Role mitochondrií a retrográdní signalizace při vývoji kvasinkových kolonií / Role mitochondria and retrograde signalization during development of yeast colony

Unicellular organisms such as yeast have been traditionally studied in shaken cultures, i.e., under condition in which they do not grow attached to solid surfaces as under natural conditions. In nature, cells only rarely live alone, but, on the other hand often create multicellular colonies or biofilms. During last years, yeasts started to be investigated also when grown on solid media. Our laboratory has previously developed special techniques for investigation of yeast colonies. These techniques allowed us to describe individual cell subpopulations within the colonies. The aim of this work was to prepare a series of mutant strains, describe morphology and ultrastructure of their colonies with the aim to contribute to understanding ofthe role of mitochondrial retrograde signalling pathway in the development of yeast colonies. This work describes expression of few selected genes (CIT2, RTG1, RTG2, and RTG3) in colonies of the parental strain BY4742 and of other mutant strains with deletion of one or more genes of RTG regulatory pathways. Playing with the subject : writing in The Pillow Book and in In the Penal Colony

This study explores the nature of writing and the sorts of presence that writing gives us access to. This understanding of writing includes not only all speaking and all writing in the narrow sense of marks on a page, but goes beyond this to include the sense in which Derrida uses the term ‘writing’ in Of Grammatology, to mean a broad and complex process of the construction of textual traces or presences necessarily brought about through the structural mechanism of difference inherent in the writing process (Derrida, 1997). This study argues that writing is a system that creates Subjects or selves as the writing happens. It suggests that writing is a remarkable site from which to explore the construction of selves, because it gives us access to (partially) identifiable presences, in the apparent absence of the writer. It goes on to demonstrate that this identity can be distinguished through written traces of difference left for the reader to decipher, by analysing different aspects of the plot and writing devices in Peter Greenaway’s film The Pillow Book and in Kafka’s short story In the Penal Colony. These two texts are considered particularly relevant to this study, in that they both explicitly deal with the contradictory nature of writing and how it relates to the Being (there or the contextualised Being of Dasein) and being (in general), the life and death, the empowerment and destruction of the Subjects that writing sets up. Both texts explore salient aspects of writing on the human body. The study uses these texts as a platform for speculation about the kind of presence that can be traced through writing, and proposes that the written Subject is multiple, contradictory and reflexive, connected and related, and that it is impermanent and has a deferred presence. Finally, this written Subject is also explored in the context of Foucault’s expositions of the self in texts such as Technologies of the self (Foucault, 1994) and ‘What is an Author?’ (Foucault, 1977) in answer to his question Who are we in the present, what is this fragile moment from which we can’t detach our identity and which will carry our identity away with itself? Expression, purification, and antimicrobial activity of avian beta-defensin-2, -6, and -12

Total RNA was extracted from chicken oviduct epithelial cells. Avian Beta-defensin (AvBD)-2, -6, and -12 cDNAs were amplified by reverse transcription-PCR and cloned into pRSET A style='msoareast-language:ZH-CN'>, a protein expression vector. The class=SpellE>hexa-histidine-tagged class=SpellE>AvBD peptides were expressed in Escherichia coli (E. coli) BL21(DE3) class=SpellE>plysS and affinity-purified. The antimicrobial activities of the recombinant AvBDs against E. coli style='msoareast-language:ZH-CN;mso-bidiont-style:italic'>, Salmonella class=SpellE>enterica style='mso-bookmark:OLE_LINK9'> serovar Typhimurium (S. class=SpellE>typhimurium), and Staphylococcus aureus style='msoareast-language:ZH-CN'> (S. aureus) were determined. style='msoareast-language:ZH-CN;mso-bidiont-style:italic'> At 8, 16 and 32 µg/ml, all three rAvBDs killed and inhibited the growth style='msoareast-language:ZH-CN'> of E. coli style='msoareast-language:ZH-CN;mso-bidiont-style:italic'>, S. typhimurium, and S. aureus. The killing of rAvBD-2, -6, and -12 against stationary phase E. coli and S. class=SpellE>aureus was pH dependent in the range investigated. style='msoareast-language:ZH-CN'> In addition, the killing-curves showed that rAvBDs exerted their antimicrobial function within 30 minutes of treatment, suggesting the fast killing mechanisms of rAvBDs.

[pt] A implementação de agricultura urbana, na forma de Quintais agroecológicos, pode ser considerada uma estratégia de adaptação às mudanças ambientais geradas a partir da urbanização. A prática da agricultura urbana traz benefícios sociais, econômicos e ambientais, já que diversas famílias a utilizam como fonte secundária de renda, de alimentação, como atividade de relaxamento, empoderamento social e cultural, regulação climática e manutenção da biodiversidade. Entretanto, essa atividade sofre com a falta de regulamentação legal e de incentivos, gerando insegurança e enfraquecimento da mesma e dos benefícios acimas citados. Nesse contexto, o fortalecimento da prática faz-se necessário através do reconhecimento e da legalização da mesma, a partir de leis que regulamentarizam a atividade, permitindo a retirada de créditos ou criação de projetos visando a maior produtividade dos quintais e engajando a população. Dessa maneira, o objetivo do trabalho foi avaliar o potencial da implementação da agricultura, na forma de Quintais Agroecológicos, servindo como estratégias de adaptação das cidades às mudanças ambientais e expansão urbana, tendo como área de estudo a colônia Juliano Moreira, Zona Oeste do Município do Rio de Janeiro, escolhida por estar inserida em um contexto de expansão urbana e apresentar a prática de agricultura urbana em sua organização. / [en] Urbanization can be considered as one of the essential processes of land-use change (Grimm et al., 2008). This process intensifies as the population grows globally, and the number of people living as cities increases. The process of urbanization of cities, besides altering the landscape, has an intense impact on the environment, as well as economic, social and environmental challenges, such as climate statistics, pollution, food and energy scarcity (Engel 2011; Cloutier et al.; 2014; IPPC 2014; Lin et al., 2015; FAO, 2016). In this sense, a change in the form of urban growth is necessary, reducing the negative impact of the urbanization process and increasing the resilience of urban systems (Newman; Jennings, 2008; Perks, 2011), which can be reached from themes such as sustainability. or sustainable development, much discussed over the last few years (Hassan; Lee, 2015). Sustainability is linked to a conscious and dynamic urban development process relating social, environmental and economic foundations, creating growth strategies and minimizing the negative impacts of urbanization, the appropriate use of resources, and the creation of resilient and sustainable environments efficient for future generations (Cloutier, et al, 2014). Urban planning and sustainability have been gaining ground in global discussions (Ziegler, 2009). They refer not only to a process of urban network management and the determination of land use functions but also territorial planning for the conservation and maintenance of resources, and the integration, articulation and cooperation among the various actors of society (Shmelev; Shemeleva, 2009; Ronconi, 2011; Stigt et al., 2013; Bento et al., 2018;).

The Mexican Herald, an English language newspaper in Mexico City during the authoritative rule of Porfirio Díaz (1895-1910), sought to introduce a vision of Mexico's development that would influence how Mexicans conceived of their country's political and cultural place within a community that transcended national boundaries. As Mexicans experienced rapid modernization led partially by foreign investors, the Herald represented the imaginings of its editors and their efforts to influence how Mexicans conceptualized their national identity and place in the world. The newspaper's editors idealized a Mexico that would follow the international model of the United States and embrace Pan-Americanism. The Herald's depictions of the ideal, future city provided an intelligible landscape to modernity. The editors' vision of modernity had significant implications for Mexican culture. The newspaper's articles and illustrations defined the parameters of modernity providing clear depictions of the physical, political, and cultural aspects of the community of modern nations.
