Efeitos do alumínio sobre a fermentação alcoólica. / Aluminum effects toward alcoholic fermentation.

O presente trabalho teve por objetivo estudar distúrbios fisiológicos e bioquímicos causados pelo alumínio em duas linhagens de Saccharomyces cerevisiae: levedura de panificação Fleischmann e a linhagem PE-2. Para tal, procurou-se simular, tanto quanto possível, as condições fisiológicas da fermentação industrial. Foram realizados 4 experimentos, empregando-se mosto semi-sintético e caldo de cana, contendo 200 g de açúcares redutores totais (ART) por litro. O alumínio foi adicionado na forma de AlCl3.6H2O nas seguintes proporções: 0 (testemunha), 50 e 100 mg/L, nos experimentos de 1 a 3, e 0 e 50 mg/L, no experimento 4, variando-se o pH dos mostos de 4,0 a 5,0. Os experimentos foram conduzidos com reciclo de células, sendo avaliados os seguintes parâmetros: rendimento em etanol, formação de glicerol e açúcares residuais, crescimento em biomassa, viabilidade celular, contaminação bacteriana, teores iniciais e finais dos carboidratos de reserva (trealose e glicogênio) e acúmulo de alumínio nas células de levedura. Concluiu-se que níveis tóxicos de alumínio podem estar presentes em mostos industriais, pois os efeitos tóxicos foram constatados em ambas linhagens, porém, a linhagem PE-2 mostrou-se mais resistente a tais efeitos quando comparada com a levedura de panificação Fleischmann. / The aim of this work was to study physiological and biochemical effects caused by aluminum (Al) in two strains of Saccharomyces cerevisiae: baker's yeast Fleischmann and strain PE-2. For such, was tried to simulate, so much as possible, the physiological conditions of the industrial process. Four experiments were performed: using semi-synthetic and cane juice containing 200g of total reducing sugar per liter at pH 4.0 and 5.0. Aluminum was added in the form of AlCl3.6H2O in the following proportions: 0 (control), 50 mg/L and 100 mg/L . The experiments were performed with cell reuse and the following parameters were analysed: ethanol production, glicerol production and residual sugars, growth of the yeast, yeast viability, bacterial contamination, trehalose and glycogen content and accumulation of aluminum in the yeast cells. It was concluded that toxic levels of aluminum could be present in industrial substrates, since toxic effects were verified for both strains. The strain PE-2 showed to be more tolerant to aluminum when compared to baker's yeast.

A fumonisina B1 (FB1) é um metabólito secundário produzido principalmente por Fusarium verticilioides em diversos tipos de alimentos, principalmente o milho, o qual constitui a base para composição de rações para várias espécies de animais domésticos. A FB1 é particularmente tóxica para suínos, cujas manifestações clínicas são evidentes em animais expostos a altas concentrações de FB1 na ração (em geral, acima de 30 mg/kg). No entanto, são escassos os estudos sobre os efeitos da FB1 em suínos alimentados com rações contendo baixas concentrações de fumonisinas, as quais são mais prováveis de serem encontradas em condições de campo. O objetivo do estudo foi avaliar os efeitos da exposição de suínos a baixos níveis de FB1 na ração, durante 28 dias, sobre o ganho de peso, consumo de ração, peso relativo de órgãos e aspectos histológicos do baço, fígado, pulmões, rins, coração e esôfago, a resposta imune e a determinação do resíduo de FB1 em materiais biológicos como plasma, urina e fezes. Vinte e quatro suínos foram distribuídos em 4 grupos experimentais e alimentados com rações contendo 0 mg (controle), 3,0 mg, 6,0 mg ou 9,0 mg FB1/kg de ração. As diferentes dietas não afetaram (P>0,05) o ganho de peso e nem o peso relativo dos órgãos analisados. Não foram constatadas lesões macroscópicas ou histopatológicas no esôfago, rins, coração e fígado. No entanto, foram observadas lesões histopatológicas nos pulmões de todos os suínos alimentados com rações contaminadas com fumonisinas, indicando que nenhum dos níveis de FB1 usados no experimento poderia ser considerado como seguro para suínos. Ainda, a FB1 não alterou as concentrações de imunoglobulinas totais, por outro lado, diminuiu a resposta imunológica dos suínos vacinados com Stellamune®, como também reduziu a expressão de citocinas pro-inflamatórias. As concentrações plasmáticas de FB1 residual encontradas permaneceram constantes ao longo dos 28 dias de exposição, nos suínos que se alimentaram com 3,0 mg FB1/kg de ração. Por outro lado, os níveis de FB1 no plasma variaram de 400,3 ± 19,6 pg/mL a 260,6 ± 61,6 pg/mL nos animais expostos a 6,0 mg FB1/kg de ração, e de 1127 ± 494,5 pg/mL a 460,7 ± 69,5 pg/mL nos animais expostos a 9,0 mg FB1/kg de ração, entre os dias 7 e 28 dias de exposição, respectivamente. Em relação aos níveis residuais de FB1 eliminados na urina, embora com variações ao longo dos 28 dias de exposição, foram encontrados níveis que variaram de 28,53 ± 17,9 ng/mL a 16,1 ± 22 ng/ml na urina dos suínos expostos com 3,0 mg FB1/kg de ração, de 47,4 ± 26,4 ng/mL a 24,12 ± 26 ng/mL nos suínos expostos a 6,0 mg FB1/kg de ração e de 75,05 ± 55,2 ng/mL a 18,9 ± 4,4 ng/mL nos suínos expostos a 9,0 mg FB1/kg de ração. Os níveis de FB1 residual eliminados nas fezes permaneceram constantes nos animais expostos com 3,0 mg FB1/kg de ração entre 2,26 ± 1,6 mg/kg e 3,25 ± 2,26 mg/kg, variando de 5,18 ± 1,6 mg/kg a 10,6 ± 2,3 mg/kg, e de 8,16 ± 6,8 mg/kg a 13,8 ± 4,8 mg/kg, nos suínos expostos a 6,0 mg FB1/kg de ração e 9,0 mg FB1/kg de ração, respectivamente. Os resíduos de FB1 encontrados nas fezes decaíram a partir de 21 dias de exposição a FB1 em suínos expostos a 6,0 mg FB1/kg de ração, porém, estes níveis permaneceram constantes entre o dia 21 e 28 em suínos expostos 9,0 mg FB1/kg de ração, indicando uma baixa absorção da toxina em todos os tratamentos. As correlações entre a FB1 eliminada nas fezes (mg/kg) e os níveis de FB1 ingeridos por suínos (mg FB1/animal) foram significativas (P<0,05) em todos os dias. Foram observadas correlações significativas (P<0,05) entre a ingestão de FB1 nos diferentes tratamentos e as concentrações de FB1 na urina e no plasma aos 7 e 28 dias, respectivamente. Deste modo, a quantificação de FB1 na urina e plasma é adequada como biomarcador da ingestão de baixos níveis FB1 no início (7 dias) e no final (28 dias) do período de intoxicação, respectivamente. São necessários novos estudos sobre os mecanismos de ação tóxica da FB1 em suínos, sobretudo em condições de exposição prolongada a níveis baixos de contaminação na ração. / Fumonisin B1 (FB1) is a secondary metabolite produced mainly by Fusarium verticilioides in various types of foods, particularly corn, which is the basis for various feed composition for domestic animals. FB1 is particularly toxic to swine, which clinical manifestations are clearly observed in animals exposed to high concentrations of FB1 in the diet (generally above 30 mg/kg). However, there are few studies on the effects of FB1 in pigs fed diets containing low concentrations of fumonisin, which are more probable to be found in field conditions. The objective of the study was to evaluate the effects of piglets exposed to low levels of FB1 in the feed for 28 days on weight gain, feed intake, relative weight of organs and histological aspects of the spleen, liver, lungs, kidneys, heart and esophagus, immune response and the determination of FB1 residue in biological materials such as plasma, urine and feces. Twenty-four pigs were divided into 4 experimental groups and fed diets containing 0 mg (control), 3.0 mg, 6.0 mg or 9.0 mg FB1/kg diet. The different diets did not affect (P> 0.05) weight gain or the relative weight of the analyzed organs. Macroscopic and microscopic lesions were not observed in the esophagus, liver, kidneys and heart. However, histopathological lesions were observed in the lungs of all pigs fed diets contaminated with fumonisin, indicating that none of FB1 levels used in the experiment could be considered as safe for pigs. Furthermore, the dietary FB1 did not alter the concentrations of total immunoglobulins, although it decreased the immune response of pigs vaccinated with Stellamune® and also reduced the expression of pro-inflammatory cytokines. The FB1 concentrations in plasma of pigs fed 3.0 mg FB1/kg diet remained constant over the 28 days of exposure. Moreover, FB1 plasma levels ranged from 400.3 ± 19.6 pg/ mL to 260.6 ± 61.6 pg/mL in animals exposed to 6.0 mg FB1/kg of diet and 1,127 ± 494.5 pg/mL to 460.7 ± 69.5 pg/mL in animals exposed to 9.0 mg FB1/kg diet, between 7 and 28 days of exposure, respectively. In relation to residual FB1 eliminated in urine, although a variation over the 28 days of exposure was observed, the levels ranged from 17.9 ± 28.53 ng/mL to 16.1 ± 22 ng/mL in urine of pigs exposed with 3.0 mg FB1/kg, 47.4 ± 26.4 ng/mL to 24.12 ± 26 ng/mL in pigs exposed to 6.0 mg FB1/kg, and 75.05 ± 55.2 ng/mL to 18.9 ± 4.4 ng/mL in pigs exposed to 9.0 mg FB1/kg of feed. Residual FB1 levels eliminated in the feces remained constant in animals exposed to 3.0 mg FB1/kg diet, with values of 2.26 ± 1.6 mg/g to 3.25 ± 2.26 mg/kg; in pigs fed 6.0 or 9.0 mg FB1/kg feed, the levels found in feces were 5.18 ± 1.6 mg/kg to 10.6 ± 2.3 mg/kg and 8.16 ± 6.8 mg/kg to 13.8 ± 4.8 mg/kg, respectively. The FB1 residues found in feces decreased from 21 days in pigs exposed to 6.0 mg FB1/kg of feed; however, these levels remained constant between 21 and 28 in pigs exposed to 9.0 mg FB1/kg of feed, indicating a low absorption of toxin in all treatments. Significant (P<0,.05) correlations were found between the FB1 excreted in feces (mg/kg) and the FB1 ingested (mg FB1/animal). The correlations between the ingested FB1 in the different treatments and the residual FB1 levels in urine or plasma were also significant (P<0.05) on days 7 and 28 of intoxication, respectively. Thus the quantification of FB1 in urine or plasm is suitable as biomarker of ingestion of low levels of FB1 at the beginning (7 days) or at the end (28 days) of the intoxication period, respectively. Further studies are needed on the mechanisms of toxic action of FB1 in pigs especially under conditions of prolonged exposure to low levels of contamination in the feed.

Evaluating the toxic effects of industrial waste from a historic landfarming site using bioassays

Thesis (MSc)--University of Stellenbosch, 2011. / ENGLISH ABSTRACT: Landfarming is a widely used method for the disposal of contaminants in the petrochemical industry. It involves ploughing the contaminants into the top soil layer allowing biological breakdown. A historically landfarmed site was identified at a South African petrol refinery. The refinery used to dispose so-called American Petroleum Institute (API) -sludge onto a landfarming site. API-sludge consisted of a mixture of oil and water soluble contaminants originating from a process of separating refinery waste from reusable water and oil. Landfarming on this site was discontinued after excessive quantities of sludge were ploughed into the soil over time and it became obvious that effective biodegradation could not take place. An environmental assessment had to be carried out to assess to what extent the soil has recovered from the contamination and after remediation was done over time. Bioassays together with chemical analyses were executed to determine the level of pollutants in the soil and to assess the integrated effects of their bioavailable fractions. The landfarming site of the refinery was divided into two sections namely, a more contaminated north-site and less contaminated south-site. Soil samples were collected from both sites as well as from an off-site (control site). The soils were analysed physically, chemically and used in the bioassays. Two additional control soils were also used, OECD-soil and LUFA2.2 soil. Chemical analysis of the site soils showed the presence of heavy metals and high levels of diesel range organic hydrocarbons. The north-site had higher levels of contaminants compared to the south-site. Three species of soil organisms were used in standardised tests: Eisenia andrei, Enchytraeus doerjesi and Folsomia candida were exposed to the respective soils to study their survival, growth, reproduction success and avoidance behavior. Exposures to both site-soils were not acutely toxic to any organisms. F. candida had a decrease in juvenile production in both north- and south-site soils (289.42 ± 58.62 and 253.33 ± 122.94 respectively) compared to the control soil (479.89 ± 30.42). E. doerjesi showed an increase in produced juveniles exposed to north- and south-site soil (339.75 ± 76.92 and 414.00 ± 17.78 ) compared to control soil (57 ± 34.39). E. andrei had similar cocoon production when exposed to south-site soil than in off-site soil (19.00 ± 5.3 and 18.5 ± 9.7 respectively) but significantly less in north-site soil (1.25 ± 0.7). Only E. doerjesi showed avoidance of north-site soil. To determine the sensitivity of the organisms to the API-sludge, they were exposed to concentration series of API-sludge-spiked control soils. The effect concentrations were calculated as the concentration of API-sludge that will decrease the studied endpoints by 50% of the control soil (EC50). The EC50s varied for each species exposed in the different control soils showing that the toxicity of the API-sludge is to a certain extent dependent on the physical soil properties of the substrate. The reproduction of F. candida were most sensitive to the API-sludge in off-site soil (EC50 = 90 mg/kg) and the E. doerjesi the least sensitive in LUFA2.2 soil (EC50 = 36000 mg/kg). Five plant species were exposed to API-sludge-spiked potting soil and the germination success, early growth rate and biomass were studied. The plants were not as sensitive to API-sludge as the soil animals. Lettuce and grass were affected the most by API-sludge and beans were the most resilient species. With the addition of low levels API-sludge to the substrate, the growth rate of beans was stimulated. This study showed that the south-site has been successfully remediated and most soil organisms exposed to these soils were not affected by the levels of toxicants present. However, exposures to north-site soil still had negative effects on soil organisms. It is recommended that hydrocarbon contamination should be further remediated in the north-site soil before landfarming should be allowed to continue. / AFRIKAANSE OPSOMMING: Ploegverwerking is ‘n algemene remediëringsmetode vir die verwerking van afvalmateriaal in petrochemiese industrieë. Dit behels die inploeg van toksiese afvalmateriaal in die boonstegrondlaag sodat dit biologies afgebreek kan word. ‘n Voorbeeld van ‘n histories ploegverwerkte grondstuk is geidentifiseer by ‘n Suid-Afrikaanse olieraffinadery. Die raffinadery het in die verlede van die grondstuk gebruik gemaak om sogenaamde Amerikaanse Petroleum Instituut-slik (API-slik) daarin te ploeg. Die API-slik bestaan uit ‘n mengsel van olie- en wateroplosbare kontaminante afkomstig van die proses waardeur die raffinadery se afvalprodukte van hernubare water en olie geskei word. Nadat oormatige konsentrasies slik in die grond ingewerk is en bioremediasie nie meer doeltreffend kon voortgaan nie, is die ploegverwerking gestaak. ‘n Omgewingimpakstudie moes uitgevoer word om te bepaal tot watter mate die grond herstel het nadat remediasie oor tyd uitgevoer is. Toksisiteitstoetse en chemiese analises is uitgevoer om die vlakke van besoedeling sowel as die biobeskikbare fraksie daarvan in die grond te bepaal. Die ploegverwerkte area van die raffinadery is in twee verdeel naamlik, ‘n meer gekontamineerde noordelike area en ‘n minder gekontamineerde suidelike area. Grondmonsters is van die onderskeie areas asook van ‘n ongekontamineerde veld (as kontrole) naby die ploegverwerkte area versamel Die gronde is fisies- en chemies geanaliseer en toksisiteitstoetse is uitgevoer. Twee addisionele kontolegronde is ook tydens die blootstellings gebruik naamlik, OECD- en LUFA2.2-grond. Die chemiese analises van die ploegverwerkte toetsgronde het getoon dat daar steeds swaarmetale en hoë vlakke van dieselgekoppelde organiese koolwaterstowwe in die gronde teenwoordig is. Kontaminante was in hoër konsentrasies teenwoordig in die grond van die noordelike gebied as in dié van die suidelike gebied. Drie spesies van grondorganismes is gebruik tydens standaard toksisitetitstoetse. Eisenia andrei, Enchytraeus doerjesi en Folsomia candida is blootgestel aan die onderskeie toets- en kontrolegronde waarna hul oorlewing, groei, voortplantingsukses en vermydingsreaksies bestudeer is. Blootstellings aan die ploegverwerkte toetsgronde het geen akute toksisiteit vir enige van die spesies getoon nie. F. candida se juveniele produksie was laer in beide noordelike- en suidelike toetsgronde (289.42 ± 58.62 en 253.33 ± 122.94 onderskeidelik) as in die kontolegrond (479.89 ± 30.42). E. doerjesi blootstellings het ‘n toename in juveniele getalle getoon in die noordelike- en suidelike toetsgronde (339.75 ± 76.92 en 414.00 ± 17.78) in vergelyking met die ongekontamineerde kontolegrond (57 ± 34.39). Kokonproduksie by E. andrei was soorgelyk in die suidelike toetsgrond en ongekontamineerde kontrolegronde (19.00 ± 5.3 en 18.5 ± 9.7 onderskeidelik) maar beduidend minder as in noordelike toetsgrond (1.25 ± 0.7). Slegs E. doerjesi het ‘n beduidende vermydingsreaksie vir die noordelike toetsgronde getoon. Om die sensitiwiteit van die organismes aan vars API-slik te bestudeer, is hulle blootgestel aan konsentrasiereekse van API-slik in die onderskeie kontrolegronde. Die effektiewe konsentrasie (EK50) is bereken as die konsentrasie van API-slik wat die bestudeerde eindpunte met 50% sal verminder in vergelyking met die kontrolegrond Die EK50-waardes vir al die spesies het verskil na blootstelling aan die onderskeie kontrolegronde. Dus, die toksisiteit van die API-slik is tot ‘n sekere mate ook afhanklik van die fisiese grondeienskappe van die blootsellingssubstraat. Die voortplanting van F. candida was die gevoeligste eindpunt vir die blootstelling aan API-slik in kontolegrond (EK50 = 90 mg/kg) en E. doerjesi was die minste gevoelig in LUFA2.2 grond (EK50 = 36000 mg/kg). Vyf plantspesies is ook blootgestel aan API-slikgekontamineerde potgrond en die saadontkiemingssukses, vroeë groeikoers en biomassa is bestudeer. Alhoewel plante nie so sensitief was vir die API-slik soos die gronddiere nie, was blaarslaai en gras die meeste geaffekteer tydens die blootstellings. Boontjies was die ongevoeligste en met die toevoeging van lae konsentrasies API-slik (2.5% API-slik), is hul groeikoers selfs gestimuleer. Uit die studie was dit duidelik dat die suidelike deel van die grondstuk meer suksesvol as die noordelike geremidieer is en dat meeste grondorganismes wat daaraan bloot gestel is nie geaffekteer is deur die vlakke van kontaminasie wat steeds teenwoordig is in die grond nie. Die toetsgronde uit die noordelike deel het egter steeds negatiewe effekte op die grondorganismes gehad. Dit word voorgestel dat die koolwaterstof kontaminasie verder geremidieër behoort te word in die noordelike deel van die grondstuk voordat verdere ploegverwerking van die afval daar gedoen word.

This medicine has a narrow therapeutic range and is usually used in a chronic way by the patients, generating the need for therapeutic monitoring by means of plasma lithium quantification. In addition, records of toxic effects from chronic medical use reinforce the need for a tool that assists in the treatment. Thus, a reliable methodology for the quantification of lithium and a therapeutic adjuvant emerge as a means to improve the quality of life of the patient. In our work, we used the flame atomic absorption spectrometer to validate the method for quantification of lithium and the results were satisfactory, because they complied with the required criteria, assuring the reliability of the method. In order to find a therapeutic adjuvant, N-acetylcysteine (NAC) was studied for presenting important characteristics against the toxic effects caused by lithium. In our results lithium presented toxic effects mainly inducing oxidative stress, while the coadministration of NAC reversed this effect in the liver, kidney and brain organs. The use of NAC as a therapeutic adjuvant has been shown to be promising, however, further studies need to be performed to better understand this relationship. / O carbonato de lítio é o principal medicamento utilizado no tratamento do transtorno bipolar. Este medicamento possuí estreita faixa terapêutica e geralmente é utilizado de forma crônica pelos pacientes, gerando a necessidade da monitorização terapêutica por meio da quantificação plasmática de lítio. Além disso, os registros de efeitos tóxicos decorrentes do uso medicinal crônico remetem a necessidade de uma ferramenta que auxilie no tratamento. Desta forma, uma metodologia confiável para a quantificação de lítio e um adjuvante terapêutico surgem como meios de melhorar a qualidade de vida do paciente. Em nosso trabalho, utilizamos o espectrômetro de absorção atômica de chama para validar um método para quantificação de lítio e os resultados foram satisfatórios, pois cumpriram os critérios exigidos, assegurando a confiabilidade do método. Com foco em buscar um adjuvante terapêutico, a N-acetilcisteína (NAC) foi estudada por apresentar características importantes contra os efeitos tóxicos causados pelo lítio. Em nossos resultados o lítio apresentou efeitos tóxicos principalmente induzindo o estresse oxidativo, enquanto que a coadministração de NAC reverteu esse efeito nos órgãos fígado, rim e cérebro. O uso da NAC como adjuvante terapêutico mostrou ser promissor, porém, mais estudos precisam ser realizados para entender melhor essa relação.

Os modelos mistos são apropriados na análise de dados longitudinais, agrupados e hierárquicos, permitindo descrever e comparar os perfis médios de respostas, levando em conta a variabilidade e a correlação entre as unidades experimentais de um mesmo grupo e entre os valores observados na mesma unidade experimental ao longo do tempo, assim como a heterogeneidade das variâncias. Esses modelos possibilitam a análise de dados desbalanceados, incompletos ou irregulares com relação ao tempo. Neste trabalho, buscou-se mostrar a flexibilidade dos modelos lineares mistos e a sua importância na análise de dados toxicológicos longitudinais. Os modelos lineares mistos foram utilizados para analisar os efeitos de dose no ganho de peso de ratos adultos machos e fêmeas, em teste de toxicidade por doses repetidas e também os efeitos de fase de gestação e dose nos perfis de pesos de filhotes de ratas tratadas. Foram comparados os modelos lineares mistos de regressão polinomial de grau 3, spline e de regressão por partes, ambos com um único ponto de mudança na idade média de abertura dos olhos dos filhotes, buscando o mais apropriado para descrever o crescimento dos mesmos ao longo do período de amamentação. São apresentados os códigos escritos no SAS/STAT para a análise exploratória dos dados, ajuste, comparação e validação dos modelos. Espera-se que o detalhamento da teoria e das aplicações apresentado contribua para a compreensão, interesse e uso desta metodologia por estatísticos e pesquisadores da área. / Mixed models are appropriate in the analysis of longitudinal, grouped and hierarchical data, allowing describe and compare the average response profiles, taking into account the variability and correlation among the experimental units of the same group and among the values observed over the time in the same experimental unit, as well as the heterogeneity of variances. These models allow the analysis of unbalanced, incomplete or irregular data with respect to time. This work aimed to show the flexibility of linear mixed models and its importance in the analysis of longitudinal toxicological data. Linear mixed models were used to evaluate the effects of doses in the body weight gain of adult male and female Wistar rats, in repeated doses toxicity test and also the effects of pregnancy period and dose in the pups growth of treated dams. It were compared the linear mixed models of third degree polynomial regression, spline and piecewise regression, both with a single point of change in the average time of pups eyes opening, searching for the most appropriate one to describe their growth along the lactation period. The SAS/STAT codes used for exploratory data analysis, comparison and validation of fitted models are presented. It is expected that the detailing of the theory and of the applications presented contribute with the understanding, interest and use of this methodology by statisticians and researchers in the area.

Os modelos mistos são apropriados na análise de dados longitudinais, agrupados e hierárquicos, permitindo descrever e comparar os perfis médios de respostas, levando em conta a variabilidade e a correlação entre as unidades experimentais de um mesmo grupo e entre os valores observados na mesma unidade experimental ao longo do tempo, assim como a heterogeneidade das variâncias. Esses modelos possibilitam a análise de dados desbalanceados, incompletos ou irregulares com relação ao tempo. Evaluation de l'exposition et des effets des éléments traces métalliques et du parasitisme chez la faune sauvage : contribution au développement d'une approche non létale / Assessment of exposure and effect of trace metals and parasitism on wildlife : contribution to the development of a non-lethal approach

En intégrant cette étude dans un contexte d'écologie du stress, l'objectif général de cette thèse est d'une part d'évaluer l'exposition et les effets des éléments traces métalliques (ETMs)et du parasitisme chez le mulot sylvestre (apodemus sylvaticus) et d'autre part de contribuer au développement de marqueurs d'exposition et d'effets toxiques non létaux. Au cours de trois sessions de capture, des individus ont été collectés dur 30 sites localisés autour de l'ancienne fonderie de Metaleurop (Nord-Pas-de-Calais) les concentrations en Cd et Pb ont été mesurées dans les organes cibles (foie et reins) et dans les poils des mulots. différents biomarqueurs d'effets toxiques létaux et non létaux ont également été mesurés. Le nombre important de mulots prélevé (n=886) a permis d'étudier l'influence des caractéristiques individuelles (âge et sexe) et des caractéristique paysagères sur les réponses des biomarqueurs. Les résultats indiquent que les concentrations en ETMs des animaux vivants sur les sites les plus contaminés s'avèrent significativement plus importantes que celles mesurées sur les autres sites. En outre, plus de 25% des mulots provenant des sites des plis pollués présente des concentrations supérieures aux seuils toxiques et ont donc un risque accru de développer des pathologies (œdème, cancers). Par ailleurs, cette étude révèle également que ETMs chez le mulot est influencée par les caractéristiques, individuelles (âge et sexe), par l'occupation des sols et qu'elle varie en fonction de la session de capture. De plus, les résultats montrent que les concentrations ETMs [...] et les caractéristiques paysagères modulent les prévalences de certains parasites. En effet, la richesse parasitaire augmente chez les individus les plus exposés aux ETMs. Ces concentrations influencent également les différents biomarqueurs d'effets létaux [...] et non létaux [...]évalués. Ces résultats illustrent donc l'intérêt du développement de biomarqueurs on létaux pour l'évaluation des effets toxiques des ETMs les plus élevées présentent des atteintes au niveau individuel [...] et au niveau cellulaire [...] Cependant, étant donnés les effets potentiels de parasitisme sur la santé de la faune sauvage, les liens de causalité entre la présence d' ETMs et les atteintes observées sont discutables. Ce travail de thèse souligne l'intérêt des approches multi-stress dans le cadre de l'évaluation de la santé de la faune sauvage. / By integrating this study in a stess-ecology Framework, the aim of this thesis is to assess exposure and effects of trace metals (TMs) and parasitism on wood mouse[…] and to contribute to the development of non-lethal exposure and toxicity makers […].This study also reveals that the accumulations of the TMs in wood mice is influenced by individual characteristics […] and landscape features and varies according to the trapping session. In addition, results show that the concentrations of TMs […] and landscape characteristics modulate the prevalence of some parasites. […].However, given the potential effects of parasitism on animals’ health, the causal links between the presence of TMs and observed alterations are questionable. This work emphasizes the importance of multi-stress approaches on wildlife’s health assessment.
