Sharpening the Sabre: Canadian Infantry Combat Training during the Second World War

During the Second World War, training was the Canadian Army’s longest sustained activity. Aside from isolated engagements at Hong Kong and Dieppe, the Canadians did not fight in a protracted campaign until the invasion of Sicily in July 1943. The years that Canadian infantry units spent training in the United Kingdom were formative in the history of the Canadian Army. Despite what much of the historical literature has suggested, training succeeded in making the Canadian infantry capable of succeeding in battle against German forces. Canadian infantry training showed a definite progression towards professionalism and away from a pervasive prewar mentality that the infantry was a largely unskilled arm and that training infantrymen did not require special expertise. From 1939 to 1941, Canadian infantry training suffered from problems ranging from equipment shortages to poor senior leadership. In late 1941, the Canadians were introduced to a new method of training called “battle drill,” which broke tactical manoeuvres into simple movements, encouraged initiative among junior leaders, and greatly boosted the men’s morale. The Canadians participated in numerous military exercises of varying sizes that exposed problems with their senior leadership. The replacement of unsuitable officers greatly enhanced the fighting potential of Canadian units and formations. As time went on, infantry training became more rigorous and realistic, and tactical concepts became increasingly sophisticated. By the time of the invasion of Normandy in June 1944, infantry training was intense, suited to units’ assigned tasks, and highly technical, which belied the false prewar assumption that the infantry was an unskilled arm. By the time Canadian divisions entered battle, they were as prepared as they would ever be. The exception to this was the training of the overseas reinforcement units, which reached an acceptable standard only in the last months of the war. This study ultimately represents a substantial contribution to understanding the history of the Canadian Army and its role in the Second World War.

Storied voices in Native American texts : Harry Robinson, Thomas King, James Welch and Leslie Marmon Silko

"Storied Voices in Native American Texts: Harry Robinson, Thomas King, James Welch and Leslie Marmon Silko" approaches Native American literatures from within an interdisciplinary framework that complicates traditional notions o f literary "origins" and canon. It situates the discussion of Native literatures in a Native American context, suggesting that contemporary Native American writing has its roots in Native oral storytelling traditions. Each of these authors draws on specific stories and histories from his or her Native culture. They also draw on European elements and contexts because these are now part o f Native American experience. I suggest that Native oral tradition is already inherently novelistic, and the stories that lie behind contemporary Native American writing explicitly connect past and present as aspects o f current Native reality. Contemporary Native American writers are continuing an on-going and vital storytelling tradition through written forms. A comparison of the texts o f a traditional Native storyteller, Robinson, with the highly literate novels of King, Welch and Silko, shows how orally told stories connect with the process o f writing. Robinson's storytelling suggests how these stories "theorize" the world as he experiences it; the Native American novel continues to theorize Native experience in contemporary times. Native writers use culturally specific stories to express an on-going Native history. Their novels require readers to examine their assumptions about who is telling whose story, and the traditional distinctions made between fact and fiction, history and story. King's Green Grass. Running Water takes stories from Western European literary traditions and Judeao-Christian mythology and presents them as part of a Native creation story. Welch's novel Fools Crow re-writes a particular episode from history, the Marias River Massacre, from a Blackfeet perspective. Silko's Almanac of the Dead recreates the Mayan creation story o f the Popol Vuh in the context o f twentiethcentury American culture. Each of these authors maintains the dialogic fluidity of oral storytelling performance in written forms and suggests that stories not only reflect the world, but that they create it in the way that Robinson understands storytelling as a form of theory. / Arts, Faculty of / English, Department of / Graduate

Compreende ainda que a relação colonial institui linhas abissais entre metrópole e colônia (SANTOS, 2002c). Esta divisão abissal produz não-existências, apagando os saberes, racionalidades e pessoas do lado colonial da linha abissal. Este trabalho se centra no apagamento epistêmico, tendo como objeto a universidade, que tem papel de destaque na afirmação do pensamento abissal que classifica o saber científico moderno como o único válido (SANTOS; MENESES, 2009). Buscando compreender como o pensamento de Boaventura de Sousa Santos tem se inserido neste debate no Brasil a dissertação analisa teses e dissertações que tratam do pensamento pós-colonial/decolonial, e têm como referencial teórico o autor. Foram identificadas contribuições valiosas de Santos tanto no sentido do diagnóstico das relações de dominação e colonialidade, evidenciando suas diferentes expressões em diferentes contextos (principalmente no caso luso-brasileiro), quanto de forma mais propositiva, a partir de sua noção de ecologia de saberes visando um pensamento pós-abissal. / This paper starts from the principle that the colonial situation, although formally ended with independence, persists in Brazil economically, socially and epistemologically (as in other countries). It also comprises that the colonial relationship establishes abyssal lines between metropolis and colony (SANTOS, 2002c). This abyssal line produces non-existences, erasing the knowledge, rationalities and people from the colonial side. This paper focuses on that epistemic erasure, having as object the University, which plays a prominent role in the affirmation of the abyssal thinking that classifies the modern scientific knowledge as the only valid one (SANTOS; MENESES, 2009). In order to comprehend how Boaventura de Sousa Santos thinking has been inserted in this debate in Brazil, the dissertation analyzes other theses and dissertations that deal with postcolonial/decolonial thinking and have, as theoretical reference, the author. Santos' valuable contributions were identified both in the sense of diagnosing the relations of domination and coloniality, highlighting their different expressions in different contexts (mainly in the Portuguese-Brazilian case) and, in a more purposeful way, based on their notion of ecology of knowledges seeking a post-abyssal thinking.

Computational analysis of transcriptional responses to the Activin signal

Die Signalwege des transformierenden Wachstumsfaktors β (TGF-β) spielen eine entscheidende Rolle bei der Zellproliferation, -migration und -apoptose durch die Aktivierung von Smad-Proteinen. Untersuchungen haben gezeigt, dass die biologischen Wirkungen des TGF-β-Signalwegs stark vom Zellkontext abhängen. In dieser Arbeit ging es darum zu verstehen, wie TGF-β-Signale Zielgene unterschiedlich regulieren können, wie unterschiedliche Dynamiken der Genexpression durch TGF-β-Signale induziert werden und auf welche Weise Smad-Proteine zu unterschiedlichen Expressionsmustern von TGF- β-Zielgenen beitragen. Der Fokus dieser Studie liegt auf den transkriptionsregulatorischen Effekten des Nodal / Activin-Liganden, der zur TGF-β-Superfamilie gehört und ein wichtiger Faktor in der frühen embryonalen Entwicklung ist. Um diese Effekte zu analysieren, habe ich kinetische Modelle entwickelt und mit den Zeitverlaufsdaten von RNA-Polymerase II (Pol II) und Smad2-Chromatin-Bindungsprofilen für die Zielgene kalibriert. Unter Verwendung des Akaike-Informationskriteriums (AIC) zur Bewertung verschiedener kinetischer Modelle stellten wir fest, dass der Nodal / Activin-Signalweg Zielgene über verschiedene Mechanismen reguliert. Im Nodal / Activin-Smad2-Signalweg spielt Smad2 für verschiedene Zielgene unterschiedliche regulatorische Rollen. Wir zeigen, wie Smad2 daran beteiligt ist, die Transkriptions- oder Abbaurate jedes Zielgens separat zu regulieren. Darüber hinaus werden eine Reihe von Merkmalen, die die Transkriptionsdynamik von Zielgenen vorhersagen können, durch logistische Regression ausgewählt. Der hier vorgestellte Ansatz liefert quantitative Beziehungen zwischen der Dynamik des Transkriptionsfaktors und den Transkriptionsantworten. Diese Arbeit bietet auch einen allgemeinen mathematischen Rahmen für die Untersuchung der Transkriptionsregulation anderer Signalwege. / Transforming growth factor-β (TGF-β) signaling pathways play a crucial role in cell proliferation, migration, and apoptosis through the activation of Smad proteins. Research has shown that the biological effects of TGF-β signaling pathway are highly cellular-context-dependent. In this thesis work, I aimed at understanding how TGF-β signaling can regulate target genes differently, how different dynamics of gene expressions are induced by TGF-β signal, and what is the role of Smad proteins in differing the profiles of target gene expression. In this study, I focused on the transcriptional responses to the Nodal/Activin ligand, which is a member of the TGF-β superfamily and a key regulator of early embryonic development. Kinetic models were developed and calibrated with the time course data of RNA polymerase II (Pol II) and Smad2 chromatin binding profiles for the target genes. Using the Akaike information criterion (AIC) to evaluate different kinetic models, we discovered that Nodal/Activin signaling regulates target genes via different mechanisms. In the Nodal/Activin-Smad2 signaling pathway, Smad2 plays different regulatory roles on different target genes. We show how Smad2 participates in regulating the transcription or degradation rate of each target gene separately. Moreover, a series of features that can predict the transcription dynamics of target genes are selected by logistic regression. The approach we present here provides quantitative relationships between transcription factor dynamics and transcriptional responses. This work also provides a general computational framework for studying the transcription regulations of other signaling pathways.

Metaphors, Myths, and Archetypes: Equal Paradigmatic Functions in Human Cognition?

The overview of contributions to metaphor theory in Chapters 1 and 2, examined in reference to recent scholarship, suggests that the current theory of metaphor derives from long-standing traditions that regard metaphor as a crucial process of cognition. This overview calls to attention the necessity of a closer inspection of previous theories of metaphor. Chapter 3 takes initial steps in synthesizing views of domains of inquiry into cognitive processes of the human mind. It draws from cognitive models developed in linguistics and anthropology, taking into account hypotheses put forth by psychologists like Jung. It sets the stage for an analysis that intends to further understanding of how the East-West dichotomy guides, influences, and expresses cognitive processes. Although linguist George Lakoff denies the existence of a connection between metaphors, myths, and archetypes, Chapter 3 illustrates the possibility of a relationship among these phenomena. By synthesizing theoretical approaches, Chapter 3 initiates the development of a model suitable for the analysis of the East-West dichotomy as exercised in Chapter 4. As purely emergent from bodily experience, however, neither the concept of the East nor the concept of the West can be understood completely. There exist cultural experiences that may, depending on historical and social context, override bodily experience inclined to favor the East over the West because of the respective connotations of place of birth of the sun and place of death of the sun. This kind of overriding cultural meaning is based on the “typical, frequently recurring and widely shared interpretations of some object, abstract entity, or event evoked in people as a result of similar experiences. To call these meanings ‘cultural meanings' is to imply that a different interpretation is evoked in people with different characteristic experiences. As such, various interpretations of the East-West image-schema exist simultaneously in mutually exclusive or competing forms, as the analysis of Gatsby and the reversal of the values of East and West in the context of colonizing and counter-colonizing attitudes suggests.

John Buchan (1875-1940) and the First World War: A Scot's Career in Imperial Britain

This dissertation examines the political career of Scottish-born John Buchan (1875-1940) who, through the avenue of the British Empire, formed political alliances that enabled him to enter into the power circles of the British government. Buchan's involvement in governmental service is illustrative of the political and financial advantages Scots sought in Imperial service. Sources include Buchan's published works, collections of correspondence, personal papers, and diaries in the holdings of the National Library of Scotland, Edinburgh. Letters and other documents pertaining to Buchan's life and career are also available in the Beaverbrook papers, Lloyd George papers, and Strachey papers at the House of Lords Record Office, London, and in the Liddle Hart Collection at King's College, London. Documents concerning Buchan's association with the War Cabinet, the Foreign Office, and the Department of Information are among those preserved at the Public Record Office, London. References to Buchan's association with the British Expeditionary Force in France are included in the holdings of the Intelligence Corps Museum, Ashford, Kent. The study is arranged chronologically, and discusses Buchan's Scottish heritage, his education, his assignment on Lord Alfred Milner's staff in South Africa, and his appointment as Director of the Department of Information during World War I. The study devotes particular attention to Buchan's leadership of the Department of Information, a propaganda arm of the British government during the First World War. Buchan consolidated independent branches of propaganda production and distribution, and coordinated the integration of information provided by the British Foreign Office, War Office, and the Department of Information's Intelligence Bureau to forward Britain's propaganda effort. The study also considers his literary contributions, his Parliamentary service, and, when raised to the peerage as Lord Tweedsmuir of Elsfield, his royal commission as Governor-General of Canada. This dissertation concludes that, while pursuing an imperial career, John Buchan established a relationship with a powerful clique that enabled him to become part of the machinery of state.

Developmental regulomes that drive tissue-specific and temporally controlled gene expression in Drosophila melanogaster

Während der Entwicklung des Organismus führen naive Zellen aufgrund eines streng regulierten Transkriptionsprogramms zu differenzierten Zelltypen und Geweben. Obwohl viele Aspekte dieses Differenzierungsprozesses noch wenig verstanden sind, ist allgemein anerkannt, dass Transkriptionsfaktoren (TFs), die mit cis-regulatorischen Modulen (CRMs), nämlich Enhancern, interagieren, einen wesentlichen Beitrag zur Regulierung der räumlich-zeitlichen Genexpression leisten. Um die regulatorischen Wechselwirkungen von Enhancern zu verstehen, verwendete ich eine Technik namens inSTEP, von zwei wichtigen neurogenen Enhancern und einem mesodermalen Enhancer zu entschlüsseln. inSTEP ist eine Abkürzung für in vivo Spatio-Temporal Enhancer Proteomics und beinhaltet die Präzipitation eines ausgewählten Enhancers zusammen mit all seinen gebundenen Elementen aus einem bestimmten Gewebe zur Identifizierung durch Massenspektrometrie (MS), wodurch die Identifizierung von regulatorischen Kandidaten ermöglicht wird, die die Neurogenese vorantreiben. Although many aspects of this differentiation process are still poorly understood, it is widely accepted that transcription factors (TFs) interacting with cis-regulatory modules (CRMs), namely enhancers, are major contributors to regulate spatio-temporal gene expression. In order to understand the regulatory interactions of enhancers, I used a technique called inSTEP to unravel the enhancer-protein interactions on two major neurogenic enhancers (for the vnd and rho genes) and one mesodermal enhancer (1070enhancer), for which no target genes are known. inSTEP is an acronym for in vivo Spatio-Temporal Enhancer Proteomics and entails precipitation of a chosen enhancer together with all its bound elements from a specific tissue, for identification by mass spectrometry (MS), thus enabling the identification of regulatory candidates driving neurogenesis. I have identified candidate regulators in the ventral column and selected ten to do follow-up experiments The knock down of at least two of the vndenhancer putative regulators, CG4707 and CG2962, led to an altered reporter gene expression pattern driven by the vndenhancer, suggesting that inSTEP is able to identify new regulatory proteins involved in the regulation of gene expression in the developing nervous system. One of the enhancers I am most interested in is an enhancer for the gene vnd, which encodes a crucial TF for neurogenesis. I have therefore expanded my project beyond the question of ‘how’ vnd expression is regulated, to also include the role Vnd itself plays in neurogenesis. I have conducted ChIP-seq experiments to elucidate the genome-wide binding profiles of Vnd and I have developed tools that will elucidate the isoform-specific role of Vnd.

El nombrar como necesidad. Ética del discurso y lenguaje inclusivo

La presente investigación tiene como finalidad reordenar los cuestionamientos planteados a la ética discursiva de Jürgen Habermas a partir de una mirada teórica y crítica sobre las funciones del lenguaje con el fin de ofrecer una nueva lectura desde la función representativa del lenguaje. Este enfoque permitirá comprender a detalle la utilidad de una perspectiva interdisciplinaria, desde la Filosofía y la Lingüística. De esta manera, se podrán rescatar nuevas contribuciones a la misma que no solo se concentren en la caracterización específica y formal que propone Habermas de tales funciones. El primer capítulo versará sobre el análisis de las funciones del lenguaje como sostén del pensamiento y como herramienta de comunicación. Sobre la base de las perspectivas que presentan las autoras Carol Gilligan e Iris Young, se establecerá un breve estado de la cuestión sobre las aristas que pueden ampliar el alcance efectivo de la ética discursiva en la realidad social. El segundo capítulo se concentrará en el estudio de la función del lenguaje como representación del mundo. A partir de la teoría lingüística de Benjamin y las recientes investigaciones acerca del lenguaje inclusivo, se ofrecerá un nuevo planteamiento que sea útil para entender la ética discursiva también como una que debe considerar los nombres, las representaciones y los contenidos de cada grupo social dentro del discurso. De tal manera, la ética discursiva puede alcanzar no solamente todos los niveles de la sociedad a la que es aplicada, sino que también puede corresponderse con los avances que brindan los actuales estudios en Lingüística de una manera más exacta.

RNA Polymerase II identifies enhancers in different states of activation

Enhancer regulieren die Transkription ihrer Zielgene und deren Expression. Sie bieten eine Bindestelle für verschiedenste Transkriptionsfaktoren (TF) und RNA Polymerase II (RNAPII) und unterstützen die Gentranskription durch das Zustandekommen von Chromatinkontakten. Zusätzlich transkribiert RNAPII in Enhancer-Regionen kurze, non-polyadenylierte Transkripte, die man Enhancer-RNA (eRNA) nennt. Der Mechanismus der RNAPII-Rekrutierung und –Regulation an Enhancern ist bisher wenig verstanden, insbesondere wie das Vorhandensein von RNAPII-Modifikationen den Chromatinstatus, -faltung sowie die Genaktivierung beeinflusst. In dieser Arbeit wurden verschiedene Ansätze der Enhancer-Bestimmung miteinander verglichen. Während eine klare Bestimmung des besten Ansatzes sich als komplex erwies, konnte gezeigt werden, dass die Bindung von RNAPII an regulatorische Regionen in Zusammenhang mit TF eine universelle Konstante darstellte. Weiterhin wurden der Status der Enhancer-gekoppelten RNAPII-Aktivierung und deren Transkriptionsaktivität untersucht. Als Hauptergebnis ergab sich, dass der RNAPII-Status mit der Enhancer-Aktivität und daraus folgend mit veränderter Transkriptionsaktivität korreliert ist. Weiterhin konnte gezeigt werden, dass das Vorhandensein extragenischer RNAPII ein neues Werkzeug zur Identifikation von regulatorischen Regionen ist. Erfolgreich konnten regulatorische Regionen in embryonalen Stammzellen der Maus sowie während der neuronalen Differenzierung vorhergesagt und mittels Enhancer-Aktivität in-vivo bestätigt werden. Dabei zeigte sich, dass im Laufe der der neuronalen Differenzierung extragenische RNAPII-Bindung spezifische Aktivierungsmuster aufweist: ihr Transkriptionslevel wird durch Kinasen feinmaschig reguliert und es werden verschiedene Formen maturierter RNA erzeugt. Zusammenfassend konnte RNAPII als Werkzeug zur Identifikation und Charakterisierung regulatorischer Regionen in verschiedenen Zelltypen ausgemacht werden. Selbst mit minimalen RNAPII-Datensätzen ist es möglich, gleichzeitig regulatorische Regionen zu identifizieren als auch ihren eigenen Aktivierungsstatus sowie den ihrer kodierender Genpromotoren zu bestimmen. / Enhancers regulate transcription of target genes and gene expression. They act as recruitment sites for multiple transcription factors (TFs) and RNA polymerase II (RNAPII) and favour transcription of target genes through chromatin contacts. RNAPII at enhancer regions transcribes short and mostly non-polyadenylated transcripts, called enhancer RNAs (eRNAs). The mechanisms of RNAPII recruitment and regulation at enhancers remain ill understood, in particular how signalling through RNAPII modifications may influence chromatin states, looping and gene activation. In this study, I compare enhancer lists defined with different approaches and find that their relation is very complex. However, I find that RNAPII binding co-occurs with TF binding at regulatory regions, independently of the identification approach used. I characterize the state of RNAPII activation at enhancers and its transcriptional activity. I find that RNAPII state reflects enhancer activation state and correlates with different transcriptional outputs. In addition, I demonstrate that extragenic RNAPII is a novel tool to identify regulatory regions. I successfully identified putative regulatory regions in mESC and during neuronal differentiation, with enhancer activity in vivo. Extragenic RNAPII regions have specific activation patterns during neuronal differentiation, are finely regulated at the transcriptional level by kinases and transcribe differently mature RNAs. In conclusion, I establish RNAPII as a tool to identify and characterise regulatory regions in a cell type of interest. With minimal RNAPII datasets it is possible to simultaneously identify regulatory regions, infer their state of activation, and the state of activation of coding gene promoters.
