A Novel Approach to Identify Candidate Imprinted Genes in Humans

Many imprinted genes are necessary for normal human development. Approximately 70 imprinted genes have been identified in humans. I developed a novel approach to identify candidate imprinted genes in humans using the premise that imprinted genes are often associated with nearby parent-of-origin-specific DNA differentially methylated regions (DMRs). I identified parent-of-origin-specific DMRs using sodium bisulfite-based DNA (CpG) methylation profiling of uniparental tissues, mature cystic ovarian teratoma (MCT) and androgenetic complete hydatidiform mole (AnCHM), and biparental tissues, blood and placenta. In support of this approach, the CpG methylation profiling led to the identification of parent-of-origin-specific differentially methylated CpG sites (DMCpGs) in known parent-of-origin-specific DMRs. I found new DMRs for known imprinted genes NAP1L5 and ZNF597. Most importantly, I discovered many new DMCpGs, which were associated with nearby genes, i.e., candidate imprinted genes. Allelic expression analyses of one candidate imprinted gene, AXL, suggested polymorphic imprinting of AXL in human blood.

It`s blameful to do nothing. You should catch the day and its possibilities. Every second, minute and day, you have the potential to be constantly productive. Do not lose a second of time that you can use to create an imprint of your existence. Do not waste your time when you have the opportunity to create something measurably valuable. Soon you are gone and the competition is great. Doing nothing is a task that fewer and fewer are devoted to, to a lesser extent. We are expected to always be accessible and keep up to date with what is happening around us. There is a constant occupation of our sense of mind, thoughtfulness and rest is unprioritized. We try to maximize the part of our time when we are productive and active to absurdity. The goal seems to make as big imprint of our existence as possible. Fast and more is applauded until the day we burn ourselves out. In my work, I investigate passivity as backlash in a faster world, in a world where focus is on production, efficiency and results. A world where everything worth valuing is measurable. What is really a desired result and is it always possible to measure? Who really gets that gain from our fast life? Can passivity and slowness act as an activist act? Can one passively create something measurable? Is burning time perhaps our time´s most provocative act? With my work, I want to question my constant bad conscience about being insufficient and my feeling of not doing enough. If I can´t do enough, I do nothing instead. / Det är skuldbelagt att göra ingenting. Du bör fånga dagen och dess möjligheter. Varje sekund, minut och dag har du potential att vara konstant produktiv. Förlora inte en sekund av tid som du kan använda till att skapa ett avtryck av din existens. Slösa inte med din tid när du har möjlighet att skapa något mätbart värdefullt. Snart är du borta och konkurrensen är stor. Att göra ingenting är en syssla som allt färre ägnar sig åt. Vi förväntas alltid vara tillgängliga och hålla oss uppdaterade om vad som händer runt oss. Det sker ständigt en ockupation av våra sinnesintryck. Eftertänksamhet och vila prioriteras bort. Vi försöker att maximera den del av vår tid då vi är produktiva och aktiva till absurditet. Målet verkar vara att hinna sätta ett så stort avtryck av vår existens som möjligt. Snabbare och mer applåderas till den dagen då vi bränner ut oss. I mitt arbete undersöker jag passivitet som motreaktion till en snabbare värld, i en värld där fokus ligger på produktion, effektivitet och resultat. I en värld där endast det som är mätbart ges ett värde. Vad är egentligen ett önskat resultat och är det alltid möjligt att mäta? Vem är det egentligen som får vinsten av vårt snabba liv? Kan passivitet och långsamhet fungera som en aktivistisk handling? Kan man passivt skapa något mätbart? Är ”att bränna tid”  kanske vår tids mest provokativa handling? Med mitt arbete vill jag ifrågasätta mitt konstanta dåliga samvete om att vara otillräcklig och min känsla av att jag inte gör tillräckligt. Om jag inte kan göra nog så gör jag istället ingenting.

Le Soulier de satin, tour à tour perçu comme oeuvre Dada, drame baroque ou pièce àclef, est une oeuvre dont l’étude de la création et la réception permet de mieuxappréhender les liens de Paul Claudel avec son temps. Dans cette pièce, Claudeleffectue la somme de l’héritage artistique de remise en cause de la société positiviste desa jeunesse et livre le récit de sa vocation poétique, enracinée dans la penséebaudelairienne et mallarméenne sur la modernité. La prégnance du paradigme picturalpour livrer son manifeste esthétique et l’utilisation de l’image sous toutes ses formespour entrer en dialogue avec le sens du texte vérifient la concordance d’intérêts entreClaudel et les artistes de sa génération. L’originalité du dramaturge réside dans sonrecours à l’image pour faire de son oeuvre un ex-voto qui retrace l’itinéraire de saconversion, l’inspiration thomiste le conduisant à privilégier l’image dans l’économiedu salut à l’oeuvre dans la pièce. L’oeuvre révèle enfin son affinité avec l’art modernepar son utilisation de procédés et de formes propres à l’art moderne, comme le collageet le montage, ou redécouverts par la modernité, comme l’empreinte ou la planéité,procédés et formes que Claudel emploie de manière polysémique et dialectique. Eninscrivant la question de sa vocation poétique, de sa conversion et de la réception de sonoeuvre par ses contemporains au coeur de la pièce, avec Le Soulier de satin, Claudeldonne une oeuvre qui s’inscrit pleinement dans le programme de la modernité tout enrévélant son originalité propre. / The Satin Slipper has been received alternately as a Dada work, a Baroque drama or a‘pièce à clé’, and studying both its first production and its reception allows one to graspbetter Claudel’s relationships with his days. In this work, Claudel adds up together theartistic legacy of his questioning the positivist society of his youth and gives thenarrative of his poetic vocation, which is rooted in the Baudelairean and Mallarméanthoughts on modernity. Claudel delivers his aesthetic manifesto through the primacy ofthe pictural paradigm and makes the reader establish a dialogue with the meaning of thetext, and all this makes him meet the concerns of the artists of his generation. Theplaywright’s originality lies in his resorting to the image to make his work into an exvoto that traces the itinerary of his conversion, as the Thomistic inspiration drives himto favour the image, in God’s plan at work in the play. Ultimately, the work discloses itscloseness to modern art through the use of devices and forms that are proper to modernart, such as collages and montages, or that were rediscovered by modernity, like theimprint or the planeness, that Claudel uses in a polysemous and dialectical way. Diploma thesis focuses on history analysis and today's CSR issues that multinational marketing corporations have to face up. It describes both CSR positives and negative responses to CSR implementation. It follows today's customer trends on markets and possibilities, how to win mainstresm customers for CSR challenges. Especially the trend of building CSR in the core business and in brands themselves is pointed out. The diploma work shows many examples of practising marketing with some social effect, from which both the company and the supported object can profit. Finally, the work describes some possibilities of CSR implementing and communicating. The practical part describes current CSR activities of Unilever company, including its Vitality mission, that is built in the whole business.

Identifikace osob pomocí otisku hlasu / Identification of persons via voice imprint

This work deals with the text-dependent speaker recognition in systems, where just a few training samples exist. For the purpose of this recognition, the voice imprint based on different features (e.g. MFCC, PLP, ACW etc.) is proposed. At the beginning, there is described the way, how the speech signal is produced. Some speech characteristics important for speaker recognition are also mentioned. The next part of work deals with the speech signal analysis. There is mentioned the preprocessing and also the feature extraction methods. The following part describes the process of speaker recognition and mentions the evaluation of the used methods: speaker identification and verification. Last theoretically based part of work deals with the classifiers which are suitable for the text-dependent recognition. The classifiers based on fractional distances, dynamic time warping, dispersion matching and vector quantization are mentioned. This work continues by design and realization of system, which evaluates all described classifiers for voice imprint based on different features.
