T lymphocytes express either a preTCR, or a clonotypic γδ TCR or αβ TCR together with the CD3-complex and the associated ζ-chain. TCR:CD3:ζ-signalling is crucial for T cell development and antigen-specific activation including proliferation, differentiation, effector functions and apoptosis of mature T cells. Protein tyrosine kinase (PTK) cascades lie at the heart of proximal TCR:CD3:ζ-signalling. The CSK-, SRC-, SYK- and TEC-family members C-terminal SRC kinase (CSK), lymphocyte-specific protein tyrosine kinase (LCK), ζ-chain associated protein tyrosine kinase of 70 kDa (ZAP-70) and interleukin-2-inducible T cell kinase (ITK), respectively, are the major T cell players. After TCR:CD3:ζ-complex triggering, activation of PTKs results in tyrosine phosphorylation signals. These include phosphorylation of immunoreceptor tyrosine-based activation motifs (ITAMs) of the CD3 and ζ-chains, and adaptor proteins that nucleate the proximal LAT:SLP-76-signalosome controlling almost all TCR:CD3:ζ-induced signalling events. These events initiate Ca2+-flux, activation of mitogen-activated protein kinases (MAPKs), activation of nuclear factor of kappa light polypeptide gene enhancer in B-cells (NF-κB), activation of nuclear factor of activated T cells (NFAT) and activator protein 1 (AP-1) as well as actin reorganization, cell-adhesion and motility. Throughout the last five decades, the immune system has been extensively investigated in vitro and in animal models such as the murine system. Additionally, studying and taking care of human primary immunodeficiency diseases (PIDs) has been seminal for our understanding of the human immune system as animal models not always recapitulate the subtleties found in men. In my doctoral thesis I report the first case of autosomal recessive human LCK-deficiency, a novel autosomal recessive mutation leading to human ZAP-70-deficiency and a novel autosomal recessive mutation leading to human ITK-deficiency. I provide detailed clinical, immunological and biochemical analyses especially of TCR:CD3:ζ-signalling and compare my findings to the well-established Lck-/-, Zap-70-/- and Itk-/- murine models.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / Trajetória do grupo de literatura Totem, que se formou na cidade mineira de Cataguases e, no período dos anos de 1960 a 80, projetou sua obra no seu estado, atingiu muitas regiões do país e chegou ao exterior, por meio da edição de jornais, cartões-poemas, revistas e livros, bem como através de volumosa correspondência. Sua contribuição, principalmente na área da poesia experimental e da abertura para outros grupos, procurou sempre a pesquisa e a reformulação da linguagem poética. Buscou-se aqui o registro da experiência de uma equipe de jovens poetas, da qual participou o autor desta dissertação, e que, a partir de uma cidade do interior de Minas Gerais, conseguiu se situar bem no meio desse caldeirão de idéias que teve início na década de 60. / The trajectory of Totem literature group, formed in Cataguases, a small town located in Minas Gerais, Brazil. From the 1960s to the 1980s, Totem circulated its work in this State, reached a lot of parts of the country and abroad, through newspapers issues, poem-cards, magazines and books, as well as a great amount of correspondence. Its contribution, specially in the area of experimental poetry and the opening of other groups, Totem was always focused in poetry language reformulation and research. This paper aimed to record the experience of a group of young poets to which the author of this thesis belonged, describing how from that small town in Minas Gerais that group succeeded in including itself among the other groups and ideas of the early 1960s.

Por fim, com base em todas as discuss?es promovidas, apresenta-se uma proposta de disciplina capaz de efetivar essa medida nas grades dos n?veis fundamental e m?dio de ensino no Brasi. / The fundamental right to education, the social nature and the demand for a positive act of the State, includes the formation of citizens and prepares education for the exercise of citizenship. Its implementation is a prerequisite for the exercise of several other rights, since there is no human dignity without an effective right to education. In this perspective, it is the role of elementary education and legal education and a diffusion of knowledge related to the constitutional content, both regarding the protection given to fundamental rights and not so important in the structures of the State and the mechanisms inherent in citizen action. Based on this assumption, we analyze the feasibility of presenting our curricula of the basic education of legal-constitutional education, with perspectives for the realization of citizenship through the right to education. Therefore, they are considered as contributions of the main critical and post-critical curricular theories, promoting an understanding of the role of the school, which no longer deserve the state ideology apparatus for a reproduction of the status quo, in the molds of the Althusserian critic Mas It should serve as a space for the formation of the humanities and the diffusion of respect for the demands and recognition of minorities. This reflection can not ignore the vast theorizing around the human rights education system, an international tendency and offer several nortes for an application of public policies tendencies to promote the diffusion of legal-constitutional education in the school environment. Once the content of legal education has been established, and as appropriate methodologies for the practice, taking into account a wide range of possibilities found in a disposition of teachers and students, as well as In building a culture of respect for human rights. In addition, they are considered as historical pedagogical experiences that have already taken place in Brazil, Brazil, Brazil, and Brazil, Senate Law no. 70/2015, which aims to amend a Law of Guidelines and Bases of Education to promote Inclusion of legal-constitutional education in Brazilian basic education curricula. Contudo, esta hipóxia se contrapõe à intensa atividade das células que requerem elevado metabolismo, gerando um estresse fisiológico para estas células presentes na interface materno-fetal. Presume-se que estas células necessitem de mecanismos apropriados de citoproteção para sua sobrevida enquanto comprometidos ativamente no suporte funcional do útero gestante. Neste sentido, o presente trabalho teve como objetivo investigar a expressão e a distribuição da proteína de choque térmico 70 (HSP70) na interface materno-fetal durante a gestação normal em camundongos e a sua possível variação em condição de estresse adicional induzido experimentalmente através da lesão embrionária. Sítios de desenvolvimento embrionário/fetal de camundongos prenhes do dia de gestação (dg) 6 ao 17 e, após 30 minutos, 1, 6 e 12 h dos animais submetidos à lesão cirúrgida do embrião (LCE) no dg 9 foram coletados para: - processamento histotécnico convencional de embebição em parafina destinados às análises citoquímicas (lectina DBA e reação de TUNEL) e imunocitoquímicas (anti-HSP72/73, anti-PCNA); - embebição em resina Lowilcryl- K4M para imunocitoquímica ultraestrutural (anti-HSP72/73); - obtenção de homogenados teciduais destinados à SDS-PAGE das frações protéicas e Westernblot (anti-HSP72/73) e, - extração de RNA de homogenados teciduais e de células uNK isoladas para análise de transcritos (HSP72 e 73) com amplificação pelo RTPCR. As análises imunocitoquímicas demonstraram que as células uNK eram as únicas células que expressavam de forma constante as isoformas HSP72/73 ao longo da gestação, sendo confirmada a expressão dos transcritos gênicos das isoformas HSP72/73 nas células uNK isoladas pelo RT-PCR. A imunomicroscopia eletrônica detectou marcação conspícua nas mitocôndrias das células uNK. A análise quantitativa demonstrou que a lesão do embrião reduz o número de células uNK positivas para HSP72/73 e, o SDS/PAGE/Western-blotting identificou as isoformas HSP72 e 73 presente nos homogenados teciduais do útero com uma perceptível redução na intensidade da banda correspondente ao HSP73 nas amostras de pós-lesão, sem afetar significativamente a isoforma HSP72. However, this hypoxia is contradictory to the great dynamism and metabolism of cells required in the pregnant uterus, conditioning a physiological stress for the cells present at the maternal-fetal interface. It is presumed these cells demand appropriate cytoprotective mechanism for their survival while are committed to actively support the pregnancy. In this way, the present work aimed to investigate the expression and distribution of the chapelone isoforms heat shock protein 72 and 73 (HSP72/73) at the maternal fetal-interface through the pregnancy in mice and its possible variations under additional stressing condition induced experimentally by embryo lesion. Embryo/fetus developing sites of pregnant mice from gestational days (gd) 6 to 17 and, after 30min, 1h and 6h of surgical embryo lesion (SEL) on gd 9 mice, were collected for: - conventional paraffin embedding for cytochemical (DBA lectin and TUNEL reaction) and immunocytochemical (anti-HSP72/73, anti-PCNA) analysis; - LR-white resin embedding for ultrastructural immunocytochemistry (anti- HSP72/73); - uterine tissue homogenates for SDS-PAGE of proteins fractions and Western-blot (anti-HSP7273) and; - RNA extratction form uterine tissue homogenates and isolated uNK cells for transcripts (HSP72 and 73) amplification by RT-PCR. The immunocytcchemical analysis showed the uNK cells as the only cell expressing constantly the HSP72/73 isoforms throughout the gestation, being confirmed the expression of both gene isoforms by RT-PCR in uNK cells. The immunoelectron microscopy detected conspicuous labeling in the mitochondria of uNK cells. The quantitative analysis demonstrated that embryo-lesion reduced the number of HSP72/73 positive uNK cells in the uterus and, SDS/PAGE and Westernblot identified the HSP72 and 73 isoforms present in the tissue homogenates with low reactive intensity of the band corresponding to HSP73 in the after-lesion samples, without affecting significantly the HSP72 isoform. The analysis of TUNEL and PCNA double labelling showed decreasing of PCNA positive-uNK cells in the uterus after embryo-lesion and increasing of TUNEL positive nuclei. These results confirms the expression of HSP72 and HSP73 isoforms in the uNK cells through the gestation and to date, this is also the first report showing HSP70 in leukocytes, suggesting a cytoptotective function to this cell while working actively as important cells supporting the pregnancy. Polidutos, tanques de armazenamento de combustíveis e outras estruturas metálicas estão em constante contato com diferentes tipos de solos, diversos horizontes ou camadas, expostos à ação corrosiva nessa interação. Isso possibilita a ocorrência de danos de conseqüências expressivas ao ambiente e à expectativa de trabalho, podendo implicar em contaminações derivadas por vazamento dos produtos de estocagem, derramamento durante transporte, acidentes causados por tubulações sem adequada manutenção além das perdas financeiras associadas. Conhecer a dinâmica de interações entre as superfícies metálicas e o entorno é fundamental a fim de prever comportamentos e desenvolver materiais melhor aplicados. Os aços classe API são utilizados em tubulações de alta e baixa pressão mantendo características de elevada resistência mecânica e à corrosão e, ainda assim, são suscetíveis de ocorrências do processo corrosivo, dado que, estão em contanto com diferentes ambientes desde regiões de mangue até atmosferas industriais. No presente caso, avaliou-se como se comportam aços API 5L X70 em contato com diferentes horizontes de alguns solos no Brasil. O estudo correlacionou a presença de espécies químicas como íons formados em solução, minerais primários e secundários. Além disso, foram avaliadas as características físicas e químicas como pH, condutividade elétrica, sólidos totais dissolvidos, entre outras, com as respostas quanto à resistência e formas de corrosão. Para tanto, foram utilizadas técnicas como difratometria de raios X, microscopia eletrônica de varredura, fluorescência por raios X por reflexão total, análise de injeção de fluxo além de análises de granulometria e gravimetria, dentre outras. Isto foi feito com a finalidade de obter a caracterização dos solos, a identificação dos produtos de corrosão, a análise da solução dos solos, a avaliação dos metais ensaiados e a classificação das formas e tipos de corrosão. Os resultados demonstraram que diferentes horizontes dos solos, assim como os diversos tipos de solos analisados, produzem um ataque diferenciado na estrutura metálica evidenciando essa seletividade. Também, o estudo demonstrou que tais respostas podem ser rapidamente obtidas com o emprego da solução do solo como meio analítico em comparação às avaliações usuais. / Pipelines, fuel storage tanks and other metallic structures are in permanent contact and exposed to different types of soils, of horizons or layers, or of soil aggressiveness. This interaction may cause expressive damages to the environment and to the planned work. Contamination may occur due to leakage of stored products, splitting during transportation, accidents caused by pipelines without extensive maintenance. The result of these accidents could be, among others, some financial losses. In order to recognize the dynamic interactions between metallic surfaces and the environment it is crucial to have preventive actions and to develop better-applied materials. API steel 5L X70 has been used in structures of low and high pressure with high mechanical strength and corrosion and, even so, it is susceptible to etching corrosion since it is in contact with different environments from mangrove regions to industrial environments. The present case evaluated the role of 5L X70 API steel in contact with different soil horizons representative of the Brazilian soil. This investigation correlated chemical species with solute ions in soil solution, secondary and primary phase minerals besides physical and chemical characteristics as pH, electric conductivity, total dissolved solids, among others, to the results of corrosion resistance and ways of corrosion. The evaluation was carried out using x-ray diffractometry, scanning electron microscopy, total reflection x-ray fluorescence, fuel injection flow besides texture and gravimetric analyses to soil characterization and mineralogy, identification of corrosion products, soil solution analyses, evaluation of tested materials and classification of ways and types of corrosion. This was an attempt to integrate the data to a better understanding of the process involving reagents and products. The results showed that different soil horizons such as different types of analyzed soils produce specific etching in metallic structures showing this selectivity. The study also shows that these results can be readily obtained with soil solution as an analytical means in comparison with nowadays evaluations.

M.A. / Christianity was not born in a vacuum, but it completely owes its historical genesis on or from Jewish religion. The Apostolic, Primitive church was initially composed of Jewish believers who had seen in and understood Jesus of Nazareth as the anticipated Messiah predicted by the Old Testament prophets and writers. The current and lamentable truth is, Jewish Religion and Christianity are now two different and separate religions. Jesus Christ, His disciples, and Paul remained Jews as far as the Jewish culture is concerned. It is difficult to separate the sacred and secular in the Jewish economy because Jewish culture and religion are intertwined. In this culture, ploughing is as sacred as worshipping. The only apparent shift in his (Paul) paradigm was in interpreting and explaining the Old Testament prophecies and ceremonial system in the light of the Jesus Christ event at the Cross. His evaluation of that "event" and the Person of Jesus of Nazareth became the point of conflict with Judaisers and Judaism that would set a stage for separation between Christianity and Jewish Religion. The Christian Church exists in the post-Cross era. This Church is faced with, among other issues and tasks, challenges of sifting and demarcating between God's injunctions and Jewish Customs.
