This dissertation thesis addresses the history of public relations of the Communist Party in Czechoslovakia in Pardubice district. It is focussed at the period of so-called normalization. Firstly, it discusses the historical context of the time for understanding the crisis of the system in 70s and 80s. The first chapters describe so-called Prague spring and military invasion in August 1968. Next chapters stress changes in politics and media after the election of Gustáv Husák to become First Secretary of the Communist Party in April, 1969. The second part of text is case study about Pardubice district. The main topic of dissertation thesis are newspaper of Pardubice district - Zář. But there are also chapters about radio, factory newspaper, regional Union of Czech Journalists or College in Pardubice and editors of student magazine. In the second part of case study attention is focussed on cultural section of newspaper Zář and its main topics - for example political anniversaries and the role of culture. In cultural section I analyze which information and how they were, or were not presented to public. Supervision and control over the media content was one of the main regime's tools to take over the culture area and its presentation.

We have tried to observe how the dynamic Brazilian economy, in which the Companhia Sider?rgica Nacional (the most important steel mill in the country) has been an important protagonist, since its creation in the 1940?s, related to the ways in which this company engendered its labor force, in clash with workers mobilizations. The most important landmark in this trajectory, after the democratic years (1945-1964), is the fracture caused by the 1964 state coup in the relationship between workers and company. The changes in CSN social policies, as well as the transformations in the economic policies implemented by the new regime, would be subject to a confrontation between different interpretations formulated by distinct agents involved in this process. In our case study, Volta Redonda, we could identify as important actors involved in the social dynamics of those years ? for example, the military ? interpreted the impacts of the coup on the working class. In the other extreme of the process, we have examined the workers experience in the town, in decade of 1970?s, looking for the rearticulation of their identitary references, after those changes in the general context, that indicate the new fractures and amalgamations inside the class occurred from that moment on / Este trabalho ? uma tentativa de articula??o entre as propostas da hist?ria social do trabalho e uma hist?ria social urbana. The 1990&#039 / s witnessed the emergence of Constructivist approach in the International Relations theory. Constructivism rejecting the rationalist precepts of Neo-Realism and Neo-Liberalism brought a sociological point of view to the world politics mentioned the role of ideational factors in the constitution process of interests and identities. Constructivism assumes that there is a mutual constitution process between states and normative structures in which states internalized existing international societal norms with efforts of international non-state actors seen as active teachers guiding states to initiate policies that are congruent with international civilized norms of behaviour. Today human rights are defined as civilized normative patterns. At this point five phase spiral model reveals itself and introduces explanations for how states understand, interpret and internalize international human rights norms. The model assumes that target state&#039 / s identity, interest and behaviors are influenced by international human rights norms as it progresses through the model&#039 / s five phased spiral process. The model in this process emphasizes roles and efforts of international organizations. v This work assesses the usefulness of the five phase spiral model and Constructivism as an explanation of the changes in the Turkish government&rsquo / s human rights practices. As case study effect of the European Union over normative transformation in the field of human rights in Turkey will be given. The thesis bringing criticisms to the model&rsquo / s assumptions in Turkey case will assert that Turkey, from the 1980 Military intervention to the end of 2007 has made progressive steps in line with the phases mentioned in the spiral model.

Turkey has been one of the few countries that signed the 1951 Convention relating to the Status of Refugees with the provision of maintaining geographical limitation to that of offering protection only to European nationals. This is, however, expected to change as Turkey heads towards EU membership. Since 1999, Turkey has been declared as a candidate country to the European Union (EU), in the Helsinki Summit. It is expected to adopt EU Asylum Acquis into its legislation and to lift the geographical limitation of the 1951 Geneva Convention. This study aims to analyze EU&rsquo / s Common Asylum Policy in order to present a comprehensive overview to EU Asylum Acquis and practices that are expected to be adopted by Turkey during the pre-accession process. The aim of this thesis is to analyze deficiencies of European Common Asylum Policy and its potential positive and negative effects on Turkey&rsquo / s asylum policy.

Noncontributory pensions, cash transfers, and documentation in Brazil and Latin America

Since 1997, fully noncontributory minimum pensions have been established in many Latin American countries, and have more recently been encouraged as a "zero pillar" of social security by the World Bank and other IFIs. These policies came into being under diverse political regimes and display a range of levels of generosity and universality. Becuase these policies are generally part of a modern bureaucratic welfare state project, they require identity documents, something that many low-income citizens do not possess. States have lowered barriers to the supply of identity documents, and new social policies, like noncontributory pensions and conditional or unconditional cash transfers, have stimulated demand for identity documents among those who do not currently have them. Brazils noncontributory pension, the BPC, has a means test and a large benefit (equivalent to the minimum wage), but requires providing identity documents for all household members. This report discusses the propagation of noncontributory pensions, then examines Brazilian government records to determine the size of the incentive to demand documents in Brazil using a logit model and a more novel survival time regression discontinuity design, raising questions of the relationships between benefit size, universality, document requirements, and poverty. / text

La hipótesis que sostendré en este trabajo es que el relato testimonial es intertextual, que recupera el discurso factual y deconstruye los sentidos institucionalizados produciendo un nuevo conocimiento. Con esos presupuestos, afirmo que el sujeto se configura en la relación intersubjetiva a la que abre el relato a partir de la memoria materializada en la escritura. Estas ideas se apoyan sobre la lectura del sistema literario de la Argentina de fin de siglo, operación que me permite recuperar ciertos textos que considero pertenecientes a una clase discursiva a la que he identificado como "relato testimonial".

Étude du rôle des gènes TGF-β1 et HSP-70 lors du processus de régénération du membre chez l’axolotl

Les urodèles amphibiens, dont fait partie l’axolotl (Ambystoma mexicanum), ont la capacité de régénérer leurs organes et membres suite à une amputation, tout au long de leur vie. La patte est l’organe dont le processus de régénération est le mieux caractérisé et ce dernier est divisé en deux phases principales. La première est la phase de préparation et commence immédiatement suite à l’amputation. Elle renferme des étapes essentielles au processus de régénération comme la guérison de la plaie et la formation d’une coiffe apicale ectodermique. Par la suite, les fibroblastes du derme et certaines cellules musculaires vont revenir à un état pluripotent via un processus appelé dédifférenciation cellulaire. Une fois dédifférenciées, ces cellules migrent et s’accumulent sous la coiffe apicale pour former le blastème. Lors de la phase de redéveloppement, les cellules du blastème se divisent puis se redifférencient pour régénérer la partie amputée. Fait intéressant, la régénération d’un membre ou la guérison d’une plaie chez l’axolotl ne mène jamais à la formation d’une cicatrice. Afin d’en apprendre plus sur le contrôle moléculaire de la régénération, les gènes Heat-shock protein-70 (Hsp-70) et Transforming growth factor-β1 (Tgf-β1) ont été sélectionnés. Ces gènes jouent un rôle important dans la réponse au stress et lors de la guérison des plaies chez les mammifères. HSP-70 est une chaperonne moléculaire qui est produite pour maintenir l’intégrité des protéines cellulaires lorsqu’un stress se présente. TGF-β1 est une cytokine produite suite à une blessure qui active la réponse inflammatoire et qui stimule la fermeture de la plaie chez les amniotes. Les résultats présentés dans cette thèse démontrent que Hsp-70 est exprimé et régulé lors du développement et de la régénération du membre chez l’axolotl. D’autre part, nos expériences ont mené à l’isolation de la séquence codante pour Tgf-β1 chez l’axolotl. Nos résultats montrent que Tgf-β1 est exprimé spécifiquement lors de la phase de préparation dans le membre en régénération. De plus, le blocage de la voie des Tgf-β avec l’inhibiteur pharmacologique SB-431542, lors de la régénération, mène à l’inhibition du processus. Ceci démontre que la signalisation via la voie des Tgf-β est essentielle à la régénération du membre chez l’axolotl. / Urodele amphibians, such as the axolotl (Ambystoma mexicanum), have the unique ability, among vertebrates, to perfectly regenerate many parts of their body throughout their life. Among the complex structures that can be regenerated, the limb is the most widely studied. Limb regeneration is divided in two main phases. The preparation phase, which begins right after amputation, includes wound healing and the formation of an apical ectodermal cap. During this phase, dermal fibroblasts and muscle cells will lose their characteristics and become pluripotent through a process called cellular dedifferentiation. The dedifferentiated cells migrate and accumulate under the apical ectodermal cap to form the blastema. During the redevelopment phase, the cells in the blastema proliferate and redifferentiate to regenerate the lost structures. It is interesting to highlight the fact that regeneration never leads to scar formation in the axolotl. In order to learn more about the molecular control of limb regeneration, the genes Heat-shock protein-70 (Hsp-70) and Transforming growth factor-β1 (Tgf- β1) were selected for their important roles in stress response and wound healing in mammals. HSP-70 is a molecular chaperone which is produced to protect cellular proteins when the cell faces a stress. TGF-β1 is a cytokine produced after wounding that activates the inflammatory response and stimulates wound closure in amniotes. Results presented in this thesis show that Hsp-70 is expressed and regulated during limb development and regeneration in the axolotl. We were also able to isolate the cDNA coding for axolotl Tgf-β1 and our results show that this gene is expressed specifically during the preparation phase of limb regeneration. Treatment of regenerating axolotls with a specific inhibitor of Tgf-β signalling, SB-431542, led to complete inhibition of regeneration. This directly implies that Tgf-β signalling is essential for limb regeneration in axolotl.

Primary T cell immunodeficiencies associated with disturbed proximal T cell receptor signalling caused by human autosomal recessive LCK, ZAP-70 and ITK-mutations

T lymphocytes express either a preTCR, or a clonotyoic γδ TCR or αβ TCR together with the CD3-complex and the associated ζ-chain. TCR:CD3:ζ-signalling is crucial for T cell development and antigen-specific activation including proliferation, differentiation, effector functions and apoptosis of mature T cells. Protein tyrosine kinase (PTK) cascades lie at the heart of proximal TCR:CD3:ζ-signalling. The CSK-, SRC-, SYK- and TEC-family members C-terminal SRC kinase (CSK), lymphocyte-specific protein tyrosine kinase (LCK), ζ-chain associated protein tyrosine kinase of 70 kDa (ZAP-70) and interleukin-2-inducible T cell kinase (ITK), respectively, are the major T cell players. After TCR:CD3:ζ-complex triggering, activation of PTKs result in tyrosine phosphorylation signals. These include phosphorylation of immunoreceptor tyrosine-based activation motifs (ITAMs) of the CD3 and ζ-chains, adaptor proteins that nucleate the proximal LAT:SLP-76-signalosome controlling almost all TCR:CD3:ζ-induced signalling events. These events initiate Ca2+-flux, activation of mitogen-activated protein kinases (MAPKs), activation of nuclear factor of kappa light polypeptide gene enhancer in B-cells (NF-κB), activation of nuclear factor of activated T cells (NFAT) and activator protein 1 (AP-1) as well as actin reorganization, cell-adhesion and motility.Througout the last five decades, the immune system has been extensively investigated in vitro and in animal models such as the murine system. Additionally, studying and taking care of human primary immunodeficiency diseases (PIDs) has been seminal for our understanding of the human immune system as animal models not always recapitulates the subtleties found in men.In my doctoral thesis I report the first case of autosomal recessive human LCK-deficiency, a novel autosomal recessive mutation leading to human ZAP-70-deficiency and a novel autosomal recessive mutation leading to human ITK-deficiency. I provide detailed clinical, immunological and biochemical analyses especially of TCR:CD3:ζ-signalling and compare my findings to the well-established Lck-/-, Zap-70-/- and Itk-/- murine models.
