Le rôle des cellules gliales de Müller dans la mort des cellules ganglionnaires de la rétine par des mécanismes cellulaires non-autonomes

Les cellules gliales sont essentielles au fonctionnement du système nerveux. Dans la rétine, les cellules gliales de Müller assurent à la fois l’homéostasie du tissu et la protection des neurones, notamment celle des cellules ganglionnaires de la rétine (CGRs). L’hypothèse principale de la thèse est que les cellules de Müller joueraient un rôle primordial dans la survie neuronale tant au plan de la signalisation des neurotrophines/proneurotrophines par suite d’une blessure que lors des mécanismes d’excitotoxicité. Contrairement au brain-derived neurotrophic factor (BDNF), le nerve growth factor (NGF) n’est pas en mesure d’induire la survie des CGRs après une section du nerf optique. Le premier objectif de la thèse a donc été de localiser les récepteurs p75NTR et TrkA du NGF dans la rétine adulte et d’établir leur fonction respective en utilisant des ligands peptidomimétiques agonistes ou antagonistes spécifiques pour chacun des récepteurs. Nos résultats ont démontré que TrkA est surexprimé par les CGRs après l’axotomie, tandis que p75NTR est spécifiquement exprimé par les cellules de Müller. Alors que NGF n’est pas en mesure d’induire la survie des CGRs, l’activation spécifique de TrkA par des ligands peptidomimétique est nettement neuroprotectrice. De façon surprenante, le blocage sélectif de p75NTR ou l’absence de celui-ci protège les CGRs de la mort induite par l’axotomie. De plus, la combinaison de NGF avec l’antagoniste de p75NTR agit de façon synergique sur la survie des CGRS. Ces résultats révèlent un nouveau mécanisme par lequel le récepteur p75NTR exprimé par les cellules gliales de Müller peut grandement influencer la survie neuronale. Ensuite, nous avons voulu déterminer l’effet des proneurotrophines dans la rétine adulte. Nous avons démontré que l’injection de proNGF induit la mort des CGRs chez le rat et la souris par un mécanisme dépendant de p75NTR. L’expression de p75NTR étant exclusive aux cellules de Müller, nous avons testé l’hypothèse que le proNGF active une signalisation cellulaire non-autonome qui aboutit à la mort des CGRs. En suivant cette idée, nous avons montré que le proNGF induit une forte expression du tumor necrosis factor α (TNFα) dans les cellules de Müller et que l’inhibition du TNF bloque la mort neuronale induite par le proNGF. L’utilisation de souris knock-out pour la protéine p75NTR, son co-récepteur sortiline, ou la protéine adaptatrice NRAGE a permis de montrer que la production de TNF par les cellules gliales était dépendante de ces protéines. Le proNGF semble activer une signalisation cellulaire non-autonome qui cause la mort des neurones de façon dépendante du TNF in vivo. L’hypothèse centrale de l’excitotoxicité est que la stimulation excessive des récepteurs du glutamate sensibles au N-Methyl-D-Aspartate (NMDA) est dommageable pour les neurones et contribue à plusieurs maladies neurodégénératives. Les cellules gliales sont soupçonnées de contribuer à la mort neuronale par excitotoxicité, mais leur rôle précis est encore méconnu. Le dernier objectif de ma thèse était d’établir le rôle des cellules de Müller dans cette mort neuronale. Nos résultats ont démontré que l’injection de NMDA induit une activation du nuclear factor κB (NF-κB) dans les cellules de Müller, mais pas dans les CGRs, et que l’utilisation d’inhibiteurs du NF-κB empêche la mort des CGRs. De plus, nous avons montré que les cellules de Müller en réaction à l’activation du NF-κB produisent la protéine TNFα laquelle semble être directement impliquée dans la mort des CGRs par excitotoxicité. Cette mort cellulaire se produit principalement par l’augmentation à la surface des neurones des récepteurs AMPA perméables au Ca2+, un phénomène dépendant du TNFα. Ces donnés révèlent un nouveau mécanisme cellululaire non-autonome par lequel les cellules gliales peuvent exacerber la mort neuronale lors de la mise en jeu de mécanismes excitotoxiques. / Glial cells are essential for the functioning of the nervous system. In the retina, the Müller glial cells ensure the homeostasis of this tissue as well as the protection of neurons including the retinal ganglion cells (RGCs). The main hypothesis of this thesis is that Müller cells play a predominant role in neuronal survival both at the levels of neurotrophin/proneurotrophin signaling following injury and excitotoxic mechanisms. Unlike the brain-derived neurotrophic factor (BDNF), the nerve growth factor (NGF) is unable to induce RGCs survival following optic nerve transection. The first objective of the thesis was therefore to describe the expression of the two receptors of NGF, p75NTR and TrkA, in the adult retina and to address their functional role by using peptidomimetic agonistic or antagonistic ligands specific for each receptor. Our results showed that TrkA is overexpressed by RGCs following axotomy, whereas p75NTR is specifically expressed by Müller cells. While NGF by itself does not promote RGC survival, selective activation of TrkA receptors using peptidomimetic ligands is markedly neuroprotective. Surprisingly, selective blockers of p75NTR, or the absence of p75NTR, protect RGCs from axotomy-induced death. Moreover, combination of NGF or TrkA agonists with p75NTR antagonists functions synergistically to enhance RGC survival. These results reveal a new mechanism by which p75NTR expression by Müller glia may profoundly influence neuronal survival. Next, we wanted to address the effect of proneurotrophins in the adult retina. We showed that injection of proNGF induces the death of RGCs in rats and mice by a p75NTR-dependent signaling mechanism. Expression of p75NTR in the adult retina being confined to Müller glial cells, we tested the hypothesis that proNGF activates a non-cell autonomous signaling pathway to induce RGC death. Consistent with this notion, we showed that proNGF induced a robust expression of tumor necrosis factor α (TNFα) in Müller cells, and that genetic or biochemical ablation of TNFα blocked proNGF-induced death of retinal neurons. Mice rendered null for p75NTR, its co-receptor sortilin, or the adaptor protein NRAGE were defective in proNGF-induced glial TNFα production and did not undergo proNGF-induced retinal ganglion cell death. We concluded that proNGF activates a non-cell autonomous signaling pathway that causes TNFα-dependent death of retinal neurons in vivo. The central hypothesis of excitotoxicity is that excessive stimulation of neuronal N-Methyl-D-Aspartate (NMDA)-sensitive glutamate receptors is harmful to neurons and contributes to a variety of neurological disorders. Glial cells have been proposed to participate in excitotoxic neuronal loss, but their precise role is poorly defined. In this in vivo study, we showed that NMDA induces a strong NF-κB activation in Müller glia, but not in retinal neurons. Intriguingly, NMDA-induced death of retinal neurons was effectively blocked by inhibitors of NF-κB activity. We demonstrated that TNFα protein produced in Müller glial cells via an NMDA-induced NF-κB dependent pathway plays a crucial role in the excitotoxic loss of retinal neurons. This cell loss occurs mainly through a TNFα-dependent increase in Ca2+-permeable AMPA receptors on susceptible neurons. Thus, our data reveal a novel non-cell-autonomous mechanism by which glial cells can profoundly exacerbate neuronal death following excitotoxic injury.

Assessing the dose received by the victims of a radiological dispersal device with Geiger-Mueller detectors

This research investigates the use of G-M counters to triage the individuals who have been exposed to a Radiological Dispersal Device (RDD). Upon being exposed to an RDD, inhalation of the airborne radionuclide is a method which someone can receive a considerable amount of dose. Bioassay via analysis of excreta is a commonly used method of determining the dose received, yet it would be cumbersome if there are a large number of people needing to be screened. An in vivo method must be considered so that a non-intrusive and more efficient triaging method can be implemented. Whole body counters are commonly used in counting facilities as an in vivo bioassay method, yet they are limited in number and not easily portable. Therefore, a more portable and more common detection device should be considered. G-M survey meters are common devices that are highly portable, making them ideal candidates to fulfill this necessity. The ease of use contributes to the viability of the device as a portable, in vivo screening device. To analyze this detector, a Monte Carlo model of the detector was created to be used in simulations with the Medical Internal Radiation Dose phantoms. The detector was placed in a few locations on the phantoms. Four locations were strategically chosen for detector placement: the posterior upper right torso, the anterior upper right torso, the lateral upper thigh, and the anterior of the neck. Six phantoms were considered: Reference Male, Female, Adipose Male, Adipose Female, Post Menopausal Adipose Female, and a Child. Six radionuclides were investigated: Am-241, Co-60, Cs-137, I-131, Ir-192, and Sr-90. The nuclides were distributed throughout the phantoms according to Dose and Risk Calculation Software, a code that determines how a radionuclide is distributed over time upon inhalation, ingestion, or injection. A set of time dependent guidelines were developed, determining the count rate per unit dose inhaled for each detector location and phantom type.

Como consequência os jovens apresentam dificuldades em compreender relações entre a cultura científica e a cultura social, levando-os muitas vezes a não conseguir refletir sobre os significados do conhecimento científico. Diante desse cenário um dos principais objetivos desse trabalho é o de refletir sobre o potencial de situações de ensino que podem ser trabalhadas em aulas de Física que visem a construção de uma cultura científica voltada à valorização de uma formação complexa. Em outras palavras, defendemos uma educação cientifica que promova nos jovens competências e autonomia de associar os saberes escolares, dando-lhes significados dentro de contexto social e cultural. Para alcançar esse objetivo os principais referenciais que fundamentam esse trabalho seguem os pressupostos teóricos do pensamento complexo proposto por Morin (2011) e do vetor epistemológico proposto por Bachelard (1978). A partir desses conceitos, apoiamo-nos nas críticas que ambos os autores fazem à concepção de educação como mera aquisição do conhecimento em um sistema fechado e na defesa de promover uma educação voltada ao desenvolvimento de um pensamento aberto e dinâmico. Para elaborar situações de ensino que contribuam para a educação Crítica, Complexa e Reflexiva (WATANABE, 2012), foi desenvolvida, em parceria com o GrECC ¿ Grupo de Ensino de Ciências e Complexidade - uma proposta de ensino que trata das questões socioambientais por meio dos conteúdos de Física. A partir das aulas originais realizou-se uma releitura da proposta à luz de reflexões sobre a construção do conhecimento científico como um processo marcado pelas incertezas e indeterminações. A releitura dessas aulas foi desenvolvida em uma escola pública com alunos de diferentes turmas do segundo ano do Ensino Médio. A partir dos resultados, nos quais é possível identificar que os estudantes passam a articular diferentes áreas do conhecimento na construção de seus argumentos diante da temática ambiental, observa-se que propostas de ensino visando uma formação voltada à ideia de complexidade na ciência, apresentam um bom resultado acerca de superar o paradigma da fragmentação e desarticulação dos saberes. / One of the problems arising in science teaching in Brazil, in general, is still training people struggling to articulate the diverse knowledge that were worked out during the long process of their school life. As a result young people have difficulties in understanding links between scientific culture and social culture, often failing to reflect on the meanings of scientific knowledge. In this scenery a major goal of this work is to reflect on the potential of teaching situations that can be worked in Physics classes aimed at building a scientific culture dedicated to the appreciation of a complex formation. In other words, we advocate a scientific education that promotes the youth skills and autonomy to associate the school knowledge, giving them meaning within social and cultural context. To achieve this objective the main benchmarks that underlie this work follow the theoretical assumptions of complex thinking proposed by Morin (1991) and epistemological vector proposed by Bachelard (1978). From these concepts, we support you in critical that both authors make the concept of education as simple acquisition of knowledge in closed system and advocacy to promote an education to the development of an open and dynamic thinking. To develop teaching situations that contribute to the formation of critical, reflective and complex, it was developed in partnership with the GrECC - Science Teaching Group and complexity- a proposal for a school that deals with environmental issues by means of Physical contents. From the original class carried out a proposal of reinterpretation in the light of reflections on the construction of scientific knowledge as a process marked by uncertainties and indeterminate. Rereading these classes it was developed in a public school with students from different second-year high school classes. From the results, where it is possible to identify which students spend articulate different areas of knowledge in building their arguments on the environmental theme, it seems that proposals for education aimed at training focused on the idea of complexity in science, have good potential to overcome the paradigm of fragmentation and disarticulation of knowledge.

Explorando os princípios da teoria constitutiva, algumas hipóteses ad hoc e, em especial, o princípio de entropia Müller-Liu, modelos termo-eletroquímicos são propostos para soluções eletrolíticas e para as regiões de bulk e dupla camada de um sistema eletroquímico. A influência das interações íon-íon e íon-solvente sobre a dinâmica da mistura é considerada através da termodinâmica de contínuos polares e as equações de balanço são apropriadamente postuladas. Além disso, equações fenomenológicas são apresentadas para uma solução eletrolítica diluída e isotrópica e as condições para o equilíbrio termodinâmico local do bulk e da dupla camada são investigadas. Comparando-se as desigualdades residuais de entropia obtidas para cada região, foi demonstrado que alguns processos, tais como os fenômenos de difusão térmica, termoeletricidade e eletroforese, desenvolvem-se somente na dupla camada. Consequentemente, o estado de equilíbrio termodinâmico local na dupla camada requer condições termodinâmicas mais severas do que no bulk. Apesar das equações constitutivas serem as mais simples possíveis, os modelos constitutivos propostos para as regiões de dupla camada e bulk são fisicamente consistentes e mais abrangentes do que os modelos eletroquímicos usuais, visto que as equações e relações termodinâmicas obtidas não se limitam às condições limites e nem de equilíbrio. Portanto, a descrição termodinâmica mostrada neste trabalho pode estimular químicos e engenheiros químicos a usar a abordagem contínua no estudo da mobilidade iônica em meios bio e geológicos, fluxos de íons e de outras espécies químicas através de membranas e processos cujas transferências de calor e massa são intensificadas por campos eletromagnéticos / Abstract: This work concerns the thermodynamics of electrochemical systems. Exploiting the principles of constitutive theory, few expedient assumptions, and, in special, the Müller-Liu entropy principle, a thermo-electrochemical continuum model is proposed for electrolyte solutions as well as for the bulk and double layer regions of an electrochemical system. The influence of ion-ion and ion-solvent interactions on the mixture dynamics is taken into account through the thermodynamics of polar materials and balance laws for an electrochemical system are accordingly stated. In addition, phenomenological equations are schemed for a dilute and isotropic electrolyte solution, and the conditions for local thermodynamic equilibrium of bulk and double layer regions are investigated. Comparing the residual entropy inequalities obtained for each region of an electrochemical system, it is shown that some mechanisms develop only in the double layer, such as the thermal diffusion, thermoelectricity and electrophoresis phenomena. As a consequence, the local thermodynamic equilibrium state in the double layer requires stricter conditions than in the bulk. Although the constitutive equations are the simplest possible, the constitutive models proposed for the double layer and bulk regions are physically consistent and more comprehensive than the usual models since the emerging equations do not constrain themselves to equilibrium neither limiting conditions. Therefore, the thermodynamic description provided in this work may stimulate chemists and chemical engineers to take advantage of it to study the flow of ions and other chemical species across cell membranes, ionic mobility in bio and geological media, and processes whose heat and mass transfers are enhanced by electromagnetic fields / Doutorado / Físico-Química / Doutora em Ciências

I detta arbete undersöks två etablerade och en experimentell metod för bestämmande av läsbarheten hos texter. Metoderna undersöks genom att de appliceras på två litterära verk: Atemschaukel av Herta Müller och Berühre mich. Nicht. av Laura Kneidl. Metoderna som undersöks är läsbarhetsformeln Flesch-Reading-Ease (meningslängd och ordlängd) och ordfrekvens (hur vanliga orden är). Dessa båda läsbarhetsundersökningar är väletablerade och har använts flitigt. I detta arbete testas även en ny möjlig läsbarhetsfaktor: Upprepningar av ord, där upprepningar av innehållsord räknas. Resultaten av analyserna enligt de olika metoderna sammanställs och jämförs för att besvara två frågor: 1. Vilket verk förefaller mest lättläst enligt de två etablerade läsbarhetsundersökningarna? 2. Samvarierar mängden upprepningar med resultaten från de andra två läsbarhetsundersökningarna? Resultaten ger en något splittrad bild: Atemschaukel är enligt Flesch-Reading-Ease det mest lättlästa verket, medan ordfrekvensen visar på motsatt förhållande: Berühre mich. Nicht. har de genomsnittligt vanligare orden. Huruvida upprepning av ord har ett samband med läsbarheten är svårt att avgöra baserat enbart på resultaten i denna undersökning. Användandet av språkliga stilfaktorer för eventuellt att läsbarhetstest inte lämpar sig för undersökning av skönlitterära texter.

Elevated amh Gene Expression in the Brain of Male Tilapia (Oreochromis niloticus) during Testis Differentiation

Anti-müllerian hormone (AMH) is expressed in male embryos and represses development of müllerian ducts during testis differentiation in mammals, birds and reptiles. Amh orthologues have been identified in teleosts despite them lacking müllerian ducts. Previously we found sexually dimorphic aromatase activity in tilapia brains before ovarian differentiation. This prompted us to search for further dimorphisms in tilapia brains during sex differentiation and see whether amh is expressed. We cloned the tilapia amh gene and found that it contains 7 exons but no spliced forms. The putative protein presents highest homologies with Amh proteins of pejerrey and medaka as compared to other Perciformes. We analysed amh expression in adult tissues and found elevated levels in testes, ovary and brain. Amh expression was dimorphic with higher levels in XY male brains at 10–15 dpf, when the gonads were still undifferentiated and gonadal amh was not dimorphic. Male brains had 2.7-fold higher amh expression than gonads. Thereafter, amh levels decreased in the brain while they were up-regulated in differentiating testes. [ES] El presente estudio se centra en el análisis de la praxis teatral, a partir del montaje esceno-performativo: Descripción de un cuadro, texto de Heiner Müller, y dirección de Madeleine Loayza, con la finalidad de exponer la importancia de la «tensión erótica» (entendida como energía vital asociada a Eros) presente en la poíesis del acontecimiento teatral. Nos enfocaremos en los dispositivos contrahegemónicos (a los que caracteriza su gesto irreverente y crítico) presentes en el fenómeno sensoperceptivo y receptivo, el mismo que involucra la relación del actor-operador/performer con los diversos elementos que intervienen en la producción de dicha poíesis y su alcance en el espectador. La investigación incluye la poíesis convivial, productiva y la poíesis receptiva enfocadas en el proceso y en el resultado. En este contexto se observa, analiza e interpreta el modo en que acontece dicha tensión, emplazándonos desde el rol de artista-investigadora/espectadora. El estudio concluye con el soporte conceptual y el diseño de la investigación-creación: Mujer en llamas; donde la investigadora se asume como artista-investigadora/directora, actriz y dramaturga. Observar cómo se constituye el fenómeno sensoperceptivo y receptivo de la poíesis, al develarse el modo en que opera la «tensión erótica» y su alcance en el espectador, en tanto dispositivo de resistencia contrahegemónico por medio de mecanismos y formas de producción liminales, para lo cual se ponen en discusión las características de operación del dispositivo, de acuerdo con las particularidades del montaje propuesto. El estudio contempla, desde una perspectiva analítica, las teatralidades contemporáneas, pertenecientes al campo de la Teatrología, con énfasis en la dimensión performativa del signo teatral que destaca los aspectos espacio/temporales y kinésicos. Nos referirnos al análisis de la dinámica de la ejecución del signo en escena (que involucra al espectador), más que limitarnos a identificar la estructura de los signos teatrales; esta perspectiva desplaza al eje de la literatura dramática como referente canónico para abordar la experiencia escénica. Enfatizar el rol del cuerpo en el fenómeno sensoperceptivo y receptivo, entre el actor y el espectador, nos remite a la visión nietzscheana que toma como referente la tragedia griega (siglos V y IV a. C), retomada luego por Artaud y otros pensadores de su tiempo desde el campo de la filosofía; esta perspectiva potencia el rol del espectador en la dinámica escénica desde la auto-alteración y la capacidad crítica y autocrítica. De allí que el concepto liminalidad presente en Descripción de un cuadro, posea un carácter político importante en tanto contrahegemónico como antiestructura. Los parámetros para observar la operatividad de la «tensión erótica» se advierten en los dispositivos poéticos, que configuran su poíesis en las modalidades productiva, receptiva y convivial. En el montaje Descripción de un cuadro, el dramaturgismo, las prácticas performáticas; el análisis del «convivio» entre los diversos materiales o matrices que intervienen en el proceso de factura de la obra, como dispositivos poéticos, constituyen los pilares que sustentan el proceso de investigación-creación. En lo que respecta al soporte conceptual y exposición del diseño del montaje Mujer en llamas, este se desarrolla a partir de la metodología que guía el enfoque del presente estudio, con el objetivo que pueda ser incorporada y replicada en otros procesos creadores. / [CA] El present estudi es centra en l'anàlisi de la praxi teatral, a partir del muntatge esceno-performatiu: Descripció d'un quadre, text de Heiner Müller, i dirección de Madeleine Loayza amb la finalitat d'exposar la importància de la «tensió eròtica» (entesa com a energia vital associada a Eros) present en la poiesis de l'esdeveniment teatral. Ens centrarem en els dispositius contrahegemónicos (caracteritzats pel caràcter irreverent i crític) presents en el fenomen sensoperceptiu i receptiu, el fenomen mateix que involucra la relació de l'actor-operador/performer amb els diversos elements que intervenen en la producció d'aquesta poíesis i el seu abast en l'espectador. La investigació inclou la poíesis convivial, productiva i la poíesis receptiva enfocades en el procés i en el resultat. En aquest context s'observa, analitza i interpreta la manera en què esdevé aquesta tensió, emplaçant-nos des del rol d'artista-investigadora/espectadora. L'estudi conclou amb el suport conceptual i el disseny de la investigació-creació: Dona en flames; on la investigadora s'assumeix com a artista-investigadora/directora, actriu i dramaturga. Observar com es constitueix el fenomen sensoperceptivo i receptiu de la poiesis, al develarse la manera en què opera la "tensió eròtica" i el seu abast en l'espectador, en tant dispositiu de resistència contrahegemònic per mitjà de mecanismes i formes de producció liminals, per a això es posen en discussió les característiques d'operació del dispositiu d'acord amb les particularitats del muntatge proposat. L'estudi contempla des d'una perspectiva analítica les teatralitats contemporànies pertanyents al camp de la Teatrología, posant especial èmfasi en la dimensió performativa del signe teatral que destaca els aspectes espai/temporals i kinèsics. Ens referirm a l'anàlisi de la dinàmica de l'execució del signe en escena (que involucra a l'espectador), més que limitar-nos a identificar l'estructura dels signes teatrals; desplaçant-nos de l'eix de la literatura dramàtica com a referent canònic a l'experiència escènica. Emfatitzar el rol del cos en el fenomen sensoperceptiu i receptiu, entre l'actor i l'espectador, ens remet a la visió nietzscheana que pren com a referent la tragèdia grega (segles V i IV a. C), represa després per Artaud desde l'escena i per altres pensadors del seu temps des del camp de la filosofia; perspectiva es potencia el rol de l'espectador en la dinàmica escènica, des de l'acte-alteració i la seua capacitat crítica i autocrítica. Per aquest motiu el concepte liminalitat present en Descripció d'un quadre, poseeix un caràcter polític important en tant contrahegemónico com a antiestructura. Els paràmetres per a observar l'operativitat de la «tensió eròtica» s'adverteixen en els dispositius poètics que configuren el seu poíesis en les modalitats productiva, receptiva i convivial. En el muntatge Descripció d'un quadre, el dramaturgismo, les pràctiques performatives; l'anàlisi del "convivio" entre els diversos materials o matrius que intervenen en el procés de creació de l'obra, com a dispositius poètics, constitueixen els pilars que sustenten el procés d'investigació-creació. Pel que fa al suport conceptual i exposició del disseny del muntatge Dona en flames, aquest es desenvolupa a partir de la metodologia que guia la perspectiva del present estudi, amb l'objectiu que puga ser incorporada i replicada en altres processos creatius. / [EN] This research focuses on the analysis of theatrical praxis, coming from the sceno-performative staging of: Description of a painting, written by Heiner Müller, and directed by Madeleine Loayza, in order to expose the importance of the "erotic tension" (understood as vital energy associated with Eros) present in the poiesis of the theatrical event. We will focus on the against-hegemonic devices (which characterizes its irreverent and critical gesture) present in the sensoperceptive and receptive phenomenon, which also involves the relationship between the actor-operator/performer with the various elements that intervene in the production the mentioned poiesis and its scope to the spectator. For this reason, the research includes convivial productive poíesis and receptive poíesis, focused on the process and the result. Under this context, the way in which this tension occurs is observed, analyzed and interpreted, summoning us from the role of artist-researcher/spectator. The study concludes with the conceptual support and design of the research-creation: Woman on fire; where the researcher is assumed as artist-searcher/director, actress and playwright. Observes how the senso-perceptive and receptive phenomenon of poíesis is constituted, the way in which "erotic tension" operates and its scope to the spectator is revealed, as a device of against-hegemonic resistance that uses mechanisms and forms of liminal production, for which the operating characteristics of the device are discussed according to the particularities of the proposed assembly. The research considers contemporary theatricalities as an analytical approach, that belong to the field of Theatrology, with emphasis on the performative dimension of the theatrical sign that highlights the spatial/temporal and kinesic aspects. We are interested in analyzing the dynamics of the enforcement of the sign on stage (which involves the viewer), rather than limiting ourselves to identify the structure of the theatrical signs; this perspective displaces the axis of dramatic literature as a canonical reference to address the stage experience. Giving emphasis to the role of the body in the sensory-perceptual and receptive phenomenon, between the actor and the spectator, refers us to the Nietzschean vision that takes as a reference the Greek tragedy (5th and 4th centuries BC), later taken up again by Artaud and other thinkers and philosophers of this period; In this way, the role of the spectator in the scenic dynamics is also enhanced, from self-alteration and their critical and self-critical capacity. Therefore, the concept of liminality in Description of a painting has an important political character as bot: against-hegemonic and anti-structural. The parameters for observing the operability of the "erotic tension" are seen in the poetic, devices that configure its poiesis in the productive, receptive and convivial modalities. In the staging of Description of a painting; the playwriting, the performative practices, the analysis of the "conviviality" between the various materials or matrices that intervene the invoice process of the work as poetic devices, constitute the pillars that support the research-creation process. Regarding the conceptual support and design exhibition for the staging of Woman on Fire, it is developed by the methodology that guides the focus of this study, with the aim that it can be incorporated and replicated in other creative processes. / Loayza Cabezas, GM.
