry, jaboticaba and strawberry). Total phenolic compound content and antioxidant activity of each fruit were evaluated, both fresh and processed by different heating methods. It was observed that jaboticaba presented the highest phenolic compound content and also the highest antioxidant activity among all red fruits studied, in all processes evaluated, except in dehydration, in which blackeberry and strawberry had higher results in the conditions studied. Besides, it could be observed that red fruits processed by microwave and dehydration presented higher phenolic compound content and higher antioxidant activity than those submitted to the other processes studied. Results obtained show that appears to be a clear influence of the kind of process used into red fruits antioxidant properties. However, more studies are needed do better understand the effects of heat processing in antioxidant activity of red fruits.

A amÃndoa de castanha de caju (ACC) à um dos principais produtos do agronegÃcio cearense, mas tambÃm à fonte de renda para pequenos produtores rurais, principalmente nos perÃodos de estiagem. A amÃndoa de castanha de caju de maior valor para comercializaÃÃo à que aquela se classifica como inteira de primeira qualidade. As amÃndoas de castanha de caju de qualidade inferior, por cor ou quebradas durante o processamento, chegam a perder 80% do seu valor comercial. Desta forma, este trabalho teve como objetivo o desenvolvimento de um Ãleo a partir da amÃndoa de castanha de caju de classes inferiores, avaliando os mÃtodos de extraÃÃo a frio (extra virgem - OEV) e com aquecimento prÃvio em micro-ondas (virgem - OV), sua composiÃÃo e caracterÃsticas quÃmicas e fÃsicas, sua estabilidade, caracterÃsticas sensoriais e bioatividade. Na seleÃÃo das amÃndoas para extraÃÃo, considerou-se as caracterÃsticas fÃsico-quÃmicas e de qualidade oxidativa, utilizaram-se seis classes inferiores: B1, S1, W4, SP1, SP2, P1 e uma classe de amÃndoa inteira de primeira qualidade (W240) como referÃncia. Considerando os parÃmetros avaliados, optou-se pelo uso das amÃndoas SP2, de menor preÃo de mercado, visto que todas as classes avaliadas apresentaram boa qualidade fÃsico-quÃmica e oxidativa, com potencial para extraÃÃo do Ãleo (lipÃdios totais superiores a 50%). Na avaliaÃÃo dos processos de extraÃÃo, observou-se maior rendimento da amostra com aquecimento prÃvio em micro-ondas. A amostra OV tambÃm apresentou maior estabilidade oxidativa com tempo de induÃÃo mÃdio de 46,25 horas, contra 39,03 horas da amostra OEV. Possivelmente a maior estabilidade se deu pela maior presenÃa de fenÃlicos totais na amostra OV (316,4 mg/100g) do que na amostra OEV (202,17 mg/100g). Ambos os Ãleos apresentaram aÃÃo antioxidante, possivelmente pela presenÃa de compostos como tocoferÃis, Ãcidos graxos mono e poli-insaturados, fitosterÃis e fenÃlicos identificados nas amostras. Na avaliaÃÃo antimicrobiana, o Ãleo OV possivelmente pelo maior teor de fenÃlicos, apresentou aÃÃo mais efetiva contra microrganismos patogÃnicos, em especial Listeria mocytogenes. Os Ãleos de amÃndoa de castanha de caju OV e OEV apresentaram aÃÃo antiproliferativa contra cÃlulas de cÃncer de mama (MCF7), pulmÃo (A549) e fÃgado (HepG2) somente em altas concentraÃÃes, superiores a 200 Âg/mL. Nos testes sensoriais com consumidores (grupos focais) e na AnÃlise Descritiva Quantitativa Otimizada percebeu-se interesse em consumir o produto caso estivesse disponÃvel no mercado, tanto por seus atributos sensoriais como pelo apelo regional do produto. Os principais atributos das amostras foram brilho, cor amarela, odor e sabor de castanha. Dentro dos aspectos avaliados, o aquecimento prÃvio com micro-ondas pode oferecer um maior rendimento de extraÃÃo, sem perdas significativas dos compostos bioativos ou diminuiÃÃo da aceitaÃÃo sensorial. / The cashew nut (CN) is a major agribusiness product of CearÃ, but it is also an income source for small farmers, especially in drought periods. The cashew nut with greater value to market is standardized as whole-grade. The cashew nut inferior grades by color or broken during processing, lose 80% of their market value. This study aimed to develop an oil from the inferior grades cashew nut, evaluating the cold extraction methods (extra virgin - EVCNO) and preheating in microwave (virgin - VCNO), its composition and chemical and physical characteristics, stability, sensory characteristics and bioactivity. In the selection of kernels for extraction, considered the physical and chemical characteristics and oxidative quality, we used lower six grades and qualities: B1, S1, W4, SP1, SP2, P1 and a whole nut-grade grade (W240) as reference. Considering the evaluated parameters, we opted for the use of SP2 kernel, due to the lower market price, since all grades and qualities evaluated showed good physical and chemical quality and oxidative, with potential for oil extraction (total lipids of over 50%). In the assessment of extraction processes, there was a higher yield of the sample with pre-heating in a microwave. The OV sample also showed greater oxidative stability with average induction time of 46.25 hours, 39.03 hours against the OEV sample, possibly given by the greater presence of phenolic compounds in sample VCNO (316.4 mg / 100g) than the sample EVCNO (202.17 mg / 100g). Both oils showed antioxidant activity, possibly due to the presence of compounds such as tocopherols, fatty acids mono and polyunsaturated, phytosterols and phenolic identified in the samples. In antimicrobial evaluation, the VCNO oil possibly due to increased phenolic content, made more effective action against pathogenic microorganisms especially Listeria mocytogenes. VCNO and EVCNO showed antiproliferative action against breast cancer cells (MCF7), lung cancer cells (A549) and liver cancer cells (HepG2) only at high concentrations greater than 200 mg/mL. In sensory tests with consumers (focus groups) and Optimized Descriptive Profile perceived interest in consuming the product if it was available in the market, both for their sensory attributes as the regional appeal of the product. The main attributes of the samples: shining, yellow color, odor and taste of nuts. Considering the evaluated aspects, preheating with microwave can offer a higher yield of extraction, without significant loss of bioactive compounds or decreased sensory acceptance.

Résumé : Lobjectif des travaux entrepris au cours de cette thèse visait à mettre en place un procédé global de conservation des graines de grenade (Punica granatum L.). Ce procédé se base essentiellement sur une déshydratation osmotique (DO), associée à un pré-traitement de congélation et un post-traitement de séchage par entrainement. Dans ce contexte, plusieurs paramètres d'optimisation du transfert de masse ont été étudiés, tels que la nature de la solution dimmersion (saccharose, glucose, glucose/saccharose et jus de datte « sous-produit » enrichi en saccharose), la température (30, 40, et 50°C) et létat du fruit (frais, congelé). En outre, nous avons mis en relation ces conditions avec certaines propriétés des graines : leur texture, leur structure, et leur couleur. Létude des paramètres de déshydratation (perte en eau (WL), gain en solides (SG), et réduction en poids (WR)) a montré quen utilisant des graines congelées et indépendamment de la température et de la solution utilisée, la majorité du transfert de masse seffectue pendant les vingt premières minutes de traitement. A lissue de cette période, la perte en eau est estimée à 46%, 41%, 39%, et 37% respectivement dans les solutions de saccharose, glucose/saccharose, de jus de datte et du glucose. La DO des graines fraîches est caractérisée par une cinétique plus lente, mais une perte finale en eau plus importante. Comme le montrent les analyses en microscopie électronique, cela sexplique par une déstructuration cellulaire survenant à la suite de la congélation des graines, ce qui vient conforter les résultats des observations microscopiques. Les mêmes techniques ont également indiqué une modification de texture/structure induite par le processus de DO. Dautre part, lutilisation dune solution de saccharose (55°Brix) et dune température de 50°C favorise un meilleur transfert de masse. La détermination des différentes fractions deau dans la graine par calorimétrie différentielle (DSC) a montré une augmentation dun facteur ~2,5 fois de la fraction deau non congelable (eau liée) et une réduction de ~3,5 fois de la fraction deau congelable (eau libre) favorisant ainsi une meilleure conservation du fruit. Le suivi de la qualité intrinsèque des graines au cours de la DO a montré une perte dune quantité non négligeable de certains composés (protéines, cendres) de la graine vers la solution, ce qui pourrait avoir une influence majeure sur la qualité organoleptique et nutritionnelle du fruit. La DO seule ne pourrait pas maintenir une stabilité du produit au cours de la conservation. En effet, lactivité deau du produit fini après DO est proche de 0,90. Ainsi, dans un but plus appliqué, un traitement complémentaire de séchage par entrainement (2 m/s durant 4 heures) a été mis en place, à différentes températures (40, 50, et 60°C), afin de réduire lactivité deau à une valeur inférieure à 0,65. Afin doptimiser le traitement de séchage, nous avons étudié en premier lieu leffet de la température sur lévolution de la matière sèche, de lactivité deau et du pourcentage de séchage des graines. Dautre part, plusieurs paramètres de qualité des graines de grenade (lactivité antioxydante, la teneur en composés phénoliques, les anthocyanines, la couleur, et la texture) ont été étudiés à différentes températures. Ce travail est une contribution à létude des propriétés physico-chimiques des graines de grenade (Punica granatum L.) au cours des procédés de congélation, de déshydratation et de séchage. Les caractéristiques du produit fini peuvent justifier de nouvelles voies de transformation et dexploitation des graines de grenade. Abstract: The aim of this work was to create a complete conservation process of pomegranate seeds (Punica granatum L.). This process is essentially based on osmotic dehydration (OD), which was associated to freezing and air-drying process. Several parameters were studied to optimize the process such as osmotic solution (sucrose, glucose, and sucrose/glucose and date juice with sucrose added), temperature (30, 40, and 50°C) and state of the fruit (fresh and frozen). All these conditions were linked to seed proprieties (texture, structure, and colour). The study of osmotic dehydration parameters (water loss (WL), solids gain (SG) and weight reduction (WR)) showed that most significant changes of mass transfer took place during the first 20 min of dewatering using frozen seeds, independently of temperature and sugar type. During this period, seeds water loss was estimated at 46% in sucrose, 41% in sucrose/glucose mix, 39% in date juice, and 37% in glucose. Mass transfer was slower starting from fresh fruit but led to a higher rate of WL at the end of the process. This fact can be explained by scanning electron microscopy, which showed damage of seed cell structure after freezing. This has practical consequences in terms of the modification of seeds texture. The same process also revealed a modification of seed texture and cell structure after osmotic dehydration. Using a sucrose solution and a temperature of 50°C favoured the best mass transfer. The determination of different water fractions of seed by differential scanning calorimetry (DSC) showed that the % of frozen water decreased 3.5 times contrary the % of unfreezable water that increased 2.5 times. This favours a better seeds conservation. During osmotic dehydration, there was a non negligible leaching of natural solutes from seeds into the solution, which might have an important impact on the sensorial and nutritional value of seeds. Using only osmotic dehydration could not maintain the stability of seeds during conservation. In fact, after the osmotic process, water activity of seeds was found to be higher than 0.9, allowing to the development of microorganisms and some undesirable reactions. As a consequence, a drying of the pomegranate seeds (during four hours) was investigated at three different temperatures (40, 50, and 60 °C) with air flow rate of 2 ms-1. Prior to the drying process, seeds were osmodehydrated in a sucrose solution (55°Brix) during 20 min at 50°C. The drying kinetics and the effects of OD and air-drying temperature on antioxidant capacity, total phenolic, colour, and texture were determined. This work is a contribution to the study of physico-chemical properties of pomegranate seeds (Punica granatum L.) during freezing, osmotic dehydration and drying. After the global process, the pomegranate seed characteristics lead to new industrial developments.

Development of antioxidant dietary fibers from wine grape pomace and their applications as functional food ingredients

Wine grape pomace (WGP), the byproduct from winemaking, is a good source of polyphenols and dietary fibers, and may be utilized as antioxidant dietary fibers (ADF) for food applications. The objectives of this thesis research were to first determine the phenolic compounds, antioxidant and antimicrobial activities in red WGP under different drying processes for long-term storage, and to further evaluate the feasibility of using WGP as a functional food ingredient in yogurt and salad dressing for enhancing the nutritional value and improving storability of the products. Two types of WGP samples, pomace containing seeds and skins (P) and pomace with skins only (S) from Pinot Noir (PN) and Merlot (M) were studied. Samples were subjected to four different drying conditions: 40 °C conventional and vacuum oven, 25 °C ambient air and freeze dry. Total phenolic content (TPC, by Folin-Ciocalteu assay), anthocyanins (ACY, by pH differential method) and flavanols content (TFC, by vanillin assay) of the samples along with their antioxidant activity (DPPH radical scavenge method, RSA) and antibacterial activity (minimum inhibition concentration, MIC) were determined during 16 weeks of storage under vacuum condition at 15±2 °C. Meanwhile, dietary fiber profile was evaluated by using gravimetric-enzyme method. Results showed that dietary fiber contents of PN-P, PN-S, M-P and M-S were 57-63% d.m. with the majority of insoluble fraction. Freeze dried WGP retained the highest bioactive compounds with TPC 21.19-67.74 mg GAE/g d.m., ACY of 0.35-0.76 mg Mal-3-glu/g d.m., TFC of 30.16-106.61 mg CE/g d.m. and RSA of 22.01-37.46 mg AAE/g d.m., followed with ambient air dried samples. Overall, TPC, TFC and RSA were higher in PN than in M, and higher in pomace than in skins, while reverse results were observed in ACY. All samples lost significant amount of bioactive compounds during storage, in which ambient air and freeze dried samples had TPC reduction of 32-56% and 35-58%, respectively at the end of 16 weeks of storage. RSA in PN-P and M-P remained more than 50 mg TE/g d.m., meaning WGP still met the criteria of ADF definition after 16 weeks of storage. WGP extracts showed higher antibacterial efficiency against L. innocua than that of E. coli with MIC of 2, 7, 3 and 8% against L. innocua, and 3, 6, 4 and 9% against E. coli for PN-P, PN-S, M-P and M-S samples, respectively. This study demonstrated that Pinot Noir and Merlot pomace are good sources of ADF even after 16 weeks of storage at 15 °C and vacuum condition. Due to the highest antioxidant activity (RSA 37.46 mg AAE/g) and dietary fiber content (61%), PN-P was selected as ADF to be fortified in yogurt and salad dressing. Three types of WGP: whole powder (WP), liquid extract (LE) and freeze dried extract (FDE) with different concentrations were incorporated into yogurt (Y), Italian (I) and Thousand Island (T) salad dressings. TPC, RSA and dietary fiber content, major quality attributes including pH and peroxide value (PV) during the shelf life and consumer acceptance of fortified products were evaluated. The highest ADF were obtained in 3% WP-Y, 1% WP-I and 2% WP-T samples with the dietary fiber contents of 1.98%, 2.12% and 1.83% and RSA of 935.78, 585.60 and 706.67 mg AAE/kg, respectively. WP fortified products had more dietary fiber content than that of LE and FDE fortified ones because of the insoluble fractions. The pH dropped from 4.52 to 4.32 for 3% WP-Y during three weeks of storage at 4 °C, but remained stable in WGP-I and WGP-T samples after four weeks of storage at 4 °C. Adding WGP resulted in 35-65% reduction of PV in all samples compared to the control. In WGP-Y, the viscosity increased, but syneresis and lactic acid percentage were stable during storage. The 1%WP-Y, 0.5%WP-I and 1%WP-T samples were mostly liked by consumers. Study demonstrated that WGP can be used as a functional food ingredient for enhancing nutraceutical content and extending shelf-life of the food products. This study provided important information about the economically feasible drying methods for retaining the bioactive compounds in WGP during processing and storage and also suggested that WGP can be utilized as antioxidant dietary fiber to be fortified in consumer products to promote nutritional benefit and extend product shelf-life. / Graduation date: 2013

Effect of Rootstock on the Fruit Quality of Mandarins "Clemenules" and "Tango", and Blood Oranges "Tarocco Rosso" and "Moro"

Tesis por compendio / [ES] La citricultura se enfrenta a escenarios ambientales cambiantes que provocan diferentes estreses bióticos y abióticos que pueden dificultar la producción o afectar a la calidad de la fruta. El patrón sobre el cual se injerta una variedad específica es una importante herramienta para mejorar su adaptabilidad a cada área de cultivo. En la presente Tesis se realiza el estudio del efecto del patrón sobre la calidad físico-química y nutricional de la fruta en variedades de gran interés comercial en la actualidad, mandarinas 'Clemenules' y 'Tango' y naranjas sanguinas 'Tarocco Rosso' y 'Moro'. En 'Clemenules' se llevó a cabo la evaluación de la calidad de la fruta de árboles injertados sobre ocho patrones en tres momentos de cosecha durante dos campañas. Los patrones Forner-Alcaide 13 y C-35 destacaron por adelantar el cambio de color, lo que es de gran interés comercial. Por otra parte, Forner-Alcaide V17 destacó por mantener niveles óptimos de acidez hasta el final de la campaña y presentar el mayor contenido en vitamina C, flavonoides, glucosa y fructosa. Carrizo Citrange también indujo altas concentraciones de sacarosa y vitamina C. 'Tango' es una mandarina de reciente introducción en el área mediterránea de gran interés por su periodo de recolección tardío. En esta Tesis se abordó el estudio de los cambios en la calidad fisicoquímica, nutricional y sensorial de la mandarina 'Tango' injertada sobre dos patrones (Carrizo Citrange y Forner-Alcaide 5) en las dos áreas principales de producción de Andalucía. Los resultados revelaron que la calidad de la fruta se vio influenciada por la localización de las parcelas, lo que se relacionó con la composición de la textura del suelo. En ambas localizaciones, Forner-Alcaide 5 indujo mayor contenido en acidez, sólidos solubles totales, sacarosa, vitamina C y ácido cítrico en la fruta. Las determinaciones físico-químicas, junto con la evaluación sensorial permitieron establecer el momento óptimo de recolección dependiendo de las diferentes condiciones estudiadas. También se ha incluido un estudio del comportamiento postcosecha de esta variedad, ya que no existían datos en nuestras condiciones de cultivo. La mandarina 'Tango' presentó síntomas externos de daños por frío a partir de los 20 días almacenada a 1°C y 5°C. Se realizó un estudio micro-estructural para caracterizar la alteración. Los frutos del patrón Forner-Alcaide 5 presentaron una menor incidencia de los daños por frío. El almacenamiento a 9°C no comprometió la calidad externa o interna. Dentro del grupo de naranjas, en los últimos años existe una creciente demanda por las naranjas sanguinas para su consumo en fresco. Para evaluar el efecto del patrón se tomaron las variedades 'Moro' y 'Tarocco Rosso' injertadas sobre ocho patrones. La calidad interna se vio afectada por el momento de cosecha, siendo más evidente en 'Moro'. En ambas variedades el patrón afectó a los parámetros de calidad durante la maduración. En 'Moro', se observó una reducción en el color del zumo debido a la degradación de los antocianos, siendo los patrones C-35, Macrophylla y Volkameriana los que mostraron un mayor descenso. Esta degradación se relacionó con los cambio de la temperatura durante el periodo de recolección. En 'Tarocco Rosso' no se evidenció ninguna degradación, lo que sugiere que esta variedad es menos sensible a los cambios de temperatura. En esta variedad Forner-Alcaide 5 y Forner-Alcaide 13 produjeron fruta con mayor contenido en antocianos y azúcares. Además se llevó a cabo el estudio de la aptitud a la frigoconservación de las variedades de sanguinas 'Tarocco Rosso' y 'Sanguinelli'. Mientras que la calidad interna no se vio afectada por el almacenamiento (1°C, 5°C y 9°C) durante 45 días, la fruta presentó síntomas de daños por frío a 1°C. 'Sanguinelli' presentó mayor incidencia que 'Tarocco Rosso'. Estas sanguinas se pueden almacenar entre 5 y 9°C / [CA] La citricultura s'enfronta constantment a escenaris ambientals canviants que provoquen diferents estressos biòtics i abiòtics. El patró sobre el qual s'empelta una varietat específica és una important eina per a millorar la seua adaptabilitat agronòmica en cada àrea de cultiu. En la present Tesi s'ha dut a terme l'estudi de l'efecte del patró sobre la qualitat físic-química i nutricional de la fruita en varietats de gran interés comercial, mandarines 'Clemenules' i 'Tango' i, taronges sanguines 'Tarocco Rosso' i 'Moro'. En 'Clemenules' es va dur a terme l'avaluació de la qualitat de la fruita d'arbres empeltats sobre huit patrons en tres moments de collita, en dues campanyes. Entre els patrons avaluats, Forner-Alcaide 13 i C-35 van destacar per avançar el canvi de color. D'altra banda Forner-Alcaide V17 va destacar per mantindre nivells òptims d'acidesa fins al final de la campanya i va presentar el major contingut en vitamina C, flavonoides, glucosa i fructosa. Carrizo Citrange també va induir altes concentracions de sacarosa i vitamina C en la fruita. 'Tango' és una mandarina de recent introducció en l'àrea mediterrània amb gran interés pel seu període de recol·lecció que comença quan finalitza el de les clementines. En la present Tesi es van estudiar els canvis en la qualitat físic-química, nutricional i sensorial de la mandarina Tango empeltada sobre dos patrons (Carrizo Citrange i Forner-Alcaide 5) durant el període de collita en les dues àrees principals de producció d'Andalusia. La qualitat de la fruita es va veure influenciada per la localització, la qual cosa es va relacionar amb la composició de la textura del sòl. En totes dues localitzacions, Forner-Alcaide 5 va ser el patró que va induir major contingut en acidesa, sòlids solubles totals, sacarosa, vitamina C i àcid cítric en la fruita. Les determinacions físic-químiques i l'avaluació sensorial van permetre establir el moment òptim de recol·lecció depenent de les diferents condicions estudiades. També s'ha inclòs un estudi del comportament postcollita d'aquesta varietat, ja que no existien dades en les nostres condicions de cultiu. La mandarina 'Tango' va presentar símptomes externs de danys per fred a partir dels 20 dies emmagatzemada a 1°C i 5°C. Es va realitzar un estudi micro-estructural per a caracteritzar l'alteració provocada per les baixes temperatures. Els fruits del patró Forner-Alcaide 5 van presentar una menor incidència dels danys per fred. L'emmagatzematge a 9°C no va comprometre la qualitat externa o interna d'aquesta varietat. Dins del grup de taronges, en els últims anys existeix una creixent demanda per les taronges sanguines pel seu consum en fresc. Per a avaluar l'efecte del patró sobre sanguines es van prendre dues varietats, 'Moro' i 'Tarocco Rosso' empeltades sobre huit patrons. La qualitat interna es va veure influenciada pel moment de collita, la qual cosa va ser més evident en la varietat 'Moro'. En totes dues varietats el patró va afectar els canvis en els paràmetres de qualitat estudiats. En 'Moro', es va observar una reducció en el color del suc degut a la degradació del antocians. Aquesta degradació es va relacionar amb el canvi de la temperatura experimentada durant el període de recol·lecció. 'Tarocco Rosso' és menys sensible als canvis de temperatura. En aquesta varietat els patrons Forner-Alcaide 5 i Forner-Alcaide 13 van produir la fruita amb major contingut en antocianos i sucres. A mes s'aporta l'estudi de l'aptitud a la frigoconservació en les sanguines 'Tarocco Rosso' i 'Sanguinelli'. Mentre que la qualitat interna no es va veure afectada per l'emmagatzematge a cap de les temperatures assajades (1°C, 5°C i 9°C) durant 45 dies, la fruita va presentar símptomes de danys per fred a 1°C. `Sanguinelli' va presentar major incidència que 'Tarocco Rosso'. La fruita es pot emmagatzemar entre 5°C i 9°C durant 30 dies en el cas de 'Sanguinelli' i fins a 45 en e / [EN] Citriculture faces changing environmental scenarios that cause biotic and abiotic stress. The rootstock onto which a specific variety is grafted is an important tool to help to improve its agronomic adaptability to each crop area. The present Thesis was carried out to study the effect of rootstock on physico-chemical and nutritional fruit quality in some varieties of commercial interest today: 'Clemenules' and 'Tango' mandarins, and 'Tarocco Rosso' and 'Moro' blood oranges. In 'Clemenules', the fruit of the trees grafted into eight rootstocks at three harvest times was evaluated by performing studies during two seasons. Forner-Alcaide 13 and C-35 Citrange stood out for their earlier color change, which is very interesting for this variety, in which early harvesting is a relevant aspect from the commercial point of view. Forner-Alcaide V17 stood out for maintaining optimum acidity levels until the season ended and presented the highest contents in vitamin C, flavonoids, glucose and fructose. Carrizo Citrange brought about high concentrations of sucrose and vitamin C in fruit. 'Tango' is a mandarin variety that has been recently introduced into the Mediterranean Region. Its harvest time is very interesting because it starts when that of clementines ends. The present Thesis studies changes in the physico-chemical, nutritional and sensorial quality of 'Tango' fruit grafted onto two rootstocks (Carrizo Citrange and Forner-Alcaide 5) during the harvest period in the two main production areas in Andalusia. The results revealed that fruit quality during harvest was influenced by the location, which was particularly related to soil texture composition. In both areas, Forner-Alcaide 5 was the rootstock that induced higher acidity content, and more total soluble solids, sucrose, vitamin C and citric acid in fruit. The physico-chemical determinations, along with the sensorial evaluation, allowed the optimum harvest time to be established depending on the different studied conditions. This Thesis also includes a study about this variety's postharvest behavior as no data are available for our crop conditions. The 'Tango' mandarin presented outer chilling injury symptoms after being stored for 20 days at 1°C and 5°C. A microstructural study was done to characterize the alteration caused by low temperatures. The Forner-Alcaide 5 rootstock fruit showed a lower chilling injury incidence. Storage at 9°C did not compromise quality fruit. Among oranges, demand for blood oranges to be eaten fresh has grown in recent years, basically due to their high content in anthocyanins and their positive effect for human health. To assess the effect that rootstock had on blood oranges, two varieties were taken, 'Moro' and 'Tarocco Rosso', grafted onto eight rootstocks. Internal quality was strongly influenced by harvest time, which was more evident for 'Moro'. In both varieties, rootstock affected changes in the quality parameter studied. In 'Moro', juice color faded as anthocyanins degraded, and rootstocks C-35 Citrange, Macrophylla and Volkameriana showed the most marked reduction. Such anthocyanin degradation was related to the change in temperature that took place during the harvest period. In 'Tarocco Rosso', anthocyanins did not undergo degradation, which suggests that this variety is less sensitive to changes in temperature. In this variety, rootstocks Forner-Alcaide 5 and Forner-Alcaide 13 gave fruit with a higher content of anthocyanins and sugars. This Thesis also includes a study of the suitability of cold storage of two blood orange varieties: 'Tarocco Rosso' and 'Sanguinelli'. Although storage at any tested temperature (1°C, 5°C and 9°C) did not affect internal quality for 45 days, fruit displayed chilling injury symptoms at 1°C, with a higher incidence for 'Sanguinelli' than for 'Tarocco Rosso'.
