Miranda, Gisele Helena Barboni 24 November 2011 (has links)
Histopathology is considered one of the most important diagnostic tools in medical routine and is characterized by the study of structural and morphological changes of the cells in biological tissues caused by diseases. Currently, the visual assessment of the pathologist is the main method used in the histopathological diagnosis of microscopic images obtained from biopsy samples. This diagnosis is usually based on the experience of the pathologist. The use of computational techniques in the processing of these images allows the identification of structural elements and the determination of inherent characteristics, supporting the study of the structural organization of tissues and their pathological changes. Also, the use of computational methods to improve diagnosis aims to reduce the subjectivity of the evaluation made by the physician. Besides, different tissue characteristics can be mapped through specific metrics that can be used in pattern recognition systems. Within this perspective, the overall objective of this work includes the proposal, the implementation and the evaluation of a methodology for the identification and analysis of histological structures. This methodology includes the specification of a method for the analysis of cervical intraepithelial neoplasias (CINs) from histopathological samples. This work was developed in collaboration with a team of pathologists. Microscopic images were acquired from blades previously stained, containing samples of biopsy examinations. For the segmentation of cell nuclei, a pipeline of morphological operators were implemented. Segmentation techniques based on color were also tested and compared to the morphological approach. For the representation of the tissue architecture an approach based on the tissue layers was proposed and implemented adopting the Delaunay Triangulation (DT) as neighborhood graph. The DT has some special properties that allow the extraction of specific metrics. Clustering algorithms and graph morphology were used in order to automatically obtain the boundary between the histological layers of the epithelial tissue. For this purpose, similarity criteria and adjacency relations between the triangles of the network were explored. The following metrics were extracted from the resulting clusters: mean degree, entropy and the occupation rate of the clusters. Finally, a statistical classifier was designed taking into account the different combinations of clusters that could be obtained from the training process. Values of accuracy, sensitivity and specificity were used to evaluate the results. All the experiments were taken in a cross-validation process (5-fold) and a total of 116 images were used. First, it was evaluated the accuracy in determining the correct presence of abnormalities in the tissue. For this, all images presenting CINs were grouped in the same class. The highest accuracy rate obtained for this evaluation was 88%. In a second step, the discrimination between the following classes were analyzed: Normal/CIN 1; CIN 1/CIN 2, and, CIN 2/CIN 3, which represents the histological grading of the CINs. In a similar way, the highest accuracy rates obtained were 73%, 77% and 86%, respectively. In addition, it was also calculated the accuracy rate in discriminating between the four classes analyzed in this work: the three types of CINs and the normal region. In this last case, it was obtained a rate of 64%.The occupation rate for the basal and superficial layers were the attributes that led to the highest accuracy rates. The results obtained shows the adequacy of the proposed method in the representation and classification of the CINs evolution in the cervical epithelial tissue.

This study was undertaken to monitor the kinetics of the transesterification reaction between soybean oil and methanol catalyzed by potassium methoxide. A series of reactions was monitored with a laser spectrometer used to analyze the absorption and scattering of light in real-time. The objectives of this study aimed to determine when the reaction came in chemical equilibrium. This objective was achieved by observing an arbitrary time interval, experimentally obtained from the graph of electrical resistance versus time, allowing optimize reaction time, by using a monitoring system that combined the information received through multiple sensors. Determine if the optical phenomenon observed during the reaction could be described by Euclidean geometry and whether it was related with the internal conical diffraction. It was found that it is not related to the internal conical diffraction, however, depending on the scale applied to observe the optical phenomenon it can be described by the Euclidean geometry. The goal of developing analytical instrumentation was achieved by constructing a non-invasive laser spectrometer. Chitosan is characterized as an excellent adsorbent material, not only for its adsorption capacity but also the low cost production. Equilibrium and kinetic studies were developed in this study to describe the mechanism of adsorption of the anionic azo dye Orange G in chitosan, with the isotherms obtained from the variation of the concentration of dye in the continuous phase. The kinetics of the process was analyzed based on models involving the adsorption of molecules of the dye in nonpolar and polar sites. Adsorption experiments were carried out in water and in saline media with different NaCl concentrations, both for the determination of the equilibrium time as isotherms for making kinetic curves in which the amount of dye adsorbed measured indirectly varied with time. The experiments revealed the opening of the biopolymer structure with increasing concentration of Orange G, accompanied by high pH values and change on the type of interaction between the dye and the adsorbent surface, suggesting behavior advocated by the Langmuir equation in a certain range of concentration of the adsorbate and following the Henry's Law at higher concentrations, from the increased number of sites available for adsorption. The studies conducted showed that the saline medium reduces the chitosan s adsorption capacity according to a certain concentration, the occurrence of the cooperative adsorption process steps kinetic mechanism suggested as a new alternative for the interpretation of the phenomenon / O fen?meno da adsor??o ? de fundamental import?ncia no tratamento de efluentes t?xteis e remo??o de corantes.

The MCM-41 has been synthesized by hydrothermal method, on which aluminum was added, in the ratio Si/Al equal to 50, to increase the generation of active acid sites in the nanotubes. The catalyst was characterized by X-ray diffraction (XRD), surface area by the BET method and, the average pore volume BJH method using the N2 adsorption, absorption spectroscopy in the infrared Fourier Transform (FT-IR) and determination of surface acidity with application of a probe molecule - n-butylamine. The catalyst showed well-defined structural properties and consistent with the literature. The overall objective was to test the Al-MCM-41 as catalyst and thermogravimetric perform tests, using two samples of heavy oil with API ? equal to 14.0 and 18.5. Assays were performed using a temperature range of 30-900 ? C and heating ratios (β) ranging from 5, 10 and 20 ?C min-1.The aim was to verify the thermogravimetric profiles of these oils when subjected to the action of the catalyst Al- MCM-41. Therefore, the percentage ranged catalyst applied 1, 3, 5, 10 and 20 wt%, and from the TG data were applied two different kinetic models: Ozawa-Flynn-Wall (OFW) and Kissinger-Akahrira-Sunose (KAS).The apparent activation energies found for both models had similar values and were lower for the second event of mass loss known as cracking zone, indicating a more effective performance of Al-MCM-41 in that area. Furthermore, there was a more pronounced reduction in the value of activation energy for between 10 and 20% by weight of the oil-catalyst mixture. It was concluded that the Al-MCM-41 catalyst has applicability in heavy oils to reduce the apparent activation energy of a catalyst-oil system, and the best result with 20% by weight of Al-MCM-41 / Os materiais mesoporosos nanoestruturados vem sendo estudados para aplica??o na ind?stria do petr?leo, em especial o Al-MCM-41, devido ?

Representations of boundary layer cloudiness and surface wind probability distributions in subtropical marine stratus and stratocumulus regions

He, Yanping 16 January 2007 (has links)
Representations of Boundary Layer Cloudiness and Surface Wind Probability Distributions in Subtropical Marine Stratus and Stratocumulus Regions Yanping He 153 pages Directed by Dr. Robert E. Dickinson A simple low cloud cover scheme is developed for the subtropical marine stratus and stratocumulus (MSC) regions. It is based on a modified CIN concept named the Lower Troposphere Available Dry Inhibition Energy (ADIN). The e-folder time for the local change of ADIN is found to be approximately 6 to 7 hours. On monthly and longer timescales, local productions of ADIN are balanced by local destructions of ADIN within lower troposphere. Dynamical transport of environmental dry static energy and surface evaporation lead to the variations of cloud top radiative cooling, which is a linear function of low cloud cover. Data analysis suggests that total ADIN dynamical transport plays the most important role in determining the seasonal variations and spatial variations of low cloud amounts¡£ The new scheme produces realistic seasonal and spatial variations of both EECRA ship observation and satellite observations in all MSC regions. It explains 25% more covariance than that using Klein-Hartmann (KH) scheme for monthly ISCCP low cloud amount near the Peruvian and Canarian region during the period from 1985 to 1997£¬it better represents the relationship between ENSO index and low cloud cover variations near the Peruvian region. When implemented into NCAR CAM3.1, it systematically reduces the model biases in the summertime spatial variations of low cloud amount and downward solar radiation in the Peruvian, California, and Canarian regions. Model simulated summertime cloud liquid water path, large scale precipitation, and surface fluxes are also significantly changed. A single predictor named Lower troposphere available thermal inhibition energy (ATIN) is also shown to be more skillful than the lower tropospheric stability in diagnosing low cloud stratiform clouds in the monthly and seasonal timescales. On synoptic timescale, dynamical transport of available dry inhibition energy and surface evaporation are better correlated with marine low cloud amount variations than ATIN and lower troposphere stability. The influence of boundary layer clouds, ocean surface SST, and large scale divergence on the stochastic dynamics of local ocean surface winds are addressed using QuikSCAT and AIRS satellite observations and a simple conceptual model in the southeast Pacific. The ocean surface pressure gradient depends on both the boundary layer height and temperature inversion strength. Marine boundary clouds are diagnosed using the cloud cover scheme developed in Chapter 2. The model successfully reproduces the observed mean state, the standard deviation, and skewness of local surface wind speeds in the southeast Pacific.

Hillmann, Elise de Castro January 2015 (has links)
Background: Most cervical cancer screening methods relies on colposcopy to confirm the diagnosis. Colposcopy suffers from the lack of availability, long patients’ displacement and waiting times. Objective: Evaluate the performance of the Cervical Digital Photography as an alternative method to colposcopy. Methods: Colposcopy and Cervical Digital Photography were performed in 228 women. The Cervical Digital Photographs were evaluated through internet by 3 colposcopy experts. The agreement between methods was calculated with kappa and percentages of agreement. Sensitivity, specificity, positive predictive value, negative predictive value and diagnostic accuracy were calculated for colposcopy and Cervical Digital Photography. Histology was the gold standard. Results: Cervical Digital Photography and colposcopy agreed in 89.9% of the cases (K=0.588). Cervical Digital Photography and colposcopy had comparable performances, sensitivity was 52.5% and 35.0%, specificity was 91.86% and 91.28%, positive predictive value was 60.0% and 48.3%, negative predictive value was 89.3% and 85.8%, and diagnostic accuracy was 84.4% and 80.7%, respectively. Conclusion: Cervical Digital Photography is a promising alternative method to colposcopy.
