It started mainly as a musical phenomenon, the punk rock, anti-conformist protest adopted by part of the young American intellectuals and then by the British. In the beggining it was against the commercialism that rock music had submitted to, afterwards it expanded to a social, cultural and artistic movement of anti-establishment youthful challenge. It grew and radicalized within the working class, mainly in Britain, afterwards it spread across all over the world seen by authorities as a subversive culture movement. In Brazil it started in many metropolitan regions of São Paulo in which the most expressivity was observed in the center region emphasized by the media factor, and in the industrial part of ABC region where it obtained more expressive and radical ways where the movement was involved with the working and union fights accurred in the late 1970s and beggining of 1980s / O Movimento Punk surge em meados dos anos 70 como um dos últimos movimentos contraculturais. Se deu primeiramente como um fenômeno musical, o punk rock, música de protesto anti-conformista adotado por uma parcela de jovens intelectuais norte-americanos e depois ingleses. A princípio foi contra a comercialização à qual o rock havia se submetido, posteriormente se expandiu como um movimento social, cultural e artístico, de contestação juvenil anti-establishment. Cresceu e se radicalizou junto à classe operária, principalmente a inglesa, depois se proliferou para o mundo todo, visto pelas autoridades como um movimento de cultura subversiva . No Brasil o movimento radicou-se nas diversas regiões metropolitanas de São Paulo, tendo maior expressividade no centro da capital enfatizado pelo fator midiático e na região industrial do ABC, onde obteve formas mais expressivas e radicais, estando envolvido com as lutas operárias e sindicais ocorridas no final da década de 70 e início dos anos de 1980. Optimering av lageraktiviteter och transporter : Flügger AB / Optimization of warehouse activities and transports : Flügger AB

Detta examensarbete har utförts under höstterminen 2008 på Högskolan i Borås på uppdrag av Flügger AB i Bollebygd. År 1994 köpte Flügger A/S upp HP Färg & Kemi och kedjan Flügger AB startades i Sverige. Idag har Flügger i Bollebygd 150 anställda, vilket gör dem till en av Bollebygds största arbetsgivare. Dagens fabrik i Bollebygd är uppdelad i fyra producerande enheter; Alkyd-, Latex-, Kemi- och Spackelfabrik. Fabrikerna rymmer produktion av vattenbaserad- och terpentinbaserad färg, träskydd, spackelprodukter, rengöringsmedel för industri och biltvättsanläggningar, samt målningsrelaterade rengöringsmedel. I Bollebygd finns även ett råvaru- och emballagelager som ansvarar för materialtillförsel till de fyra produktionsenheterna. Syftet med detta arbete är att hitta förbättringar och effektiviseringsförslag för nuvarande interna transporter från råvaru- och emballagelager. Målsättningen är även att hitta lösningar för mellanlagringsplatser, samt förslag till utformning av råvaru- och emballagelager. En ytterligare uppgift är att hitta ett tillvägagångssätt som förbättrar beställningsrutiner och returhantering. Behovssvängningar skall jämnas ut genom att sprida ut de interna beställningarna till råvaru- och emballagelagret. I och med att företaget är certifierat enligt ISO 14 001 skall ovanstående punkter utföras med hänsynstagande till miljön. Genom intervjuer med de anställda på Flügger har information erhållits om de problem som finns inom internlogistik och lagerhantering. Intervjuer har även gjorts för att ta vara på de eventuella förbättringsförslag de anställda besitter. För att få en sådan korrekt helhetsbild som möjligt har både lagerpersonal och fabrikspersonal intervjuats. Observationer har gjorts på lagret såväl som på produktionsenheterna. På lagret har även tidtagning av lageraktiviteter utförts. En klar bild av Flüggers interna logistik och lageraktiviteter har erhållits under arbetets gång. Utifrån den bilden dras slutsatsen att aktiviteterna kan effektiviseras och att Flügger med relativt små medel kan förändra bilden till det bättre. Ett antal förbättringsförslag har framtagits inom de olika problemområdena. Dessa innehåller bland annat en presentation av lagerutformning, beställningsrutin samt effektivisering av lageraktiviteter.

Expression, purification and characterisation of the Cystic Fibrosis Transmembrane conductance Regulator (CFTR) in Saccharomyces cerevisiae

Mutations in the eukaryotic integral membrane protein Cystic Fibrosis Transmembrane conductance Regulator (CFTR) cause the hereditary disease cystic fibrosis (CF). CFTR functions as an ion channel at the surface of epithelial cells and regulates the movement of chloride ions and water across the plasma membrane. CFTR is difficult to express and purify in heterologous systems due to its propensity to form insoluble aggregates and its susceptibility to degradation. Obtaining good yields of highly purified CFTR has proven problematic and contributes to our limited understanding of the structure and function of the protein. The most prevalent disease causing mutation, F508del, results in misfolded CFTR which is particularly unstable and is quickly targeted for degradation by the host system and is prevented from being trafficked to the plasma membrane. There are limited treatment options for patients with the F508del mutation and it is therefore of significant interest within CF research. New methods and assays are required to identify potential compounds which could correct the F508del mutation. This thesis investigates the use of Saccharomyces cerevisiae to express and purify codon optimised recombinant CFTR. The use of a green fluorescent protein (GFP) tag enabled quick and simple detection of CFTR in whole cells and after extraction from the plasma membrane. By optimising the culture conditions for CFTR expression and detergent solubilisation conditions, relatively high yields of full-length protein were obtained. When used as a chemical chaperone at the time of inducing CFTR expression, glycerol increased yields of full-length protein. Degradation of CFTR could be limited by inducing expression at an optimal cell density and by harvesting cells within a specific time window. CFTR was extracted by solubilisation in the mild detergent dodecyl-β-D-maltopyranoside (DDM) in the presence of up to 1 M NaCl with up to ~87% efficiency in some cases. Using a gene optimisation strategy in which additional purification tags and a yeast Kozak-like sequence were added, the human CFTR (hCFTR) protein was expressed and purified. Fluorescence microscopy revealed CFTR localisation at the periphery of yeast cells. Immunoaffinity chromatography facilitated by the GFP tag at the C terminus of CFTR produced protein of up to 95% purity. An assessment of the thermal stability of this highly purified CFTR using a fluorescent probe binding assay revealed a denaturation midpoint (Tm) of ~43 degC. The ability of this assay to determine the stability of CFTR is encouraging and there is the potential to further develop it in a high-throughput manner to identify compounds which stabilise the F508del protein and which may hold the key to developing new treatments for CF.

Syfte och frågeställningar Syftet med studien är att undersöka statisk och dynamisk balansförmåga för individer med varierad grad av funktionsnedsättning genom uppmätt balansförmåga, upplevd balansförmåga samt eventuella korrelationer mellan balansförmåga och upplevd balansförmåga. Frågeställningar som studien avser att besvara: • Vilket värde får deltagarnas statiska och dynamiska balansförmåga utifrån två av testen i Movement ABC? •Hur skattar deltagarna sin individuella upplevda statiska och dynamiska balansförmåga? • Hur förhåller sig eventuella korrelationer mellan deltagarnas utförda förmåga och skattningen av utförandet? Metod Metoden baseras på två balanstester från testbatteriet Movement ABC. De två balanstesterna, både statiskt och dynamiskt, kompletterades med ett individuellt subjektivt skattningsformulär angående upplevd balansförmåga. Resultaten analyserades med Spearman´s korrelationsanalys. Resultat De beskrivande resultaten angående statisk balans uppmätte 5/8 deltagare som Mycket bra/OK respektive dynamisk balans uppmätte 3/8 deltagare som Mycket bra/OK. Resultaten från det individuella självskattningsformuläret skattade 7/8 deltagare Mycket bra/OK. Inga signifikanta resultat framkom i korrelationsanalysen efter att empirin analyserats, då urvalsgruppen inte var tillräckligt i antal för att fastställa någon statistisk skillnad. Slutsats Deltagarna i studien uppvisade en högre prestation i det statiska kontra det dynamiska testet. Majoriteten av deltagarna skattade sin balansförmåga som Mycket bra och OK oavsett resultat på utförd balanstest. Då studien bestod av en liten urvalsgrupp visade analysen inte på några signifikanta resultat dras inga större slutsatser. / Aim The purpose of this study is to examine the static and dynamic balance ability of individuals with various kinds of impairments by looking into their state of balance, their experienced state of balance, and determine potential correlations between their state of balance and experienced state of balance. This study aims to answer the following questions: • Based on the two tests of the Movement ABC, what will the participants’ static and dynamic balance ability scores be? • How will the participants estimate their experienced static and dynamic state of balance? • Is there a potential correlation between the participants’ test scores and their estimations? Method The method is based on the two balance tests from the Movement Assessments Battery for Children. These two tests, both static and dynamic, are complemented with an individual subjective estimation form regarding experienced state of balance. The results are analyzed through Spearman’s correlation analysis. Results The descriptive results regarding static balance measured 5/8 participants as Very good / OK and dynamic balance measured 3/8 participants as Very good / OK. Results from the individual self-assessment form estimated 7/8 participants Very good / OK. No significant results were found in the correlation analysis after the empirical analysis, since the selection group was not sufficient in number to determine any statistical difference. Conclusion The participants showed a higher performance in the static versus dynamic test. The majority of participants estimated their balance ability as Very good and OK regardless of the results on the performed balance test. Since the study consisted of a minor selection group, the analysis did not show any significant results, no major conclusions were drawn.

The bilingual mind...simultaneous and sequential processing and spelling ability in monolingual English and bilingual Afrikaans-English children

This has far-reaching and challenging implications for cognitive models of spelling. Specifically, bilingualism exhibits a pervasive influence on children’s literacy development (Bialystok, 2002). The majority of research on children’s spelling has been conducted internationally with monolingual English children. From international literature, cognitive processing (simultaneous processing and sequential processing) has been identified as an important area for consideration in the spelling acquisition process of English children (Kaufman & Kaufman, 1983b). Simultaneous processing is important for whole word spelling, whilst sequential processing is important for decoding letter sound correspondences. Cross-linguistic research demonstrates a bias towards one or the other spelling strategy may be tied to the depth of a language’s orthography, possibly due to the different demands the language orthography places on how children learn to spell (Frost et al., 1987; Wimmer & Hummer, 1990, 1994; Goswami et al., 1998). The present study examined the relationship between simultaneous and sequential processing and spelling in Grade 3 monolingual English-speaking children and bilingual Afrikaans-English speaking children at one point in time. Thirty bilingual Afrikaans-English children (Afrikaans first language, English second language) and were learning to spell in Afrikaans and in English simultaneously, and thirty monolingual (English first language) learning to spell in English. Simultaneous and sequential processing subtests of the Kaufman Assessment Battery (K-ABC) were administered to the monolingual and to the bilingual children. Monolingual Englishspeaking children received the English word and non-word spelling tests, while the bilingual Afrikaans-English children were asked to spell English and Afrikaans words and non-words (Klein, 1993). The results suggest that lexical (logographic or simultaneous) and non-lexical (alphabetic or sequential) routes are available in English and Afrikaans, but orthography did exert an influence on cognitive processing strategies. Sequential processing demonstrates a higher relationship than simultaneous processing with spelling in English and Afrikaans, although sequential processing contributes more to spelling in a shallow orthography, because the reliable relationship between spelling supports easier and faster computation than in an opaque orthography. Additionally, the results demonstrate that in the bilingual Afrikaans-English children spelling in a second language (L2) rely on spelling skills in a first language (L1), even when the same teaching strategies are used for spelling instruction. Orthography as a tool of academic literacy instruction, influences whether the transfer of spelling skills has a positive or negative influence on spelling in English as a second language in bilingual Afrikaans-English children with a transparent L1. A dual-route model that incorporates the influence of orthographic depth is supported (Seymour, Bunce & Evans, 1992). The present research study concludes that (1) simultaneous processing and sequential processing influence and predict the production of spelling in L1 and L2 in both English and Afrikaans alphabetic orthographies that differ in orthographic transparency, (2) orthographic demands of learning to spell in different orthographies varies and influences cognitive processing resources and decoding skills, which may provide an indication of a cumulative or challenging development of L2 spelling skills particularly when the L1 is transparent. The present research has implications for assessment, traditional spelling models and teaching bilingual children learning to spell in a second language, which is orthographically opaque relative to their transparent mother tongue.

Phosphate Signaling Through Alternate Conformations of the PstSCAB Phosphate Transporter

Phosphate is an essential compound for life. Escherichia coli employs a signal transduction pathway that controls the expression of genes that are required for the high-affinity acquisition of phosphate and the utilization of alternate sources of phosphorous. These genes are only expressed when environmental phosphate is limiting. The seven genes for this signaling pathway encode the two-component regulatory proteins PhoB and PhoR, as well as the high-affinity phosphate transporter PstSCAB and an auxiliary protein called PhoU. As the sensor kinase PhoR has no periplasmic sensory domain, the mechanism by which these cells sense environmental phosphate is not known. This paper explores the hypothesis that it is the alternating conformations of the PstSCAB transporter which are formed as part of the normal phosphate transport cycle that signal phosphate sufficiency or phosphate limitation. We tested two variants of PstB that are predicted to lock the protein in either of two conformations for their signaling output. We observed that the pstBQ160K mutant, predicted to reside in an inward facing, open conformation signaled phosphate sufficiency whereas the pstBE179Q mutant, predicted to reside in an outward facing, closed conformation signaled phosphate starvation. Neither mutant showed phosphate transport.

A novel sequential ABC algorithm with applications to the opioid crisis using compartmental models

The abuse of and dependence on opioids are major public health problems, and have been the focus of intense media coverage and scholarly inquiry. This research explores the problem in Iowa through the lens of infectious disease modeling. We wanted to identify the current state of the crisis, factors affecting the progression of the addiction process, and evaluate interventions as data becomes available. We introduced a novel sequential Approximate Bayesian Computation technique to address shortcomings of existing methods in this complex problem space, after surveying the literature for available Bayesian computation techniques. A spatial compartmental model was used which allowed forward and backward progression through susceptible, exposed, addicted, and removed disease states. Data for this model were compiled over the years 2006-2016 for Iowa counties, from a variety of sources. Prescription overdose deaths and treatment data were obtained from the Iowa Department of Public Health, possession and distribution arrest data were acquired from the Iowa Department of Public Safety, a measure of total available pain reliever prescriptions was derived from private health insurance claims data, and population totals were obtained from the US Census Bureau. Inference was conducted in a Bayesian framework. A measure called the empirically adjusted reproductive number which estimates the expected number of new users generated from a single user was used to examine the growth of the crisis. Results expose the trend in recruitment of new users, and peak recruitment times. While we identify an overall decrease in the rate of spread during the study period, the scope of the problem remains severe, and interesting outlying trends require further investigation. In addition, an examination of the reproductive numbers estimated for contact within and between counties indicates that medical exposure, rather than spread through social networks, may be the key driver of this crisis.
