Initially, it has been evaluated the best pH for each coagulant studied to then optimize the concentration of the coagulants and settling time. For this finality, it has been used velocity of rapid mixing (VMR) and slow (VML) at 100 rpm and 20 rpm, respectively and time of rapid mixing (TMR) and slow (TML) for 2 minutes and 20minutes, respectively. The parameters analyzed were pH after treatment, absorbance at a wavelength of maximum absorption for each dye present in the wastewater (Procion Orange HER and Procion Red HE-7B), color, turbidity and COD. In the test for optimization of coagulants concentration and settling time, also was evaluated the amount of sludge generated at each concentration studied. Thus, organic coagulants showed superior results in comparison with inorganic coagulants in the treatment of textile wastewater, especially MO-KCl, which showed removal of 86,8% for absorbance of the dye Procion Orange HER, 75,65% for absorbance of the dye Procion Red HE-7B, 70,67%forcolor, 85,97% for COD and 64,07% for turbidity. Based on the results obtained, MO-KCl presented a promising coagulant in the treatment of textile wastewater. / O presente estudo avaliou a eficiência dos coagulantes orgânicos sementes de Moringa oleifera lam extraídas em soluções salinas de cloreto de sódio e cloreto de potássio 1,0 (MO-NaCl e MO-KCl) respectivamente, além dos inorgânicos sulfato de alumínio e cloreto férrico no tratamento de água residuária têxtil. Inicialmente, avaliou-se o melhor pH de trabalho para cada coagulante estudado, para, em seguida, otimizar a concentração do coagulante e o tempo de sedimentação. Para isto, utilizou-se velocidade de mistura rápida (VMR) e lenta (VML) de 100 rpm e 20 rpm respectivamente, e tempo de mistura rápida (TMR) e lenta (TML) de 2 minutos e 20 minutos, respectivamente. Os parâmetros analisados foram pH após tratamento, absorbância no comprimento de onda máximo de cada corante presente na água residuária (Laranja Procion HER e Vermelho Procion HE-7B), cor, turbidez e DQO. Além destes, no teste para otimização da concentração de coagulante e tempo de sedimentação, avaliou-se a quantidade de lodo gerado em cada concentração estudada. Assim, os coagulantes orgânicos apresentaram resultado superior aos inorgânicos no tratamento da água residuária têxtil, com destaque para a MO-KCl, que apresentou remoção de 86,8% para absorbância do corante Laranja Procion HER, 75,65% para absorbância do corante Vermelho Procion HE-7B, 70,67% para cor aparente, 85,97% para DQO e 64,07% para turbidez. Com base nos resultados obtidos a MO-KCl pode ser considerada um coagulante promissor para o tratamento da água residuária têxtil.

Micro-organismos bioprotetores em salames com baixo teor de sal e seus efeitos sobre a qualidade global e sensorial / Bacteriocin-producingmicroorganismsin low-sodium salami and their effects on global and sensory quality

As mudanças dos hábitos de consumo do consumidor, força o desenvolvimento de produtos industrializados mais saudáveis. Assim, pesquisadores em ciência e tecnologia de produtos cárneos, têm focado parte de suas pesquisas no desenvolvimento de produtos com níveis reduzidos de sódio. O presente estudo teve como objetivo buscar por micro-organismos bioprotetores e proteolíticos para o uso em salames com baixo teor de cloreto de sódio (NaCl). O primeiro capitulo consiste em uma revisão de literatura, sendo o segundo o desenvolver salames italianos com 1,0% NaCl com a adição ou não de cloreto de potássio (KCl) e cloreto de cálcio (CaCl2) com características sensoriais semelhantes aos descritos no salames comerciais com 2,5% de NaCl adicionado. Os resultados mostraram diferenças de pH e Aw, devido à redução de NaCl. As misturas substitutas de sal (KCl/CaCl2) nos salames não afetaram tecnologicamente a elaboração de salames. Para os consumidores, esta substituição não acarretou diferenças significativas (p> 0,05) na aparência (cor), porém observou-se diferenças nos demais atributos. Com relação ao teor de sal, os consumidores consideraram os tratamentos com baixo teor de NaCl adicionados dos substitutos KCl e CaCl2 como ideais. No terceiro capitulo, o objetivo foi analisar os aldeídos produzidos pela lipólise e proteólise assim como a aceitação sensorial de salames tipo Italiano com reduzido conteúdo de sódio. Das seis amostras elaboradas, todas foram monitoradas quanto ao pH e atividade de água e após a maturação do salame, foi realizada analise do perfil de voláteis e comparado com a sensorial. Os resultados mostraram a existência de uma relação entre a concentração de aldeídos e aceitação sensorial. Com a finalidade de elucidar a proteólise dos micro-organismos escolhidos, foi realizado no quarto capitulo um estudo com modelos Beaker Sausage (BS) para simular a produção de salame com baixo conteúdo de sódio para avaliar a produção de aminoácidos e peptídeos de culturas starters bacteriocigenicas. O BS contendo Enterococcus mundtii CRL35 apresentou a maior variedade de peptídeos pequenos, seguido pela BS contendo L. curvatus CRL1862. Embora o modelo de carne inoculadas com Lactobacillus plantarum CRL681 produziu o maior número de peptídeos derivados de proteínas sarcoplasmáticas. A pequena quantidade considerável de péptido produzido no BS inoculado permitiu inferir que os conteúdos mais baixos de sódio não afetam negativamente a ação das peptidases microbianas. O objetivo do último capitulo, foi avaliar o efeito das alegações de saúde sobre as aceitação dos salames tipo Italiano com reduzido conteúdo de sódio. Neste estudo, foi avaliada a expectativa gerada pelas alegações de saúde e os resultados mostraram que as alegações tiveram pouco efeito na aceitação dos consumidores decorrentes em valores de aceitação baixo para o protótipo de laboratório. Assim, pode-se concluir que a produção de salames tipo Italiano com teores reduzidos de NaCl, adicionados de CaCl2+ KCl é viável com a pouca adição destes substitutos e o uso de culturas starters bacteriocigênicas, que também são altamente proteolíticas de ser melhor estudado a fim de adequar a proteólise ideal para a formação dos compostos desejáveis no salame com baixo teor de sódio. / Changes in consumers\' eating habits and lifestyle have required the development of healthier processed products. Thus, researchers in science and meat products technology has focused their studies on the development of products with reduced sodium contents. This study investigated the use of bioprotectants and proteolytic microorganisms for low-sodium NaCl in salami. The chapter 1 consists in a literature review. In the second chapter, the objective was to develop Italian salami with 1.0% NaCl with or without the addition of potassium chloride (KCl) and calcium chloride (CaCl2) with sensory characteristics similar to those described in the commercial salami with 2.5% NaCl added. The results showed differences in pH and water activity (Aw) due to the reduction of NaCl. Salt substitutes chloride (KCl / CaCl2) in salamis did not technologically affectsalami preparation. For consumers, this replacement did not cause significant differences (p> 0.05) in appearance (color), but there were differences in other attributes. Regarding the salt content, consumers considered treatment with low NaCl content and added with KCl and CaCl2 as ideal substitutes. In the third chapter, the goal was to analyze the aldehydes produced by lipolysis and proteolysis as well as sensory acceptance of the Italian salami type with reduced sodium content. Allsix samples preparedwere monitored for pH and Aw and after salami maturation, thevolatile profile analysis was performed and compared with the sensory analysis. The results show the existence of a relationship between the concentration of aldehydes and sensory acceptance. In order to elucidate proteolysis of selected microorganisms, in the fourth chapter a study of models Beaker Sausage (BS) was conducted to simulate production of low-sodium salami to evaluate the production of amino acids and peptides of bacteriocigenicstarter cultures. The BS containing Enterococcus mundtii CRL35 showed the greatest variety of small peptides, followed by BS containing Lactobacilluscurvatus CRL1862. However, the meat model inoculated with Lactobacillus plantarum CRL681 produced the largest number of sarcoplasmic proteins derived from peptides. A considerable number of peptides produced in the BS inoculated allowed to infer that lower sodium content did not affect the action of microbial peptidases. The purpose of the last chapter was to evaluate the effect of health claims on the acceptance of Italian-type salami with reduced sodium content. In this study, the expectation generated by health claims wereanalyzed and the results showed that health claims had little effect on the increase of consumer acceptance for the prototype developed in the laboratory. Thus, it can be concluded that the production of Italian-type salami with reduced levels of NaCl, KCl + CaCl2 added is feasible but low addition of these substitutesmust be considered. Moreover, the use of bacteriocigenicstarter cultures, highly proteolytic to achieve optimal proteolysis in the formation of the desired compounds, should adjusted to low-sodiumsalami.

La expansividad de suelos es un fenómeno ampliamente reconocido, al cual se le atribuye una copiosa cantidad de problemas estructurales suscitados a nivel mundial. Precisamente, la incidencia de su comportamiento en los antecedentes históricos registrados, propició una marcada tendencia científica por estudiar los factores desencadenantes, así como encontrar técnicas y/o materiales eficientes para su estabilización. En ese afán, la presente investigación pretende evaluar la contribución del cloruro de potasio en el comportamiento expansivo de suelos arcillosos de alta plasticidad. Para ello, se efectuaron ensayos de identificación mineralógica, caracterización física y determinación del grado de expansión, obteniendo como principal resultado una significativa reducción de los parámetros expansivos del suelo. / Soil swelling is a widely known problem, which is related to a huge number of structural problems all over the world. In fact, due to its negative behavior in structures, which have been recorded many times in preceding documents, experts have focused their aim on studying the factors that made expansive soils extremely dangerous, as well to find techniques and/or materials that assure the efficiency of their stabilization. In that commitment, this investigation pretends to evaluate the effects of potassium chloride in the expansive behavior of high-plasticity clay soils. La expansividad de suelos es un tema que ha generado sumo interés entre los especialistas geotecnistas dada la incidencia de su comportamiento en los daños estructurales experimentados históricamente a nivel mundial. Al respecto, numerosos estudios han centrado el foco de su investigación en evaluar la contribución de ciertos aditivos químicos sobre las propiedades de desempeño de los suelos problemáticos en materia, sumiéndose en una búsqueda constante de materiales alternativos sostenibles y eficientes para su estabilización. En este contexto, el presente proyecto de investigación se enfoca en evaluar el efecto de la adición de un agente estabilizador, como el cloruro de potasio KCl, en el grado de expansión de suelos arcillosos con alto potencial de hinchamiento en la Urb. Los Vencedores 2da Etapa, en la ciudad de Talara. Para este fin, se realizaron, en primera instancia, ensayos de identificación mineralógica y caracterización física para una muestra inalterada en bloque (Mib) extraída de la zona de estudio. Adicionalmente, se efectuaron ensayos de determinación del potencial de expansión para muestras patrón y grupos posprueba con niveles de presencia de cloruro de potasio en concentraciones de 2%, 4%, 6% y 8% en peso. Los resultados obtenidos demuestran la efectividad del agente químico propuesto en la inhibición de la expansión de los suelos arcillosos, con un porcentaje de reducción máximo equivalente a 92% para la muestra con 6% de cloruro de potasio, siendo esta la concentración óptima. / The issue of the swelling soils has generated a significant interest among the specialists in geotechnics given the incidence of its behavior in the structural damages ever experienced in history. In this regard, multiple studies have focused their research on evaluation of the contribution of some chemical additives in the development of properties of problematic soils in matter, getting into a constant search of alternative sustainable and efficient materials for its stabilization. In this environment, the present project focuses on evaluate the effect on the addition of a stabilizing agent, like potassium chloride (KCl), in the expansion rate of clayey soils with high swelling potential in “Urb. Los Vencedores 2da Etapa, Talara”. For that purpose, there has been made, for instance, mineralogical identification and physical characterization tests for an unspoilt sample in bulk extracted from the study area. In addition, there were made tests to determinate the swelling potential for pattern samples plus post-tests groups with potassium chloride presence in 2%, 4%, 6% and 8% concentration. Results shown that the chemical agent proposed in the inhibition of swelling clayey soils was effective, with a maximum reduction of 92% for the sampling and 6% of potassium chloride, making this concentration the ideal one. / Tesis

Postnatale Entwicklung der striatalen GABAergen Interneurone im dtsz Hamster als Dystoniemodell: Untersuchungen des Homöodomänproteins Nkx 2.1, des Kalium-Chlorid-Kotransporters KCC2, der Carboanhydrase CAH7 und des Wachstumsfaktors BDNF

Einleitung: Bei der Dystonie handelt es sich um eine Erkrankung des zentralen Nervensystems. Sie ist charakterisiert durch ungewollte, dauerhafte oder wiederkehrende Muskelkontraktionen, die zu abnormalen Bewegungsabläufen und Haltungen führen. Sie ist die dritthäufigste Bewe-gungsstörung beim Menschen. Bisherige Befunde beim Menschen und Untersuchungen an Tier-modellen weisen u.a. auf eine besondere Bedeutung der Basalganglien-Thalamus-Schleife hin, die an der Kontrolle von willkürlichen und unwillkürlichen Bewegungen beteiligt ist. So konnten bei unterschiedlichen Tiermodellen Veränderungen im Striatum (STR), der Eingangsstruktur der Basalganglien, nachgewiesen werden. Beim dtsz Hamster, einem gut etablierten Tiermodell für die paroxysmale Dystonie, konnte neben vielen striatalen Veränderungen eine Reduktion der GABAergen Interneurone (IN), wie Parvalbumin-positive (PV+) IN, zum Zeitpunkt der maximalen Ausprägung der Dystonie gezeigt werden. Ziele der Untersuchung: Die Gründe für den Mangel an striatalen GABAergen IN bei der dtsz Hamstermutante sollten weiter untersucht werden, indem der Frage nachzugehen war, ob beim dtsz Hamster eine Migrations- oder Ausreifungsstörung der IN vorliegt. Dazu wurde das Homöodomänprotein Nkx 2.1, als Marker für aus dem medialen Ganglienhügel eingewanderte IN, im STR der dtsz Hamstermutante untersucht. Die Expression des Brain-derived neurotrophic factors (BDNF), des Kalium-Chlorid-Kotransporters 2 (KCC2) und die zytosolische Carboanhydrase vom Isotyp 7 (CAH7) wurden als Indikatoren für die Ausreifung von GABAergen IN herangezogen. Tiere, Material und Methoden: Die Untersuchungen wurden vergleichend an dtsz Hamstern und Kontrollhamstern durchgeführt. Beim dtsz Hamster zeigt die Dystonie einen altersabhängigen Verlauf (Beginn: ca. 16. Lebenstag (LT); Maximum: 30.-42. LT; Remission: 70. LT). Deshalb wurden als Untersuchungszeitpunkte der 18. LT und der 33. LT gewählt. Um die Migration der striatalen IN zu untersuchen, wurde im STR bei 33 Tage alten Hamsterns die Dichte der immun-histochemisch markierten Nkx 2.1-positiven Zellen stereologisch ermittelt. Der mRNA-Gehalt wurde relativ mittels „quantitativer Echtzeit-PCR“ (qPCR) bestimmt. Zusätzlich wurde die mRNA-Expression von Nkx 2.1 bei 18 Tage alten Tieren untersucht. Von KCC2 und CAH7 wurde die mRNA mittels qPCR bei 18 und 33 Tage alten Hamstern im STR untersucht. Die Expression von BDNF wurde mittels ELISA-System im Kortex (Cx), STR und im restlichen Gehirngewebe („R“) bei 33 und 18 Tage alten Tieren bestimmt. Der BDNF-mRNA Gehalt wurde im Cx (18. und 33. LT) und im STR (33. LT) untersucht. Des Weiteren sollte BDNF bei 33 Tage alten Hamstern mittels immunhistochemischer Markierung im Cx und STR untersucht werden. Die Untersuchung von BDNF im Cx ist deshalb wichtig, weil BDNF vom Cx in das STR trans-portiert wird. Zusätzlich wurde Parvalbumin (PV) zusammen mit Nkx 2.1 immunhistochemisch markiert und die mRNA-Expression von PV bei 18 und 33 Tage alten Tieren bestimmt. Ergebnisse: Für Nkx 2.1 konnte kein Unterschied in der Zelldichte zwischen dtsz- und Kontroll-hamstern gefunden werden. Ebenfalls gab es weder bei 18 noch bei 33 Tage alten Tieren einen Unterschied in der Nkx 2.1-mRNA-Expression. Unterschiede in der mRNA-Expression von KCC2 und CAH7 im STR (18. und 33. LT) lagen auch nicht vor. Die Expression der PV-mRNA im STR bei 33 Tage alten Tieren war jedoch erwartungsgemäß vermindert. Auf mRNA-Ebene konnte im Cx für BDNF kein Unterschied zwischen dtsz- und Kontrolltiergruppe gefunden wer-den. Bei beiden Tiergruppen wurde mittels ELISA im STR mehr BDNF nachgewiesen als im Cx und im R (18. und 33. LT) nachweisbar. Dies lag daran, dass die BDNF-Expression nur bei den Kontrolltieren am 33. LT im Vergleich zum 18. LT herunterreguliert war. Die Ergebnisse der BDNF-Immunhistologie waren in Hinblick auf die Spezifität zweifelhaft. Schlussfolgerung: Die Nkx 2.1 Daten lassen auf eine ungestörte Migration striataler IN bei der dtsz Mutante schließen. Wahrscheinlich ist eine Ausreifungsstörung für den Mangel an GABAer-gen IN verantwortlich. Die Ergebnisse von KCC2 und CAH7 zeigen, dass keine generelle Ausreifungsstörung von GABAergen Neuronen vorliegt, wobei dies für kleinere Subpopulationen nicht ausgeschlossen werden kann. Entgegen der Arbeitshypothese konnte keine Verringerung sondern eine leichte Erhöhung von BDNF zum 33. LT bei der dtsz Hamstermutante festgestellt werden. Eine mögliche Erklärung könnte sein, dass BDNF auf Grund der verzögert einsetzenden Entwicklung der IN nicht herunterreguliert wird. Die Gründe für diese Erhöhung wie auch weitere Marker für die neuronale Ausreifung werden durch weiterführende Studien untersucht.

However, too much can cause health problems to their consumers, such as hypertension. The objective of this research was partially replace sodium chloride by potassium chloride and a mixture of oregano (Origanum vulgare), basil (Ocimum basilicum) and rosemary (Rosmarinus officinalis), named Herbal Mix, initially in chicken breast fillets to determine an optimization interval of the variables to be tested in fresh sausages. Treatments were obtained using a Central Composite Rotational Design with three factors, resulting in 17 trials. Fillet of chicken breast with various flavors underwent Quantitative Descriptive Analysis for raising and quantify their sensory attributes. Subsequently, analyzes were carried out the physical parameters of quality (water holding capacity, cooking loss, shear force and color), to verify the influence of sodium chloride (NaCl), potassium chloride (KCl) and Herbal Mix on physical and sensory characteristics of meat. To check the possibility of further substitution of sodium chloride, found optimization intervals were studied in the development of fresh sausages, meat product to develop a low sodium content. The formulations of NaCl, KCl and Herbal Mix were obtained in a randomized mixture of three factors, the type simplex centroid, totaling 10 trials. The sausages were analyzed by sensory acceptance test (appearance, texture, flavor, global acceptance, purchase intention and intensity of salt), physical-chemical analysis (moisture, ash, proteins, lipids, pH and water activity) and physical (water holding capacity, cooking loss, shear force and color) to verify the influence of mixing of the quality factors of the samples. In the first study it was observed that, at 5% significance level, the response variables characteristic flavor, taste bitter and hardness influenced the statistical variables NaCl and Herbal Mix, while the attribute color gold was influenced by variables KCl and Herbal Mix . As for the physical parameters, only shear force of the treatments was influenced by variables, specifically KCl and Herbal Mix. To optimize a majority of the variables is chosen to optimize the following range, replacing initially 33% NaCl: 5-30 g / kg meat, 2-12 g / kg meat and 5-12 g / kg meat. Regarding the study of the reduction of optimized intervals showed that only the sensory analysis global acceptance and purchase intention influenced statistical variables. The physical-chemical properties, only the moisture and ash content were significantly influenced by the studied variables. None of the physical parameters was influenced by statistical factors. The experiment allowed a reduction of 20% sodium chloride without significant changes in quality of the sausage. With respect to experiment with chicken breast fillets, the use of potassium chloride and Herbal mix can replace sodium chloride up to 44,4%. / O cloreto de sódio é um componente essencial na condimentação dos alimentos, especialmente de carnes e seus derivados. Entretanto, seu excesso pode causar problemas de saúde a seus consumidores, a exemplo da hipertensão arterial. Objetivou-se nesta pesquisa substituir parcialmente o cloreto de sódio por cloreto de potássio e uma mistura de orégano (Origanum vulgare), manjericão (Ocimum basilicum) e alecrim (Rosmarinus officinalis), denominada de Mix de Ervas, inicialmente em filés de peito de frango, para determinar um intervalo de otimização das variáveis a serem testadas em linguiças de frango. Os tratamentos foram obtidos segundo um Delineamento Composto Central Rotacional com três fatores, totalizando 17 ensaios. Os filés de peito de frango com as diversas condimentações foram submetidos à Análise Descritiva Quantitativa para levantar e quantificar os seus atributos sensoriais. Posteriormente, foram realizadas as análises dos parâmetros físicos de qualidade (Capacidade de retenção de água, Perda de peso por cocção, Força de cisalhamento, e Cor objetiva), para verificar a influência do cloreto de sódio (NaCl), cloreto de potássio (KCl) e Mix de Ervas sobre as características físicas e sensoriais da carne. Para verificar a possibilidade de uma maior substituição de cloreto de sódio, os intervalos de otimização encontrados foram estudados no desenvolvimento de linguiças de frango, para desenvolver um produto cárneo com baixo teor de sódio. As formulações de NaCl, KCl e Mix de Ervas foram obtidas segundo um delineamento em mistura de três fatores, do tipo centroide simplex, totalizando 10 ensaios. As linguiças foram submetidas à análise sensorial mediante teste de aceitação (Aparência, Textura, Sabor, Aceitação Global, Intenção de compra e Intensidade de sal), análises físico-químicas (umidade, cinzas, proteínas, lipídeos, pH e atividade de água) e físicas (capacidade de retenção de água, perda de peso por cocção, força de cisalhamento e cor objetiva) para verificar a influência da mistura dos fatores na qualidade das amostras. No primeiro estudo foi observado que, em nível de 5% de significância, as variáveis resposta Sabor Característico, Gosto Amargo e Dureza sofreram influência estatística das variáveis NaCl e Mix de Ervas, enquanto o atributo Cor Dourada foi influenciado pelas variáveis KCl e Mix de Ervas. Quanto aos parâmetros físicos, apenas a Força de Cisalhamento dos tratamentos foi influenciada pelas variáveis, especificamente o KCl e o Mix de Ervas. Para otimizar a maioria das variáveis, foi escolhido o seguinte intervalo de otimização, que substituiu inicialmente 33% de NaCl: 5 a 30 g/kg de carne, 2 a 12 g/kg de carne e 5 a 12 g/kg de carne. Quanto ao estudo da redução dos intervalos otimizados observou-se que na Análise Sensorial apenas a Aceitação Global e a Intenção de compra sofreram influência estatística das variáveis. Nas análises físico-químicas, apenas a umidade e o teor de cinzas foram estatisticamente influenciados pelas variáveis em estudo. Nenhum dos parâmetros físicos sofreu influência estatística dos fatores. O experimento possibilitou uma redução de até 20% de cloreto de sódio sem alterações significativas de qualidade das linguiças. Com relação ao experimento com filés de peito de frango, o uso de cloreto de potássio e Mix de Ervas pode substituir o cloreto de sódio em até 44,4%.

Postnatale Entwicklung der striatalen GABAergen Interneurone im dtsz Hamster als Dystoniemodell: Untersuchungen des Homöodomänproteins Nkx 2.1, des Kalium-Chlorid-Kotransporters KCC2, der Carboanhydrase CAH7 und des Wachstumsfaktors BDNF

Einleitung: Bei der Dystonie handelt es sich um eine Erkrankung des zentralen Nervensystems. Sie ist charakterisiert durch ungewollte, dauerhafte oder wiederkehrende Muskelkontraktionen, die zu abnormalen Bewegungsabläufen und Haltungen führen. Sie ist die dritthäufigste Bewe-gungsstörung beim Menschen. Bisherige Befunde beim Menschen und Untersuchungen an Tier-modellen weisen u.a. auf eine besondere Bedeutung der Basalganglien-Thalamus-Schleife hin, die an der Kontrolle von willkürlichen und unwillkürlichen Bewegungen beteiligt ist. So konnten bei unterschiedlichen Tiermodellen Veränderungen im Striatum (STR), der Eingangsstruktur der Basalganglien, nachgewiesen werden. Beim dtsz Hamster, einem gut etablierten Tiermodell für die paroxysmale Dystonie, konnte neben vielen striatalen Veränderungen eine Reduktion der GABAergen Interneurone (IN), wie Parvalbumin-positive (PV+) IN, zum Zeitpunkt der maximalen Ausprägung der Dystonie gezeigt werden. Ziele der Untersuchung: Die Gründe für den Mangel an striatalen GABAergen IN bei der dtsz Hamstermutante sollten weiter untersucht werden, indem der Frage nachzugehen war, ob beim dtsz Hamster eine Migrations- oder Ausreifungsstörung der IN vorliegt. Dazu wurde das Homöodomänprotein Nkx 2.1, als Marker für aus dem medialen Ganglienhügel eingewanderte IN, im STR der dtsz Hamstermutante untersucht. Die Expression des Brain-derived neurotrophic factors (BDNF), des Kalium-Chlorid-Kotransporters 2 (KCC2) und die zytosolische Carboanhydrase vom Isotyp 7 (CAH7) wurden als Indikatoren für die Ausreifung von GABAergen IN herangezogen. Tiere, Material und Methoden: Die Untersuchungen wurden vergleichend an dtsz Hamstern und Kontrollhamstern durchgeführt. Beim dtsz Hamster zeigt die Dystonie einen altersabhängigen Verlauf (Beginn: ca. 16. Lebenstag (LT); Maximum: 30.-42. LT; Remission: 70. LT). Deshalb wurden als Untersuchungszeitpunkte der 18. LT und der 33. LT gewählt. Um die Migration der striatalen IN zu untersuchen, wurde im STR bei 33 Tage alten Hamsterns die Dichte der immun-histochemisch markierten Nkx 2.1-positiven Zellen stereologisch ermittelt. Der mRNA-Gehalt wurde relativ mittels „quantitativer Echtzeit-PCR“ (qPCR) bestimmt. Zusätzlich wurde die mRNA-Expression von Nkx 2.1 bei 18 Tage alten Tieren untersucht. Von KCC2 und CAH7 wurde die mRNA mittels qPCR bei 18 und 33 Tage alten Hamstern im STR untersucht. Die Expression von BDNF wurde mittels ELISA-System im Kortex (Cx), STR und im restlichen Gehirngewebe („R“) bei 33 und 18 Tage alten Tieren bestimmt. Der BDNF-mRNA Gehalt wurde im Cx (18. und 33. LT) und im STR (33. LT) untersucht. Des Weiteren sollte BDNF bei 33 Tage alten Hamstern mittels immunhistochemischer Markierung im Cx und STR untersucht werden. Die Untersuchung von BDNF im Cx ist deshalb wichtig, weil BDNF vom Cx in das STR trans-portiert wird. Zusätzlich wurde Parvalbumin (PV) zusammen mit Nkx 2.1 immunhistochemisch markiert und die mRNA-Expression von PV bei 18 und 33 Tage alten Tieren bestimmt. Ergebnisse: Für Nkx 2.1 konnte kein Unterschied in der Zelldichte zwischen dtsz- und Kontroll-hamstern gefunden werden. Ebenfalls gab es weder bei 18 noch bei 33 Tage alten Tieren einen Unterschied in der Nkx 2.1-mRNA-Expression. Unterschiede in der mRNA-Expression von KCC2 und CAH7 im STR (18. und 33. LT) lagen auch nicht vor. Die Expression der PV-mRNA im STR bei 33 Tage alten Tieren war jedoch erwartungsgemäß vermindert. Auf mRNA-Ebene konnte im Cx für BDNF kein Unterschied zwischen dtsz- und Kontrolltiergruppe gefunden wer-den. Solvent–Solute Interaction : Studied by Synchrotron Radiation Based Photo and Auger Electron Spectroscopies

Aqueous solutions were studied using photoelectron and Auger spectroscopy, based on synchrotron radiation and a liquid micro-jet setup. By varying the photon energy in photoelectron spectra, we depth profiled an aqueous tetrabutylammonium iodide (TBAI) solution. Assuming uniform angular emission from the core levels, we found that the TBA+ ions were oriented at the surface with the hydrophobic butyl arms sticking into the liquid. We investigated the association between ions and their neighbors in aqueous solutions by studying the electronic decay after core ionization. The (2p)−1 decay of solvated K+ and Ca2+ ions was studied. The main features in the investigated decay spectra corresponded to two-hole final states localized on the ions. The spectra also showed additional features, related to delocalized two-hole final states with vacancies on a cation and a neighboring water molecule. These two processes compete, and by comparing relative intensities and using the known rate for the localized decay, we determined the time-scale for the delocalized process for the two ions. We compared to delocalized electronic decay processes in Na+, Mg2+, and Al3+, and found that they were slower in K+ and Ca2+, due to different internal decay mechanisms of the ions, as well as external differences in the ion-solute distances and interactions. In the O 1s Auger spectra of aqueous metal halide solutions, we observed features related to delocalized two-hole final states with vacancies on a water molecule and a neighboring solvated anion. The relative intensity of these feature indicated that the strength of the interaction between the halide ions and water correlated with ionic size.
